RESUMO

Marine algae-based drug discovery has recently received a lot of attention. This study was conducted to extract laminarin-enriched solvent extracts from Padina tetrastromatica and Sargassum cinereum and to evaluate their anticancer activity against the HeLa cell line in vitro (MTT assay). Furthermore, their toxicity was determined through a zebra fish model study. P. tetrastromatica and S. cinereum biomasses have a higher concentration of essential biomolecules such as carbohydrates, protein, and crude fiber, as well as essential minerals (Na, Mg, K, Ca, and Fe) and secondary metabolites. Methanol extracts, in particular, contain a higher concentration of vital phytochemicals than other solvent extracts. The laminarin quantification assay states that methanol extracts of P. tetrastromatica and S. cinereum are rich in laminarin, which is primarily confirmed by FTIR analysis. In an anticancer study, laminarin-MeE from P. tetrastromatica and S. cinereum at concentrations of 750 and 1000 µg mL-1 demonstrated 100% activity against HeLa cells. The Zebra fish model-based toxicity study revealed that the laminarin-enriched MeE of P. tetrastromatica and S. cinereum is non-toxic. These findings revealed that the laminarin-enriched MeE of P. tetrastromatica and S. cinereum has significant anticancer activity without causing toxicity.

LC-MS-assisted metabolomic profiling of the Red Sea-derived brown algae Sargassum cinereum "Sargassaceae" dereplicated eleven compounds 1-11. Further phytochemical investigation afforded two new aryl cresol 12-13, along with eight known compounds 14-21. Both new metabolites, along with 19, showed moderate in vitro antiproliferative activity against HepG2, MCF-7, and Caco-2. Pharmacophore-based virtual screening suggested both 5-LOX and 15-LOX as the most probable target linked to their observed antiproliferative activity. The in vitro enzyme assays revealed 12 and 13 were able to inhibit 5-LOX more preferentially than 15-LOX, while 19 showed a convergent inhibitory activity toward both enzymes. Further in-depth in silico investigation revealed the molecular interactions inside both enzymes' active sites and explained the varying inhibitory activity for 12 and 13 toward 5-LOX and 15-LOX.

Seaweed extracts of Sargassum cinereum was used as a reducing agent in the eco-friendly extracellular synthesis of silver nanoparticles from an aqueous solution of silver nitrate (AgNO3). High conversion of silver ions to silver nanoparticles was achieved with a reaction temperature of 100(°) and a seaweed extract concentration of 10% with a residential time of 3 h. Formation of silver nanoparticles was characterised by spectrophotometry and the scanning electron microscope. The average particles size was ranging from 45 to 76 nm. Antimicrobial activities indicate the minimum inhibitory concentration of biologically synthesised nanoparticles tested against the pathogen Staphylococcus aureus with 2.5 µl (25 µg/disc). High inhibitions over the growth of Enterobacter aerogenes, Salmonella typhi and Proteus vulgaris were witnessed against the concentrations of 100 µg/disc. Promising potential and the future prospects of S. cinereum nanoparticles in pharmaceutical research are the highlights in this paper.
