RESUMO

In China, aquatic supply chain network design does not include the green concept or the coordination of environmental and economic performance. Sea cucumber (Apostichopus japonicus) is an aquatic product of high economic value; however, studies on sea cucumber supply chain network optimization are lacking. This study is the first to design the sea cucumber supply chain and construct an optimization model. Considering the characteristics of the sea cucumber industry, LCA for Experts software and the CML-IA-Aug. 2016-world method were used to assess each aquaculture model's global warming potential (GWP), as the environmental performance indicator. In addition, multi-objective genetic algorithm (MOGA) coupled with Modified Technique for Order of Preference by Similarity to Ideal Solution (M-TOPSIS) integrates yield production, economic benefits, and environmental performance. The results demonstrated that cage seed rearing (CSR) combined bottom sowing aquaculture (BSA) represents the best production strategy upstream of the sea cucumber supply chain. In the downstream, the best proportion of sales channels in supermarkets, boutique stores and online shops accounted for 14.79 %, 58.02 % and 27.19 % of the production, respectively. The proposed optimization scenario 4 (S4) can increase product profit by 27.88 % and reduce GWP by 56.89 %. The following improvement measures are proposed: using sea cucumber aquaculture industry standards (cleaner production and green supplier selection) to regulate the behavior of enterprises, adopting an ecological and green production strategy, eliminating high-energy consumption and high emission production practices, and promoting widespread adoption of green consumption concepts. Finally, these measures may improve the sea cucumber supply chain, achieve coordinated environmental and economic performance development in the sea cucumber industry, and provide guidance for green optimization of other aquatic product supply chains in China.

RESUMO

The tensile strength and modulus of elasticity of a jute/polylactic acid (PLA) composite were found to vary nonlinearly with the loading angle of the specimen through the tensile test. The variation in these properties was related to the fiber orientation distribution (FOD) and fiber length distribution (FLD). In order to study the effects of the FOD and FLD of short fibers on the mechanical properties and to better predict the mechanical properties of short-fiber composites, the true distribution of short fibers in the composite was accurately obtained using X-ray computed tomography (XCT), in which about 70% of the jute fibers were less than 300 µm in length and the fibers were mainly distributed along the direction of mold flow. The probability density functions of the FOD and FLD were obtained by further analyzing the XCT data. Strength and elastic modulus prediction models applicable to short-fiber-reinforced polymer (SFRP) composites were created by modifying the laminate theory and the rule of mixtures using the probability density functions of the FOD and FLD. The experimental measurements were in good agreement with the model predictions.

RESUMO

Fundamental to all living organisms and living soft matter are emergent processes in which the reorganization of individual constituents at the nanoscale drives group-level movements and shape changes at the macroscale over time. However, light-induced degradation of fluorophores, photobleaching, is a significant problem in extended bioimaging in life science. Here, we report opening a long-time investigation window by nonbleaching phase intensity nanoscope: PINE. We accomplish phase-intensity separation such that nanoprobe distributions are distinguished by an integrated phase-intensity multilayer thin film (polyvinyl alcohol/liquid crystal). We overcame a physical limit to resolve sub-10 nm cellular architectures, and achieve the first dynamic imaging of nanoscopic reorganization over 250 h using PINE. We discover nanoscopic rearrangements synchronized with the emergence of group-level movements and shape changes at the macroscale according to a set of interaction rules with importance in cellular and soft matter reorganization, self-organization, and pattern formation.

RESUMO

Cypermethrin (CYP), a pyrethroid insecticide, exerts the detrimental effect on the reproductive system, while astaxanthin (AST), a xanthophyll carotenoid, possesses the powerful antioxidant property and can protect oocyte maturation. However, the toxicity of CYP and the protective role of AST against CYP during oocyte maturation remain unclear. Here, porcine oocytes were applied to investigate the potential effects and underlying mechanisms of CYP and AST during oocyte maturation. This work demonstrated that CYP significantly decreased oocyte maturation rate and subsequent embryo development in a dose-dependent manner (P < 0.05). And, CYP obviously induced the overproduction of reactive oxygen species and the reduction of glutathione content by downregulating the expression of redox genes in oocytes (P < 0.05). Moreover, CYP significantly caused oocyte DNA damage and disturbed the function of endoplasmic reticulum by altering the transcription of DNA damage repair and endoplasmic reticulum stress related genes (P < 0.05). Whereas CYP-exposed oocytes were treated with AST, these defects caused by CYP were significantly ameliorated (P < 0.05). In conclusion, this study demonstrated that CYP exerted the toxic effect on porcine oocytes, while AST effectively alleviated CYP-induced defects. This work provides a potential strategy to prevent pesticide toxicity and protect oocyte maturation in mammalian reproduction.

RESUMO

Dynamic detection in challenging lighting environments is essential for advancing intelligent robots and autonomous vehicles. Traditional vision systems are prone to severe lighting conditions in which rapid increases or decreases in contrast or saturation obscures objects, resulting in a loss of visibility. By incorporating intelligent optimization of polarization into vision systems using the iNC (integrated nanoscopic correction), we introduce an intelligent real-time fusion algorithm to address challenging and changing lighting conditions. Through real-time iterative feedback, we rapidly select polarizations, which is difficult to achieve with traditional methods. Fusion images were also dynamically reconstructed using pixel-based weights calculated in the intelligent polarization selection process. We showed that fused images by intelligent polarization selection reduced the mean-square error by two orders of magnitude to uncover subtle features of occluded objects. Our intelligent real-time fusion algorithm also achieved two orders of magnitude increase in time performance without compromising image quality. We expect intelligent fusion imaging photonics to play increasingly vital roles in the fields of next generation intelligent robots and autonomous vehicles.

RESUMO

Twenty percent of breast cancer (BC) patients develop distant metastasis for which there is no cure. Mesenchymal stem/stromal cells (MSCs) in the tumor microenvironment were shown to stimulate metastasis, but the mechanisms are unclear. Here, we identified and quantified cancer cells engulfing stromal cells in clinical samples of BC metastasis by dual immunostaining for EZH2 and ALDH1 expression. Using flow cytometry and a microfluidic single-cell paring and retrieval platform, we show that MSC engulfment capacity is associated with BC cell metastatic potential and generates cells with mesenchymal-like, invasion, and stem cell traits. Whole-transcriptome analyses of selectively retrieved engulfing BC cells identify a gene signature of MSC engulfment consisting of WNT5A, MSR1, ELMO1, IL1RL2, ZPLD1, and SIRPB1. These results delineate a mechanism by which MSCs in the tumor microenvironment promote metastasis and provide a microfluidic platform with the potential to predict BC metastasis in clinical samples.

RESUMO

Vibration frequencies and modes for the thickness-shear vibrations of infinite partially-electroded circular AT-cut quartz plates are obtained by solving the two-dimensional (2D) scalar differential equation derived by Tiersten and Smythe. The Mathieu and modified Mathieu equations are derived from the governing equation using the coordinate transform and the collocation method is used to deal with the boundary conditions. Solutions of the resonant frequencies and trapped modes are validated by those results obtained from COMSOL software. The current study provides a theoretical way for figuring out the vibration analysis of circular quartz resonators.

RESUMO

OBJECTIVE: To identify the casual mutation of a Chinese pedigree with hypertrophic cardiomyopathy (HCM), and to analyze the genotype-phenotype relationship. METHODS: The coding exons of 26 reported disease genes were sequenced by targeted resequencing in the proband and the identified mutation were detected with bi-directional Sanger sequencing in all family members and 307 healthy controls. The genotype-phenotype correlation was analyzed in the family. RESULTS: A missense mutation (c.2191C > T, p. Pro731Ser) in the 20th exon of MYH7 gene was identified. This mutation was absent in 307 healthy controls and predicted to be pathogenic by PolyPhen-HCM. Totally 13 family members carried this mutation, including 10 patients with HCM and 3 asymptomatic mutation carriers. The proband manifested severe congestive heart failure and 8 patients expressed various clinical manifestations of heart failure, including dyspnea, palpitations, chest pain, amaurosis or syncope. Five patients were diagnosed as HCM at the age of 16 or younger. One family member suffered sudden cardiac death. CONCLUSIONS: The Pro731Ser of MYH7 gene mutation is a causal and malignant mutation linked with familiar HCM.

RESUMO

The protein p26 is a small heat shock protein that functions as a molecular chaperone to protect embryos by preventing irreversible protein damage during embryonic development. A 542 bp fragment of the p26 gene was cloned and sequenced. The fragment encoded 174 amino acid residues and the amino acid sequence contained the alpha-crystallin domain. Phylogenetic analysis showed that eight Artemia populations were divided into four major groups. Artemia sinica (YC) belonged to the East Asia bisexual group. Expression of the p26 gene at different developmental stages of A. sinica was quantified using real-time quantitative polymerase chain reaction followed by cloning and sequencing. The relationship between the quantity of p26 gene expression and embryonic development was analyzed. The results indicated that massive amounts of p26 were expressed during the development of A. sinica. At the developmental stage of 0 h, A. sinica expressed the highest level of p26. As development proceeded, expression levels of the p26 gene reduced significantly. There was a small quantity of p26 gene expression at the developmental stages of 16 h and 24 h. We concluded that p26 might be involved in protecting the embryo from physiological stress during embryonic development.
