RESUMO

The aim of this study was to produce feed based on locally sourced ingredients for tambaqui farming in Amazon. Diets were formulated with increasing levels (0, 25, 50, 75 and 100%) of defatted black soldier fly larvae meal (BSFL) as a replacement for fish meal (FM), and cassava by-products in the same proportion (tuber residues, peel and leaves). A conventional diet (CO) was used as the control. Juvenile tambaqui (24.61 ± 1.14 g) were housed in 24 tanks in a recirculation aquaculture system. Neither diet rejection nor mortality were observed. Fish fed cassava by-products showed similar feed conversion rates (FCR 1.76); however, these values were worse than those observed in fish fed the CO (FCR 1.33). No differences were observed in the whole-body composition of the fish. The fillets of fish fed cassava by-products had a yellow color due the carotenoids present in the leaves. Dietary BSFL and cassava by-products can contribute to the sustainability of Amazonian aquaculture. Further studies with a lower proportion of cassava leaves in the diet formulation are recommended so as to ensure enhanced diet digestibility and less impact on the color of the fillets.

The commercial activity of the grey mullet (known as Tainha: TAI) and Tambaqui (TAM) generates tons of waste that can be turned into valuable resources. Therefore, this work aimed to chemically characterize and quantify the fatty acids profiles of the two fishes. GCMS quantification was performed by using calibration curves built from a standard that contains 19 FAME. The analysis revealed that visceral wastes from both fishes contain 16 fatty acids (FA) consisting of saturated (SFA), monounsaturated (MUFA) and polyunsaturated (PUFA). However, their compositions were different as FA side chains in TAI and TAM contain 12 to 20 and 13 to 22 carbon atoms, respectively. Also, the SFA amount in TAI was greater than in TAM. On the other hand, TAM is richer in MUFA and PUFA compared to TAI. Both have similar chemical compositions of ω-3 and ω-6 in PUFA and ω-5, ω-7, and ω-9 in MUFA.

Acanthocephalosis in tambaqui Colossoma macropomum culture is a limiting factor for the production of the species, being the most recurrent, which makes it necessary to develop strategies to control parasitosis even in the early stages of parasite development. This study aimed to elucidate the morphological characteristics of eggs of the acanthocephalan Neoechinorhynchus buttnerae and the ovicidal effect of the essential oils of Lippia alba, Lippia sidoides, and Lippia gracilis. The parasites were collected from hosts naturally infected with N. buttnerae, and the eggs obtained from the female 'parasites' abdominal cavity were added to 12-well plates containing the essential oils at different dilutions. After the eggs (N = 300) were separated and measured (length and width) with the aid of an optical microscope coupled to a digital program, the viability of the treated eggs was monitored 24 h after treatments, L. gracilis essential oil was 100% effective at the highest concentration, whereas L. sidoides essential oil showed over 50% efficacy at 2 mg/ml, reaching 100% efficacy at all higher concentrations. Lippia. alba essential oil did not show satisfactory efficacy in preventing N. buttnerae egg hatching.

In the present study, two approaches were followed to evaluate the metabolic responses of tambaqui (Colossoma macropomum), a frugivorous species, to intraperitoneal (IP) administration of glucose (GLU) and fructose (FRU) in fed (FED) and 10-day fasted (FAST) fish. Glucose and fructose tolerance tests were performed to assess the carbohydrate utilization and complementary NMR-metabolomics analyses were done to elucidate the impacts of sugar mobilization on the metabolic profile of plasma, liver and muscle. Blood was sampled from FED groups at 0, 3, 6 and 24 h; and at 0 and 24 h from FAST groups. Significant differences were observed in the hyperglycaemic peak between sugars at 3 h (GLU - 13.7 ± 2.0 mM vs. FRU - 8.7 ± 1.1 mM; saline 6.3 ± 0.6 mM) and on the return to normoglycaemia (GLU - 8.5 ± 2.2 mM vs. FRU - 5.2 ± 0.9 mM; saline 4.9 ± 0.6 mM) 6 h after IP on the FRU fish. The NMR-metabolomics approach allowed to conclude that tambaqui seems to be more responsive to the feeding regime (FED vs. FAST) than to the injected sugar (FRU vs. GLU). From the studied tissues, plasma showed no significant variations between feeding regimes at 24 h after IP, while muscle and liver revealed some variations on the final metabolome profile between FED and FAST groups. The metabolome variations between feeding regimes are indicative of changes on the amino acid utilization. Fish from FAST group seem to utilize amino acids as energy source rather than for protein synthesis and muscle growth. Variations on glucose concentration in muscle can also indicate different utilization of the sugars depending on the feeding regime.

BACKGROUND: Tambaqui (Colossoma macropomum, Cuvier, 1818) is the most economically important native freshwater fish species in Brazil. It can reach a total length of over 1 m and a weight of over 40 kg. The species displays a clear sex dimorphism in growth performance, with females reaching larger sizes at harvest. In aquaculture, the production of monosex populations in selective breeding programmes has been therefore identified as a key priority. RESULTS: In the present study, a genetic linkage map was generated by double digest restriction-site associated DNA (ddRAD) sequencing from 248 individuals sampled from two F1 families. The map was constructed using 14,805 informative SNPs and spanned 27 linkage groups. From this, the tambaqui draft genome was improved, by ordering the scaffolds into chromosomes, and sex-linked markers were identified. A total of 235 markers on linkage group 26 showed a significant association with the phenotypic sex, supporting an XX/XY sex determination system in the species. The four most informative sex-linked markers were validated on another 206 sexed individuals, demonstrating an accuracy in predicting sex ranging from 90.0 to 96.7%. CONCLUSIONS: The genetic mapping and novel sex-linked DNA markers identified and validated offer new tools for rapid progeny sexing, thus supporting the development of monosex female production in the industry while also supporting breeding programmes of the species.

INTRODUCTION: Members of the genus Mymarothecium Kritsky, Boeger & Jégu, 1996 infect characiform fish in the Amazon River Basin. Although the Peruvian Amazon hosts rich fauna of fish, the taxonomic diversity of parasitic organisms such as Mymarothecium has been few explored. A new species of Mymarothecium in Colossoma macropomum Cuvier, from the southeast of the Peruvian Amazon, is described. METHODS: Monogeneans were isolated from gills of C. macropomum, fixed in formaldehyde in 4% solution. For morphological examinations were stained with Gomori's trichrome and others were clarified with Hoyer's medium. RESULTS: Mymarothecium is the third species of the genus recorded in C. macropomum. These new species have been characterized by a slightly sigmoid male copulatory organ (MCO) and the accessory piece with sinuous distal rod with hook-shaped process and a bifurcation in the middle part. Furthermore, the anteromedial projection of the ventral rod has a small conspicuous filament in both anchors. CONCLUSIONS: With the description of this new species, eight species of Mymarothecium have been reported in fish of the Serrasalmidae family originating in the Amazon.

Abstract The aim of this study was to evaluate the association between proteins in the seminal plasma of tambaqui Colossoma macropomum (Cuvier, 1818) with seminal quality indicators after thawing. The semen was cryopreserved with a dilution based on BTS with 8% DMSO. A 200 µL sample of semen from each animal was diluted in 800 µL BTS, centrifuged at 800 rpm, and the supernatant was cryopreserved to further analyze of the protein profile of seminal plasma through one-dimensional electrophoresis (SDS-PAGE). After 15 days of cryopreservation, a cryopreserved semen straw was thawed to analyze both qualitative and quantitative parameters. When considering all collections, the SDS-PAGE identified 15 protein bands in the seminal plasma of tambaqui. When the interaction (presence or absence) between proteins observed in the seminal plasma and the post thawed spermatic parameters was evaluated, we observed a great influence of the presence of proteins on spermatic quality. A greater (P<0.05) fertilization rate was observed with the presence of proteins 12, 34, 44, 85, and 90 kDa. Proteins in seminal plasma of tambaqui influenced the spermatic quality after thawing, and thus, they can be utilized as an indicator of sperm quality, especially the proteins with a molecular weight ≤ 50 kDa.


Resumo O objetivo desse estudo foi de avaliar a associação entre a presença de proteínas no plasma seminal do tambaqui Colossoma macropomum (Cuvier, 1818) com indicadores de qualidade seminal pós-descongelamento. O semen foi criopreservado com diluidor a base de BTS com 8% DMSO. Uma amostra de 200 µL de semen de cada animal foi diluída em 800 µL de BTS, e centrifugada em 800 rpm, e somente o sobrenadante foi criopreservado para posterior análise do perfil proteico do plasma seminal, através da eletroforese unidimensional (SDS-PAGE). Decorridos 15 dias da criopreservação, uma palheta com semen criopreservado foi descongelado para análise dos parâmetros quali-quantitativos. Considerando todas as coletas, o SDS-PAGE identificou 15 bandas proteicas no plasma seminal do tambaqui. Quando se avaliou a interação (presença ou ausência) das proteínas encontradas no plasma seminal, com os parâmetros espermáticos pós-descongelamento, observou-se grande influência da presença das proteínas na qualidade espermática. Observou-se maior taxa de fertilização (P<0,05) com a presença das proteínas 12, 34, 44, 85 e 90 kDa. As proteínas do plasma seminal de tambaqui influenciaram na qualidade espermática após descongelamento, podendo ser utilizadas como indicadores para a qualidade espermática após descongelamento, principalmente as proteínas com peso molecular ≤50 kDa.

Fish use spatial cognition based on allocentric cues to navigate, but little is known about how environmental enrichment (EE) affects learning and memory in correlation with hematological changes or gene expression in the fish brain. Here we investigated these questions in Colossoma macropomum (Teleostei). Fish were housed for 192 days in either EE or in an impoverished environment (IE) aquarium. EE contained toys, natural plants, and a 12-h/day water stream for voluntary exercise, whereas IE had no toys, plants, or water stream. A third plus maze aquarium was used for spatial and object recognition tests. Compared with IE, the EE fish showed greater learning rates, body length, and body weight. After behavioral tests, whole brain tissue was taken, stored in RNA-later, and then homogenized for DNA sequencing after conversion of isolated RNA. To compare read mapping and gene expression profiles across libraries for neurotranscriptome differential expression, we mapped back RNA-seq reads to the C. macropomum de novo assembled transcriptome. The results showed significant differential behavior, cell counts and gene expression in EE and IE individuals. As compared with IE, we found a greater number of cells in the telencephalon of individuals maintained in EE but no significant difference in the tectum opticum, suggesting differential plasticity in these areas. A total of 107,669 transcripts were found that ultimately yielded 64 differentially expressed transcripts between IE and EE brains. Another group of adult fish growing in aquaculture conditions were either subjected to exercise using running water flow or maintained sedentary. Flow cytometry analysis of peripheral blood showed a significantly higher density of lymphocytes, and platelets but no significant differences in erythrocytes and granulocytes. Thus, under the influence of contrasting environments, our findings showed differential changes at the behavioral, cellular, and molecular levels. We propose that the differential expression of selected transcripts, number of telencephalic cell counts, learning and memory performance, and selective hematological cell changes may be part of Teleostei adaptive physiological responses triggered by EE visuospatial and somatomotor stimulation. Our findings suggest abundant differential gene expression changes depending on environment and provide a basis for exploring gene regulation mechanisms under EE in C. macropomum.

The present study was developed to characterize, at the species level, 34 strains of Aeromonas spp., previously isolated from stressed tambaqui fish (Colossoma macropomum), to elucidate virulence factors, as well as their antibiotic resistance profile. Amplification of the gyrB gene identified the strains as A. hydrophila, A. dhakensis, A. caviae, A. veronii and A. jandaei. Bacterial virulence was evaluated by enzymatic assays for phenotypical production of hemolysins, proteases and lipases followed by the search for genes codifying the enzymes ß-hemolysin, serine protease and lipase. Phenotypical production of virulence factors was diversified and proteolytic activity demonstrated to be a common expression among the strains. On the other hand, the lip gene encoding extracellular lipase was the most expressed. Furthermore, A. hydrophila was the most prevalent species isolated from tambaqui in our work.

Long-chain (C20-24) polyunsaturated fatty acids (LC-PUFA) are physiologically important nutrients for vertebrates including fish. Previous studies have addressed the metabolism of LC-PUFA in the Amazonian teleost tambaqui (Colossoma macropomum), an emerging species in Brazilian aquaculture, showing that all the desaturase and elongase activities required to convert C18 polyunsaturated fatty acids (PUFA) into LC-PUFA are present in tambaqui. Yet, elongation of very long-chain fatty acid 4 (Elovl4) proteins, which participate in the biosynthesis of very long-chain (>C24) saturated fatty acids (VLC-SFA) and very long-chain polyunsaturated fatty acids (VLC-PUFA), had not been characterized in this species. Here, we investigate the repertoire and function of two Elovl4 in tambaqui. Furthermore, we present the first draft genome assembly from tambaqui, and demonstrated the usefulness of this resource in nutritional physiology studies by isolating one of the tambaqui elovl4 genes. Our results showed that, similarly to other teleost species, two elovl4 gene paralogs termed as elovl4a and elovl4b, are present in tambaqui. Tambaqui elovl4a and elovl4b have open reading frames (ORF) of 948 and 912 base pairs, encoding putative proteins of 315 and 303 amino acids, respectively. Functional characterization in yeast showed that both Elovl4 enzymes have activity toward all the PUFA substrates assayed (18:3n-3, 18:2n-6, 18:4n-3, 18:3n-6, 20:5n-3, 20:4n-6, 22:5n-3, 22:4n-6 and 22:6n-3), producing elongated products of up to C36. Moreover, both Elovl4 were able to elongate 22:5n-3 to 24:5n-3, a key elongation step required for the synthesis of docosahexaenoic acid via the Sprecher pathway.

Although the tambaqui (Colossoma macropomum) is the most cultivated native fish species in Brazil, estimated breeding values for growth traits are rarely used for selection of superior individuals in commercial fingerling production. This study aimed to estimate the (co)variance components of growth traits. Body weight, length and width of 2500 tambaqui were determined at tagging and at 6 and 12 months after tagging in a commercial breeding programme in Brazil. Heritability estimates were low for traits measured at tagging (0.10 to 0.19) and moderate to high for traits measured at 6 and 12 months (0.23 to 0.81). Common full-sib effects were high at tagging (>73%), low at 6 months and negligible at 12 months. Positive genetic correlations were found among growth traits at 12 months (0.84 to 0.99) and between growth traits at 6 and 12 months (0.80 to 0.92). These results show that animal selection can be performed at 6 months after tagging. Expected genetic gains for growth traits ranged from 8% to 31%. A simulation of the sex ratio was performed, as individuals did not reach sexual maturity during the experimental period. Because of the sexual dimorphism, more accurate heritability estimates were obtained when considering the female proportion to be 90% in the high-weight group. The findings indicate that it is possible to obtain considerable genetic gains in growth by selecting for growth traits. The development of a tool to determine the sex of animals at early stages can improve the response to selection in tambaqui.

Roundup® (RD) is a glyphosate-based herbicide used to control weeds in agriculture, and fishponds. In the Amazon, hypoxia is a natural phenomenon in flooded areas. Beyond the challenge of hypoxia, fish need to cope with the use of pesticides as RD that increases in the aquatic environment through the leaching of agricultural areas, and in aquaculture fish tanks. Thus, there is a need to better understand the combined effects of hypoxia and RD contamination for aquatic biota. The aim of this study was to investigate the effects of Roundup® (RD) and subsequent acute hypoxia in the gene expression, genotoxicity, histological and physiological responses of Colossoma macropomum. Fish were individually exposed to four different treatments during 96 h: normoxia (N), hypoxia (H), RD plus normoxia (NRD), and RD plus hypoxia (HRD) (RD concentration represents 75% of LC50 - nominal concentration 15 mg L-1 to C. macropomum). HRD fishes presented down-regulation of hif-1α gene and ras oncogene, while NRD fish presented overexpression of ras; no difference occurred in hif-1α gene expression in both normoxia treatments. The glutathione-S-transferase and catalase activities increased in HRD fish liver compared to NRD. Otherwise, there was no difference in lipoperoxidation (LPO) between all treatments. Genetic Damage Index, measured throughout comet assay in erythrocytes of all treatments, presented similar values, excepted by fish exposed to NRD. As regard as hypoxic exposure, hypoxic fish presented significantly lower values, compared to HRD fishes. An increase in liver histological injuries occurred in H and HRD fish groups. In conclusion, we may affirm that C. macropomum is sensitive concerning RD contamination and that this sensitivity increases when combined with hypoxia.

Este trabalho teve como objetivo avaliar a fauna parasitária de tambaquis na região do Baixo São Francisco-AL/SE-Brasil e correlacionar os índices de prevalência e intensidade média com fatores bióticos e abióticos. Foram coletados 252 espécimes para análise parasitológica de 10 pisciculturas. Os parasitos foram contabilizados, identificados, e determinaram-se os índices de prevalência e intensidade média, que foram correlacionados com fatores bióticos e abióticos. Dos peixes coletados, 65,5% estavam parasitados por pelo menos um táxon. Foram encontrados 10 táxons: Monogeneas, Ichthyophthirius multifiliis, tricodinídeos, Piscinoodinium pillulare, Ichthyobodo sp., Dolops carvalhoi, Lernaea cyprinacea, Procamallanus (Spirocamallanus) inopinatus, Henneguya sp. e Myxobolus sp. As maiores prevalências foram encontradas para Monogeneas (49,2%) e Myxobolus sp. (31,5%). Correlações negativas entre prevalência e fatores bióticos (peso e comprimento) foram observadas para Monogeneas (r2= -0,49; r2= -0,43), Myxobolus sp. (r²= -0,46; r²= -0,39) e Henneguya sp. (r²= -0,41; r²= -0,39). O fator abiótico temperatura apresentou correlação negativa com as prevalências de Lernaea cyprinacea (r= -0,39) e tricodinídeos (r= -0,33), enquanto a condutividade elétrica apresentou correlação positiva (r= 0,40) com a prevalência de tricodinídeos. Conclui-se que a fauna parasitária dos tambaquis cultivados na região do Baixo São Francisco é diversificada e com a carga parasitária dependente da qualidade de água e do estágio de desenvolvimento dos peixes.(AU)


This study investigated the parasitic fauna of tambaquis reared in lower Sao Francisco region-Al/SE-Brazil correlating parasitic indices to abiotic and biotic factors. A total of 252 specimens of tambaqui were collected in ten fish farms for parasitological analysis. The parasites were counted, identified and the parasitological indices were determined and correlated to biotic and abiotic factors. Of all collected fish, 65,5 % were parasitized by at least one taxon. Ten taxa were found: Monogeneans, Ichthyophthirius multifiliis, trichodinids Piscinoodinium pillulare, Ichthyobodo sp, Dolops carvalhoi, Lernaea cyprinacea, Procamallanus (Spirocamallanus) inopinatus, Henneguya sp. and Myxobolus sp. The higher prevalences were found to monogeneans (49.2%) and Myxobolus sp. (31.5%). Negative correlation of prevalence and biotic factor (weight and length) were observed to monogeneans (r 2 = -0.49, r 2 = -0.43), Myxobolus sp (r²= -0.46; r²= -0.39) and Henneguya sp (r²= -0.41; r²= -0.39). Abiotic factor of temperature presented a negative correlation to prevalence of Lernaea cyprinacea and trichodinids (r= -0.39 e r= -0.33, respectively) and the electric conductivity presented positive correlation to trichodinids (r= 0.40). It was concluded that parasitic fauna of tambaquis cultured in Lower São Francisco region is diversified and the parasitic load dependent on water parameters and fish growth.(AU)

In vertebrates, the essential fatty acids (FA) that satisfy the dietary requirements for a given species depend upon its desaturation and elongation capabilities to convert the C18 polyunsaturated fatty acids (PUFA), namely linoleic acid (LA, 18:2n-6) and α-linolenic acid (ALA, 18:3n-3), into the biologically active long-chain (C20-24) polyunsaturated fatty acids (LC-PUFA), including arachidonic acid (ARA, 20:4n-6), eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3). Recent studies have established that tambaqui (Colossoma macropomum), an important aquaculture-produced species in Brazil, is a herbivorous fish that can fulfil its essential FA requirements with dietary provision C18 PUFA LA and ALA, although the molecular mechanisms underpinning such ability remained unclear. The present study aimed at cloning and functionally characterizing genes encoding key desaturase and elongase enzymes, namely fads2, elovl5 and elovl2, involved in the LC-PUFA biosynthetic pathways in tambaqui. First, a fads2-like desaturase was isolated from tambaqui. When expressed in yeast, the tambaqui Fads2 showed Δ6, Δ5 and Δ8 desaturase capacities within the same enzyme, enabling all desaturation reactions required for ARA, EPA and DHA biosynthesis. Moreover, tambaqui possesses two elongases that are bona fide orthologs of elovl5 and elovl2. Their functional characterization confirmed that they can operate towards a variety of PUFA substrates with chain lengths ranging from 18 to 22 carbons. Overall our results provide compelling evidence that demonstrates that all the desaturase and elongase activities required to convert LA and ALA into ARA, EPA and DHA are present in tambaqui within the three genes studied herein, i.e. fads2, elovl5 and elovl2.

BACKGROUND: In recent years, tracing of alimentary produce of animal origin has become increasingly important, for economic, food safety and ecological reasons. The tambaqui, Colossoma macropomum, is the native fish most farmed in Brazil. The reliable identification of the origin of tambaquis (wild or farmed) offered for sale to the general public has become necessary to satisfy regulatory norms and uphold consumer confidence. Molecular methods based on the analysis of DNA sequences have often been used to evaluate the potential for tracing farmed fish, given their reliability and precision. RESULTS: Full likelihood and Bayesian approaches proved to be the most efficient for the identification, respectively, of individuals and populations for most of the fish sampled from seven hatcheries and one wild stock. The exclusion method and genetic distances were the least effective approaches for the identification of individuals and populations. The Bayesian method identified correctly more than 99% of the fry from most stocks, except those of the Santarém hatchery and River Amazon wild stock, which presented the best results for individual identification. CONCLUSIONS: The identification of populations was effective for most hatcheries, although the identification of individuals from most stocks was hampered by the reduced genetic variability. © 2018 Society of Chemical Industry.

Abstract The effects of extreme droughts on freshwater fish remain unknown worldwide. In this paper, we estimated the condition factor, a measure of relative fitness based on the relationship of body weight to length, in four fish species representing two trophic levels (omnivores and piscivores) from Amazonian floodplain lakes for three consecutive years: 2004, 2005 (an anomalous drought year), and 2006. The two omnivores, Colossoma macropomum and Mylossoma duriventre, exhibited trends consistent with their life cycles in 2004 and 2006: high values during the hydrologic seasons of high water, receding water, and low water, with a drop following reproduction following the onset of rising water. However during the drought year of 2005 the condition factor was much lower than normal during receding and low water seasons, probably as a result of an abnormal reduction in resource availability in a reduced habitat. The two piscivorous piranhas, Serrasalmus spilopleura and S. elongatus, maintained relatively stable values of condition factor over the hydrologic cycles of all three years, with no apparent effect of the drought, probably because the reduction in habitat is counterbalanced by the resulting increase in relative prey density. We suggest that if predictions of increasing drought in the Amazon are correct, predatory species may benefit, at least in the short run, while omnivores may be negatively affected.


Resumo Os efeitos de secas extremas sobre peixes de água doce ainda é globalmente desconhecido. Neste artigo, nós estimamos o fator de condição, uma medida relativa de bem estar baseada na relação entre e peso do corpo e o comprimento, em quatro espécies de peixes de lagos da Amazônia, pertencentes a dois níveis tróficos (onívoros e carnívoros), capturadas durante três anos consecutivos: 2004, 2005 (quando ocorreu uma seca anômala) e 2006. As duas espécies onívoras, Colossoma macropomum e Mylossoma duriventre exibiram tendências consistentes com seu ciclo de vida nos anos de 2004 e 2006: valores mais altos na cheia, vazante e seca, seguida por uma queda na enchente que coincide com a estação reprodutiva. No entanto, o fator de condição foi muito menor durante a seca e a enchente em 2005, provavelmente devido a redução anormal na disponibilidade de recursos em um habitat reduzido. As duas espécies piscívoras, as piranhas Serrasalmus spilopleura e S. elongatus, mantiveram relativa estabilidade nas estimativas do fator d condição, ao longo do ciclo hidrológico dos três anos, sem efeito aparente da seca extrema, provavelmente porque a redução do habitat foi contrabalanceada pelo aumento na densidade de presas. Caso as predições de aumento de frequência de secas extremas venha a se confirmar, acreditamos que espécies predadoras poderão ser beneficiadas, ao menos no curto prazo, enquanto as onívoras serão negativamente afetadas.

Abstract The physiological responses of juvenile tambaqui (Colossoma macropomum) fed commercial feed supplemented with different concentrations of camu camu (Myrciaria dubia) were evaluated. The design was completely randomized, with treatments arranged in a factorial design with three proportions of camu camu (15%, 30% and 45%) and a control treatment (100% commercial diet), with four replicates per treatment. A total of 96 tambaqui specimens were used, with a mean initial weight of 11.69 ± 2.68 g and a mean length of 7.06 ± 0.44 cm. After 30 days, hematological parameters, metabolic variables, growth and fish swimming performance were evaluated. The different proportions of camu camu in the diet did not cause significant changes to the tambaqui's hematological parameters during the feeding period, except for hemoglobin (Hb) concentration and mean corpuscular hemoglobin concentration (MCHC) after the 30th day, and hematocrit (Ht) after the swimming stress test, which increased significantly (p < 0.05). The significant increases in metabolic variables, such as cortisol, glucose, proteins and triglycerides, and in hematologic variables after the Ucrit test reflect, respectively, biochemical adaptations for maintenance of the energy mobilization process and a regulatory necessity in tissue oxygen demand during intense exercise. Fish fed 15% and 30% camu camu gained the most weight and achieved the best swimming performance, respectively. The results for camu camu concentrations above 30% suggest a saturation of its intrinsic properties in the diet at this level and a loss of nutrients from the commercial feed replaced by the fruit, reducing productive performance and nutritional assimilation.


Resumo As respostas fisiológicas de juvenis de tambaqui (Colossoma macropomum) alimentados com ração comercial suplementada com diferentes concentrações de camu camu (Myrciaria dubia) foram avaliados. As amostras analisadas foram inteiramente casualizadas, com os tratamentos arranjados em esquema fatorial com três proporções de camu camu (15%, 30% e 45%) e um tratamento controle (ração comercial 100%), com quatro repetições por tratamento. Um total de 96 amostras de tambaqui foram utilizadas, com um peso médio inicial de 11,69 ± 2,68 g e um comprimento médio de 7,06 ± 0,44 cm. Após 30 dias, foram avaliados os parâmetros hematológicos, variáveis ​​metabólicas, crescimento e desempenho natatório de peixes. As diferentes proporções de camu camu na dieta não causou alterações significativas nos parâmetros hematológicos dos tambaquis durante o período de alimentação, com exceção de hemoglobina (Hb) e concentração de hemoglobina corpuscular média (CHCM), após o 30º dia, e hematócrito (Ht), após o teste de natação de estresse, que aumentou de forma significativa (p <0,05). Os aumentos significativos nas variáveis ​​metabólicas, como o cortisol, glicose, proteínas e triglicerídeos, e nas variáveis ​​hematológicas após o teste Ucrit reflete, respectivamente, adaptações bioquímicas para a manutenção do processo de mobilização de energia e uma necessidade de regulamentação na demanda de oxigênio nos tecidos durante o exercício intenso. Os peixes alimentados com 15% e 30% de camu camu obtiveram mais peso e melhor desempenho natatório, respectivamente. Os resultados para as concentrações camu camu superiores a 30% indicam uma saturação das suas propriedades intrínsecas na dieta, a este nível e uma perda de nutrientes a partir da ração comercial substituído pelo fruto, reduzindo o desempenho produtivo e assimilação nutricional.

The family Neoechinorhynchidae includes seven species of Neoechinorhynchus from freshwater fishes of Brazil. Although several Neoechinorhynchus species are cited infecting different fish species in Brazil, there is a lack of information concerning to their life cycle and the identification of the intermediate hosts. Thus, the aim of the present study was to describe the development of Neoechinorhynchus buttnerae in its intermediate host collected in a fish farm located in Rio Preto da Eva, Amazonas, Brazil. To verify the presence of N. buttnerae in the fish pond, twenty Colossoma macropomum were captured and analyzed, being corroborated the presence of this parasite species. Samples of plankton were also collected, finding the ostracod Cypridopsis vidua as the intermediate host. For the description of the larvae development, a laboratory experimental procedure was conducted by feeding the collected ostracods with the eggs of the adult specimens taken from the sampled fish. To observe the stages of development an artificial hatch was performed. Every stage of development was photographed, measured, drawn and described. The time of development of the immature stages of N. buttnerae was 29 days, reporting the stages: acanthor, acanthella (with eight developmental changes) and cystacanth. As high infections by N. buttnerae causes morphological damages to the intestine and may compromise the quality of C. macropomum and in consequence the production of fish farmers in the Brazilian Amazon region, the knowledge of its intermediate host and the understanding of its life cycle represents a useful information to prevent and combat infections by this parasite.

Avaliou-se o efeito dos níveis de metionina+cistina digestível para juvenis de tambaqui na fase inicial de desenvolvimento (3,03±0,06g) durante 45 dias. Foram utilizados 160 juvenis de tambaqui distribuídos em DIC, com cinco tratamentos (níveis de metionina+cistina digestível = 0,66; 0,73; 0,80; 0,87; 0,94%), quatro repetições e oito animais por unidade experimental. As dietas utilizadas foram isoproteicas (25%) e isoenergéticas (3200kcal.kg-1). Os resultados obtidos foram submetidos aos testes de homogeneidade, normalidade, à análise de variância e à análise de regressão. Os parâmetros físicos e químicos da água do sistema de recirculação ficaram dentro da faixa de tolerância para a espécie. Os níveis de metionina+cistina digestíveis avaliados não influenciaram (P>0,05) a sobrevivência, o índice hepatossomático, o índice de gordura visceral e o consumo de ração. Entretanto, ganho de peso e taxa de eficiência proteica apresentaram aumento linear (P<0,05) com aumento da metionina + cistina na ração. Verificou-se que os níveis de metionina + cistina para o tambaqui não foram completamente atendidos, sendo recomendada a realização de novos estudos.(AU)


This study aimed to determine digestible methionine+cystine requirement of tambaqui juveniles in the early stage of development (3.03 ± 0.06 g) lasting 45 days. 160 tambaqui juveniles were distributed into DIC, with five treatments (methionine + cystine = 0.66; 0.73; 0.80; 0.87; 0.94%), four replications and eight animals per experimental unit. The basal diets were isonitrogenous (25% crude protein) and isocaloric (3200kcal.kg-1). The results were submitted to the homogeneity test, normality test, analysis of variance, and regression analysis. The physical and chemical parameters of water from the water recirculation system were within those recommended for the species. Digestible methionine+cystine levels did not influence (P>0.05) the survival, hepatosomatic index, visceral fat index and feed intake. However, weight gain and protein efficiency ratio exhibit a linear increase (P<0.05) with increase in methionine and cystine in the feed. Our results showed that the methionine + cystine levels for tambaqui were not fully met, and new studies are recommended.(AU)

The study describes the morphological changes associated with parasitism by the intestinal acanthocephalan Neoechinorhynchus buttnerae in tambaqui juveniles Colossoma macropomum farmed in an excavated nursery, in Manaus (Amazon) in September 2013. After fish biometrics, analysis of macroscopic changes in morphology and counting of parasites, bowel fragments were fixed and submitted to histological and histochemical processing. All fish analyzed had acanthocephalans in the intestine; intestinal loops were milky white in color, with the presence of nodules with heavy parasitism. The changes in tissues that form the intestine varied according to the arrangement of the parasites: either free in the intestinal lumen or fixed by the proboscis on the organ wall. In the first case, the changes found were flaking, abrasion, compression, hypertrophy of goblet cells and disappearance of the villi on the mucosa, leukocytic cell infiltration in the submucosa, and muscle layer thickening. In the second case, in addition to these, other changes were observed as metaplasia in muscle tissue with its replacement by a loose connective tissue with severe leukocytic infiltration, edema in blood vessels, and necrotic foci. The histochemical analysis revealed that positive Alcian Blue mucosal cells (pH 2.5) were more expressive in parasitized intestines than in intestines not parasitized by N. buttnerae.
