Biological nitrogen removal process could be affected due to the presence of heavy metals owing to their toxicity and accumulation in the sludge. In this study, the impact of Cu2+ shock on a long-term nitritation operation was investigated in an air-lift reactor with self-recirculation. Both the dynamics of microbial community and inhibition kinetics under Cu2+ stress were ascertained. The results showed that Cu2+ exerted severe inhibition on nitritation performance of an air-lift reactor (ALR) at 25 mg/L. The corresponding NH4+-N removal efficiency decreased to below 50%, which was mainly due to the variation of microbial community structure, especially the inhibition of nitrifiers like Nitrosomonas (the relative abundance decreased from 30% to 1% after Cu2+ inhibition). Kinetic parameters were obtained and compared after fitting the Haldane model. The long-term Cu2+ stress on the ALR aggravated the ammonium affinity and the resistance to substrate self-inhibition of the nitritation sludge, but reduced the resistance to Cu2+ inhibition. Furthermore, Cu2+ acted as uncompetitive inhibitor on nitritation process. Our results provide new insights into the nitritation characteristics under long-term Cu2+ stress.

OBJECTIVE: Pleuropulmonary blastoma (PPB) is a rare malignant tumor in childhood that is highly invasive and has poor prognosis. We retrospectively analyzed patients with PPB who died within 30 days in hopes of providing a basis for improving diagnosis and treatment. METHODS: We retrospectively reviewed six children with PPB who died within 30 days of admission at our hospital from January 2004 to March 2018, including their clinical features, pathological diagnosis, course of treatment, and major causes of death. RESULTS: Six patients (two female, four male; median age, 38 months) were included. All patients presented with respiratory symptoms. Chest imaging showed that all tumors had diameters greater than 10 cm, with varying degrees of serous effusion. Four patients underwent ultrasound-guided fine-needle aspiration (FNA), one patient underwent exploratory thoracotomy, and one underwent partial tumor resection. Five cases were type III, and another was type II. Four patients developed adverse events while waiting for pathological results after FNA, and four patients were treated with chemotherapy but their tumors failed to decrease in size one to two weeks later. The median hospitalization to death time was 17 days (range, 5-24 days). CONCLUSIONS: PPB often presents with respiratory symptoms that rapidly develop into respiratory distress. The rapid tumor enlargement contributes to the disease's progression. Chemoreduction in such tumors is not obviously effective, and the mortality rate is high. The main causes of death were respiratory failure and sepsis. Further clinical studies will be required to evaluate the role of initial biopsy compared with upfront total excision.

BACKGROUND/AIMS: Salvia miltiorrhiza (SM) contains four major aqueous active ingredients, which have been isolated, purified and identified as danshensu (DSS), salvianolic acid A (Sal-A), salvianolic acid B (Sal-B) and protocatechuic aldehyde (PAL), totally abbreviated as SABP. Although SM is often used to treat various cardiovascular diseases in traditional Chinese medicine, the efficacy and function of optimal compatibility ratio of SM's active ingredients (SABP) in the prevention and treatment of cardiovascular diseases remain uncertain. This study investigated antihypertensive effect and underlying mechanisms of SABP vs. SM lyophilized powder (SMLP) in spontaneously hypertensive rats (SHR) and to establish the ratio of the optimal compatibility of DSS, Sal-A, Sal-B and PAL in improving cardiovascular functions. METHODS: The SHRs were treated with either SABP or SMLP and their systolic blood pressures (SBP) were monitored. The isolated thoracic aorta of SHRs was segregated for immunohistochemistry, Hematoxylin-Eosin stain and mRNA and protein expression of NOX4, TGF-ß1, Col-I, ET-1, α-SMA and Smad7. Moreover, the adventitial fibroblasts (AFs) were isolated and cultured from SD rats' aorta and the reactive oxygen species (ROS) production was determined after SABP or SMLP treatment. RESULTS: SABP, but not SMLP, significantly reduced SBP, which were accompanied by the inhibited morphological changes in the thoracic aorta and the reduced mRNA and protein expression of NOX4, TGF-ß1, Col-I, ET-1 and α-SMA, but the increased Smad 7 expression in SHRs. Moreover, SABP also resulted in a decreased ROS production in AFs of SD rats. CONCLUSIONS: These results indicate that SABP, but not SMLP, treatment potently inhibits hypertension through improvements of vascular remodeling and oxidative stress. The present study provides new evidence that the efficacy and function from optimal compatibility ratio of SM active ingredients is much better than its lyophilized powder, which represents a strategy to develop SM's new beneficial effect in improving cardiovascular functions.

Increasing atmospheric nitrogen (N) deposition has the potential to alter plant diversity and thus the function and stability of terrestrial ecosystems. N-limited alpine ecosystems are expected to be particularly susceptible to increasing N deposition. However, little is known about the critical loads and saturation thresholds of ecosystem responses to increasing N deposition on the Tibetan Plateau, despite its importance to ecosystem management. To evaluate the N critical loads and N saturation thresholds in an alpine ecosystem, in 2010, we treated an alpine meadow with five levels of N addition (0, 10, 20, 40, and 80 kg N ha(-1) year(-1)) and characterized plant and soil responses. The results showed that plant species richness and diversity index did not statistically vary with N addition treatments, but they both changed with years. N addition affected plant cover and aboveground productivity, especially for grasses, and soil chemical features. The N critical loads and saturation thresholds, in terms of plant cover and biomass change at the community level, were 8.8-12.7 and 50 kg N ha(-1) year(-1) (including the ambient N deposition rate), respectively. However, pronounced changes in soil inorganic N and net N mineralization occurred under the 20 and 40 kg N ha(-1) year(-1) treatments. Our results indicate that plant community cover and biomass are more sensitive than soil to increasing N inputs. The plant community composition in alpine ecosystems on the Qinghai-Tibetan Plateau may change under increasing N deposition in the future.

Microglia-mediated inflammatory responses have been shown to play a crucial role in Parkinson's disease. In addition, exosomes derived from mesenchymal stem cells have shown anti-inflammatory effects in the treatment of a variety of diseases. However, whether they can protect neurons in Parkinson's disease by inhibiting microglia-mediated inflammatory responses is not yet known. In this study, exosomes were isolated from human umbilical cord mesenchymal stem cells and injected into a 6-hydroxydopamine-induced rat model of Parkinson's disease. We found that the exosomes injected through the tail vein and lateral ventricle were absorbed by dopaminergic neurons and microglia on the affected side of the brain, where they repaired nigral-striatal dopamine system damage and inhibited microglial activation. Furthermore, in an in vitro cell model, pretreating lipopolysaccharide-stimulated BV2 cells with exosomes reduced interleukin-1ß and interleukin-18 secretion, prevented the adoption of pyroptosis-associated morphology by BV2 cells, and increased the survival rate of SH-SY5Y cells. Potential targets for treatment with human umbilical cord mesenchymal stem cells and exosomes were further identified by high-throughput microRNA sequencing and protein spectrum sequencing. Our findings suggest that human umbilical cord mesenchymal stem cells and exosomes are a potential treatment for Parkinson's disease, and that their neuroprotective effects may be mediated by inhibition of excessive microglial proliferation.

The study is to investigate the effect of angiotensin II (Ang II) and its receptor blockers on migration and endothelin-1 (ET-1) expression of rat vascular adventitial fibroblast subpopulations. Vascular adventitial fibroblasts were individually expanded by using cloning rings, and the effects of Ang II on the migration of adventitial fibroblast subpopulations were evaluated by Transwell. Fluorescence quantitative-PCR detected the expression of preproET-1 mRNA induced by Ang II, and its receptor antagonists losartan and PD-123319. The concentration of ET-1 was determined by ELISA. It showed that spindle shaped and epithelioid shaped cells were isolated by using cloning rings, named as spindle cells and round cells. RT-PCR showed that fibroblast subpopulations did not have leukocytes, endothelial cells and smooth muscle cells, namely pure cell lines. Compared with respective control cells, two subpopulations had transferring ability. Ang II significantly improved round cells migration in a concentration-dependent manner, and had no obvious influence on spindle cells migration. Ang II (1 x 10(-8) - 1 x 10(-6) mol x L(-1)) significantly increased the expression of preproET-1 mRNA in round cells (P < 0.01), and had no significant effect on the expression of preproET-1 mRNA in spindle cells. Losartan blocked the expression of preproET-1 mRNA induced by Ang II in round cells, and had no significant effect on the expression of preproET-1 mRNA in spindle cells. The effects of Ang II and ET-1 receptor inhibitors on the release of ET-1 were similar to the expression of preproET-1 mRNA. The results indicate that there are two cell subpopulations: round cells and spindle cells in rat vascular adventitial fibroblasts. Ang II significantly improved cells migration, and increased the expression of ET-1 in round cell subpopulation. It suggested that there may be different migratory mechanisms in two cell subpopulations, and the two subpopulations may play a different role in vascular remodeling and reparative process.

Recovering nitrogen and phosphorus from waste water in the form of struvite is an effective way to recycle resources. The insufficient purity of the resulting struvite and the large loss of nitrogen and phosphorus are the challenges at present. Therefore, it is urgent to develop innovative method in struvite crystallization process for efficient nitrogen and phosphorus recovery. This study proposed a crystallization method to reduce the loss of nitrogen and phosphorus by a struvite fluidized bed reactor (FBR) with optimized structure and operation conditions. The properties of struvite obtained under various conditions in the reactor were studied, and the internal operating conditions of the reactor were simulated with COMSOL Multiphysics to verify the effectiveness of the reactor optimization. This reactor achieved stable operation under the conditions of N/P = 1:1 and pH = 9.0. The purity of struvite obtained reached 98.5%, the conversion rate of ammonia nitrogen reached 97.2%, and struvite crystals could grow to 84 µm within 24 h. The simulation results showed that the Venturi tubes installed at multiple locations increased the turbulent energy to 4 × 10-4 m2/s2, which greatly improved the mass transfer efficiency. The trajectory of the crystal particles was consistent with the fluid flow field, which promoted the purification and growth of the crystal. In general, the new FBR with enhanced external recirculation would be a very feasible way to improve crystal growth and crystal purification of struvite, and it could enhance the recovery efficiency of nitrogen and phosphorus with reduced cost.

Parkinson's disease (PD) is a progressively debilitating neurodegenerative condition that leads to motor and cognitive dysfunction. At present, clinical treatment can only improve symptoms, but cannot effectively protect dopaminergic neurons. Several reports have demonstrated that human umbilical cord mesenchymal stem cells (hucMSCs) afford neuroprotection, while their application is limited because of their uncontrollable differentiation and other reasons. Stem cells communicate with cells through secreted exosomes (Exos), the present study aimed to explore whether Exos secreted by hucMSCs could function instead of hucMSCs. hucMSCs were successfully isolated and characterized, and shown to contribute to 6-hydroxydopamine (6-OHDA)-stimulated SH-SY5Y cell proliferation; hucMSC-derived Exos were also involved in this process. The Exos were purified and identified, and then labeled with PKH 26, it was found that the Exos could be efficiently taken up by SH-SY5Y cells after 12 h of incubation. Pretreatment with Exos promoted 6-OHDA-stimulated SH-SY5Y cells to proliferate and inhibited apoptosis by inducing autophagy. Furthermore, Exos reached the substantia nigra through the blood-brain barrier (BBB) in vivo, relieved apomorphine-induced asymmetric rotation, reduced substantia nigra dopaminergic neuron loss and apoptosis, and upregulated the level of dopamine in the striatum. These results demonstrate that hucMSCs-Exos have a treatment capability for PD and can traverse the BBB, indicating their potential for the effective treatment of PD.

The toxicity of hexavalent chromium (Cr(VI)) is one of the challenges in implementing Anammox process to ammonium-rich wastewater treatment. However, the response of Anammox process to Cr(VI) stress and the inhibition mechanism remain unclear. Here, two Anammox UASB reactors were operated for 285 days under different Cr(VI) stresses. The results showed Anammox performance was not affected at low Cr(VI) concentration (i.e., 0-0.5 mg L-1), but was severely inhibited at 0.8 mg L-1. Attempts to domesticate Anammox process to higher Cr(VI) by lowering nitrogen loading rate were failed. Examination of Cr(VI) fate showed the occurrence of extracellular and intracellular Cr(VI) reduction to Cr(III). The inhibition was ascribed to the significant intracellular Cr(VI) reduction, accounting for 99.78% of the total Cr(VI) reduction. Moreover, under long-term Cr(VI) exposure, most nitrite was oxidized to nitrate. But microbial community showed no enrichment of Cr(VI) reducing bacteria and other nitrogen transformation-related bacteria.

High adsorption capacity, good biocompatibility and low cost are highly demanded for biofilter used in ammonium-rich wastewater treatment. In this study, we used SEM, BET, XRD and 16S rRNA to document the evidence for good performance in adsorption and biodegradation in aged refuse. Parallel experiment between raw and inert refuse showed ammonium adsorption occurred at the initial week, with the highest ammonium removal efficiency of 90.36%, but saturated during the subsequent long-term operation. Meanwhile, over 6months' operation of an aged refuse biofilter was conducted to confirm that nitrification was the main pathway of ammonium conversion. The maximum nitrogen loading rate could reach up to as high as 1.28kg/m3/d, with ammonium removal efficiency at 99%. Further, high nitrifier biodiversity were detected with 'Nitrosomonas' and 'Nitrospira' in domination in the refuse. However, Nitrospira would outcompete Nitrosomonas under the oxygen limiting condition and resulted in the failure of partial nitrification. The physicochemical and biological analysis show that biodegradation is the main ammonium conversion pathway, which is the critical finding of this work. This investigation would help to accelerate the application of the aged refuse process in ammonium-rich wastewater treatment.

Increasing evidence indicates that disruption of normal iron homeostasis may contribute to pathological development of Alzheimer's disease. Icariin, astragalus, and puerarin have been shown to suppress iron overload in the cerebral cortex and improve spatial learning and memory disorders in Alzheimer's disease mice, although the underlying mechanism remains unclear. In the present study, APPswe/PS1ΔE9 transgenic mice were administered icariin, astragalus, and puerarin (120, 80, and 80 mg/kg, respectively, once a day, for 3 months). Iron levels were detected by flame atomic absorption spectroscopy. Interleukin-1ß, interleukin-6, and tumor necrosis factor-α levels were measured in the cerebral cortex by enzyme linked immunosorbent assay. Glutathione peroxidase and superoxide dismutase activity and malondialdehyde content were determined by colorimetry. Our results demonstrate that after treatment, iron levels and malondialdehyde content are decreased, while glutathione peroxidase and superoxide dismutase activities are increased. Further, interleukin-1ß, interleukin-6, and tumor necrosis factor-α levels were reduced. These results confirm that compounds of icariin, astragalus, and puerarin may alleviate iron overload by reducing oxidative stress and the inflammatory response.

In recent decades, global warming has become an indisputable fact on the Tibetan Plateau. Alpine ecosystems are very sensitive to global warming, and the impact may depend on the degree of atmospheric nitrogen (N) deposition. The previous studies have paid more attention to year-round warming, but the effect of winter warming has been unstudied. In this study, a manipulative experiment was conducted, consisting of warming and N addition. It was carried out since 2010 in an alpine meadow, and three types of warming treatments were set up: no warming (NW), year-round (YW), and winter warming (WW). Warming significantly increased air and soil temperature, but decreased soil moisture. Under no N addition, YW showed significantly decreased ecosystem respiration (Reco) in 2012, and WW decreased Reco in 2014. Under N addition, neither YW nor WW had significant effects on Reco, indicating that N addition compensated the negative effect of warming on Reco. Annually, YW and WW decreased ecosystem carbon (C) emissions, and the extent of the reduction was even larger under WW. Under no N addition, both YW and WW significantly decreased aboveground biomass. Moreover, especially under no N, YW and WW significantly decreased soil inorganic N. WW also had negative effects on soil microbial biomass C. Structure equation modeling showed that soil moisture was the most important factors controlling Reco, and soil inorganic N content and microbial biomass C could explain 46.6% and 16.8% of the variation of Reco. The findings indicate that soil property changes under warming had substantial effects on ecosystem C efflux. The inhibitory effects of winter warming on ecosystem C efflux were mainly attributed to the decline of soil N and microbial biomass. Thus, the effects of winter warming on ecosystem C emissions in this semiarid alpine meadow are not as serious as expected and largely depend on N deposition.

A long-term experiment, involving exogenous N addition and simulated warming, was conducted in an alpine meadow in Damxung, northern Tibet, to study how warming and N addition influence community production and biomass allocation. The results showed that warming resulted in a warm but dry microsite, that was, air temperature increased by 1.6 ℃ and soil surface temperature increased by 1.4 ℃, and soil water content decreased by 4.7%. Under no N addition treatments, warming significantly decreased plant aboveground biomass by 61.5%, 108.8% and 77.1% in 2012, 2013 and 2014, respectively. Under high N treatments (40 and 80 kg N·hm-2·a-1), warming had no significant effect on aboveground biomass. These findings indicated that the effect of warming might be dependent on N addition level, and N addition could compensate for soil N loss caused by warming. Warming led to an increase in root/shoot by 98.6%, 60.7% and 97.8% in 2012, 2013 and 2014 under no N addition treatments, respectively. Under the ambient condition, the change percentages of aboveground and belowground biomass of plant communities first increased and then decreased along an N gradient, with the saturation thresholds of above- and below-ground biomass for N addition 56.0 and 55.5 kg N·hm-2·a-1, respectively. Under the warming condition, above- and belowground biomass increased linearly with increasing N addition. These findings suggested that warming modulated the response patterns of alpine meadows to exogenous N input, which was mainly caused by decreased soil inorganic N under warming. The estimation of N thresholds highlights that alpine meadows are more sensitive to future N deposition than other types of grasslands.

The adsorption behavior of Zn2+ in four different biological sludge systems, i.e. activated sludge, denitrification sludge, short-cut nitrification sludge, and anammox granules, was investigated. The results indicated that all sludge samples possessed considerable potential for Zn2+ adsorption. Short-cut nitrification sludge possessed the highest Zn2+ maximum adsorption capacity (qm) of 36.4 mg g SS-1, which was much higher than other sludges applied (12.8-14.7 mg g SS-1). Besides, the adsorption rate for short-cut nitrification sludge was fastest among the four types of sludge after fitting with a pseudo-second-order rate equation. Comparing with the physicochemical properties of the four sludges, the soluble extracellular polymeric substances (EPS), especially soluble polysaccharide (PS), played a prior role in binding metal cations (i.e., Zn). The present study also showed that with less than 30% of Zn2+ trapped by EPS, 61.6-71.9% of Zn2+could be harvested directly by cells, indicating that the protecting capability by EPS was limited. IMPORTANCE: Childhood solid tumors account for the highest proportion of childhood cancers and are one of the leading causes of death in childhood. However, their pathogenesis is unclear. OBJECTIVE: To explore prenatal and perinatal risk factors for solid malignancies in children. METHODS: We enrolled 71 consecutive pediatric patients (44 boys and 27 girls; median age, 30 months) with solid tumors who were diagnosed and treated at our center from January 2013 to December 2016 as the case group. We also enrolled 211 age- and residence-matched healthy children (ratio of approximately 3:1 with the case group) as the control group. We conducted a questionnaire-based survey with the parents of these 282 children. Univariate and multivariate conditional logistic regression analyses of the collected data were performed. RESULTS: Confirmed solid malignancies included neuroblastoma (n = 32), rhabdomyosarcoma (n = 18), retinoblastoma (n = 7), renal tumors (n = 3), and other tumors (n = 11). Risk factors for solid childhood tumors in the univariate analysis were the parents' age, gravidity, parity, abortion history, vaginal bleeding, family history of malignancy, and prenatal use of folic acid or hematinics/iron supplements (P < 0.05), and those in the multivariate analysis were higher parity (odds ratio [OR], 2.482; 95% confidence interval [CI], 1.521-4.048), family history of malignancy (OR, 3.667; 95% CI, 1.679-8.009), and prenatal use of hematinics/iron supplements (OR, 2.882; 95% CI, 1.440-5.767). In contrast, use of prenatal folic acid was protective (OR, 0.334; 95% CI, 0.160-0.694). INTERPRETATION: A family history of malignancy, use of prenatal hematinics/iron supplements, and higher parity are risk factors for solid childhood tumors, whereas use of prenatal folic acid is a protective factor.

Global climate warming and increasing nitrogen (N) deposition, as controversial global environmental issues, may distinctly affect the functions and processes of terrestrial ecosystems. It has been reported that the Qinghai-Tibet Plateau has been experiencing significant warming in recent decades, especially in winter. Previous studies have mainly focused on the effects of warming all the year round; however, few studies have tested the effects of winter warming. To investigate the effects of winter warming and N addition on plant community structure and species composition of alpine meadow, long-term N addition and simulated warming experiment was conducted in alpine meadow from 2010 in Damxung, northern Tibet. The experiment consisted of three warming patterns: Year-round warming (YW), winter warming (WW) and control (NW), crossed respectively with five N gradients: 0, 10, 20, 40, 80 kg N·hm-2·a-1. From 2012 to 2014, both warming and N addition significantly affected the total coverage of plant community. Specifically, YW significantly decreased the total coverage of plant community. Without N addition, WW remarkably reduced the vegetation coverage. However, with N addition, the total vegetation coverage gradually increased with the increase of N level. Warming and N addition had different effects on plants from different functional groups. Warming significantly reduced the plant coverage of grasses and sedges, while N addition significantly enhanced the plant coverage of grasses. Regression analyses showed that the total coverage of plant community was positively related to soil water content in vigorous growth stages, indicating that the decrease in soil water content resulted from warming during dry seasons might be the main reason for the decline of total community coverage. As soil moisture in semi-arid alpine meadow is mainly regulated by rainfalls, our results indicated that changes in spatial and temporal patterns of rainfalls under the future climate change scenarios would dramatically influence the vegetation coverage and species composition. Additionally, the effects of increasing atmospheric N deposition on vegetation community might also depend on the change of rainfall patterns.

OBJECTIVE: Radial corrective osteotomy is an established but challenging treatment for distal radial malunion. There is an ongoing discussion about whether an opening or closing-wedge osteotomy between should employed. The purpose of the present study was to retrospectively compare the clinical and radio graphic results between conventional opening-wedge osteotomy and closing-wedge technique. METHODS: From January 2004 and December 2012,42 patients with extra-articular distal radial malunion were managed with corrective osteotomy and were followed for a minimum of one year. Twenty-two patients (5 males and 17 females, ranging in age from 25 to 75 years old) were managed with radial opening-wedge osteotomy and implanting of interpositional bone graft or bone-graft substitute, and twenty patients (4 males and 16 females, ranging in age from 19 to 79 years) were managed with simultaneous radial closing-wedge and ulnar shortening osteotomy without bone graft. The selection of the surgical procedure was determined by the surgeon. Each patient was evaluated on the basis of objective radio graphic measurements, and functional outcomes were determined on the basis of clinical examinations, including range of wrist motion, grip strength, pain-rating score, Mayo wrist score, and Disabilities of the Arm, Shoulder and Hand (DASH) score. RESULTS: The mean duration of follow-up was 36 months (ranged, 12 to 101 months) for the opening-wedge cohort and 28 months (ranged, 12 to 87 months) for the closing-wedge cohort. The two techniques were comparable in terms of complications. Post-operative volar tilt and ulnar variance were improved significantly in each cohort. The ulnar variance was more frequently restored to within defined criteria (22.5 to 0.5 mm) in the closing-wedge cohort than that in the opening-wedge cohort. The post-operative mean extension-flexion are of the wrist and Mayo wrist score were significantly better in the closing-wedge cohort. Differences in the pronation-supination arc, grip strength, pain-rating score, and DASH scores between these two cohorts were not significant. CONCLUSION: The closing wedge osteotomy technique is an effective reconstructive procedure for the treatment of extra-articular distal radial malunion. It is significantly better than the opening-wedge osteotomy technique in terms of the restoration of ulnar variance, the extension-flexion arc of wrist motion, and the Mayo wrist score.

Alzheimer's disease (AD) is a neurodegenerative brain disorder and the most common cause of dementia. New treatments for AD are required due to its increasing prevalence in aging populations. The present study evaluated the effects of the active components of Epimedium, Astragalus and Radix Puerariae on learning and memory impairment, ß-amyloid (Aß) reduction and brain iron load in an APPswe/PS1ΔE9 transgenic mouse model of AD. Increasing evidence indicates that a disturbance of normal iron homeostasis may contribute to the pathology of AD. However, the underlying mechanisms resulting in abnormal iron load in the AD brain remain unclear. It has been hypothesized that the brain iron load is influenced by the deregulation of certain proteins associated with brain iron metabolism, including divalent metal transporter 1 (DMT1) and ferroportin 1 (FPN1). The present study investigated the effects of the active components of Epimedium, Astragalus and Radix Puerariae on the expression levels of DMT1 and FPN1. The treatment with the active components reduced cognitive deficits, inhibited Aß plaque accumulation, reversed Aß burden and reduced the brain iron load in AD model mice. A significant increase was observed in the levels of DMT1-iron-responsive element (IRE) and DMT1-nonIRE in the hippocampus of the AD mouse brain, which was reduced by treatment with the active components. In addition, the levels of FPN1 were significantly reduced in the hippocampus of the AD mouse brain compared with those of control mice, and these levels were increased following treatment with the active components. Thus, the present study indicated that the active components of Epimedium, Astragalus and Radix Puerariae may exert a neuroprotective effect against AD by reducing iron overload in the AD brain and may provide a novel approach for the development of drugs for the treatment of AD.

Abnormally increased levels of iron in the brain trigger cascade amplification in Alzheimer's disease patients, resulting in neuronal death. This study investigated whether components extracted from the Chinese herbs epimedium herb, milkvetch root and kudzuvine root could relieve the abnormal expression of iron metabolism-related protein in Alzheimer's disease patients. An APPswe /PS1ΔE9 double transgenic mouse model of Alzheimer's disease was used. The intragastric administration of compounds from epimedium herb, milkvetch root and kudzuvine root improved pathological alterations such as neuronal edema, increased the number of neurons, downregulated divalent metal transporter 1 expression, upregulated ferroportin 1 expression, and inhibited iron overload in the cerebral cortex of mice with Alzheimer's disease. These compounds reduced iron overload-induced impairment of the central nervous system, indicating a new strategy for developing novel drugs for the treatment of Alzheimer's disease.

Apurinic/apyrimidinic endonuclease 1 (APE-1), a kind of multifunctional protein widely-distributed in the body, plays an essential role in the DNA base excision repair and serves as multiple possible roles in the response of human cancer to radiotherapy and chemotherapy. In this work, an ultrasensitive solid-state electrochemiluminescence (ECL) immunosensor is designed to determine APE-1 based on the new Ru(bpy)3(2+)/bi-arginine system. The bi-arginine (bi-Arg) is decorated on the Au nanoparticles functionalized magnetic Fe3O4/reduced graphene oxide (bi-Arg/Au@Fe3O4-rGO) according to the self-assembling and covalent cross-linking interaction to obtain the functionalized nanocomposite of bi-Arg/Au@Fe3O4-rGO. Herein, the bi-Arg/Au@Fe3O4-rGO plays not only an amplification label to enhance the ECL signal of Ru(bpy)3(2+) due to the coreactant of bi-Arg but also an ideal nanocarrier to load numerous secondary antibody. Based on sandwich-type immunoassay format, this proposed method offers a linear range of 1.0fgmL(-1)-5.0pgmL(-1) and an estimated detection limit of 0.3fgmL(-1) for the APE-1. Moreover, the reagentless ECL immunosensor also exhibits high sensitivity, excellent selectivity and good stability, which has greatly potential development and application in clinical diagnostics, immunology and biomedical research.
