RESUMO

Allium roseum is amongst the most important wild medicinal plants. It is known for its diverse biological properties, including antioxidant, antibacterial and antidiabetic activities. In this work, the polysaccharides (PARLs) were ultrasonically extracted from Allium roesum leaves then purified and analyzed by several techniques. Chemical composition and GC-MS analysis showed that the obtained polysaccharides were composed mainly of glucose (40.20 %), mannose (25.30 %), fructose (10.60 %) and galacturonic acid (15.11 %). Moreover, PARLs exhibited a potent antioxidant effect with higher capacities up to 69.61 % and 71.72 % for DPPH and ABTS free radicals, respectively. Furthermore, PARLs significantly modulated inflammatory response by reducing TNF-α, IL-6, and IL-8 pro-inflammatory mediators and promoting the anti-inflammatory IL-10 mediator in LPS stimulated THP-1 derived macrophages. The in-vivo tests proved that the extract was able to decrease carrageenan-induced rat paw swelling by around 68.15 % after 4 h of treatment. PARLs, significantly reduced the growth of U87 (glioblastoma) and IGROV-1 cancer cells with IC50 values of about 4.27 and 7.89 mg/mL respectively. This research clearly shows that Allium roseum polysaccharides can be used as natural antioxidants with anti-inflammatory and anticancer properties.

Epidemiologic studies keep up the proposition that Allium vegetables can lower the risk of cancers. Acute myeloid leukemia (AML) cells exhibit high proliferative potency and have a reduced capacity of undergoing apoptosis and maturation. The beneficial effects of Allium seem related to the organosulfur products generated upon processing of these species. For this purpose, the aim of this study was to test Allium roseum fresh (FAE), crude (CAE) and dried (DAE) aqueous extracts for activity against the human acute leukemia cell line (U937). As assessed by flow cytometry, inhibited cell proliferation was in a dose-dependent manner. Firstly, study showed that cell growth was inhibited with 20 mg/mL using FAE and CAE (60% and 73% respectively). Secondly, our experiments clearly indicate that all A. roseum extracts do not induce cell apoptosis. This was confirmed by the soft binding of Annexin V to phosphatidylserine. Finally, the high expression of macrophage's marker CD11 associated with adequate morphological changes proves clearly the differentiation aspect produced by A. roseum extract. Taken together, these data suggest that A. roseum could be a promising candidate for the alternative medicine in the field of cancer therapy.

Allium roseum is one of the medicinal plants of the Liliaceae family, widely used in the food industry and traditional medicine. It is known for its various biological properties, such as its antioxidant, antiviral, antidiabetic, and anti-inflammatory activities. The present work aims to extract the polysaccharides from Allium roseum leaves and evaluate their antioxidant activities and hepatoprotective effects in vivo. Three polysaccharides from the leaves of Allium roseum were sequentially extracted in three media: water, chelating, and basic, respectively. They were characterized by size exclusion chromatography, gas chromatography mass spectrometry, FTIR-ATR, and NMR spectroscopy (1D and 2D). The different polysaccharides principally consist of glucose, galactose, mannose, rhamnose, xylose, and galacturonic acid. The antioxidant activity and hepatoprotective effect of the extracts against Cd-caused oxidative stress in liver mouse were tested. Cd treatment, during 24 h, enhanced significantly lipid peroxidation by a high production of malondyaldehyd (MDA) and superoxide dismutase (SOD) activity. In contrast, catalase activity (CAT) was decreased after the same period of exposure to the metal. The polysaccharides pre-treatment improved the antioxidant defense system to a great degree, mainly explained by the modulating levels of oxydative stress biomarkers (MDA, SOD, and CAT). This research clearly shows that Allium roseum polysaccharides, especially those extracted in aqueous medium, can be used as natural antioxidants with hepatoprotective properties.

This study gives a new insight into the direct supplementation of Allium roseum leaves in double cream cheese. Allium roseum leaves were added to double cream cheese as a powder and a fresh paste. Based on the formulation calculation and on sensory analyses, doses of 6% paste and 0.8% powder were used to formulate the flavored cheeses. The sensory characteristics of the two double cream cheeses were assessed and compared to a plain formula. Cheese samples, stored at 5 °C, were sampled every 0, 4, 8, 12 and 15 days and analyzed for pH, fat, dry contents, yeasts and molds and total coliforms. The positive effects of Allium roseum supplementation, either as a powder or as a fresh paste, have been proven, based on sensorial, physicochemical, and microbiological qualities. The shelf life of cheese samples was also determined through an accelerated shelf life test and the Arrhenius equation. The experiments were conducted at 5, 15, and 25 °C for 15 days. The results showed a significant shelf life extension for flavored double cream cheeses (12 days) versus the plain formula (10 days). The use of Allium roseum leaves, as a natural preservative, seems to be a promising trend for the formulation of similar dairy products.

The present study deals with the valorization of an edible spontaneous plant of the Tunisian arid areas: Allium roseum. This plant is traditionally used for therapeutic and culinary uses. Thin-layer drying behavior of Allium roseum leaves was investigated at 40, 50 and 60 °C drying air temperatures and 1 and l.5 m/s air velocity, in a convective dryer. The increase in air temperature significantly affected the moisture loss and reduced the drying time while air velocity was an insignificant factor during drying of Allium roseum leaves. Five models selected from the literature were found to satisfactorily describe drying kinetics of Allium roseum leaves for all tested drying conditions. Drying data were analyzed to obtain moisture diffusivity values. During the falling rate-drying period, moisture transfer from Allium roseum leaves was described by applying the Fick's diffusion model. Moisture diffusivity varied from 2.55 × 10(-12) to 8.83 × 10(-12) m(2)/s and increased with air temperature. Activation energy during convective drying was calculated using an exponential expression based on Arrhenius equation and ranged between 46.80 and 52.68 kJ/mol. All sulfur compounds detected in the fresh leaves were detected in the dried leaves. Convective air drying preserved the sulfur compounds potential formation.
