RESUMO

Prior studies have shown that experts possess an excellent ability for action anticipation. However, it is not clear how experts process the discrepancies between predicted outcomes and actual outcomes. Based on Bayesian theory, Experiment 1 in the current study explored this question by categorizing unexpected outcomes into gradually increasing discrepancies and comparing the performance of experts and novices on a congruence discrimination task. Our behavioral analysis revealed that experts outperformed novices significantly in detecting these discrepancies. The following electroencephalogram study in Experiment 2 was conducted focused exclusively on experts to examine the role of theta wave oscillations within the mid-frontal cortex in processing varying levels of discrepancy. The results showed that reaction time and theta oscillations gradually increased as the magnitude of discrepancy increased. These findings indicate that compared to the novices, experts have a better ability to perceptual the discrepancy. Also, the magnitude of discrepancies induced an increase in mid-frontal theta in experts, providing greater flexibility in their response strategies.

Large capacitive loading of electrodes induces massive error current and imperfect settling in the electrochemical signal acquisition process, leading to inaccurate acquisition results. To efficiently mitigate this inaccuracy, this paper presents a current-and-voltage dual-mode acquisition technique in which a voltage front-end (VFE) is employed to acquire the electrode voltage error and compensate the nonlinearity induced by the electrode capacitive loading. Therefore, the gain and bandwidth requirements of the current front end (CFE) can be relaxed to reduce the complexity and power consumption. With a relieved gain requirement, an inverter-based capacitive trans-impedance amplifier (IB-CTIA) is adopted to boost the input transconductance for low-noise design. By reusing the supply current, the IB-CTIA effectively achieves a low input-referred current noise of 3.9 pArms and a dynamic range (DR) of 126 dB with only 18-µW static power. The prototype chip is fabricated in a 180-nm CMOS process. Interleukin-6 immunoassays (IL-6) are implemented to verify the chip's performance. With the proposed nonlinear error compensation, the correlation coefficient of the detection result is improved from 0.951 to 0.980 and the limit of detection (LoD) is reduced from 8.31 pg/mL to 6.90 pg/mL.

Cisplatin is commonly used for the chemotherapy of tongue squamous cell carcinoma (TSCC); however, adverse side effects and drug resistance impact its therapeutic efficacy. Capsaicin is an active ingredient in chili peppers that exerts antitumor effects, whether it exerts antitumor effects on cisplatin-resistant cells remains unknown. Therefore, in this study, we investigated the effect of capsaicin on cisplatin resistance in TSCC cells and explored the underlying mechanisms. A cisplatin-resistant TSCC cell line was established by treated with increasing cisplatin concentrations. Combined treatment with cisplatin and capsaicin decreased the glucose consumption and lactate dehydrogenase activity and increased the adenosine triphosphate production both in vitro and in vivo, suggesting the inhibition of the Warburg effect. Moreover, this combined treatment induced cell apoptosis and significantly upregulated the levels of proapoptotic proteins, such as Bax, cleaved caspase-3, -7, and -9, and apoptosis-inducing factor. In contrast, levels of the antiapoptotic protein, Bcl-2, were downregulated. Additionally, LKB1 and AMPK activities were stimulated, whereas those of AKT and mTOR were suppressed. Notably, AMPK knockdown abolished the inhibitory effects of capsaicin and cisplatin on the AKT/mTOR signaling pathway and Warburg effect. Overall, combined treatment with capsaicin and cisplatin reversed cisplatin resistance by inhibiting the Warburg effect and facilitating mitochondrial-dependent apoptosis via the AMPK/AKT/mTOR axis. Our findings suggest combination therapy with capsaicin and cisplatin as a potentially novel strategy and highlight capsaicin as a promising adjuvant drug for TSCC treatment.

Purpose: Parent involvement is crucial for tailored early intervention programs. The Hanen More Than Words (HMTW) program is a parent-implemented language intervention for autistic children. The current study examined the effectiveness of the HMTW program delivered online among Chinese families. Methods: Using a randomized controlled trial design, 22 Chinese families of autistic children in Hong Kong completed the trial. Baseline and post-intervention assessments were conducted to measure changes in parent-child interaction, parents' use of linguistic facilitation techniques (LFTs), and children's communication skills. Additionally, the influence of parental self-efficacy and parenting stress on treatment outcomes was explored. Results: The intervention group demonstrated significant improvements in parent-child attention synchrony. Although the treatment effect on children's spontaneous communication was not significant, the intervention group showed a larger effect size compared to the controls. The treatment outcomes were mainly influenced by the parents' initial levels of self-efficacy but not by parenting stress. Conclusion: These findings provide preliminary evidence of the effectiveness of the online-delivered HMTW program for Chinese parents of autistic children. Further research involving a larger sample and focusing on long-term effects is needed.

The congruency judgments in action understanding helps individuals make timely adjustments to unexpected occurrence, and this process may be influenced by emotion. Previous research has showed contradictory effect of emotion on conflict processing, possibly due to the degree of relevance between emotion and task. However, to date, no study has systematically manipulated the relevance to explore how emotion affects congruency judgments in action understanding. We employed a cue-target paradigm and controlled the way emotional stimuli were presented on the target interface, setting up three experiments: emotion served as task-irrelevant distractor, task-irrelevant target and task-relevant target. The results showed that when emotion was irrelevant to the task, it impaired congruency judgements performance, regardless of a distractor or a target, while task-relevant emotion facilitated this process. These findings indicate that the impact of emotion on congruency judgements during action understanding depends on the degree of emotion-task relevance.

Objective: Time perception is a critical point for curling athletes to have in order to successfully complete interactions between themselves and their environment. Exploring the relationship between the accuracy of duration judgment and curling athletes' performance is helpful to reveal the influencing factors on their performance and to provide a reference for the training of athletes' delivery performance. Methods: Thirty curling athletes and 30 non-athletes were recruited as participants. Using 3D modeling technology, curling videos of different situations were presented to the participants as stimulus information, and the participants were required to complete the duration judgment task. The neural activation of the participants during the entire process of duration judgment was recorded using electroencephalogram (EEG) equipment. The performance of the 30 curlers participating in the experiment was measured. Variance analyses were conducted on the collected behavioral and EEG data, and correlation and regression analyseswere conducted between behavioral data and delivery performance. Results: The accuracy of the distance judgment of curlers was higher than that of non-curlers (P < 0.05). In the stimulus video presentation stage, the power in the alpha band of curlers was higher than that of non-athletes (P < 0.05). In the task decision stage, the power in the alpha band of curlers was higher than that of non-athletes (P < 0.05), and the power in the theta band was higher than that of non-athletes (P < 0.05). There was a correlation between the accuracy of the curlers' perception of specific situational time intervals and the accuracy of delivery (P < 0.05). Regression analysis results were y = 3.422 + 1.415x. Conclusion: The accuracy of curling athletes' duration judgment is high in a specific situation. There is a correlation between the accuracy of duration judgment and delivery performance in a specific situation: the higher the accuracy of specific duration perception, the higher the performance accuracy of delivery. The cognitive strategies adopted by curlers differ from those adopted by non-athletes in the completion of duration judgment. Specifically, in a specific situation, fewer attention resources are utilized in the stimulus presentation and decision-making stages, while more memory resources are utilized in the decision-making stage to ensure higher accuracy of interval judgment. This study provides a new idea for exploring the causes of curling athletes' excellent technical performance and provides a reference for future curling research on competition training practice. Given the limitations of mobile EEG devices in this study, future studies can measure neural activity during actual delivery preparation and execution in an environment of high ecological validity to obtain more direct evidence.

Congruency monitoring of action occurs in individuals with relevant motor experience while observing actions. However, it remains unclear whether congruency monitoring can occur at the motor level and the extent to which expertise contributes. Here, we examined the behavioral performance and electrophysiological brain activity of individuals with and without domain-specific expertise when judging the action congruency of occluded video clips of a snowboard halfpipe trick and normal walking. For the halfpipe trick, experts exhibited better task performance and greater midline theta oscillations before possible incongruency compared with controls. Source reconstruction for the theta oscillation revealed a stronger activation in the middle and superior frontal gyrus for experts in response to incongruency compared with controls. Incongruent halfpipe actions elicited a higher N400 amplitude in experts compared with congruent actions, while no such differences were observed in controls. Source reconstruction revealed the activation in the board frontal regions and middle temporal gyrus for experts. These findings suggest that congruency monitoring can occur at the motor level during action observations and is modulated by individual expertise. The modulation of expertise reflects in the special N400 effect and midline theta oscillation.

Although motor performance training often involves action observation, it has been controversial whether individual aesthetic sport athletes benefit more from watching videos of discrete maneuvers (DMs) or continuous runs (CRs). In the present study, half-pipe snowboarding athletes completed a visual 2-back task with DM and CR conditions. To explore the neural mechanisms of action memory processing, brain hemodynamic activity during the task was monitored with functional near-infrared spectroscopy (fNIRS). Compared to watching CR videos, watching DM videos tended to yield better action memory performance and greater activation in the middle temporal gyrus to these athletes, suggesting that watching DM videos may have a tendency to improve action memory more effectively. Evidence of two pathways underlying half-pipe snowboarding action processing was obtained. Watching of CR videos and watching of DM videos might be associated with activation of more sensorimotor regions and more semantic regions, respectively, during memory consolidation.

The caveolin-3 (CAV3) protein is known to be specifically expressed in various myocytes, and skeletal muscle consumes most of the blood glucose as an energy source to maintain normal cell metabolism and function. The P104L mutation in the coding sequence of the human CAV3 gene leads to autosomal dominant disease limb-girdle muscular dystrophy type 1C (LGMD-1C). We previously reported that C2C12 cells transiently transfected with the P104L CAV3 mutant exhibited decreased glucose uptake and glycogen synthesis after insulin stimulation. The present study aimed to examine whether the P104L mutation affects C2C12 cell glucose metabolism, growth, and proliferation without insulin stimulation. C2C12 cells stably transfected with CAV3-P104L were established, and biochemical assays, western blot analysis and confocal microscopy were used to observe glucose metabolism as well as cell growth and proliferation and to determine the effect of the P104L mutation on the PI3K/Akt signaling pathway. Without insulin stimulation, C2C12 cells stably transfected with the P104L CAV3 mutant exhibited decreased glucose uptake and glycogen synthesis, decreased CAV3 expression and reduced localization of CAV3 and GLUT4 on the cell membrane. The P104L mutant significantly reduced the cell diameters, but accelerated cell proliferation. Akt phosphorylation was inhibited, and protein expression of GLUT4, p-GSK3ß, and p-p70s6K, which are molecules downstream of Akt, was significantly decreased. The CAV3-P104L mutation inhibits glycometabolism and cell growth but accelerates C2C12 cell proliferation by reducing CAV3 protein expression and cell membrane localization, which may contribute to the pathogenesis of LGMD-1C.
