Objective: To evaluate the effects of a 4-week intervention combining small-sided games (SSGs) and high-intensity interval training (HIIT) on physical fitness in collegiate male soccer players. Methods: Twenty-one soccer players were randomly assigned to either the HIIT + SSGs group (n = 11) or a control group (n = 10). Physical fitness was assessed at baseline and 1-week post-intervention, including countermovement jump (CMJ), change of direction (COD) test, sprint test, repeated sprint ability (RSA) test, and 30-15 Intermittent Fitness Test (30-15IFT). The intervention comprised eight sessions over 4 weeks: four SSGs and four HIIT. Results: The intervention group showed small to moderate improvements: mean RSA improved by 4.5% (p = 0.07), CMJ increased by 3.2% (p = 0.12), and 30-15IFT scores enhanced by 6.8% (p = 0.09). Key predictors of group membership included heart rate load per minute (OR 1.602) and various GPS variables. Conclusion: The 4-week intervention combining SSGs with HIIT did not produce statistically significant improvements in most physical fitness variables compared to the control group. Although there were positive trends in variables such as RSA and 30-15IFT, these changes were modest and not statistically significant. The results suggest that while the combined SSGs and HIIT approach shows potential, its impact on physical fitness over a 4-week period is limited, with some variables, like CMJ, even showing decreases.

The study aim was to explore how Sámi healthcare staff experience working as an ethnic minority in the Norwegian healthcare system. This was a qualitative focus group and individual interview study inspired by interpretive description, with 14 participants. The results indicate that Sámi healthcare staff experience various degrees of cultural load in their jobs. This was shown through the two themes: "Feeling responsible for Sámi patients and non-Sámi colleagues" and "Feeling exhausted as a Sámi healthcare worker." To provide culturally safe care for all Sámi patients and their families, it is vital to ensure the well-being of the small number of Sámi healthcare personnel. Therefore, we emphasize the need for training programs for non-Sámi healthcare staff to provide them with the knowledge needed to support their encounters with Sámi patients in culturally safe ways. Sámi healthcare staff cannot take all responsibility for teaching their non-Sámi colleagues and acting as cultural mediators in all situations that non-Sámi staff find challenging. The risk of cultural load and burnout is very present. Nursing departments in universities and leaders in primary and secondary healthcare need to address these issues in order to ensure culturally safe care to all patients and support Sámi healthcare staff.

BACKGROUND: The escalating global burden of stress and depression underscores an urgent need to unravel their complex interrelationships and underlying mechanisms. This investigation delves into the intricate dynamics between stress and depression, spotlighting the Neuroimmunoinflammatory Stress Model (NIIS), which elucidates the pivotal role of cellular and molecular pathways in mediating these conditions. METHODS: Through an exhaustive review of literature spanning epidemiology, neurobiology, and psychoneuroimmunology, this study synthesizes the current understanding of stress and depression. It accentuates the definitional scopes, interplay, and intricacies of the NIIS model, which integrates neuroimmune-inflammatory responses into the conceptual framework of the stress-depression interaction. RESULTS: By identifying stress as a multifactorial reaction to perceived adversities and depression as a manifestation of prolonged stress exposure, our analysis foregrounds the NIIS model. This paradigmatic model reveals the transition from normal stress responses to pathological neuroinflammatory pathways, highlighting neurotransmitter imbalances, disruptions in neuronal and glial homeostasis, and ensuing low-grade neuroinflammation as key factors in the pathogenesis of depression under chronic stress conditions. The NIIS model identifies prolonged cellular pro-inflammatory stress of neurons and microglia as a fundamental pathological subsystem of many neuropsychiatric disorders. In turn, neuroinflammation and associated neurodegenerative processes are complications of chronic psychoemotional stress, which can clinically manifest as depression. CONCLUSIONS: The NIIS model views depression as the terminal stage of chronic stress, pathogenetically linked to latent neuroinflammation. This insight not only advances our understanding of their etiopathogenesis but also paves the way for developing precise therapeutic interventions.

Sleep disorders are prevalent and interfering conditions that affect people living with HIV (PLWH) at higher rates than the general population. Lower quality sleep has been associated with poorer health-related quality of life and immune function in PWH, though sleep is typically assessed subjectively. The current study aimed to examine the association between objective sleep/wake patterns measured via actigraphy with HIV outcomes. Participants (N = 87) were recruited from a public, urban HIV clinic located in the Southeastern United States. Participants were instructed to wear actigraphy monitors for one week (Range: 5-8 days). Log viral load and absolute CD4 were obtained via medical chart review. Linear regression analyses predicting HIV RNA Viral Load (log transformed) and CD4 Count were employed with three actigraphy sleep variables: sleep efficiency, wake after sleep onset (WASO), and sleep quantity. Backward entry regression with both significant actigraphy predictors, sleep efficiency and WASO, included as predictors resulted in sleep efficiency remaining in the model and WASO being removed. Separate models revealed that each one-unit increase in sleep efficiency was associated with a b = 0.032-point decrease in the log-transformed HIV RNA viral load (p = 0.03) and for each one-unit increase in wake after sleep onset (WASO) was associated with a b = 0.35-point increase in the log-transformed HIV RNA viral load (p = 0.04). Sleep quantity, however, was not, and none were associated with absolute CD4 count. The findings add to the evidence for an association of objectively measured poorer sleep efficiency being associated with higher HIV RNA viral load. Implications for clinical practice include assessing and addressing sleep efficiency as part of comprehensive clinical HIV care.

Partial nitritation-anammox (PN/A) is a cost-effective technology in high ammonia-nitrogen wastewater treatment. However, PN/A is prone to instability as the ammonia-nitrogen sharply fluctuates. In this study, a packed bed reactor is employed to construct a single-stage PN/A system to investigate the operational characteristics and explore the denitrification mechanism. The effluent NH4+-N concentration, ammonia nitrogen removal rate (ARE), and total nitrogen removal rate (TNR) could be sustained at about 60 mg/L, 80%, and over 70%, respectively, when the influent nitrogen load rate (NLR) is changed from 0.733 to 0.879 kg-N/m3/day. Both ARE and TNR are decreased when NLR continues increasing to 1.026 kg-N/m3/day. The influent NLR decreases from 0.879 to 0.147 kg-N/m3/day, and ARE and TNR reached 98% and 85.4%, respectively. Therefore, the denitrification effect of the reactor could be recovered, and the excellent nitrogen removal capacity could be obtained within a wide range of influent NLR. Moreover, the high-throughput sequencing and metagenomic testing indicate that the Proteobacteria and Planctomycetes that the PN/A functional strains (i.e., ammonia-oxidizing bacteria (AOB) and anammox bacteria (AnAOB)) account for 38.8% in the sludge. The relative abundance of Nitrospira containing the nitrite-oxidizing bacteria (NOB) has dropped to 0.01%, and the functional gene nxr of the nitrite oxidation process is also inhibited. The relative expression of the functional gene is dominated by the short-range nitritation and anammox oxidation, which demonstrates that the nitrogen removal is mainly dominated by nitritation-anammox.

OBJECTIVE: Analyze the association between histopathology, seizure outcomes, and drug load of antiseizure medications (ASMs) 5-8 years after epilepsy surgery to inform preoperative decision-making and consultation. METHODS: In this retrospective, non-interventional, single-center study, patients who visited the epilepsy clinic at West China Hospital, Sichuan University from Jan 1, 2015 to Dec 31, 2020 were assessed. Patients with postoperative histopathology after epilepsy resection were included and categorized into 13 etiological groups. The primary outcomes were achieving Engel class 1 at 1, 2, 3, 5, and 8 years postoperative. Secondary outcomes included the use of ASMs and comparison of postoperative seizure outcomes between adults and children. Univariate and multivariable analyses were conducted to explore the association between clinical characteristics such as histopathology and seizure outcomes. RESULTS: A total of 315 patients were include. Patients with embryonic dysplastic neuroepithelial tumor (DNT) achieved the best seizure outcomes (84.6 % Engel class 1). DNT (odds ratio, OR=0.103, 95 %CI=0.012-0.899), cavernous hemangiomas (OR=0.140, 95 %CI=0.024-0.819) and meningioma (OR=0.137, 95 %CI=0.021-0.910) were independently associated with a higher probability of seizure-free outcome. The results of epileptic seizures in adult and pediatric groups with different pathologies were significantly different, and the preoperative and postoperative ASM dosages were also different among adult patients with various etiologies. Additionally, multivariate analysis showed that early age at onset (adjusted hazard ratio (HR) = 1.754, 95 % CI=1.049-2.934, P=0.032), late surgical age (HR=0.569, 95 %CI=0.339-0.954, P=0.032), and longer duration from seizure onset to surgery (HR=1.735, 95 % CI=1.028-2.928, P=0.039) were independent predictors of unfavorable outcomes in epileptic seizures. CONCLUSIONS: we demonstrated that the seizure outcomes of focal epilepsy have high pathological specificity, with histopathological diagnosis serving as a crucial and independent determinant of seizure outcome. Surgical assessment should be contemplated for all patients with presumed refractory focal epilepsy, irrespective of their age.

Visceral leishmaniasis (VL) is a severe infectious disease caused by protozoan parasites of the Leishmania donovani complex. Blood cytokine concentrations in VL patients can inform us about underlying immunopathogenesis and may serve as a biomarker for treatment effectiveness. However, cytokine levels have not yet been studied in VL patients from Kenya, where case load is high. This study measured the serum cytokine profile, blood parasite load and clinical and haematological features of VL patients from West Pokot County, Kenya, over the course of treatment with sodium stibogluconate and paromomycin (SSG-PM). VL patients recruited at the hospital presented with splenomegaly and weight loss, and frequently had pancytopenia and anaemia. Median Leishmania parasite load in blood, determined with real-time polymerase chain reaction, was 2.6 × 104 parasite equivalents mL−1. Compared to endemic healthy controls, serum interferon gamma (IFN-γ), interleukin 5 (IL-5), IL-6, IL-10, IL-12p70, IL-17A and IL-27 were significantly elevated in untreated VL patients. Severe VL was associated with higher IL-10 and lower IFN-γ levels. After 17 daily injections with SSG-PM, disease symptoms disappeared, leukocyte and thrombocyte counts significantly increased, and blood parasite load decreased to undetectable levels in all VL patients. There was a significant decrease in IL-10 and IL-6, whereas IL-17A levels increased; the remaining cytokines showed no significant concentration change during treatment. In conclusion, the results suggest that SSG-PM treatment of VL patients from West Pokot was effective. Moreover, both inflammatory and regulatory immune responses appeared to decrease during treatment, although the increase in IL-17A could reflect a partial continuation of immune activation.

Objective: Mild metabolic acidosis may adversely affect cardiovascular risk factors, and diet-dependent acid-base load may impair mental health and sleep quality. The aim of this study was to investigate the effects of dietary acid load on cardiometabolic risk factors, psychological resilience, and sleep quality in adolescents with obesity. Methods: 205 adolescents with obesity (105 males, 100 females) aged 13-18 years participated in the study. Participants' biochemical parameters, anthropometric measurements and blood pressures were measured. Three-day retrospective food intake records were collected from the adolescents, and potential renal acid load (PRAL), net endogenous acid production (NEAP), and dietary acid load (DAL) were derived from food intake records. Psychological resilience levels of adolescents were assessed by the Child and Youth Resilience Measure (CYRM-12) and sleep quality was assessed by the Pittsburgh Sleep Quality Index (PSQI). Results: It was found that BMI, fat mass, fat percentage, fasting insulin, triglyceride, systolic blood pressure, HOMA-IR and PSQI scores were significantly higher and psychological resilience levels were significantly lower in high tertiles of dietary acid load (p<0.05). Adolescents in the lowest tertile of dietary acid load scores had higher consumption of whole grains, vegetables, dairies, legumes, and higher intakes of potassium and calcium than adolescents in the highest tertile of the dietary acid load scores (p<0.05). Red meat, white meat consumption and sodium intake were higher in adolescents in the high tertiles (p<0.05). Energy intakes were found to be significantly lower in the first tertile of PRAL and DAL scores compared to the other tertiles (p<0.05). According to the linear regression model, an increase in NEAP, PRAL and DAL scores leads to a decrease in psychological resilience score and an increase in PSQI and HOMA-IR scores (p<0.05). Conclusion: High dietary acid load is associated with high cardiometabolic risk, insulin resistance, and low psychological resilience and poor sleep quality.

OBJECTIVE: To investigate the risk factors and prognostic factors that affect the long-term clinical outcomes of acute retinal necrosis (ARN). METHODS: A retrospective study of patients with ARN who underwent treatment and completed follow-up in our ophthalmology department from 2011 to 2021 was conducted. The incidence and risk factors of retinal detachment (RD) and prognostic factors affecting long-term clinical outcomes, such as late-onset RD and final vision loss (< 20/200), were analyzed. RESULTS: Totally 59 ARN patients (65 eyes) with an average follow-up of 48.9 months were enrolled. During the follow-up period, RD occurred in 34 eyes (52.3%). The risk factors for RD included quadrants of involved retinal necrosis (odds ratio [OR], 4.181; 95% confidence interval [CI], 1.950-10.834) and initial intraocular viral load (OR, 1.721; 95% CI, 1.071-3.083). Early intravitreal antiviral treatment (OR, 1.204; 95% CI, 1.040-1.480) was independently associated with a decreased risk of late-onset RD. The factors independently associated with an increased risk of final vision loss were worse initial visual acuity (OR, 3.895; 95% CI, 1.551-13.662) and late-onset RD (OR, 8.043; 95% CI, 1.380-67.216). In addition, we utilized the fluctuating magnitude of viral load to quantify the extent of its reduction in comparison to its original value following the initial intravitreal antiviral injection (IAI). This ratio was strongly related to initial intraocular IL-8 concentration (Spearman correlation coefficient=-0.741, P = 0.000) and moderately related to the initial degree of aqueous flare (Spearman correlation coefficient=-0.508, P = 0.010). CONCLUSION: RD is a common and severe complication of ARN with multiple risk factors, such as initial retinitis involvement area and initial intraocular viral load. Active local antiviral therapy may reduce the risk of late-onset RD. The antiviral medication should be adjusted according to the inflammatory state. Therefore, timely detection of causative viruses and intensive systemic and local antiviral therapy is crucial for preserving visual function in ARN patients.

Osgood Schlatter's disease (OSD) is characterized by pain at the tibial tuberosity provoked during knee-loading activities, and is common in adolescent athletes. The aim of this study was to characterize clinical, pain and ultrasound imaging characteristics in participants with OSD compared to controls. This cross-sectional study included adolescents diagnosed with OSD and matched controls. Following baseline evaluation including ultrasound, participants completed the following aggravating activities in a randomized order: single-leg isometric knee hold, single-leg squat, single-leg vertical jump, hopping, running, cutting, lunges, and walking. Participants rated pain intensity on a numeric rating scale (0-10; no pain to worst pain imaginable) and localization during activities. We included 35 participants with OSD (48.5% females, age 13.0 [SD 1.5]) and 21 controls (47.6% females, age 13.4 [SD1.4]). Doppler signal was more prevalent in OSD participants at the tendon (77% vs. 30%) and tuberosity (29% vs. 10%). Tendon thickness was greater in OSD at distal (mean difference = 4.5 mm 95% CI 1.5-7.5) and proximal sites (mean difference = 4.2 95% CI 0.1-8.3). Aggravating activities induced higher pain in OSD. The greatest differences between OSD and control were the dynamic single-leg squat (mean difference = 4.2 (95% CI 3.22-5.1)). Pain was localized at the tibial tuberosity and patellar tendon during activities. Sex, sports participation, bilateral pain, and Doppler were associated with greater pain during aggravating activities. Single-leg activities loading the tibial tuberosity through the tendon appear to provoke OSD-related pain more than other sports specific movements. This may be useful to guide adolescents on which activities are likely to aggravate pain.

Root-associated microbiota profoundly affect crop health and productivity. Plants can selectively recruit beneficial microbes from the soil and actively balance microbe-triggered plant-growth promotion and stress tolerance enhancement. The cost associated with this is the root-mediated support of a certain number of specific microbes under nutrient limitation. Thus, it is important to consider the dynamic changes in microbial quantity when it comes to nutrient condition-induced root microbiome reassembly. Quantitative microbiome profiling (QMP) has recently emerged as a means to estimate the specific microbial load variation of a root microbiome (instead of the traditional approach quantifying relative microbial abundances) and data from the QMP approach can be more closely correlated with plant development and/or function. However, due to a lack of detailed-QMP data, how soil nutrient conditions affect quantitative changes in microbial assembly of the root-associated microbiome remains poorly understood. A recent study quantified the dynamics of the soybean root microbiome, under unbalanced fertilization, using QMP and provided data on the use of specific synthetic communities (SynComs) for sustaining crop productivity. In this editorial, we explore potential opportunities for utilizing QMP to decode the microbiome for sustainable agriculture.

It has been assumed that the COVID-19 vaccination reduces the risk of transmission to others. Results during the delta predominance show that the viral load in the vaccinated population is not consistently lower compared to the unvaccinated, and during the omicron predominance, the viral load was even somewhat higher. Levels of infectious SARS-CoV-2 were partly lower in the vaccinated population. Viral loads were mostly lower in re-infections compared to breakthrough infections. Viral clearance including the detection of infectious virus has mostly been described to be faster in the vaccinated population suggesting a shorter duration as a possible source for transmission. The epidemiological relevance of this finding remains uncertain. Approximately half of the transmission studies found lower secondary attack rates from the fully vaccinated population, but the results are probably best explained by the vaccination status of the contact population. Public health data from the UK show that the number of COVID-19 cases is higher among the fully vaccinated and boosted population who might be possible sources, in contrast to lower case numbers within the first three months among the vaccinated obtained in phase 3 trials on symptomatic cases. Overall, there is no convincing evidence that the COVID-19 vaccination significantly reduces the risk to transmit SARS-CoV-2 to others.

Introduction: Physical exercise and gravitational load affect the activity of the cardiovascular system. How these factors interact with one another is still poorly understood. Here we investigate how the cardiovascular system responds to leg-press exercise during head-down tilt, a posture that reduces orthostatic stress, limits gravitational pooling, and increases central blood volume. Methods: Seventeen healthy participants performed leg-press exercise during head-down tilt at different combinations of resistive force, contraction frequency, and exercise duration (30 and 60 s), leading to different exercise power. Systolic (sBP), diastolic (dBP), mean arterial pressure (MAP), pulse pressure (PP) and heart rate (HR) were measured continuously. Cardiovascular responses were evaluated by comparing the values of these signals during exercise recovery to baseline. Mixed models were used to evaluate the effect of exercise power and of individual exercise parameter on the cardiovascular responses. Results: Immediately after the exercise, we observed a clear undershoot in sBP (Δ = -7.78 ± 1.19 mmHg), dBP (Δ = -10.37 ± 0.84 mmHg), and MAP (Δ = -8.85 ± 0.85 mmHg), an overshoot in PP (Δ = 7.93 ± 1.13 mmHg), and elevated values of HR (Δ = 33.5 ± 0.94 bpm) compared to baseline (p < 0.0001). However, all parameters returned to similar baseline values 2 min following the exercise (p > 0.05). The responses of dBP, MAP and HR were significantly modulated by exercise power (correlation coefficients: rdBP = -0.34, rMAP = -0.25, rHR = 0.52, p < 0.001). All signals' responses were modulated by contraction frequency (p < 0.05), increasing the undershoot in sBP (Δ = -1.87 ± 0.98 mmHg), dBP (Δ = -4.85 ± 1.01 and Δ = -3.45 ± 0.98 mmHg for low and high resistive force respectively) and MAP (Δ = -3.31 ± 0.75 mmHg), and increasing the overshoot in PP (Δ = 2.57 ± 1.06 mmHg) as well as the value of HR (Δ = 16.8 ± 2.04 and Δ = 10.8 ± 2.01 bpm for low and high resistive force respectively). Resistive force affected only dBP (Δ = -4.96 ± 1.41 mmHg, p < 0.0001), MAP (Δ = -2.97 ± 1.07 mmHg, p < 0.05) and HR (Δ = 6.81 ± 2.81 bpm, p < 0.0001; Δ = 15.72 ± 2.86 bpm, p < 0.0001; Δ = 15.72 ± 2.86 bpm, p < 0.05, depending on the values of resistive force and contraction frequency), and exercise duration affected only HR (Δ = 9.64 ± 2.01 bpm, p < 0.0001). Conclusion: Leg exercises caused only immediate cardiovascular responses, potentially due to facilitated venous return by the head-down tilt position. The modulation of dBP, MAP and HR responses by exercise power and that of all signals by contraction frequency may help optimizing exercise prescription in conditions of limited orthostatic stress.

Objectives: Pupil dilation is controlled both by sympathetic and parasympathetic nervous system branches. We hypothesized that the dynamic of pupil size changes under cognitive load with additional false feedback can predict individual behavior along with heart rate variability (HRV) patterns and eye movements reflecting specific adaptability to cognitive stress. To test this, we employed an unsupervised machine learning approach to recognize groups of individuals distinguished by pupil dilation dynamics and then compared their autonomic nervous system (ANS) responses along with time, performance, and self-esteem indicators in cognitive tasks. Methods: Cohort of 70 participants were exposed to tasks with increasing cognitive load and deception, with measurements of pupillary dynamics, HRV, eye movements, and cognitive performance and behavioral data. Utilizing machine learning k-means clustering algorithm, pupillometry data were segmented to distinct responses to increasing cognitive load and deceit. Further analysis compared clusters, focusing on how physiological (HRV, eye movements) and cognitive metrics (time, mistakes, self-esteem) varied across two clusters of different pupillary response patterns, investigating the relationship between pupil dynamics and autonomic reactions. Results: Cluster analysis of pupillometry data identified two distinct groups with statistically significant varying physiological and behavioral responses. Cluster 0 showed elevated HRV, alongside larger initial pupil sizes. Cluster 1 participants presented lower HRV but demonstrated increased and pronounced oculomotor activity. Behavioral differences included reporting more errors and lower self-esteem in Cluster 0, and faster response times with more precise reactions to deception demonstrated by Cluster 1. Lifestyle variations such as smoking habits and differences in Epworth Sleepiness Scale scores were significant between the clusters. Conclusion: The differentiation in pupillary dynamics and related metrics between the clusters underlines the complex interplay between autonomic regulation, cognitive load, and behavioral responses to cognitive load and deceptive feedback. These findings underscore the potential of pupillometry combined with machine learning in identifying individual differences in stress resilience and cognitive performance. Our research on pupillary dynamics and ANS patterns can lead to the development of remote diagnostic tools for real-time cognitive stress monitoring and performance optimization, applicable in clinical, educational, and occupational settings.

Reproductive strategies of sexually dimorphic plants vary in response to the environment. Here, we ask whether the sexual systems of Fagopyrum species (i.e., selfing homostylous and out-crossing distylous) represent distinct adaptive strategies to increase reproductive success in changing alpine environments. To answer this question, we determined how spatial and temporal factors (e.g., elevation and peak flowering time) affect reproductive success (i.e., stigmatic pollen load) in nine wild Fagopyrum species (seven distylous and two homostylous) among 28 populations along an elevation gradient of 1299-3315 m in the Hengduan Mountains, southwestern China. We also observed pollinators and conducted hundreds of hand pollinations to investigate inter/intra-morph compatibility, self-compatibility and pollen limitation in four Fagopyrum species (two distylous and two homostylous). We found that Fagopyrum species at higher elevation generally had bigger flowers and more stigmatic pollen loads; late-flowering individuals had smaller flowers and lower pollen deposition. Stigmatic pollen deposition was more variable in distylous species than in homostylous species. Although seed set was not pollen-limited in all species, we found that fruit set was much lower in distylous species, which rely on frequent pollinator visits, than in homostylous species capable of autonomous self-pollination. Our findings that pollination success increases at high elevations and decreases during the flowering season suggest that distylous and homostylous species have spatially and temporally distinct reproductive strategies related to environment-dependent pollinator activity.

Understanding genome-wide diversity, inbreeding, and the burden of accumulated deleterious mutations in small and isolated populations is essential for predicting and enhancing population persistence and resilience. However, these effects are rarely studied in limestone karst plants. Here, we re-sequenced the nuclear genomes of 62 individuals of the Begonia masoniana complex (B. liuyanii, B. longgangensis, B. masoniana and B. variegata) and investigated genomic divergence and genetic load for these four species. Our analyses revealed four distinct clusters corresponding to each species within the complex. Notably, there was only limited admixture between B. liuyanii and B. longgangensis occurring in overlapping geographic regions. All species experienced historical bottlenecks during the Pleistocene, which were likely caused by glacial climate fluctuations. We detected an asymmetric historical gene flow between group pairs within this timeframe, highlighting a distinctive pattern of interspecific divergence attributable to karst geographic isolation. We found that isolated populations of B. masoniana have limited gene flow, the smallest recent population size, the highest inbreeding coefficients, and the greatest accumulation of recessive deleterious mutations. These findings underscore the urgency to prioritize conservation efforts for these isolated population. This study is among the first to disentangle the genetic differentiation and specific demographic history of karst Begonia plants at the whole-genome level, shedding light on the potential risks associated with the accumulation of deleterious mutations over generations of inbreeding. Moreover, our findings may facilitate conservation planning by providing critical baseline genetic data and a better understanding of the historical events that have shaped current population structure of rare and endangered karst plants.

Maritime studies, encompassing a range of disciplines, increasingly rely on advanced data analytics, particularly in the context of navigation. As technology advances, the statistical averaging of large datasets has become a critical component of these analyses. However, recent studies have highlighted discrepancies between statistical predictions and observable realities, especially in high-stress environments like port approach procedures conducted by marine pilots. This study analyzed physiological responses recorded during simulation exercises involving experienced marine pilots. The focus was not on the specific outcomes of the simulations but on the potential faults arising from conventional statistical signal processing, particularly mean-centered approaches. A large dataset of signals was generated, including one signal with a dominant characteristic intentionally designed to introduce imbalance, mimicking the uneven distribution of real-world data. Initial analysis suggested that the average physiological response of the pilots followed an S-shaped curve, indicative of a psycho-physiological reaction to stress. However, further post hoc analysis revealed that this pattern was primarily influenced by a single participant's data. This finding raises concerns about the generalizability of the S-curve as a typical stress response in maritime pilots. The results underscore the limitations of relying solely on conventional statistical methods, such as mean-centered approaches, in interpreting complex datasets. The study calls into question the validity of standardizing data interpretations based on dominant characteristic curves, particularly in environments as intricate as maritime navigation. The research highlights the need for a re-evaluation of these methods to ensure more reliable and nuanced conclusions in maritime studies. This study contributes to the ongoing discourse on data interpretation in maritime research, emphasizing the critical need to re-assess conventional statistical signal processing techniques. By recognizing the potential pitfalls in data generalization, the study advocates for more robust analytical approaches to better capture the complexities of real-world maritime challenges.

BACKGROUND: Limited research has investigated the association between training load and performance of basketball players during games. Little is known about how different indicators of player performance are affected by internal and external loads. The purpose of this study was to determine whether external and internal loads influence basketball players' performance during games. METHOD: This longitudinal study involved 20 professional male basketball players from a single team, classified as first-level athletes by the Chinese Basketball Association. During 34 games, external load was measured as PlayerLoad using micro-sensors, while internal load was assessed using session rating of perceived exertion (sRPE). Player performance was quantified using three metrics: Efficiency, Player Index Rating (PIR), and Plus-Minus (PM). Pearson correlation coefficients were calculated to assess the strength of the relationships between training loads and performance metrics. Linear mixed-effects models were applied to further analyze the influence of internal and external loads on basketball performance. RESULTS: Pearson correlation analysis revealed moderate positive correlations between both sRPE and PlayerLoad with Efficiency and PIR. Specifically, sRPE (r = 0.52) and PlayerLoad (r = 0.54) were both significantly correlated with Efficiency. For PIR, sRPE (r = 0.50) and PlayerLoad (r = 0.56) also demonstrated moderate correlations. These correlations were further substantiated by linear mixed-effects models, which showed that sRPE (ß = 2.21, p < 0.001) and PlayerLoad (ß = 1.87, p = 0.004) had significant independent effects on Efficiency. Similarly, sRPE (ß = 2.15, p < 0.001) and PlayerLoad (ß = 2.36, p < 0.001) significantly predicted PIR. Additionally, a significant interaction effect between PlayerLoad and sRPE was found on Plus-Minus (ß = -2.49, p < 0.001), indicating that the combination of high physical and psychological loads negatively impacted overall team performance. However, the correlation strengths for Plus-Minus were relatively low (sRPE: r = 0.16; PlayerLoad: r = 0.10). CONCLUSION: Both external and internal loads positively contribute to performance, the integration of objective (accelerometry) and subjective (sRPE) measures of load provides a comprehensive understanding of the physiological and psychological demands on athletes, contributing to more effective training regimens and performance optimization.

As interest in eco-friendly work vehicles grows, research on the powertrains of eco-friendly tractors has increased, including research on the development of eco-friendly vehicles (tractors) using hydrogen fuel cell power packs and batteries. However, batteries require a long time to charge and have a short operating time due to their low energy efficiency compared with hydrogen fuel cell power packs. Therefore, recent studies have focused on the development of tractors using hydrogen fuel cell power packs; however, there is a lack of research on powertrain performance analysis considering actual working conditions. To evaluate vehicle performance, an actual load measurement during agricultural operation must be conducted. The objective of this study was to conduct an efficiency analysis of powertrains according to their power source using data measured during agricultural operations. A performance evaluation with respect to efficiency was performed through comparison and an analysis with internal combustion engine tractors of the same level. The specifications of the transmission for hydrogen fuel cell and engine tractors were used in this study. The power loss and efficiency of the transmission were calculated using ISO 14179-1 equations, as shown below. Plow tillage and rotary tillage operations were conducted for data measurement. The measurement system consists of four components. The engine data load measurement was calculated using the vehicle's controller area network (CAN) data, the axle load was measured using an axle torque meter and proximity sensors, and fuel consumption was measured using the sensor installed on the fuel line. The calculated capacities, considering the engine's fuel efficiency for plow and rotary tillage operations, were 131.2 and 175.1 kWh, respectively. The capacity of the required power, considering the powertrain's efficiency for hydrogen fuel cell tractors with respect to plow and rotary tillage operations, was calculated using the efficiency of the motor, inverter, and power pack, and 51.3 and 62.9 kWh were the values obtained, respectively. Considering these factors, the engine exhibited an efficiency of about 47.9% compared with the power pack in the case of plow tillage operations, and the engine exhibited an efficiency of about 29.3% in the case of rotary tillage operations. A hydrogen fuel cell tractor is considered suitable for high-efficiency and eco-friendly vehicles because it can operate on eco-friendly power sources while providing the advantages of a motor.

Maglev vehicles apply the entire vehicle load uniformly onto bridges through levitation forces. In assessing the dynamic characteristics of the maglev train-bridge coupling system, it is reasonable to simplify the distributed levitation force as a concentrated force. This article theoretically derives the analytical response of bridge dynamics under the action of a single constant force and conducts numerical simulations for a moving single constant force and a series of equally spaced constant forces passing over simply supported beams and two-span continuous beams, respectively. The topic of discussion is the response of bridge dynamics when different degrees of force concentration are involved. High-precision displacement and acceleration sensors were utilized to conduct tests on the Shanghai maglev line to verify the accuracy of the simulation results. The results indicate that when simplifying the distributed levitation force into a concentrated force model, a frequency ratio can be used to analyze the conditions for resonance between the train and the bridge and to calculate the critical speed of the train; the levitation distribution force of a high-speed maglev vehicle can be simplified into four groups of concentrated forces based on the number of levitation frames to achieve sufficient accuracy, with the dynamic response of the bridge being close to that under distributed loads.
