RESUMO

This study aims to assess hepatoprotective properties of M. rotundifolia. Decoction was used to prepare the aqueous extract. The preliminary cytotoxicity evaluated against Caco 2 and RAW 264 cells demonstrate no cytotoxic effect. The preventive impact of the extract against liver damage was evaluated by examining blood levels of AST, ALT, ALP, total proteins, and histological alterations in liver tissues. Thirty albino rats were separated into five groups: the first served as normal group, the second was injected by olive oil (3 ml/kg), and the third was injected by CCL4 (3 ml/kg). However, groups IV and V received daily doses of 250 and 500 mg extract/kg bw, respectively before CCL4 injection. The results showed that the administration of the extract led to a marked improvement in plasma biochemical markers and a reduction in symptoms of CCL4-induced liver damage. The extract exhibits hepatoprotective activity, which may be attributed to its phytochemical components.

Mints are aromatic plants of Lamiaceae, globally known for the phytochemical-rich essential oils. Most of the cultivated mints are menthol-rich, whereas spearmint being the only dominant carvone-rich species. In this study, another carvone-rich mint Mentha rotundifolia (L.) Huds., a native of temperate region was assessed for its acclimation in sub-tropical environment to see any possible changes in specialized metabolite accumulation. Plants grown under open environment was compared with glasshouse grown plants where, temperature, humidity and photoperiods were uniformly maintained. Thickened leaves with increased cuticular wax load (2.82 folds) and anthocyanin accumulation (202.97 µg/g) in the widened stems were observed in plants grown in open environment, while higher chlorophyll contents were exhibited by the glasshouse-grown plants. Enhanced antioxidant capacity in open environment, correlated with elevated concentration (86.4% increase for caffeic acid) of wall-bound phenolics was observed. Increased proline, hydrogen peroxide and malondialdehyde contents in open environment indicated the plant's ability to cope up with abiotic stress. Higher amounts of terpenes like (-)-carvone (2.68 folds) and D-limonene (1.35 folds) were found in both internal volatile pool and essential oil of glasshouse-grown plants. Histochemical study of glandular trichomes also supported this finding. In conclusion, glasshouse-grown plants showed relatively better growth and higher terpene contents, nevertheless the plant survived well in warmer environment, with increased antioxidant capacities and phenolic contents. Future study includes mass propagation of this species in different geographical locations with distinct climatic variations to determine the suitable sub-tropical locations for cultivation as a potential alternative to spearmint for commercial-scale (-)-carvone production. Introduction: Aging is often linked to oxidative stress, where the body experiences increased damage from free radicals. Plants are rich sources of antioxidants, playing a role in slowing down aging and supporting the proper functioning and longevity of cells. Our study focuses on exploring the impact of Mentha rotundifolia (MR) and Salvia officinalis (SO) hydrosols on aging-related comorbidities. Methods: The chemical composition of MR and SO hydrosols was analyzed by gas chromatography coupled to mass spectrometry. 2,2-Diphenyl 1-picrylhydrazyl and 2,20-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid radicals scavenging assays were used to assess their in vitro antioxidant activity, and heat induced albumin denaturation test was used to evaluate their anti-inflammatory activity. Subsequently, we administered 5% of each plant hydrosol in the drinking water of 18-month-old rats for six months. We then conducted behavioral tests, including open field, dark/light box, rotarod, and Y-maze assessments, and measured biochemical parameters in plasma, liver and brain tissues. Results and discussion: At two years old, animals treated with MR and SO hydrosols displayed fewer physical and behavioral impairments, along with well-preserved redox homeostasis in comparison with animals in the control group. These results highlighted the significance of MR and SO hydrosols in addressing various aspects of age-related comorbidities. The study suggests that these plant-derived hydrosols may have potential applications in promoting healthy aging and mitigating associated health challenges.

Introduction: Oxidative stress plays a causal role in neurodegenerative diseases. The aim of this study is to evaluate the antioxidant and neuroprotective effects of Mentha rotundifolia (L.) Huds (M. rotundifolia), a widely used Moroccan plant in traditional medicine. Methods: The chemical composition of M. rotundifolia aqueous extract was analyzed by liquid chromatography coupled to mass spectrometry (LC-MS). 2,2-diphenyl 1-picrylhydrazyl (DPPH) and 2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid (ABTS+) assays were used to assess its in vitro antioxidant activity. H2O2 was utilized to induce oxidative stress and neurotoxicity in vivo. Behavioral changes were evaluated using Open Field, Y-maze and Rotarod tests. Hyperalgesia was assessed using the tail immersion test. Results and discussion: The LC-MS/MS analysis revealed high content of kaempferol glucuronide (85%) at the extract. IC50 values of the DPPH and ABTS were 26.47 and 41.21 µg/mL, respectively. Pre-treatments with M. rotundifolia extract attenuated the behavioral changes induced by H2O2. In addition, the latency of tail withdrawal increased significantly in the treated groups suggesting central analgesic effect of M. rotundifolia extract. Moreover, the extract attenuated the deleterious effects of H2O2 and improved all liver biomarkers. The obtained results suggested that M. rotundifolia had remarkable antioxidant and neuroprotective effects and may prevent oxidative stress related disorders.

Phenolic profiling of ten plant samples of Mentha rotundifolia (L.) Huds. collected from different bioclimatic areas of Tunisia, was for the first time carried out by using a fast ultra-high-performance liquid chromatography (UHPLC)-high resolution tandem mass spectrometry (HRMS/MS) method on a Q Exactive platform equipped with an electrospray ionization (ESI) source. An intraspecific, interpopulation variability was evidenced and a total of 17 polyphenolic metabolites were identified and quantified by using the UHPLC-HRESIMS/MS method, here validated for specificity, linearity, limit of detection, limit of quantitation, accuracy and precision. The quantitative method resulted sensitive at the nM level and reliable for rapid polyphenol quantification in vegetal matrices. The metabolomic study allowed us to identify a new compound, named salvianolic acid W, which was isolated and characterized mainly by NMR and MS analysis. A statistical correlation of the phenolic composition with antioxidant and anti-acetylcholinesterase activities was provided.

Background The Mentha rotundifolia L. (Lamiaceae family), is a medicinal herb used since the ancient times as an antiseptic, analgesic and anti-inflammatory agent. In the present work, metabolomic profiling of two Mentha rotundifolia L. ecotypes leaf tissues spontaneously growing in the North of Tunisia was achieved. Methods Phenolic contents (TPC, TFC and TTC) were assessed using colorimetric methods. Metabolomic profiling of leaf tissues extracts was assessed based on Gas Chromatography-Mass Spectrometry (GC/MS) analysis. The antioxidant ability of M. rotundifolia extracts was achieved based on two test systems namely DPPH and FRAP assays. Antimicrobial activity against a set of Gram negative and Gram positive bacteria was estimated by measuring ID, MIC and MBC values. Results Fifty metabolites were identified as belonging mainly to phenolics, fatty acids, terpenes, steroids and aldehydes classes with qualitative and quantitative variability. Most of the identified compounds are reputed bioactive with potent antioxidant, antimicrobial and anti-inflammatory among others effects. To confirm these findings common in vitro biological activities were achieved. The investigated extracts showed significant antioxidant abilities based on both 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Ferric reducing antioxidant potential (FRAP) assays. Furthermore, the extracts revealed promising antimicrobial ability against tested Gram+ and Gram- bacterial strains (ID: 12.5-14.5 mm, MIC: 3.125-25 (µg/mL), MBC: 6.25-100 (µg/mL)). Conclusions Based on our findings Mentha rotundifolia L. leaves extracts present a potential source of natural antioxidants and diverse bioactive compounds which could be used in green pharmacy, food preservation, alternative medicine and natural therapies.

A new microwave-assisted extraction (MAE) method using ethanol as solvent has been optimized by means of a Box-Behnken experimental design for the enhanced extraction of bioactive terpenoids from Mentha rotundifolia leaves; 100°C, 5 min, 1.125 g dry sample: 10 mL solvent and a single extraction cycle were selected as optimal conditions. Improved performance of MAE method in terms of extraction yield and/or reproducibility over conventional solid-liquid extraction and ultrasound assisted extraction was also previously assessed. A comprehensive characterization of MAE extracts was carried out by GC-MS. A total of 46 compounds, mostly terpenoids, were identified; piperitenone oxide and piperitenone were the major compounds determined. Several neophytadiene isomers were also detected for the first time in MAE extracts. Different procedures (solid-phase extraction and activated charcoal (AC) treatment) were also evaluated for clean-up of MAE extracts, with AC providing the highest enrichment in bioactive terpenoids. Finally, the MAE method here developed is shown as a green, fast, efficient and reproducible liquid extraction methodology to obtain M. rotundifolia bioactive extracts for further application, among others, as food preservatives.

Chemical composition and antifungal activity of essential oils of Algerian Mentha species were studied. Chemical compositions of different Mentha species oils (Mentha rotundifolia, M. spicata, M. pulegium, and M. piperita) were investigated by capillary GC and GC/MS, and their antifungal activities were evaluated by means of paper disc diffusion method and minimum inhibitory concentration (MIC) assays. In total, 98 components from all Mentha species were identified. All oils were rich in monoterpene-oxygenated components. In addition, we reported fumigant antifungal activity of Algerian Mentha essential oils against four fungi: Botrytis cinerea, Penicillium expansum, Monilinia laxa, and M. fructigena. All oils demonstrated very good inhibition especially against B. cinerea, M. laxa, and M. fructigena. Both Monilinia fungi were extremely sensitive to all Algerian Mentha oils, which suggests that Mentha essential oils have the potential to be used as bio-pesticides to protect fruit trees, such as apple and pear trees, and provides an alternative to chemical pesticides.

Background It is important to consider the optimum conditions and processing factors (like solvent type) influencing activity of plant antioxidants for utilization in food and biological systems. Methods The antioxidant capacity and phenolic content of two Mentha species, namely, Mentha pulegium L. (MP) and Mentha rotundifolia (L.) Huds (MR), were studied and six solvent systems were used. The total antioxidant capacity of the mint species extracts was evaluated using phosphomolybdenum method and the free radical-scavenging capacity by 2,2-diphenyl-1-picrylhydrazyl radical-scavenging assay. Results The efficiency of the used solvents to extract phenols from the two species varied considerably. The highest total phenolic content was obtained from methanol extract of MP (25.3±1.3 mg GAE/gdw) and total flavonoid content from methanol extract of MR (10.1±0.1 mg QE/gdw). High phenol content was significantly correlated with high antioxidant capacity. The methanol extracts showed the highest radical scavenging activity. All the extracts showed variable antioxidant capacity by the formation of phosphomolybdenum complex. Acetone extract of MP and methanol extract of MR exhibited marked reducing power in this method. Conclusions Our findings identified the appropriate solvent for extracting MP and MR phenolics which might provide a rich source of natural antioxidants.

CONTEXT: Mints (Lamiaceae) are used as traditional remedies for the treatment of several diseases. Their extracts are recognized as anti-inflammatory compounds. OBJECTIVE: This study characterized the cytotoxic effects of Mentha spicata L. (MS), Mentha pulegium L. (MP) and Mentha rotundifolia (L). Huds (MR) on macrophage cells (RAW264.7; U937) and determined their impact on apoptosis and autophagy, which can play a role in controlling inflammation. MATERIALS AND METHODS: The extracts were prepared in culture medium and tested from 25 to 400 µg/mL after 24-48 h of treatment. To show the effect of the aqueous ethanol (50%) extracts on apoptosis and authophagy, the presence of cleaved caspase-3, and the conversion of LC3-I to LC3-II was evaluated by Western blotting. RESULTS: Compared with the MTT assay, crystal violet showed a pronounced decrease in the number of cells with all extracts at 48 h. Calculated IC50 values were 257.31, 207.82 and 368.02 µg/mL for MS, MP and MR, respectively. A significant increase in PI positive cells was observed with all extracts at 200-400 µg/mL. Mitochondrial dysfunctions and nuclear morphological changes were detected with MS and MR extracts at 400 µg/mL. At this concentration, no cleaved caspase-3 was found whereas stabilized caspase-3 in its dimeric form was identified. MS and MR extracts also favour LC3-I to LC3-II conversion which is a criterion of autophagy. CONCLUSIONS: The cytotoxic profiles depend on the extracts considered; MS extract showed the strong activity. However, all the mint extracts studied interact with the apoptotic and autophagic pathways at elevated concentrations.
