Background and Objectives: The incidence of diabetic osteoporosis, an important complication of diabetes mellitus, is increasing gradually. This study investigated the combined effect of the Zuogui pill (ZGP) and eldecalcitol (ED-71), a novel vitamin D analog, on type 2 diabetic osteoporosis (T2DOP) and explored their action mechanism. Materials and Methods: Blood glucose levels were routinely monitored in db/db mice while inducing T2DOP. We used hematoxylin and eosin staining, Masson staining, micro-computed tomography, and serum biochemical analysis to evaluate changes in the bone mass and blood calcium and phosphate levels of mice. Immunohistochemical staining was performed to assess the osteoblast and osteoclast statuses. The MC3T3-E1 cell line was cultured in vitro under a high glucose concentration and induced to undergo osteogenic differentiation. Quantitative real-time polymerase chain reaction, Western blot, immunofluorescence, ALP, and alizarin red staining were carried out to detect osteogenic differentiation and PI3K-AKT signaling pathway activity. Results: ZGP and ED-71 led to a dramatic decrease in blood glucose levels and an increase in bone mass in the db/db mice. The effect was strongest when both were used together. ZGP combined with ED-71 promoted osteoblast activity and inhibited osteoclast activity in the trabecular bone region. The in vitro results revealed that ZGP and ED-71 synergistically promoted osteogenic differentiation and activated the PI3K-AKT signaling pathway. The PI3K inhibitor LY294002 or AKT inhibitor ARQ092 altered the synergistic action of both on osteogenic differentiation. Conclusions: The combined use of ZGP and ED-71 reduced blood glucose levels in diabetic mice and promoted osteogenic differentiation through the PI3K-AKT signaling pathway, resulting in improved bone mass. Our study suggests that the abovementioned combination constitutes an effective treatment for T2DOP.

The intestinal tract is an essential component of the body's immune system, and is extremely sensitive to exposure of ionizing radiation. While ionizing radiation can effectively induce multiple forms of cell death, whether it can also promote ferroptosis in intestinal cells and the possible interrelationship between ferroptosis and intestinal immune function has not been reported so far. Here, we found that radiation-induced major ultrastructural changes in mitochondria of small intestinal epithelial cells and the changes induced in iron content and MDA levels in the small intestine were consistent with that observed during cellular ferroptosis, thus suggesting occurrence of ferroptosis in radiation-induced intestinal damage. Moreover, radiation caused a substantial increase in the expression of ferroptosis-related factors such as LPCAT3 and ALOX15 mRNA, augmented the levels of immune-related factors INF-γ and TGF-ß mRNA, and decreased the levels of IL-17 mRNA thereby indicating that ionizing radiation induced ferroptosis and impairment of intestinal immune function. Liproxstatin-1 is a ferroptosis inhibitor that was found to ameliorate radiation-induced ferroptosis and promote the recovery from immune imbalances. These findings supported the role of ferroptosis in radiation-induced intestinal immune injury and provide novel strategies for protection against radiation injury through regulation of the ferroptosis pathway.

Thymol is a phenolic compound with various pharmacological activities such as anti-inflammatory, anti-bacterial and anti-tumor effects. However, the effect of thymol on bladder cancer cell growth is still elusive. The purpose of this study is to investigate the efficacy of thymol in bladder cancer cells and its underlying mechanism. Thymol inhibited bladder cancer cell proliferation in a dose and time-dependent manner. We also observed cell cycle arrest at the G2/M phase after the treatment of thymol. Moreover, thymol could induce apoptosis in bladder cancer cells via the intrinsic pathway along with caspase-3/9 activation, release of cytochrome c and down-regulation of anti-apoptotic Bcl-2 family proteins. The activation of JNK and p38 was also critical for thymol-induced apoptosis since it was abrogated by the treatment of JNK inhibitor (SP600125), and p38 inhibitor (SB203580) but not ERK inhibitor (SCH772984). Furthermore, the generation of ROS (reactive oxygen species) was detected after the treatment of thymol. ROS scavenger NAC (N-acetyl cysteine) could block the thymol-triggered apoptosis and activation of MAPKs. These findings offer a novel therapeutic approach for bladder cancer.

How does organizational digitalization enable enterprises to achieve sustainable development? To explore this question, in this paper, sustainable development performance is characterized by the corporate's financial performance and environmental performance respectively, and the intermediary role of green technology innovation is explored. Taking China's listed companies from 2011 to 2020 as the research sample, the empirical results suggest the following: (a) corporate digitalization positively affects corporate sustainability in terms of its financial performance and environmental performance; (b) exploratory green-tech innovation fully mediates the relationship between digitalization and corporate sustainability, while the exploitative counterpart partially mediates this relationship and negatively affects financial performance; (c) state-owned enterprises tend to improve environmental performance through digitalization accelerating green-tech innovation process, while non-state-owned enterprises pay more attention to financial performance. Moreover, enterprises in high-tech industries focus on both financial performance and environmental performance, while enterprises in non-high-tech industries emphasize environmental performance. Therefore, this research can be conducive for enterprises to make full use of digitalization and green technology innovation to achieve sustainable development including the improvement of financial performance and environmental performance.

The hydrogen evolution reaction (HER) is important for "green" hydrogen production from water electrolysis. Nowadays, there is an urgent need to construct highly efficient electrocatalysts to boost the HER and achieve hydrogen production. Herein, we present the preparation of a new composite Co-Mo bimetallic sulfide supported on carbon cloth (MoS2/CoS2/CC) via a one-pot hydrothermal sulfurization strategy using (C3H5N2)6[CoMo12O40]·10H2O (CoMo12) as a metal source and thiourea as a sulfur source. The obtained MoS2/CoS2/CC catalyst exhibited outstanding HER ability, with an overpotential of 69 mV when the current density is 10 mA cm-2 in KOH solution, showing comparable performance with those of the advanced Pt/C electrodes tested under the same conditions. Additionally, the results of XRD after the catalytic reaction showed that the electrode had excellent stability in the electrolyte of 1.0 M KOH.

PURPOSE: The aim of the present study was to investigate the injuries of spleen and intestinal immune system induced by 2 Gy 60Co γ ray in mice. MATERIALS AND METHODS: A total of 120 Balb/c mice were randomly divided into two groups: blank control (Ctrl) and model (IR). The IR mice were exposed to a single dose of total body irradiation (2 Gy, dose rate: 1 Gy/min) and sacrificed on 1st, 3rd, 7th, 14th and 21st day after irradiation. The indicators including general observations and body weight, the changes in peripheral hemogram, spleen index, histopathology examination and lymphocyte subsets of spleen. As well as the count and subsets of lymphocyte in gut-associated lymphoid tissue. RESULTS: Compared with the Ctrl group, the body weight, spleen index, peripheral blood cell and splenocyte amounts, intraepithelial lymphocytes number decreased significantly after exposure, accompanied by a notable decreased count of lymphocytes in Peyer's patch and mesenteric lymph nodes. Moreover, ionizing radiation also broke the balance of CD4+/CD8+ and increased the Treg proportion in spleen, which then triggered immune imbalance and immunosuppression. In general, the spleen injuries occurred on 1st day after exposure, worse on 3rd day, and were relieved on 7th day. The intestinal immune injuries were observed on 1st day, and attenuated on 3rd day. On 21st day after exposure, the spleen volume and index have returned to normal, except for the distribution of lymphocyte subpopulations. Furthermore, all indicators of gut-associated lymphoid tissue, except for mesenteric lymph nodes lymphocyte count, had returned to normal levels on 21st day. CONCLUSION: In conclusion, our data showed the injuries of spleen and intestinal immune system induced by 2 Gy 60Co γ ray whole-body irradiation. These findings may provide the bases for further radiation protection in the immunity.

ETHNOPHARMACOLOGICAL RELEVANCE: Ionizing radiation (IR) has found widespread application in modern medicine. As a result, radiotherapy inevitably causes spermatogenic cell injury. Many Chinese herbal prescriptions or natural extracts have the potential to protect against radiation injury. AIM OF THE STUDY: We used GC-2spd cells to investigate the effects and potential mechanisms of YQJD decoction on protecting spermatogenic cells from ionizing radiation injury. MATERIALS AND METHODS: Firstly, the GC-2spd cells were irradiated with 60Co γ-rays (1 Gy, 2 Gy, 4 Gy and 8 Gy) to establish an in vitro model of radiation injury. After that, Cells were divided into six groups: negative control group (NC group), model group (IR group), positive drug group (IRA group), high-dose YQJD decoction (IRH group), medium-dose YQJD decoction (IRM group), and low-dose YQJD decoction group (IRL group). DNA damage, oxidative damage and inflammatory factors were measured. Cell apoptosis and cell cycle were detected by Flow cytometry. Transmission electron microscopy was performed to observe the morphological changes. RESULTS: After irradiation with 60CO γ-ray, the results indicated that the damage of spermatocyte was significantly induced by radiation exposure over 4 Gy. Furthermore, ionizing radiation could make DNA damage and oxidative stress in in GC-2spd cells. In addition, 60CO γ-ray also caused the increase of IL-1ß, IL-6 and TNF-α and the change of cell cycle. However, the application of YQJD decoction inhibited the damage and apoptosis of GC-2spd cells in the aspects of anti-oxidation, promoting DNA damage repair and regulating inflammatory reaction. CONCLUSIONS: Taken together, the protective effects of YQJD decoction on 60CO γ-ray induced spermatocyte injury were confirmed in this study. This exploration might provide a new strategy for the application of Chinese herbs in radioprotection.

ETHNOPHARMACOLOGICAL RELEVANCE: Irradiation-induced immunosuppression often occurs during radiotherapy in patients, which would increase the risk of opportunistic infections. Many Chinese herbal prescriptions or natural extracts have recently attracted increased radiation protection and therapy attention due to their low toxicity. AIM OF THE STUDY: The present study aimed to investigate the protective effects of Yiqi Jiedu (YQJD) decoction on spleen injury induced by 2 Gy 60Co γ ray in mice. MATERIALS AND METHODS: A total of 180 Balb/c mice were randomly divided into five groups: blank control (Ctrl), model (IR), positive drug (IRA), low-dose YQJD decoction (IRL), and high-dose YQJD decoction (IRH). After a ten-day intervention, mice were exposed to a single dose of total body irradiation (2 Gy) and sacrificed on the 1st, 3rd, and 7th day after irradiation. The indicators include general observations and body weight, changes in peripheral hemogram, index and histopathology examination of the spleen, distribution of lymphocyte subsets, cytokine levels, and apoptosis in the spleen. RESULTS: In comparison to the Ctrl group, the body weight, spleen index, peripheral blood cell, and splenocyte quantities decreased significantly after exposure, accompanied by a notable increase of apoptosis in spleen cells. Moreover, ionizing radiation also broke the balance of CD4+/CD8+, Th1/Th2, and Th17/Treg, triggering immune imbalance and immunosuppression. The above injuries occurred on the 1st day after exposure, worsened on the 3rd, and were relieved on the 7th day. However, the pretreatment of YQJD decoction increased the spleen index, improved the spleen structure, and inhibited radiation-induced apoptosis after exposure. Additionally, YQJD decoction has shown its ability to promote immunological balance recovery following exposure by regulating CD4+/CD8+, Th1/Th2, and Th17/Treg ratios, which may minimize the risk of infection. In addition, the high-dose of YQJD decoction showed a better protective effect than the low-dose group. CONCLUSION: The protective effects of YQJD decoction on 2 Gy 60Coγray induced spleen injury were confirmed in this study. This mechanism may be related to inhibiting apoptosis and modulating immune balance. This exploration might provide new insights into the use of Chinese herbs on radioprotection of the immune system.

Ionizing radiation damage refers to acute, delayed, or chronic tissue damage associated with ionizing radiation. Specific or effective therapeutic options for systemic injuries induced by ionizing radiation have not been developed. Studies have shown that Chinese herbal Medicine or Chinese Herbal Prescription exhibit preventive properties against radiation damage. These medicines inhibit tissue injuries and promote repair with very minimal side effects. This study reviews traditional Chinese herbal medicines and prescriptions with radiation protective effects as well as their mechanisms of action. The information obtained will guide the development of alternative radioprotectants.

8­Gingerol, which is extracted from ginger (Zingiber officinale Roscoe), has been shown to possess antioxidant and anti­inflammatory properties. However, the antitumor effect of 8­gingerol has not been fully elucidated. The aim of the present study was to investigate the therapeutic potential of 8­gingerol against colorectal cancer (CRC). The results demonstrated that 8­gingerol significantly inhibited cell proliferation in CRC cell models. Treatment of CRC cells with 8­gingerol resulted in dose­dependent decreases in migration and invasion. The inhibitory effect of 8­gingerol on CRC cell growth was attributed to cell cycle arrest and increased apoptosis. Moreover, to the best of our knowledge, the present study was the first to demonstrate that 8­gingerol acted as an inhibitor of epidermal growth factor receptor (EGFR) signaling. 8­Gingerol inhibited CRC cell proliferation and migration by targeting the EGFR/signal transducer and activator of transcription/extracellular signal­regulated kinase pathway, and the effects of 8­gingerol depended on the expression of EGFR. Moreover, 8­gingerol reduced the effective dosage of 5­fluorouracil and, thereby, the toxicity of drug combination therapy. These data suggest that 8­gingerol may be a promising candidate for the development of novel anticancer agents against CRC.

OBJECTIVE: Yiqi Jiedu (YQJD) decoction is a Chinese herbal prescription, based on an experienced expert of traditional Chinese medicine. It is used for the injuries caused by radiotherapy. The current study was designed to investigate the protective effects of YQJD decoction on radiation damage of testis in mice, and to explore its potential mechanisms. METHODS: Mice were randomly divided into blank control group (Ctrl), model group (IR), positive drug group (IRA), and YQJD decoction group (IRY). After 10-day period intervention, they were whole-body irradiated with 2 Gy 60Co γ-rays and sacrificed on 7th day after irradiation. The indicators including the index and histopathology examination of testis, spermatogenic cell types and apoptosis, and the expression of TLR5, MyD88, NF-κB, TNF-α, IL-6 and Bcl-2 in testis. RESULTS: The testis atrophied significantly on 7th day of exposure to radiation, while YQJD decoction promoted the recovery of testis index and structure. Moreover, spermatogenic cell types and apoptosis had significant changes after irradiation. YQJD decoction protected the testicular function of spermatogenesis, as while as reduced the apoptosis rate of spermatogenic cells. In addition, RT-PCR and immunohistochemical analysis showed that YQJD decoction up-regulated the expression of TLR5 in testis. The levels of TLR5's downstream factors were also up-regulated in YQJD decoction group, which indicated that TLR5 signaling pathway might play an important role in the protective effects of YQJD decoction. CONCLUSIONS: The results showed that YQJD decoction attenuated irradiation induced testis injury in mice. Its potential mechanism was related to TLR5 signaling pathway.

OBJECTIVE: To investigate the intervention effect of Wujia Bugu Recipe (WBR, consisting of Radix Rehmanniae Praeparata, Radix Achyranthis bidentate, Radix Astragali, Radix Angelicae sinensis, acetolytic substance of Concha Ostreae) on the intestinal absorption of exogenous calcium in rats under simulated Weightlessness manner. METHODS: Twenty-one male Wistar rats were randomly divided into 3 groups, the blank group, the model group, and the tested group treated with WBR via gastric perfusion. Simulated weightlessness manner was formed by tail suspending after rats' entry into an one-week adaptation stage. And a definite dose of 41Ca tracer was given to each rat by gastric administration on the 11th day of the weightless stage. Their 72 h feces were collected for measuring ratio of 41Ca/40Ca by accelerator mass spectrometry (AMS), the total calcium in feces was determined by inductively coupled plasma-atomic emission spectroscopy (ICP-AES), the 41Ca feces content (FC) and intestinal absorption amount (AA), and then the calcium intestinal absorption ratio (AR) were analyzed and calculated. RESULTS: 41Ca/40Ca ratio and 41Ca FC were higher, 41Ca AA and AR were lower in the model group than those in the blank group, the differences between groups were significant (P <0.01); while in the tested group, the differences of all above-mentioned indices with those in the blank group were insignificant, although the 41Ca/40Ca ratio showed somewhat increased, yet the increment was less than that in the model group (P <0.05). CONCLUSION: Simulated weightlessness could lead to decrease of intestinal calcium absorption, which could be improved to some extent by Chinese herbal medicine.

UNLABELLED: To study the effect of a Chinese herbal prescription on external calcium deposition to weight-bearing bone in simulated weightlessness rats. METHOD: Twenty-one male Wistar rats were divided into 3 groups: control group, tail suspension group, tail suspension with Chinese medicine group which takes a Chinese herbal prescription extract (containing Radix Rehmanniae Preparata, Radix Acanthopanacis Bidentatae, Radix Astragali, Radix Angelicae Sinensis, Concha Ostreae prepared by acetic acid) by intragastric administration. After 1 week adaption, there start off 3 weeks simulated weightlessness by tail suspension. At the eleventh day of simulated weightlessness, every rat was given one equal dose of 41Ca tracer by intragastric administration. Right femurs were separated as experiment over, and the ratio of 41Ca to 40Ca (41Ca/40Ca) was measured by accelerator mass spectrometry (AMS), while total femur calcium was measured by inductively coupled plasma-atomic emission spectroscopy (ICP-AES). Femur 41Ca deposition amount (DA) and femur 41Ca deposition ratio (DR) were calculated. RESULT: The results showed that compared with control group, 41Ca/40Ca decreased significantly (P < 0.001) in tail suspension group, while in tail suspension with Chinese medicine group, it significantly increased (P < 0.05). DA and DR were both decreased significantly (P < 0.001) in tail suspension group, but no significant change in tail suspension with Chinese medicine group as compared with control group. Compared with tail suspension group, DA and DR increased significantly (P < 0.001) in tail suspension with Chinese medicine group. CONCLUSION: Simulated weightlessness by tail suspension can cause decreased deposition of external calcium to weight-bearing bone, and the Chinese herbal prescription in this trial is effective to prevent the decrease. Moreover, multiple mechanisms may contribute to weightlessness induced osteoporosis, besides calcium deposition disturbance.

OBJECTIVE: To study the effects of Chinese medicine compound on bone metabolism of weightlessness rats simulated by tail suspension. METHOD: Fifty Wistar rats were divided into 5 groups randomly: control group, model group, and low does, medium dose and high does treated group. The experiment period lasted 21 days. After the Chinese compound prescription and distilled water were orally given to treated groups, and control and model group for 7 days, respectively, the tail suspension experiment was performed for treated and model group, meanwhile administration of Chinese compound prescription and distilled water still lasted until the end of the experiment. Blood serum was collected for determination of alkaline phosphatase (ALP), bone gla protein (BGP), tartrate-resistant acid phosphatase (TRAP), femoral bone HOP. The changes of bone mineral density (BMD) of femoral bone and lumbar vertebra were observe. RESULT: Compared with control group, the ALP level of model group was markedly decreased (P < 0.05), no change of BGP, TRAP was not observed, the BMD of femoral bone and lumbar vertebra were decreased remarkably (P < 0.05), Compared with model group, the change of ALP level of treated groups was not significant for all treated groups, the BGP level and BMD for medium dose group were increased (P < 0.05), the TRAP level for medium dose and high does groups was decreased (P < 0.05) CONCLUSION: The Chinese compound prescription can improve the bone formation and prevent bone loss via inhibiting bone absorption and improving ossify, bone mineral deposition and mineralization as well as increasing BMD, which leads to prevention and treatment of bone loss.

Microgravity has been previously demonstrated to induce skeletal muscle atrophy, loss of muscle force and disorders in myogenesis and metabolism. Current pharmacological strategies exhibit poor efficacy. Bu Zhong Yi Qi decoction (BZ) is a well­known traditional Chinese medicine decoction used for myasthenia gravis. In the present study, its effect on unloading induced muscle atrophy was investigated. The mousetail suspension model was used to simulate weightlessness induced muscle atrophy. The results indicated that BZ could significantly protect muscles from simulated weightlessness­induced atrophy. To elucidate the underlying mechanisms, drugCIPHER­CS methods were introduced to predict its potential targets, significantly enriched pathways and biological processes. The results demonstrated that the calcium signaling pathway, citrate cycle, biosynthetic and lipid metabolic process are affected by BZ. Among the targets, nuclear receptor corepressor 1 (NCoR1) is one of the most important proteins involved in myogenesis and metabolism. The results indicated that BZ significantly downregulated NCoR 1 expression, and further induced muscle differentiation and metabolism by regulating NCoR1­associated gene expression in vivo and in vitro. In summary, the present study indicated that may be effective in combating weightlessness­induced muscle atrophy. Combined with bioinformatics, the underlying mechanism for this decoction was investigated, which provided an improved understanding of this decoction.

Recent studies indicate important roles for long noncoding RNAs (lncRNAs) as essential regulators of myogenesis and adult skeletal muscle regeneration. However, the specific roles of lncRNAs in myogenic differentiation of adult skeletal muscle stem cells and myogenesis are still largely unknown. Here we identify a lncRNA that is specifically enriched in skeletal muscle (myogenesis-associated lncRNA, in short, lnc-mg). In mice, conditional knockout of lnc-mg in skeletal muscle results in muscle atrophy and the loss of muscular endurance during exercise. Alternatively, skeletal muscle-specific overexpression of lnc-mg promotes muscle hypertrophy. In vitro analysis of primary skeletal muscle cells shows that lnc-mg increases gradually during myogenic differentiation and its overexpression improves cell differentiation. Mechanistically, lnc-mg promotes myogenesis, by functioning as a competing endogenous RNA (ceRNA) for microRNA-125b to control protein abundance of insulin-like growth factor 2. These findings identify lnc-mg as a novel noncoding regulator for muscle cell differentiation and skeletal muscle development.

OBJECTIVE: To study the effects of Qilan Tangzhining capsule on glucose and lipid metabolism in diabetes mellitus and hyperlipemia. METHOD: Rats were fed with high sugar and fat food for 2 months, then 2% streptozocin (STZ, 30 mg kg(-1)) was injected intraperitoneally to induce hyperglycemia and hyperlipemia. After the model rats were administrated drugs, the blood glucose, the serum lipids, and the histopathology of the liver and pancreas were observed. RESULT: Qilan Tangzhining capsule could decrease the FBG and the serum TC, TG, LDL-C, increase HDL-C, and improve the histopathologic injury of the liver and pancreas. CONCLUSION: Qilan Tangzhining capsule can not only adjust the blood glucose and lipid, but also improve the histopathology injury of the liver and pancreas.

OBJECTIVE: To investigate the behavioral changes and the levels of monoamine neurotransmitters in the anterior cortex in the olfactory bulb damage rats after being treated with Guanyu capsules (GYC). METHOD: Open-field test and step-down passive avoidance test were used to observe the behavior in model rats. HPLC-ECD was used to analyze the influences of GYC on the levels of monoamine neurotransmitters. RESULT: In the model rats, there was a characteristic hyperactivity in the Open-field and learning deficits in step-down passive avoidance (P < 0.01). The contents of 5-HT reduced, and the rate of DOPAC/DA increased significantly (P < 0.01). GYC 1.2, 0.6 g x kg(-1) could correct behavioral changes increase the contents of 5-HT, and decrease DOPAC/DA level (P < 0.01). CONCLUSION: GYC can correct behavioral changes in rats model of olfactory bulb damage, and regulating 5-HT and DA metabolism in cortex is one of the antidepressive mechanisms of GYC.

Weightlessness environment can lead to the muscle atrophy and body fluid distribution upward,which can cause the bone calcium metabolism disorder and always accompanied by the loss of bone microstructure and increased rate of bone fracture. Under microgravity,the astronauts are much easier to decrease the Ca2+ ion in bone, which can cause serious osteoporosis. However the bone lost is not equilibrium, it is especially serious in the mechanism loading bone and the recovery process is more difficult. These are very different from the osteoporosis in older people and postmenopausal osteoporosis. It is necessary to find an optimal method to due with it. In traditional Chinese medicine theory,the kidney stores "Jing" and dominates the bone, thus a lot of bone related diseases can be treated through the kidney. A lot of clinical practices have also proved that the Chinese herbs used under the guidance of basic Chinese medicine theory are always good at the treatment of common osteoporosis. In simulated weightlessness experiment, people found that the kidney nourishment drugs do can prevent the decrease of BMD. So in this article we want to review the causes of weightlessness and the potentials applications of tradition Chinese medicine in the treatment of weightlessness osteoporosis.
