RESUMO

Plant roots and soil prokaryotes primarily interact with each other in the rhizosphere. Changes in the rhizosphere prokaryotic structure are influenced by several factors. In this study, the community structure of the Potentilla anserina L. rhizosphere prokaryotes was identified and evaluated by high-throughput sequencing technology in different continuous cropping fields and developmental stages of the plant. In total, 2 archaeal (Euryarchaeota and Thaumarchaeota) and 26 bacterial phyla were identified in the P. anserina rhizosphere. The bacterial community was mainly composed of Acidobacteria, Actinobacteria, Bacteroidetes, Chloroflexi, Gemmatimonadetes, Planctomycetes, Proteobacteria, and Verrucomicrobia. Moreover, the prokaryotic community structure of the rhizosphere varied significantly during plant development. Our results provide new insights into the dynamics of the P. anserina rhizosphere prokaryotic community and may provide useful information for enhancing the growth and development of P. anserina through artificial control of the soil prokaryotes.

Background: The genus Potentilla is closely related to that of Fragaria, the economically important strawberry genus. Potentilla micrantha is a species that does not develop berries but shares numerous morphological and ecological characteristics with Fragaria vesca. These similarities make P. micrantha an attractive choice for comparative genomics studies with F. vesca. Findings: In this study, the P. micrantha genome was sequenced and annotated, and RNA-Seq data from the different developmental stages of flowering and fruiting were used to develop a set of gene predictions. A 327 Mbp sequence and annotation of the genome of P. micrantha, spanning 2674 sequence contigs, with an N50 size of 335,712, estimated to cover 80% of the total genome size of the species was developed. The genus Potentilla has a characteristically larger genome size than Fragaria, but the recovered sequence scaffolds were remarkably collinear at the micro-syntenic level with the genome of F. vesca, its closest sequenced relative. A total of 33,602 genes were predicted, and 95.1% of bench-marking universal single-copy orthologous genes were complete within the presented sequence. Thus, we argue that the majority of the gene-rich regions of the genome have been sequenced. Conclusions: Comparisons of RNA-Seq data from the stages of floral and fruit development revealed genes differentially expressed between P. micrantha and F. vesca.The data presented are a valuable resource for future studies of berry development in Fragaria and the Rosaceae and they also shed light on the evolution of genome size and organization in this family.

Plants can plastically respond to light competition in three strategies, comprising vertical growth, which promotes competitive dominance; shade tolerance, which maximises performance under shade; or lateral growth, which offers avoidance of competition. Here, we test the hypothesis that plants can 'choose' between these responses, according to their abilities to competitively overcome their neighbours. We study this hypothesis in the clonal plant Potentilla reptans using an experimental setup that simulates both the height and density of neighbours, thus presenting plants with different light-competition scenarios. Potentilla reptans ramets exhibit the highest vertical growth under simulated short-dense neighbours, highest specific leaf area (leaf area/dry mass) under tall-dense neighbours, and tend to increase total stolon length under tall-sparse neighbours. These responses suggest shifts between 'confrontational' vertical growth, shade tolerance and lateral-avoidance, respectively, and provide evidence that plants adopt one of several alternative plastic responses in a way that optimally corresponds to prevailing light-competition scenarios.

BACKGROUND: Different methods have been used to map species and habitat distributions. In this paper, similarity-based reasoning-a methodological approach that has received less attention-was applied to estimate the distribution and coverage of Dasiphora fruticosa for the region in the Baltic states where grows the most abundant population of this species. METHODS: Field observations, after thinning to at least 50 m interval, included 1480 coverage estimations in the species presence locations and 8317 absence locations. Species coverage for the 750 km2 of directly unobserved area was calculated using machine learning in the similarity-based prediction system Constud. Separate predictive sets of site features (e.g. land cover, soil type) and exemplar weights were calibrated for spatial partitions of the study area (probable presence region, unclear region, proved absence region). A modified version of the Gower's distance metric, as used in Constud, is described. RESULTS: The resulting maps depicted the predicted coverage, the certainty of decision when predicting presence or absence, and the mean similarity to the exemplar locations used while predicting. Coverage prediction errors were smaller in the unclear partition-where the species was mostly absent-than in the probable presence partition, where coverage ranged from 0 to 90%. CONCLUSIONS: We call for methodological comparisons using the same data set.

Environmental factors may influence types and contents of active substances. This study investigated the influence of environmental factors on the active substance contents and antioxidant activity of Potentilla fruticosa L. from different regions of China. Also, HPLC fingerprint similarity analysis (SA) coupled with hierarchical cluster analysis (HCA) and discriminant analysis (DA) were further introduced for the accurate classification and quality assessment of P. fruticosa. The results showed that altitude was significantly and negatively correlated to the content of tannin (P < 0.05). Annual sunshine duration and altitude were significantly and positively correlated to the flavonoids content, rutin content and antioxidant activity (P < 0.05). Annual mean temperature was significantly and negatively correlated to the content of total phenolics, while altitude was significantly and positively correlated to the content of total phenolics (P < 0.05). Eight samples were unambiguously separated into three groups. Two types of discriminant functions with a 100% discrimination ratio were constructed. All data consistently supported the conclusion that P. fruticosa produced from Kangding, Sichuan Province had high quality among all samples, therefore, Kangding in Sichuan Province with favorable environmental conditions is recommended as a preferable production location.

Growth of herbaceous plants responds sensitively and rapidly to climate variability. Yet, little is known regarding how climate warming influences the growth of herbaceous plants, particularly in semi-arid sites. This contrasts with widely reported tree growth decline and even mortality in response to severe water deficits due to climate warming around the world. Here, we use the relatively novel approach of herb-chronology to analyze the correlation between climatic factors and annual ring width in the root xylem of two perennial forb species (Medicago sativa, Potentilla chinensis) in the Loess Plateau of China. We show that warming-induced water deficit has a significant negative effect on the growth of herbaceous plants in the Loess Plateau. Our results indicate that the growth of forbs responds rapidly and sensitively to drought variability, implying that water availability plays a dominant role in regulating the growth of herbaceous plants in semi-arid areas. If warming and drying in the Loess Plateau continue in the future, further affects the growth of herbaceous plants, potentially driving regional changes in the relationship between herbaceous vegetation and climate.

Biomass partitioning has been explored across various biomes. However, the strategies of allocation in plants still remain contentious. This study investigated allocation patterns of above- and belowground biomass at the community level, using biomass survey from the Tibetan Plateau. We explored above- and belowground biomass by conducting three consecutive sampling campaigns across shrub biomes on the northeast Tibetan Plateau during 2011-2013. We then documented the above-ground biomass (AGB), below-ground biomass (BGB) and root: shoot ratio (R/S) and the relationships between R/S and environment factors using data from 201 plots surveyed from 67 sites. We further examined relationships between above-ground and below-ground biomass across various shrub types. Our results indicated that the median values of AGB, BGB, and R/S in Tibetan shrub were 1102.55, 874.91 g m-2, and 0.85, respectively. R/S showed significant trend with mean annual precipitation (MAP), while decreased with mean annual temperature (MAT). Reduced major axis analysis indicated that the slope of the log-log relationship between above- and belowground biomass revealed a significant difference from 1.0 over space, supporting the optimal hypothesis. Interestingly, the slopes of the allometric relationship between log AGB and log BGB differed significantly between alpine and desert shrub. Our findings supported the optimal theory of above- and belowground biomass partitioning in Tibetan shrub, while the isometric hypothesis for alpine shrub at the community level.

The study evaluated the levels of nine metals, namely Ca, Cd, Fe, K, Mg, Mn, Pb, Tl, and Zn, in soils and tissues of ten plant species growing spontaneously on heaps left by historical mining for Zn-Pb ores. The concentrations of Cd, Pb, Tl, and Zn in heap soils were much higher than in control soils. Plants growing on heaps accumulated excessive amounts of these elements in tissues, on average 1.3-52 mg Cd kg(-1), 9.4-254 mg Pb kg(-1), 0.06-23 mg Tl kg(-1) and 134-1479 mg Zn kg(-1) in comparison to 0.5-1.1 mg Cd kg(-1), 2.1-11 mg Pb kg(-1), 0.02-0.06 mg Tl kg(-1), and 23-124 mg Zn kg(-1) in control plants. The highest concentrations of Cd, Pb, and Zn were found in the roots of Euphorbia cyparissias, Fragaria vesca, and Potentilla arenaria, and Tl in Plantago lanceolata. Many species growing on heaps were enriched in K and Mg, and depleted in Ca, Fe, and Mn. The concentrations of all elements in plant tissues were dependent on species, organ (root vs. shoot), and species-organ interactions. Average concentrations of Ca, K, and Mg were generally higher in shoots than in roots or similar in the two organs, whereas Cd, Fe, Pb, Tl, and Zn were accumulated predominantly in the roots. Our results imply that heaps left by historical mining for Zn-Pb ores may pose a potential threat to the environment and human health.

Clonal plants spreading horizontally and forming a network structure of ramets exhibit complex growth patterns to maximize resource uptake from the environment. They respond to spatial heterogeneity by changing their internode length or branching frequency. Ramets definitively root in the soil but stay interconnected for a varying period of time thus allowing an exchange of spatial and temporal information. We quantified the foraging response of clonal plants depending on the local soil quality sampled by the rooting ramet (i.e. the present information) and the resource variability sampled by the older ramets (i.e. the past information). We demonstrated that two related species, Potentilla reptans and P. anserina, responded similarly to the local quality of their environment by decreasing their internode length in response to nutrient-rich soil. Only P. reptans responded to resource variability by decreasing its internode length. In both species, the experience acquired by older ramets influenced the plastic response of new rooted ramets: the internode length between ramets depended not only on the soil quality locally sampled but also on the soil quality previously sampled by older ramets. We quantified the effect of the information perceived at different time and space on the foraging behavior of clonal plants by showing a non-linear response of the ramet rooting in the soil of a given quality. These data suggest that the decision to grow a stolon or to root a ramet at a given distance from the older ramet results from the integration of the past and present information about the richness and the variability of the environment.

BACKGROUND: The responses of plant seeds and seedlings to changing atmospheric nitrogen (N) deposition and precipitation regimes determine plant population dynamics and community composition under global change. METHODOLOGY/PRINCIPAL FINDINGS: In a temperate steppe in northern China, seeds of P. tanacetifolia were collected from a field-based experiment with N addition and increased precipitation to measure changes in their traits (production, mass, germination). Seedlings germinated from those seeds were grown in a greenhouse to examine the effects of improved N and water availability in maternal and offspring environments on seedling growth. Maternal N-addition stimulated seed production, but it suppressed seed mass, germination rate and seedling biomass of P. tanacetifolia. Maternal N-addition also enhanced responses of seedlings to N and water addition in the offspring environment. Maternal increased-precipitation stimulated seed production, but it had no effect on seed mass and germination rate. Maternal increased-precipitation enhanced seedling growth when grown under similar conditions, whereas seedling responses to offspring N- and water-addition were suppressed by maternal increased-precipitation. Both offspring N-addition and increased-precipitation stimulated growth of seedlings germinated from seeds collected from the maternal control environment without either N or water addition. Our observations indicate that both maternal and offspring environments can influence seedling growth of P. tanacetifolia with consequent impacts on the future population dynamics of this species in the study area. CONCLUSION/SIGNIFICANCE: The findings highlight the importance of the maternal effects on seed and seedling production as well as responses of offspring to changing environmental drivers in mechanistic understanding and projecting of plant population dynamics under global change.

BACKGROUND AND AIMS: High alpine environments are characterized by short growing seasons, stochastic climatic conditions and fluctuating pollinator visits. These conditions are rather unfavourable for sexual reproduction of flowering plants. Apomixis, asexual reproduction via seed, provides reproductive assurance without the need of pollinators and potentially accelerates seed development. Therefore, apomixis is expected to provide selective advantages in high-alpine biota. Indeed, apomictic species occur frequently in the subalpine to alpine grassland zone of the European Alps, but the mode of reproduction of the subnival to nival flora was largely unknown. METHODS: The mode of reproduction in 14 species belonging to seven families was investigated via flow cytometric seed screen. The sampling comprised 12 species typical for nival to subnival plant communities of the European Alps without any previous information on apomixis (Achillea atrata, Androsace alpina, Arabis caerulea, Erigeron uniflorus, Gnaphalium hoppeanum, Leucanthemopsis alpina, Oxyria digyna, Potentilla frigida, Ranunculus alpestris, R. glacialis, R. pygmaeus and Saxifraga bryoides), and two high-alpine species with apomixis reported from other geographical areas (Leontopodium alpinum and Potentilla crantzii). KEY RESULTS: Flow cytometric data were clearly interpretable for all 46 population samples, confirming the utility of the method for broad screenings on non-model organisms. Formation of endosperm in all species of Asteraceae was documented. Ratios of endosperm : embryo showed pseudogamous apomixis for Potentilla crantzii (ratio approx. 3), but sexual reproduction for all other species (ratios approx. 1·5). CONCLUSIONS: The occurrence of apomixis is not correlated to high altitudes, and cannot be readily explained by selective forces due to environmental conditions. The investigated species have probably other adaptations to high altitudes to maintain reproductive assurance via sexuality. We hypothesize that shifts to apomixis are rather connected to frequencies of polyploidization than to ecological conditions.

Background and Aims Game theoretical models predict that plants growing in dense stands invest so much biomass in height growth that it trades-off with investment in other organs such as the leaves, leading to decreased plant production. Using the stoloniferous plant Potentilla reptans, we tested the hypothesis that genotypes investing more in the petioles in response to increased density show a greater decrease in total plant mass. We also tested whether a greater increase in mother ramet investment would lead to a greater decrease in investment in vegetative propagation. Methods To uncouple costs and benefits of investments in petioles, ten genotypes that were known to differ in their response to shading signals were grown in monogenotypic stands at two different densities. Key Results Genotypes differed in their increase in petiole investment in response to an increase in density, but not in their decrease in total plant mass or root mass. Total lamina area per plant did not differ significantly between the densities, nor did the mass invested in the laminae per unit of total plant mass. Genotypes differed considerably in the change in vegetation height and petiole investment, but this was not significantly negatively correlated with the change in total plant mass. The genotypes did differ in the change of mass investment in the mother ramet: a greater increase in investment in the mother ramet was correlated to a greater decrease in vegetative propagation. Conclusions While a greater increase in height investment did not lead to a greater decrease in biomass production, it did lead to a decrease in vegetative propagation. This ability to change allocation towards the mother ramets may imply that competition within a stand of stoloniferous plants does not necessarily result in lower total biomass production due to increased height investment.

To date, little is still known about how alpine species occurring in the Qinghai-Tibetan Plateau (QTP) responded to past climatic oscillations. Here, by using variations of the chloroplast trnT-L, we examined the genetic distribution pattern of 101 individuals of Potentilla glabra, comprising both the interior QTP and the plateau edge. Phylogenetic and network analyses of 31 recovered haplotypes identified three tentative clades (A, B and C). Analysis of molecular variance (amova) revealed that most of the genetic variability was found within populations (0.693), while differentiations between populations were obviously distinct (F(st)= 0.307). Two independent range expansions within clades A and B occurring at approximately 316 and 201 thousand years ago (kya) were recovered from the hierarchical mismatch analysis, and these two expansions were also confirmed by Fu's F(S) values and 'g' tests. However, distant distributions of clade C and private haplotypes from clades A and B suggest that they had survived the Last Glacial Maximum (LGM) and previous glaciers in situ since their origins. Our findings based on available limited samples support that multiple refugia of a few cold-enduring species had been maintained in the QTP platform during LGM and/or previous glacial stages.

Using a new experimental set up, the way in which height growth of stoloniferous plants is adjusted to that of their neighbours, as well as differences between genotypes in their ability to keep up with neighbour height growth were tested. Five Potentilla reptans genotypes inherently differing in petiole length were subjected to three experimental light gradients, involving light intensity and red : far-red ratio. Each plant was placed in a vertically adjustable cylinder of green foil, and the treatments differed in the speed of cylinder height increase and final height. Total weight of plants decreased from the 'Slow' to the 'Fast' treatment, while petiole length increased. Leaves reaching the top of the cylinder stopped petiole elongation, resulting in similar final heights for all genotypes in the 'Slow' treatment. In the 'Fast' treatment only the fastest-growing genotype maintained its position in the top of the cylinder and genotypes differed strongly in final height within the cylinders. Plants adjust their height growth to that of the surrounding vegetation, leading to height convergence in short light gradients that slowly increase. These adjustments and genotypic differences in ability to keep up with fast-growing neighbours can influence the outcome of competition for light.

BACKGROUND AND AIMS: Because plants protect each other from wind, stand density affects both the light climate and the amount of mechanical stress experienced by plants. But the potential interactive effects of mechanical stress and canopy shading on plant growth have rarely been investigated and never in stoloniferous plants which, due to their creeping growth form, can be expected to respond differently to these factors than erect plants. METHODS: Plants of ten genotypes of the stoloniferous species Potentilla reptans were subjected to two levels of mechanical stress (0 or 40 daily flexures) and two levels of spectral shading (15 % of daylight with a red:far red ratio of 0.3 vs. 50 % daylight and a red:far red ratio of 1.2). KEY RESULTS: Mechanically stressed plants produced more leaves with shorter more flexible petioles, more roots, and more but less massive stolons. Responses to spectral shading were mostly in the opposite direction to thigmomorphogenesis, including the production of thinner, taller petioles made of more rigid tissue. The degree of thigmomorphogenesis was either independent of light climate or stimulated by spectral shading. At the genotypic level there were no clear correlations between responses to shade and mechanical stress. CONCLUSIONS: These results suggest that in stoloniferous plants mechanical stress results in clones with a more compact, shorter shoot structure and more roots. This response does not appear to be suppressed by canopy shading, which suggests that wind shielding (reduced mechanical stress) by neighbours in dense vegetation serves as a cue that induces shade avoidance responses such as increased petiole elongation.

RESUMO

The ability of Potentilla griffithii Hook var. velutina Cardot to hypaeraccumulate zinc (Zn) was identified through field survey and hydroponic experiments. Our results showed that P. griffithii could be classified as a new Zn hyperaccumulator. Zn concentrations in the shoots of P. griffithii averaged 6250 mg kg(-1) (3870-8530 mg kg(-1)) growing in Zn-rich soils. The highest Zn concentration was observed in the leaves of P. griffithii at 22,990 mg kg(-1). The fact that P. griffithii was able to grow in a mining soil with a Zn concentration of 193,000 mg kg(-1) without showing a major sign of phytotoxicity demonstrated its high tolerance to Zn. When growing in hydroponic systems, P. griffithii accumulated a maximum 26700 mg kg(-1) zinc concentration in the shoots, indicating the ability of this species to effectively take up and translocate Zn. Translocation factors (the ratio of Zn concentration in shoot to root) of 1.1 to 1.6 were obtained. Compared to the control, dry biomass of P. griffithii in 160 mg L(-1) Zn treatment increased 66.6% (P < 0.05). The time-course experiment showed that the maximum Zn concentration at 100 mg L(-1) Zn treatment was found at 16 d, much later than that of the 10 mg L(-1) Zn treatment, which might be an attribution of a accumulating mechanism or detoxification of a plant. The report of a new Zn hyperaccumulator provides a new plant species for the phytoremediation of contaminated soil and for the research on mechanisms of Zn hyperaccumulation in plants.

RESUMO

The clonal growth of Potentilla anserina on degraded and non-degraded Kobresia humilis meadow soil was studied by a transplanting experiment in the field. No significant differences in numbers of stolons, height, and leave size per mother ramet were observed between the two soils, but the numbers of leaves per mother ramet, length and width of stolon, and spacer length were significantly different. There were more leaves per mother ramet, longer stolon and spacer, and wider stolon on degraded soil, where available nutrient was poor than in non-degraded soil. Under degraded alpine meadow soil condition, the clonal plant species might produce much more photosynthetic product to support stolon growth by the increase of leave numbers per mother ramet, and the longer and wider stolon could intensify the foraging ability of the mother ramet that would benefit to daughter ramet. Both mother and daughter ramet of Potentilla anserina invested much more biomass to their underground part (root system) in non-degraded soil to increase the survival rate of daughter ramet.
