RESUMO

Transition milk (TM, defined here as the second through fourth milkings after calving) supplies additional fat, protein, and immunoglobulins to the calf compared with milk replacer at industry-suggested feeding rates (∼14% solids). Our objective was to determine whether 9 feedings of TM on d 2 through 4 of life increase the growth rate and overall health of calves. Holstein heifer calves on a commercial farm were randomly assigned to 1 of 3 diets (n = 35/diet): milk replacer (MR; Purina Warm Front BOV MOS Medicated Milk Replacer, St. Louis, MO), TM, or a 50:50 blend of MR and colostrum replacer (MCR; Alta HiCal Colostrum Powder Replacer, the Saskatoon Colostrum Company Ltd., Saskatoon, SK, Canada). The TM was harvested from Holstein cows on the farm, pooled, and pasteurized at 71.7°C for 15 s. Nutrient composition on a dry matter basis of TM was 25.9% fat, 41.8% protein, and 14% solids; MR was 10.3% fat, 27.8% protein, and 14% solids; and MCR was 14.6% fat, 38.6% protein, and 15% solids. All calves received IgG-enriched colostrum replacer for the first 2 feedings after birth. Subsequently, calves were fed 1.9 L of MR, TM, or MCR 3 times per day for 3 d (starting on d 2). After initial diets ended, calves were fed and managed similarly. Body weights (d 1, 7, 14, 21, and 56), blood samples (d 1, 7, 14, and 21), and daily health scores (scale of 0 to 3, with 0 representing normal or healthy and 3 representing severe symptoms or ill) were collected through weaning at 56 d. All except 1 calf achieved successful transfer of passive immunity, with serum IgG values greater than 10.0 mg/mL. From birth through weaning, calves fed TM and MCR gained 3 kg more total body weight than those fed MR (34.3, 34.3, and 31.3 kg, respectively). Increased metabolizable energy (using NRC 2001 recommendations) in TM accounts for 0.68 kg of the increased gain compared with MR. Treatment did not alter health scores for ears, eyes, or feces. Haptoglobin concentrations were lower in TM and MCR than in MR calves (4.63, 3.62, and 7.54 µg/mL, respectively), whereas lipopolysaccharide binding protein concentrations were not different. In conclusion, compared with MR alone, feeding TM or MR with colostrum replacer for 3 d increased growth rate of calves throughout the preweaning period.

microRNAs (miRNAs) are conserved non-coding small nucleotide molecules found in nearly all species and breastmilk. miRNAs present in breastmilk are very stable to freeze-thaw, RNase treatment, and low pH as they are protected inside exosomes. They are involved in regulating several physiologic and pathologic processes, including immunologic pathways, and we have demonstrated better immune response to vaccines in piglets fed with human milk (HM) in comparison to dairy-based formula (MF). To understand if neonatal diet impacts circulatory miRNA expression, serum miRNA expression was evaluated in piglets fed HM or MF while on their neonatal diet at postnatal day (PND) 21 and post-weaning to solid diet at PND 35 and 51. MF fed piglets showed increased expression of 14 miRNAs and decreased expression of 10 miRNAs, relative to HM fed piglets at PND 21. At PND 35, 9 miRNAs were downregulated in the MF compared to the HM group. At PND 51, 10 miRNAs were decreased and 17 were increased in the MF relative to HM suggesting the persistent effect of neonatal diet. miR-148 and miR-181 were decreased in MF compared to HM at PND 21. Let-7 was decreased at PND 35 while miR-199a and miR-199b were increased at PND 51 in MF compared to HM. Pathway analysis suggested that many of the miRNAs are involved in immune function. In conclusion, we observed differential expression of blood miRNAs at both PND 21 and PND 51. miRNA found in breastmilk were decreased in the serum of the MF group, suggesting that diet impacts circulating miRNA profiles at PND 21. The miRNAs continue to be altered at PND 51 suggesting a persistent effect of the neonatal diet. The sources of miRNAs in circulation need to be evaluated, as the piglets were fed the same solid diet leading up to PND 51 collections. In conclusion, the HM diet appears to have an immediate and persistent effect on the miRNA profile and likely regulates the pathways that impact the immune system and pose benefits to breastfed infants.

A growing need exists for the development of practical feeding strategies to mitigate methane (CH4) emissions from cattle. Therefore, the objective of this study was to evaluate the influence of milk replacer feeding intensity (MFI) in calves on CH4 emission, rumen development, and performance. Twenty-eight female newborn Holstein calves were randomly assigned to 2 feeding groups, offered daily either 10% of the body weight (BW) in colostrum and subsequently 10% of the BW in milk replacer (MR; 10%-MR), or 12% of the BW in colostrum followed by 20% of the BW in MR (20%-MR). In wk 3, half of each feeding group was equipped with a permanent rumen cannula. Both groups were weaned at the end of wk 12. Hay and calf starter (mixture of pelleted grains) were offered from d 1 until wk 14 and 16, respectively. A total mixed ration was offered from wk 11 onward. Feed intake was measured daily and BW, anatomical measures, and rumen size weekly. Methane production and gastrointestinal passage rate were measured pre-weaning in wk 6 and 9 and post-weaning in wk 14 and 22, with additional estimation of organic matter digestibility. Rumen fluid, collected in wk 1, 2, 3, 6, 9, 14, 18, and 22, was analyzed for volatile fatty acid concentrations. Although the experimental period ended in wk 23, rumen volume of 17 calves was determined after slaughter in wk 34. Data was analyzed using ANOVA for the effects of feeding group, cannulation, and time, if applicable. Dry matter intake (DMI) of solid feed (SF) in 20%-MR animals was lower pre-weaning in wk 6 to 10 but mostly higher post-weaning. From wk 6 onward, anatomical measures and BW were greater in 20%-MR animals, and only the differences in body condition score gradually ceased post-weaning. Following the amount of SF intake, 10%-MR calves emitted more CH4 pre-weaning in wk 9, whereas post-weaning the 20%-MR group tended to have higher levels. Methane emission intensity (CH4/BW) was lower pre-weaning in 20%-MR animals but was comparable to the 10%-MR group post-weaning. Methane yield (CH4/DMI of SF) and estimated post-weaning organic matter digestibility were not affected by MFI. Rumen size normalized to heart girth was greater in 10%-MR calves from wk 5 to 10, but differences did not persist thereafter. In wk 34, rumen volume was higher in 20%-MR calves, but normalization to BW revealed no difference between feeding groups. In conclusion, high MFI reduces CH4 emission from calves pre-weaning, although this effect ceases post-weaning.

Importance: Breast milk substitutes (BMS) are important nutritional products evaluated in clinical trials. Concerns have been raised about the risk of bias in BMS trials, the reliability of claims that arise from such trials, and the potential for BMS trials to undermine breastfeeding in trial participants. Existing clinical trial guidance does not fully address issues specific to BMS trials. Objectives: To establish new methodological criteria to guide the design, conduct, analysis, and reporting of BMS trials and to support clinical trialists designing and undertaking BMS trials, editors and peer reviewers assessing trial reports for publication, and regulators evaluating the safety, nutritional adequacy, and efficacy of BMS products. Design, Setting, and Participants: A modified Delphi method was conducted, involving 3 rounds of anonymous questionnaires and a face-to-face consensus meeting between January 1 and October 24, 2018. Participants were 23 experts in BMS trials, BMS regulation, trial methods, breastfeeding support, infant feeding research, and medical publishing, and were affiliated with institutions across Europe, North America, and Australasia. Guidance development was supported by an industry consultation, analysis of methodological issues in a sample of published BMS trials, and consultations with BMS trial participants and a research ethics committee. Results: An initial 73 criteria, derived from the literature, were sent to the experts. The final consensus guidance contains 54 essential criteria and 4 recommended criteria. An 18-point checklist summarizes the criteria that are specific to BMS trials. Key themes emphasized in the guidance are research integrity and transparency of reporting, supporting breastfeeding in trial participants, accurate description of trial interventions, and use of valid and meaningful outcome measures. Conclusions and Relevance: Implementation of this guidance should enhance the quality and validity of BMS trials, protect BMS trial participants, and better inform the infant nutrition community about BMS products.

The milk-fed calf has a requirement for amino acids (AA) instead of crude protein (CP); however, most milk replacers (MR) are still formulated for CP concentrations. Previous work has demonstrated that feeding a modified MR (24:20; CP:fat @ 0.64 kg/d) improved calf growth performance compared with standard (20:20 @ 0.57 kg/d) and accelerated MR (26:16 @ 0.78 kg/d). The 56-d study objective was to determine if feeding an MR formulated using synthetic AA to achieve the AA concentrations of a 24:20 MR while reducing CP results in similar or enhanced growth performance and/or reduce cost compared with standard MR formulations. Eighty 3- to 5-d-old Holstein bull calves received in two lots (40) within the same week were blocked by body weight (BW) and randomly assigned to one of four MR treatments consisting of 20:20 (20), 22:20 (22), 24:20 (24), and a 22:20 having the AA concentrations of the 24, but with reduced CP (22AA). All MR contain decoquinate and were fed at 0.57 kg/calf daily split into 2×/d feeding for 14 d via bucket, increased to 0.85 kg/calf daily in two feedings until 35 d, and then fed 1×/d at 0.41 kg/calf daily with weaning after day 42. Calves were housed in straw-bedded hutches with ad libitum access to water and pelleted calf starter (CS). All data were statistically analyzed as a randomized complete block design with block considered random with week as a repeated measurement. Initial BW was similar (P > 0.10) across all treatments (42.4 ± 2.2 kg). Calves fed 22AA MR demonstrated greater (P < 0.05) BW compared with calves fed the 24 MR, and calves fed the 20 and 22 MR being intermediate and similar (P > 0.10; 78.7, 78.8, 76.5, and 81.8 kg for 20, 22, 24, and 22AA, respectively). CS intake was greater (P < 0.05) for calves fed 22AA (0.74, 0.78, 0.65, and 0.81 kg/d) compared with calves fed the 20 and 24, but similar (P > 0.10) to calves fed 22. Calves fed 24 MR demonstrated the lowest CS intake. This study demonstrates that similar growth performance can be achieved by feeding an AA fortified MR having a lesser CP concentration, which might reduce feed costs.

Successful passive transfer of antibodies in neonatal calves can be achieved by feeding an adequate quantity and quality of maternal colostrum (MC) or colostrum replacer (CR). An alternative could be feeding low-quality maternal colostrum (LMC) with added IgG from a CR. The objective of this study was to determine if a commercial whey-based CR product containing low levels of casein (Premolac PLUS Bovine IgG; Zinpro Corporation, Eden Prairie, MN) fed to replace MC or supplement LMC could lead to adequate serum IgG levels and apparent efficiency of absorption (AEA) in neonatal dairy calves. Holstein calves (n = 20 per treatment) were separated from their dam after birth and randomly assigned to be fed 3.79 L of MC (106 g/L of IgG; 401 g of IgG fed), LMC (30 g/L IgG) supplemented with CR (41 g/L IgG; 154 g of IgG total fed; LMC-CR), or 1.3 L of 1 of 2 levels of CR (110 or 150 g of IgG fed; CR-110 or CR-150) within 1.5 h of birth. Colostrum was obtained from the first (MC) or second and third milkings (LMC) of cows from Pennsylvania State University dairy and pooled by source into large batches. Blood samples were taken from calves before colostrum feeding and 24 h after birth and were analyzed for serum total protein, total IgG, hematocrit, and Brix percentage. Calves fed MC had higher 24-h IgG values (means ± SEM) than calves fed LMC-CR (27.04 ± 1.07 vs. 22.33 ± 1.08 mg/mL, respectively). Feeding 150 g of IgG from CR led to higher 24-h serum IgG values than feeding 110 g of IgG (16.90 ± 1.09 vs. 12.79 ± 1.08 mg/mL). Serum IgG levels were different between the CR-fed calves and the calves fed LMC-CR and MC, but all had average values >10 mg/mL IgG. Calves fed LMC-CR had greater AEA than calves fed MC (54.58 ± 2.39 vs. 24.38 ± 2.36%, respectively). Among calves fed CR-110 or CR-150, AEA did not differ. Serum total protein and Brix percentage had strong correlations with actual IgG values across the entire study. We found no differences in average daily gain or health variables measured, and no differences in final hip width, withers height, or body weight for calves fed MC, LMC-CR, CR-150, or CR-110. These results indicate that CR can be fed successfully as an alternative to MC or as a supplement to colostrum with low IgG.

The aim of this study was to evaluate effects of milk replacer (MR) feeding rate and processing of corn in calf starter (CS) on growth performance, nutrient digestibility, and rumen and fecal fibrolytic bacteria in dairy calves. Holstein male calves (n = 48, 2-3 d of age) were randomly assigned to 1 of 4 treatments with a 2 × 2 factorial arrangement of MR level of 0.749 kg of MR/d (LO) or up to 1.498 kg of MR/d (HI); and whole corn or flaked corn in textured CS. Calves were weaned by reducing MR offered by 50% during wk 6. Intakes of MR and CS were recorded daily, whereas body weight (BW) was measured weekly. Rumen fluid and fecal matter were collected at wk 5 and 8 to quantify fibrolytic bacteria and nutrient digestibility. Data were analyzed as a completely randomized design using mixed model ANOVA. Repeated measures were used as appropriate. Calves fed HI had greater average daily gain than calves fed LO at wk 2, 3, 4, and 5, yet at wk 7 calves fed HI had lower average daily gain compared with calves fed LO. Starter intake was greater for calves fed LO compared with HI at wk 4, 5, 6, and 7. During wk 5 and 8, calves fed LO had increased ADF and NDF digestibility compared with calves fed HI. During wk 5, dry matter and organic matter digestibility were lower for LO-fed calves compared with HI-fed calves, but during wk 8 the opposite was observed, with HI-fed calves having lower dry matter and organic matter digestibility than LO-fed calves. At wk 5, Clostridium cluster IV and Butyrivibrio fibrisolvens proportions in rumen fluid tended to be higher and Clostridium cluster IV, Fecalibacterium sp., and Prevotella sp. proportions in fecal matter were higher in calves fed LO compared with HI. From wk 8 to 16, dry matter intake was unaffected by treatment; however, energy efficiency was greater in calves fed LO, causing LO calves to have higher BW gain during this period. Greater starter digestibility was observed for calves fed LO versus HI in concert with increased fibrolytic bacteria proportions (wk 5) in fecal and rumen samples, which resulted in greater postweaning BW gain and similar BW and frame measurements by 16 wk of age. Overall the results show that rate of MR feeding has a larger effect than the processing of corn in CS on performance, fiber digestibility, and rumen and fecal fibrolytic bacterial communities.

Newborn Holstein male calves (n = 50) born on a single dairy farm were assigned randomly at birth to receive 3 feedings of 1.8 L of pooled maternal colostrum (MC) at 1, 6, and 12 h of age or 1 feeding of 500 g of a colostrum replacer reconstituted to 1.8 L at 1 h of age, followed by 2 feedings of 227 g of a commercial milk replacer (MR) reconstituted to 1.8 L at 6 and 12 h of age (CR). All feedings were administered by esophageal feeder. At 2 to 3 d of age, calves were transported to the experimental facility and assigned within colostrum group to receive 0.66 kg/d dry matter (DM) of MR to 39 d, and then 0.33 kg/d to 42 d (MRM) or 0.77 kg/d of MR DM to d 13, 1.03 kg/d for 22 d, and 0.51 kg/d for 7 d (MRH). The MR contained 25.8% crude protein and 17.6% crude fat (DM basis) and was based on whey proteins and lard as the primary fat source. Calf starter (21.7% crude protein, 15.7% neutral detergent fiber, 37.4% starch, DM basis) and water were available for ad libitum consumption throughout the 56-d study. Serum IgG and total protein were measured at 2 to 3 d of age. Intakes of MR and calf starter were monitored daily. Calf health and fecal scores were also monitored daily. Body weight was measured weekly, and hip width and body condition score were monitored every 2 wk. Digestion of DM, organic matter, crude protein, and ether extract were determined at 1 and 3 wk from 5 calves randomly selected within treatment and using chromic oxide as a digestibility marker added to the MR. Calves fed CR had lower serum IgG and total protein than calves fed MC. Also, calves fed CR grew more slowly, consumed less calf starter, and were less efficient to 56 d than calves fed MC. The number of days calves were treated with veterinary medications was higher when calves were fed CR. Calves fed MC-MRH gained more BW than other calves from 3 to 8 wk of age. Calves fed CR-MRH consumed less calf starter than other calves during wk 7 and 8. Digestion of nutrients at 1 and 3 wk of the study was unaffected by type of colostrum or level of MR fed and did not change from 1 to 3 wk. Over the first 2 mo of life, the calves fed MRH consumed less calf starter than calves fed MRM, but average daily gain or hip width change did not differ. One feeding of CR followed by 2 feedings of MR in the first 24 h likely reduced absorption of IgG from CR and contributed to differences in health and growth. Differences in animal performance observed in this study were unrelated to MR digestibility.

Growth and the digestibility of nutrients can be greatly affected by diet preweaning and the rate at which calves are weaned. A 2 × 2 factorial design [moderate (MOD) or high (HI) milk replacer (MR) feeding rates and abrupt (AB) or gradual (GR) weaning] was used to compare these effects. Calves (n = 50) were randomly assigned to 1 of 4 treatments: MOD-AB, MOD-GR, HI-AB, and HI-GR. Calves assigned to MOD-AB were fed 0.66 kg of MR for the first 42 d and then 0.33 kg for the last 7 d; those assigned to MOD-GR were fed 0.66 kg of MR for 28 d, 0.33 kg for 14 d, and 0.17 kg for the last 7 d; those assigned to HI-AB were fed 0.66 kg of MR for 7 d, 0.82 kg for 7 d, 1.1 kg for 28 d, and 0.66 kg for the last 7 d; and those assigned to HI-GR were fed 0.66 kg of MR for 7 d, 0.82 kg for 7 d, 1.1 kg for 14 d, 0.66 kg for 14 d, and 0.33 kg for the last 7 d. All calves received the same MR [25% crude protein (CP), 17% fat; dry matter (DM) basis] and were given ad libitum access to water and a textured starter (42% starch and 20% CP). On d 26 to 30 and d 45 to 49, a fecal sample was taken from 5 calves in each treatment via the rectum to estimate apparent digestibility coefficients (dC). Apparent dC of DM, organic matter, and fat were greater for HI versus MOD calves. Apparent dC of neutral detergent fiber, acid detergent fiber, and sugar were greater for MOD versus HI calves. Apparent dC of DM was greater for AB versus GR calves [90.9, 89.0 ± 0.5384 (standard error)], and the apparent dC of acid detergent fiber was greater for GR versus AB calves (39.5, 32.3 ± 1.67). Feed efficiency was greater for HI versus MOD and AB versus GR. There were no significant differences between CP or starch dC based on treatment, and no interactions were observed. Starter consumption was greater for MOD calves compared with HI calves, and GR calves consumed more than AB calves. These results suggest that providing calves high amounts of MR preweaning enhanced readily available nutrient dC, but providing moderate amounts of MR resulted in increased fibrous fraction dC. Accordingly, the AB weaning strategy had higher dC for DM and organic matter, but there was a depression in fiber dC.

The aim of this study was to determine the effect of docosahexaenoic acid-rich algae (DHA-RA) supplementation in milk replacer (MR) on performance, selected cytokine expression in lymphocytes, and blood immunoglobulin concentration in newborn dairy calves. Forty female Holstein-Friesian calves (8.6 ± 0.8 d old and 41.1 ± 4.3 kg; mean ± standard deviation) were blocked by date of birth and allocated into 4 experimental groups (10 animals/group): (1) not supplemented with DHA-RA, (2) supplemented with 9 g of DHA-RA/d in MR, (3) supplemented with 18 g of DHA-RA/d in MR, and (4) supplemented with 27 g of DHA-RA/d in MR. Milk replacer was fed in an amount equal to 900 g of MR powder/d (as fed), 2 times a d, for 49 d. Starter mixture (SM) was fed ad libitum beginning on d 15 of the study. Each calf was in the study over a period of 49 d. The MR and SM intake and fecal score were recorded daily and body weight was recorded weekly. Blood samples were collected before the morning feeding, at the beginning of the study, every consecutive week, and at the end of the study for morphology and smear analysis, serum immunoglobulin level (IgG, IgA, and IgM), and lymphocyte isolation. The mRNA isolated from lymphocytes was checked for TNFα, IL-1ß, IL-6, and p65 expression. Average daily gain between d 1 to 14 of the study increased quadratically with increasing dose of DHA-RA. However, average daily gain between d 15 to 49 of the study tended to linearly decrease and over the whole study linearly decreased with increasing dose of DHA-RA. The MR intake decreased linearly between d 1 to 14 of the study and over the whole study, and mean SM intake decreased quadratically with increasing dose of DHA-RA. Feed efficiency increased quadratically and fecal score decreased quadratically during the first 14 d of the study. Increasing dose of DHA-RA led to cubic changes in feed efficiency and fecal score between d 15 and 49 of the study. Overall, over the whole study period a tendency was observed for lower fecal score for the DHA-RA supplemented groups. Interleukin-1ß mRNA expression decreased linearly, whereas the mRNA expression of p65 and TNFα as well as serum IgG concentration tended to decrease linearly with increasing dose of supplemental DHA-RA. No effect of group was found on IgA and IgM serum level and the majority of blood parameters. Altogether, treatment worsened production variables but seemed to have a beneficial effect on the immune system of calves.

The objectives of this study were to evaluate calf performance and diet digestibility when feeding a milk replacer (MR; 25% crude protein, 18% fat, dry matter basis) at a moderate (MOD) and high (HI) rate, along with MOD with neomycin-oxytetracycline inclusion (1.43 each g/kg; NTMOD). Male Holstein calves (n = 48; 45 ± 1.0 kg of body weight; 3 to 4 d of age) were housed in individual pens for 56 d. All calves were initially fed 0.66 kg of dry matter from MR for the first p.m. and following a.m. feeding, then randomly assigned to 1 of 2 feeding programs: (1) 0.66 kg of dry matter/d for first 39 d divided into 2 equal a.m. and p.m. meals and 0.33 kg for 3 d fed once daily in the a.m. feeding only (MOD, NTMOD); (2) 0.85 kg of dry matter/d for the first 4 d divided into 2 equal a.m. and p.m. meals, 1.07 kg of dry matter/d for the next 31 d divided into 2 equal a.m. and p.m. meals, and 0.48 kg for 7 d fed once daily in the a.m. feeding only (HI). Calves were fed a textured starter containing whole grains (20% crude protein, 43% starch, dry matter basis). Fecal samples were collected for 5 individual calves per treatment between d 47 to 51 to estimate digestibility. Calves were moved into groups by treatment (4 calves/pen) at 56 d and fed the same starter blended with 5% hay until d 112. Data were analyzed as a completely randomized design with repeated measures when applicable using mixed models. Milk replacer dry matter intake averaged 27.4 kg for MOD and NTMOD and 40.3 kg for HI per calf. Preweaning starter intake was less for calves fed HI versus MOD. Preweaning calf BCS change was greater for calves fed HI versus MOD. Preweaning medical treatment days were fewer for calves fed NTMOD versus MOD. Postweaning starter intake was less for calves fed HI versus MOD. Postweaning average daily gain was less for calves fed NTMOD versus MOD. Over the 56-d nursery trial, calves fed HI had less starter intake than calves fed MOD. Digestibility of dry matter, organic matter, acid detergent fiber, and neutral detergent fiber were greater for calves fed MOD versus NTMOD or HI during d 47 to 51 postweaning. Additionally, digestibility of fat was greater for calves fed MOD versus HI. During d 56 to 112, hip width change was greater for calves fed MOD versus NTMOD or HI. Over 0 to 112 d, body weight gain was 100, 98, and 102 kg, whereas hip width change was 10.2, 9.0, and 9.2 cm for calves fed MOD, NTMOD, and HI, respectively. Any preweaning improvements from feeding NTMOD or HI over MOD were lost during the postweaning period, which could be partially explained by reductions in diet digestibility.

Selection for prolificacy in modern pig farming has resulted in increasing litter sizes. Since rearing large litters is challenging, artificial rearing of piglets with a milk replacer is an alternative strategy. It is hypothesized that the development of the piglets' mucosa-associated lymphoid tissues (MALT) is affected by these artificial conditions. Therefore, the stereologically estimated volumes of the tonsil of the soft palate, and the lingual, nasopharyngeal and paraepiglottic tonsils, as well as the jejunal and ileal Peyer's patches were statistically compared at day 21 postpartum between six conventionally reared piglets and six piglets that were artificially reared from day 7 onwards. In addition, six 7-day-old sow-fed piglets were examined to evaluate the effect of age. All tonsils and Peyer's patches significantly increased in volume with age. The rearing strategy had no significant effect on the volumes of the tonsil of the soft palate and the lingual tonsil. The former tonsil was by far the largest with a mean volume of 967.2 ± 122.4 mm3 and 822.3 ± 125.4 mm3 in the conventionally and artificially reared piglets, respectively. The lingual tonsil only measured 9.4 ± 6.4 mm3 and 6.3 ± 2.6 mm3 in conventionally and artificially reared groups, respectively. In contrast, the rearing strategy did affect the volumes of the nasopharyngeal and paraepiglottic tonsils, which had a mean volume of 137.1 ± 32.4 mm3 and 84.4 ± 26.9 mm3, and 30.7 ± 7.8 mm3 and 20.0 ± 3.9 mm3 in conventionally and artificially reared piglets, respectively. The rearing strategy did not affect the development of the Peyer's patches. At day 21, the jejunal Peyer's patches of the conventionally and artificially reared piglets presented a volume of 1.6 ± 0.4 cm3 and 1.3 ± 0.2 cm3, respectively. The volumes of the ileal Peyer's patch amounted to 15.1 ± 3.0 cm³ in conventionally reared piglets and 12.0 ± 2.6 cm³ in artificially reared piglets at day 21. The results showed that artificial rearing hampers the morphological development of the tonsils that are exposed to inhaled antigens, but the voluminous lymphoid tissues that sample oral antigens are not influenced. Since it is unlikely that the observed differences in both tonsils are due to the milk replacer, artificial rearing could be a valuable alternative for raising large litters. In addition, the presence of developing MALT in piglets allows for investigating the value of nasal and oral vaccination in this species for human or veterinary purposes.

In recent years, lactose-free and low-lactose infant formulas have been increasingly used. The impact of using different carbohydrates than lactose on later cognition of formula-fed infants remains, however, unknown. We examined the effects of providing formulas containing either digestible maltodextrin or lactose as main carbohydrate source (28% of total nutrient composition) on cognitive performance of piglets. Piglets received the formulas from 1 to 9 weeks of age and, starting at 12 weeks, were individually tested in a spatial holeboard task (n = 8 pens/formula), in which they had to learn and memorize a configuration of baited buckets. After 28 acquisition trials, piglets were subjected to 16 reversal trials in which the location of the baited buckets was changed. Piglets fed the maltodextrin-based formula had higher reference memory (RM) scores than piglets fed the lactose-based formula towards the end of acquisition. During the switch of configuration, piglets offered the maltodextrin-based formula tended to have higher RM scores and make fewer RM errors than piglets offered the lactose-based formula. Working (short-term) memory was not affected by the formulas. Compared to lactose, the use of maltodextrin in milk formulas improved long-term spatial memory of piglets, even weeks after the end of the intervention.

BACKGROUND: Breast milk contains important nutrients and immunoregulatory factors that are essential for newborn infants. Recently, epidemiological studies suggested that breast-feeding prevents a wide range of infectious diseases and lowers the incidence of infant allergic diseases. METHODS: To examine the effects of breast milk on immunological development in infancy, we established an artificial rearing system for hand-feeding mice and compared mouse pups fed with either breast milk or milk substitute. All mice were killed at 14 days of age and immune cells in the thymus, spleen, and small intestine were examined on flow cytometry. RESULTS: The number of thymocytes was higher whereas that of total immune cells of peripheral lymphoid tissues was lower in mice fed breast milk compared with milk substitute-fed mice. In peripheral lymphoid tissues, the proportion of B cells was higher and that of CD8+ T cells, macrophages, dendritic cells, and granulocytes was significantly lower in breast milk-fed mice. The same alteration in immune cells of the thymus and peripheral lymphoid tissues in milk substitute-fed mice was also observed in pups reared by mother mice treated with anti-transforming growth factor-ß (anti-TGF-ß) monoclonal antibody. CONCLUSIONS: Breast milk regulates the differentiation and expansion of innate and adaptive immune cells partly due to TGF-ß. Hence, TGF-ß in breast milk may be a new therapeutic target for innate immune system-mediated diseases of infancy.

Cow's milk allergy is among the more frequent food allergies in infants and children. Because its suspicion stems from a plethora of symptoms, it is frequently reported. However, the development of a rigorous diagnostic pathway will reduce the diagnosed children to less than 50% of those reported. Cow's milk allergy is the only specific food allergy for which an EBM guideline exists. According to the guidelines (Diagnosis and Rationale for Action against Cow's Milk Allergy), a diagnostic process based on the pre-test probability of this condition is available. Treatments include avoidance, the substitution of cow's milk with an appropriate formula, and in some cases, oral immunotherapy. Treatment choice is also guided by these guidelines.

BACKGROUND: The objective of this randomized controlled trial was to determine the effect of feeding a commercial lacteal-derived colostrum replacer (CR) or pooled maternal colostrum (MC) on preweaning morbidity, growth and mortality in Holstein heifer calves. A total of 568 calves were randomly assigned to be fed either 3.8 L of pooled MC or two doses (200 g IgG) of a CR. Calves were monitored daily for preweaning morbidity until weaning at 60 d old. Birth and weaning weights were measured to estimate growth rates. RESULTS: Calves fed CR were significantly less likely to be affected with a diarrhea event (OR = 0.58; 95% CI, 0.38 to 0.88; P value = 0.011) and had a higher rate of daily weight gain (0.051 kg/day; 95% CI, 0.03 to 0.08; P value <0.001) compared to calves fed pooled MC. Use of lacteal-derived colostrum replacer was not significantly associated with respiratory disease (OR = 1.01; 95% CI 0.67 to 1.51; P value = 0.974 ), omphalitis (OR = 0.93; 95% CI 0.06 to 14.86; P value = 0.956), or mortality (HR = 0.71; 95% CI 0.27 to 1.92; P value = 0.505) in the study calves. CONCLUSIONS: The lacteal-derived CR fed at the study dose was a viable colostrum alternative in the event of poor quality pooled MC for the prevention of preweaning diarrhea and resulted in higher growth rates in comparison to calves fed pooled MC in the study herd.

This study was undertaken to assess the effect of milk replacer (MR) containing Lactobacillus acidophilus and a mix of Bifidobacterium animalis subsp. lactis and Bifidobacterium longum subsp. longum on lamb immune response and on lamb meat quality. A 6-week-trial was conducted on 40 male Comisana lambs, divided into four groups, fed maternal milk (MM), MR, MR with L. acidophilus supplementation (MRL) and MR with a mix (1 : 1) of B. animalis subsp. lactis and B. longum subsp. longum supplementations (MRB). Lambs fed MR containing a mix of bifidobacteria showed the highest in vivo cellular immune response to phytohemagglutinin, whereas MM and MRB showed the highest antibody response to ovalbumin. At day 11 of the trial, MRL displayed the highest value of Interleukin-10; differences disappeared among groups subsequently. Blood cholesterol levels in lambs fed MR containing L. acidophilus was almost halved compared with that found in MM and MR groups. Meat from artificially reared lambs was characterized by trans-11 18:1 and total conjugated 18:2n-6, whereas meat from the dam-suckled lambs was characterized by 14:0, cis-9 14:1 and 16:0. Polyunsaturated to saturated fatty acid ratio was higher in meat of MR, MRL and MRB than in MM lambs. Meat from artificially reared lamb fed MR containing probiotics showed an improved fatty acid profile for human diet.

In order to evaluate the effects of an elevated amount of modified milk replacer on body weight, daily gain, starter intake, plasma endocrine parameters and expression of nutrient transporters in small intestinal epithelia, Holstein bull calves (n=24) were fed for 60days either with the usual amount of 24% crude protein (CP) and 20% fat milk (CF) replacer (C group), or with a double amount of a modified milk replacer of 28% CP and 16% CF (E group). Body weight from D20 to D60 and daily gain before D40 was greater or tended to be greater for the E group than the C group. Plasma concentrations of insulin-like growth factor-1 (IGF-I) and insulin were greater for the E group than the C group on D28 but not on D56, without changing plasma growth hormone levels. Gene expression for sodium-dependent glucose transporter 1 and fatty acid translocase (CD36) was altered in day- and intestine-dependent manners. From these findings, we conclude that an elevated intake of milk replacer given up to 40days old is sufficient to enhance body weight, which may be associated with increased plasma IGF-I concentrations, in Holstein bulls.

P-glycoprotein (Pgp), a product of the multi-drug resistance gene MDR1a, is a broad specificity efflux ATP cassette transmembrane transporter that is predominantly expressed in epithelial tissues. Because mdr1a(-/-) mice tend to develop spontaneous colitis in bacteria-dependent manner, Pgp is believed to have a role in protection of the intestinal epithelium from luminal bacteria. Here we demonstrate that levels of Pgp in the small intestine of newborn rodents dramatically increase during breastfeeding, but not during formula feeding (FF). In rats and mice, levels of intestinal Pgp peak on days 3-7 and 1-5 of breastfeeding, respectively. The mdr1a(-/-) neonatal mice subjected to FF, hypoxia, and hypothermia have significantly higher incidence and pathology, as well as significantly earlier onset of necrotizing enterocolitis (NEC) than congenic wild type mice. Breast-fed mdr1a(-/-) neonatal mice are also more susceptible to intestinal damage caused by the opportunistic pathogen Cronobacter sakazakii that has been associated with hospital outbreaks of NEC. Breast milk, but not formula, induces Pgp expression in enterocyte cell lines in a dose- and time-dependent manner. High levels of ectopically expressed Pgp protect epithelial cells in vitro from apoptosis induced by C. sakazakii. Taken together, these results show that breast milk-induced expression of Pgp may have a role in the protection of the neonatal intestinal epithelium from injury associated with nascent bacterial colonization.

OBJECTIVE: To evaluate longevity, milk production, and breeding performance in adult Holstein cows fed either a plasma-derived commercial colostrum replacer (CR) or raw bovine maternal colostrum (MC) at birth. DESIGN: Randomized controlled clinical trial. ANIMALS: 497 heifer calves born in 12 commercial dairies located in Minnesota and Wisconsin. PROCEDURES: All calves were separated from their dams within 30 to 60 minutes after birth and systematically assigned to be fed either MC (control group [n = 261 calves]) or CR (treatment group [236]). Calves were observed from birth up to adulthood (approx 54 months old), during which time death and culling events plus milk yield and breeding performance data were collected. Time to death, time to culling, time to death or culling combined, time to first calving, and time to conception intervals were evaluated by use of proportional hazards survival analysis models. Number of times inseminated per conception and lifetime milk yield (up to 54 months old) were evaluated by use of general linear models. RESULTS: Cows fed CR as calves at the time of birth were no different than cows fed MC as calves with respect to overall risk of death, culling, or death or culling combined (from birth to 54 months of follow-up and from first calving to 54 months old); lifetime milk yield; and breeding performance. CONCLUSIONS AND CLINICAL RELEVANCE: No difference was detected in overall risk of death or culling, milk production, or reproductive performance between cows fed CR and those fed MC as calves at birth.
