ABSTRACT

The endemic chub Squalius tenellus (Heckel, 1843) was introduced more than 100 years ago to Lake Blidinje (Bosnia-Herzegovina). Only 1 species of enteric helminth was found in a sample of 35 chubs, the tapeworm Caryophyllaeus brachycollis (Janiszewska, 1953). The paper includes histopathological investigation with identification of innate immune cells involved in host reaction and molecular data allowed correct designation of the cestode species. Of 35 specimens of chub examined, 21 (60%) harboured individuals of C. brachycollis and a total of 1619 tapeworms were counted, the intensity of infection ranged from 1 to 390 worms per fish (46.2 ± 15.3, mean ± s.e.). Histopathological and ultrastructural investigations showed strict contact between the worm's body and the epithelia and increase in the number of mucous cells, rodlet cells among the epithelial cells. Within the tunica propria-submucosa, beneath the site of scolex attachment, numerous neutrophils and mast cells were noticed. This is the first study of the occurrence of C. brachycollis in chub from Lake Blidinje and on the response of the innate immune cells of S. tenellus to this tapeworm. Interestingly, in 3 very heavily infected chubs, perforation of the intestinal wall was documented; this is uncommon among cestodes which use fish as a definitive host.

ABSTRACT

Sculpins (coastrange and slimy) and sticklebacks (ninespine and threespine) are widely distributed fishes cohabiting 2 south-central Alaskan lakes (Aleknagik and Iliamna), and all these species are parasitized by cryptic diphyllobothriidean cestodes in the genus Schistocephalus. The goal of this investigation was to test for host-specific parasitic relationships between sculpins and sticklebacks based upon morphological traits (segment counts) and sequence variation across the NADH1 gene. A total of 446 plerocercoids was examined. Large, significant differences in mean segment counts were found between cestodes in sculpin (mean = 112; standard deviation [s.d.] = 15) and stickleback (mean = 86; s.d. = 9) hosts within and between lakes. Nucleotide sequence divergence between parasites from sculpin and stickleback hosts was 20.5%, and Bayesian phylogenetic analysis recovered 2 well-supported clades of cestodes reflecting intermediate host family (i.e. sculpin, Cottidae vs stickleback, Gasterosteidae). Our findings point to the presence of a distinct lineage of cryptic Schistocephalus in sculpins from Aleknagik and Iliamna lakes that warrants further investigation to determine appropriate evolutionary and taxonomic recognition.

ABSTRACT

BACKGROUND: The common cat tapeworm, Hydatigera taeniaeformis, is widely distributed across many regions of the world but its global genetic diversity, ecology, and other biological features are not fully understood. The predator-prey relationship of felids and rodents sustains the transmission in both sylvatic and synanthropic or urban settings. This study was carried out in three urban market centres in Tamale, Ghana to determine the prevalence of H. taeniaeformis infection in urban rats and characterise the local population genetics using the mitochondrial nad1 gene sequences. METHODS: From January to May 2023, rats were captured and euthanized to identity and isolate the H. taeniaeformis metacestodes. The full mitochondrial nad1 gene was amplified, sequenced and used to characterise the cestode population. RESULTS: In total 64/80 rats from the three market centres were found naturally infected with metacestodes of H. taeniaeformis. Averagely, infected rats harboured two cysts with one exceptional case of 25 cysts in one liver. Each cyst harboured one metacestode. The DNA sequences revealed four haplotypes in a star-like distribution network across the three market centres. All four haplotypes were found to be unique, clustered with H. taeniaeformis sensu stricto (s.s.) global isolates and had only marginal variation among themselves. CONCLUSION: This is the first account of the genetic variability within H. taeniaeformis in Ghana and would contribute to ongoing and future studies on African regional biogeographic distribution of the common cat tapeworm.

ABSTRACT

Despite the importance of the electric catfish (Malapterurus electricus) and the African giant catfish (Heterobranchus bidorsalis) in the foodweb of Lake Nasser, Egypt, little is known about their diseases and parasitic fauna. This work describes, for the first time, cestodiasis in M. electricus and H. bidorsalis. Corallobothrium solidum and Proteocephalus sp. were identified morphologically and molecularly from M. electricus and H. bidorsalis, respectively. Using PCR, sequencing, and phylogenetic analysis, the two cestodes shared rRNA gene sequence similarities yet were unique and the two new sequences for the proteocephalid genera were submitted to the GenBank database. The prevalence of infection was 75% and 40% for the two fish species, respectively. Infections significantly increased in the summer and spring and were higher in female fish than in male fish. The intestine was the preferred site of the two adult cestodes. However, in the case of C. solidum some larval cestodes were found outside the intestine in between the skin and abdominal musculature, attached to the mesentery, and within intestinal tunica muscularis. Desquamation of the intestinal epithelium and inflammation at the site of infection in addition to congestion of the intestinal wall of the tapeworm infected fish were evident, indicating that C. solidum and Proteocephalus sp. impacted the infected fish. The larval stages of C. solidum attempted to penetrate the intestine and sometimes they were encircled within fibrous layers infiltrated with inflammatory cells. The infected fish's musculature was free of cestode infections. Preventive measures should be implemented to prevent the spread of infections.

ABSTRACT

This study investigated the prevalence, morphology, molecular identification, and histopathological effects of larval tapeworms (plerocercoids) infecting the skeletal muscles of the Indian halibut (Psettodes erumei) collected from the coastal waters of the Arabian Gulf. Numerous oval or round blastocysts, measuring 13-26 mm, were found embedded within the muscular tissues of the Indian halibut, rendering the fish unsuitable for human consumption. Morphological and molecular analyses identified the plerocercoids as Dasyrhynchus giganteus (family Dasyrhynchidae), with an overall prevalence of 15.4%. The seasonal prevalence was the highest in summer (14.6%), followed by spring (10.6%), winter (4.4%), and autumn (3.5%). Infection rates increased with fish size. Histopathological examination revealed fibrous connective tissue capsules surrounding the larvae, causing muscular atrophy and degenerative changes, with few inflammatory eosinophilic cells. Molecular and phylogenetic analysis of the 28S rDNA gene sequences confirmed the specimens as D. giganteus, clustered closely with other sequences of D. giganteus with 100% bootstrap values. This study provided valuable insights into the parasitic infection dynamics, seasonal variation, molecular identification, and histopathological effects, highlighting the importance of monitoring fish for food safety and public health implications.

ABSTRACT

Diphyllobothriosis, a fish-borne zoonosis in South America, is mainly caused by the Pacific broad tapeworm Adenocephalus pacificus Nybelin, 1931, a parasite of considerable concern in fishery resources due to its impact on public health. A new diphyllobothrid, Diphyllobothrium sprakeri Hernández-Orts et al. Parasites Vectors 14:219, 2021, was recently described from sea lions from the Pacific Coast, but marine fish acting as intermediate hosts are unknown. The objective of this study was to confirm the presence of plerocercoid larvae of Diphyllobothriidae Lühe, 1910 (Cestoda: Diphyllobothriidea) in nine fish species of commercial importance in Peru. Of a total of 6999 fish (5861 Engraulis ringens, 853 Sciaena deliciosa, 6 Sciaena callaensis, 171 Scomber japonicus, 40 Trachurus murphyi, 40 Ariopsis seemanni, 18 Merluccius peruanus, 5 Sarda chiliensis, and 5 Coryphaena hippurus), 183 were infected with plerocercoid larvae, representing a total prevalence of 2.61% and a mean intensity of 3.2. Based on mtDNA cox1 sequences of 43 plerocercoids, a phylogenetic analysis revealed that 41 belong to A. pacificus and two to D. sprakeri. These findings are first molecular data for D. sprakeri larvae, and the infections of E. ringens and T. murphyi by plerocercoid larvae represent the first records of intermediate/paratenic hosts for this species. Hence, the findings of the current study enhance our understanding of the presence of diphyllobothriid species in commercial fish from the Southeastern Pacific Ocean and their potential impact on seafood safety for local human populations.

ABSTRACT

A comparative analysis of taxonomic diversity on shrew cestodes among four islands in the Sea of Japan and the Sea of Okhotsk (Sakhalin, Kunashir, Hokkaido, and Moneron) was performed. Cestode species shared among the islands were identified and their host specificity was investigated. On Sakhalin Island, 33 species of the families Hymenolepididae, Dilepididae and Mesocestoididae were recorded in four shrew species (Sorex caecutiens, S. gracillimus, S. minutissimus and S. unguiculatus). In S. caecutiens, S. gracillimus, and S. unguiculatus on Kunashir Island, 22 species of the same families were found and, on Hokkaido Island, 23 species of the families Hymenolepididae and Dilepididae were recorded. On Moneron Island, three species of cestodes were registered in S. tundrensis. The Sakhalin-Hokkaido-Kunashir complex of shrew cestodes includes eastern-Palearctic, trans-Palearctic and endemic species. High endemism (~22%) of shrew tapeworms in the Sakhalin-Kunashir-Hokkaido Islands was noted as compared to continental territories. The different numbers of cestode species in S. unguiculatus (31), S. caecutiens (29), S. gracillimus (19) and S. minutissimus (1) were found. It was concluded that the cestodes species diversity of shrews of Sakhalin-Kunashir-Hokkaido depended primarily on the history of island formation, their modern physical and geographical features, the abundance of definitive and intermediate cestodes hosts and, to a lesser extent, on the size and remoteness of the islands from the mainland and the diversity of host species.

ABSTRACT

The tapeworms of Moniezia spp. are heteroxenous parasites and their adult forms occur in ruminants' alimentary tract. They steal a significant portion of hosts' nourishment initiating monieziasis, thereby inflicting economic losses in animal rearing. Despite their high economic importance, the molecular characterization and taxonomic status of these parasites have remained poorly understood. In the present study, cestodes were isolated from the sheep and goats' intestines and were stained with Gower's carmine. Upon careful evaluation of morphological characters, 2 species Moniezia denticulata and Moniezia expansa were identified. The genomic DNA was extracted and polymerase chain reaction (PCR) amplified targeting regions of mitochondrial cytochrome c oxidase subunit 1 (cox1), small subunit ribosomal RNA (SSU rRNA) and internal transcribed spacer 1­5.8S rRNA (ITS1­5.8S rRNA) genes followed by sequencing. The partial sequences of cox1, SSU rRNA and ITS1­5.8S rRNA genes of M. denticulata generated in the present study revealed that even though they share high similarities with M. benedeni (93.2% cox1; 92.6% SSU rRNA; 84.70% ITS1­5.8S rRNA) and M. expansa (88.85% cox1; 92.27% SSU rRNA; 81.70% ITS1­5.8S rRNA), they are not identical to them. In the maximum likelihood phylogenetic trees, M. denticulata and M. expansa consistently appeared as distinct species from each other. The high values of pairwise divergence between these 2 species collected in the present study confirmed their separate identity. The present study reports the first molecular characterization of M. denticulata with reference to M. expansa infecting sheep and goats in India.

ABSTRACT

A lack of accurate information on the prevalence and distribution of Anoplocephala spp. infections on horse farms has led to insufficient attention to tapeworm control and increasing horse anoplocephaloses in Europe. Our study aimed to examine the occurrence of Anoplocephala spp. infection using coprological, serum- and saliva-based antibody detection methods and to analyze the risk factors associated with tapeworm infection in domestic horses in Slovakia. Fecal, serum, and saliva samples were collected from 427 horses from 31 farms in Slovakia. Additionally, a questionnaire study was conducted to collect information on tapeworm distribution on horse farms and analyze risk factors associated with infection. Fecal samples were examined by the mini-FLOTAC and the double centrifugation/combined sedimentation-flotation techniques. Serum and saliva samples were analyzed by ELISA to determine antibody levels against Anoplocephala spp. The effects of variables associated with an individual horse were tested for the positive result of the saliva ELISA test on Anoplocephala spp. Cestode eggs were detected in 1.99% of fecal samples (farm prevalence 12.90%), with no differences between the two coprological methods. Serum-based tapeworm ELISA results revealed that 39.39% of horses tested positive (farm prevalence 83.87%); while saliva-based tapeworm ELISA results revealed 56.95% positive horses (farm prevalence 96.77%). Binary logistic regression analysis revealed four meaningful predictors that significantly impacted the likelihood of detecting tapeworm infection in horses: horse age, pasture size, anthelmintic treatment scheme, and access to pasture. The influences of other variables associated with an individual horse were not significantly associated with detecting tapeworm infection.

ABSTRACT

We report molecular identification of an adult Spirometra mansoni tapeworm retrieved from a crab-eating fox (Cerdocyon thous) in Colombia, confirming presence of this parasite in South America. This tapeworm is the causative agent of human sparganosis, commonly reported from Southeast Asia, and represents the second congeneric species with known zoonotic potential in the Americas.

ABSTRACT

Although Joyeuxiella pasqualei is frequently detected in cats from Mediterranean Europe, information on its biology is still scarce. This cestode is relatively less frequently reported in dogs, possibly because it is often misdiagnosed with the better-known Dipylidium caninum. The occurrence of J. pasqualei proglottids in a dog living in a closed environment triggered us to delve into the biology of this cestode by collecting biological samples from lizards and a road-killed cat. Two reptile species, Podarcis siculus (Lacertidae), and Tarentola mauritanica (Geckonidae) were also collected in the garden and its surroundings. In addition, experimental infections with eggs obtained from gravid proglottids were performed in laboratory mice, and Tenebrio molitor (Coleoptera: Tenebrionidae) beetles. Proglottids from the dog's feces and adult cestodes detected at necroscopy of a cat were morphologically identified as J. pasqualei. Two out of 13 T. mauritanica collected in the garden had natural infections of J. pasqualei cysts in the liver and attached to the intestine. All P. siculus lizards and experimentally infected rodents and beetles were negative. DNA sequences obtained from J. pasqualei showed the highest nucleotide similarities with Versteria sp., Echinococcus sp., Raillietina sonini, Taenia polyacantha and D. caninum. Data herein provided show the inability of rodents to become infected by direct ingestion of gravid proglottids, suggesting a need for an invertebrate first intermediate host in the life cycle. Thus, more research study is advocated to better understand the biology of J. pasqualei such as its first intermediate host and its mechanism of transmission in reptiles and rodents.

ABSTRACT

The cestode Schistocephalus solidus is a common parasite in freshwater threespine stickleback populations, imposing strong fitness costs on their hosts. Given this, it is surprising how little is known about the timing and development of infections in natural stickleback populations. Previous work showed that young-of-year stickleback can get infected shortly after hatching. We extended this observation by comparing infection prevalence of young-of-year stickleback from 3 Alaskan populations (Walby, Cornelius and Wolf lakes) over 2 successive cohorts (2018/19 and 2019/20). We observed strong variation between sampling years (2018 vs 2019 vs 2020), stickleback age groups (young-of-year vs 1-year-old) and sampling populations.

ABSTRACT

The tapeworms of fishes (Chondrichthyes and Actinopterygii) account one-third (1670 from around 5000) of the total tapeworm (Platyhelminthes: Cestoda) species diversity. In total 1186 species from 9 orders occur as adults in elasmobranchs (sharks, rays and chimaeras), and 484 species from 8 orders mature in ray-finned fishes (referred to here as teleosts). Teleost tapeworms are dominated by freshwater species (78%), but only 3% of elasmobranch tapeworms are known from freshwater rays of South America and Asia (Borneo). In the last 2 decades, vast progress has been made in understanding species diversity, host associations and interrelationships among fish tapeworms. In total, 172 new species have been described since 2017 (149 from elasmobranchs and 23 from teleosts; invalidly described taxa are not included, especially those from the Oriental region). Molecular data, however, largely limited to a few molecular markers (mainly 28S rDNA, but also 18S and cox1), are available for about 40% of fish tapeworm species. They allowed us to significantly improve our understanding of their interrelationships, including proposals of a new, more natural classification at the higher-taxonomy level (orders and families) as well as at the lower-taxonomy level (genera). In this review, we summarize the main advances and provide perspectives for future research.

Plerocerci of the trypanorhynch cestode Progrillotia dasyatidis Beveridge, Neifar & Euzet, (Progrillotiidae) were isolated from the gallbladder of the three-spined stickleback Gasterosteus aculeatus from a small freshwater marsh in the Lake Atanasovsko Wetlands, a coastal area adjacent to the Black Sea coast, Bulgaria. The parasite was recorded in five out of 134 fish individuals studied (prevalence 3.73%, intensity 1-7, mean intensity 2.40 ± 1.17 and mean abundance 0.09 ± 0.06). A description of the plerocerci is presented, expanding data on intraspecific variation. The present report is the first record of P. dasyatidis from G. aculeatus (new host record) and from Bulgaria (new geographical record). Recording only plerocerci with evaginated scoleces in the sticklebacks is consistent with the hypothesis that teleosts are paratenic hosts and not intermediate hosts of P. dasyatidis.

ABSTRACT

Parasite infracommunities tend to be stochastic in nature, although environmental characteristics such as the type of water source in streams and host traits can have an effect on the biotic assemblages and by extension the parasite fauna. We examined the effect of water source and the rate of adult fish migration on the metazoan parasite infracommunities of conspecific juvenile brown trout, Salmo trutta L. among streams flowing into Lake Lucerne (Switzerland). Juvenile (1 to 2-year old) fish harboured higher parasite species richness in groundwater-fed than in surface water-fed streams, whereas the rate of fish migration did not affect infracommunity richness. Heteroxenous species were more common in groundwater-fed streams with high and medium rates of trout migration, whereas infracommunities in surface water-fed streams and streams with low rates of fish migration were dominated by one monoxenous parasite or lacked infections. Similarity in the parasite infracommunity composition of juvenile trout across streams was explained by the interaction between type of water source and adult migration rates. Our conclusions support that similarity in the parasite composition of resident freshwater conspecifics can be predicted by the local environmental settings and host migratory behaviour, whereas parasite richness is mainly influenced by the environmental characteristics.

ABSTRACT

Helminth infections in wood mice (n = 483), trapped over a period of 26 years in the woods surrounding Malham Tarn in North Yorkshire, were analysed. Although 10 species of helminths were identified, the overall mean species richness was 1.01 species/mouse indicating that the helminth community was relatively depauperate in this wood mouse population. The dominant species was Heligmosomoides polygyrus, the prevalence (64.6%) and abundance (10.4 worms/mouse) of which declined significantly over the study period. Because of the dominance of this species, analyses of higher taxa (combined helminths and combined nematodes) also revealed significantly declining values for prevalence, although not abundance. Helminth species richness (HSR) and Brillouin's index of diversity (BID) did not show covariance with year, neither did those remaining species whose overall prevalence exceeded 5% (Syphacia stroma, Aonchotheca murissylvatici and Plagiorchis muris). Significant age effects were detected for the prevalence and abundance of all higher taxa, H. polygyrus and P. muris, and for HSR and BID, reflecting the accumulation of helminths with increasing host age. Only two cases of sex bias were found; male bias in abundance of P. muris and combined Digenea. We discuss the significance of these results and hypothesize about the underlying causes.

ABSTRACT

Raillietina saudiae is a well-studied avian gastrointestinal parasite belonging to the family Davaineidae and is the most prevalent cyclophyllid tapeworm infecting pigeon in Saudi Arabia. The present study considered as a complementary analysis of Al-Quraishy et al. (2019; Parasitol Int 71, 59-72) with molecular studies for two ribosomal DNA genes employed for precise recognition of this Raillietina species. The annotated partial 18S and 28S rDNA gene regions were found to be 888 and 900 bp long that utilized further to elucidate their genetic relationships at species level using maximum likelihood method. The query sequence of R. saudiae is well aligned and placed within the Davaineidae family, with the same clade of all species of Raillietina that well separated from other cyclophyllidean cestodes especially taeniid and hymenolepid species. Sequence data recorded the monophyly of Raillietina species. The current phylogeny supports the usage of the partial 18S and 28S rDNA genes as reliable markers for phylogenetic reconstructions.

ABSTRACT

Red foxes (Vulpes vulpes) are the most prevalent wild carnivores in the world and definitive hosts of many pathogenic parasites for humans and farm animals. These animals travel great distances in search of prey and nests, and cause contamination of large geographic areas with parasites. For this reason, monitoring the parasitic pathogens of red foxes is particularly important in terms of public and animal health. The goal of this study was to determine the intestinal helminths and molecular characterization of Taenia species of red foxes in Turkey. In this study, 103 red fox intestines obtained from 29 provinces of Turkey were examined with sedimentation and counting technique. Collected helminths were diagnosed according to their morphologic features. Additionally, further molecular analysis (PCR and DNA sequencing) was performed for the identification of Taeniid cestodes. At the end of the study, it was determined that 87.37% (90/103) of red foxes were infected with at least one helminth species. Detected helminths and their prevalence's were Mesocestoides sp. (56.31%), Joyeuxiella echinorhynchoides (33%), Taenia polyacantha (15.53%), Dipylidium caninum (0.97%), Pterygodermatites affinis (51.45%), Toxascaris leonina (45.63%), Uncinaria stenocephala (33%), Oxynema numicidum (20.38%), Toxocara canis (14.56%), Ancylostoma caninum (12.62%), and Trichuris vulpis (1.94%), respectively. Additionally, Pachysentis sp. (37.69%), Centrorhynchus sp. (0.97%) (Acantocephala), and nymphs of Linguatula serrata (20.38%) (Arthropoda) were also detected in the same intestinal samples. This is the most comprehensive study that has been conducted on the intestinal helminthes of red foxes in Turkey. To the best of our knowledge, molecular characterization of T. polyacantha and the detection of O. numicidum, A. caninum, Pachysentis sp., and Centrorhynchus sp. are the first reports in red foxes in Turkey. Our study revealed that red foxes are important hosts for many intestinal helminth species and are link between domestic and sylvatic cycles of these parasites.

ABSTRACT

As training in helminthology has declined in the medical microbiology curriculum, many rare species of zoonotic cestodes have fallen into obscurity. Even among specialist practitioners, knowledge of human intestinal cestode infections is often limited to three genera, Taenia, Hymenolepis and Dibothriocephalus. However, five genera of uncommonly encountered zoonotic Cyclophyllidea (Bertiella, Dipylidium, Raillietina, Inermicapsifer and Mesocestoides) may also cause patent intestinal infections in humans worldwide. Due to the limited availability of summarized and taxonomically accurate data, such cases may present a diagnostic dilemma to clinicians and laboratories alike. In this review, historical literature on these cestodes is synthesized and knowledge gaps are highlighted. Clinically relevant taxonomy, nomenclature, life cycles, morphology of human-infecting species are discussed and clarified, along with the clinical presentation, diagnostic features and molecular advances, where available. Due to the limited awareness of these agents and identifying features, it is difficult to assess the true incidence of these 'forgotten' cestodiases as clinical misidentifications are likely to occur. Also, the taxonomic status of many of the human-infecting species of these tapeworms is unclear, hampering accurate species identification. Further studies combining molecular data and morphological observations are necessary to resolve these long-standing taxonomic issues and to elucidate other unknown aspects of transmission and ecology.

ABSTRACT

This study provides insights about the diversity, prevalence and distribution of alpine wild galliformes gastrointestinal parasite community, trying to fill a gap in the scientific information currently available in scientific literature. The analysis included three host species: 77 rock partridge (Alectoris graeca saxatilis), 83 black grouse (Tetrao tetrix tetrix) and 26 rock ptarmigan (Lagopus muta helveticus) shot during the hunting seasons 2008-2015. Parasites isolated were Ascaridia compar, Capillaria caudinflata and cestodes. The rock ptarmigan was free from gastrointestinal parasites, whereas the most prevalent helminth (37%) was A. compar in both black grouse and rock partridge. C. caudinflata occurrence was significantly higher in black grouse (prevalence = 10%, mean abundance = 0.6 parasites/sampled animal) than in rock partridge (prevalence = 1.20%, mean abundance = 0.01 parasites/sampled animal). Significant differences were detected among hunting districts. A. compar was found with a significant higher degree of infestation in the hunting districts in the northern part of the study area whereas cestodes abundance was higher in Lanzo Valley. Quantitative analysis of risk factors was carried out using a generalized linear model (GLM) only on the most common parasite (A. compar). Latitude was the only factors associated with infestation risk (OR = 52.4). This study provides information on the composition and variability of the parasite community in the alpine Galliformes species.
