Raman spectroscopy has emerged as a crucial mineral analysis technique in planetary surface exploration missions. Nonetheless, the inherently low Raman scattering efficiency of planetary silicate materials makes it challenging to extract enough Raman information. Theoretical and experimental studies of the remote Raman scattering properties of planetary materials are also urgent requirements for future lunar and planetary explorations. Here, Shandong University Remote Raman Spectrometer (SDU-RRS) was developed to demonstrate the feasibility of lunar remote Raman technology and conduct preliminary research on remote Raman scattering properties. SDU-RRS utilizes a pulsed 532 nm laser, a non-focal Cassegrain telescope, a volume phase holographic grating, an intensified charge-coupled device, and the time-gating technique to detect weak-signal silicate minerals. The spectral resolution obtained with atomic emission lamps was <4.91 cm-1, and the wavelength accuracy was <1 cm-1, across the spectral range of 241-2430 cm-1. SDU-RRS can detect natural augite within a feldspar-olivine-augite matrix at a concentration of 20 % at ∼1 m under ambient lighting conditions. A series of experiments were conducted to evaluate the influence of measurement conditions and physical matrix effects on acquired Raman signals, either qualitatively or quantitatively, on geological materials. The study indicates that the transmission of Raman-scattered light conforms to Lambert's cosine law, and a linear correlation exists between Raman intensity and laser power. The study also evaluated the impact of grain size, surface roughness, porosity, and shadow-hiding effects. Reducing grain size decreases Raman intensity and broadens Raman spectra. These characteristics are essential for achieving definitive mineralogical information from granular materials by remote Raman spectroscopy in lunar and planetary explorations.

Subsurface exploration of ice-covered planets and moons presents communications challenges because of the need to communicate through kilometers of ice. The objective of this task is to develop the capability to wirelessly communicate through kilometers of ice and thus complement the potentially failure-prone tethers deployed behind an ice-penetrating probe on Ocean Worlds. In this paper, the preliminary work on the development of wireless deep-ice communication is presented and discussed. The communication test and acoustic attenuation measurements in ice have been made by embedding acoustic transceivers in glacial ice at the Matanuska Glacier, Anchorage, Alaska. Field test results show that acoustic communication is viable through ice, demonstrating the transmission of data and image files in the 13-18 kHz band over 100 m. The results suggest that communication over many kilometers of ice thickness could be feasible by employing reduced transmitting frequencies around 1 kHz, though future work is needed to better constrain the likely acoustic attenuation properties through a refrozen borehole.

The low organic matter content in the hyperarid core of the Atacama Desert, together with abrupt temperature shifts and high ultraviolet radiation at its surface, makes this region one of the best terrestrial analogs of Mars and one of the best scenarios for testing instrumentation devoted to in situ planetary exploration. We have operated remotely and autonomously the SOLID-LDChip (Signs of Life Detector-Life Detector Chip), an antibody microarray-based sensor instrument, as part of a rover payload during the 2019 NASA Atacama Rover Astrobiology Drilling Studies (ARADS) Mars drilling simulation campaign. A robotic arm collected drilled cuttings down to 80 cm depth and loaded SOLID to process and assay them with LDChip for searching for molecular biomarkers. A remote science team received and analyzed telemetry data and LDChip results. The data revealed the presence of microbial markers from Proteobacteria, Acidobacteria, Bacteroidetes, Actinobacteria, Firmicutes, and Cyanobacteria to be relatively more abundant in the middle layer (40-50 cm). In addition, the detection of several proteins from nitrogen metabolism indicates a pivotal role in the system. These findings were corroborated and complemented on "returned samples" to the lab by a comprehensive analysis that included DNA sequencing, metaproteomics, and a metabolic reconstruction of the sampled area. Altogether, the results describe a relatively complex microbial community with members capable of nitrogen fixation and denitrification, sulfur oxidation and reduction, or triggering oxidative stress responses, among other traits. This remote operation demonstrated the high maturity of SOLID-LDChip as a powerful tool for remote in situ life detection for future missions in the Solar System.

The 2020s could be called, with little doubt, the "Mars decade". No other period in space exploration history has experienced such interest in placing orbiters, rovers and landers on the Red Planet. In 2021 alone, the Emirates' first Mars Mission (the Hope orbiter), the Chinese Tianwen-1 mission (orbiter, lander and rover), and NASA's Mars 2020 Perseverance rover reached Mars. The ExoMars mission Rosalind Franklin rover is scheduled for launch in 2022. Beyond that, several other missions are proposed or under development. Among these, MMX to Phobos and the very important Mars Sample Return can be cited. One of the key mission objectives of the Mars 2020 and ExoMars 2022 missions is the detection of traces of potential past or present life. This detection relies to a great extent on the analytical results provided by complementary spectroscopic techniques. The development of these novel instruments has been carried out in step with the analytical study of terrestrial analogue sites and materials, which serve to test the scientific capabilities of spectroscopic prototypes while providing crucial information to better understand the geological processes that could have occurred on Mars. Being directly involved in the development of three of the first Raman spectrometers to be validated for space exploration missions (Mars 2020/SuperCam, ExoMars/RLS and RAX/MMX), the present review summarizes some of the most relevant spectroscopy-based analyses of terrestrial analogues carried out over the past two decades. Therefore, the present work describes the analytical results gathered from the study of some of the most distinctive terrestrial analogues of Martian geological contexts, as well as the lessons learned mainly from ExoMars mission simulations conducted at representative analogue sites. Learning from the experience gained in the described studies, a general overview of the scientific outcome expected from the spectroscopic system developed for current and forthcoming planetary missions is provided.

The Mars 2020 Perseverance rover landed on February 18, 2021, and has started ground operations. The ExoMars Rosalind Franklin rover will touch down on June 10, 2023. Perseverance will be the first-ever Mars sample caching mission-a first step in sample return to Earth. SuperCam and Scanning Habitable Environments with Raman & Luminescence for Organics & Chemicals (SHERLOC) on Perseverance, and Raman Laser Spectrometer (RLS) on Rosalind Franklin, will comprise the first ever in situ planetary mission Raman spectroscopy instruments to identify rocks, minerals, and potential organic biosignatures on Mars' surface. There are many challenges associated when using Raman instruments and the optimization and quantitative analysis of resulting data. To understand how best to overcome them, we performed a comprehensive Raman analysis campaign on CanMars, a Mars sample caching rover analog mission undertaken in Hanksville, Utah, USA, in 2016. The Hanksville region presents many similarities to Oxia Planum's past habitable conditions, including liquid water, flocculent, and elemental compounds (such as clays), catalysts, substrates, and energy/food sources for life. We sampled and conducted a complete band analysis of Raman spectra as mission validation analysis with the RLS ExoMars Simulator or RLS Sim, a breadboard setup representative of the ExoMars RLS instrument. RLS Sim emulates the operational behavior of RLS on the Rosalind Franklin rover. Given the high fidelity of the Mars analog site and the RLS Sim, the results presented here may provide important information useful for guiding in situ analysis and sample triage for caching relevant for the Perseverance and Rosalind Franklin missions. By using the RLS Sim on CanMars samples, our measurements detected oxides, sulfates, nitrates, carbonates, feldspars, and carotenoids, many with a higher degree of sensitivity than past results. Future work with the RLS Sim will aim to continue developing and improving the capability of the RLS system in the future ExoMars mission.

A remote Raman prototype with a function of excitation energy adjusting for the purpose of obtaining a Raman signal with good signal-to-noise ratio (SNR), saving power consumption, and possibly avoiding destroying a target by high energy pulses, which may have applications for Chinese planetary explorations, has been setup and demonstrated for detecting different minerals. The system consists of a spectrograph equipped with a thermoelectrically cooled charge-coupled device (CCD) detector, a telescope with 150 mm diameter and 1500 mm focus length, and a compact 1064 nm Nd:YAG Q-switched laser with an electrical adjusted pulse energy from 0 to 200 mJ/pulse. A KTP crystal was used for second harmonic generation in a 1064 nm laser to generate a 532 nm laser, which is the source of Raman scatting. Different laser pulse energies and integration time were used to obtain distinguishable remote Raman spectra of various samples. Results show that observed remote Raman spectra at a distance of 4 m enable us to identify silicates, carbonates, sulfates, perchlorates, water/water ice, and organics that have been found or may exist on extraterrestrial planets. Detailed Raman spectral assignments of the measured planetary materials and the feasible applications of remote Raman system for planetary explorations are discussed.

The analysis of robotic systems (e.g. landers and rovers) involved in sampling operations on planetary bodies is crucial to ensure mission success, since those operations generate forces that could affect the stability of the robotic system. This paper presents MISTRAL (MultIdisciplinary deSign Tool for Robotic sAmpLing), a novel tool conceived for trade space exploration during early conceptual and preliminary design phases, where a rapid and broad evaluation is required for a very high number of configurations and boundary conditions. The tool rapidly determines the preliminary design envelope of a sampling apparatus to guarantee the stability condition of the whole robotic system. The tool implements a three-dimensional analytical model capable to reproduce several scenarios, being able to accept various input parameters, including the physical and geometrical characteristics of the robotic system, the properties related to the environment and the characteristics related to the sampling system. This feature can be exploited to infer multidisciplinary high-level requirements concerning several other elements of the investigated system, such as robotic arms and footpads. The presented research focuses on the application of MISTRAL to landers. The structure of the tool and the analysis model are presented. Results from the application of the tool to real mission data from NASA's Phoenix Mars lander are included. Moreover, the tool was adopted for the definition of the high-level requirements of the lander for a potential future mission to the surface of Saturn's moon Enceladus, currently under investigation at NASA Jet Propulsion Laboratory. This case study was included to demonstrate the tool's capabilities. MISTRAL represents a comprehensive, versatile, and powerful tool providing guidelines for cognizant decisions in the early and most crucial stages of the design of robotic systems involved in sampling operations on planetary bodies.

Enceladus, an icy moon of Saturn, is a compelling destination for a probe seeking biosignatures of extraterrestrial life because its subsurface ocean exhibits significant organic chemistry that is directly accessible by sampling cryovolcanic plumes. State-of-the-art organic chemical analysis instruments can perform valuable science measurements at Enceladus provided they receive sufficient plume material in a fly-by or orbiter plume transit. To explore the feasibility of plume sampling, we performed light gas gun experiments impacting micrometer-sized ice particles containing a fluorescent dye biosignature simulant into a variety of soft metal capture surfaces at velocities from 800 m ⋅ s-1 up to 3 km ⋅ s-1 Quantitative fluorescence microscopy of the capture surfaces demonstrates organic capture efficiencies of up to 80 to 90% for isolated impact craters and of at least 17% on average on indium and aluminum capture surfaces at velocities up to 2.2 km ⋅ s-1 Our results reveal the relationships between impact velocity, particle size, capture surface, and capture efficiency for a variety of possible plume transit scenarios. Combined with sensitive microfluidic chemical analysis instruments, we predict that our capture system can be used to detect organic molecules in Enceladus plume ice at the 1 nM level-a sensitivity thought to be meaningful and informative for probing habitability and biosignatures.

Mass spectrometry (MS) has played a remarkable role in exploring the chemical make-up of our solar system. In situ probes were historically developed to analyze inorganic/elemental compositions while leveraging native ions or harsh ionization methods to aid in exploring astrophysics applications (e.g., heliophysics). The part played by MS is demonstrated in a majority of scientific payloads focused on exploration, particularly at the turn of the century with missions including Cassini-Huygens, Rosetta, and now Mars Science Laboratory. Plasma mass spectrometers have grown more sophisticated to interrogate fundamental inorganic analysis (e.g., solar wind and magnetospheres) including both native ions and neutrals. Cosmic dust floating in-between and orbiting planetary bodies has been targeted by unique sampling via impact ionization. More complex systems rely on landed planetary instrumentation with lessons learned from pioneering missions in the 1970s and 1980s to near neighbors Mars and Venus. Modern probes have expanded applicable target chemicals by recognizing the needs to provide for molecular analyses, extended mass range, and high resolution to provide unequivocal detection and identification. Notably, as the field surrounding astrobiology has gained momentum, so has the in situ detection of complex molecular chemistry including the chemical evolution of organic molecules. Mission context often includes long term timelines from spacecraft launch to arrival and additionally the diverse target environments across various planets. Therefore, customized experimental designs for space MS have been born of necessity. To this point, the development of MS instrumentation on Earth has now far outpaced development for experiments in space. Therefore, exciting developments lie ahead among various international space agencies conducting current and future mission planning with increasingly enhanced instrumentation. For instance, near-neighbor Mars has entertained considerable attention with complex MS instrumentation with laser desorption ionization aboard the Mars Organic Molecule Analyzer instrument. To study comets, the Rosetta mission employs a secondary ionization mechanism. Meanwhile, the various moons of Jupiter and Saturn have intriguing surface and subsurface properties that warrant more advanced analyzer systems. Instrumentation design will continue to evolve as requirements develop and this review serves as a reflection of the contribution of in situ MS to space exploration in the past 20 years and the anticipated contribution yet to come. © 2020 John Wiley & Sons Ltd. Raman spectroscopy is a technique that can detect and characterize a range of molecular compounds such as water, water ice, water-bearing minerals, and organics of particular interest to planetary science. The detection and characterization of these molecular compounds, which are indications of habitability on planetary bodies, have become an important goal for planetary exploration missions spanning the solar system. Using a compact portable remote Raman system consisting of a 532 nm neodymium-doped yttrium aluminum garnet- (Nd:YAG-) pulsed laser, a 3-in. (7.62 cm) diameter mirror lens and a compact spectrograph with a miniature intensified charge coupled device (mini-ICCD), we were able to detect water (H2O), water ice (H2O-ice), CO2-ice, hydrous minerals, organics, nitrates, and an amino acid from a remote distance of 122 m in natural lighting conditions. To the best of our knowledge, this is the longest remote Raman detection using a compact system. The development of this uniquely compact portable remote Raman system is applicable to a range of solar system exploration missions including stationary landers for ocean worlds and lunar exploration, as they provide unambiguous detection of compounds indicative of life as well as resources necessary for further human exploration.

The Earth's moon is currently an object of interest of many space agencies for unmanned robotic missions within this decade. Besides future prospects for building lunar gateways as support to human space flight, the Moon is an attractive location for scientific purposes. Not only will its study give insight on the foundations of the Solar System but also its location, uncontaminated by the Earth's ionosphere, represents a vantage point for the observation of the Sun and planetary bodies outside the Solar System. Lunar exploration has been traditionally conducted by means of single-agent robotic assets, which is a limiting factor for the return of scientific missions. The German Aerospace Center (DLR) is developing fundamental technologies towards increased autonomy of robotic explorers to fulfil more complex mission tasks through cooperation. This paper presents an overview of past, present and future activities of DLR towards highly autonomous systems for scientific missions targeting the Moon and other planetary bodies. The heritage from the Mobile Asteroid Scout (MASCOT), developed jointly by DLR and CNES and deployed on asteroid Ryugu on 3 October 2018 from JAXA's Hayabusa2 spacecraft, inspired the development of novel core technologies towards higher efficiency in planetary exploration. Together with the lessons learnt from the ROBEX project (2012-2017), where a mobile robot autonomously deployed seismic sensors at a Moon analogue site, this experience is shaping the future steps towards more complex space missions. They include the development of a mobile rover for JAXA's Martian Moons eXploration (MMX) in 2024 as well as demonstrations of novel multi-robot technologies at a Moon analogue site on the volcano Mt Etna in the ARCHES project. Within ARCHES, a demonstration mission is planned from the 14 June to 10 July 2021,1 during which heterogeneous teams of robots will autonomously conduct geological and mineralogical analysis experiments and deploy an array of low-frequency antennas to measure Jovian and solar bursts. The Mars 2020 Perseverance rover is equipped with a next-generation engineering camera imaging system that represents an upgrade over previous Mars rover missions. These upgrades will improve the operational capabilities of the rover with an emphasis on drive planning, robotic arm operation, instrument operations, sample caching activities, and documentation of key events during entry, descent, and landing (EDL). There are a total of 16 cameras in the Perseverance engineering imaging system, including 9 cameras for surface operations and 7 cameras for EDL documentation. There are 3 types of cameras designed for surface operations: Navigation cameras (Navcams, quantity 2), Hazard Avoidance Cameras (Hazcams, quantity 6), and Cachecam (quantity 1). The Navcams will acquire color stereo images of the surface with a 96 ∘ × 73 ∘ field of view at 0.33 mrad/pixel. The Hazcams will acquire color stereo images of the surface with a 136 ∘ × 102 ∘ at 0.46 mrad/pixel. The Cachecam, a new camera type, will acquire images of Martian material inside the sample tubes during caching operations at a spatial scale of 12.5 microns/pixel. There are 5 types of EDL documentation cameras: The Parachute Uplook Cameras (PUCs, quantity 3), the Descent stage Downlook Camera (DDC, quantity 1), the Rover Uplook Camera (RUC, quantity 1), the Rover Descent Camera (RDC, quantity 1), and the Lander Vision System (LVS) Camera (LCAM, quantity 1). The PUCs are mounted on the parachute support structure and will acquire video of the parachute deployment event as part of a system to characterize parachute performance. The DDC is attached to the descent stage and pointed downward, it will characterize vehicle dynamics by capturing video of the rover as it descends from the skycrane. The rover-mounted RUC, attached to the rover and looking upward, will capture similar video of the skycrane from the vantage point of the rover and will also acquire video of the descent stage flyaway event. The RDC, attached to the rover and looking downward, will document plume dynamics by imaging the Martian surface before, during, and after rover touchdown. The LCAM, mounted to the bottom of the rover chassis and pointed downward, will acquire 90 ∘ × 90 ∘ FOV images during the parachute descent phase of EDL as input to an onboard map localization by the Lander Vision System (LVS). The rover also carries a microphone, mounted externally on the rover chassis, to capture acoustic signatures during and after EDL. The Perseverance rover launched from Earth on July 30th, 2020, and touchdown on Mars is scheduled for February 18th, 2021.

Between February 1 and 28, 2018, the Austrian Space Forum, in cooperation with the Oman Astronomical Society and research teams from 25 nations, conducted the AMADEE-18 mission, a human-robotic Mars expedition simulation in the Dhofar region in the Sultanate of Oman. As a part of the AMADEE-18 simulated Mars human exploration mission, the Remote Science Support team performed analyses of the Dhofar area (Oman) in an effort to characterize the region as a potential Mars analog site. The main motivation of this research was to study and register selected samples collected by analog astronauts during the AMADEE-18 mission with laboratory analytical methods and techniques comparable with those that are likely to be used on Mars in the future. The 25 samples representing unconsolidated sediments obtained during the simulated mission were studied by using optical microscopy, Raman spectroscopy, X-ray diffraction, laser-induced breakdown spectroscopy, and laser-induced fluorescence spectroscopy. The principal results show the existence of minerals and alteration processes related to volcanism, hydrothermalism, and weathering. The analogy between the Dhofar region and the Eridana Basin region of Mars is clearly noticeable, particularly as an analog for secondary minerals formed in a hydrothermal seafloor volcanic-sedimentary environment. The synergy between the techniques used in the present work provides a solid basis for the geochemical analyses and organic detection in the context of future human-robotic Mars expeditions. AMADEE-18 has been a prime test bed for geoscientific workflows with astrobiological relevance and has provided valuable insights for future space missions.

An essential characteristic that an exploration robot must possess is to be autonomous. This is necessary because it will usually do its task in remote or hard-to-reach places. One of the primary elements of a navigation system is the information that can be acquired by the sensors of the environment in which it will operate. For this reason, an algorithm based on convolutional neural networks is proposed for the detection of rocks in environments similar to Mars. The methodology proposed here is based on the use of a Single-Shot-Detector (SSD) network architecture, which has been modified to evaluate the performance. The main contribution of this study is to provide an alternative methodology to detect rocks in planetary images because most of the previous works only focus on classification problems and used handmade feature vectors.

Metabolt is a portable soil incubator to characterize the metabolic activity of microbial ecosystems in soils. It measures the electrical conductivity, the redox potential, and the concentration of certain metabolism-related gases in the headspace just above a given sample of regolith. In its current design, the overall weight of Metabolt, including the soils (250 g), is 1.9 kg with a maximum power consumption of 1.5 W. Metabolt has been designed to monitor the activity of the soil microbiome for Earth and space applications. In particular, it can be used to monitor the health of soils, the atmospheric-regolith fixation, and release of gaseous species such as N2, H2O, CO2, O2, N2O, NH3, etc., that affect the Earth climate and atmospheric chemistry. It may be used to detect and monitor life signatures in soils, treated or untreated, as well as in controlled environments like greenhouse facilities in space, laboratory research environments like anaerobic chambers, or simulating facilities with different atmospheres and pressures. To illustrate its operation, we tested the instrument with sub-arctic soil samples at Earth environmental conditions under three different conditions: (i) no treatment (unperturbed); (ii) sterilized soil: after heating at 125 °C for 35.4 h (thermal stress); (iii) stressed soil: after adding 25% CaCl2 brine (osmotic stress); with and without addition of 0.5% glucose solution (for control). All the samples showed some distinguishable metabolic response, however there was a time delay on its appearance which depends on the treatment applied to the samples: 80 h for thermal stress without glucose, 59 h with glucose; 36 h for osmotic stress with glucose and no significant reactivation in the pure water case. This instrument shows that, over time, there is a clear observable footprint of the electrochemical signatures in the redox profile which is complementary to the gaseous footprint of the metabolic activity through respiration.

RESUMO

The objective of this work is to present the first analytical and experimental results obtained with a 3D heat flux sensor for planetary regolith. The proposed structure, a sphere divided in four sectors, is sensible to heat flow magnitude and angle. Each sector includes a platinum resistor that is used both to sense its temperature and provide heating power. By operating the sectors at constant temperature, the sensor gives a response that is proportional to the heat flux vector in the regolith. The response of the sensor is therefore independent of the thermal conductivity of the regolith. A complete analytical solution of the response of the sensor is presented. The sensor may be used to provide information on the instantaneous local thermal environment surrounding a lander in planetary exploration or in small bodies like asteroids. To the best knowledge of the authors, this is the first sensor capable of measuring local 3D heat flux.

Analog research of human or combined human and robotic missions is an established tool to explore the workflows, instruments, risks, and challenges of future planetary surface missions in a representative terrestrial environment. Analog missions that emulate selected aspects of such expeditions have risen in number, expanded their range of disciplines covered, and seen a significant increase in their operational and programmatic impact on mission planning. We propose a method to compare analog missions across agencies, disciplines, and complexities/fidelities to improve scientific output and mission safety and maximize effectiveness and efficiency. This algorithm measures mission performance, provides a tool for an objective postmission evaluation, and catalyzes programmatic progress. It does not evaluate individual sites or instruments but focuses at mission level. By applying the algorithm to several missions, we compare the missions' performance for benchmarking purposes. Methodically, a combination of objective data sets and questionnaires is used to evaluate three areas: two sections of closed and quantitative questions and a third section dedicated to the level or representativeness of the test site. By using a weighted metric, the complexity and fidelity of a mission are compared with reference missions, which yield strengths and weaknesses in mission planning.

Sulfate and iron oxide deposits in Río Tinto (Southwestern Spain) are a terrestrial analog of early martian hematite-rich regions. Understanding the distribution and drivers of microbial life in iron-rich environments can give critical clues on how to search for biosignatures on Mars. We simulated a robotic drilling mission searching for signs of life in the martian subsurface, by using a 1m-class planetary prototype drill mounted on a full-scale mockup of NASA's Phoenix and InSight lander platforms. We demonstrated fully automated and aseptic drilling on iron and sulfur rich sediments at the Río Tinto riverbanks, and sample transfer and delivery to sterile containers and analytical instruments. As a ground-truth study, samples were analyzed in the field with the life detector chip immunoassay for searching microbial markers, and then in the laboratory with X-ray diffraction to determine mineralogy, gas chromatography/mass spectrometry for lipid composition, isotope-ratio mass spectrometry for isotopic ratios, and 16S/18S rRNA genes sequencing for biodiversity. A ubiquitous presence of microbial biomarkers distributed along the 1m-depth subsurface was influenced by the local mineralogy and geochemistry. The spatial heterogeneity of abiotic variables at local scale highlights the importance of considering drill replicates in future martian drilling missions. The multi-analytical approach provided proof of concept that molecular biomarkers varying in compositional nature, preservation potential, and taxonomic specificity can be recovered from shallow drilling on iron-rich Mars analogues by using an automated life-detection lander prototype, such as the one proposed for NASA's IceBreaker mission proposal.

Perchlorate anions are produced by chemical industries and are important contaminants in certain natural ecosystems. Perchlorate also occurs in some natural and uncontaminated environments such as the Atacama Desert, the high Arctic or the Antarctic Dry Valleys, and is especially abundant on the surface of Mars. As some bacterial strains are capable of using perchlorate as an electron acceptor under anaerobic conditions, their detection is relevant for environmental monitoring on Earth as well as for the search for life on Mars. We have developed an antibody microarray with 20 polyclonal antibodies to detect perchlorate-reducing bacteria (PRB) strains and two crucial and highly conserved enzymes involved in perchlorate respiration: perchlorate reductase and chlorite dismutase. We determined the cross-reactivity, the working concentration, and the limit of detection of each antibody individually and in a multiplex format by Fluorescent Sandwich Microarray Immunoassay. Although most of them exhibited relatively high sensitivity and specificity, we applied a deconvolution method based on graph theory to discriminate between specific signals and cross-reactions from related microorganisms. We validated the system by analyzing multiple bacterial isolates, crude extracts from contaminated reactors and salt-rich natural samples from the high Arctic. The PRB detecting chip (PRBCHIP) allowed us to detect and classify environmental isolates as well as to detect similar strains by using crude extracts obtained from 0.5 g even from soils with low organic-matter levels (<103 cells/g of soil). Our results demonstrated that PRBCHIP is a valuable tool for sensitive and reliable detection of perchlorate-reducing bacteria for research purposes, environmental monitoring and planetary exploration.

This paper presents a case study of microbe transportation in the Mars-satellites system. We examined the spatial distribution of potential impact-transported microbes on the Martian moons using impact physics by following a companion study (Fujita et al., in this issue). We used sterilization data from the precede studies (Patel et al., 2018; Summers, 2017). We considered that the microbes came mainly from the Zunil crater on Mars, which was formed during 1.0-0.1 Ma. We found that 70-80% of the microbes are likely to be dispersed all over the moon surface and are rapidly sterilized due to solar and galactic cosmic radiation except for those microbes within a thick ejecta deposit produced by natural meteoroids. The other 20-30% might be shielded from radiation by thick regolith layers that formed at collapsed layers in craters produced by Mars rock impacts. The total number of potentially surviving microbes at the thick ejecta deposits is estimated to be 3-4 orders of magnitude lower than at the Mars rock craters. The microbe concentration is irregular in the horizontal direction due to Mars rock bombardment and is largely depth-dependent due to the radiation sterilization. The surviving fraction of transported microbes would be only ∼1 ppm on Phobos and ∼100 ppm on Deimos, suggesting that the transport processes and radiation severely affect microbe survival. The microbe sampling probability from the Martian moons was also investigatesd. We suggest that sample return missions from the Martian moons are classified into Unrestricted Earth-Return missions for 30 g samples and 10 cm depth sampling, even in our conservative scenario. We also conducted a full statistical analysis pertaining to sampling the regolith of Phobos to include the effects of uncertainties in input parameters on the sampling probability. The most likely probability of microbial contamination for return samples is estimated to be two orders of magnitude lower than the 10-6 criterion defined by the planetary protection policy of the Committee on Space Research (COSPAR).
