RESUMO

The setts of the European badger Meles meles can be cohabited during reproductive season by the red fox Vulpes vulpes and raccoon dog Nyctereutes procyonoides. There is no information on the possible impact of both species on the size of badgers' litter. The aim of the study was to show the influence of cohabitation of the same setts by badger, raccoon dog and fox on the litter size. The research was conducted in 2012-2014 and 2018 in the lowland forests of western Poland. We conducted the survey of setts by direct observations and analysis of photographic material from trap cameras during mid-April-July each year. We recorded 85 badger litters, 18 fox litters, and 15 raccoon dog litters. Average litter size was 1.71 (±0.90), 2.44 (±1.34) and 4.93 (±2.76) litter mates in badgers, foxes and raccoon dogs, respectively for all observed pairs. Badger litter size did not differ between setts used only by badgers including pairs with no cubs (1.66 ± 0.98) and cohabited with foxes (1.90 ± 0.32) or raccoon dogs (1.88 ± 0.81). However, foxes reared even more cubs in setts cohabited with badgers than when badger was absent (2.90 ± 1.37 vs. 1.88 ± 1.13 respectively). In the case of raccoon dogs, there were no differences in the mean number of their cubs in setts with badgers (5.25 ± 2.92) and without badgers (4.57 ± 2.76). The results indicate that the cohabitation of setts by badgers, foxes and raccoon dogs does not affect litter size negatively.

We analyzed the genotypes of three pregnant females and their litters to investigate the phenomenon of multiple paternity in wild raccoon dogs (Nyctereutes procyonoides) using 17 microsatellite markers. If a female has mated with only one male during estrus, then the maximum number of paternal alleles will not exceed two among littermates with the same father. The results revealed two out of three litters had three or four paternal alleles at one or five microsatellite loci. Therefore, the female had mated with more than one male during estrus. To the best of our knowledge, the present study is the first to report the possibility of multiple paternity in wild raccoon dogs.

Vertical seed dispersal, i.e. seed dispersal towards a higher or lower altitude, is considered a critical process for plant escape from climate change. However, studies exploring vertical seed dispersal are scarce, and thus, its direction, frequency, and mechanisms are little known. In the temperate zone, evaluating vertical seed dispersal of animal-dispersed plants fruiting in autumn and/or winter is essential considering the dominance of such plants in temperate forests. We hypothesized that their seeds are dispersed towards lower altitudes because of the downhill movement of frugivorous animals following the autumn-to-winter phenology of their food plants which proceeds from the mountain tops to the foot in the temperate zone. We evaluated the vertical seed dispersal of the autumn-fruiting wild kiwi, Actinidia arguta, which is dispersed by temperate mammals. We collected dispersed seeds from mammal faeces in the Kanto Mountains of central Japan and estimated the distance of vertical seed dispersal using the oxygen isotope ratios of the dispersed seeds. We found the intensive downhill seed dispersal of wild kiwi by all seed dispersers, except the raccoon dog (bear: mean -393.1 m; marten: -245.3 m; macaque: -98.5 m; and raccoon dog: +4.5 m). Mammals with larger home ranges dispersed seeds longer towards the foot of the mountains. Furthermore, we found that seeds produced at higher altitudes were dispersed a greater distance towards the foot of the mountains. Altitudinal gradients in autumn-to-winter plant phenology and other mountain characteristics, i.e. larger surface areas and more attractive human crops at lower altitudes compared to higher altitudes, were considered drivers of downhill seed dispersal via animal movement. Strong downhill seed dispersal by mammals suggests that populations of autumn-to-winter fruiting plants dispersed by animals may not be able to sufficiently escape from current global warming in the temperate zone.

We studied the food habits of the Japanese marten (Martes melampus) and the raccoon dog (Nyctereutes procionoides) at the Tama Forest Science Garden, western Tokyo, Japan by fecal analysis in 2014/15. Martens were dependent on fruits throughout the year and showed less marked seasonal changes. Raccoon dogs were less dependent on fruits than martens were, and more dependent on mammals in spring, insects in summer and winter, and seeds throughout the year. Martens fed on more fruits containing tiny seeds, such as Actinidia arguta and Stachyurus praecox, whereas raccoon dogs fed on more large-seeded fruits, such as Ginkgo biloba and Diospyros kaki. Martens fed on more fruits that grew at the forest edges, whereas raccoon dogs fed on more fruits growing inside the forest.

The raccoon dog (Nyctereutes procyonoides) is an invasive canid originating from eastern Asia. Here, we review its physiological adaptations to wintering, with an emphasis on northern Europe, where the raccoon dog spends the coldest part of the year in winter sleep. The timing of physiological changes related to wintering is connected to photoperiod by melatonin. In preparation to winter, raccoon dogs display autumnal hyperphagia and fattening probably regulated by the interaction of several peptide hormones. Sufficient fat deposition is essential for survival through the cold season and for reproduction in spring. The wintering strategy includes alternating periods of physical activity and passivity. Effective arousal and foraging during warmer bouts are enabled by normoglycaemia. During active periods, raccoon dogs are opportunistic participants in the food web, and they mainly utilize ungulate carcasses, plant material, and small mammals. Preferred wintertime habitats include watersides, forests, wetlands, and gardens. However, many food items become limited in mid-winter and snow restricts foraging leading to a negative energy balance. During passivity, energy is preserved by denning and by modest metabolic suppression, probably enabled by decreased thyroid hormone levels. Sleepiness and satiety could be maintained by high growth hormone and leptin concentrations. Several hormones participate in the extension of phase II of fasting with selective fatty acid mobilization and efficient protein conservation. The blood count, organ function tests, bone mass, and bone biomechanical properties exhibit high resistance against catabolism, and breeding can be successful after significant weight loss. The flexible physiological response to wintering is probably one reason enabling the successful colonization of this species into new areas.

Sarcoptic mange is a highly contagious zoonotic skin disease that can have severe effect on population dynamics of many wild mammals. However, very little is known about its effect on the activity and space use of infected animals. In this study we equipped two raccoon dogs (Nyctereutes procyonoides) in a mange outbreak area with radio-collars and observed the spatial and temporal effects of sarcoptic mange to these individuals. Initially, the raccoon dogs had no external symptoms of mange infection, but developed these during the study period. One of the raccoon dogs died just 32 and the other 52days after collaring. During a relatively short period before their death, there was an abrupt and drastic decline in their home range size. For one of the animals it started about 1 month before its death and the home range size reduced >1000×, whereas for the other raccoon dog it took place within the last week, decreasing >200×. The daily covered distances also declined considerably. These results indicate that at the later stage sarcoptic mange affected the physiological state of the diseased animals so forcefully that made them almost immobile. Our results show for the first time how rapid and severe could the impact of sarcoptic mange be for one of its most important reservoir and vector species.

The raccoon dog (Nyctereutes procyonoides) is a promising animal model capable of preventing disuse-induced osteoporosis. Previous data suggest that this species resembles bears in the preservation of bone mass and biomechanical properties during prolonged passivity and catabolism. This longitudinal study examined the osteological properties of tibiae in farm-bred raccoon dogs that were either fed or fasted (n=6 per group) for a 10 week period. Peripheral quantitative computed tomography was utilized and plasma markers of bone turnover measured before fasting and at 9 weeks followed by mechanical testing (three-point bending), micro-computed tomography and Fourier transform infrared imaging at 10 weeks. Passive wintering with prolonged catabolism (body mass loss 32%) had no significant effects on bone mineralization, porosity or strength. The concentration of C-terminal telopeptide of type I collagen, indicative of bone resorption, increased in the plasma of the fasted raccoon dogs, while the bone formation markers were unchanged. The levels of 25-hydroxyvitamin D were reduced in the fasted animals. Based on these data, the preservation of bone in wintering raccoon dogs shares characteristics with that of bears with no apparent decrease in the formation of bone but increased resorption. To conclude, raccoon dogs were able to minimize bone loss during a 10 week period of catabolism and passivity.

Knowledge of space use is central to understand animals' role in ecosystems. The raccoon dog Nyctereutes procyonoides is considered as one of the most influential alien mesopredator species in Europe, having the potential to cause loss of local biodiversity and act as a vector for zoonotic diseases. We collared 12 animals to study their home range and habitat use in two areas with different management regimes in Estonia: in a protected natural area and in an intensively managed area. From May to October raccoon dogs inhabiting the natural area had considerably smaller home ranges compared to the managed area, 193.3ha±37.3SD and 391.9ha±292.9SD, respectively. This result contradicts somewhat earlier findings in other European raccoon dog populations, where the home range sizes in natural areas in summer and autumn period have usually been larger compared to managed areas. In both study areas raccoon dogs preferred watersides, where amphibians and other semi-aquatic prey are abundant, to other habitats available in their home ranges. We also studied movements of a raccoon dog pair in the managed study area in winter period. Due to mild weather conditions during the study period, raccoon dogs changed their resting sites quite often, covering a relatively large 599 ha area from November 2012 to January 2013, indicating the absence of usual winter lethargy during the mild winters.

The ranges of pronation/supination of forearms in raccoons, raccoon dogs and red pandas were nondestructively examined. Three carcasses of each species were used for CT analysis, and the left forearms were scanned with a CT scanner in two positions: maximal supination and maximal pronation. Scanning data were reconstructed into three-dimensional images, cross-sectional images were extracted at the position that shows the largest area in the distal part of ulna, and then, the centroids of each cross section of the radius and ulna were detected. CT images of two positions were superimposed, by overlapping the outlines of each ulna, and then, the centroids were connected by lines to measure the angle of rotation, as an index of range of mobility. The measurements in each animal were analyzed, using the Tukey-Kramer method. The average angle of rotation was largest in raccoons and smallest in raccoon dogs, and the difference was significant. In the maximally pronated forearm of all species, the posture was almost equal to the usual grounding position with palms touching the ground. Therefore, the present results demonstrate that the forearms of raccoons can supinate to a greater degree from the grounding position with palms on the ground, as compared with those of raccoon dogs and red pandas.

BACKGROUND: The aim of the study was to evaluate the quality of fresh raccoon dog semen and raccoon dog semen stored at 4 °C. The qualitative evaluation was based on apoptosis in the sperm cells, which was tested by the Annexin V/Pi assay, the TUNEL method and JC-1. In addition, the suitability of the semen for insemination and its effect on reproduction in females were determined in relation to the time of storage. RESULTS: During cold storage of the semen, in the samples from all groups a gradual decrease was noted in the percentage of live cells, and an increase in the percentage of cells with abnormal morphology, exhibiting changes typical of late-stage apoptosis (V+/PI+), and of necrotic cells (V-/PI+). There was a significant increase in the percentage of ApoBrDu + sperm cells, while the mitochondrial membrane potential of the sperm decreased significantly after 12 h of storage at 4 °C in the case of lower-quality semen and after 48 h in the case of semen of good quality. As the percentage of sperm with DNA and cell membrane damage increased and the mitochondrial membrane potential decreased, there was an increase in AspAT and acrosin activity. The increase in the percentage of apoptotic sperm in the raccoon dog semen stored at 4 °C resulted in a decrease in the number of females with cubs. CONCLUSIONS: Identification of apoptotic changes in sperm by flow cytometry using the annexin assay, the TUNEL assay and evaluation of mitochondrial membrane potential can be recommended for determination of the suitability of raccoon dog semen for artificial insemination. The study shows that fresh raccoon dog semen should not be used for insemination more than 48 h after collection in the case of semen of very high quality, or after more than 24 h in the case of semen of poorer quality. Cytometric methods of semen analysis should also be used to evaluate various extenders of raccoon dog semen and methods of cryopreservation in terms of ensuring sperm viability, fertilization capacity, and suitability for insemination.

Medium-sized carnivorous mammals are important seed dispersers of fleshy fruits. The raccoon dog (Nyctereutes procyonoides viverrinus) often feeds on fleshy fruits and forms latrines. This behavior may potentially lead to seed dispersal. To determine if this is the case, we studied 1) seed recovery in the droppings of raccoon dogs, and 2) the transportation of seeds between habitats using plastic markers in a western suburb of Tokyo, Japan. In total, 32,473 seeds of 50 plant taxa were recovered from 120 raccoon dog droppings during a year, and 95.7% of the seeds were found to be those of fleshy fruits. The species most frequently recovered were the eurya (Eurya japonica, 52.6%), the brambles (Rubus spp., 17.4%), and the black night shade (Solanum nigrum, 16.0%). A total of 7,412 plastic markers were embedded in baits at 14 bait plots and were recovered in the feces of the raccoon dogs at 22 latrines. The "transportation rates" were calculated in 50-m distance classes and found that most seeds (43.5%) were deposited within 50 m from the bait point, suggesting very short seed dispersal distances. Inter-habitat transportation was observed: 64.9% of the retrieved markers deposited in the forest were transported to other places within the forest. In contrast, almost all of the markers (99.4%) deposited in the open site were transported within the same habitat. These findings suggest that the seeds of forest plants bearing berries can be dispersed out of the forest to open areas by raccoon dogs.

The raccoon dog (Nyctereutes procyonoides) is a canid species with autumnal fattening and prolonged wintertime fasting. Nonpathological body weight cycling and the ability to tolerate food deficiency make this species a unique subject for studying physiological mechanisms in energy metabolism. AMP-activated protein kinase (AMPK) is a cellular energy sensor regulating energy homeostasis. During acute fasting, AMPK promotes fatty acid oxidation and enhances glucose uptake. We evaluated the effects of prolonged fasting on muscle energy metabolism in farm-bred raccoon dogs. Total and phosphorylated AMPK and acetyl-CoA carboxylase (ACC), glucose transporter 4 (GLUT 4), insulin receptor and protein kinase B (Akt) protein expressions of hind limb muscles were determined by Western blot after 10 weeks of fasting. Plasma insulin, leptin, ghrelin, glucose and free fatty acid levels were measured, and muscle myosin heavy chain (MHC) isoform composition analyzed. Fasting had no effects on AMPK phosphorylation, but total AMPK expression decreased in m. rectus femoris, m. tibialis anterior and m. extensor digitorum longus resulting in a higher phosphorylation ratio. Decreased total expression was also observed for ACC. Fasting did not influence GLUT 4, insulin receptor or Akt expression, but Akt phosphorylation was lower in m. flexor digitorum superficialis and m. extensor digitorum longus. Three MHC isoforms (I, IIa and IIx) were detected without differences in composition between the fasted and control animals. The studied muscles were resistant to prolonged fasting indicating that raccoon dogs have an effective molecular regulatory system for preserving skeletal muscle function during wintertime immobility and fasting.

The study was conducted to investigate the effect of dietary manganese (Mn) on reproductive performance of female Ussuri raccoon dogs (Nyctereutes procyonoides) during the breeding season. Healthy female Ussuri raccoon dogs (n=72) were randomly divided into six groups of twelve each. The six experimental diets were formulated to contain graded amounts of Mn (0, 40, 80, 120, 200 and 400mg/kg of diet; Groups A through F, respectively). Litter size of Group D was greater than that of Groups A, B, C and E (P<0.05), with Group E having the smallest litter size. Values of number born alive were affected by different amounts of organic Mn. Number of pups born alive in Group D was greater than that of Groups B, C, E and F (P<0.05). Number of pups weaned alive for Group D was greater than that of Groups A, B, C and E (P<0.05). Mn supplementation of the control diet (containing 24.32 mg/kg from raw materials) with 120 mg/kg of Mn was adequate for female Ussuri raccoon dogs during the breeding season, based on positive effects of reproduction performance.

The raccoon dog (Nyctereutes procyonoides) is an introduced species in Europe with a continually expanding range. Since the species is capable of affecting local ecosystems and is a vector for a number of severe zoonotic diseases, it is important to understand its food habits. Raccoon dog diet was studied in Estonia by examining the contents of 223 stomach samples collected during the coldest period of the year, August to March, in 2010-2012. The most frequently consumed food categories were anthropogenic plants (e.g. cereals, fruits; FO = 56.1%) and carrion (e.g. carcasses of artiodactyls and carnivores; FO = 48.4%). Carrion was also the only food category that was consumed significantly more frequently by raccoon dogs exhibiting symptoms of sarcoptic mange than by uninfected animals. Small mammals, which represent intermediate hosts for the zoonotic tapeworm Echinococcus multilocularis, were more commonly recorded in samples also containing anthropogenic plants than expected by chance. Comparison of raccoon dog and red fox (Vulpes vulpes) diet in Estonia revealed higher overlap than found elsewhere in Europe, with 'carrion' and 'anthropogenic plants' making up the bulk of both species' diet; however, raccoon dogs were more omnivorous than red foxes. Our results suggest that while the use of most food categories reflects the phenology of natural food sources, 'anthropogenic plants' and 'carrion' provide an essential resource for raccoon dogs during the coldest period of the year, with the latter resource especially important for individuals infected with sarcoptic mange. Since both of these food categories and small mammals are often found at supplementary feeding sites for wild boar (Sus scrofa), this game management practice may facilitate high densities of mesocarnivores and promote the spread of some severe zoonotic diseases, including alveolar echinococcosis, trichinellosis, rabies and sarcoptic mange.

Male sexually-selected traits often impose an increased risk of predation on their bearers, causing male-biased predation. We investigated whether males of the sap-feeding Japanese rhinoceros beetle Trypoxylus dichotomus were more susceptible to predation than females by comparing the morphology of beetles caught in bait traps with the remains of beetles found on the ground. The males of this species are larger than the females and have a horn on the head. We found that predation pressure was greater for males than for females, and that larger individuals of both sexes were more vulnerable to predation. We identified two predators, the raccoon dog Nyctereutes procyonoides and jungle crow Corvus macrorhynchos, by monitoring sap-site trees with infrared video cameras. Raccoon dogs visited sap-site trees at night, while crows came after daybreak. The highest frequency of visits by both predators was observed in the first half of August, which matches the peak season of T. dichotomus. Raccoon dogs often left bite marks on the remains of prey, whereas crows did not. Bite marks were found on most of the remains collected at two distant localities, which suggested that predation by raccoon dogs is common. Size- and sex-dependent differences in the conspicuousness and active period of T. dichotomus probably explain these biased predation patterns. Our results suggest that having a large horn/body is costly in terms of the increased risk of predation. Predation cost may act as a stabilizing selection pressure against the further exaggeration of male sexual traits.

Despite a marked increase in the focus toward biodiversity conservation in fragmented landscapes, studies that confirm species breeding success are scarce and limited. In this paper, we asked whether local (area of forest patches) and landscape (amount of suitable habitat surrounding of focal patches) factors affect the breeding success of raccoon dogs (Nyctereutes procyonoides) in Tokyo, Central Japan. The breeding success of raccoon dogs is easy to judge as adults travel with pups during the breeding season. We selected 21 forest patches (3.3-797.8 ha) as study sites. In each forest patch, we used infra-red-triggered cameras for a total of 60 camera days per site. We inspected each photo to determine whether it was of an adult or a pup. Although we found adult raccoon dogs in all 21 forest patches, pups were found only in 13 patches. To estimate probability of occurrence and detection for raccoon in 21 forest fragments, we used single season site occupancy models in PRESENCE program. Model selection based on AIC and model averaging showed that the occupancy probability of pups was positively affected by patch area. This result suggests that large forests improve breeding success of raccoon dogs. A major reason for the low habitat value of small, isolated patches may be the low availability of food sources and the high risk of being killed on the roads in such areas. Understanding the effects of local and landscape parameters on species breeding success may help us to devise and implement effective long-term conservation and management plans.

BACKGROUND: A multi-faceted approach was used to investigate the wintertime ecophysiology and behavioral patterns of the raccoon dog, Nyctereutes procyonoides, a suitable model for winter sleep studies. By utilizing GPS tracking, activity sensors, body temperature (Tb) recordings, change-point analysis (CPA), home range, habitat and dietary analyses, as well as fatty acid signatures (FAS), the impact of the species on wintertime food webs was assessed. The timing of passive bouts was determined with multiple methods and compared to Tb data analyzed by CPA. RESULTS: Raccoon dogs displayed wintertime mobility, and the home range sizes determined by GPS were similar or larger than previous estimates by radio tracking. The preferred habitats were gardens, shores, deciduous forests, and sparsely forested areas. Fields had close to neutral preference; roads and railroads were utilized as travel routes. Raccoon dogs participated actively in the food web and gained benefit from human activity. Mammals, plants, birds, and discarded fish comprised the most important dietary classes, and the consumption of fish could be detected in FAS. Ambient temperature was an important external factor influencing Tb and activity. The timing of passive periods approximated by behavioral data and by CPA shared 91% similarity. CONCLUSIONS: Passive periods can be determined with CPA from Tb recordings without the previously used time-consuming and expensive methods. It would be possible to recruit more animals by using the simple methods of data loggers and ear tags. Hunting could be used as a tool to return the ear-tagged individuals allowing the economical extension of follow-up studies. The Tb and CPA methods could be applied to other northern carnivores.

The objective of this study was to investigate immunolocalization of steroidogenic enzymes 3ßHSD, P450c17 and P450arom and their expression during the breeding season in wild male raccoon dogs. The testicular weight, size and seminiferous tubule diameters were measured, and histological and immunohistochemical observations of testes were performed. The messenger RNA expression (mRNA) of 3ßHSD, P450c17 and P450arom was measured in the testes during the breeding season. 3ßHSD was found in Leydig cells during the breeding and non-breeding seasons with more intense staining in the breeding season. P450c17 was identified in Leydig cells and spermatids in the breeding season, whereas it was present only in Leydig cells in the non-breeding season. The localization of P450arom changed seasonally: no immunostaining in the non-breeding season; more extensive immunostaining in Leydig cells, Sertoli cells and elongating spermatids in the breeding season. In addition, 3ßHSD, P450c17 and P450arom mRNA were also expressed in the testes during the breeding season. These results suggested that seasonal changes in testicular weight, size and seminiferous tubule diameter in the wild raccoon dog were correlated with spermatogenesis and immunoreactivity of steroidogenic enzymes and that steroidogenic enzymes may play an important role in the spermatogenesis and testicular recrudescence and regression process.

The objectives of this study were to (1) assess year-round behaviors and activity patterns of captive raccoon dogs (Nyctereutes procyonoides) and (2) characterize the species' reproductive endocrinology. Behaviors and activity patterns of 12 (5.7) animals were recorded over a 1-year period. During that time, fecal samples were collected 2-7 times/week from 16 (7.9) individuals (six of these were included in the behavioral study) for the analysis of testosterone, progesterone and estrogen metabolite concentrations. Activity pattern and excretion of gonadal steroids followed a seasonal pattern. Specifically, dogs were cathemeral in summer, and primarily nocturnal in winter. In the males, testosterone concentrations were at baseline from April through September, began to rise in October and reached peak concentrations in February (P<0.05). In the females, elevated estrogen (P<0.05) was observed in March followed by an increase in progestagen concentrations from March through May (P<0.05) in both pregnant and pseudopregnant animals. Gender significantly influenced monthly testosterone/estrogen ratio (P<0.01); values were higher in males than in females throughout the year with overall percentage of overlapping values between males and females being 28%. In summary, this study characterized cirannual fluctuations in behaviors and gonadal steroid metabolites in the raccoon dog maintained in captivity. Because there is no obvious sexual dimorphism, the differences in testosterone/estrogen ratio may be useful for gender differentiation (72% accuracy), especially among individuals living in the wild.

Nine skull samples of the beaver Castor fiber, six samples of the raccoon dog Nyctereutes procyonoides, and six samples of the American mink Neovison vison were studied using phenetic and craniometric methods. Analysis of the phenofund structure suggests that in all of the studied species the emergence of novel character variations does not lead to their fixation with a significant frequency. Considerable morphological variability emerges in the contact zone of different autochtonous populations, of wild and breeding forms, as well as in geographically and reproductively isolated small groups of individuals. Morphological differences of introduced animals fit into the conception of species polymorphism and are smoothed over when separate colonies merge into metapopulations, which does not lead to the emergence of novel stable taxa.
