The evaluation of cytotoxic and apoptotic activities of silver nanoparticles (Ag-NPs) synthesized by aqueous extract of Prosopis farcta was investigated against lung (A549) and colon (HT-29) cell lines. The cytotoxic activity of nanoparticles was performed using MTT assay, while their apoptotic activity was tested using TUNEL method. The obtained results of MTT showed that the cell viability of A549 was dependent on the nanoparticles concentration and incubation time. Therefore, although the cytotoxic effect increased as the Ag-NPs concentration and incubation time heightened, yet the viability of HT-29 cells seems to be dependent only on the incubation time. The apoptotic results of the nanoparticles showed more than 50% of apoptosis on A549 and HT-29 cell lines, which in this case, HT-29 demonstrated 100% apoptosis at concentrations of more than 400 µg/ml. It seems that Ag-NPs synthesized using P. farcta extract can serve as anti-cancer agent in the treatment many cancers through creating or discovering new drug forms.

BACKGROUND: Pollens from mesquite (Prosopis juliflora) are potent allergen responsible in causing immediate hypersensitivity reactions in susceptible people in tropical countries. OBJECTIVE: This study aimed to clone, express and purify the mesquite pollen profilin (Pro j 2) as well as evaluating its nucleotide sequence homology in order to predict allergenic cross-reactivity with profilins of common allergenic plants. METHODS: Immunoblotting assay and specific ELISA were applied to determine the immunoreactivity of sera from 35 patients who were allergic to mesquite pollen. The mesquite profilin-coding sequence was cloned into PTZ57R/T vector and amplified. The cDNA of mesquite pollen profilin was then expressed in Escherichia coli using pET-21b (+) vector and puri?ed by one-step Ni2+ a?nity chromatography. IgE binding capacity of the recombinant mesquite profiling (rPro j 2) was analyzed by specific ELISA, immunoblotting, and inhibition assays. RESULTS: cDNA nucleotide sequencing revealed an open reading frame of 399bp encoding for 133 amino acids which belongs to the profilin family. Seventeen patients (17/35, 48.57%) had significant specific IgE level for rPro j 2. Immunodetection and inhibition assays indicated that puri?ed rPro j 2 might be similar as that in the crude extract. CONCLUSION: Pro j 2, as a new allergen from mesquite pollen, was produced in E. coli with an IgE-reactivity similar to that of its natural counterpart. The amino acid sequences homology analysis of mesquite profilin and several profilin molecules from other plants showed high degree of cross-reactivity among plant-derived profilins from unrelated families.

As vagens de Prosopis juliflora (algaroba) causam sinais nervosos em bovinos, devidos a lesões no núcleo motor do trigêmeo, e cólicas em cavalos devidos à formação de fitobezoários no intestino. O presente trabalho objetivou estudar a toxicidade das vagens de P. juliflora para equinos e comprovar que a utilização 30% de vagens adicionadas na ração de bovinos não causa intoxicação. Para isso foram realizados três experimentos. No Experimento 1, dois bovinos ingeriram, durante um ano, vagens de algaroba, em quantidade equivalente a 30% do total da matéria seca ingerida. Nenhum animal experimental apresentou sinais nervosos. No Experimento 2, dois cavalos receberam vagens em quantidade equivalente a 1% do peso corporal (pc) durante quatro meses e em quantidade equivalente a 1,5% do pc durante outros três meses. No experimento 3, dois equinos receberam vagens ad libitum, durante um mês. Em nenhum dos equinos foram observados sinais nervosos nem cólicas devidas à formação de fitobezoários. Esses resultados sugerem que as vagens de algoraba, apesar de sua toxicidade, podem ser utilizadas na alimentação de bovinos em confinamento ou semi-confinamento, nas concentrações de 30% da alimentação por períodos de até um ano. Não há restrições para a administração de vagens de algaroba em equinos confinados; no entanto, permanecem as restrições para o pastejo de equídeos em áreas invadidas por algaroba. Sugere-se que as vagens perdem a sua capacidade de formar fitobezoários em consequência do armazenamento. Bovinos não devem permanecer em áreas invadidas por P. juliflora, quando a planta está frutificando, por mais de 30 dias.


The ingestion of pods of Prosopis juliflora causes nervous signs in cattle due to lesions in the trigeminal nuclei, and colics in horses due to the formation of phytobezoars in the gut. The objective of this research was to study the toxicity for of P. juliflora pods in horses, and to establish if 30% of pods in the feed are not toxic for cattle. Three experiments were performed. In Experiment 1, two steers were fed, during one year, with food containing 30% of P. juliflora pods. None of the experimental animals showed nervous signs. In Experiment 2, two horses received pods of P. juliflora in amount equivalent to 1% of their bw during four months. For other 3 months this amount was increased to 1.5% bw. In Experiment 3, two horses received P. juliflora pods ad libitum during 30 days. None of the horses showed nervous signs or colic due to the presence of phytobezoars in the gut. These results suggest that P. juliflora pods, despite their toxicity, can be used to feed cattle at concentrations of 30% of the food during one year. There are no restrictions for the use of P. juliflora pods in horses in confined or semi-confined systems, but it is not recommended to maintain equidae grazing in areas where P. juliflora is fructificating. It is suggested that there is a loss in the capacity of the pods to form phytobezoars after harvesting. Cattle may be kept in areas invaded by P. juliflora, during fructification, for no more than 30 days.

Descrevem-se três surtos de intoxicação por vagens de Prosopis juliflora no Sertão e Agreste de Pernambuco, na região semi-árida, em animais pastejando áreas invadidas pela planta ou que ingeriram as vagens como alimento concentrado. Em duas fazendas nas que a doença ocorria esporadicamente foram observados casos individuais. Em outra, o surto afetou um rebanho de 1206 bovinos, dos quais adoeceram 112 (9,28 por cento) e morreram 84 (6,96 por cento), enquanto os demais 28 (2,32 por cento) recuperaram-se e ganharam peso após a retirada das vagens da alimentação. Clinicamente observou-se, principalmente, perda de peso progressiva, atrofia da musculatura da face e masseter, mandíbula pendulosa, protrusão de língua, dificuldade de apreensão e mastigação dos alimentos, torção da cabeça para mastigar ou ruminar, salivação excessiva, disfagia e hipotonia lingual. Nos exames laboratoriais constatou-se anemia e hipoproteinemia. Na necropsia havia caquexia e diminuição de volume e coloração acinzentada dos músculos masseteres. Na histologia observou-se degeneração de neurônios do núcleo motor do trigêmeo, degeneração Walleriana do nervo trigêmeo e atrofia muscular por denervação do músculo masseter com substituição por tecido fibroso. Recomendam-se medidas para a profilaxia da intoxicação e discute-se a necessidade de desenvolver pesquisas para determinar a viabilidade econômica e sustentabilidade da utilização da algaroba como alimento animal ou humano e para produção de carvão, lenha ou madeira.


Three outbreaks of poisoning by Prosopis juliflora pods are reported in the semiarid region of the state of Pernambuco, Northeastern Brazil, in cattle grazing in fields invaded by the plant or ingesting mesquite beans as a concentrate food. In two farms the disease occurred sporadically. In another, 112 (9.28 percent) cattle out of 1206 were affected, 84 (6.96 percent) died due to emaciation, and 28 (2.32 percent) gained weight after the pods had been withdrawn from the feed. Main clinical signs were progressive weight loss, atrophy of the masseter muscles, dropped jaw, tongue protrusion, difficulties in prehending food, tilting the head during mastigation or rumination, salivation, impaired swallowing, and decreased tone of the tongue. The hematology reveals hypoproteinemia and anemia. Gross lesions were emaciation and reduction in size of the masseter muscles, which appear thinner than normal and grayish due muscular atrophy. Degeneration of neurons of the trigeminal motor nuclei, Wallerian degeneration of the trigeminal nerve roots, and muscular atrophy of the masseter muscles with substitution by fibrous tissue were observed on histologic examination. For the prevention of the poisoning is necessary to limit the amount of mesquite beans in animal nutrition. It is also necessary to develop research to determine the economic and sustainability of the use of Prosopis juliflora for animal food, human food or other uses such as charcoal, wood and fuel wood.

Intoxication by pods of Prosopis juliflora (mesquite beans) causes an impairment of cranial nerve function in cattle and goats. In goats, vacuolation of neurons in the trigeminal motor nuclei has been reported. To study the lesions in cattle caused by consumption of P. juliflora pods and dry ground pods, eight 6- to 12-month-old male cattle were divided into 4 groups: group 1 was fed a ration containing 50% of pods; groups 2 and 3 received a ration containing 50 and 75% of dry ground pods, respectively; group 4 was the control. After 200 days, all cattle were killed and sampled for histologic evaluation. Samples of the trigeminal motor nucleus were examined by electron microscopy. All cattle from groups 1, 2, and 3 showed clinical signs resulting from impaired function of cranial nerves V, IX, X, and XII, starting 45-75 days after consumption of the plant. The main histologic lesions were vacuolation and loss of neurons in trigeminal motor nuclei and other motor cranial nerve nuclei with Wallerian-like degeneration in the cranial nerves. Mild denervation atrophy was observed in the masseter and other masticatory muscles. On electron microscopy, neurons of the trigeminal nuclei had markedly swollen mitochondria, with the mitochondrial cristae displaced peripherally, disoriented and disintegrating. Intoxication by P. juliflora seems to have a novel pathogenesis, characterized by a selective, primary, chronic, and progressive injury to mitochondria of neurons of the trigeminal and other cranial nerve nuclei. Cranial nerve degeneration and denervation atrophy of the muscles occurs as a consequence of the neuronal lesion.

Prosopis juliflora is largely used for feeding cattle and humans. Neurological signals have been reported in cattle due to intoxication with this plant. In this study, an alkaloidal fraction (AF) obtained from P. juliflora pods was tested on astrocyte primary cultures. Astrocytes display physiological functions essential to development, homeostasis and detoxification in the central nervous system (CNS). These cells are known for their role on energetic support and immune response in the CNS. Concentrations between 0.03 to 30 µg/ml AF were assayed for 24 - 72 h. The mitochondrial activity, assayed by MTT test, showed cytotoxicity at 30 µg/ml AF after 24 h. At concentrations ranging between 0.3 - 3 µg/ ml, the AF induced an increase on mitochondrial activity, indicating cell reactivity. lmmunocytochemistry assay for GFAP cytoskeletal protein, revealed alterations on cell morphology after treatment with 0.3 - 3 µg/ ml AF for 72 h. This result corroborates with western blot analysis when ceUs treated with 0.3 - 3 I-µg/ml AF for 72 h showed GFAP upregulation. The vimentin expression was not significantly altered in all tested concentrations. These results suggest that alkaloids induce astrocyte reactivity and might be involved in the neurotoxic effects induced by P. juliflora consumption.


A Prosopis juliflora é amplamente utilizada na alimentação humana e de várias espécies animais, especialmente bovinos. Quadros de intoxicação por esta planta, nesta espécie, têm sido relatados, principalmente quando a mesma é oferecida como única fonte alimentar, desencadeando uma doença de sintomatologia nervosa. Neste estudo, objetivou-se avaliar os efeitos in vitro da fração de alcalóides totais (F A) extraída das vagens da Prosopis juliflora utilizando cultura primária de astrócitos obtidos do córtex cerebral de ratos como modelo de estudo. A avaliação da atividade mitocondrial pelo teste do MTT demonstrou a citotoxicidade em 30 µg/ ml da FA após 24 h. As concentrações de 0,3 e 3 µg/ ml da FA induziram um aumento da atividade mitocondrial, indicando reatividade celular. Testes imunocitoquimicos para a GFAP, principal proteína de citoesqueleto de astrócitos, revelaram alterações morfológicas nas células após tratamento com 0,3 e 3 µg/ml da FA por 72 h. Tais resultados são consoantes à análise desta proteína por westernblot, quando as culturas foram tratadas com 0,3 e 3 µg/ml da FA por 72 h, demonstrando interferências na regulação da expressão da GFAP. A expressão de vimentina não foi significativamente alterada em nenhuma das concentrações testadas. Estes resultados sugerem que os alcalóides da P.juliflora induzem a reatividade astrocitária, o que pode estar envolvido nos efeitos neurotóxicos providos pelo consumo desta planta.

BACKGROUND: Allergen skin test reactivity and total serum IgE are objective measures used to characterize and help diagnose allergic diseases. Cross-sectional studies have shown that overall aeroallergen skin test reactivity increases throughout childhood. However, little attention has been paid to whether individual aeroallergen remittance occurs, which could distort or mask relationships to disease. OBJECTIVE: To access the incidence and remittance of skin test reactions to individual allergens in children aged 6-11 years. METHODS: Longitudinal sensitization to six aeroallergens and total IgE were assessed in 828 children raised in the semi-arid US southwest at ages 6 and 11 years. RESULTS: New sensitization (to any allergen) between 6 and 11 years occurred in 30.2% of children compared with 39.7% before age 6 years. The rate of complete remittance from positive to negative between ages 6 and 11 years was 8.2%, and total IgE at age 6 years was not predictive. Remittance rates for individual allergens were high and variable (19-49%). The perennial allergens Bermuda and Alternaria were early sensitizers and had low remittance rates. Early sensitization to the four seasonal allergens was less common and more subject to remittance with the bulk of sensitization occurring between 6 and 11 years. CONCLUSION: This study shows that sensitization to individual aeroallergens in childhood is dynamic and indicates the limitation of single point assessment of skin test reactivity.
