This research communication reports the effects of a compound enzyme preparation consisting of fibrolytic (cellulase 3500 CU/g, xylanase 2000 XU/g, ß-glucanase 17 500 GU/g) and amylolytic (amylase 37 000 AU/g) enzymes on nutrient intake, rumen fermentation, serum parameters and production performance in primiparous early-lactation (47 ± 2 d) dairy cows. Twenty Holstein-Friesian cows in similar body condition scores were randomly divided into control (CON, n = 10) and experimental (EXP, n = 10) groups in a completely randomized single-factor design. CON was fed a basal total mixed ration diet and EXP was dietary supplemented with compound enzyme preparation at 70 g/cow/d. The experiment lasted 4 weeks, with 3 weeks for adaptation and then 1 week for measurement. Enzyme supplementation significantly increased diet non-fibrous carbohydrates (NFC) content as well as dry matter intake (DMI) and NFC intake (P < 0.05). EXP had increased ruminal butyrate and isobutyrate percentages (P < 0.01) but decreased propionate and valerate percentages (P < 0.05), as well as increased serum alkaline phosphatase activity and albumin concentration (P ≤ 0.01). Additionally, EXP had increased milk yield (0.97 kg/d), 4% fat corrected milk yield and energy corrected milk yield, as well as milk fat and protein yield (P < 0.01). In conclusion, dietary supplementation with a fibrolytic and amylolytic compound enzyme preparation increased diet NFC content, DMI and NFC intake, affected rumen fermentation by increasing butyrate proportion at the expense of propionate, and enhanced milk performance in primiparous early-lactation dairy cows.

Calving interval (CI) data (N = 37,263) from 17,319 cows born 2008-2018 in six herds were assessed. The data were made available by the National Association of Hungarian Holstein Friesian Breeders in Hungary. The effects of some genetic and environmental factors, population genetic parameters, breeding value (BV) of sires, and phenotypic and genetic trends of the CI were estimated. The GLM method was used for studying different effects on the CI. BLUP animal model was used for heritability (h2) and BV estimation. Linear regression analyses were applied for the trend calculation. The mean of the CI was 412.2 ± 2.0 days. The h2 of the CI proved to be low (0.07 ± 0.01 and 0.08 ± 0.01). There were relatively high differences among the sires in the estimated BV. Based on the phenotypic trend calculation, the CI of cows showed decreasing direction by an average of 1.80 days per year (R2 = 0.94; p < 0.01). In the case of genetic trend calculation, the average BV of sires in the CI has decreased -4.94 and -0.31 days per year (R2 = 0.91 and 0.41; p < 0.01).

BACKGROUND: The insulin-like growth factor (IGF-I) and growth hormone (GH) genes have been identified as major regulators of milk yield and composition, and reproductive performance in cattle. Genetic variations/polymorphism in these genes have been found to influence milk production, yield and quality. This investigation aimed to explore the association between IGF-I and GH polymorphisms and milk yield and composition, and reproductive performance in a herd consisting of 1000 Holstein-Friesian (HF) dairy cattle from El-Alamia farm. The experimental animals were 76 ± 7.25 months in age, with an average live weight of 750 ± 50.49 kg, and raised under the same conditions of feeding and weather. The studied animals were divided into three categories; high producers (n = 280), medium producers (n = 318) and low producers (n = 402). RESULTS: The digestion of 249 bp for IGF-I-SnaBI using the Restriction-fragment-length-polymorphism (RFLP) technique yielded two alleles; T (0.59) and C (0.41) and three genotypes; TT (0.52), TC (0.39) and CC (0.09) and this agrees with the results of DNA/gene sequencing technique. The sequencing analysis of the IGF-I gene revealed polymorphism in position 472 (C > T). Nucleotide sequencing of the amplified fragment of the IGF-I gene of different genotypes was done and submitted to the NCBI GenBank with Accession no. MH156812.1 and MH156811.1. While the digestion of 432 bp for GH-AluI using the RFLP technique yielded two alleles; A (0.81) and G (0.19) and two genotypes; AA (0.77) and AG (0.23) and this agrees with the results of DNA/gene sequencing technique. The sequencing analysis of the GH gene revealed polymorphism in the position 1758 C > G and in turn led to changes in amino acid sequence as Alanine for (A) compared to Glycine for (G). Nucleotide sequencing of the amplified fragment of the GH gene was done and submitted to the NCBI GenBank with Accession no. MH156810.1. The results of this study demonstrate the effects of variants of the GH-IGF-I somatotrophic axis on milk production and composition traits in commercial HF cattle. The greatest values of milk yield and reproductive performance were observed on IGF-I-SnaBI-TC and GH-AluI-AG genotypes. While the greatest % fat and % protein values were observed on IGF-I-SnaBI-CC and GH-AluI-AA genotyped individuals. CONCLUSION: The genetic variation of the studied genes can be utilized in selecting animals with superior milk yield, composition and reproductive performance in Holstein-Friesian Dairy Cattle under subtropical conditions.

Global warming caused by climate change is a challenge for dairy farming, especially in sub-Saharan countries. Under high temperatures and relative humidity, lactating dairy cows suffer from heat stress. The objective of this study was to investigate the effects and relationship of heat stress (HS) measured by the temperature-humidity index (THI) regarding the physiological parameters and milk yield and composition of lactating Holstein Friesian crossbred dairy cows reared in the humid coastal region of Tanzania. A total of 29 lactating Holstein Friesian x Zebu crossbred dairy cows with 50% (HF50) and 75% (HF75) Holstein Friesian gene levels in the second and third months of lactation were used. The breed composition of Holstein Friesians was determined based on the animal recording system used at the Tanzania Livestock Research Institute (TALIRI), Tanga. The data collected included the daily temperature, relative humidity, daily milk yield, and physiological parameters (core body temperature, rectal temperature, respiratory rate, and panting score). THI was calculated using the equation of the National Research Council. The THI values were categorized into three classes, i.e., low THI (76-78), moderate THI (79-81), and high THI (82-84). The effects of THI on the physiological parameters and milk yield and composition were assessed. The effects of the genotype, the parity, the lactation month, and the interaction of these parameters with THI on the milk yield, milk composition, and physiological parameters were also investigated. The results show that THI and its interaction with genotypes, parity, and the lactation month had a highly significant effect on all parameters. THI influenced (p Ë 0.05) the average daily milk yield and milk fat %, protein %, lactose %, and solids-not-fat %. As the THI increased from moderate to high levels, the average daily milk yield declined from 3.49 ± 0.04 to 3.43 ± 0.05 L/day, while the fat % increased from 2.66 ± 0.05% to 3.04 ± 0.06% and the protein decreased from 3.15 ± 0.02% to 3.13 ± 0.03%. No decline in lactose % was observed, while the solid-not-fat % declined from 8.56 ± 0.08% to 8.55 ± 0.10% as the THI values increased from moderate to high. Also, the THI influenced physiological parameters (p Ë 0.05). The core body temperature (CBT), rectal temperature (RT), respiratory rate (RR) and panting score (PS) increased from 35.60 ± 0.01 to 36.00 ± 0.01 °C, 38.03 ± 0.02 to 38.30 ± 0.02 °C, 62.53 ± 0.29 to 72.35 ± 0.28 breaths/min, and 1.35 ± 0.01 to 1.47 ± 0.09, respectively, as the THI increased from low to high. The THI showed a weak positive correlation with the average daily milk yield and fat percentage, whereas the protein, lactose, and solids-not-fat percentages showed negative relationships with THI (p ≤ 0.05). CBT, RT, RR, and PS showed positive relationships (p ≤ 0.05) with THI. These negative relationships indicate that there is an antagonistic correlation between sensitivity to HS and the level of production. It is concluded that the THI, the genotype, the parity, and the lactation month, along with their interactions with THI, significantly influenced the milk yield, milk composition, and physiological parameters of lactating Holstein Friesian dairy crosses at THI thresholds ranging from 77 to 84.

Low birth weight (BW) calves exhibit higher mortality rates, reduced body weights at parturition, lower first-lactation milk yields, and longer parturition to first insemination intervals. In human medicine, small for gestational age (SGA) births are associated with increased perinatal morbidity and long-term metabolic risks. This study aimed to define SGA in Holstein Friesian (HF) calves, evaluate their body measurements and proportions, and identify its prenatal risk factors. Four linear regression models were built with weight as a function for gestation length for bull and heifer calves born from nulli- or multiparous dams. Calves with a BW below the 10th percentile were classified as SGA. Differences in body measurements were analyzed using ANOVA, and logistic regression models identified prenatal risk factors to be born SGA. Gestation length, calf sex, and dam parity were crucial variables in defining SGA. SGA calves had significantly smaller body measurements (p < 0.001) and larger body proportions (p < 0.001) compared to average and large calves. For nulliparous dams, a higher 2nd trimester temperature-humidity index (p = 0.032) and older age at parturition (>26 months, p = 0.026) significantly increased the birth of SGA calves. For multiparous dams, both low (<5800 kg, p = 0.049) and high (6700-8600 kg, p = 0.027) milk yields during gestation lead to more SGA births, although very high-yielding dams (>8600 kg) did not birth more SGA calves. This study establishes SGA in HF calves, suggests SGA calves are asymmetrical with evidence of "brain sparing", and highlights the impact of prenatal factors on calf size at birth. Further research is needed to determine the long-term effects of being born SGA on growth, reproductive performance, and productivity.

BACKGROUND: The milk's nutritional value is determined by its constituents, including fat, protein, carbohydrates, and minerals. The mammary gland's ability to produce milk is controlled by a complex network of genes. Thereby, the fat, protein, and lactose synthesis must be boost in milk to increase milk production efficiency. This can be accomplished by fusing genetic advancements with proper management practices. Therefore, this study aimed to investigate the association between the Lipoprotein lipase (LPL), kappa casein CSN3, and Glucose transporter 1 (GLUT1) genes expression levels and such milk components as fat, protein, and lactose in different dairy breeds during different stages of lactation. METHODS: To achieve such a purpose, 94 milk samples were collected (72 samples from 36 multiparous black-white and red-white Holstein-Friesian (HF) cows and 22 milk samples from 11 Egyptian buffaloes) during the early and peak lactation stages. The milk samples were utilized for milk analysis and genes expressions analyses using non- invasive approach in obtaining milk fat globules (MFGs) as a source of Ribonucleic acid (RNA). RESULTS: LPL and CSN3 genes expressions levels were found to be significantly higher in Egyptian buffalo than Holstein-Friesian (HF) cows as well as fat and protein percentages. On the other hand, GLUT1 gene expression level was shown to be significantly higher during peak lactation than early lactation. Moreover, lactose % showed a significant difference in peak lactation phase compared to early lactation phase. Also, fat and protein percentages were significantly higher in early lactation period than peak lactation period but lactose% showed the opposite pattern of Egyptian buffalo. CONCLUSION: Total RNA can be successfully obtained from MFGs. The results suggest that these genes play a role in glucose absorption and lactose synthesis in bovine mammary epithelial cells during lactation. Also, these results provide light on the differential expression of these genes among distinct Holstein-Friesian cow breeds and Egyptian buffalo subspecies throughout various lactation phases.

The serious drawback underlying the biological annotation of whole-genome sequence data is the p >> n problem, which means that the number of polymorphic variants (p) is much larger than the number of available phenotypic records (n). We propose a way to circumvent the problem by combining a LASSO logistic regression with deep learning to classify cows as susceptible or resistant to mastitis, based on single nucleotide polymorphism (SNP) genotypes. Among several architectures, the one with 204,642 SNPs was selected as the best. This architecture was composed of two layers with, respectively, 7 and 46 units per layer implementing respective drop-out rates of 0.210 and 0.358. The classification of the test data resulted in AUC = 0.750, accuracy = 0.650, sensitivity = 0.600, and specificity = 0.700. Significant SNPs were selected based on the SHapley Additive exPlanation (SHAP). As a final result, one GO term related to the biological process and thirteen GO terms related to molecular function were significantly enriched in the gene set that corresponded to the significant SNPs. Our findings revealed that the optimal approach can correctly predict susceptibility or resistance status for approximately 65% of cows. Genes marked by the most significant SNPs are related to the immune response and protein synthesis.

We evaluated the relationship between decreased pregnancy-associated glycoprotein (PAG) levels, inflammatory parameters (serum amyloid A [SAA] and milk amyloid A [MAA]), postpartum inflammatory conditions (mastitis, ketosis, and follicular cysts), and the FOXP3 gene. Nineteen Holstein-Friesian cows were included in this study. Up to approximately eight weeks after delivery, weekly health examinations were performed for mastitis and ketosis, and reproductive organ ultrasonography was performed. The decreasing PAG rate was negatively correlated with SAA concentration (r = -0.493, p = 0.032). Cows with mastitis exhibited a slower trend of PAG decrease (p = 0.095), and a greater percentage of these cows had MAA concentrations above 12 µg/mL (p = 0.074) compared with those without mastitis. A negative correlation, although nonsignificant (r = -0.263, p = 0.385), was observed between the day-open period and decreased PAG rate. The day-open period was correlated with the presence or absence of follicular cysts (p = 0.046). Four cows that developed follicular cysts were homozygous for the G allele of the FOXP3 gene related to repeat breeders. These results indicate a relationship between a decreased PAG rate and inflammatory status during the postpartum period. Thus, suppressing inflammation during the perinatal period may improve reproductive efficiency in the dairy industry.

A stop-gain mutation (rs715966442; BTA11: 1,02,463,944 nucleotide position) in transcription termination factor, RNA polymerase I (TTF1) gene causes abortion in Holstein Friesian (HF) cattle. A PCR-restriction fragment length polymorphism (PCR-RFLP)-based genetic test has been developed and validated to screen the TTF1 mutation locus in HF cattle. The mutation locus was screened in 80 HF and HF crossbreds using the protocol, which revealed two animals as carriers of the mutant TTF1 allele. The test employed is cost-effective, rapid and precise and can be utilized as an effective tool for the screening of TTF1 mutation carriers in HF cattle population.

The dairy industry is reshaping itself and becoming commercialized in Pakistan due to the increased demand for milk to overcome the shortage. Exotic breeds such as Holstein Friesian, a high milk producing breed have started being reared more on farms in Pakistan. Along with other issues, mastitis does affects the milk production of this breed. The objective of this study was to evaluate the milk composition in terms of bacterial communities in Holstein Friesian reared in Punjab, Pakistan and alteration in microbial composition with healthy and mastitic udder. Milk samples (n = 36) from farms rearing Holstein Friesian were collected. Among these samples, 05 samples from each three groups, HHF(healthy), CHF (clinical mastitis) and SHF (subclinical mastitis), based on their udder health condition, were processed using the 16 S r=RNA gene based technique. Diversity assessment as carried out by alpha diversity indices showed that milk samples from the udder infected with clinical mastitis were the least diverse and those from the healthy udder were more diverse. Beta diversity across samples showed a scattered pattern suggesting overlap amongst bacterial communities across different groups samples as depicted by PCA plots of beta diversity indices. The taxonomic profile revealed that Proteobacteria Firmicutes, Bacteroidota and Actinobacteriota were the major phyla detected across all groups. Proteobacteria dominated the HHF and SHF group while abundance of Firmicutes was higher in CHF group. Differences at other levels including order, genus and species were also recorded. The overall picture concludes that diverse microbiota is associated with different udder health conditions.

To improve the genomic evaluation of milk-related traits in Holstein-Friesian (HF) cattle it is essential to identify the associated candidate genes. Novel SNP-based analyses, such as the genetic mapping of inherited diseases, GWAS, and genomic selection, have led to a new era of research. The aim of this study was to analyze the association of each individual SNP in Serbian HF cattle with milk production traits and inbreeding levels. The SNP 60 K chip Axiom Bovine BovMDv3 was deployed for the genotyping of 334 HF cows. The obtained genomic results, together with the collected phenotypic data, were used for a GWAS. Moreover, the identification of ROH segments was performed and served for inbreeding coefficient evaluation and ROH island detection. Using a GWAS, a polymorphism, rs110619097 (located in the intron of the CTNNA3 gene), was detected to be significantly (p < 0.01) associated with the milk protein concentration in the first lactation (adjusted to 305 days). The average genomic inbreeding value (FROH) was 0.079. ROH islands were discovered in proximity to genes associated with milk production traits and genomic regions under selection pressure for other economically important traits of dairy cattle. The findings of this pilot study provide useful information for a better understanding of the genetic architecture of milk production traits in Serbian HF dairy cows and can be used to improve lactation performances in Serbian HF cattle breeding programs.

The transition period in high-yielding dairy cows is a critical phase marked by an elevated risk of oxidative stress. This study evaluated the effect of oral selenitetriglyceride supplementation on oxidative stress management in periparturient cows. A controlled experiment was conducted on 12 cows, divided into two groups: the experimental group (STG) received selenitetriglycerides (0.5 mg Se/kg BW), while the control group (CON) was given a placebo, starting 12 days before calving until the calving day. Blood and liver tissue samples were collected at predetermined intervals around the time of parturition. The study observed a significant increase in serum selenium levels and NEFA stabilization in the STG group compared with the control. Antioxidant parameters indicated elevated GSH-Px and CAT concentrations in the STG group. Liver gene expression analysis revealed a significant increase in SOD2 mRNA levels in the STG group (FC = 4.68, p < 0.01). Conversely, GSH-Px3 expression significantly decreased (FC = 0.10, p < 0.05) on the 7th day postpartum in the CON group. However, SOD1, SOD3, and CAT expressions remained stable in both groups. These findings highlight the beneficial role of selenitetriglycerides in enhancing antioxidant capacity and influencing specific gene expressions associated with oxidative stress management in dairy cows during the peripartum period.

AIMS: To use a farm-based survey to identify characteristics of the New Zealand dairy system associated with the risk of spontaneous humeral fracture in dairy heifers. METHODS: A questionnaire was designed and made available in print and online to collect information from dairy farmers and/or veterinarians, across New Zealand, about the management and nutrition of cows from birth to first lactation. Data were collected from July 2019 to March 2020 from farms that either had recorded (case farms) or not recorded (control farms) cases of humeral fractures in dairy heifers. RESULTS: A total of 68 completed questionnaires were returned, with 35 responses from case farms and 33 responses from control farms. Twenty-six responses (38%) were from the South Island (13 case farms and 13 control farms) and 38 responses (56%) were from the North Island (20 case farms and 18 control farms). For four questionnaires (6%) farm location was not given. Adjusting for the effect of age when calves accessed pasture, case farms had increased odds of having Holstein-Friesian Jersey crossbreed cows as the predominant breed (OR = 9.7; 95% CI = 3.1-36.0; p < 0.001). Adjusting for the effect of breed, allowing calves access to pasture a week later decreased the odds of being a case farm (OR = 0.68; 95% CI = 0.47-0.90; p = 0.006). CONCLUSIONS: Cows being Holstein-Friesian Jersey crossbreed was identified as a possible risk factor associated with spontaneous humeral fracture in dairy heifers in New Zealand. Given the small sample size, the likely multifactorial aetiology for humeral fractures, and the non-randomised survey, this risk factor, and the possible association between age at turn out and herd production with humeral fractures, all require further investigation.

Selection for resilience indicator (RIND) traits in Holstein cattle is becoming an important breeding objective as the worldwide population is expected to be exposed to increased environmental stressors due to both climate change and changing industry standards. However, genetic correlations between RIND and productivity indicator (PIND) traits, which are already being selected for and have the most economic value, are often unfavorable. As a result, it is necessary to fully understand these genetic relationships when incorporating novel traits into selection indices, so that informed decisions can be made to fully optimize selection for both groups of traits. In the past 2 decades, there have been many estimates of RIND traits published in the literature, albeit in small populations. To provide valuable pooled summary estimates, a random-effects meta-analysis was conducted for heritability and genetic correlation estimates for PIND and RIND traits in worldwide Holstein cattle. In total, 926 heritability estimates for 9 PIND and 27 RIND traits, along with 362 estimates of genetic correlation (PIND × RIND traits) were collected. Resilience indicator traits were grouped into the following subgroups: Metabolic Diseases, Hoof Health, Udder Health, Fertility, Heat Tolerance, Longevity, and Other. Pooled estimates of heritability for PIND traits ranged from 0.201 ± 0.05 (energy-corrected milk) to 0.377 ± 0.06 (protein content), while pooled estimates of heritability for RIND traits ranged from 0.032 ± 0.02 (incidence of lameness, incidence of milk fever) to 0.497 ± 0.05 (measures of body weight). Pooled estimates of genetic correlations ranged from -0.360 ± 0.25 (protein content vs. milk acetone concentration) to 0.535 ± 0.72 (measures of fat-to-protein ratio vs. milk acetone concentration). Additionally, out of 243 potential genetic correlations between PIND and RIND traits that could have been reported, only 40 had enough published estimates to implement the meta-analysis model. Our results confirmed that the interactions between PIND and RIND traits are complex, and all relationships should be evaluated when incorporating novel traits into selection indices. This study provides a valuable reference for breeders looking to incorporate RIND traits for Holstein cattle into selection indices.

Bovine leukemia virus (BLV) is a retrovirus that primarily infects dairy cows. Although few studies have also used the tax gene, phylogenetic studies of BLV use mostly the env gene. The aim of this work was to establish the circulating genotypes of BLV in specialized dairy cattle from Antioquia, Colombia. Twenty blood samples from Holstein Friesian cows were collected, and their DNA was isolated. A PCR was performed for a partial region of the env and tax genes. A phylogenetic analysis was carried out using the maximum likelihood and Bayesian methods for both genes. Nineteen sequences were identified as genotype 1 by env and tax genes. Only one sequence was clustered with genotype 3 and had the highest proportion of different nucleotide sites compared to other strains. Four amino acid substitutions in the 134 amino acid residue fragment of the Env protein were identified in the Colombian sequences, and three new amino acid substitutions were reported in the 296 amino acid residue fragment of the Tax protein. R43K (Z finger), A185T (Activation domain), and L105F changes were identified in the genotype 3 sample. This genotype has been reported in the United States, Japan, Korea, and Mexico, but so far, not in Colombia. The country has a high rate of imported live animals, semen, and embryos, especially from the United States. Although it is necessary to evaluate samples from other regions of the country, the current results indicate the presence of two BLV genotypes in specialized dairy herds.

Abstract Background: Farm profitability depends on raising healthy calves, which rests on successful feeding and management programs. Objective: To determine the effects of weaning weight on growth performance, feed efficiency, behavioral traits, as well as feeding cost of Holstein-Friesian and Brown Swiss calves. Methods: Sixty-seven newborn dairy calves were allocated into four weaning-weight groups [60 kg (n=18), 65 kg (n=19), 70 kg (n=18) and 75 kg (n=12)]. Body weights (BW) and measurements were recorded at birth, weaning, and four months of age. An instantaneous sampling method was used to collect behavioral data. Results: Daily weight gain of Brown Swiss calves was higher than Holsteins between weaning and four months of age (p<0.05). During pre-weaning, daily weight gain of calves weaned at 60 kg was lower than the other three groups (p<0.05). Feed efficiency of Holstein-Friesian was higher than Brown Swiss calves in the period between birth and four months of age (p<0.05). Chest depth of calves weaned at 60 kg was (p<0.05) lower compared to the other groups. Water drinking behavior during the pre-weaning period was notably higher in calves weaned at 75 kg (p<0.05). Conclusions: Our results suggest that dairy calves can be weaned at 60 kg of BW without detrimental effects on performance, and their feed cost per kg weight gain is significantly lower compared to higher weaning weights.
Resumen Antecedentes: La crianza de terneros sanos es uno de los principales requisitos para asegurar la rentabilidad de la granja, y depende en gran medida del éxito en los programas de alimentación y manejo de los terneros. Objetivo: Determinar los efectos del peso al destete sobre el rendimiento del crecimiento, la eficiencia alimenticia, las características de comportamiento y el costo de alimentación de terneros Holstein-Friesian y Pardo suizo. Métodos: Sesenta y siete terneros lecheros recién nacidos se distribuyeron en cuatro grupos diferentes de peso al destete [60 kg (n=18), 65 kg (n=19), 70 kg (n=18) y 75 kg (n=12)]. Los pesos y medidas corporales se registraron al nacimiento, al destete, y a los cuatro meses de edad. Se utilizó un método de muestreo instantáneo para recopilar los datos de comportamiento. Resultados: La ganancia diaria de peso de los terneros Pardo suizo entre el destete y los cuatro meses de edad fue mayor que la de los Holstein-Friesian (p<0,05). Además, las ganancias diarias de peso de los terneros destetados con 60 kg de peso corporal (PC) fueron menores que las de los otros tres grupos durante el período previo al destete (p<0,05). La eficiencia alimenticia de los terneros Holstein-Friesian fue mayor que la de los Pardo suizo en el período entre nacimiento y cuatro meses de edad (p<0,05). La profundidad del pecho de los terneros en el grupo destetado a 60 kg fue menor que en otros grupos (p<0,05). El comportamiento de consumo de agua fue notablemente superior en los terneros destetados a 75 kg de PC durante el período previo al destete (p<0,05). Conclusiones: Nuestros resultados sugieren que los terneros lecheros pueden destetarse a los 60 kg de PC sin efectos perjudiciales sobre su rendimiento, y su costo de alimento por 1 kg de aumento de peso es significativamente menor en comparación con mayores pesos de destete.
Resumo Antecedentes: A criação de bezerros saudáveis é um dos principais requisitos para garantir a rentabilidade dos empreendimentos e depende muito do sucesso nos programas de alimentação e manejo dos bezerros. Objetivo: Determinar os efeitos do peso ao desmame sobre o desempenho de crescimento, eficiência alimentar, características comportamentais e custo de alimentação de bezerros da raça Holstein-Friesian e Pardo suíço. Métodos: Sessenta e sete bezerros recém-nascidos foram alocados em quatro diferentes grupos de peso ao desmame [60 kg (n=18), 65 kg (n=19), 70 kg (n=18) e 75 kg (n=12)]. Os pesos e medidas corporais foram determinados ao nascimento, desmame e 4 meses de idade. O método de amostragem instantânea foi utilizado para coletar os dados comportamentais. Resultados: O ganho de peso diário dos bezerros Pardos suíços entre o desmame e os 4 meses de idade foi significativamente maior do que os da Holstein-Friesian (p<0,05). Além disso, o ganho de peso diário dos bezerros desmamados com 60 kg de peso corporal (PC) foi significativamente (p<0,05) menor do que os outros três grupos no período pré-desmame. A eficiência alimentar dos bezerros Holstein-Friesian foi significativamente maior do que a dos bezerros Pardo suíço nos períodos entre o nascimento e os 4 meses de idade (p<0,05). O crescimento na profundidade do peito dos bezerros do grupo desmamado com 60 kg de PC foi significativamente (p<0,05) menor do que os demais grupos. Apenas o comportamento de beber foi notavelmente maior (p<0,05) nos bezerros desmamados com 75 kg de PC no período pré-desmame. Conclusões: Os resultados deste estudo sugerem que bezerros leiteiros podem ser desmamados com 60 kg de PC sem qualquer efeito prejudicial sobre seu desempenho e o custo de alimentação para ganho de peso de 1 kg é significativamente menor em comparação com pesos de desmame avançado.

An important factor for dairy cattle farmers is the profitability of cattle rearing, which is influenced by the animals' health and reproductive parameters, as well as their genomic stability and integrity. Bovine viral diarrhea (BVD) negatively affects the health of dairy cattle and causes reproductive problems. The aim of the study was to identify genomic instability in cows with reproductive disorders following infection with the BVD virus. The material for analysis was peripheral blood from Holstein-Friesian cows with reproductive problems, which had tested positive for BVD, and from healthy cows with no reproductive problems, which had tested negative for BVD. Three cytogenetic tests were used: the sister chromatid exchange assay, fragile sites assay, and comet assay. Statistically significant differences were noted between the groups and between the individual cows in the average frequency of damage. The assays were good biomarkers of genomic stability and enabled the identification of individuals with an increased frequency of damage to genetic material that posed a negative impact on their health. The assays can be used to prevent disease during its course and evaluate the genetic resistance of animals. This is especially important for the breeder, both for economic and breeding reasons. Of the three assays, the comet assay proved to be the most sensitive for identifying DNA damage in the animals.

This study focused on assessing blood metabolites and somatic cell count (SCC) during the transition period in Achai and Holstein Friesian cows. Blood samples were collected from cows of both breeds on days -21, -14, -7 (pre-partum period), on day 0 (parturition day) and then on days 7, 14 and 21 (post-partum period). Additionally, milk samples were obtained on the day of parturition and on days 7, 14 and 21 post-partum for SCC assessment. Among Holstein cows, blood glucose levels showed a significant increase on day -21 before calving, and a decrease on the 14th and 21st days post-partum. Similarly, the blood triglycerides concentration exhibited a significant rise in Holstein cows 21 days before calving, while Achai cows experienced a significant decline in blood triglycerides on the day of parturition and throughout the entire post-partum period. Comparing Holstein and Achai cows, blood cholesterol was significantly higher in Holstein cows on day -21 before calving, whereas Achai cows had significantly lower blood cholesterol levels on the day of parturition. Regarding blood protein, Holstein cows exhibited significantly higher levels on day 14 post-partum, while Achai cows had lower protein levels 7 days before calving. The results further demonstrated that SCC was significantly elevated in Holstein cows on day 21 of parturition and lower on the day of parturition in Achai cows. Overall, the metabolic profile trend and SCC were comparable, yet the studied parameters were more pronounced in Holstein Friesian compared to Achai cows.

BACKGROUND: The Kumamoto strain of Japanese Brown (JBRK) cattle is a sub-breed of Wagyu and has a different genetic background than that of Japanese Black (JB) cattle. Bovine leukemia virus (BLV) is the pathogen causing enzootic bovine leukosis (EBL), the predominant type of bovine leukosis (BL). EBL is one of the most common bovine infectious diseases in dairy countries, including Japan. Some host genetic factors, including the bovine leukocyte antigen (BoLA)-DRB3 gene, have been associated with the proviral load (PVL) of BLV and/or onset of EBL. Here, we determined the number of BL cases by analyzing prefectural case records in detail. We measured the PVL of BLV-infected JBRK cattle and compared it with that obtained for other major breeds, JB and Holstein-Friesian (HF) cattle. Finally, the relationship between PVL levels and BoLA-DRB3 haplotypes was investigated in BLV-infected JBRK cattle. RESULTS: We determined the number of BL cases recorded over the past ten years in Kumamoto Prefecture by cattle breed. A limited number of BL cases was observed in JBRK cattle. The proportion of BL cases in the JBRK was lower than that in JB and HF. The PVL was significantly lower in BLV-infected JBRK cattle than that in the JB and HF breeds. Finally, in BLV-infected JBRK cattle, the PVL was not significantly affected by BoLA-DRB3 alleles and haplotypes. BoLA-DRB3 allelic frequency did not differ between BLV-infected JBRK cattle with low PVL and high PVL. CONCLUSIONS: To our knowledge, this is the first report showing that BL occurred less in the JBRK population of Kumamoto Prefecture. After BLV-infection, the PVL was significantly lower in JBRK cattle than that in JB and HF breeds. The genetic factors implicated in maintaining a low PVL have yet to be elucidated, but the BoLA-DRB3 haplotypes are likely not involved.

Introduction: Cattle health and welfare are monitored via the analysis of the haematological profile, and it shows cattle's ability to adapt to changing environmental conditions, pregnancy and lactation; profile changes also indicate reproductive disorders. The literature lacks reports of the examination of the haematological profile in cows up to the 50th day of pregnancy (dop). Therefore, this research examined that in cows up to this pregnancy stage. Material and Methods: A total of 101 Polish Holstein-Friesian black-and-white cows were divided into groups. The control groups consisted of non-pregnant heifers (group C00) and non-pregnant cows (group C0), and the experimental groups were pregnant heifers (group T1 at dop ≤ 28 and group T2 at dop ≥ 29-dop < 45) and pregnant cows (group T3 at dop ≥ 29-dop ≤ 50). In addition, the T3 group was divided into cows pregnant for up 45 dop and cows between 45 and 50 dop. Blood samples were collected in March and April 2021 from each animal and analysed. A transrectal ultrasound examination was performed to detect and confirm pregnancy. Results: Statistically significant differences (P ≤ 0.01) between the group of cows at dop < 45 dop and those at dop ≥ 45-dop ≤ 50 dop were noted in granulocyte percentage (GRA%), white and red blood cell counts (WBC/RBC), platelets (PLT), platelet distribution width (PDW), haematocrit (HCT) and lymphocyte percentage (LYM%). No statistically significant differences were found in the mean corpuscular haemoglobin, monocytes (MON), monocyte percentage (MON%), mean platelet volume (MPV), thrombocrit or red blood cell distribution width (RDW). Similar statistically significant differences (P ≤ 0.01) emerged between the groups of heifers in PLT, GRA, RBC, lymphocytes, LYM% and HCT, and no significant differences were found between MPV, MON, MON% or RDW. Conclusion: Examining the haematological profile in high-yielding cattle is vital in maintaining herd profitability and high reproduction, which depend on the quick diagnosis of disorders facilitated by haematology. This study analysed the haematology profile of dairy cattle at dop ≤ 50 for the first time, indicating changes in lymphocyte levels, which suggests that the animals experienced direct stress during the study.
