Purpose: Dysregulation of glutamatergic neurotransmission (GN) is linked to sympathetic-respiratory overactivity and hypertension. We investigated whether maternal protein restriction is able to alter GN into the nucleus of the solitary tract (NTS) in adult offspring.Methods: Wistar rat dams were fed with control (NP; 17% protein) or low-protein (LP; 8% protein) diet during pregnancy and lactation, and their offspring were evaluated at 70-90d old. Direct measurements of mean arterial pressure (MAP), heart rate (HR), respiratory frequency (RF) and respiratory (RV) and cardiac (CV) variabilities were assessed in consciousness. The evaluation of GN into NTS over cardiovascular system were assessed by microinjections of unilateral glutamate (L-glu 0.5 nmol/100nL) and bilateral kynurenic acid (Kyn 2.5 nmol/50nL). The NP and LP groups were compared using unpaired Student's t-test where p < 0.05 was considered significant.Results: The LP exhibited higher MAP at rest (p = 0.03) and after L-glu microinjection (p = 0.04), as well as an increase over HR after Kyn microinjection when compared to the NP (p = 0.049). In the RV, the LP group showed an increase of the component-standard deviation 1 (p = 0.037) at rest. In the CV, the LP presented an increase of the low frequency (LF) component of the pulse interval (PI) (p = 0.034), a decrease of high frequency (HF) of the PI (p = 0.034), beyond an increased LF/HF ratio of the PI (p = 0.027) when compared to the NP. The kynurenic acid microinjection did not produce changes in RV or CV (p > 0.05).Conclusions: Altered GN into the NTS may contribute to augmented blood pressure in protein-restricted offspring.

BACKGROUND AND AIMS: Maternal undernutrition induces development of the arterial hypertension. We investigated the effects of a maternal low-protein diet on cardiovascular autonomic control in the offspring. METHODS AND RESULTS: Male Wistar rats were divided into two groups according to the diets of their mothers during gestation and lactation: the control (normal protein, NP, 17% casein; n = 14) and low-protein (LP, 8% casein; n = 14) groups. Direct measurements of arterial pressure (AP) were recorded from wakeful 90-day-old male offspring. The LP offspring presented higher mean AP than did the NP rats (NP: 93 ± 4 vs. LP: 113 ± 2 mmHg; p < 0.05), whereas the heart rate (HR) was similar in the two groups. In the spectral analysis, the LP group showed higher power at low (NP: 1.98 ± 0.25 vs. LP: 3.7 ± 0.3 mmHg²; p < 0.05) and high (NP: 1.28 ± 0.18 vs. LP: 2.13 ± 0.42 mmHg²; p < 0.05) frequencies of systolic arterial pressure (SAP). In the pulse interval, the LP group presented an increase in the LF/HF ratio (NP: 0.32 vs. LP: 0.56; p < 0.05). After propranolol (4 mg/kg, intravenous (iv)), the bradycardia was higher in the LP group (NP: -36 ± 8 vs. LP: -94 ± 12 bpm; p < 0.05), after methylatropine (2 mg/kg, iv), the tachycardia was similar to NP group. After administration of the ganglionic blocker (hexamethonium; 25 mg/kg, iv), the LP animals showed larger delta variation in the AP (NP: -33.7 ± 5 vs. LP: -53.6 ± 4 mmHg; p < 0.05). CONCLUSION: The rats subjected to protein malnutrition presented an increase in the cardiovascular sympathetic tone, which contributed to the elevated AP observed in these animals.

AIM: To assess and compare the flexibility and torsional resistance of PathFile, RaCe ISO 10 and Scout RaCe instruments in relation to stainless steel K-File hand instruments. METHODOLOGY: Rotary PathFile (sizes 13, 16 and 19; .02 taper), Race ISO 10 (size 10; 0.02, 0.04 and 0.06 tapers), Scout RaCe (sizes 10, 15 and 20; 0.02 taper) and hand K-File (sizes 10, 15 and 20; 0.02 taper) instruments were evaluated. Alloy chemical composition, phases present and transformation temperatures were determined for the NiTi instruments. For all instruments, diameters at each millimetre from the tip as well as cross-sectional areas at 3 mm from the tip were measured based on ANSI/ADA Specification No. 101 using image analysis software. Resistance to bending and torsional resistance were determined according to specification ISO 3630-1. Vickers microhardness measurements were also taken in all instruments to assess their strength. Data were analysed using analysis of variance (α = 0.05). RESULTS: The alloys used in the manufacture of the three types of NiTi instruments had approximately the same chemical composition, but the PathFile instruments had a higher Af transformation temperature and contained a small amount of B19' martensite. All instruments had diameter values within the standard tolerance. The bending and torsional resistance values were significantly increased relative to the instrument diameter and cross-sectional area. CONCLUSIONS: PathFile instruments were the most flexible and the least torque resistant, whilst the stainless steel instruments were the least flexible although they were more torque resistant than the NiTi instruments.

Alternariaster was erected in 2007 to accommodate Alternaria helianthi, a fungal species known to cause leaf spots on Helianthus annuus (sunflower). It was segregated from Alternaria based on conidial morphology. Recently an unknown alternaria-like dematiaceous fungus was found associated with leaf spots on Bidens sulphurea (yellow cosmos) in Brazil. Based on a multi-gene phylogeny of parts of the ITS and LSU genes, this fungus was placed within the Leptosphaeriaceae with Alternariaster helianthi as its closest neighbour. Additional genes sequenced, RPB2 and GAPDH, confirmed this close relationship. The fungus on B. sulphurea has smaller conidia, 50-97.5 × 12.5-20 µm, compared to Al. helianthi, 80-160 × 18-30 µm, and lacks oblique or transverse septa which can be present in Al. helianthi. Pathogenicity studies on 18 plant species belonging to the Compositae showed that the B. sulphurea fungus only infected B. sulphurea, whereas Al. helianthi infected H. annuus and Galinsoga quadriradiata, a yet unreported host of Al. helianthi. The fungus causing disease on B. sulphurea is hence closely related but phylogenetically, morphologically and pathologically distinct from Al. helianthi, and therefore newly described as Alternariaster bidentis. The collection of a second species in the genus Alternariaster and the multigene phylogenetic analysis of these two species, confirmed Alternariaster to be a well-delimited genus in the Leptosphaeriaceae rather than the Pleosporaceae, to which Alternaria belongs.

RESUMEN

Cellular materials have a wide range of applications, including structural optimization and biomedical applications. Due to their porous topology, which promotes cell adhesion and proliferation, cellular materials are particularly suited for tissue engineering and the development of new structural solutions for biomechanical applications. Furthermore, cellular materials can be effective in adjusting mechanical properties, which is especially important in the design of implants where low stiffness and high strength are required to avoid stress shielding and promote bone growth. The mechanical response of such scaffolds can be improved further by employing functional gradients of the scaffold's porosity and other approaches, including traditional structural optimization frameworks; modified algorithms; bio-inspired phenomena; and artificial intelligence via machine learning (or deep learning). Multiscale tools are also useful in the topological design of said materials. This paper provides a state-of-the-art review of the aforementioned techniques, aiming to identify current and future trends in orthopedic biomechanics research, specifically implant and scaffold design.

The aim of our study was to validate the use of the standardized Radiological Society of North America (RSNA) reporting system in individuals with known lung cancer who presented to the emergency department with suspected COVID-19. We included patients aged 18 years or older from the Cancer Institute of the State of São Paulo (ICESP) with a confirmed diagnosis of lung cancer, admitted to the emergency department and undergoing chest computed tomography (CT) for suspicion of COVID-19. Comparison between SARS-CoV2 RT-PCR across RSNA categories was performed in all patients and further stratified by diagnosis of lung cancer progression. Among 58 individuals included in the analysis (65±9 years, 43% men), 20 had positive RT-PCR. Less than a half (43%) had no new lung findings in the CT. Positive RT-PCR was present in 75% of those with typical findings according to RSNA and in only 9% when these findings were classified as atypical or negative (P<0.001). Diagnostic accuracy was even higher when stratified by the presence or absence of progressive disease (PD). Extent of pulmonary inflammatory changes was strongly associated with higher mortality, reaching a lethality of 83% in patients with >25% of lung involvement and 100% when there was >50% of lung involvement. The lung involvement score was also highly predictive of prognosis in this population as was reported for non-lung cancer individuals. Collectively, our results demonstrated that diagnostic and prognostic values of chest CT findings in COVID-19 are robust to the presence of lung abnormalities related to lung cancer.

Although the gold standard for HRV registration is the electrocardiogram (ECG), several applications (APPs) have been increasingly developed. The evaluation carried out by these devices must be compatible with ECG standards. The aim of this study was to compare the data obtained simultaneously with ECG and APP with chest heart rate transmitters. Fifty-six healthy individuals (28 men and 28 women) were evaluated at rest through a short simultaneous HRV measurement with both devices. Data from both acquisition systems were analyzed separately using their own analysis software and exported and analyzed using a validated software. Signal recordings were compatible between the two acquisition systems (Pearson r=0.99; P<0.0001). Although a high correlation was found for the HRV variables obtained in the time domain (Spearman r=0.99; P<0.0001), the correlation decreased in the frequency domain (Pearson r=0.85; P<0.0001) when two software programs were used. Comparison of the averages of spectral analysis parameters also showed differences when HRV data were analyzed separately in each device for low-frequency (LF) and high-frequency (HF) bands. Although the portability of these mobile devices allows for optimal HRV evaluation, the direct analysis obtained from these devices must be carefully evaluated with respect to frequency domain parameters.

RESUMEN

This work deals with the finite element (FE) implementation of a biphasic poroelastic formulation specifically developed to address the intricate behaviour of the Intervertebral Disc (IVD) and other highly hydrated soft tissues. This formulation is implemented in custom FE solver V-Biomech, being the validation performed with a lumbar IVD model, which was compared against the analogous FE model of Williams et al. and the experiments of Tyrrell et al. Good agreement with these benchmarks was achieved, meaning that V-Biomech and its novel poroelastic formulation are a viable alternative for simulation of biphasic soft tissues.

Ixora coccinea, known as jungle geranium, flame of the woods, or flame of the forest, is a shrub in the Rubiaceae native to Eastern India and is widely cultivated for its showy red globose inflorescences. It was introduced in Brazil in the beginning of the 19th century where it became a popular ornamental (2). Recently, leaf spots were observed on I. coccinea foliage in Brazil. Severely diseased plants were unsightly with loss of foliage and remaining foliage was heavily spotted and yellowed. Sparse colonies of a fungus were consistently observed to be associated with diseased tissues. Slides were prepared by scraping colonized surfaces with a scalpel and mounting the fungal structures in lactophenol. Observations, measurements, and photographs were prepared with a light microscope (Olympus BX 50). The fungus was isolated in pure culture in plates containing V8 juice agar and incubated at 25°C. Pathogenicity was verified by placing pure culture plugs, obtained from actively growing colonies, onto healthy leaves of I. coccinea still attached to living plants left in a dew chamber. After 48 h, plants were removed and transferred to a greenhouse. Controls consisted of leaves of two separate healthy plants on which sterile culture medium plugs were deposited. Typical leaf spot symptoms appeared 11 days after inoculation and sporulation was observed after 30 days, but was not observed on the controls. Lesions on leaves were subcircular to polygonal or irregular, reddish brown to dark brown, 3.5 to 12 mm in diameter, coalescing, and leading to premature abscission. Fungus morphology was as follows: stromata absent; sporulation nearly indistinct under the dissecting microscope; conidiophores hypophyllous, either emerging from the stomata in small fascicles or isolate and formed from external mycelium, obclavate to cylindrical, 15 to 60 × 3.0 to 6.0 µm, 3 to 7 septate, obliquely constricted at septae, simple or branched, thin walled, brown, smooth; conidiogenous loci inconspicuous, not darkened, unthickened; conidia obclavate to cylindrical, straight to slightly curved, 17.5 to 89.0 × 3.0 to 5.0 µm, apex obtuse, base obconic-truncate, 1 to 7 obliquely septate, subhyaline, thin walled, eguttulate, smooth. Except for minor differences on conidiophore and conidial length, morphology was as described for Pseudocercospora ixoricola (4). Representative samples collected in the states of Minas Gerais (VIC 31216 - May 2009) and Rio de Janeiro (VIC 31217 - July 2009) were deposited in the local herbarium. Additional ad hoc observations indicate a widespread distribution of this disease in southeastern Brazil. Until now the sole cercosporoid fungus recorded on I. coccinea was P. ixorae from Asia and North America. There is also a record of this fungal species from an Ixora sp. from Brazil (1). Features such as the absence of stromata and the shape and size of conidiophores and conidia, separates it readily from P. ixoricola and P. ixoriana (a third species of Pseudocercospora known to be associated with plants in the genus Ixora). To our knowledge, this is the first record of P. ixoricola in South America and the first time it is recorded as a pathogen of I. coccinea. PURPOSE: As the promoter of the stent expansion, the balloon plays a very important role, offering a strong influence on the deployment process. Balloon-artery interaction is pointed as a probable cause of restenosis, stressing the relevance of balloon modeling when simulating the stenting procedure. In this work, an in-silico study of the balloon modeling strategies is performed. METHODS: Ultrasonic-microcasting is a novel technology that allows obtaining stents manufactured in magnesium alloys, being suggested as a promising solution. However, this technique demands superior stent strut thickness, which may have an impact on the stent deployment procedure. The influence of the balloon modeling is studied through the simulation of different balloon geometries (open- or taper-ended) and material constitutive model (linear elastic or hyperelastic) on the expanded configuration of a stent manufactured through ultrasonic-microcasting. RESULTS: The results obtained suggest that the choice of balloon type has small impact in terms of demanded pressure to inflate the balloon and in the stent final radius achieved at fully-expanded configuration. Additionally, it was proved that the balloon-type influences the stent expanded profile along its length and diameter as a result of the different deformation behavior exhibited by the balloon. CONCLUSION: The hyperelastic taper-ended balloon suggests being the model that better correlates with both experimental and clinical results regarding the expanded balloon profile during the procedure.

Tooth loss is a problem that affects both old and young people. It may be caused by several conditions, such as poor oral hygiene, lifestyle choices or even diseases like periodontal disease, tooth grinding or diabetes. Nowadays, replacing a missing tooth by an implant is a very common process. However, many limitations regarding the actual strategies can be enumerated. Conventional screwed implants tend to induce high levels of stress in the peri-implant bone area, leading to bone loss, bacterial bio-film formation, and subsequent implant failure. In this sense, root-analogue dental implants are becoming promising solutions for immediate implantation due to their minimally invasive nature, improved bone stress distribution and because they do not require bone drilling, sinus lift, bone augmentation nor other traumatic procedures. The aim of this study was to analyse and compare, by means of FEA, the stress fields of peri-implant bone around root-analogue and screwed conventional zirconia implants. For that purpose, one root-analogue implant, one root-analogue implant with flaps, two conventional implants (with different threads) and a replica of a natural tooth were modelled. COMSOL was used to perform the analysis and implants were subjected to two simultaneous loads: 100 N axially and 100 N oblique (45°). RESULTS: revealed that root-analogue implants, namely with flaps, should be considered as promising alternatives for dental implant solutions since they promote a better stress distribution in the cortical bone when compared with conventional implants.

The present study aims at developing a computational framework with experimental validation to determine the mechanical properties of zirconia foams for bone tissue engineering. A micro-CT based finite element model that allows characterizing the mechanical property of such cellular structures is developed. Micro-CT images are filtered to vanish noises and smooth boundaries before constructing 3D zirconia foams using an adaptive Body-Centered Cubic background lattice. In addition to micro-CT images, the local material property at the scaffold struts is measured using a micro-indentation test, which shows a considerable difference with that of common zirconia owing to the manufacturing process. The computational model also takes the plastic deformation of material into account employing the Voce law, a nonlinear isotropic hardening law, as well as Von-mises yield criterion. Zirconia foams with different pore sizes are manufactured using the replica method and their mechanical properties determined experimentally. Such experimental outcomes are to validate and demonstrate the capability of the developed model, which can be used for pre-operational evaluations and preclinical tests of zirconia scaffolds. The stress magnitude and distribution within the scaffold as well as plastic strains and flow stress of the zirconia scaffold are computed and analysed. Using the proposed approach, a deep insight into the association of macroscopic behaviour of the scaffold to microscopic features, e.g. strut waviness, Plateau border, thickness variation of cells, irregularity, microstructural variability, imperfections and strut's material property associated with to the manufacturing procedure, can be gained.

RESUMEN

Pulmonary arterial hypertension (PAH), characterized by localized increased arterial blood pressure in the lungs, is a slow developing long-term disease that can be fatal. PAH is characterized by inflammation, vascular tone imbalance, pathological pulmonary vascular remodeling, and right-sided heart failure. Current treatments for PAH are palliative and development of new therapies is necessary. Recent and relevant studies have demonstrated that epigenetic processes may exert key influences on the pathogenesis of PAH and may be promising therapeutic targets in the prevention and/or cure of this condition. The aim of the present mini-review is to summarize the occurrence of epigenetic-based mechanisms in the context of PAH physiopathology, focusing on the roles of DNA methylation, histone post-translational modifications and non-coding RNAs. We also discuss the potential of epigenetic-based therapies for PAH.

The aim of this study was to investigate the reproductive, biochemical, and hematological outcomes of pregnant rats exposed to protein restriction. Wistar rat dams were fed a control normal-protein (NP, 17% protein, n=8) or a low-protein (LP, 8% protein, n=14) diet from the 1st to the 20th day of pregnancy. On the 20th day, the clinical signs of toxicity were evaluated. The pregnant rats were then anesthetized and blood samples were collected for biochemical-hematological analyses, and laparotomy was performed to evaluate reproductive parameters. No sign of toxicity, or differences (P>0.05) in body weight gain and biochemical parameters (urea, creatinine, albumin, globulin, and total protein) between NP and LP pregnant dams were observed. Similarly, hematological data, including red blood cell count, white blood cell count, hemoglobin, hematocrit, red blood cell distribution width (coefficient of variation), mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, % lymphocytes, absolute lymphocyte count, platelet count, and mean platelet volume were similar (P>0.05) at the end of pregnancy. Reproductive parameters (the dam-offspring relationship, ovary mass, placenta mass, number of corpora lutea, implantation index, resorption index, and the pre- and post-implantation loss rates) were also not different (P>0.05) between NP and LP pregnant dams. The present data showed that a protein-restricted diet during pregnancy did not alter reproductive, biochemical, and hematological parameters and seems not to have any toxic effect on pregnant Wistar rats.

PURPOSE: The purpose of the present study was to evaluate the distribution of stresses and consequent bone volume affected surrounding external hexagon or Morse taper dental implant systems by finite element analysis. MATERIAL AND METHODS: Two different dental implant-abutment designs were assessed: external hexagon or Morse taper joints. A mandibular bone model obtained from a computed tomography scan was used. The implant-abutment systems were axially or obliquely (45°) loaded on 150 N relatively to the central axis of the implant. The von Mises stresses were analysed in terms of magnitude and volume of affected surrounding bone. RESULTS: The von Mises equivalent values found on the cortical bone were higher than that recorded on the trabecular bone. Additionally, the bone volume associated with high stress values was higher in cortical and trabecular bone for oblique loading compared to axial loading. The values of von Mises equivalent stress around Morse taper implant-abutment system were lower on both axial and oblique loads than those recorded for external hexagon implant-abutment systems. CONCLUSIONS: Morse taper implant joints revealed a proper biomechanical behavior when compared to external hexagon systems concerning a significant volume of surrounding peri-implant bone subjected to lower stresses values.

The present study investigated the impact of a western diet during gestation and lactation on the anthropometry, serum biochemical, blood pressure and cardiovascular autonomic control on the offspring. Male Wistar rats were divided into two groups according to their mother's diet received: control group (C: 18% calories of lipids) and westernized group (W: 32% calories of lipids). After weaning both groups received standard diet. On the 60th day of life, blood samples were collected for the analysis of fasting glucose and lipidogram. Cardiovascular parameters were measured on the same period. Autonomic nervous system modulation was evaluated by spectrum analysis of heart rate (HR) and systolic arterial pressure (SAP). The W increased glycemia (123±2 v. 155±2 mg/dl), low-density lipoprotein (15±1 v. 31±2 mg/dl), triglycerides (49±1 v. 85±2 mg/dl), total cholesterol (75±2 v. 86±2 mg/dl), and decreased high-density lipoprotein (50±4 v. 38±3 mg/dl), as well as increased body mass (209±4 v. 229±6 g) than C. Furthermore, the W showed higher SAP (130±4 v. 157±2 mmHg), HR (357±10 v. 428±14 bpm), sympathetic modulation to vessels (2.3±0.56 v. 6±0.84 mmHg2) and LF/HF ratio (0.15±0.01 v. 0.7±0.2) than C. These findings suggest that a western diet during pregnancy and lactation leads to overweight associated with autonomic misbalance and hypertension in adulthood.
