RESUMO

PURPOSE: Cosmocercids are common nematodes that parasitize the digestive tract and lungs of amphibians and reptiles around the world. They are commonly found in leptodactylid and bufonid anurans in South America, primarily in Brazil and Argentina. This paper describes a new species of genus Cosmocerca based on specimens collected in a microhylid from the Dry Chaco ecoregion. METHODS: A total of 18 specimens of Elachistocleis haroi were collected in October 2011 in Chaco province and in December 2021 and March 2022 in Formosa province, Argentina, both areas of the Dry Chaco ecoregion. The morphology of the nematodes was studied in detail using light microscopy and scanning electron microscopy. RESULTS: One hundred and fifty-one nematodes were collected from the large intestines of E. haroi. Based on the morphology of the caudal papillae (rosette papillae and plectanes), the presence of two spicules and the absence of caudal alae in males, and the presence of two prodelphic ovaries in females, these specimens were allocated to the genus Cosmocerca. However, they presented unique characteristics that differentiate them from all known species. Cosmocerca wichiorum sp. nov. is similar to C. archeyi, C. australis, C. sardiniae, C. zugi, and C. leytensis by having four pairs of plectanes, but can be easily distinguished from all of them by the type, number, and arrangement of the precloacal (non-plectanes), adcloacal and postcloacal papillae, by the length of the spicules and by its different zoogeographical distribution. CONCLUSION: This is the 14th species of the genus Cosmocerca described in South America and the first one for the microhylid E. haroi from the Dry Chaco ecoregion.

Morphological examination was made of the larval forms of Grassenema procaviae (Cosmocercoidea: Atractidae), an autoinfective and viviparous nematode parasite in the stomach of Cape hyrax (Procavia capensis). Three different larval stages (second-, third- and fourth-stages), and the adult stage were found among the worms collected at necropsy of 3 hosts, which were reared in a zoo in Japan. Molting phases between the larval stages and the final molt to the adult stage were also observed. It was considered that the gravid female delivers the second-stage larva, which develops to the adult stage through 3 molts. The cephalic structure was identical throughout the second to adult stages; all with transparent filaments extending from the mouth. Because starch grains were frequently found attached to the filaments and the worm intestinal lumen also contained starch grains ingested, the filaments were surmised to act as nutrient catchers.

Cosmocercoid nematodes are common parasites in the digestive tract of amphibians. However, our knowledge of the species diversity, genetic data and molecular phylogeny of the superfamily Cosmocercoidea are far from being well understood. In the present study, large numbers of cosmocercoid nematodes were collected from the fine-spined frog Sylvirana spinulosa (Smith) (Anura: Ranidae) and the white-spotted thigh tree-frog Polypedates megacephalus (Hallowell) (Anura: Rhacophoridae) in Guangxi Province, China. Integrated morphological and genetic evidence reveals these nematode specimens to be a new species of the genus Aplectana, A. dayaoshanensis n. sp. (Cosmocercoidea: Cosmocercidae). The molecular characterization of small ribosomal DNA (18S), internal transcribed spacer (ITS) and large ribosomal DNA (28S) of A. dayaoshanensis n. sp., together with the 28S of A. chamaeleonis (Baylis, 1929) (collected from Hyperolius kivuensis Ahl in Rwanda), were reported for the first time. Moreover, phylogenetic analyses using maximum likelihood (ML) inference based on 18S + 28S and ITS sequence data, respectively, both supported the family Cosmocercidae to be a monophyletic group and the family Kathlaniidae to be a paraphyletic group. Our phylogenetic results rejected the monophyly of the genus Aplectana. The present results contribute to the knowledge of the species diversity and genetic data of cosmocercoid nematodes, and preliminarily revealed the phylogenetic relationships of the major families and some genera in the Cosmocercoidea.

The superfamily Cosmocercoidea comprises three families: Cosmocericidae, Kathlaniidae and Atractidae. Information on the nucleotide sequences of the Cosmocercoidea is quite limited, and the molecular classification of the whole superfamily has been slow to progress. The genus Grassenema of the family Atractidae is a parasitic nematode group that occurs in the digestive tract of hyraxes and includes three species: Grassenema procaviae, G. dendrohyraci, and G. hyracis. The type species of the genus, G. procaviae, was isolated from the digestive tract of Cape hyraxes (Procavia capensis) and has the potential to cause gastric ulcers. Although G. procaviae is a common parasite of Cape hyraxes, no genetic information for the parasite is currently available. In this study, we obtained the first genomic sequences of G. procaviae and performed detailed morphological observations. Furthermore, molecular phylogenetic analysis was performed, and the taxonomic position of the parasite was evaluated using 18S and 28S rDNA sequences. Those data will be useful for molecular identification of G. procaviae and future phylogenetic analysis within the Atractidae.

Probstmayria gombensis File, 1976 (Nematoda: Cosmocercoidea: Atractidae) individuals discharged in the feces of eastern chimpanzees, Pan troglodytes schweinfurthii, in Bulindi, Uganda, were examined morphologically. Adults and fourth-stage larvae, all females, found in the feces, and the third-stage larvae excised from the uterus of the gravid females were described. By close observation of the molting worms, it was considered that the uterine third-stage larvae attain molting phase, and then are laid to become fourth-stage larvae. Nutrients required for larval development in the uterus seem to be supplied by the mother after the eggshell is formed. After some growth in the host intestine, the fourth-stage larvae undergo the final molt to the adult stage. The genital primordium was very small in the early fourth-stage larvae but rapidly developed with embryonation in the pre-molt and molting phases. Such precocity suggests parthenogenetic reproduction without insemination by males. This style may enhance rapid autoinfection in the host intestine under the condition of male worm scarcity. Several ellipsoidal pseudocoelomocytes with granules of unknown function were found ventral to the intestine of the adults, fourth-stage larvae, and third-stage larvae.

The marine toad Rhinella marina (Linnaeus) (Anura, Bufonidae) is a notorious, exotic amphibian species in Australia. However, our present knowledge of the composition of the nematode fauna of R. marina is still not complete. In the present study, a new cosmocercid nematode, Cosmocerca multipapillata sp. nov., was described using both light and scanning electron microscopy, based on specimens collected from R. marina in Australia. Cosmocerca multipapillata sp. nov. can be easily distinguished from its congeners by the body size, the presence of lateral alae and well sclerotized gubernaculum, the number and arrangement of plectanes and rosettes and the length of spicules, oesophagus and tail.

Species of MegalobatrachonemaYamaguti, 1941 (Ascaridida: Cosmocercoidea) are important nematode parasites in amphibians and reptiles. However, the phylogenetic relationship of its included two subgenera Megalobatrachonema and Chabaudgolvania remains unclear. In the present study, a new species of Megalobatrachonema, M. (Chabaudgolvania) wangi sp. nov., was described based on the specimens collected from the lesser spiny frog Quasipaa exilispinosa (Liu & Hu) (Amphibia: Anura) in China. The ribosomal [large ribosomal DNA (28S) and internal transcribed spacer (ITS1-5.8S-ITS2)] and mitochondrial [12S small subunit ribosomal DNA and cytochrome c oxidase subunit 1 (cox1)] target regions of the new species and M. (Chabaudgolvania) terdentatum, together with the 12S region of M. (Megalobatrachonema) hainanensis, were amplified and sequenced for molecular identification and phylogeny. Moreover, in order to clarify the systematic position of the new species and the phylogenetic relationship of the two subgenera Megalobatrachonema and Chabaudgolvania, phylogenetic analyses based on 28S + ITS1-5.8S-ITS2 + 12S sequence data were performed using maximum likelihood (ML) inference and Bayesian inference (BI). The molecular phylogenetic results conflicted with the current classification and challenged the validity of the subgenus Chabaudgolvania, that should be a synonym of the subgenus Megalobatrachonema. The presence or absence of valves in the oesophageal bulb as a key criterion for delimitation of the two subgenera Megalobatrachonema and Chabaudgolvania seems to be unreliable.

The fish fauna in the State of Acre represents 10·7% of all fish species recorded from Brazil, but, despite this, there are few fish parasite studies in this area. The recent expansion of fish farming in Acre prompted a need for helminthological studies of the most commonly consumed fish species in the area, Pimelodus blochii (Pimelodidae). The aim of this study was to analyse the helminth fauna of P. blochii from the Acre and Xapuri Rivers in Northwestern Brazil. Numerous nematodes were collected from the intestine and two species of the family Atractidae were identified: Rondonia rondoni Travassos, 1920 and Orientatractis moraveci n. sp. The new species is distinguished from its congeners mainly by having: 10 pairs of caudal papillae (3 pairs pre-cloacal, 2 pairs ad-cloacal and 5 pairs post-cloacal); unequal spicules of 161-198 and 69-100 µ m long; and a gubernaculum 38-58 µ m long with an antero-lateral process. Morphological and ultrastructural data on O. moraveci n. sp. and R. rondoni are presented, in addition to new genetic data based on partial 18S rDNA and 28S rDNA. The taxonomic status of Labeonema synodontisi (Vassiliadès, 1973) is discussed, suggesting that it should be returned to the genus Raillietnema.
