RESUMO

OBJECTIVES: To explore the relationship between nocturnal levels of stress-related hormones and different sleep-wake states in chronic insomnia disorder (CID) patients. METHODS: Thirty-three CID patients and 34 good sleepers were enrolled and completed assessment of sleep log, Pittsburgh Sleep Quality Index and Insomnia Severity Index. During a-overnight polysomnography monitoring, the patients' vein bleeds were continually collected at different time points (pre-sleep, deep-sleep, 5-min or 30-min waking, and morning waking-up). The control subjects' bleeds were collected only at 22:00 and morning waking-up. The serum hormones were detected using enzyme-linked immunosorbent assay. RESULTS: Compared with at pre-sleep, the level of cortisol was significantly higher at morning waking-up respectively in two-group subjects (Ps < 0.001), with insignificant inter-group differences in cortisol, corticotropin releasing hormone and copeptin at the two time-points. In the patients, the nocturnal secretion curves of three hormones were similar, with the highest concentration at morning waking-up, followed by 30-min waking, 5-min waking, pre-sleep, and deep-sleep. The patients' cortisol (Z = 79.192, P < 0.001) and copeptin (Z = 12.333, P = 0.015) levels were statistically different at different time-points, with higher cortisol at morning waking-up relative to deep-sleep, pre-sleep and 5-min waking (Ps < 0.05), and at 30-min waking relative to deep-sleep and pre-sleep (Ps < 0.05), and higher copeptin at morning waking-up relative to deep-sleep (P < 0.05). CONCLUSIONS: In CID, the nocturnal wakes were instantaneously accompanied by high level, and deep sleep was accompanied by the lowest levels, of stress-related hormones, especially in cortisol, supporting the insomniac hypothesis of increased nocturnal pulse-release of cortisol.

BACKGROUND AND OBJECTIVE: Rumination, a common factor of chronic insomnia disorder (CID) caused by cognitive-emotional arousal, is associated with an increased amount of rapid eye movement (REM) sleep. However, the specific subtypes, such as phasic REM and tonic REM, that contribute to the increased REM sleep have not been reported. This study aimed to determine the association between rumination and different REM sleep subtypes in patients with CID. METHODS: This study enrolled 35 patients with CID and 27 age- and sex-matched healthy controls. The Immersion-Rumination Questionnaire evaluated participants' rumination, and the Insomnia Severity Index was used to assess insomnia severity. Finally, polysomnography was used to monitor objective sleep quality and quantification of different types of REM. RESULTS: The CID patients had higher rumination scores than the healthy controls. They had a shorter REM sleep duration, less phasic REM, a lower percentage of phasic REM time, and a higher percentage of tonic REM time. Spectral analysis revealed that the patients affected by insomnia had higher ß power during REM sleep, higher ß and σ power during phasic REM sleep, and higher ß, and γ power during tonic REM sleep. Partial correlation analysis showed that rumination in the CID patients correlated negatively with the duration of phasic REM sleep. Additionally, rumination correlated negatively with δ power in REM sleep and positively with ß power in REM sleep, tonic REM sleep, phasic REM sleep, N3and N2 sleep in the patients with CID. CONCLUSION: The CID patients had stronger rumination, reduced total and phasic REM sleep, and the stronger rumination was, the shorter phasic REM was and the higher fast (ß) wave power in REM sleep.

Purpose: To investigate changes and links of stress and high sleep reactivity (H-SR) on the macro-structure and orderliness of sleep and cortisol levels in good sleepers (GS). Patients and Methods: Sixty-two GS (18-40 years old) were recruited, with 32 in the stress group and 30 in the control group. Each group was further divided into H-SR and low SR subgroups based on the Ford Insomnia Response to Stress Test. All participants completed two nights of polysomnography in a sleep laboratory. Before conducting polysomnography on the second night, the stress group completed the Trier Social Stress Test and saliva was collected. Results: The duration of NREM sleep stages 1, 2 (N1, N2) and rapid eye movement sleep (REM) decreased, and the values of approximate entropy, sample entropy, fuzzy entropy, and multiscale entropy increased under stress and SR effects. Stress increased rapid eye movement density, and H-SR increased cortisol reactivity. Conclusion: Stress can damage the sleep and increase cortisol release in GS, especially those with H-SR. N1, N2 and REM sleep are more easily affected, while NREM sleep stage 3 sleep is relatively stable.

In this study, surface imprinting, magnetic separation, and fluorescent detection were integrated to develop a dual-recognition sensor (MF-MIPs), which was used for highly selective and sensitive detection of 4-nitrophenol (4-NP) in food samples. Silane-functionalized carbon dots (Si-CDs) participated in the imprinting process and were uniformly distributed into the MIPs layers. MF-MIPs sensor exhibited a high fluorescence response and selectivity based on the dual-recognition mechanism of imprinting recognition and fluorescence identification. The relative fluorescence intensity of MF-MIPs sensor presented a good linear relationship in the range of 0.08-10 µmol·L-1 with a low limit of detection (23.45 nmol·L1) for 4NP. MF-MIPs sensor showed high anti-interference, as well as excellent stability and reusability. The 4-NP recovery from spiked food samples ranged from 93.20 to 102.15%, and the relative standard deviation was lower than 5.0%. Therefore, MF-MIPs sensor may be a promising method for 4-NP detection in food samples.

In this study, a pectin was extracted from Akebia trifoliata var. australis fruit peel waste using water solution, and its physicochemical properties were evaluated. The pectin was rich in galacturonic acid (GalA) content (76.68%). The degree of esterification (DE) and molecular weight (Mw) were 37.60% and 29,890 Da, respectively. The pectin structure was determined using Fourier transform-infrared (FT-IR) and Hydrogen nuclear magnetic resonance (H-NMR). The pectin exhibited an amorphous nature, negative charge, and good solubility. The pectin was then used as a wall-material to coat curcumin-loaded zein nanoparticles for the first time. The obtained nanoparticles (curcumin-loaded core-shell nanoparticle, CLCSNs) exhibited a core (zein)-shell (pectin) structure and a spherical shape with an average diameter of 230 nm. The electrostatic attraction, hydrogen bonding, and intermolecular interaction were involved in the CLCSNs formation. A high encapsulation efficiency (EE, 89.65%) and loading capacity (LC, 10.35%) of the CLCSNs were obtained for the curcumin. The solubility, stability, antioxidant activity, and in vitro bioavailability of the curcumin were significantly increased after loading into the CLCSNs. Therefore, this sustainable pectin from Akebia trifoliata var. australis fruit peel waste represents a promising natural macromolecule for use in the pharmaceutical and food industries.

To achieve a rapid, sensitive, and economical method for the detection of ascorbic acid (AA) in the presence of Fe3+, a nitrogen and sulfur co-doped carbon dots (N,S-co-CDs) based fluorescence sensing system was developed. In this work, N,S-co-CDs were successfully synthesized via a one-step microwave-assisted method within 2.5 min using ammonium citrate and L-cysteine as precursors. The fluorescence of N,S-co-CDs was quenched (off ) by Fe3+ through a static-quenching mechanism. Subsequently, the fluorescence was recovered (on) after introducing AA into the quenched system, which was attributed to the reduction effect of AA for Fe3+. Therefore, a switch-on sensor (N,S-co-CDs/Fe3+ system) was developed for AA detection. Under optimal conditions, the limit of detection (LOD) of 2.31 µmol/L for AA was obtained over a linear range from 0 to 150 µmol/L. Furthermore, the proposed sensing method was successfully applied to detect AA in processed fruit juice with satisfactory results. The most important is that the sensor derived from a microwave-assisted method has simple and eco-friendly synthesis processes, is rapid, and has high detection efficiency. Therefore, such a switch-on sensor may be a promising candidate sensor for AA detection in processed fruit samples.
