RESUMO

The global decline of pollinators has become a major concern for the scientific community, policymakers, and the general public. Among the main drivers of diminishing bee populations is the widespread use of agrochemicals. To gain a comprehensive understanding of the foraging dynamics of bees at agrochemical-contaminated areas, it is essential to consider both environmental conditions and the specific foraging ecology of bee species. For the first time, we conducted a semi-field study to investigate whether stingless bees exhibit a preference for food contaminated with agrochemicals compared to non- contaminated food, under natural weather conditions. Colonies of Plebeia lucii Moure, 2004 were placed in a greenhouse and subjected to a preference test, where bees were given the freedom to choose between contaminated or non-contaminated food sources following a preliminary training period. Within the greenhouse, we placed feeders containing realistic concentrations of an insecticide (acephate: 2 mg a.i./L), a herbicide (glyphosate: 31.3 mg a.i./L), or a mixture of both, alongside non-contaminated food. Environmental variables (temperature, humidity, and light intensity) were monitored throughout the experiment. At higher temperatures, the foragers preferred food containing the mixture of both agrochemicals or uncontaminated food over the other treatments. At lower temperatures, by contrast, the bees preferred food laced with a single agrochemical (acephate or glyphosate) over uncontaminated food or the agrochemical mixture. Our findings indicate that agrochemical residues in nectar pose a significant threat to P. lucii colonies, as foragers do not actively avoid contaminated food, despite the detrimental effects of acephate and glyphosate on bees. Furthermore, we demonstrate that even minor, natural fluctuations in environmental conditions can alter the colony exposure risk. Despite the interplay between temperature and bees' preference for contaminated food, foragers consistently collected contaminated food containing both agrochemicals, whether isolated or in combination, throughout the whole experiment.

In this study, oxidative stress and lipid peroxidation in honey bee larvae, pupae and the midguts of adult bees were investigated during a one-year exposure to radiofrequency electromagnetic fields (RF-EMFs) at a frequency of 900 MHz under field conditions. The experiment was carried out on honey bee colonies at three locations with electric field levels of 30 mV m-1, 70 mV m-1 and 1000 mV m-1. Antioxidant enzymes, glutathione-S-transferase (GST), catalase (CAT) and superoxide dismutase (SOD) and thiobarbituric acid reactive substances (TBARS) as indicators of lipid peroxidation were measured spectrophotometrically. The GST activity within the same developmental stage showed no significant differences regardless of electric field level or sampling time. The highest GST activity was found in the pupae, followed by activity in the larvae and midguts. Both CAT activity and TBARS concentration were the highest in the midguts, regardless of field level and sampling time. The larvae showed a significantly higher TBARS concentration at the location with an electric field level of 1000 mV m-1 compared to the locations with lower levels. Our results show that RF-EMFs at a frequency of 900 MHz can cause oxidative stress in honey bees, with the larval stage being more sensitive than the pupal stage, but there was no linear relationship between electric field level and effect in any of the developmental stages.

Global contraction of biodiversity pushed most members of Felidae into threatened or endangered list except the domestic cat (Felis catus) thence preferred as the best model for conservation studies. One of the emerging conservation strategies is vitrification of ovarian tissue which is field-friendly but not yet standardized. Thus, our main goal was to establish a suitable vitrification protocol for feline ovarian tissue in field condition. Feline ovarian tissue fragments were punched with biopsy punch (1.5 mm diameter) and divided into 4 groups. Group 1 was fresh control (Fr), while the other three were exposed to 3 vitrification protocols (VIT_CT, VIT_RT1 and VIT_RT2). VIT_CT involved two step equilibrations in solutions containing dimethyl sulfoxide (DMSO) and ethylene glycol (EG) for 10 min each at 4 °C. VIT_RT1 involved three step equilibration in solutions containing DMSO, EG, polyvinylpyrrolidone and sucrose for 14 min in total at room temperature, while in VIT_RT2 all conditions remained the same as in VIT_RT1 except equilibration timing which was reduced by half. After vitrification and warming, fragments were morphologically evaluated and then cultured for six days. Subsequently, follicular morphology, cellular proliferation (expression of Ki-67, MCM-7) and apoptosis (expression of caspase-3) were evaluated, and data obtained were analysed using generalised linear mixed model and chi square tests. Proportions of intact follicles were higher in Fr (P = 0.0001) and VIT_RT2 (P = 0.0383) in comparison to the other protocols both post warming and after the six-day culture. Generally, most follicles remained at primordial state which was confirmed by the low expression of Ki-67, MCM-7 markers. In conclusion, VIT_RT2 protocol, which has lower equilibration time at room temperature has proven superior thus recommended for vitrification of feline ovarian tissue.

The "oviposition preference-offspring performance" hypothesis proposes that females lay their eggs in habitats that maximize the fitness of their offspring. The aim of this study was to assess the oviposition site selection by Aedes aegypti females and the success of their larvae in habitats with different detritus accumulation times, under conditions representative of the natural spatial variability of detritus quality and quantity in the Metropolitan Area of Buenos Aires, Argentina. Two experiments were performed, one assessing oviposition site selection and the other analyzing developmental success. In both experiments, two levels of detritus accumulation time were compared, one with short-time detritus accumulation (2 weeks), and the other with long-time detritus accumulation (8 weeks). Naturally fallen detritus was used in both experiments, collected in ten sites across the Metropolitan Area of Buenos Aires. In the oviposition experiment, two contiguous ovitraps corresponding to each accumulation time were placed at each of the ten sites and the number of eggs received for each accumulation time was compared. In the development experiment, always 19 larvae were raised in containers of both accumulation times and overall performance was compared using an integrated index that considers survival, development time and female size. A large variability in the amount of detritus collected at the different sites was observed. The number of eggs was significantly higher in the long-time than in the short-time detritus accumulation containers, and approximately proportional to the amount of detritus in each ovitrap. The performance was not affected by the detritus accumulation time, but a better performance was detected in containers that received a higher amount of organic detritus, regardless of the accumulation time. Leaves were on average the most abundant type of detritus, with an average of 53 % of the total detritus collected. The amount of leaves added 2 weeks before hatching showed a positive effect on larval performance. Our results do not support the "oviposition preference-offspring performance" hypothesis, since Ae. aegypti females laid eggs in containers where larvae did not show a better performance. Moreover, at larval densities related to the number of eggs actually laid in each of the accumulation times, it is expected that the performance would be even worse in the most selected containers, due to the density-dependent effects of crowding. Since the results obtained reflect the natural heterogeneity of the environmental conditions in the region studied, they might be a fairly good indicator of what occurs in natural larval habitats.

Background and Aim: A combined microbial and cytological examination of uterine samples is the main diagnostic method for endometritis in mares. This study aimed to describe a procedure for using the same uterine cytobrush (CB) for both bacteriological and cytological evaluation. Material and Methods: The procedure consists of rolling the CB onto a sterilized glass slide immediately after collection and before the transfer into a sterile saline solution. In Experiment 1, a comparison between bacteriological results of the cotton swab (CS) and CB or pellet was made in 10 mares; in Experiment 2, bacteriological and cytological results were compared between different processing methods of CB in 28 mares; in other 6 mares, a CB was processed for cytology only, to investigate the reasons for the low cellularity of the pellet. Results: The agreement between culture results from the CB and CS was evaluated, and a comparison between the cytological data obtained by different processing methods of CB was performed. The perfect agreement between the CB and CS microbiological results was found. The described procedure enables useful diagnostic smears for cytology. Moreover, the seeding of both the tip of CB and the saline solution used for the transport produced accurate bacteriological results. Conclusion: The protocol described in this study for the use of CB for both cytological and bacteriological analysis could be used for the diagnosis of endometritis. To maximize diagnostic sample quality, cytology slides must be prepared with meticulous care in the field to preserve cellular integrity and minimize artifacts.

BACKGROUND: Malaria is a globally distributed infectious disease. According to the World Health Organization, Angola is one of the six countries that account for over half the global malaria burden in terms of both malaria cases and deaths. Diagnosis of malaria still depends on microscopic examination of thin and thick blood smears and rapid diagnostic tests (RDTs), which often lack analytical and clinical sensitivity. Molecular methods could overcome these disadvantages. The aim of this study was to evaluate, for the first time to our knowledge, the performance of a loop-mediated isothermal amplification (LAMP) for the diagnosis of malaria in an endemic area in Cubal, Angola, and to assess the reproducibility at a reference laboratory. METHODS: A total of 200 blood samples from patients attended at Hospital Nossa Senhora da Paz, Cubal, Angola, were analysed for Plasmodium spp. detection by microscopy, RDTs, and LAMP. LAMP assay was easily performed in a portable heating block, and the results were visualized by a simple colour change. Subsequently, the samples were sent to a reference laboratory in Spain to be reanalysed by the same colorimetric LAMP assay and also in real-time LAMP format. RESULTS: In field tests, a total of 67/200 (33.5%) blood samples were microscopy-positive for Plasmodium spp., 98/200 RDT positive, and 112/200 (56%) LAMP positive. Using microscopy as reference standard, field LAMP detected more microscopy-positive samples than RDTs (66/67; 98% vs. 62/67; 92.5%). When samples were reanalysed at a reference laboratory in Spain using both colorimetric and real-time assays, the overall reproducibility achieved 84.5%. CONCLUSIONS: This is the first study to our knowledge in which LAMP has been clinically evaluated on blood samples in a resource-poor malaria-endemic area. The colorimetric LAMP proved to be more sensitive than microscopy and RDTs for malaria diagnosis in field conditions. Furthermore, LAMP showed an acceptable level of reproducibility in a reference laboratory. The possibility to use LAMP in a real-time format in a portable device reinforces the reliability of the assay for molecular diagnosis of malaria in resource-poor laboratories in endemic areas.

INTRODUCTION: The risk of traumatic injury in football has been suggested to be affected by field conditions. This study, therefore, aimed to investigate whether near surface water content of the football field, influenced the risk of traumatic injuries during a youth football tournament. METHODS: At Dana Cup, an annual international youth football tournament in Denmark, all injuries were registered and classified at the on-site emergency department over 7 years. The incidence rate of traumatic injury was computed. Meteorological data and soil characteristics were used to simulate near surface water content of the playing fields. The incidence rate ratio (IRR) between water content and injury incidence rate was analyzed using a multivariate Poisson regression, controlling for tournament stage and demographical parameters. RESULTS: About 2704 injuries were recorded corresponding to a risk time of 284 905 player hours. An inverse relation between water contents and the incidence rate of traumatic injury was found (IRR = 0.24 95% CI 0.1-0.7, p = 0.01). The incidence rate of traumatic injury increased with advancing tournament stage, that is, for the finals (IRR = 4.30 95% CI 2.8-6.6, p < 0.001). Also we found an interaction between dry fields and the final stage of tournament, further exacerbating this effect. CONCLUSIONS: Our data suggest that the incidence rate of traumatic injury is increased with drier field conditions and advanced tournament stages.

Ensuring food security with severe shortages of freshwater and drastic changes in climatic conditions in arid countries requires the urgent development of feasible and user-friendly strategies. Relatively little is known regarding the impacts of the co-application (Co-A) of salicylic acid (SA), macronutrients (Mac), and micronutrients (Mic) through foliar (F) and soil (S) application strategies on field crops under arid and semiarid climatic conditions. A two-year field experiment was designed to compare the impacts of seven (Co-A) treatments of this strategy, including a control, FSA + Mic, FSA + Mac, SSA + FMic, SSA + FSA + Mic, SSA + Mic + FSA, and SSA + Mic + FMac + Mic on the agronomic performance, physiological attributes, and water productivity (WP) of wheat under normal (NI) and limited (LMI) irrigation conditions. The results reveal that the LMI treatment caused a significant reduction in various traits related to the growth (plant height, tiller and green leaf numbers, leaf area index, and shoot dry weight), physiology (relative water content and chlorophyll pigments), and yield components (spike length, grain weight and grain numbers per spike, thousand-grain weight, and harvest index) of wheat by 11.4-47.8%, 21.8-39.8%, and 16.4-42.3%, respectively, while WP increased by 13.3% compared to the NI treatment. The different Co-A treatments have shown a 0.2-23.7%, 3.6-26.7%, 2.3-21.6%, and 12.2-25.0% increase in various traits related to growth, physiology, yield, and WP, respectively, in comparison to the control treatment. The SSA+ FSA + Mic was determined as the best treatment that achieved the best results for all studied traits under both irrigation conditions, followed by FSA + Mic and SSA + Mic + FSA under LMI in addition to FSA + Mac under NI conditions. It can be concluded that the Co-A of essential plant nutrients along with SA accomplished a feasible, profitable, and easy-to-use strategy to attenuate the negative impacts of deficit irrigation stress, along with the further improvement in the growth and production of wheat under NI conditions.

The chestnut moth (Cydia splendana Hübner) and the chestnut weevil (Curculio elephas Gyllenhal) cause serious damage to European producers and companies that transform and market the chestnut. The objective of the present work was to evaluate, under field conditions, the possibilities of Beauveria bassiana (Bals.-Criv.) Vuill. to infect and kill the larvae of the two main carpophagous pests of European chestnut, in treatments directed at the soil. For this purpose, the surfaces of vases were sprayed with two concentrations of conidia/mL 5 × 107 (T1) and 1 × 108 (T2). The control (T0) was sprayed with distilled water. Larval mortality and infection were evaluated on five dates (D8 to D220). Confirmation of the fungus present in the larva was performed by molecular analysis. The results obtained are promising for the use of B. bassiana as a biological control agent against these key pests of the chestnut crop. There were no significant differences in mortality between the T1 and T2 modalities, however, they were significantly higher than the control. In the case of total mortality (dead and infected larvae), no significant differences were observed for C. elephas either. In the case of C. splendana, the T2 modality obtained better results in terms of total mortality.

In this study, we applied an inductively coupled, radio frequency oxygen plasma to maize seeds and investigated its effects on seedling emergence, plant number at tasseling, and crop yield of maize in realistic field conditions. Maize seeds of seven different hybrids were treated over two harvest years. In addition to plasma-treated seeds, a control sample, fungicide-treated seeds, an eco-layer, and a plasma and eco-layer combination, were planted. Seedling emergence, plant number at tasseling (plants/m2), and yield (kg/ha), were recorded. In the first harvest year, results were negatively affected by the presence of an insect pest. In the second harvest year, plant number and yield results were more uniform. In both years, for two and three hybrids, respectively, the highest yield arose from plants from plasma-treated seeds, but the differences were only partially significant. Considering our results, plasma treatment of maize seeds appears to have a positive effect on the yield of the plant.

Freshwater shortage and inadequate nutrient management are the two major challenges for sustainable wheat production in arid agro-ecosystems. Relatively little is known about the positive roles of the application methods for the combination of salicylic acid (SA) and plant nutrients in sustaining wheat production under arid climatic conditions. A two-year field study was undertaken to assess the impact of seven treatments for the integrated application of SA, macronutrients, and micronutrients on the morpho-physiological traits, yield, and irrigation water use efficiency (IWUE) of wheat subjected to full (FL) and limited (LM) irrigation regimes. The results showed that the LM regime caused a significant reduction in different plant growth traits, relative water content, chlorophyll pigments, yield components, and yield, while a significant increase was observed in IWUE. The sole application of SA or co-application with micronutrients through soil did not significantly affect the studied traits under the FL regime, while they achieved some improvement over untreated plants under the LM regime. Based on the different multivariate analyses, the soil and foliar applications for the combinations of SA and micronutrients, as well as a foliar application for the combinations of SA, macronutrients, and micronutrients were identified as an efficient option for mitigating the negative impacts of water deficit stress and enhancing the growth and production of wheat under normal conditions. In conclusion, the results obtained herein indicated that the co-application of SA and macro- and micronutrients is an effective option to greatly enhance and improve the growth and production of wheat crops in water-scarce countries of arid regions, such as Saudi Arabia, while an appropriate application method for this combination was required for positive effects.

The heterogeneity of Italian manufacturing processes results in the production of a large variety of pork products. In Sardinia, boars are raised and butchered to produce charcuterie. These animals are castrated before slaughter as androstenone would otherwise taint the meat, rendering it unfit for human consumption. However, to date, the literature concerning surgical orchiectomy in adult boars is limited. The goal of this study is to assess whether a sutureless swine orchiectomy procedure is feasible. Additionally, this study aims to determine the appropriate traction force needed to tie knots in the deferens duct of pigs of different weights and ages. Two groups were created: the first (n = 91) underwent orchiectomy by suture ligation; the second (n = 20) was castrated using the sutureless technique. Deferens ducts of animals in the first group (n = 182) were collected following castration, and their tensile strength was measured. Pearson's linear correlation was used to determine the relationship between the maximum traction force and weight and age groups. A correlation of 0.99 and 0.96 was shown between traction force and age and traction force and weight, respectively. In accordance with these results, sutureless castration was performed on 20 animals, calibrating the pulling force needed according to the age and weight of the boars. No complications were observed during the feasibility study, thus validating sutureless orchiectomy in adult boars.

The obstacles in Phakopsora pachyrhizi management result especially from susceptible soybean genotypes and resistant fungal strains. The objective of the current study was to evaluate the applicability of the emission of extremely low and specific frequencies by Effatha technology in the soybean Asian rust control, nutrition, and its impact on yield. The in-vivo test followed the detached leaves method, with six treatments: frequencies 1 and 2 individually and in association; the conventional chemical treatment (fungicide azoxystrobin + benzovindiflupyr); and witnesses in presence and absence of the fungus. Frequency 1 relates to inhibition of the enzyme succinate dehydrogenase and 2 to ubiquinone oxidase. In the field, frequencies 1 and 2 associated (with the same fungicidal action of the in-vivo study); nutritional frequency; application of azoxystrobin + benzovindiflupyr + mancozeb, and control without application were evaluated. In vivo, the fungicide provided 85% control of the disease symptoms, against 65% of frequencies 1 and 2 in association, which showed a higher efficiency compared to the isolated frequencies. In the field, the rate of increase of symptoms were reduced by all treatments compared to the control. At the end of the soybean cycle, the conventional fungicide resulted in 33% severity against 56% of frequencies 1 and 2 associated, and 69.2% of the control. The emission of the frequency for increased nutrient efficiency stood out positively on yield in relation to all the other ones. The conventional application provided the highest weight of 1,000 grains, possibly a direct reflection of the better control of the disease.

Tropical grasslands are very important to global carbon and water cycles. C4 plants have increased heat tolerance and a CO2 concentrating mechanism that often reduces responses to elevated concentrations of CO2 ([CO2]). Despite the importance of tropical grasslands, there is a scarcity of studies that elucidate how managed tropical grasslands will be affected by elevated [CO2] and warming. In our study, we used a combination of a temperature-free air-controlled enhancement (T-FACE) and a free-air carbon dioxide enrichment (FACE) systems to increase canopy temperature and [CO2] under field conditions, respectively. We warmed a field-grown pasture dominated by the C4 tropical forage grass Megathyrsus maximus by 2°C above ambient under two levels of [CO2] (ambient (aC) and elevated (eC - 600 ppm) to investigate how these two factors isolated or combined regulate water relations through stomatal regulation, and how this combination affects PSII functioning, biochemistry, forage nutritive value, and digestibility. We demonstrated that the effects of warming negated the effects of eC in plant transpiration, water potential, proline content, and soil moisture conservation, resulting in warming canceling the eCO2-induced improvement in these parameters. Furthermore, there were additive effects between eC and warming for chlorophyll fluorescence parameters and aboveground nutritive value. Warming sharply intensified the eCO2-induced decrease in crude protein content and increases in forage fibrous fraction and lignin, resulting in a smaller forage digestibility under a warmer CO2-enriched atmosphere. Our results highlight the importance of multifactorial studies when investigating global change impacts on managed ecosystems and the potential consequences for the global carbon cycle like amplification in methane emissions by ruminants and feeding a positive climate feedback system.

Efficacy of the essential oil of Lippia sidoides to control the bovine tick Rhipicephalus (Boophilus) microplus was evaluated in tests on naturally infested cattle under field conditions. Twenty cattle were allocated to one of two groups (Control and Treated). Animals in the treated group were sprayed with three liters of 1 % L. sidoides solution (10,000 ppm) and efficacy was calculated from teleogynous counts between 4.5 and 8 mm in length before (days -3, - 2 and - 1) and after treatment (days +3, +7, +14, +21 and +28). The essential oil of L. sidoides showed an average efficacy of 50.7 % between days + 3 and + 21, with greater efficacy in the larvae (63.2 % - p = 0.0001).

Maize yields worldwide are limited by foliar diseases that could be fungal, oomycete, bacterial, or viral in origin. Correct disease identification is critical for farmers to apply the correct control measures, such as fungicide sprays. Deep learning has the potential for automated disease classification from images of leaf symptoms. We aimed to develop a classifier to identify gray leaf spot (GLS) disease of maize in field images where mixed diseases were present (18,656 images after augmentation). In this study, we compare deep learning models trained on mixed disease field images with and without background subtraction. Performance was compared with models trained on PlantVillage images with single diseases and uniform backgrounds. First, we developed a modified VGG16 network referred to as "GLS_net" to perform binary classification of GLS, which achieved a 73.4% accuracy. Second, we used MaskRCNN to dynamically segment leaves from backgrounds in combination with GLS_net to identify GLS, resulting in a 72.6% accuracy. Models trained on PlantVillage images were 94.1% accurate at GLS classification with the PlantVillage testing set but performed poorly with the field image dataset (55.1% accuracy). In contrast, the GLS_net model was 78% accurate on the PlantVillage testing set. We conclude that deep learning models trained with realistic mixed disease field data obtain superior degrees of generalizability and external validity when compared to models trained using idealized datasets.

Microplastics (MPs) as emerging persistent pollutants are ubiquitous in terrestrial environments. The effects of MPs input on soil dissolved organic matter (DOM) yet remain largely unclear, which limits our ability to predict how soil carbon dynamics will respond to the intensifying terrestrial plastic contamination, especially under the context of climate change. Here, a long-term field experiment with MPs addition treatment in soils spanning cold temperate zone to the tropics in China was conducted to evaluate the effects of MPs on DOM composition and to explore the spatial and temporal distribution patterns and relevant mechanistic controls of DOM responses after MPs input. We report that the DOM composition in soils with MPs addition was significantly changed relative to the ambient, in which tryptophan-like substances were decreased and humic-like substances were increased. Moreover, we find more apparent transformations of DOM composition in soils for longer treatment time, suggesting a long-lasting effect of MPs on DOM. The overall impact of MPs on DOM is more pronounced in lower latitudes, and nutrient availability and latitude-related climatic variables are associated with the influence degree of MPs input on soil DOM.

Increasing populations and temperatures are expected to escalate food demands beyond production capacities, and the development of maize lines with better performance under heat stress is desirable. Here, we report that constitutive ectopic expression of a heterologous glutaredoxin S17 from Arabidopsis thaliana (AtGRXS17) can provide thermotolerance in maize through enhanced chaperone activity and modulation of heat stress-associated gene expression. The thermotolerant maize lines had increased protection against protein damage and yielded a sixfold increase in grain production in comparison to the non-transgenic counterparts under heat stress field conditions. The maize lines also displayed thermotolerance in the reproductive stages, resulting in improved pollen germination and the higher fidelity of fertilized ovules under heat stress conditions. Our results present a robust and simple strategy for meeting rising yield demands in maize and, possibly, other crop species in a warming global environment.

In the literature, little information is available on the effect of Selenium (Se) on durum wheat yield and grain quality performances. A field investigation was conducted to explore the effect of exogenous Se foliar supply on two types of durum wheat germplasm; i.e., 16 advanced lines and nine modern varieties. The Se effect was assessed on grain yield as well as on technological quality traits (moisture, protein and gluten contents, Zeleny sedimentation index, and deformation energy) in two contrasting environments in Tunisia, namely Kef-Boulifa (semi-arid region) and Beja (sub-humid region). The results displayed significant effects of environments, Se foliar application, and cultivars on grain yield and quality attributes. For grain yield performance, the beneficial effect of Se was more pronounced under the Kef-Boulifa environment, and conversely for the grain quality. A genetic variation was observed within and among the two environments under both Se treatments (with and without Se). Notably, the Se-treated advanced lines displayed the highest grain yield under Kef-Boulifa and Beja conditions. Although these cultivars showed better grain quality in both sites, the modern varieties valorized the Se foliar application better. Cultivars that recorded the highest values for the studies attributes were not necessarily those that valorized the Se supply better. Interestingly, some advanced lines have noted superiority compared to the modern varieties. In this study, cultivars that combine both good yield and good grain quality were determined for semi-arid (L11, L1, Dhahbi, and Maali) and sub-humid (L2, L14, L6, L3, Salim, and INRAT 100) zones. The screening results provide genetic material that could be exploited in breeding programs to improve Se use efficiency.

Creating germplasm banks of wild species, such as the Iberian red Deer (Cervus elaphus hispanicus) can be challenging. One of the main difficulties is the obtention and cryopreservation of good-quality reproductive cells when the spermatozoa are obtained from epididymides after death. To avoid a loss of seminal quality during transport, developing alternative methods for cooling and freezing sperm samples under field conditions is necessary. The objective of this study was to evaluate the effects of different durations of equilibrium and different techniques of cooling and freezing on Iberian red deer epididymal sperm quality after thawing to optimize the processing conditions in this species. Three experiments were carried out: (I) evaluation of refrigeration in straws or tubes of 15 mL; (II) study of equilibration period (0, 30, 60, or 120 min); and (III) comparison of four freezing techniques (liquid nitrogen vapor in a tank (C), liquid nitrogen vapor in a polystyrene box (B), dry ice (DY), and placing straws on a solid metallic plate floating on the surface of liquid nitrogen (MP)). For all experiments, sperm motility and kinematic parameters, acrosomal integrity, sperm viability, mitochondrial membrane potential, and DNA integrity were evaluated after thawing. All statistical analyses were performed by GLM-ANOVA analysis. Samples refrigerated in straws showed higher values (p ≤ 0.05) for mitochondrial activity and lower values (p ≤ 0.05) for apoptotic cells. Moreover, the acrosome integrity showed significant differences (p ≤ 0.05) between 0 and 120 min, but not between 30 and 60 min, of equilibration. Finally, no significant differences were found between freezing in liquid nitrogen vapors in a tank or in a box, although there was a low quality after thawing when the samples were cryopreserved in dry ice or by placing straws on a solid metallic plate floating on the surface of liquid nitrogen. In conclusion, under field conditions, it would be possible to refrigerate the sperm samples by storing them in straws with a 120 min equilibration period and freezing them in liquid nitrogen vapors in a tank or box.
