RESUMO

This study examined the impact of sertraline, an antidepressant common in treated wastewater, on the host-parasite dynamics between parasitic freshwater mussel (Unio tumidus, Unionidae) larvae (glochidia) and their host fish (Squalius cephalus, Cyprinidae). Employing a full-factorial design, both fish and glochidia were subjected to sertraline at the combinations of 0 µg L-1 (control), 0.2 µg L-1 (environmentally relevant concentration), and 4 µg L-1 (elevated concentration, short-term exposure of the parasite). The results showed that long-term host exposure (involving intensive sertraline accumulation in the fish brain) marginally increased subsequent glochidia attachment success by 2 %, while parasite exposure at the same environmentally relevant concentrations had no detectable effect. There was also no effect of exposure of glochidia to 0.2 µg L-1 of sertraline on their viability and encapsulation success during the initial parasitic stage. However, a significant alteration in attachment behavior, marked by a 3.3 % increase in attachment success and changes in the glochidia spatial distribution on the host body, was noted after 24 h of glochidia exposure to 4 µg L-1 of sertraline. Importantly, this study provides the first evidence of sertraline transfer from exposed glochidia to nonexposed host fish, as indicated by elevated levels of sertraline (12.8 ng g-1) in the brain tissue of nonexposed hosts. These findings highlight the subtle yet significant effects of pharmaceutical pollutants on freshwater ecosystems but also underscore the importance of understanding the unexpected dynamics of such contamination to predict and address future ecological changes.

Acanthocephalans' position in food webs, in close interaction with free-living species, could provide valuable information about freshwater ecosystem health through the viability of the parasites' host populations. We explored Pomphorhynchus laevis cystacanths' and adults' intensities of infection, and the prevalence of infected hosts respectively in their Gammarus pulex intermediate hosts and Squalius cephalus definitive hosts in a Mediterranean river. First, we analysed the relationship between P. laevis intensity of infection, its two hosts populations and the other acanthocephalan species found (Pomphorhynchus tereticollis and Polymorphus minutus). Second, we characterised the influence of bacteriological, physicochemical and biological water parameters on these acanthocephalans, and their intermediate and definitive hosts. This research highlights that P. laevis infection was closely related to their two preferential hosts population in the river. Moreover, P. laevis intensity of infection was positively correlated with organic pollution in the river but negatively correlated with biodiversity and with ecological indexes of quality. Pomphorhynchus laevis could thus benefit from moderate freshwater pollution, which promotes their tolerant intermediate and definitive hosts.

Eutrophication seriously threatens the ecological quality and biodiversity of running waters. In nutrient-enriched streams and shallow rivers, eutrophication leads to excessive periphyton growth and, in turn, biological clogging, oxygen depletion in the hyporheic zone and finally a reduction in the hyporheic habitat quality. Top-down control of the food-web by manipulating fish stocks, similar to the biomanipulation successfully applied in lakes, offers a promising approach to mitigating the effects of eutrophication in shallow rivers, especially those in which major reductions in nutrient input are not feasible. We conducted a reach-scale experiment over 4 years in a medium-sized eutrophic river to assess whether the top-down effects of two important large European cypriniform fish species, herbivorous common nase (Chondrostoma nasus) and omnivorous European chub (Squalius cephalus), would mitigate the effects of eutrophication. The enhancement of fish stocks was expected to reduce biological clogging, via the top-down control of periphyton by benthic grazing and enhanced bioturbation, thus increasing oxygen availability in the hyporheic zone as well as water exchange between the surface water and the hyporheic zone. As expected, enhancing the stocks of nase and chub increased both oxygen availability and vertical exchange flux of water in the upper layer of the hyporheic zone. However, periphyton biomass (chlorophyll a) was significantly reduced only in deeper pool habitat. Thus, while experimental biomanipulation in a shallow river significantly mitigated the effects of eutrophication in the hyporheic zone, top-down effects on periphyton biomass were rather small. Overall, to our knowledge, our results provide first evidence that the biomanipulation achieved by enhancing herbivorous and omnivorous fish stocks has the potential to mitigate the effects of eutrophication in medium-sized European rivers.

The metacercariae of Clinostomum species which known as yellow grubs have zoonotic potential by infecting humans. In the present study, a total of 403 freshwater fish specimens belonging to different genera from Central Anatolia Region of Turkey were examined for yellow grub metacercariae infections. Only three specimens belonging to Squalius cephalus were found to be infected with metacercariae with a prevalence on this host species of 2.4% and an overall prevalence of 0.7%. All the metacercariae were morphologically identified as Clinostomum complanatum. Partial fragments of mitochondrial cytochrome oxidase I (mt-COI) gene and internal transcribed spacer 2 (ITS-2) were amplified for sequence and phylogenetic analyses. The sequence analyses of ITS-2 and mt-COI revealed three and nine polymorphic sites leading to detection of four and five haplotypes within the related gene regions, respectively. Moreover, the intraspecific genetic distances for C. complanatum isolates ranged from 0.0 to 0.7% for ITS-2 and 0.0 to 1.4% for mt-COI data sets. Consequently, the present study has provided first combined morphologic and molecular data on C. complanatum infecting Turkish freshwater fishes.

This research aimed to find out the levels of Iron (Fe), Zinc (Zn), and Cupper (Cu) in the flesh of Chub (Squalius cephalus) and in the Karakaya Dam Lake water in which they live. The findings were evaluated in terms of the risk of consuming of this fish as human food. In addition, the changes of the heavy metal level of fish with weight, length and sex of them. The accumulation factor for each metal was also found. The level of heavy metals in the flesh of fish were found higher than that in the water. The level of heavy metals changed with the weight, length and sex of fish. The results were compared with acceptable values for heavy metals given by FAO/WHO to detect whether the flesh of chub has any risk for consumption as human food.

Assessing the impacts of invasive organisms is a major challenge in ecology. Some widespread invasive species such as crayfish are potential competitors and reciprocal predators of ecologically and recreationally important native fish species. Here, we examine the effects of signal crayfish (Pacifastacus leniusculus) on the growth, diet, and trophic position of the chub (Squalius cephalus) in four rivers in Britain. Growth rates of 0+ chub were typically lower in sympatric populations with signal crayfish compared with allopatric populations, and this effect could be traced through to 2+ chub in one river. However, growth rates of older chub (5+ to 6+) were typically higher in the presence of crayfish. Sympatry with crayfish resulted in lower chub length-at-age and mass-at-age in half of the rivers sampled, with no change detected in the other rivers. Stable isotope analyses (δ13C and δ15N) revealed that both chub and crayfish were omnivorous, feeding at multiple trophic levels and occupying similar trophic positions. We found some evidence that chub trophic position was greater at invaded sites on one river, with no difference detected on a second river. Mixing models suggested crayfish were important food items for both small and large chub at invaded sites. This study provides evidence that invasive species can have both positive and negative effects on different life stages of a native species, with the net impact likely to depend on responses at the population level.

Predicting how elevated temperatures from climate change alter host-parasite interactions requires understandings of how warming affects host susceptibility and parasite virulence. Here, the effect of elevated water temperature and parasite exposure level was tested on parasite prevalence, abundance and burden, and on fish growth, using Pomphorhynchus laevis and its fish host Squalius cephalus. At 60 days post-exposure, prevalence was higher at the elevated temperature (22 °C) than ambient temperature (18 °C), with infections achieved at considerably lower levels of exposure. Whilst parasite number was significantly higher in infected fish at 22 °C, both mean parasite weight and parasite burden was significantly higher at 18 °C. There were, however, no significant relationships between fish growth rate and temperature, parasite exposure, and the infection parameters. Thus, whilst elevated temperature significantly influenced parasite infection rates, it also impacted parasite development rates, suggesting warming could have complex implications for parasite dynamics and host resistance.

Diet-tissue discrimination factors (Δ(13)C and Δ(15)N) are influenced by variables including the tissues being analysed and the taxon of the consumer and its prey. Whilst differences in Δ(13)C and Δ(15)N are apparent between herbivorous and piscivorous fishes, there is less known for omnivorous fishes that consume plant and animal material. Here, the omnivorous cyprinid fishes Barbus barbus and Squalius cephalus were held in tank aquaria and exposed to three diets that varied in their constituents (plant based to fishmeal based) and protein content (13% to 45%). After 100 days and isotopic replacement in fish tissues to 98%, samples of the food items, and dorsal muscle, fin tissue and scales were analysed for δ(13)C and δ(15)N. For both species and all diets, muscle was always enriched in δ(15)N and depleted in δ(13)C compared with fin tissue and scales. Across the different diets, Δ(13)C ranged between 2.0‰ and 5.6‰ and Δ(15)N ranged between 2.0‰ and 6.9‰. The diet based on plant material (20% protein) always resulted in the highest discrimination factors for each tissue, whilst the diet based on fishmeal (45% protein) consistently resulted in the lowest. The discrimination factors produced by non-fish diets were comparatively high compared with values in the literature, but were consistent with general patterns for some herbivorous fishes. These outputs suggest that the diet-tissue discrimination factors of omnivorous fishes will vary considerably between animal and plant prey, and these specific differences need consideration in predictions of their diet composition and trophic position.

The Rhine catchment in Switzerland has been transformed by a chain of hydroelectric power stations. We addressed the impact of fragmentation on the genetic structure of fish populations by focusing on the European chub (Squalius cephalus). This fish species is not stocked and copes well with altered habitats, enabling an assessment of the effects of fragmentation per se. Using microsatellites, we genotyped 2133 chub from 47 sites within the catchment fragmented by 37 hydroelectric power stations, two weirs and the Rhine Falls. The shallow genetic population structure reflected drainage topology and was affected significantly by barriers to migration. The effect of power stations equipped with fishpasses on genetic differentiation was detectable, albeit weaker than that of man-made barriers without fishpasses. The Rhine Falls as the only long-standing natural obstacle (formed 14 000 to 17 000 years ago) also had a strong effect. Man-made barriers also exacerbated the upstream decrease in allelic diversity in the catchment, particularly when lacking fishpasses. Thus, existing fishpasses do have the desired effect of mitigating fragmentation, but barriers still reduce population connectivity in a fish that traverses fishpasses better than many other species. Less mobile species are likely to be affected more severely.

Here we report the genetic variability and presence of mtDNA-like sequences of Pomphorhynchus laevis from the chub, Squalius cephalus, caught at the sampling sites along the Sava River and its tributary the Sutla River in Croatia. Sequences of the cytochrome c oxidase subunit 1 (COI) gene of the recovered P. laevis specimens were used for haplotype network construction and phylogenetic analysis. These analyses showed that some specimens contained mitochondrial-like sequences, and they uncovered the existence of a Sava River basin strain different from known strains of P. laevis. This is the first time that P. laevis has been shown to contain mtDNA-like sequences, suggesting the need to exercise caution during COI analyses of P. laevis using universal primers. Highly conserved sequences of two nuclear markers, the ITS region and 18S rRNA, were not helpful for understanding genetic variability or differentiating strains. Furthermore, analysis of the dynamics of P. laevis infections in S. cephalus from the Sava and Sutla Rivers showed decreased prevalence and abundance at sites with inferior water quality, positive association of parasite abundance with fish size, and no clear association of parasite abundance with fish condition index or sex.

In the present study, the importance of considering fish intestinal parasites i.e. the acanthocephalans in metal exposure assessment was estimated under low metal contamination conditions. Two acanthocephalan species, Pomphorhynchus laevis and Acanthocephalus anguillae were examined in 59 specimens of European chub (Squalius cephalus L.) sampled at 5 locations along the Sava River, Croatia. Concentrations of essential (Cu, Mn) and non-essential (Ag, Cd, Pb) metals were higher in intestinal parasites than chub gastrointestinal tissue, but levels of essential metals Fe and Zn were comparable or lower in parasites, respectively. The highest accumulation in both acanthocephalan species was found for non-essential metals and followed the order: Ag>Pb>Cd. Higher infection intensity with P. laevis allowed us to present their spatial metal distribution and evaluate the influence of P. laevis on metal levels and sub-cellular biological responses (total protein and metallothionein levels) in the host infected with P. laevis. Even in the river section with low metal contamination, parasitism affected metal levels, resulting in lower Cu, Cd and Pb concentrations in chub infected with P. laevis than in uninfected chub. Although total protein and metallothionein levels remained constant in infected and uninfected chub, acanthocephalans should be considered a potential confounding factor in metal exposure assessments. Moreover, P. laevis-chub system can be suggested as an appropriate tool in biomonitoring, since in both species increased Cu and Cd concentrations towards the downstream locations were found. Higher Cu and Cd levels in P. laevis suggest acanthocephalans to be sensitive bioindicators if low metal levels have to be detected.
