RESUMO

The copepod family Shiinoidae Cressey, 1975 currently comprises nine species of teleost parasites with unusual morphology and a unique attachment mechanism. Female shiinoids possess greatly enlarged antennae that oppose a rostrum, an elongate outgrowth of cuticle that originates between the antennules. The antennae form a moveable clasp against the rostrum which they use to attach to their host. In this study, we use micro-computed tomography (microCT) to examine specimens of Shiinoa inauris Cressey, 1975 in situ attached to host tissue in order to characterize the functional morphology and specific muscles involved in this novel mode of attachment and to resolve uncertainty regarding the segmental composition of the regions of the body. We review the host and locality data for all reports of shiinoids, revise the generic diagnoses for both constituent genera Shiinoa Kabata, 1968 and Parashiinoa West, 1986, transfer Shiinoa rostrata Balaraman, Prabha & Pillai, 1984 to Parashiinoa as Parashiinoa rostrata (Balaraman, Prabha & Pillai, 1984) n. comb., and present keys to the females and males of both genera.

In order to study the trophic level of small tuna species and their contribution to the carbon flow in pelagic food webs, an analysis of carbon and nitrogen stable isotopes was carried out. The investigation was focused on four small tuna species (Auxis rochei, Auxis thazard, Euthynnus alletteratus and Sarda sarda) commonly harvested in the northeast Atlantic Ocean. The isotope analysis showed how the results for S. sarda are different from the rest of the species analysed, with a higher trophic level, similar to other major tuna species. The greatest niche overlap in δ13C and δ15N occurs among A. rochei, A. thazard and E. alletteratus. Auxis rochei and E. alletteratus showed a size-dependent variability in δ15N, and in δ13C for S. sarda. The small tuna S. sarda exhibits the highest migration rates among various geographical areas in comparison to other small pelagic tunas, and the seasonal variability of isotope values in the area studied can be attributed to the incorporation of larger individuals with a higher lipid content. The results of this work provide new information on the ecological role played by small tuna in food webs, which is more complex and varied than currently thought. This knowledge is essential for a more effective management of fisheries.

The Serra Spanish mackerel, Scomberomorus brasiliensis, exemplifies a predator resource historically targeted by artisanal fisheries on the Brazilian coast. The present work analyzes landings in three periods within a 50-year timescale on the Parana coast, Southern Brazil, aiming to evaluate whether historical production has supposedly declined. Simultaneously, the diet was analyzed to confirm carnivorous habits and evaluate the trophic level in this region. Surprisingly, the results show that from the 1970's to 2019 Serra Spanish mackerel production grew relatively to other resources, as well as in individual values. The trophic level was calculated as 4.238, similar to other Scomberomorus species, consisting of a case where landings increase over time, despite the high trophic level and large body size of the resource. The results agree with a recent global assessment that has demystified a necessary correlation between high trophic level and overexploitation, but possible factors acting on the present findings are discussed.


Resumo Na pesca, o fenômeno fishing down food webs, ou 'pescando teias tróficas abaixo', expressa a redução do nível trófico na composição das capturas, e tem origem tradicionalmente atribuída à sobrepesca. Sob intenso extrativismo, a resiliência das espécies carnívoras, predadoras de topo, é particularmente afetada, sendo seus estoques os primeiros a entrarem em depleção. A cavala, ou serra, ou sororoca, Scomberomorus brasiliensis, é exemplo de recurso predador historicamente alvo da pesca artesanal, de pequena escala, na costa brasileira. O presente trabalho analisa os valores de desembarque do recurso em três períodos ao longo de 50 anos na costa paranaense, sul do Brasil, com objetivo de avaliar se sua produção tem efetivamente diminuído. Ainda, estuda-se a dieta da espécie na região, para confirmar seus hábitos carnívoros e avaliar o nível trófico na região. Os resultados mostram que, surpreendentemente, dos anos 1970 para 2019 a produção de S. brasiliensis tem aumentado em valores absolutos, bem como relativamente à de outros recursos, em que pese o nível trófico 4,238, similar a outras espécies do gênero. Constata-se que a pesca da cavala na região de estudo constitui um caso de aumento nos desembarques a despeito de elevado nível trófico e grande tamanho corpóreo. Os resultados concordam com recente levantamento em nível mundial, que nega existir correlação necessária entre sobrepesca e nível trófico alto, mas fatores que podem explicar os resultados do presente trabalho são discutidos.

One of the most promising aquaculture species is the Atlantic bluefin tuna (Thunnus thynnus) with high market value; disease control is crucial to prevent and reduce mortality and monetary losses. Microsporidia (Fungi) are a potential source of damage to bluefin tuna aquaculture. A new microsporidian species is described from farmed bluefin tunas from the Spanish Mediterranean. This new pathogen is described in a juvenile associated with a highly severe pathology of the visceral cavity. Whitish xenomas from this microsporidian species were mostly located at the caecal mass and ranged from 0.2 to 7.5 mm. Light and transmission electron microscopy of the spores revealed mature spores with an average size of 2.2 × 3.9 µm in size and a polar filament with 13-14 coils arranged in one single layer. Phylogenetic analysis clustered this species with the Glugea spp. clade. The morphological characteristics and molecular comparison confirm that this is a novel microsporidian species, Glugea thunni. The direct life-cycle and the severe pathologies observed makes this parasite a hard risk for bluefin tuna cultures.

Scombridae fish (tuna, bonito and mackerel) have significant ecological and economic values. They are very appreciated by consumers worldwide for their high-quality flesh and for their high nutritional value. However, consumption of Scombridae fish is potentially hazardous. Indeed, several cases of infections and toxicity linked to the consumption of Scombridae fish as raw, or processed food products have been reported worldwide. In this review, we presented the most common health risks associated with Scombridae fish consumption. Diseases associated with the consumption of these fish are generally infectious or toxic and are caused by biological hazards, such as bacteria, viruses, parasites or chemicals hazards that enter the body through contaminated fish (polycyclic aromatic hydrocarbons, histamine) or by physical contaminants, such as heavy metals. The risks of contamination exist throughout the food chain, from primary production to the preparation of products for consumption.

Aquaculture is an increasingly important food resource, but its sustainability is often limited by disease. In Scombridae fishes, puffy snout syndrome (PSS) is a debilitating condition where tumor-like collagenous growths form around the eyes, nares, and mandibles which impair vision and feeding and frequently lead to mortality. While PSS is considered an infectious or metabolic disease, no disease agents or promoters have been identified. Here, we used electron microscopy (EM) to describe the cellular pathology and search for etiological agents of PSS in Pacific mackerel Scomber japonicus, the first use of this approach for PSS. We examined aquaculture specimens across a range of apparent PSS severity, comparing the results to both wild and aquaculture asymptomatic mackerel. EM imagery consistently revealed viral-like particles in PSS samples, as well as the uniform absence of bacteria, protists, fungi, and other multicellular parasites. In addition to viral-like particles, symptomatic fish had a higher mean percentage of swollen and disintegrating mitochondria than both asymptomatic aquaculture and wild mackerel. This suggests that degraded mitochondria may be related to PSS and could be important to further understanding the origin, promoters, and prevention of PSS. This study serves as a first step in identifying the etiological agents of PSS.

Polybrominated diphenyl ethers (PBDEs) and their methoxylated analogues (MeO-BDEs) are widely distributed in the environment. The main concern about the presence of PBDEs and MeO-BDEs in fish is due to their potential endocrine disruption effects in the specimens, and their potential risk to the health of human consumers. Considering these concerns, the goal of this study was to investigate the occurrence of PBDEs and MeO-BDEs in muscle tissues of blue shark (BSH), Prionace glauca, and yellowfin tuna (YFT), Thunnus albacares, caught in the Equatorial Atlantic Ocean (EAO), North-eastern Brazilian waters, and to evaluate the potential risk of human exposure by consumption. Muscle tissues of YFT and BSH were extracted using a Soxhlet apparatus and an Accelerated Solvent Extractor (ASE), respectively. PBDEs and MeO-BDEs were analysed by GC-NCI-MS. The complete mitochondrial genome of the doubled-lined mackerel, Grammatorcynus bilineatus, was determined by the combination of high-throughput sequencing (HTS) and Sanger sequencing. The constructed mitochondrial genome of G. bilineatus was 16,537 bp in length, which harbors a canonical 37 genes (13 proteins, two ribosomal RNAs, and 22 tRNAs) and two non-coding regions (origin of light-strand replication (OL) and the D-loop control region). Among 38 genes, nine were encoded on its light strand (L), while the other 28 were on its heavy strand (H). Besides COX1 (GTG) and ATP6 (CTG), the other eleven protein-coding genes (PCGs) begin with a typical start codon (ATG). The phylogenetic tree showed that G. bilineatus was not clustered with the other species in the Scombridae, forming a clade for Grammatorcynus. The genetic information of G.bilineatus will provide useful information for the scientific management and conservation of the species in the genus.

Feeding ecology of Atlantic bluefin tuna (ABFT, Thunnus thynnus), from the Gulf of Saint Lawrence, Canada, was assessed by stomach content and stable isotope analyses. Stomachs were collected from 161 individuals averaging 249 ± 20 cm in curved fork length and 248 ± 64 in round weight. The fish were caught by commercial fishing boats using rod and reel gear during fall 2015-2017. Liver and muscle samples were taken from 133 fish for stable isotope analysis. Atlantic mackerel (Scomber scombrus) was the most abundant prey item in 2015 (%Alimentary index, %AI = 88.94), whereas in 2016 and 2017 the ABFT diet was mainly composed of Atlantic herring (Clupea harengus) (%AI = 56.73 and %AI = 77.23, respectively). PERMANOVA analysis revealed significant differences in the dietary composition between years. δ15N values varied between 12.3 and 15.4‰ for muscle and between 11.1 and 14.6‰ for liver. δ13C values ranged from -19.9 to -16.5‰ for muscle and from -20.7 to -17.3‰ for liver. PERMANOVA analysis also detected inter-annual isotopic differences in both muscle and liver samples (p < 0.05). Daily rations (DR) were similar in 2015 and 2016 (DR = 1.02 ± 0.32 and DR = 1.10 ± 0.21, respectively), but higher in 2017 (DR = 1.73 ± 0.54).

Many large predatory fishes are in decline and tuna sustainability is high on the global agenda. Slender tuna (SLT), Allothunnus fallai, is data-poor and a rare contemporary example of a globally unexploited temperate tuna. This study analysed 214 otoliths for age and growth of fish collected in the South Atlantic. Observed ages varied between 9 and 42 years for a size range of 68-90 cm fork length. We reveal important life history data for SLT before exploitation and underline the relevance of data-poor stocks in understanding wider questions for exploited tuna.

Globalization of seafood product marketing caused the increase of request of an effective fish traceability that enhances the consumer confidence in food safety. In this study, an integrated multi analytical approach based on two different and independent analytical techniques (carbon and nitrogen stable isotopes and fatty acids analysis) was applied in order to identify different fish species and trace their geographical provenience. The investigation was focused on four species (Thunnus thynnus, Thunnus alalunga, Auxis rochei and Scomber scombrus) belonging to the Scombridae family. The DNA barcoding method confirmed genus and species for S. scombrus and A. rochei, but only genus for T. alalunga and T. thynnus. Carbon and nitrogen stable isotopes results evidenced different fish diets and trophic positions, whereas fatty acids analysis displayed that the unsaturated prevailed (∼60 %) over the saturated compounds with a variation among the species and the geographical area in particular for docosahexaenoic and eicosapentaenoic acids percentage. The principal component analysis applied to stable isotopes and fatty acids evidenced a good discrimination among species and their geographical catching area. This multi-disciplinary analytical approach could represent a promising tool to identify the commercial fish and trace their origin in order to guarantee the health of consumers.

The Strait of Gibraltar (SoG) houses a large diversity of marine species including large pelagic animals that use this area as permanent or temporary habitat. In particular, the SoG is home to a population of adult and sub-adult Atlantic bluefin tuna (Thunnus thynnus) that occurs there for most of the year. To gain further insight into the life history of this ABFT population, seasonal feeding patterns were investigated combining stomach content analysis (SCA) and stable isotopes analysis (SIA) of muscle and liver tissues. Our results reveal seasonal variations in feeding patterns, which are evidenced by a significant shift in the diet composition and the highest food consumption rates that take place in spring, prior to the spawning season. These changes in feeding habits are consistent with the varying energetic needs along the species' annual biological cycle. The present observations confirm the SoG as an important foraging ground for ABFT resident populations and may have further implications for the management of the resource.

Chronic Kiss1 administration strongly promotes gonadal development in immature chub mackerel (cm) (Scomber japonicus). Here, we performed an Alanine scanning (Ala-scanning) of Kiss1 to determine its key residues. Additionally, we examined functional peptides from 16 Scombridae species to develop maturation-inducing super-analogs that can be used universally in Scombridae species. In the Ala-scanning of Kiss1-15 (QDMSSYNFNSFGLRY), substitution of Gln1 and Asp2 did not affect agonistic activity. This suggests that peptides could be downsized. Furthermore, it is possible that Phe8 can be substituted by unnatural amino acids that are difficult to degrade. In molecular cloning, only Scomber showed a 16-residue form as a putative mature peptide. The other genera, did not have a His residue at the N-terminal, which indicated that the functional peptide was 15 residues and the second and third residues from the N-terminal showed variation between interspecies. Next, we examined the binding affinity of various synthetic Kiss1 core peptides in Scombridae interspecies using an SRE-Luc reporter system. We cloned Kiss1 receptors (KissR1) from bluefin tuna (bft) (Thunnus orientalis) and Japanese Spanish mackerel (jsm) (Scomberomorus niphonius) for the first time. In binding affinity with cmKissR1, bftKissR1, and jsmKissR1, the species specificity of the second residue from the N-terminus in each ligand could be ignored, but the difference in the third residue strongly affected receptor binding. Scombridae species possess the same Kiss1 system but the structure of the functional peptide might be species-specific.

Foraging habits of skipjack tuna, SKJ (Katsuwonus pelamis), were investigated in three Spanish marine regions (Balearic Sea, Alboran Sea and Gulf of Cadiz) using stomach content (SCA), stable isotope (SIA), and fatty acid (FA) analyses. The three methodological approaches yielded significant differences among locations. All the studied areas appear to serve as important foraging grounds for SKJ, but the diet composition significantly varied among them. The predominant prey species in the respective locations were the North Atlantic krill, Meganyctiphanes norvegica, in the Balearic Sea, the flyingfish, Exocœtus volitans, in the Alboran Sea, and the anchovy, Engraulis encrasicolus, in the Gulf of Cadiz. Regional differences were also found in δ13C and δ15N values; furthermore, the analysis of standard ellipse areas (SEAc) based on isotopic data showed that the broadest niche corresponded to SKJ from the Gulf of Cadiz, with no significant isotopic niche overlap between areas. Consistent with the SCA and stable isotope results, the FA composition varied among areas. Docosahexaenoic acid (DHA, 22:6n-3) was the most abundant FA in the SKJ muscle, showing its highest values in samples from the Gulf of Cadiz; this FA was the most effective in segregating SKJ geographical groups. In conclusion, this study provides evidence that SKJ in eastern Atlantic and western Mediterranean waters can easily adapt their diet to local prey availability. Furthermore, stable isotope signatures and FA profiles of muscle tissue prove to be reliable trophic markers that allow separation of populations occurring in different marine areas.

Measuring the demographic parameters of exploited populations is central to predicting their vulnerability and extinction risk. However, current rates of population decline and species loss greatly outpace our ability to empirically monitor all populations that are potentially threatened.The scale of this problem cannot be addressed through additional data collection alone, and therefore it is a common practice to conduct population assessments based on surrogate data collected from similar species. However, this approach introduces biases and imprecisions that are difficult to quantify. Recent developments in hierarchical modelling have enabled missing values to be reconstructed based on the correlations between available life-history data, linking similar species based on phylogeny and environmental conditions.However, these methods cannot resolve life-history variability among populations or species that are closely placed spatially or taxonomically. Here, theoretically motivated constraints that align with life-history theory offer a new avenue for addressing this problem. We describe a Bayesian hierarchical approach that combines fragmented, multispecies and multi-population data with established life-history theory, in order to objectively determine similarity between populations based on trait correlations (life-history trade-offs) obtained from model fitting.We reconstruct 59 unobserved life-history parameters for 23 populations of tuna that sustain some of the world's most valuable fisheries. Testing by cross-validation across different scenarios indicated that life-histories were accurately reconstructed when information was available for other populations of the same species. The reconstruction of several traits was also accurate for species represented by a single population, although credible intervals increased dramatically. Synthesis and applications. The described Bayesian hierarchical method provides access to life-history traits that are difficult to measure directly and reconstructs missing life-history information useful for assessing populations and species that are directly or indirectly affected by human exploitation of natural resources. The method is particularly useful for examining populations that are spatially or taxonomically similar, and the reconstructed life-history strategies described for the principal market tunas have immediate application to the world-wide management of these fisheries.

BACKGROUND: Digenean trematodes of the family Didymozoidae are tissue parasites that are particularly common in scombrid fish. These parasites can reduce the commercial value of fish, especially the ones occurring in the muscle. PURPOSE: In the present study and for the first time, we report the occurrence of didymozoids in muscle tissue of Atlantic chub mackerel (Scomber colias) collected in northeast Atlantic (Portuguese coast). METHODS: The entire musculature of 64 Scomber colias was removed and examined macroscopically and under a stereomicroscope to search for parasites. RESULTS: The prevalence (3.1%) and abundance (0.03) of infection detected were quite low, but the parasites cause muscle softening decreasing the quality of the infected fish. If the infection levels increase, this may constitute a cause of concern for the fisheries and canning industry. CONCLUSION: Didymozoids identified in this study are closely related to muscle parasites detected in other fish species of the genus Scomber, most probably the same species. Phylogenetic analysis also corroborates the hypothesis that muscle-parasitizing didymozoids are distinct from the ones parasitizing other fish organs.

Vertebrate bone is composed of three main cell types: osteoblasts, osteoclasts and osteocytes, the latter being by far the most numerous. Osteocytes are thought to play a fundamental role in bone physiology and homeostasis, however they are entirely absent in most extant species of teleosts, a group that comprises the vast majority of bony 'fishes', and approximately half of vertebrates. Understanding how this acellular (anosteocytic) bone appeared and was maintained in such an important vertebrate group has important implications for our understanding of the function and evolution of osteocytes. Nevertheless, although it is clear that cellular bone is ancestral for teleosts, it has not been clear in which specific subgroup the osteocytes were lost. This review aims to clarify the phylogenetic distribution of cellular and acellular bone in teleosts, to identify its precise origin, reversals to cellularity, and their implications. We surveyed the bone type for more than 600 fossil and extant ray-finned fish species and optimised the results on recent large-scale molecular phylogenetic trees, estimating ancestral states. We find that acellular bone is a probable synapomorphy of Euteleostei, a group uniting approximately two-thirds of teleost species. We also confirm homoplasy in these traits: acellular bone occurs in some non-euteleosts (although rarely), and cellular bone was reacquired several times independently within euteleosts, in salmons and relatives, tunas and the opah (Lampris sp.). The occurrence of peculiar ecological (e.g. anadromous migration) and physiological (e.g. red-muscle endothermy) strategies in these lineages might explain the reacquisition of osteocytes. Our review supports that the main contribution of osteocytes in teleost bone is to mineral homeostasis (via osteocytic osteolysis) and not to strain detection or bone remodelling, helping to clarify their role in bone physiology.

RESUMO

Hermaphroditic gonads were collected from three skipjack tuna Katsuwonus pelamis in the western and central Pacific Ocean. Both ovarian and testicular tissues were found in the same gonad lobe for each of the three fish. Histological sections of the hermaphroditic gonad from one K. pelamis showed postovulatory follicles, which is evidence of spawning.
