RESUMO

Previously, we found that 5-(2,6-dimethoxybenzoylamino)-3-phenylisoxazoles (IOXs) inhibit chitin synthesis in the cultured integument of Chilo suppressalis. In this study, IOXs with various substituents at the para-position of the 3-phenyl ring were synthesized, and the concentrations required to inhibit chitin synthesis to 50% (IC50) were determined for all compounds. The introduction of halogens-such as F, Cl, and Br-and small alkyls-such as Me, Et, Pr, and n-Bu-at the 3-phenyl ring slightly enhanced the activity. However, the activity decreased drastically with the introduction of NO2, CF3, and t-Bu. The quantitative analysis of the substituent effect at the 3-phenyl ring on chitin-synthesis inhibition using the Hansch-Fujita method showed that the hydrophobic substituent with the optimum value was favored for the activity, but the bulky substituent in terms of E s was detrimental to the activity.

BACKGROUND: To control subterranean termite pests, chitin synthesis inhibitor (CSI) baits have been widely applied. Despite CSI baits having low impacts on the environment, they require a lengthy time period to eliminate colonies. 20-hydroxyecdysone (20E) was proposed to speed up the baiting process as it showed faster mortality than CSI baits. However, the efficacy of 20E has previously not been tested at the colony level prior to applying in the field. RESULTS: We compared the effect of 20E, 20E + noviflumuron, noviflumuron and untreated control using colonies of Coptotermes formosanus. Our result revealed that both 20E and 20E + noviflumuron did not accelerate colony elimination and termite activity remained relatively stable during the observation periods. To determine the limited effects of 20E, we further investigated feeding duration and consumption amount of 20E with different concentrations (control, 100 and 1000 ppm) for 10 days. Termites ceased feeding after 1 day in 100 and 1000 ppm treatment and 100% mortality was observed within 10 days in 1000 ppm 20E, while mortality in the 100 ppm 20E treated group was much lower than that in the 1000 ppm group. Furthermore, no termites molted in the control and termites died from hyperecdysonism in 1000 ppm 20E treatment, whereas about 20% of termites molted in 100 ppm 20E. CONCLUSION: This study demonstrated that 20E may not be suitable as a sole active ingredient to accelerate elimination of a subterranean termite colony, while CSI baits and lower concentrations of 20E may reduce the lengthy time period in colony elimination. © 2023 Society of Chemical Industry.

Cockroaches, widespread pests found in metropolitan areas, are known as vectors of various disease agents, including viruses, fungi and antibiotic-resistant bacteria, as well as causing allergies in humans. Insect growth regulators have been used in pest management for several decades. These insecticides disrupt insect development and reproduction. Chitin synthesis inhibitors interfere with chitin biosynthesis in insects, causing abortive moulting and mortality, as well as inhibiting egg fertility, and larval hatching in insects. In this research, we evaluated the various effects of diflubenzuron, a chitin synthesis inhibitor, on synthetic pyrethroid-resistant German cockroach (Blattella germanica L. Blattodea: Ectobiidae), including ootheca production, oothecal viability, ootheca incubation time, the number of nymphs emerging from the ootheca and survivorship of nymphs. The cockroaches were fed diets that contained diflubenzuron, which was added to solid bait (impregnated fish food) and ingestible aqueous bait (impregnated cotton). Three concentrations (0.5%, 1% and 2%) were used in the experiments. As a result, diflubenzuron treatment led to ootheca production ranging from 60% to 100%; statistically, no difference was found between the treatment and the control groups. The number of nymphs emerging from the first and second ootheca was reduced by 40%-100% in the diflubenzuron-treated groups compared with the control. Nymphs exposed to diflubenzuron-impregnated solid bait and ingestible aqueous bait experienced mortality exceeding 92.1% and 66.27% within 15 days, respectively. In conclusion, diflubenzuron is a potential insecticide for use in cockroach baits to control B. germanica, as it caused high nymphal and embryonic mortality in the synthetic pyrethroid-resistant population and decreased the number of nymphs emerging from the ootheca.

The traditional stake survey and in-ground (IG) monitoring stations have been ineffective in aggregating the Asian subterranean termite, Coptotermes gestroi (Wasmann) in southeastern Florida. In this study, we used both IG and above-ground (AG) Sentricon stations to monitor and bait C. gestroi, and as expected, none of the 83 IG stations was intercepted. Despite this, AG bait stations with 0.5% noviflumuron were successfully used to eliminate C. gestroi colonies. From 2 field experiments, the mean colony elimination time (±SD) using AG baits were 6.4 ± 3.8 wk (n = 4) and 8.0 ± 2.1 wk (n = 12), respectively. Such results were compatible with baiting studies against field colonies of C. gestroi elsewhere, that is, 4-9 wk. The successful rates in monitoring and baiting of C. gestroi with IG stations in other regions also varied, which may be due to the variabilities in tunnel geometry of this species in different environments. In areas with established C. gestroi populations, routine inspection for signs of activity in structures and surrounding trees can be a critical component for pest control providers for early detection of infestation and colony elimination with AG bait stations.

BACKGROUND: High-levels of etoxazole resistance have not yet been frequently reported in Panonychus citri. Although a highly resistant strain was discovered in 2014, etoxazole resistance has not become a significant problem in areas of citrus production in Japan. A target site mutation in chitin synthase 1 (CHS1), I1017F, is a major etoxazole-resistance factor in Tetranychus urticae. To investigate the mechanisms of etoxazole resistance and the dispersal of resistance genes, we analyzed target-site mutations in a highly resistant strain and their geographical distribution in Japan. RESULTS: High-level etoxazole resistance was completely recessive. The I1017F mutation was detected in CHS1 of the highly resistant strain, and its frequency was correlated with the hatchability of eggs treated with etoxazole. Sequencing and variant frequency analyses of local populations by quantitative polymerase chain reaction revealed that I1017F is restricted to the Ariake Sea area of Kyushu Island. Although a new nonsynonymous substitution, S1016L, accompanied by I1017F was found in CHS1 of the highly resistant strain, CRISPR/Cas9 engineering of flies showed that S1016L had no effect on the etoxazole resistance conferred by I1017F. CONCLUSION: I1017F is a major target site mutation that confers high-level etoxazole resistance on P. citri. Dispersion of I1017F possibly was suppressed as a result of the completely recessive inheritance of resistance together with low gene flow between local populations. © 2022 Society of Chemical Industry.

The efficacy of three groups of insect growth regulators, namely juvenile hormone mimics (methoprene and pyriproxyfen), chitin synthesis inhibitors (diflubenzuron and novaluron), and molting disruptor (cyromazine) was evaluated for the first time, against Aedes albopictus Skuse (Diptera: Culicidae) larvae from 14 districts in Sabah, Malaysia. The results showed that all field populations of Ae. albopictus were susceptible towards methoprene, pyriproxyfen, diflubenzuron, novaluron, and cyromazine, with resistance ratio values ranging from 0.50-0.90, 0.60-1.00, 0.67-1.17, 0.71-1.29, and 0.74-1.07, respectively. Overall, the efficacy assessment of insect growth regulators in this study showed promising outcomes and they could be further explored as an alternative to conventional insecticides.

Chitin synthesis inhibitors (CSI) are supposed to inhibit formation of chitin microfibrils in newly synthesized cuticle during molting process. Conversely, there has been comparatively few data on morphological effects of CSI on non-target insect organs. In this work, the effects of the CSI novaluron on behavior and midgut of A. aegypti were evaluated. Toxicity bioassays revealed that novaluron is toxic to A. aegypti larva with LC50 = 18.57 mg L-1 when exposed in aqueous solution for 24 h. Novaluron treated larvae were less active and spent more time resting compared to the control group. Histopathology showed that midguts of novaluron-treated larvae had cytoplasm vacuolization and damaged brush border. Cytotoxic effects in midguts of treated larvae induced necrosis, autophagy and damage to mitochondria. Despite being chitin synthesis inhibitor, novaluron did not induce alterations in the integument of A. aegypti larvae. Fluorescence microscopy revealed that the number of digestive cells were higher in novaluron-treated larvae than in control, in response to digestive cell apoptosis. The present study highlights the importance of novaluron against A. aegypti larvae by causing injuries to non-target organs, altering behaviors, inducing cell death and inhibiting cell proliferation.

The environmental pollution, evolution of resistance, and risks to human and aquatic animal health associated with pesticide application have attracted much attention globally. Herein, we tested the capacity of diallyl trisulfide (DAT) from garlic essential oil to control the destructive stored-product pest, Sitotroga cerealella. The effects of DAT on the total content of cuticular chitin and structure of adults S. cerealella were evaluated. This study was the first to investigate changes in chitin structure in adults due to exposure to DAT through Fourier-transform infrared spectroscopy, thermogravimetric analysis, X-ray diffraction, and differential scanning calorimetry. The results of these analyses revealed that the cuticular chitin content of pests decreased after DAT treatment. DAT treatment also reduced thermal stability and crystallinity of chitin. These findings indicate that DAT is a potent biopesticide that is active against the moth, and establishes the basis for its use as an IPM and alternative to chitin synthesis inhibitors.

BACKGROUND: Insect growth regulators disrupt insect development and reproduction. Chitin synthesis inhibitors (CSIs) allow the insect to grow normally, but because chitin is an essential component of the cuticle, formation of a new cuticle and ecdysis are prevented and the insect dies. CSIs can also kill embryos by disrupting their normal development. We evaluated the potential utility of novaluron in bait formulations against the German cockroach (Blattella germanica L.). RESULTS: The minimum novaluron intake that interfered with molting and reproduction was assessed by exposing nymphs and adult females to novaluron. Results showed that 1 day of feeding on 0.1% novaluron was sufficient to disrupt molting in nymphs and prevent adult females from developing viable oothecae. The long-term effects on gravid females were investigated by feeding females 0.1% novaluron for different 5-day intervals during successive stages of gestation. Results demonstrated that gravid females fed novaluron during any period of gestation were able to produce viable eggs. To determine if ingestion of novaluron affected mating success and fertility of adult males, males were fed novaluron and then allowed to mate with untreated virgin females. Males that fed on novaluron successfully mated, and the females produced viable oothecae. Finally, direct comparisons revealed that novaluron is equally effective by ingestion and topical application. CONCLUSIONS: Novaluron caused mortality in nymphs and interfered with ootheca production in adult females, but only before they formed an ootheca. It successfully reduced German cockroach populations in cages and has potential to be incorporated in cockroach baits.

The residual efficacy of novaluron on concrete, metal and wood was evaluated against last-instar larvae of Oryzaephilus surinamensis (L.), Tribolium castaneum (Herbst), and Trogoderma granarium Everts. The larvae and food provided for survival were exposed to surfaces pretreated at rates of 0.053, to 0.209 mg/m2 and bioassays were conducted from 0- to 16-weeks post-treatment. Percentage emergence of adults was recorded after 30 days (d). On all surfaces at week 0, no O. surinamensis or T. castaneum adults emerged, and ≤3.3% emergence of T. granarium was found at 0.209 mg/m2. Novaluron significantly reduced the percentage emergence of adults of the three species compared to a distilled water control for the first 12 weeks on all the tested surfaces as the residual efficacy declined at a low rate during initial weeks and then at a high rate in the final weeks of the 16-week study. Reductions to emergence were most persistent on metal surfaces, with mean percentages of adult emergence of ≤18.3 in week 12, followed by concrete (≤32.5) and wood (≤45.0) for all species at novaluron application rates of 0.209 mg/m2. For >4-weeks protection, higher application rates would be needed to avoid buildup of pest populations and reductions in profitability. Such results can be helpful for the management of O. surinamensis, T. castaneum, and T. granarium as the costs of commonly used insecticides against postharvest insect pests and the resistance of these insects to the pesticides gradually increase in mills, warehouses, and food storage facilities.

Disruption in chitin regulation by using chitin synthesis inhibitor (novaluron) was investigated to gain insights into the biological activity of chitinase in red palm weevils, an invasive pest of date palms in the Middle East. Impact of novaluron against ninth instar red palm weevil larvae was examined by dose-mortality response bioassays, nutritional indices, and expression patterns of chitinase genes characterized in this study. Laboratory bioassays revealed dose-dependent mortality response of ninth-instar red palm weevil larvae with LD50 of 14.77 ppm of novaluron. Dietary growth analysis performed using different doses of novaluron (30, 25, 20, 15, 10, and 5 ppm) exhibited very high reduction in their indexes such as Efficacy of Conversion of Digested Food (82.38%) and Efficacy of Conversion of Ingested Food (74.27%), compared with control treatment. Transcriptomic analysis of red palm weevil larvae characterized numerous genes involved in chitin degradation including chitinase, chitinase-3-like protein 2, chitinase domain-containing protein 1, Endochitinase-like, chitinase 3, and chitin binding peritrophin-a domain. However, quantitative expression patterns of these genes in response to novaluron-fed larvae revealed tissue-specific time-dependent expression patterns. We recorded overexpression of all genes from mid-gut tissues. Growth retarding, chitin remodeling and larvicidal potential suggest novaluron as a promising alternate for Rhynchophorus ferrugineus management.

Benzoylphenyl ureas inhibit chitin synthesis and interfere with the molting process in arthropods. In this study, the effect of diflubenzuron on third-instar larvae of Aedes aegypti was evaluated. The susceptibility to the product was determined, and the alterations generated were shown through light and electron microscopy. LC50 and LC90 were 0.23 and 0.47 ppm, respectively. The main alterations observed were the incapacity to complete the molt, a reduction of mobility, the fragmentation of the old cuticle, a division of the body segments that was not evident, and the deformation of the caudal structures. Images of the ultrastructure are included, where breaking zones in the cuticle were observed, separation of the cuticle, the epidermis and the muscles, and these latter with a disorganized arrangement. In low concentrations, from 0.15 ppm, diflubenzuron causes alterations in the behavior and morphology of Ae. aegypti.

The objective of bait application envisioned by early researchers was to eliminate the source of infestation, the colony, but because of the lack of adequate evaluation tools, results of field trials with mirex baits in the 1960s were mostly inconclusive. On-the-ground monitoring stations and mark-recapture protocol developed in the 1970s marked the turning point in the field studies of termite baits. Results of field studies with metabolic inhibitors and chitin synthesis inhibitors (CSIs) in the 1990s indicated that a bait toxicant has to be slow-acting and nonrepellent, and its lethal time has to be dose independent. A recent discovery that termites return to the central nest to molt and CSI-poisoned termites die near the royal pair further explains the success of CSI baits in eliminating colonies. Owing to the availability of durable baits that require less-frequent site inspection, more termite control professionals have adopted baiting systems in recent years.

Abstract This study involved two field trials with the aim of evaluating the efficacy of diflubenzuron, via mineral supplementation, against Haematobia irritans parasitizing cattle. Concomitantly with the main trial, a stall test was conducted to ascertain the effects of a different formulation with the same active ingredient against Rhipicephalus microplus, along with the action of diflubenzuron on the reproductive parameters of R. microplusfemales that had naturally detached from cattle. Against H. irritans, it was observed that the efficacy indexes fordiflubenzuron were low (≤ 31.3% or 44.6%) or null (0.0%) throughout the study. The anti- R. microplus efficacy of diflubenzuron, at weekly intervals, ranged from 0.0 to 13.7% over the entire experimental period. Null efficacy (0.0%) was registered for diflubenzuron in relation to the reproductive parameters of R. microplusfemales that had naturally detached from cattle. The different diflubenzuron formulations, administered via mineral salt supplementation, did not show satisfactory efficacy indexes against H. irritans and R. microplus parasitizing cattle, within the experimental design of the present study. In addition, this agent did not present any deleterious effects on the reproductive parameters of R. microplus females.


Resumo O objetivo deste estudo foi avaliar a eficácia do diflubenzuron, administrado via suplementação mineral, contra Haematobia irritans parasitando bovinos, em dois testes à campo. Concomitantemente, foi realizado testes em estábulo para determinar os efeitos de uma formulação diferente, com o mesmo princípio ativo, contra Rhipicephalus microplus , bem como a ação do diflubenzuron nos parâmetros reprodutivos de fêmeas de R. microplus recolhidas após desprendimento natural do hospedeiro bovino. Contra H. irritans, foi observado que foram baixos (≤ 31,3% ou 44,6%) ou nulos (0,0%) os índices de eficácia do diflubenzuron. A eficácia anti-R. microplus do diflubenzuron, observada em intervalos semanais, variaram de 0,0% a 13,7% durante todo o período experimental. Com relação aos parâmetros reprodutivos das fêmeas de R. microplus recolhidas, foi observada eficácia nula (0,0%) para o diflubenzuron. Conclui-se que as diferentes formulações administradas via sal mineral no atual estudo, contra H. irritans e R. microplus parasitando bovinos, não apresentaram eficácia satisfatória. Este agente também não mostrou efeito deletério sobre os parâmetros reprodutivos de fêmeas de R. microplus.

Fluid baits comprised of 10% dry medium (impregnated with 0.5% hexaflumuron) and 90% methylcel solution were injected into foraging galleries of the Formosan subterranean termite, Coptotermes formosanus Shiraki (Blattodea: Rhinotermitidae), in trees and buildings. Elimination times at two of the six baited sites were longer than expected, possibly due to the application disturbance or the presence of an old and declining colony, but fluid bait treatments eliminated all baited colonies. Fluid baits were injected through drill holes behind infested wood surface, and bypassed the difficulties for installing above-ground (AG) bait stations on uneven surface or at tight corners. Because a large quantity of fluid baits could be applied at once, termite control professionals may be able to avoid revisiting the site to add more baits. The absence of unsightly AG station in a house will also make fluid baits more acceptable to homeowners than AG baits. Currently, local applications of liquid termiticides are common practices because termite control professionals are often urged by homeowners to take remedial actions when termites are found in a house, but such treatments only drive termites away from the active loci without affecting the termite colony(s). Remedial treatments with fluid baits would result in immediate bait consumption and shorten colony elimination time with or without the use of in-ground baiting systems. Introduction of fluid baits will add another tool to the termite control industry and will make it more plausible for the homeowners to accept baiting technology.

To date, few studies have evaluated chitin synthesis inhibitors against bed bugs, although they would provide an alternative mode of action to circumvent insecticide resistance. Acute and sublethal effects of lufenuron were evaluated against two strains of the common bed bug. Combined acute and sublethal effects were used to calculate effective doses. The dose that was effective against 50% of Harlan strain bed bugs was 0.0081% (w/v), and was much higher against Bradenton strain bed bugs (1.11% w/v). Sublethal doses were chosen to determine the effect that leg abnormalities had on pulling force. Both Harlan and Bradenton strain bed bugs had significantly lower locomotion ability (p < 0.0001) following topical application of lufenuron. The observed sublethal effects that limit locomotion could prevent bed bugs from moving within a domicile and taking a blood meal, subsequently reducing a bed bug population over time.

Bistrifluron, a benzoylphenyl urea compound, was evaluated for efficacy against Coptotermes lacteus (Froggatt), a mound-building species in southern Australia. Bistrifluron bait (tradename Xterm) was delivered as containerized pellets inserted into plastic in-ground feeding stations implanted in the sides of mounds. Termites actively tunneled in the gaps between pellets and removed bait from the canisters. Two separate trials were conducted, one commencing on 22 September 2011 and the second commencing on 30 November 2011. In trial 1, all 13 treated colonies (seven single and six double treatments) were eliminated within 19 wk, while all five untreated colonies remained healthy. In trial 2, all four treated colonies were eliminated within 14 wk. In trial 1, bait consumed or removed in treated mounds averaged 105 g for single treatments and 147 g for dual treatments, and overall ranged from 7 to 309 g (70-3,090 mg bistrifluron). In trial 2, the four treated colonies removed an average of 85 g of bait. At the time mounds were dismantled, all showed signs of inattention: external cracking, delamination, and general external weathering. Mound repair and temperature profile data indicate that colony decline commenced much earlier than 19 wk and 14 wk, respectively, for trials 1 and 2, from as early as 4 wk onward. The ability of colonies to repair mound damage was impaired as early as 4 wk in some colonies, and mean internal mound temperatures in treated mounds began declining from 8 wk onward and clearly diverged from mean temperatures of untreated mounds thereafter.

We report on the efficacy of 0.5 M (61,000 ppm) erythritol (E) in Truvia Baking Blend, 10 ppm lufenuron (L), and their combination (LE) to reduce egg and larval densities of wild populations of Drosophila suzukii (Matsumura) infesting fields of rabbiteye blueberries (Vaccinium virgatum) and blackberries (Rubus sp.). Formulations included the active ingredients (lufenuron, erythritol, or both), sugar (in control and erythritol treatments), and Dawn hand-soap applied to plants with pressurized 3-gallon garden spray tanks. The three chemical treatments (E, L, and LE) had no effect on D. suzukii ovipositing in blackberry and blueberry fruit, but they did reduce larval infestation by 75%, particularly densities of first and second instars. Erythritol and lufenuron were equally efficacious compounds as a D. suzukii ovicide and larvicide, but they did not display additive or synergistic activity. Extremely high larval mortality in control fruits show an age structure heavily skewed toward egg output.

Baiting systems have been introduced using slow-acting bait toxicants to provide environment-friendly and target-specific termite management. In the present study, the Exterra termite bait system (USA) with chlorfluazuron, a chitin synthesis inhibitor, as the active ingredient was tested against termite colonies. Ten residential areas of Lahore, infested with subterranean termites were selected for the study. The study period was from 2013 to 2015. In-ground stations were installed at 10 sites and above-ground stations were only installed at four test sites. Requiem termite bait was prepared according to the label instructions. Results showed that the range of termite activity was between 30 and 214 d to first termite activity on underground monitors with a mean of 78.23 ± 6.44. Timeline graphs also show activity of termites and active ingredient placement for each of the stations at each site. As termite feeding activity in the stations increased, there was a decrease in termite activities in wooden structures, followed by cessation of termite feeding and foraging activity noted in the building structures. It was concluded that a termite baiting system in Pakistan has the potential to suppress and reduce termite populations, when foraging termites feed on the active ingredient and share with nest mates through trophallaxis by installing more bait stations and prolonging baiting period.

BACKGROUND: An understanding of the genetic basis of insect resistance to insecticides is important for the establishment of insect resistance management (IRM) strategies. In this study we evaluated the inheritance pattern of resistance to the chitin synthesis inhibitor lufenuron in Spodoptera frugiperda. RESULTS: The LC50 values (95% CI) were 0.23 µg lufenuron mL(-1) water (ppm) (0.18-0.28) for the susceptible strain (SUS) and 210.6 µg mL(-1) (175.90-258.10) for the lufenuron-resistant strain (LUF-R), based on diet-overlay bioassay. The resistance ratio was ≈ 915-fold. The LC50 values for reciprocal crosses were 4.89 µg mL(-1) (3.79-5.97) for female LUF-R and male SUS and 5.74 µg mL(-1) (4.70-6.91) for female SUS and male LUF-R, indicating that the inheritance of S. frugiperda resistance to lufenuron is an autosomal, incompletely recessive trait. Backcrosses of the progeny of reciprocal crosses with the parental LUF-R showed a polygenic effect. The estimated minimum number of independent segregations was in the 11.02 range, indicating that resistance to lufenuron is associated with multiple genes in S. frugiperda. CONCLUSIONS: Based on genetic crosses, the inheritance pattern of lufenuron resistance in S. frugiperda was autosomal, incompletely recessive and polygenic. Implications of this finding to IRM are discussed in this paper.
