Background: Intestinal metaplasia (IM) of gastric epithelium has traditionally been regarded as an irreversible stage in the process of the Correa cascade. Exploring the potential molecular mechanism of IM is significant for effective gastric cancer prevention. Methods: The GSE78523 dataset, obtained from the Gene Expression Omnibus (GEO) database, was analyzed using RStudio software to identify the differently expressed genes (DEGs) between IM tissues and normal gastric epithelial tissues. Subsequently, gene ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, Gene Set Enrichment Analysis (GESA), and protein-protein interaction (PPI) analysis were used to find potential genes. Additionally, the screened genes were analyzed for clinical, immunological, and genetic correlation aspects using single gene clinical correlation analysis (UALCAN), Tumor-Immune System Interactions Database (TISIDB), and validated through western blot experiments. Results: Enrichment analysis showed that the lipid metabolic pathway was significantly associated with IM tissues and the apolipoprotein B (APOB) gene was identified in the subsequent analysis. Experiment results and correlation analysis showed that the expression of APOB was higher in IM tissues than in normal tissues. This elevated expression of APOB was also found to be associated with the expression levels of hepatocyte nuclear factor 4A (HNF4A) gene. HNF4A was also found to be associated with immune cell infiltration to gastric cancer and was linked to the prognosis of gastric cancer patients. Moreover, HNF4A was also highly expressed in both IM tissues and gastric cancer cells. Conclusion: Our findings indicate that HNF4A regulates the microenvironment of lipid metabolism in IM tissues by targeting APOB. Higher expression of HNF4A tends to lead to a worse prognosis in gastric cancer patients implying it may serve as a predictive indicator for the progression from IM to gastric cancer.

Population aging and global warming have become everyday concerns of all countries. Based on the panel data of 30 provinces in China from 2003 to 2019, this paper uses the panel fixed effect model and two-stage least square method to analyze the effect of population aging on domestic energy carbon emissions of urban and rural residents. On this basis, the threshold regression model is introduced to explore the heterogeneity of the effect under different aging levels. The results show that (1) the progress of population aging at the overall level will significantly increase the level of carbon emissions from household energy consumption. At the regional level, the effect of population aging on carbon emissions from household energy consumption in rural areas is higher than in urban areas. (2) Population aging has a nonlinear effect on the carbon emissions of residential energy consumption. For urban areas, when the level of population aging crosses the threshold, its marginal impact on living carbon emissions in urban areas is further enhanced. In contrast, the opposite is true in rural areas. (3) Heterogeneity analysis results show that the impact of population aging on residential energy carbon emissions differs in different regions at the national and rural levels but does not show regional heterogeneity at the urban level.

BACKGROUND: Artificial intelligence represents an emerging area with promising potential for improving colonoscopy quality. AIMS: To develop a colon polyp detection model using STFT and evaluate its performance through a randomized sample experiment. METHODS: Colonoscopy videos from the Digestive Endoscopy Center of the First Affiliated Hospital of Anhui Medical University, recorded between January 2018 and November 2022, were selected and divided into two datasets. To verify the model's practical application in clinical settings, 1500 colonoscopy images and 1200 polyp images of various sizes were randomly selected from the test set and compared with the STFT model's and endoscopists' recognition results with different years of experience. RESULTS: In the randomized sample trial involving 1500 colonoscopy images, the STFT model demonstrated significantly higher accuracy and specificity compared to endoscopists with low years of experience (0.902 vs. 0.809, 0.898 vs. 0.826, respectively). Moreover, the model's sensitivity was 0.904, which was higher than that of endoscopists with low, medium, or high years of experience (0.80, 0.896, 0.895, respectively), with statistical significance (P < 0.05). In the randomized sample experiment of 1200 polyp images of different sizes, the accuracy of the STFT model was significantly higher than that of endoscopists with low years of experience when the polyp size was ≤ 0.5 cm and 0.6-1.0 cm (0.902 vs. 0.70, 0.953 vs. 0.865, respectively). CONCLUSIONS: The STFT-based colon polyp detection model exhibits high accuracy in detecting polyps in colonoscopy videos, with a particular efficiency in detecting small polyps (≤ 0.5 cm)(0.902 vs. 0.70, P < 0.001).

BACKGROUND: MicroRNAs (miRNAs) are associated with cancer progression. MiR-140-3p is a tumor suppressor. Nevertheless, its function in non-small cell lung cancer (NSCLC) is unclear. METHODS: MiR-140-3p expression in NSCLC clinical specimens was examined using the TCGA database and real-time PCR. NSCLC cell proliferation and apoptosis were investigated after the miRNA overexpression. Then, mineral dust-induced gene (MDIG) levels in NSCLC clinical specimens were monitored by real-time PCR and western blotting. Bioinformatics predicated the binding of miR-140-3p to MDIG, and their relationship was validated by luciferase reporter assay. The miR-140-3p/MDIG axis was further validated through rescue experiments. The involvement of STAT3 signaling in the actions of miR-140-3p/MDIG axis was investigated. RESULTS: MiR-140-3p was decreased in NSCLC tissues and negatively correlated with MDIG expression. Additionally, it was also lower in high-grade specimens than in low-grade ones. MiR-140-3p restrained cell proliferation, facilitated apoptosis, and inhibited STAT3 signaling in NSCLC. Interestingly, MDIG was a target of this miRNA. Furthermore, MDIG upregulation abolished miR-140-3p's effect on cell proliferation, apoptosis, and STAT3 pathway in NSCLC cells. CONCLUSION: MiR-140-3p restrained NSCLC development through the regulation of the STAT3 pathway by targeting MDIG. This axis may be a promising target for NSCLC treatment.

RESUMO

Autophagy is a physiological mechanism in which cells degrade themselves and quickly recover the degraded cell components. Recent studies have shown that autophagy plays an important role in the occurrence, development, treatment, and prognosis of colorectal cancer. In the early stages of colorectal cancer, autophagy can inhibit the production and development of tumors through multiple mechanisms such as maintaining DNA stability, inducing tumor death, and enhancing immune surveillance. However, as colorectal cancer progresses, autophagy may mediate tumor resistance, enhance tumor metabolism, and other pathways to promote tumor development. Therefore, intervening in autophagy at the appropriate time has broad clinical application prospects. This article summarizes the recent research progress of autophagy and colorectal cancer and is expected to provide new theoretical basis and reference for clinical treatment of colorectal cancer.

Renal fibrosis is a common pathological feature of chronic kidney disease (CKD) with various etiologies, which seriously affects the structure and function of the kidney. Pregnane X receptor (PXR) is a member of the nuclear receptor superfamily and plays a critical role in regulating the genes related to xenobiotic and endobiotic metabolism in mammals. Previous studies show that PXR is expressed in the kidney and has protective effect against acute kidney injury (AKI). In this study, we investigated the role of PXR in CKD. Adenine diet-induced CKD (AD) model was established in wild-type and PXR humanized (hPXR) mice, respectively, which were treated with pregnenolone-16α-carbonitrile (PCN, 50 mg/kg, twice a week for 4 weeks) or rifampicin (RIF, 10 mg·kg-1·d-1, for 4 weeks). We showed that both PCN and RIF, which activated mouse and human PXR, respectively, improved renal function and attenuated renal fibrosis in the two types of AD mice. In addition, PCN treatment also alleviated renal fibrosis in unilateral ureter obstruction (UUO) mice. On the contrary, PXR gene deficiency exacerbated renal dysfunction and fibrosis in both adenine- and UUO-induced CKD mice. We found that PCN treatment suppressed the expression of the profibrotic Wnt7a and ß-catenin in AD mice and in cultured mouse renal tubular epithelial cells treated with TGFß1 in vitro. We demonstrated that PXR was colocalized and interacted with p53 in the nuclei of tubular epithelial cells. Overexpression of p53 increased the expression of Wnt7a, ß-catenin and its downstream gene fibronectin. We further revealed that p53 bound to the promoter of Wnt7a gene to increase its transcription and ß-catenin activation, leading to increased expression of the downstream profibrotic genes, which was inhibited by PXR. Taken together, PXR activation alleviates renal fibrosis in mice via interacting with p53 and inhibiting the Wnt7a/ß-catenin signaling pathway.

The purpose of this study is to explore the influence of Foreign Direct Investment (FDI) on rural-urban migrants' physical health and its influencing mechanism. A total of 134,920 rural-urban migrant samples are matched based on the China Migrants Dynamic Survey in 2017 and the China Urban Statistical Yearbook in 2016. On the basis of the samples, a Binary Probit Model is used to explore the relationship between the degree of FDI and rural-urban migrants' physical health. The results show that compared with migrants who lived in cities with a lower FDI level, rural-urban migrants who lived in cities with a higher FDI level are better in physical health. The results of the mediation effect model show that the degree of FDI has a significant positive impact on employment rights and benefits the protection of rural-urban migrants, improving rural-urban migrants' physical health, which means employment rights and benefits protection plays an intermediary role in the process of FDI affecting rural-urban migrants' physical health. Therefore, when formulating public policies such as plans to improve the physical health of rural-urban migrants, not only the availability of medical services for rural-urban migrants needs to be improved, but the positive spillover effect of FDI should be taken into account. By doing so, FDI can positively affect the physical health of rural-urban migrants.

Carbon trading price (CTP) prediction accuracy is critical for both market participants and policymakers. As things stand, most previous studies have only focused on one or a few carbon trading markets, implying that the models' universality is insufficient to be validated. By employing a case study of all carbon trading markets in China, this study proposes a hybrid point and interval CTP forecasting model. First, the Pearson correlation method is used to identify the key influencing factors of CTP. The original CTP data is then decomposed into multiple series using complete ensemble empirical mode decomposition with adaptive noise. Following that, the sample entropy method is used to reconstruct the series to reduce computational time and avoid overdecomposition. Following that, a long short-term memory method optimized by the Adam algorithm is established to achieve the point forecasting of CTP. Finally, the kernel density estimation method is used to predict CTP intervals. On the one hand, the results demonstrate the proposed model's validity and superiority. The interval prediction model, on the other hand, reflects the uncertainty of market participants' behavior, which is more practical in the operation of carbon trading markets.

Background: Exploring the potential mechanism of cholangiocarcinoma (CCA) metabolic reprogramming is significant for guiding clinical treatment. However, related research and exploration are still lacking. Therefore, we aimed to identify a reliable metabolism-related gene or biomarker of CCA using bioinformatics analysis. Methods: The GSE26566, GSE45001, and GSE132305 datasets were obtained from the Gene Expression Omnibus (GEO) database. Differently expressed genes (DEGs) between CCA tissues and adjacent tissues were screened out. The key gene was identified through enrichment and functional analysis, and its immune and clinical correlation was investigated utilizing the Tumor Immune Evaluation Resource (TIMER2.0), the Tumor-Immune System Interactions Database (TISIDB), the Gene Expression Profiling Interactive Analysis (GEPIA2), and the Kaplan-Meier Plotter. Finally, immunohistochemistry (IHC) was performed to validate the results. Results: By analysis, the expression of FBJ murine osteosarcoma viral oncogene homolog B (FOSB) was significantly downregulated in CCA tissues when compared with adjacent tissues. Moreover, the expression levels of FOSB positively correlated with tumor-infiltrating immune cells in most tumors, and patients with high FOSB expression tended to have a better prognosis. The FOSB and SIRT3/HIF1A axes have similar expression trends and metabolic functions in CCA cells, and the correlation between of them was preliminarily explored by IHC experiments. Conclusions: The expression levels of FOSB are closely related to the prognosis of CCA patients, which may be a predictive indicator for prognosis and immunotherapy.

The sound of the pig is one of its important signs, which can reflect various states such as hunger, pain or emotional state, and directly indicates the growth and health status of the pig. Existing speech recognition methods usually start with spectral features. The use of spectrograms to achieve classification of different speech sounds, while working well, may not be the best approach for solving such tasks with single-dimensional feature input. Based on the above assumptions, in order to more accurately grasp the situation of pigs and take timely measures to ensure the health status of pigs, this paper proposes a pig sound classification method based on the dual role of signal spectrum and speech. Spectrograms can visualize information about the characteristics of the sound under different time periods. The audio data are introduced, and the spectrogram features of the model input as well as the audio time-domain features are complemented with each other and passed into a pre-designed parallel network structure. The network model with the best results and the classifier were selected for combination. An accuracy of 93.39% was achieved on the pig speech classification task, while the AUC also reached 0.99163, demonstrating the superiority of the method. This study contributes to the direction of computer vision and acoustics by recognizing the sound of pigs. In addition, a total of 4,000 pig sound datasets in four categories are established in this paper to provide a research basis for later research scholars.

Background: In order to maintain high standards of healthcare, it is necessary for medical departments to provide high-quality and affordable medical services to local residents. This has been widely accepted in developed countries, while the medical treatment systems in developing countries remain to be improved. This research is based on a pilot of a hierarchical medical system in Shanghai, China, to evaluate the effects on policy of medical reform in developing countries. Methods and results: By means of the difference-in-differences (DID) method, the causal relationship between medical care services' improvement and hierarchical medical systems' implementation could be identified. This project also explores the differential effects of policy intervention and confirms that the pilot showed a significant improvement in medical performance in central districts while the result remains uncertain in terms of suburban districts. Furthermore, the dynamic effect of a hierarchical medical system has also been identified with the event study method, while the policy pilot only had short-term effects on local medical resources' improvement. In order to ascertain the function mechanisms of hierarchical medical systems and explain why the policy pilot only had short-term effects, this project also conducts influencing mechanism analysis with the triple-differences method (also known as difference-in-difference-in-differences or DDD method). According to the empirical results, there is no direct evidence indicating the hierarchical medical system could bring obvious benefits from the perspectives of patients and medical institutions. Conclusions: For better implementation of hierarchical medical systems in the future, long-term supervision mechanisms should be given more attention in the enforcement process of hierarchical medical systems. At the same time, more safeguarding measures should be implemented, such as supervising the payment systems of the medical institution and conducting performance evaluation.

Alzheimer's disease (AD) is a neurodegenerative disease, characterized by memory loss and cognitive deficits accompanied by neuronal damage and cholinergic disorders. Sesamol, a lignan component in sesame oil, has been proven to have neuroprotective effects. This research aimed to investigate the preventive effects of sesamol on scopolamine (SCOP)-induced cholinergic disorders in C57BL/6 mice. The mice were pretreated with sesamol (100 mg/kg/d, p.o.) for 30 days. Behavioral tests indicated that sesamol supplement prevented SCOP-induced cognitive deficits. Sesamol enhanced the expression of neurotrophic factors and postsynaptic density (PSD) in SCOP-treated mice, reversing neuronal damage and synaptic dysfunction. Importantly, sesamol could balance the cholinergic system by suppressing the AChE activity and increasing the ChAT activity and M1 mAChR expression. Sesamol treatment also inhibited the expression of inflammatory factors and overactivation of microglia in SCOP-treated mice. Meanwhile, sesamol improved the antioxidant enzyme activity and suppressed oxidative stress in SCOP-treated mice and ameliorated the oxidized cellular status and mitochondrial dysfunction in SCOP-treated SH-SY5Y cells. In conclusion, these results indicated that sesamol attenuated SCOP-induced cognitive dysfunction via balancing the cholinergic system and reducing neuroinflammation and oxidative stress.

Methionine is an essential amino acid, involved in the promotion of growth, immunity, and regulation of energy metabolism. Over the decades, research has long focused on the beneficial effects of methionine supplementation, while data on positive effects of methionine restriction (MR) were first published in 1993. MR is a low-methionine dietary intervention that has been reported to ameliorate aging and aging-related health concomitants and diseases, such as obesity, type 2 diabetes, and cognitive disorders. In addition, MR seems to be an approach to prolong lifespan which has been validated extensively in various animal models, such as Caenorhabditis elegans, Drosophila, yeast, and murine models. MR appears to be associated with a reduction in oxidative stress via so far mainly undiscovered mechanisms, and these changes in redox status appear to be one of the underlying mechanisms for lifespan extension and beneficial health effects. In the present review, the association of methionine metabolism pathways with redox homeostasis is described. In addition, the effects of MR on lifespan, age-related implications, comorbidities, and diseases are discussed.

Spider silks exhibit excellent mechanical properties and have promising application prospects in engineering fields. Because natural spider silk fibers cannot be manufactured on a large scale, researchers have attempted to fabricate bio-inspired spider silks. However, the fabrication of bio-inspired spider silks with dynamically tunable mechanical properties and stimulation-response characteristics remains a challenge. Herein, the 4D printing of shape memory polyurethane is employed to produce dynamic bio-inspired spider silks. The bio-inspired spider silks have two types of energy-absorbing units that can be adjusted, one by means of 4D printing with predefined nodes, and the other through different stimulation methods to make the bio-inspired spider silks contract and undergo spiral deformation. The shape morphing behaviors of bio-inspired spider silks are programmed via pre-stress assemblies enabled by 4D printing. The energy-absorbing units of bio-inspired spider silks can be dynamically adjusted owing to stress release generated with the stimuli of temperature or humidity. Therefore, the mechanical properties of bio-inspired spider silks can be controlled to change dynamically. This can further help in developing applications of bio-inspired spider silks in engineering fields with dynamic changes of environment.

As the proportion of renewable energy gradually increases, it brings challenges to the stable operation of the combined heat and power (CHP) system. As an important flexible resource, energy storage (ES) has attracted more and more attention. However, the profit of energy storage can't make up for the investment and operation cost, and there is a lack of measurement system for multiple values, which seriously hinders the development of energy storage industry. Based on this, this paper makes a quantitative analysis on the system value of multiple energy storage in CHP. Firstly, the uncertain output of renewable energy is characterized by Kullback-Leibler (KL) divergence, and a two-level dispatching model is constructed based on the distributionally robust optimization method, so as to study the optimal operation strategy of the ES-CHP system. Secondly, based on the system value theory, this paper analyzes the system value of multiple energy storage, including internal value and external value, and constructs the value quantitative model, respectively. Finally, in a typical ES-CHP system, the system value of multiple energy storage is quantified. The effectiveness of the two-level model constructed in this paper can be seen from the simulation results, and the influence of different electricity prices on the system value of multiple energy storage is further analyzed.

Transcranial magneto-acoustic electrical stimulation (TMAES) is a novel method of brain nerve regulation and research, which uses induction current generated by the coupling of ultrasound and magnetic field to regulate neural electrical activity in different brain regions. As the second special envoy of nerve signal, calcium plays a key role in nerve signal transmission. In order to investigate the effect of TMAES on prefrontal cortex electrical activity, 15 mice were divided into control group, ultrasound stimulation (TUS) group and TMAES group. The TMAES group received 2.6 W/cm 2 and 0.3 T of magnetic induction intensity, the TUS group received only ultrasound stimulation, and the control group received no ultrasound and magnetic field for one week. The calcium ion concentration in the prefrontal cortex of mice was recorded in real time by optical fiber photometric detection technology. The new object recognition experiment was conducted to compare the behavioral differences and the time-frequency distribution of calcium signal in each group. The results showed that the mean value of calcium transient signal in the TMAES group was (4.84 ± 0.11)% within 10 s after the stimulation, which was higher than that in the TUS group (4.40 ± 0.10)% and the control group (4.22 ± 0.08)%, and the waveform of calcium transient signal was slower, suggesting that calcium metabolism was faster. The main energy band of the TMAES group was 0-20 Hz, that of the TUS group was 0-12 Hz and that of the control group was 0-8 Hz. The cognitive index was 0.71 in the TMAES group, 0.63 in the TUS group, and 0.58 in the control group, indicating that both ultrasonic and magneto-acoustic stimulation could improve the cognitive ability of mice, but the effect of the TMAES group was better than that of the TUS group. These results suggest that TMAES can change the calcium homeostasis of prefrontal cortex nerve clusters, regulate the discharge activity of prefrontal nerve clusters, and promote cognitive function. The results of this study provide data support and reference for further exploration of the deep neural mechanism of TMAES.

Intermittent fasting (IF) has been studied for its effects on lifespan and the prevention or delay of age-related diseases upon the regulation of metabolic pathways. Mitochondria participate in key metabolic pathways and play important roles in maintaining intracellular signaling networks that modulate various cellular functions. Mitochondrial dysfunction has been described as an early feature of brain aging and neurodegeneration. Although IF has been shown to prevent brain aging and neurodegeneration, the mechanism is still unclear. This review focuses on the mechanisms by which IF improves mitochondrial function, which plays a central role in brain aging and neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease, and Huntington's disease. The cellular and molecular mechanisms of IF in brain aging and neurodegeneration involve activation of adaptive cellular stress responses and signaling- and transcriptional pathways, thereby enhancing mitochondrial function, by promoting energy metabolism and reducing oxidant production.

Abiotic stress affects metabolic processes in plants and restricts plant growth and development. In this experiment, Caucasian clover (Trifolium ambiguum M. Bieb.) was used as a material, and the CDS of TaMYC2, which is involved in regulating the response to abiotic stress, was cloned. The CDS of TaMYC2 was 726 bp in length and encoded 241 amino acids. The protein encoded by TaMYC2 was determined to be unstable, be highly hydrophilic, and contain 23 phosphorylation sites. Subcellular localization results showed that TaMYC2 was localized in the nucleus. TaMYC2 responded to salt, alkali, cold, and drought stress and could be induced by IAA, GA3, and MeJA. By analyzing the gene expression and antioxidant enzyme activity in plants before and after stress, we found that drought and cold stress could induce the expression of TaMYC2 and increase the antioxidant enzyme activity. TaMYC2 could also induce the expression of ROS scavenging-related and stress-responsive genes and increase the activity of antioxidant enzymes, thus improving the ability of plants to resist stress. The results of this experiment provide references for subsequent in-depth exploration of both the function of TaMYC2 in and the molecular mechanism underlying the resistance of Caucasian clover.

Caucasian clover is the only perennial herb of the genus Leguminous clover with underground rhizomes. However, we know very little about its development process and mechanism. Transcriptome studies were conducted on the roots of Caucasian clover without a rhizome (NR) at the young seedling stage and the fully developed rhizome, including the root neck (R1), main root (R2), horizontal root (R3), and rhizome bud (R4), of the tissues in the mature phase. Compared with the rhizome in the mature phase, NR had 893 upregulated differentially expressed genes (DEGs), most of which were enriched in 'phenylpropanoid biosynthesis', 'phenylalanine metabolism', 'DNA replication' and 'biosynthesis of amino acids'. A higher number of transcription factors (AP2/ERF, C2H2 and FAR1) were found in NR. There were highly expressed genes for R4, such as auxin response factor SAUR, galacturonosyltransferase (GAUT), and sucrose synthase (SUS). Phenylpropanoids are very important for the entire process of rhizome development. We drew a cluster heat map of genes related to the phenylpropanoid biosynthesis pathway, in which the largest number of genes belonged to COMT, and most of them were upregulated in R4.

Collaborative governance plays an important role in air pollution prevention (APP). This paper aims to evaluate the policy effects of collaborative governance for APP in the Yangtze River Delta (YRD) region, analyze the collaborative mechanism from the perspective of institutional collective action (ICA) theory, and discuss its implications for improving the policy effects and regional collaboration. First, the causal relationship between collaborative environmental governance and air pollutant reduction is confirmed using the regression discontinuity design (RDD). However, it is also established that regional APP policies initiated by the Regional Cooperation Office of Yangtze River Delta (RCOYRD) did not have a long-term effect on reducing air pollutants. Second, initiative heterogeneity and administrative hierarchy heterogeneity of policy effects between cities are analyzed. It demonstrates that the mechanism formulated by RCOYRD has very limited impact on the reduction of transaction costs and collaborative risks of collaborative environmental governance. Third, clear division of responsibilities, balanced cost-benefit sharing, and coordinated enforcement between local governments are needed to improve the policy effects of collaborative governance for APP.
