RESUMO

Gram-negative, aerobic, motile by means of two or more polar or subpolar flagella, rod-shaped strain NS12-5T and Gram-negative, facultatively anaerobic, yellow-coloured, rod-shaped strain RP8T were isolated from rice rhizosphere soil and fermented fruits of Liriope platyphylla in the Republic of Korea, respectively. The result of phylogenetic analyses based on 16S rRNA gene sequences showed that strain NS12-5T was most closely related to Ideonella aquatica 4Y11T with 99.79 % sequence similarity. The average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between strain NS12-5T and species of the genus Ideonella were 75.6-91.7 % and 20.3-43.9 %, respectively. Growth occurred at 15-40 °C and pH 5-11, and NaCl was not needed for growth. The major fatty acids of strain NS12-5T were summed feature 3 (comprising C16 : 1 ω7c and/or C16 : 1 ω6c) and C16 : 0, and the major polar lipids were phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol. The DNA G+C content of strain NS12-5T was 69.03 mol%. The result of phylogenetic analyses based on 16S rRNA gene sequences revealed that strain RP8T was most closely related to Spirosoma aureum BT328T with 96.01 % sequence similarity. The ANI and dDDH values between strain RP8T and reference strains of the genus Spirosoma were 72.9-76.4 % and 18.6-20.0 %, respectively. Growth occurred at 15-37 °C and pH 5-11, and NaCl was not needed for growth. The major fatty acids of strain RP8T were summed feature 3 (comprising C16 : 1 ω7c and/or C16 : 1 ω6c), C16 : 1 ω5c and iso-C15 : 0. RESUMO

Two novel strains (HMF3257T and HMF4905T), isolated from freshwater and bark samples, were investigated to determine their relationships within and between species of the genus Spirosoma by using a polyphasic approach. They were aerobic, Gram-stain-negative, non-motile and rod-shaped bacteria. The major fatty acids (>10%) in both strains were identified as summed feature 3 (C16 : 1 ω7c and/or C16 : 1 ω6c) and C16 : 1 ω5c, while strains HMF3257T and HMF4905T contained a moderately high amount of C16 : 0 and iso-C15 : 0, respectively. The predominant respiratory quinone was MK-7 for both strains. In addition to phosphatidylethanolamine and one unidentified glycolipid, the polar lipid profile of strain HMF3257T consisted of three unidentified aminophospholipids, one unidentified aminolipid and two unidentified polar lipids, and that of strain HMF4905T consisted of one unidentified aminophospholipid, two unidentified aminolipids and three unidentified polar lipids. The DNA G+C contents of strains HMF3257T and HMF4905T were 47.2 and 46.4 mol%, respectively. Phylogenetic analysis based on 16S rRNA gene sequences showed that strains HMF3257T and HMF4905T are closely related to Spirosoma migulaei 15J9-8T (97.0 % sequence similarity), while sharing 97.4 % sequence similarity with each other. The average nucleotide identity value between strains HMF3257T and HMF4905T was 81.1 %, and the digital DNA-DNA hybridization value between these two strains was 24.4 %. Based on the above data, strains HMF3257T and HMF4905T represent two novel members within the genus Spirosoma, for which the names Spirosoma telluris sp. nov. and Spirosoma arboris sp. nov. are proposed, respectively. The type strain of S. telluris is HMF3257T (=KCTC 62463T=NBRC 112670T) and type strain of S. arboris is HMF4905T (=KCTC 72779T=NBRC 114270T).

RESUMO

A novel Gram-stain negative, aerobic, rod-shaped, asporogenous, propanil-degrading bacterial strain, TY50T, was isolated from a herbicide-contaminated soil in Nanjing, China. Strain TY50T was found to grow optimally at pH 9.0, 30 °C and in the absence of NaCl. The G + C content of the total DNA was determined to be 55.5 mol%. The 16S rRNA gene sequence of strain TY50T shows high identity to that of Spirosoma lacussanchae CPCC 100624T (99.3%), Spirosoma metallicum PR1014kT (94.8%) and Spirosoma soli MIMBbqt12T (94.6%). DNA-DNA hybridization indicated that the isolate had relatively low levels of DNA-DNA relatedness with S. lacussanchae CPCC 100624T (48.3%). Average nucleotide identity and the digital DNA-DNA hybridizations for draft genomes between strain TY50T and S. lacussanchae CPCC 100624T were 93.2% and 51.0%, respectively. The major cellular fatty acids of strain TY50T were identified as C16:1ω5c (24.5%) and summed feature 3 (C16:1ω6c/C16:1ω7c, 40.7%). MK-7 was found to be the predominant respiratory quinone. The major polar lipid profile includes phosphatidylethanolamine, an unidentified lipid and an unidentified aminolipid. These chemotaxonomic data support the affiliation of strain TY50T with the members of the genus Spirosoma. Strain TY50T can be distinguished from its close phylogenetic neighbours based on its phenotypic, genotypic, and chemotaxonomic features. Therefore, strain TY50T represents a novel member of the genus Spirosoma, for which the name Spirosoma sordidisoli sp. nov. is proposed. The type strain is TY50T (= KCTC 62494T = CCTCC AB 2018041T).

RESUMO

Between 2014 and 2016, 16 Gram-stain-negative, aerobic, rod-shaped and yellow-orange pigmented bacteria were isolated from exposed soils from the Utsteinen region, Sør Rondane Mountains, East Antarctica. Analysis of their 16S rRNA gene sequences revealed that the strains form a separate cluster in the genus Spirosoma, with Spirosoma rigui KCTC 12531T as its closest neighbour (97.8 % sequence similarity). Comparative genome analysis of two representative strains (i.e. R-68523T and R-68079) of the new group with the type strains of Spirosoma rigui (its closest neighbour) and Spirosoma linguale (type species of the genus), yielded average nucleotide identity values of 73.9-78.7 %. Digital DNA-DNA reassociation values of the two strains and these type strains ranged from 20.3 to 22.0 %. The predominant cellular fatty acids of the two novel strains were summed feature 3 (i.e. C16 : 1 ω7c and/or iso-C15 2-OH), C16 : 1 ω5c, C16 : 0 and iso-C15 : 0. The new Spirosoma strains grew with 0-0.5 % (w/v) NaCl, at pH 6.5-8.0 and displayed optimum growth between 15 and 25 °C. Based on the results of phenotypic, genomic, phylogenetic and chemotaxonomic analyses, the new strains represent a novel species of the genus Spirosoma for which the name Spirosoma utsteinense sp. nov. is proposed. The type strain is R-68523T (=LMG 31447T=CECT 9925T).

RESUMO

A Gram-negative bacterium, strain HA7T, was isolated from the microhabitat of common hazel (Corylus avellana L.) pollen. HA7T was found to be an aerobic, rod-shaped, catalase-positive, oxidase-negative bacterium with an optimum growth temperature of 25 °C and pH of 7. The nearly complete 16S rRNA gene sequence of HA7T strain showed the closest similarities to Spirosoma linguale DSM 74T (97.4 %) and Spirosoma fluviale DSM 29961T (97.43 %). The major fatty acids (>5 %) were C16 : 1ω5c, summed feature 3 (C16 : 1ω7c and/or iso-C15 : 0 2-OH), iso-C15 : 0 and iso-C17 : 0 3-OH. The major polar lipids were an unidentified aminophospholipid and phosphatidylethanolamine. The major respiratory quinone detected was menaquinone MK-7 (95 %). The draft genome sequence included 8 794 837 bases, which contained 3665 predicted coding sequences and had a G+C content of 47.9 mol%. The genome-based comparison between HA7T and S. linguale DSM 74T and S. fluviale DSM 29961T with pairwise average nucleotide identity indicated a clear distinction, between 76.2-76.3 %. Moreover, the digital DNA-DNA relatedness of HA7T to these strains was 26.5 and 25.1 %. Based on the differential genotypic, phenotypic and chemotaxonomic properties to closely related type strains, strain HA7T ought to be assigned with the status of a new species, for which the name Spirosomapollinicola sp. nov. is proposed. The type strain is HA7T (DSM 105799T=LMG 30282T).

A Gram-negative, motile by gliding, rod-shaped, aerobic bacterium, designated S7-3-19T, was isolated from apple orchard soil in Gyeongsangnam-do Province, Republic of Korea, and characterized taxonomically by using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequencing showed that strain S7-3-19T belonged to the family Cytophagaceae and was most closely related to Spirosoma linguale DSM 74T (96.38 %), Spirosoma fluviale MSd3T (96.38 %), Spirosoma pulveris JSH5-14T (96.35 %) and Spirosoma radiotolerans DG5AT (96.24 %). Chemotaxonomic characteristics supported the classification of strain S7-3-19T within the genus Spirosoma. Summed feature 3 (C16 : 1ω7c/C16 : 1ω6c; 46.7 %) and C16 : 1ω5c (23.8 %) were the major fatty acids. Phosphatidylethanolamine, an unidentified aminophospholipid, an unidentified phospholipid and two unidentified lipids were the major polar lipids. Menaquinone with seven isoprene units was the predominant respiratory quinone. The G+C content of the genomic DNA of strain S7-3-19T was 48.6 mol%. On the basis of its phenotypic properties, genotypic distinctiveness and chemotaxonomic features, strain S7-3-19T represents a novel species of the genus Spirosoma, for which the name Spirosomahorti sp. nov. is proposed. The type strain is S7-3-19T (=KCTC 52728T=JCM 32131T).

RESUMO

A Gram-negative, non-motile, rod-shaped, aerobic bacterial strain, designated S2-3-6T, was isolated from soil in Jeollabuk-do province, South Korea, and was characterized taxonomically using a polyphasic approach. According to a comparative 16S rRNA gene sequence analysis, strain S2-3-6T belonged to the family Cytophagaceae and was most closely related to Spirosoma endophyticum EX36T (98.2%), Spirosoma fluviale MSd3T (96.0%), and Spirosoma linguale DSM 74T (95.7%). Strain S2-3-6T contained summed feature 3 (C16:1 ω7c/C16:1 ω6c), C16:1 ω5c, and C16:0 N alcohol as major cellular fatty acids, MK-7 as the predominant respiratory quinone, and phosphatidylethanolamine as the main polar lipid. The DNA G+C content of strain S2-3-6T was 47.8%. Phenotypic and chemotaxonomic data supported the affiliation of strain S2-3-6T to the genus Spirosoma. However, the DNA-DNA relatedness between strain S2-3-6T and Spirosoma endophyticum KACC 17920T was 27%, clearly showing that the isolate constitutes a new genospecies. Strain S2-3-6T could be clearly differentiated from its closest neighbors based on its phenotypic, genotypic, and chemotaxonomic features. Therefore, strain S2-3-6T represents a novel species of the genus Spirosoma, for which the name Spirosoma jeollabukense sp. nov. is proposed. The type strain is S2-3-6T (= KCTC 52725T = JCM 32129T).

RESUMO

A Gram-stain-negative, non-motile, non-spore-forming, rod-shaped, aerobic bacterium, designated 15J8-9T, was isolated from a sandy beach in Jeju Island, South Korea. The isolate was able to grow between 10 and 30 °C, pH 5-8, and in presence of 0-1% (w/v) NaCl. Based on 16S rRNA gene phylogenetic analysis, the novel strain was closely related to members of the genus Spirosoma (96.1-90.9% similarities) and showed highest sequence similarity to Spirosoma panaciterrae DSM 21099T (96.1%). The G + C content of the genomic DNA of strain 15J8-9T was 45.1 mol%. The isolate contained menaquinone MK-7 as the predominant respiratory quinone, phosphatidylethanolamine as the major polar lipid, and summed feature 3 (C16:1 ω6c/C16:1 ω7c; 28.0%), C16:1 ω5c (23.4%), iso-C15:0 (13.5%), and C16:0 (11.5%) as the major fatty acids that supported the affiliation of strain 15J8-9T to the genus Spirosoma. The isolate could be differentiated clearly from recognized Spirosoma species on the basis of several phenotypic, genotypic and chemotaxonomic features. Therefore, strain 15J8-9T is considered to represent a novel species of the genus the genus Spirosoma, for which the name Spirosoma harenae sp. nov. is proposed. The type strain is 15J8-9T (= KCTC 52030T = JCM 31993T).
