RESUMO

Paralytic shellfish toxins (PSTs) are potent neurotoxins produced by certain microalgae, particularly dinoflagellates, and they can accumulate in shellfish in coastal seawater and thus pose significant health risks to humans. To explore the relationship between toxicity and PST profiles in seawater and mussels, the spatiotemporal variations in PST concentrations and profiles were investigated along the southern coast of Korea under peak PST levels during spring. Seawater and mussel samples were collected biweekly from multiple stations, and the toxin concentrations in the samples were measured. Moreover, the dinoflagellate community composition was analyzed using next-generation sequencing to identify potential PST-producing species. The PST concentrations and toxin profiles showed substantial spatiotemporal variability, with GTX1 and GTX4 representing the dominant toxins in both samples, and C1/2 tending to be higher in seawater. Alexandrium species were identified as the primary sources of PSTs. Environmental factors such as water temperature and salinity influenced PST production. This study demonstrates that variability in the amount and composition of PSTs is due to intricate ecological interactions. To mitigate shellfish poisoning, continuous monitoring must be conducted to gain a deeper understanding of these interactions.

As a kind of obligate biotrophic fungus, Puccinia striiformis f. sp. tritici (Pst) secretes vast effectors via haustoria to host cells during the infection to inhibit host defense responses and promote fungal invasion. In this study, based on the completion of genome sequencing and haustorial transcriptome sequencing of Pst, we identified a Pst effector (Hasp155) that is significantly induced in the early stage of Pst infection to wheat. The 18 N-terminal amino acids of Hasp155 encoded a signal peptide with a secretory function. Transient expression of Hasp155 in Nicotiana benthamiana inhibited Bax-induced cell death as well as chitin-triggered callose deposition and defense-related gene expression. Moreover, delivery of the Hasp155 protein into wheat cells via type three secretion systems (TTSS) led to reduced plant immunity to nonpathogenic bacteria and to the avirulent Pst race with decreased H2O2 accumulation and promoted Pst development. Furthermore, transgenic overexpression of Hasp155 significantly renders wheat resistance susceptible, resulting in a decreased defense response and increased Pst pathogenicity. Overall, these results indicate that Hasp155 is an important effector of Pst pathogenicity by suppressing plant immunity.

The study of interactions between copepods of the genus Acartia and toxic dinoflagellates of the genus Alexandrium has been an important topic during the last four decades. Feeding behavior and physiological responses of copepods have been studied in laboratory and field experiments, sometimes with contradictory results. More recently, an evolutionary adaptive mechanism leading to enhanced tolerance of Alexandrium toxins in a population of Acartia experiencing chronic exposure to these dinoflagellates has been reported. In the present work, we collected data from the existing studies on the effects of Alexandrium on feeding, reproduction and mortality of Acartia. With these data, we performed a systematic review consisting of a secondary analysis employing general or generalized linear models, weighting data from different studies by the reciprocal of their standard deviation. Our first aim was to overcome shortcomings of individual studies: limited ranges of the variables and overlooked variables (experiment length, population adaptation). These shortcomings could have led to inconsistent conclusions by missing heterogeneous patterns in copepod responses and in the interactions between variables. Our second aim was to test the enhanced physiological performance of chronically exposed relative to naïve copepod populations over a wide geographic range. We found that the feeding rate is enhanced by increased food biomass, irrespective of the food type. Toxins do not have a clear effect on egg production and have a bi-phasic effect on egg hatching success, which was negative above a specific threshold. Toxins also increased mortality. Experiment length had a positive effect on egg production and negative on egg hatching. Naïve copepod populations showed consistently lower ingestion of Alexandrium and egg hatching rates, thereby supporting the spread of the aforementioned mechanism across populations over a wide geographic range.

Microseira wollei, a globally distributed freshwater bloom-forming benthic cyanobacterium, is known for its production of cyanotoxins and taste and odor (T&O). While CYN (Cylindrospermopsin)-producing populations of M. wollei are confined to Australia, PST (Paralytic shellfish toxins)-producing populations have been exclusively documented in North America. In this study, four benthic cyanobacterial strains, isolated from West Lake in China, were identified as M. wollei based on morphological and phylogenetic analyses. Detection of sxtA gene and UPLC-MS/MS analysis conclusively confirmed the PST-producing capability of M. wollei CHAB5998. In the phylogenetic tree of 16S rDNA, M. wollei strains formed a monophyletic group with two subclades. Notably, non-PST-producing Chinese strains clustered with Australian strains in Clade II, while all other strains, including PST-producing ones, clustered in Clade I. Additionally, CHAB5998 contains ten PST variants, of which STX, NEO, GTX2, GTX3, GTX5 and C1 were identified for the first time in M. wollei. Sequence analysis of PST biosynthetic gene cluster (sxt) genes indicated potential base variations, gene rearrangements, insertions, and deletions in the strain CHAB5998. Also, sxt gene has a longer evolutionary history in M. wollei than that in cyanobacteria from Nostocales. Multiple recombination breakpoints detected in sxt genes and the inconsistency in the topology of the phylogenetic trees between sxt and 16S rDNA suggested that multiple horizontal gene transfers (HGT) have occurred. Overall, the present study marks the first documented occurrence of PST-producing M. wollei outside of North America and identifies it as the first toxic freshwater benthic cyanobacterium in China. This revelation implies that benthic cyanobacteria may pose a higher environmental risk in China than previously acknowledged.

OBJECTIVE: To systematically review the literature on the efficacy of addressing glenohumeral internal rotation deficit (GIRD) and risk of upper-extremity injury in overhead athletes. DATA SOURCES: A search was conducted for relevant studies published in PubMed, Medline, CINAHL, Cochrane, Embase, Ovid, Google Scholar, and Web of Science. STUDY SELECTION: The review focused on randomized controlled trials (RCTSs) and quasi-experiments conducted in English language that assessed the effectiveness of GIRD and the risk of upper-extremity injury in athletes performing overhead movements. The review included 7 RCTs and 2 quasi-experiments out of 5403, which involved a total of 360 participants. DATA EXTRACTION: Two reviewers independently screened the articles, assessed methodological quality, and extracted data for analysis. The review was conducted and reported according to the Preferred Reporting Items for Systematic Reviews. All studies were assessed in duplicate for risk of bias using the Physiotherapy Evidence Database Scale for RCTs. DATA SYNTHESIS: The efficacy of different types of techniques was evaluated, including joint mobilization, sleeper stretch, cross-body stretch, myofascial release, kinesio taping, and rigid taping. These techniques showed improvement in pain score and range of motion. Furthermore, self-myofascial release tends to improve internal rotation; sleeper stretch and cross-body stretch tend to improve internal rotation with 40 percent decline in pain. However, kinesio taping and rigid taping showed positive results for internal rotation. Acute results determined that the metabolic equivalent (MET) group had significantly more horizontal adduction range of motion posttreatment compared with the control group (P=.04). No significant differences existed between MET and joint mobilizations or between joint mobilizations and the control group for horizontal adduction (P>.16). No significant between-group differences existed acutely for internal rotation (P>.28). There were no significant between-group differences for either horizontal adduction or internal rotation at the 15-minute posttests (P>.70). CONCLUSIONS: The study evaluated the efficacy of various techniques in improving pain score and range of motion in individuals with GIRD. Joint mobilization, sleeper stretch, cross-body stretch, myofascial release, kinesio taping, and rigid taping all showed improvements in pain score and range of motion. However, no significant between-group differences were found for horizontal adduction or internal rotation at the 15-minute posttests. These findings suggest that a combination of these techniques may be effective in treating individuals with GIRD.

RESUMO

One of the major plant stress level indicators is reactive oxygen species (ROS). They have been known to play a central role in regulating plant responses to various environmental stresses. This book chapter specifically covers abiotic stress induced by a drought hormone abscisic acid and biotic stress induced by Pseudomonas syringe DC3000 on single cell-type guard cells. We describe in detail the measurement of ROS production starting from sample preparation to data analysis by fluorescence intensity acquisition using ImageJ software. We discussed the problems faced while performing the experiment and addressed how to overcome them by providing specific guidelines to ensure high quality repeatable data.

Stomatal immunity plays an important role during bacterial pathogen invasion. Abscisic acid (ABA) induces plants to close their stomata and halt pathogen invasion, but many bacterial pathogens secrete phytotoxin coronatine (COR) to antagonize ABA signaling and reopen the stomata to promote infection at early stage of invasion. However, the underlining mechanism is not clear. SAD2 is an importin ß family protein, and the sad2 mutant shows hypersensitivity to ABA. We discovered ABI1, which negatively regulated ABA signaling and reduced plant sensitivity to ABA, was accumulated in the plant nucleus after COR treatment. This event required SAD2 to import ABI1 to the plant nucleus. Abolition of SAD2 undermined ABI1 accumulation. Our study answers the long-standing question of how bacterial COR antagonizes ABA signaling and reopens plant stomata during pathogen invasion.

To reveal genetic diversity for effective resistance to five foliar diseases and toxic aluminum ions, the entire collection of wheat species from the N.I. Vavilov All-Russian Institute of Plant Genetic Resources (VIR) originating from Ethiopia and Eritrea were studied regarding their traits. The collection contains 509 samples of four wheat species (Triticum aestivum-122 samples; T. aethiopicum-340 samples; T. polonicum-6 samples; and T. dicoccum-41 samples). The majority of accessions are new entries of landraces added to the Vavilov collection as a result of the Russian-Ethiopian expedition in 2012. Wheat seedlings were inoculated with causal agents of leaf rust (Pt), powdery mildew (Bgt), Septoria nodorum blotch (SNB), and dark-brown leaf spot blotch (HLB). The types of reaction and disease development were assessed to describe the levels of resistance. All samples of T. aethiopicum were also screened for seedling and adult resistance to Pt, Bgt, and yellow rust (Pst) under field conditions after double inoculation with the corresponding pathogens. To study tolerance to abiotic stress, seedlings were grown in a solution of Al3+ (185 µM, pH 4,0) and in water. The index of root length was used to characterize tolerance. Seedlings belonging to only two accessions out of those studied-k-68236 of T. aethiopicum and k-67397 of T. dicoccum-were resistant to Pt at 20 °C but susceptible at 25 °C. Specific molecular markers closely linked to the five genes for Pt resistance effective against populations of the pathogen from the northwestern region of Russia were not amplified in these two entries after PCR with corresponding primers. Four entries of T. dicoccum-k-18971, k-18975, k-19577, and k-67398-were highly resistant to Bgt. All samples under study were susceptible to HLB and SNB. Under field conditions, 15% of the T. aethiopicum samples were resistant to Pst, both at the seedling and the flag leaf stages, but all were susceptible to the other diseases under study. Among the evaluated samples, 20 entries of T. aestivum, 1 of T. polonicum (k-43765), and 2 of T. dicoccum (k-18971, k-67397) were tolerant to aluminum ions. The identified entries could be valuable sources for the breeding of T. aestivum and other wheats for resistance to biotic and abiotic stresses.

Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) is able to infect many economically important crops and thus causes substantial losses in the global agricultural economy. Pst DC3000 can be divided into virulent lines and avirulent lines. For instance, the pathogen effector avrRPM1 of avirulent line Pst-avrRpm1 (Pst DC3000 avrRpm1) can be recognized and detoxified by the plant. To further compare the pathogenicity mechanisms of virulent and avirulent Pst DC3000, a comprehensive analysis of the acetylome and succinylome in Arabidopsis thaliana was conducted following infection with virulent line Pst DC3000 and avirulent line Pst-avrRpm1. In this study, a total of 1625 acetylated proteins encompassing 3423 distinct acetylation sites were successfully identified. Additionally, 229 succinylated proteins with 527 unique succinylation sites were detected. A comparison of these modification profiles between plants infected with Pst DC3000 and Pst-avrRpm1 revealed significant differences. Specifically, modification sites demonstrated inconsistencies, with a variance of up to 10% compared to the control group. Moreover, lysine acetylation (Kac) and lysine succinylation (Ksu) displayed distinct preferences in their modification patterns. Lysine acetylation is observed to exhibit a tendency towards up-regulation in Arabidopsis infected with Pst-avrRpm1. Conversely, the disparity in the number of Ksu up-regulated and down-regulated sites was not as pronounced. Motif enrichment analysis disclosed that acetylation modification sequences are relatively conserved, and regions rich in polar acidic/basic and non-polar hydrophobic amino acids are hotspots for acetylation modifications. Functional enrichment analysis indicated that the differentially modified proteins are primarily enriched in the photosynthesis pathway, particularly in relation to light-capturing proteins. In conclusion, this study provides an insightful profile of the lysine acetylome and succinylome in A. thaliana infected with virulent and avirulent lines of Pst DC3000. Our findings revealed the potential impact of these post-translational modifications (PTMs) on the physiological functions of the host plant during pathogen infection. This study offers valuable insights into the complex interactions between plant pathogens and their hosts, laying the groundwork for future research on disease resistance and pathogenesis mechanisms.

Temperature is one of the most prominent environmental factors that influence plant immunity. Depending on the plant-pathogen system, increased temperature may inhibit or enhance disease resistance or immunity in plants. Measuring the effect of temperature on plant immunity is the first step toward revealing climate effects on plant-pathogen interactions and molecular regulators of temperature sensitivity of plant immunity. Quantification of plant disease resistance or susceptibility under different temperatures can be accomplished by assessing pathogen growth over time in infected plants or tissues. Here, we present a protocol for quantifying pathogen growth in the most studied system of Arabidopsis thaliana and Pseudomonas syringae pathovar tomato (Pst) DC3000. We discuss important factors to consider for assaying pathogen growth in plants under different temperatures. This protocol can be used to assess temperature sensitivity of resistance in different plant genotypes and to various pathovars.

The S-locus lectin receptor kinases (G-LecRKs) have been suggested as receptors for microbe/damage-associated molecular patterns (MAMPs/DAMPs) and to be involved in the pathogen defense responses, but the functions of most G-LecRKs in biotic stress response have not been characterized. Here, we identified a member of this family, G-LecRK-I.2, that positively regulates flg22- and Pseudomonas syringae pv. tomato (Pst) DC3000-induced stomatal closure. G-LecRK-I.2 was rapidly phosphorylated under flg22 treatment and could interact with the FLS2/BAK1 complex. Two T-DNA insertion lines, glecrk-i.2-1 and glecrk-i.2-2, had lower levels of reactive oxygen species (ROS) and nitric oxide (NO) production in guard cells, as compared with the wild-type Col-0, under Pst DC3000 infection. Also, the immunity marker genes CBP60g and PR1 were induced at lower levels under Pst DC3000 hrcC- infection in glecrk-i.2-1 and glecrk-i.2-2. The GUS reporter system also revealed that G-LecRK-I.2 was expressed only in guard cells. We also found that G-LecRK-I.2 could interact H+-ATPase AHA1 to regulate H+-ATPase activity in the guard cells. Taken together, our results show that G-LecRK-I.2 plays an important role in regulating stomatal closure under flg22 and Pst DC3000 treatments and in ROS and NO signaling specifically in guard cells.

BACKGROUND: The sarcoplasmic reticulum (SR) Ca2+ ATPase (SERCA2a) depression substantially contributes to diastolic dysfunction in heart failure (HF), suggesting that SERCA2a stimulation may be a mechanism-based HF therapy. Istaroxime is a drug endowed with both a SERCA2a stimulatory activity and a Na+/K+ pump inhibitory activity for acute HF treatment. Its main metabolite PST3093 shows a more favorable therapeutic profile as compared to the parent drug, but it is still unsuitable for chronic usage. Novel PST3093 derivatives have been recently developed for oral (chronic) HF treatment; compound 8 was selected among them and here characterized. METHODS: Effects of compound 8 were evaluated in a context of SERCA2a depression, by using streptozotocin-treated rats, a well-known model of diastolic dysfunction. The impact of SERCA2a stimulation by compound 8 was assessed at the cellular level ad in vivo, following i.v. infusion (acute effects) or oral administration (chronic effects). RESULTS: As expected from SERCA2a stimulation, compound 8 induced SR Ca2+ compartmentalization in STZ myocytes. In-vivo echocardiographic analysis during i.v. infusion and after repeated oral administration of compound 8, detected a significant improvement of diastolic function. Moreover, compound 8 did not affect electrical activity of healthy guinea-pig myocytes, in line with the absence of off-target effects. Finally, compound 8 was well tolerated in mice with no evidence of acute toxicity. CONCLUSIONS: The pharmacological evaluation of compound 8 indicates that it may be a safe and selective drug for a mechanism-based treatment of chronic HF by restoring SERCA2a activity.

To understand environmental effects affecting paralytic shellfish toxin production of Centrodinium punctatum, this study examined the growth responses, and toxin contents and profiles of a C. punctatum culture exposed to drastic changes of temperature (5-30 °C) and salinity (15-40). C. punctatum grew over a temperature range of 15-25 °C, with an optimum of 20 °C., and over a salinity range of 25-40, with optimum salinities of 30-35. This suggests that C. punctatum prefers relatively warm waters and an oceanic habitat for its growth and can adapt to significant changes of salinity levels. When C. punctatum was cultivated at different temperature and salinity levels, the PST profile included four major analogs (STX, neoSTX, GTX1 and GTX4, constituted >80 % of the profile), while low amounts of doSTX and traces of dc-STX and dc-GTX2 were also observed. Interestingly, though overall toxin contents did not change significantly with temperature, increases in the proportion of STX, and decreases in proportions in GTX1 and GTX4 were observed with higher temperatures. Salinity did not affect either toxin contents or profile from 25 to 35. However, the total toxin content dropped to approximately half at salinity 40, suggesting this salinity may induce metabolic changes in C. punctatum.

BACKGROUND: Pod shell thickness (PST) is an important agronomic trait of peanut because it affects the ability of shells to resist pest infestations and pathogen attacks, while also influencing the peanut shelling process. However, very few studies have explored the genetic basis of PST. RESULTS: An F2 segregating population derived from a cross between the thick-shelled cultivar Yueyou 18 (YY18) and the thin-shelled cultivar Weihua 8 (WH8) was used to identify the quantitative trait loci (QTLs) for PST. On the basis of a bulked segregant analysis sequencing (BSA-seq), four QTLs were preliminarily mapped to chromosomes 3, 8, 13, and 18. Using the genome resequencing data of YY18 and WH8, 22 kompetitive allele-specific PCR (KASP) markers were designed for the genotyping of the F2 population. Two major QTLs (qPSTA08 and qPSTA18) were identified and finely mapped, with qPSTA08 detected on chromosome 8 (0.69-Mb physical genomic region) and qPSTA18 detected on chromosome 18 (0.15-Mb physical genomic region). Moreover, qPSTA08 and qPSTA18 explained 31.1-32.3% and 16.7-16.8% of the phenotypic variation, respectively. Fifteen genes were detected in the two candidate regions, including three genes with nonsynonymous mutations in the exon region. Two molecular markers (Tif2_A08_31713024 and Tif2_A18_7198124) that were developed for the two major QTL regions effectively distinguished between thick-shelled and thin-shelled materials. Subsequently, the two markers were validated in four F2:3 lines selected. CONCLUSIONS: The QTLs identified and molecular markers developed in this study may lay the foundation for breeding cultivars with a shell thickness suitable for mechanized peanut shelling.

Evaluating the stray light cancellation performance of an optical system is an essential step in the search for superior optical systems. However, the existing evaluation methods, such as the Monte Carlo method and the ray tracing method, suffer from the problems of vast arithmetic and cumbersome processes. In this paper, a method for a rapid stray light performance evaluation model and quantitatively determining high-magnitude stray light outside the field of view are proposed by adopting the radiative transfer theory based on the scattering property of the bidirectional scattering distribution function (BSDF). Under the global coordinates, based on the derivation of the light vector variation relationship in the near-linear system, the specific structural properties of the off-axis reflective optical system, and the specular scattering properties, a fast quantitative evaluation model of the optical system's stray light elimination capability is constructed. A loop nesting procedure was designed based on this model, and its validity was verified by an off-axis reflective optical system. It successfully fitted the point source transmittance (PST) curve in the range of specular radiation reception angles and quantitatively predicted the prominence due to incident stray light outside the field of view. This method does not require multiple software to work in concert and requires only 10-5 orders of magnitude of computing time, which is suitable for the rapid stray light assessment and structural screening of off-axis reflective optical systems with a good symmetry. The method is promising for improving imaging radiation accuracy and developing lightweight space cameras with low stray light effects.

By relating genetic divergence at neutral loci, phenotypic variation, and geographic and environmental distances, it is possible to dissect micro-evolutionary scenarios involving natural selection and neutral evolution. In this work, we tested the patterns of intraspecific genetic and phenotypic variation along an elevational gradient, using Dianthus virgineus as study system. We genotyped genome-wide SNPs through ddRAD sequencing and quantified phenotypic variation through multivariate morphological variation. We assessed patterns of variation by testing the statistical association between genetic, phenotypic, geographic, and elevational distances and explored the role of genetic drift and selection by comparing the Fst and Pst of morphometric traits. We revealed a weak genetic structure related to geographic distance among populations, but we excluded the predominant role of genetic drift acting on phenotypic traits. A high degree of phenotypic differentiation with respect to genetic divergence at neutral loci allowed us to hypothesize the effect of selection, putatively fuelled by changing conditions at different sites, on morphological traits. Thus, natural selection acting despite low genetic divergence at neutral loci can be hypothesized as a putative driver explaining the observed patterns of variation.

As the non-imaging light of optical instruments, stray light has an important impact on normal imaging and data quantification applications. The FY-3D Medium Resolution Spectral Imager (MERSI) operates in a sun-synchronous orbit, with a scanning field of view of 110° and a surface imaging width of more than 2300 km, which can complete two coverage observations of global targets per day with high detection efficiency. According to the characteristics of the operating orbit and large-angle scanning imaging of MERSI, a stray light radiation model of the polar-orbiting spectrometer is constructed, and the design requirements of stray light suppression are proposed. Using the point source transmittance (PST) as the merit function of the stray light analysis method, the instrument was simulated with all stray light suppression optical paths, and the effectiveness of stray light elimination measures was verified using the stray light test. In this paper, the full-link method of "orbital stray light radiation model-system, internal and external simulation design-system analysis and actual test comparison verification" is proposed, and there is a maximum decrease in the system's PST by about 10 times after applying the stray light suppression's optimization design, which can provide a general method for stray light suppression designs for polar-orbit spectral imagers.

Phage therapy, particularly for infections due to multi-drug-resistant bacteria, is increasingly gaining in importance. Consecutively, there is a rising need for phage testing in routine diagnostic laboratories. The incubation time of phage susceptibility testing for detecting lytic phage activity on phage/host strain combinations was evaluated. A standardized approach for routine diagnostic laboratories provided reliably detectable lysis zones within 8 hours.

This study presents the first evidence that a diverse suite of phycotoxins is not only being actively produced by the toxigenic algal communities in the Canadian Arctic waters, but is also entering the marine food web. We detected measurable amounts of Amnesic Shellfish Toxins (ASTs) and Paralytic Shellfish Toxins (PSTs), as well as trace amounts of other lipophilic toxin groups including pectenotoxins, yessotoxins, and cyclic imines, in bivalves collected from the Canadian Beaufort Sea in 2014 and 2018. There appear to be species-specific differences in accumulation and retention of AST by Arctic bivalves, with significantly higher concentrations recorded in Nuculanidae than Propeamussiidae, likely reflecting physiological and allometric differences. We further confirm the omnipresence of potentially toxic taxonomically-versatile phytoplankton communities in the western Canadian Arctic comprising Pseudo-nitzschia delicatissima group, P. obtusa, Dinophysis acuminata, Prorocentrum minimum, Alexandrium tamarense, and Gymnodinium spp. Although measurements of actual toxicity levels and profiles of these species at the time of sampling fall outside of the scope of this study, we show that high abundance and competitive success of known AST-producers, Pseudo-nitzschia spp., are possible in Canadian Arctic waters. In 2014, a strong dominance of Pseudo-nitzschia spp. was observed at a few shallow coastal stations, representing nearly 40% of the total phytoplankton cell abundances with > 106 cells/L at the depth of maximum chlorophyll a. We further describe oceanographic conditions conducive to high abundances of toxin-producing algae, indicating that temperature is likely a key factor. Even though measured AST and PST concentrations in bivalve tissue remained well below the Health Canada's levels at which monitored fisheries would close, i.e., 5% and 4%, respectively, their presence demonstrate that phycotoxin accumulation is occurring in food webs of the Canadian Beaufort Sea. Yet, the phycotoxin production controls and trophic transfer mechanisms remain unknown. Canadian Arctic marine ecosystems are rapidly changing and temperatures are expected to continue to increase. Given that these changes simultaneously affect multiple, and often co-occurring, species of primary producers, adaptive capacity is likely to play an important role in the structure of phytoplankton communities in the Canadian Arctic.
