RESUMEN

Stage Ta/T1 urothelial carcinoma of the bladder (Ta/T1 BC) has a marked tendency to recurrence. Long noncoding RNA HOTAIR has been reported to be expressed in some human cancers such as breast cancer, and it may be positively correlated with patient's prognosis. The aim of our study was to evaluate the prognostic value of HOTAIR in Ta/T1 BC. HOTAIR expression in Ta/T1 BC tissues and adjacent normal tissues was collected from 110 patients and measured by real-time quantitative PCR. The relationships between HOTAIR and the clinical pathological characteristics of Ta/T1 BC patients were analyzed. Immunohistochemistry was done to detect the protein of Wnt inhibitory factor 1 (WIF-1) as well. Ninety out of 110 specimens were detected in HOTAIR high expression. Histological grade and expression levels of HOTAIR were positively correlated with the recurrence rate. HOTAIR expression (hazard ratio 4.712; 95 % CI 2.894-8.714; P < 0.001) was an independent predictor of recurrence rate in multivariate Cox regression analysis. HOTAIR expression is correlated with patients' poor prognosis. A significant inverse correlation between HOTAIR and WIF-1 expression was demonstrated in Ta/T1 BC tissues. The expression levels of HOTAIR are an independent prognostic factor of recurrence in Ta/T1 BC patients.

Water in the mantle transition zone and the core-mantle boundary plays a key role in Earth's stratification, volatile cycling, and core formation. If water transportation is actively running between the aforementioned layers, the lower mantle should contain water channels with distinctive seismic and/or electromagnetic signatures. Here, we investigated the electrical conductivity and sound velocity of ε-FeOOH up to 71 GPa and 1800 K and compared them with global tomography data. An abrupt three-order jump of electrical conductivity was observed above 50 GPa, reaching 1.24(12) × 103 S/m at 61 GPa. Meanwhile, the longitudinal sound velocity dropped by 16.8% in response to the high-to-low spin transition of Fe3+. The high-conductivity and low-sound velocity of ε-FeOOH match the features of heterogenous scatterers in the mid-lower mantle. Such unique properties of hydrous ε-FeOOH, or possibly other Fe-enriched phases can be detected as evidence of active water transportation in the mid-lower mantle.

NaHCO3 with three particle size distributions was employed to determine the minimum inerting concentration (MIC, g/m3) for the explosion of 5µm and 30µm aluminum dust in a standard 20L spherical chamber and thus examine the effect of particle size on the inhibition efficiency. Results showed that the MIC significantly increases as the aluminum particle size decreases from 30µm to 5µm. For 30µm aluminum, the MIC dramatically decreased with the reduction in the NaHCO3 particle size. By contrast, for 5µm aluminum, the MIC was nearly independent of the particle size of NaHCO3 in the range studied. Time-scale analysis indicated that the decomposition of NaHCO3 must be faster than the aluminum combustion reaction for effective chemical inhibition. Scanning electron microscopy showed that the particles of the explosion residues of a NaHCO3/Al mixture were considerably larger than those of pure aluminum explosion residues. A diameter ratio ßmix was defined to evaluate the degree of incomplete reaction promoted by NaHCO3. The composition of the explosion products was analyzed by X-ray photoelectron spectroscopy, and the data revealed that Na2CO3 and Al2O3 are the major species of the products. An inhibition mechanism was proposed based on these results.

The suppressions of ultrafine water mists containing additives (NaCl and NaHCO3) on 100 nm, 5 µm, and 30 µm polymethyl methacrylate (PMMA) dust explosions were experimentally studied in a dust-explosion apparatus. High-speed photography showed that maximum vertical positions and flame propagation velocities were significantly decreased by suppression with ultrafine water mist/additives. Flame propagation velocities in 100 nm, 5 µm, and 30 µm dust explosions suppressed by the ultrafine pure water mist were reduced by 48.2%, 27.7%, and 15.3%, respectively. Maximum temperatures and temperature rising rates measured by a fine thermocouple in nano- and micro-PMMA dust explosions were also significantly decreased. It was proved that the addition of NaCl and NaHCO3 improved the suppression effects of the ultrafine pure water mist. The improvement of explosion suppression by an 8% NaHCO3 mist was superior to that of a 16% NaCl mist. The suppression mechanisms of ultrafine water mist/additives are further discussed by analyzing the physical and chemical effects.

BACKGROUND AND AIMS: Matrix metalloproteinase 14 (MMP14) has been identified to play a significant role in several types of cancers, but little is known about the significance of MMP14 in nasopharyngeal carcinoma (NPC) patients. The aim of this study was to explore the association of MMP14 expression with clinicopathologic features and prognosis in NPC. METHODS: MMP14 mRNA and protein expressions were examined in NPC and nasopharyngeal tissues through real-time PCR and immunohistochemistry. Meanwhile, the relationship of MMP14 expression levels with clinical features and prognosis of NPC patients was analyzed. RESULTS: MMP14 mRNA expression was markedly higher in NPC tissues than in nasopharyngeal epithelium tissues (p = 0.002). Using immunohistochemistry, staining for MMP14 protein was found in the normal nasopharyngeal epithelial cells and malignant epithelial cells, but increased expression of MMP14 was observed in NPC samples compared with normal nasopharyngeal epithelium samples (p = 0.027). In addition, high levels of MMP14 protein were positively correlated with the status of clinical stage (p = 0.009), N classification (p = 0.006), and distant metastasis (p = 0.005) of NPC patients. Patients with higher MMP14 expression had a significantly shorter overall survival time than did patients with low MMP14 expression. Multivariate analysis indicated that the level of MMP14 expression was an independent prognostic indicator (p < 0.001) for the survival of patients with NPC. CONCLUSIONS: MMP14 overexpression is a potentially unfavorable prognostic factor for NPC patients.

Engineering 3D perfusable vascular networks in vitro and reproducing the physiological environment of blood vessels is very challenging for tissue engineering and investigation of blood vessel function. Here, we engineer interconnected 3D microfluidic vascular networks in hydrogels using molded sodium alginate lattice as sacrificial templates. The sacrificial templates are rapidly replicated in polydimethylsiloxane (PDMS) microfluidic chips via Ca⁺²-crosslinking and then fully encapsulated in hydrogels. Interconnected channels with well controlled size and morphology are obtained by dissolving the monolayer or multilayer templates with EDTA solution. The human umbilical vein endothelial cells (HUVECs) are cultured on the channel linings and proliferated to form vascular lumens. The strong cell adhesion capability and adaptive response to shear stress demonstrate the excellent cytocompatibility of both the template and template-sacrificing process. Furthermore, the barrier function of the endothelial layer is characterized and the results show that a confluent endothelial monolayer is fully developed. Taken together, we develop a facile and rapid approach to engineer a vascular model that could be potentially used in physiological studies of vascular functions and vascular tissue engineering.
