RESUMEN

The sex pheromone for the winter moth, Operophtera brumata (L.), has been identified as the novel compound (Z,Z,Z)-1,3,6,9-nonadecatetraene. The male moths respond to the pheromone at low temperatures (4 degrees to 15 degrees C) and exhibit an upper response limit that coincides with the lower response limit for other reported moth sex pheromone systems. The pheromone attracted two other geometrid species, O. bruceata (Bruce spanworm) and O. occidentalis.

Current-voltage (J-V) characteristics of poly(3-hexylthiophene) (P3HT) are studied at different temperatures upto high voltages approximately 20V in the hole-only device configuration. The characteristics are studied in the temperature range 310-210K. In the intermediate voltage range the J-V characteristics follow J proportional to V(l+1), where l > 1. As the voltage increases to high values J still varies as a power law i.e. as V(m), but contrary to the literature result m becomes < 2. This behavior is explained theoretically in terms of non-zero injection Schottky barriers. The complete analytical expressions for the actual trap filled limit voltage (V'(TFL)) and J-V curves beyond V'(TFL) are presented.

We report here the crystallographic structure determination of an autoprocessed (Ser221Cys)-subtilisin E-propeptide complex at 2.0 A resolution. The subtilisin domain sequence has a single substitution (Ser221Cys) which has been shown to block the maturation process prior to degradation of the propeptide domain (77 residues) that acts as an intramolecular chaperon. This mutation, however, did not prevent the enzyme from cleaving its propeptide domain with a 60-80% efficiency. The current determination is the first example of a subtilisin E-propeptide complex which has been autoprocessed. A previous structure determination of a BPN'-prosegment complex has been reported in which the subtilisin domain was extensively mutated and a calcium binding loop was deleted. Further, in this earlier determination, the complex was formed by the addition of separately expressed propeptide domain. The structure determination reported here provides additional information about the nature of the interaction between the subtilisin and propeptide domains in this complex.

We have earlier established in tissues of several mammalian animal species the existence of a novel membrane bound enzyme termed 7,8-diacetoxy-4-methylcoumarin (DAMC): protein transacetylase (TAase) that possibly transfers acetyl groups from the model acetoxy drug (DAMC) to certain enzyme protein viz. glutathione S-transferase (GST), cytochrome P-450 and NADPH cytochrome C reductase leading to the drastic modulation of their catalytic activities. We have in this report extended the studies to human tissue and characterized TAase from placenta. For this purpose placental microsomes were preincubated with DAMC along with the receptor protein (cytosolic GST) followed by the addition of the substrates of GST in order to quantify the catalytic activity of GST, the extent of inhibition of GST served as a measure of TAase. Placental TAase was also found to irreversibly activate NADPH cytochrome C reductase by DAMC. Placental enzyme activated the reductase even at very low concentration of DAMC. Iodoacetamide nearly abolished the placental TAase suggesting the presence of active thiol group in the enzyme and the TAase demonstrated hyperbolic kinetics. Kinetic constants obtained by varying the concentrations of either of the substrates DAMC or cytosolic GST characterized TAase catalysed reaction as the bimolecular reaction. Further studies are in progress to delineate the physiological importance of TAase in placenta.

Changes in blood glucose and glucose tolerance by oral glucose tolerance test (OGTT) after 40 days of yoga therapy in 149 non-insulin-dependent diabetics (NIDDM) were investigated. The response to yoga in these subjects was categorized according to a severity scale index (SSI) based on area index total (AIT) under OGTT curve. One hundred and four patients showed a fair to good response to the yoga therapy. There was a significant reduction in hyperglycemia and AIT with decrease in oral hypoglycemia and AIT with decrease in oral hypoglycemic drugs required for maintenance of normoglycemia. It is concluded that yoga, a simple and economical therapy, may be considered a beneficial adjuvant for NIDDM patients.

Ethidium forms a crystalline complex with the dinucleoside monophosphate, uridylyl (3'-5') adenosine (UpA). The complex crystallizes in the monoclinic space group P2l with unit cell dimensions, a = 13.704 A, b = 31.674 A, c = 15.131 A, beta = 113.9 degrees. This light atom structure has been solved to atomic resolution and refined by full matrix least squares to a residual of 0.12, using 3,034 observed reflections. The asymmetric unit consists of two ethidium molecules, two UpA molecules and 19 solvent molecules, a total of 145 non-hydrogen atoms. The two UpA molecules are hydrogen-bonded together by Watson-Crick type base pairing. Base-pairs in this duplex are separated by 6.7 A; this reflects intercalative binding by one of the ethidium molecules. The other ethidium molecule stacks on either side of the intercalated base-paired dinucleoside monophosphate, being related by a unit cell translation along the a axis. The conformation of the sugar-phosphate backbone accompanying intercalation has been accurately determined in this analysis, and contains the mixed sugar-puckering pattern: C3' endo (3'-5') C2' endo. This same structural feature has been observed in the ethidium-iodoUpA and ethidium-iodoCpG complexes, and exists in two additional structures containing ethidium-CpG. Taken together, these studies confirm our earlier sugar-puckering assignments and demonstrate that iodine covalently bound to the C5 position on uridine or cytosine does not alter the basic sugar-phosphate geometry or the mode of ethidium intercalation in these model studies. We have proposed this stereochemistry to explain the intercalation of ethidium (as well as other simple intercalators) into both DNA and into double-helical RNA, and discuss this aspect of our work further in this paper and in the accompanying papers.

This paper describes two complexes containing ethidium and the dinucleoside monophosphate, cytidylyl(3'-5')guanosine (CpG). Both crystals are monoclinic, space group P2l, with unit cell dimensions as follows: modification 1: a = 13.64 A, b = 32.16 A, c = 14.93 A, beta = 114.8 degrees and modification 2: a = 13.79 A, b = 31.94 A, c = 15.66 A, beta = 117.5 degrees. Each structure has been solved to atomic resolution and refined by Fourier and least squares methods; the first has been refined to a residual of 0.187 on 1,903 reflections, while the second has been refined to a residual of 0.187 on 1,001 reflections. The asymmetric unit in both structures contains two ethidium molecules and two CpG molecules; the first structure has 30 water molecules (a total of 158 non-hydrogen atoms), while the second structure has 19 water molecules (a total of 147 non-hydrogen atoms). Both structures demonstrate intercalation of ethidium between base-paired CpG dimers. In addition, ethidium molecules stack on either side of the intercalated duplex, being related by a unit cell translation along the a axis. The basic feature of the sugar-phosphate chains accompanying ethidium intercalation in both structures is: C3' endo (3'-5') C2' endo. This mixed sugar-puckering pattern has been observed in all previous studies of ethidium intercalation and is a feature common to other drug-nucleic acid structural studies carried out in our laboratory. We discuss this further in this paper and in the accompanying papers.

Thalassaemic children being multi-transfused are at increased risk of parenterally transmissible hepatitis viruses and majority of them prone to develop chronic liver disease. The study is designed to find out the prevalence of hepatitis B virus (HBV), hepatitis C virus (HCV) and hepatitis D virus (HDV) seromarkers and correlation of age, sex, number of transfusions and the viral aetiology in thalassaemics of central India. One hundred and four thalassaemic children were subjected to clinical, biochemical and serological analysis for the HBV, HCV and HDV viruses. The chi(2) test was applied to check the statistical significance of different variables. In the present study HBV markers were detected in 57 (56%) of the subjects while anti-HCV antibodies were observed in 21% of the patients. However, only four subjects were detected hepatitis B surface antigen (HBsAg) reactive but none of them were reactive for anti-HDV antibodies. Forty patients had raised alanine transaminase (ALT) levels and among them two were HBsAg reactive, 16 were anti-HBc antibody positive and 14 were anti-HCV reactive. The prevalence of hepatitis viruses and raised ALT levels are found to be significantly associated with the increasing age and number of blood units transfused to them. The present findings also document the excellent contribution of stringent screening of blood units and HBV vaccination programme for containing the HBV infection among thalassaemics.

Oesophageal electrocardiograms were recorded in 25 normal persons, 20 patients with chronic bronchitis and predominant emphysema without right ventricular hypertrophy and 25 patients with chronic cor pulmonale as an end result of chronic obstructive pulmonary disease. At ventricular level, patients with chronic cor pulmonale had higher P wave height, lower Q wave depth, lower R wave height, deeper S waves and lower R/S ratio as compared to the group with emphysema. In terms of the transitional zone, patients with cor pulmonale had smaller Q waves and a lower Q/R ratio. At atrial level, the patients with cor pulmonale had higher heights of the P and R waves and a lower Q/R ratio. All these differences were highly significant statistically (P less than 0.001). An R/S ratio of less than 2.5 at ventricular level had high sensitivity (96.0%), specificity (100%) and accuracy (98%) in diagnosing right ventricular hypertrophy in cases of chronic obstructive pulmonary disease. This criterion obtained from the oesophageal electrocardiogram is much more sensitive and accurate than those obtained routinely from the surface electrocardiogram in diagnosing right ventricular hypertrophy in cases of chronic obstructive pulmonary disease.

RESUMEN

In the Marinelli chair, a geometry widely used in whole body counting, the lower part of the leg is seen quite inefficiently by the detector. The present paper describes an attempt to modify the standard chair geometry to minimise this limitation. The subject sits crossed-legged in the "Buddha Posture" in the standard chair. Studies with humanoid phantoms and a volunteer sitting in the Buddha posture show that this modification brings marked improvement over the Marinelli chair both from the point of view of sensitivity and uniformity of spatial response.

The available limited anatomical and physiological data have been compiled for Indian adult as a part of ongoing efforts in the country to formulate an Indian Reference Man for radiation protection purposes. The parameters included weight, height, body surface area, weight of various organs (brain, heart, kidney, lung, liver etc.), total body water, extra-cellular water, blood volume, specific gravity, total lung capacity, vital capacity etc. The data have been drawn from post-mortem studies (566 males and 185 females) and other data from published work on 433 healthy male and 137 female volunteers. Data gaps have been filled by the method of pooling wherever necessary. This paper presents the intercomparison of the present data with the data available from other sources. The data from various Indian sources indicate a similar trend. However, a significant difference (P less than 0.01) was observed in organ weights and total body weights, in comparison to the Bhabha Atomic Research Centre (BARC) values. The present data are not comprehensive enough to suggest definitive norms of the Indian Reference Man.

RESUMEN

An ethanolic extract of the flowers of Calotropis procera was investigated for anti-inflammatory, antipyretic, analgesic and antimicrobial activities. The plant extract reduced the paw swelling induced by carrageenan by 37%, fever in rats by 40% and showed some weak effect in rats on the writhing induced by acetic acid. The growth of both gram-positive and gram-negative bacteria was significantly inhibited. A dose-dependent effect on prostaglandin release was also observed.

An ethanolic extract of the rhizomes of Zingiber officinale was investigated for anti-inflammatory, analgesic, antipyretic, antimicrobial and hypoglycaemic activities. In rats, the extract reduced carrageenan-induced paw swelling and yeast-induced fever but was ineffective in suppressing the writhing induced by intraperitoneal acetic acid. The extract produced blood glucose lowering in rabbits. The growth of both Gram-positive and Gram-negative bacteria was significantly inhibited. A dose-dependent inhibition of prostaglandin release effect was observed using rat peritoneal leucocytes.

In the present study, the ethanolic extract of the whole plant parts (root, stem leaves and pods) of Cassia italica (Mill.) Lam. ex F.W. Ander (Leguminosae) was investigated for bioactivities: namely anti-inflammatory, antipyretic, analgesic and prostaglandin (PG) release by rat peritoneal leucocytes, antineoplastic and antiviral. In rats, the extracts reduced carrageenin-induced paw swelling (100 mg/kg bw-31%) and fever (100 mg/kg bw-37%). The extract showed weak effect on writhing induced by acetic acid. A dose-dependent inhibition of PG release effect was observed using rat peritoneal leucocytes.

PIP: Applicants for therapeutic abortion at an outpatient unit of a general hospital in the capital city of Newfoundland were interviewed, and the contraception related findings are presented. The total number of therapeutic abortions performed in 1977 in the province was 493, of which 416 (84%) were performed in Grace General Hospital, where the study was conducted. The 416 women ranged in age from 14-45 years. 72% were under age 24. 41% had achieved only elementary school education. 65% came from the capital city and from the surrounding areas, within a radius of 100 miles. The rest of the women had traveled up to 700 miles or more to procure the service. Of the total of 416, 10, or 2%, claimed that they had no knowledge that there are ways to prevent conception. The 2nd group of 170 women (41%) knew that contraceptive methods did exist but for various reasons had never used any method at any time in their lives. The 3rd group of 236 (57%) consisted of those who had used some form of contraception. This group was further divided: 181 women (44%) had used birth control methods within the last 6 months or earlier but not at the time of conception; the rest had been consistent contraceptors but the method had failed. Of the 10 women who claimed they had absolutely no knowledge of contraception, 6 were below age 17, 7 were unmarried, and 5 were Roman Catholics. Approximately 1/3 of the 170 women who knew of contraceptive methods but did not use them were under age 16 and would have required parental consent to receive contraceptive counseling and/or prescriptions. For approximately 2/3 of this group, birth control methods were available and accessible since they were above the age of 17 and could have sought contraceptive counseling. The reasons this group of women gave for not using contraceptives fell under 3 main headings. 1/3 attributed it to ignorance and/or carelessness. Another 1/3 consisted of those who had not planned on having sexual intercourse. The rest consisted mainly of young women whose parental attitude had deterred them from seeking help with birth control. The 181 women who had used some form of contraception within the last 6 months or earlier but not at the time of conception were mainly over age 17. More than half of them were unmarried. Less than 1/3 had elementary school education, and half had high school or more education. The complaint of side effects caused by the method was the most common reason for non-use among this group. In most instances the method complained about was oral contraception. More than half of the consistent users were married and almost 3/4 had high school or better education. The contraceptive that appears to have had the highest failure rate among the consistent users was the condom. Almost 1/3 of the women claimed that they had depended on the condom. The oral contraceptive, the IUD, and irreversible methods such as tubal ligation and vasectomy had a failure rate of 2% each.^ieng

A study of the effect of yoga therapy programme on 46 indoor patients of chronic bronchial asthma on exercise capacity, pulmonary functions and blood gases was conducted. Exercise capacity was measured by 3 tests: (i) 12 min walk test (12-md); (ii) physical fitness index (PFI) by modified Harvard step test; and (iii) Exercise-Liability index (ELI). Yoga therapy programme resulted in a significant increase in the pulmonary functions and exercise tolerance. A one-year follow-up study showed a good to fair response with reduced symptoms score and drug requirements in these subjects. It is concluded that yoga therapy is beneficial for bronchial asthma.

The recovery of pulmonary function were studied in fifty patients of acute bronchial asthma receiving a standard therapeutic regime. Sixty-two per cent of patients had achieved 50% of their total improvement in peak expiratory flow rate (PEFR) within 24 hours (fast responders) as against slow responders. Duration of asthma, characteristics of present exacerbation, mean pulse rate and presence of pulsus paradoxus on admission did not differ in fast and slow responders. The rise in PEFR within 4 hours of starting treatment was highly significantly correlated with a higher PEFR at 24 hours and a faster recovery. The mean arterial PaCO2 was higher (P less than 0.02) in slow responding group and they were slightly older (P less than 0.01), had lower mean FVC (P less than 0.01), mean FEV1 (P less than 0.02) and PEFR (P less than 0.001). The mean PaO2 of less than 80 mm Hg at 48 hours was more common in those with delayed recovery of PEFR.

Data on heights and weights of Indian males and females of various ages are presented. The weights of specific organs obtained from post-mortem records of accident cases of Indian adults and children are also presented to provide some inputs towards formulation of an Indian Reference Man for radiation protection purposes. An intercomparison of the present data with the data available from other sources including those for ICRP Reference Man, Woman, and Child has been made. The daily intakes of air, water, and dietary constituents of an average Indian adult are also given.

Haematologic changes were studied in 16 albino rabbits during acclimatisation, deacclimatisation, and reinduction to hypobaric hypoxia. Of these, four animals died during reinduction. Haemoglobin, haematocrit, and red blood cell count attained maximum values by the 15th day during acclimatisation and by the 10th day during reinduction. Reticulocyte count reached maximum level by the fifth day, both during acclimatisation and reinduction. All these parameters returned to initial control levels by the 10th day of deacclimatisation. The erythrocytes became macrocytic during acclimatisation and remained so during deacclimatisation and reinduction. Blood volume increased on acclimatisation, returned to control values during deacclimatisation, and remained low at this level on reinduction. Red cell mass increased both during acclimatisation and reinduction and returned to control levels on deacclimatisation. Plasma volume did not change on acclimatisation and deacclimatisation, but was reduced on reinduction. The erythrocyte life span and erythrocyte glutamic oxaloacetic transaminase (EGOT) decreased during acclimatisation and reinduction. There was no change in RBC osmotic fragility, indicating no change in the integrity of the red cell wall. The deaths during reinduction were possibly due to haemoconcentration and increased blood viscosity as a result of reduction in plasma volume. It has been concluded that reinduction to hypoxia after a phase of deacclimatisation imposes a more severe stress than experienced during acclimatisation.
