BACKGROUND: Hematologic and blood biochemical values are key tools for assessing primate health. A long-term behavioral study of howler monkeys at a single site (La Pacífica, Guanacaste, Costa Rica), afforded the opportunity to develop baseline values for a large group of animals, evaluating differences between adult males and females and comparing to a report in the same population two decades later. METHODS: In 1998, 64 free-ranging mantled howler monkeys were anesthetized and sampled for hematologic and biochemical analysis. RESULTS: Blood analysis is reported for 29 adult females, 9 juvenile females, 19 adult males and 3 juvenile males. Four adults were excluded due to external injury or disease. There were few significant differences between adult females, juvenile females, and adult males. CONCLUSIONS: Baseline blood parameters are useful for determining normal values for howler monkey populations. The values for total protein, blood urea nitrogen, glucose, liver enzymes and potassium differed from a later study in 2019 may indicate changes that are influencing howler monkey health.

Over a 25-mo span from a single zoologic collection, two bufflehead ducklings (Bucephala albeola) presented with pelvic limb paresis and were euthanized. On postmortem examination, the first duckling had intralesional fungal hyphae consistent with Aspergillus sp. in the spinal vertebrae and within pulmonary granulomas. In the second duckling, evidence of a thoracic spinal lesion was detected antemortem by using thermographic imaging. At postmortem examination, fungal hyphae consistent with Mucor sp. were found within the vertebrae. Although fungal infections of the respiratory system are commonly reported in waterfowl, infections that involve the spinal cord and vertebrae are unusual. These cases highlight the importance of consideration of axial skeleton fungal disease in neurologic presentations and the use of thermography for noninvasive diagnostic screening.

This study evaluated selected bacteriologic and nutritional components of a commercially prepared ground raw horsemeat diet as part of a raw meat safety program. Six lots of meat were analyzed in triplicate. Frozen meat samples were thawed for 44 hr at 5°C. Meat samples were tested at three times during thawing (t = 0, 24, 44 hr) for selected bacteria. Samples were screened for Salmonella sp. using an enzyme-linked immunosorbent assay. Escherichia coli and total coliform bacteria were quantified using a ready-made culture medium system. Proximate, energy, macro and trace mineral composition was determined at a reference laboratory. Salmonella sp. antigen was detected in one sample of meat at t = 0 hr. Frozen meat samples had low average maximum expected numbers of E. coli and coliforms. The average maximum number of E. coli did not change significantly at t = 24 or 44 hr, but the average maximum number of coliforms increased significantly by t = 44 hr. These bacteriologic tests were easily incorporated into a raw meat safety program. Median concentrations of moisture, dry matter, crude protein, crude fat, ash, calcium, and phosphorus conformed to the guaranteed analysis but median crude fiber exceeded the guaranteed maximum. Median magnesium, potassium, and sodium concentrations conformed to the approximate nutrient content. Median concentrations of copper exceeded, whereas iron, zinc, and manganese fell below, the approximate nutrient content. Median copper and manganese concentrations exceeded the National Research Council's recommendation for adult domestic cats, whereas iron and zinc were below the National Research Council's recommendations for adult cats.

Capture myopathy and associated death have been reported with capture and restraint of greater flamingos (Phoenicopterus ruber roseus) and lesser flamingos (Phoeniconaias minor). In chickens (Gallus gallus), blood lactate concentration levels have been used as indicators of muscle damage. Lactate has also been used to predict survival in humans and dogs. The goals of this study were to validate two common methods for measuring lactate (i-STAT and VetTest analyzers) in flamingo plasma by comparing measurements to a reference analyzer; and to correlate blood lactate concentration levels in captured flamingos with the duration and difficulty of capture as a possible indicator of capture myopathy. Twenty-seven banked flamingo plasma samples were run in triplicate on each of the three blood analyzers. Values from the i-STAT analyzer were consistently lower than those from the ABL analyzer, while values from the VetTest were consistently higher than those from the ABL analyzer. However, there was a good level of correlation between all three analyzers. Two of the three analyzers were determined to have acceptable total allowable error levels, calculated at 3.6% for the ABL and 10.7% for the VetTest. For clinical purposes, both the i-STAT and the VetTest analyzers provide adequate evaluation of lactate levels when serial samples are measured on the same analyzer. After validating the assay, 34 captive flamingos were captured for routine examinations. Blood lactate concentration levels were positively correlated with the length of time of the individual capture, but lactate did not increase significantly as capture difficulty increased. Only one animal was considered to have a difficult capture. No flamingos demonstrated clinical signs of capture myopathy during this study. Further research is required to determine if blood lactate concentration is a useful indicator of capture myopathy.

Rapid and sensitive diagnostic assays for the detection of tuberculous mycobacteria in elephants are lacking. DNA extraction with PCR analysis is useful for tuberculosis screening in many species but has not been validated on elephant trunk wash samples. We estimated the analytical sensitivity and specificity of three DNA extraction methods to detect Mycobacterium tuberculosis complex organisms in trunk wash specimens. A ZR soil microbe DNA kit (ZR) and a traditional salt and ethanol precipitation (TSEP) approach were evaluated under three different treatment conditions: heat treatment, phenol treatment, and contamination with Mycobacterium avium. A third approach, using a column filtration method, was evaluated for samples contaminated with soil. Trunk wash samples from uninfected elephants were spiked with various concentrations of M. bovis cells and subjected to the described treatment conditions prior to DNA extraction. Extracted DNA was amplified using IS6110-targeted PCR analysis. The ZR and TSEP methods detected as low as 1 to 5 M. bovis cells and 10 M. bovis cells, respectively, per 1.5 ml of trunk wash under all three conditions. Depending on the amount of soil present, the column filtration method detected as low as 5 to 50 M. bovis cells per 1.5 ml of trunk wash. Analytical specificity was assessed by DNA extraction from species of nontuberculous mycobacteria and amplification using the same PCR technique. Only M. bovis DNA was amplified, indicating 100% analytical specificity of this PCR technique. Our results indicate that these DNA extraction techniques offer promise as useful tests for detection of M. tuberculosis complex organisms in elephant trunk wash specimens.

A 19-yr-old intact male Coquerel's sifaka (Propithecus coquereli) was evaluated for a crusting facial dermatopathy. Fungal culture and histopathology of skin biopsies were consistent with dermatophytosis caused by Trichophyton mentagrophytes, and treatment with the antifungal medication terbinafine was initiated. After 1 mo of treatment, all clinical signs had resolved and a fungal culture of the skin was negative. The sifaka was treated with terbinafine for a total of 81 days. Two additional fungal cultures were taken and found to be negative for the presence of dermatophytes, the last culture being taken 1 mo after discontinuation of terbinafine. To the authors' knowledge, this is the first reported case of dermatophytosis in a prosimian species and the first reported treatment of a prosimian with the antifungal drug terbinafine.

Telazol has been commonly used for field anesthesia of wild lemurs, including ring-tailed lemurs (Lemur catta). Telazol alone provides good induction, but doesn't cause adequate muscle relaxation and sedation for collecting consistent somatic measurements and high-quality dental impressions that are sometimes needed. Variability in induction response has been seen between individuals that have received similar dosages, with young lemurs seeming to need more anesthetic than mature lemurs. This investigation evaluated Telazol induction in young (2.0-4.9 yr) and mature (> or = 5.0 yr) ring-tailed lemurs and compared postinduction supplementation with medetomidine or medetomidine-butorphanol. Forty-eight lemurs were anesthetized with Telazol administered via blow dart; then, 20 min after darting, they were supplemented via hand injection with either medetomidine (0.04 mg/ kg) or medetomidine-butorphanol (0.04 mg/kg and 0.2 mg/kg, respectively). The odds ratio for young lemurs to need more than one dart for induction, relative to mature lemurs, was 3.8, even though the initial dose of Telazol received by young lemurs (19 +/- 7 mg/kg) was significantly higher than the initial dose administered to mature lemurs (12 +/- 5 mg/kg). The total Telazol dosage was also significantly different between young lemurs (33 +/- 15 mg/kg) and mature lemurs (18 +/- 9 mg/kg). Both medetomidine and medetomidine-butorphanol provided good muscle relaxation and sedation for all procedures. Physiologic values were similar between the two protocols. Oxygen saturation by pulse oximetry was generally good, although there were a few SaO2 values < 90%. Recoveries were smooth, but long. Time to head up was correlated with total Telazol dosage in mature lemurs. In young lemurs, time to standing was correlated with Telazol induction dosage and time of last Telazol administration. Lemurs that received hand injections of Telazol took longer to recover than those that did not. Further refinements are needed to increase induction reliability and to decrease recovery time, particularly in young lemurs.

West Nile virus (WNV) was first documented in North America in New York City in 1999. Several deaths attributable to WNV have been reported in captive thick-billed parrots (Rhynchopsitta pachyrhyncha), an endangered psittacine native to North America. The serologic responses in 12 captive adult thick-billed parrots after a series of three initial WNV vaccine injections with annual boosters over 6 yr was evaluated. In addition, the serologic responses of 11 thick-billed parrot chicks following an initial vaccination series to determine if there were seroconversions were also reported. Most adults (67%) had seroconverted after 5 yr of annual vaccination, with a median titer of 1:80 (range 1:40-1:160) for those that seroconverted. After the first year, birds were likely naturally exposed to WNV, which limited interpretation of titers. None of the chicks seroconverted during the initial three-vaccine series; only two of four chicks (50%) had seroconverted when tested at the 1-yr yearly booster, and at 2 yr, three of four chicks had seroconverted. Although some birds had detectable antibody titers, it is unclear whether this vaccine can reliably provide protection against WNV in thick-billed parrots.

Wild ring-tailed lemurs (Lemur catta) can be anesthetized with Telazol via blow dart, but improved techniques are needed so that each lemur is reliably induced with a single dart. Medetomidine-butorphanol (MB) is a good supplemental protocol to be administered once the lemurs are captured, but other protocols may provide longer periods of sedation and immobility. One possible way of increasing the efficacy of each dart is to increase the time it is retained in the leg. In this investigation, a "double-sleeve" technique was used to try to increase the time of dart retention. This technique used a standard silicone sleeve on the needle, along with a second sleeve at the needle hub. Induction values were compared between lemurs darted with double-sleeve needles and those induced with needles that each had a single silicone sleeve. Once the lemurs were induced, supplementation with MB (0.04 mg/kg and 0.2 mg/kg) was compared with supplementation with ketamine-medetomidine (KM) (10 mg/ kg and 0.04 mg/kg). Twenty-three lemurs were darted with Telazol by using single-sleeve needles, and 24 were darted with double-sleeve needles. The number of darts per lemur and number of escapes were not different between animals darted with a single sleeve compared with a double-sleeve; thus, there were no significant improvements in induction success with the double-sleeve technique. Adequate sedation and muscle relaxation were achieved with both MB and KM; however, lemurs that received MB were more relaxed and needed fewer additional supplements that those that received KM. Single-sleeve dart needles are recommended for Telazol induction of ring-tailed lemurs via blow dart and MB is preferable to KM for supplemental sedation and muscle relaxation.

This study compared immobilization of gemsbok (Oryx gazella) using thiafentanil-xylazine (TX) versus carfentanil-xylazine (CX). Twelve adult gemsboks were divided into two groups, TX and CX. Each group received either 6 mg thiafentanil (0.036 microg/kg (0.032-0.040 microg/kg) and 20 mg xylazine (TX) or 3.6 mg carfentanil (0.021 mg/kg [DOSAGE ERROR CORRECTED] (0.017-0.024 mg/kg) [DOSAGE ERROR CORRECTED] and 20 mg xylazine (CX). Once handled, physiologic measurements were taken every 5 min for 20 min. There were few physiologic differences between the groups, although TX gemsboks were more hypertensive. Quality of induction, quality of recovery, depth of anesthesia, and muscle relaxation were subjectively assessed. Although TX gemsboks showed a trend for more-rapid induction and recovery postantagonist administration, this difference was not statistically significant. Similarly, there was a trend for TX animals to have smoother inductions and improved immobilization quality, but only the quality of induction was significantly different. These trends suggest that TX is preferable to CX when anesthetizing adult gemsboks.

Two White's tree frogs (Litoria caerulea) housed at a zoological park died after a short period of lethargy, weight loss, and edema. Detailed postmortem examinations were performed on both frogs, including bacterial cultures and complete histologic examinations. Intracytoplasmatic as well as free protozoan parasites were identified in multiple organs from both frogs. The parasites were identified within erythrocytes, leukocytes, endothelial cells, and hepatocytes. Immunohistochemistry demonstrated a cross-reaction with Toxoplasma gondii antisera. Parasite ultrastructural analysis was performed by transmission electron microscopy. The parasites demonstrated an apical complex containing a conoid, rhoptries, and micronemes, demonstrating it was a member of the phylum Apicomplexa. In addition, the parasites had bipolar paranuclear bodies, organelles that are typical of coccidian sporozoites. The organisms were tentatively identified as members of the genus Lankesterella on the basis of histologic and ultrastructural morphology. A portion of the 18s ribosomal RNA (rRNA) gene was amplified via a polymerase chain reaction, sequenced, and used in a Basic Local Alignment Search Tool search of the GenBank database. The 18s rRNA gene sequence was found to be most similar to gene sequences isolated from Lankesterella organisms (88%). In aggregate, these data support the classification of these protozoa as a novel species of Lankesterella. A causal relationship between frog morbidity and protozoal parasitism was not determined. This is the first report of Lankesterella sp. in White's tree frogs.

Surgical procedures of the wing are commonly performed in companion, captive, and wild avian species. To develop a clinically applicable brachial plexus nerve block technique for perioperative analgesia in birds, 8 adult female mallard ducks (Anas platyrhynchos) were anesthetized and used in several local anesthetic trials with bupivacaine (2 or 8 mg/kg) or a combination of lidocaine (15 mg/kg) and epinephrine (3.8 microg/kg) perineurally; equal volumes of saline were administered as control treatments. Both axillary and dorsal approaches to the brachial plexus were evaluated. With the axillary approach, radial and ulnar compound nerve action potentials (CNAP), sensory nerve conduction velocities (SNCVs), and cord dorsum potentials (CDPs) were recorded after distal sensory nerve stimulation. Values were recorded before and at 5, 30, and 60 minutes after injection of local anesthetic or saline. Birds were monitored for the presence of a wing droop and a change in motor function on recovery from anesthesia. Results were highly variable for all techniques. No technique significantly decreased CDPs or resulted in consistent wing droop. Radial and ulnar CNAPs, SNCVs, and CDPs were consistently recorded in all birds. Variable results might indicate that the treatment, concentration, or volume of local anesthetic used was ineffective in producing local anesthesia. Electrodiagnostic methods used in these ducks to assess loss of sensory nerve conduction might not be sensitive enough to assess the effects of local anesthesia. Further research is needed to identify methods for assessing the efficacy of brachial plexus nerve blockade in birds.

This study was conducted to evaluate whether the administration of naloxone (NAL) had an impact on the speed or quality of recovery in servals anesthetized with ketamine, butorphanol, and medetomidine. Twenty-two clinically healthy, captive servals (Leptailurus serval) were anesthetized for routine physical examinations using ketamine (3 mg/kg), medetomidine (0.03 mg/kg), and butorphanol (0.3 mg/kg). Eleven animals were administered atipamezole (5 mg/mg medetomidine) and NAL (0.1 mg/mg butorphanol), while the other 11 were administered atipamezole (5 mg/mg medetomidine) and sterile saline (SAL). There was no statistically significant difference in recovery time between the NAL and the SAL group; similarly, there were no significant differences between groups at any time point for the measures of quality of recovery used in this study. This investigation was unable to detect a clinically significant difference in anesthetic recovery for servals that received NAL relative to those that did not. These results suggest that butorphanol does not routinely need to be antagonized in captive servals that are anesthetized with these dosages of ketamine, medetomidine, and butorphanol. It may be more important to administer opioid antagonists in free-ranging animals, where residual sedation would be more detrimental. In these cases, a long-acting antagonist, such as naltrexone, may be indicated. An opioid antagonist, such as NAL or naltrexone, should always be available for emergency administration whenever opioids are used. Additionally, this study provided physiologic data for servals anesthetized with a ketamine-medetomidine-butorphanol combination.

PROCEDURES: Anesthesia was induced and maintained with isoflurane in oxygen. In the first bird that was anesthetized, end-tidal isoflurane concentration was maintained at 1.0% for 15 minutes. After this period of anesthetic equilibration, an end-tidal gas sample was obtained for verification of isoflurane concentration. A toe was pinched to determine the bird's response to pain, and the bird was then allowed to recover from aesthesia. To determine MAC, a so-called up-and-down approach was subsequently used in all 15 birds. Compared with the isoflurane concentration used for MAC determination in the first bird, maintenance isoflurane concentration for the second bird was increased by approximately 10% if the first bird reacted and decreased by approximately 10% if the first bird did not react to a toe pinch. These steps were then followed until all 15 birds had been anesthetized. Crossover events occurred when birds in sequence had discordant results (ie, 1 reactor and 1 nonreactor). The MAC was defined as the mean of the isoflurane concentrations measured during these crossover events. RESULTS: Mean MAC of isoflurane in thick-billed parrots was estimated to be 1.07% (95% confidence interval, 0.97% to 1.16%). CONCLUSIONS AND CLINICAL RELEVANCE: Isoflurane MAC appears to be lower in thick-billed parrots than the MAC determined for other bird species. Determination of the species-specific requirements of thick-billed parrots should allow isoflurane anesthesia to be performed more safely in this endangered species.

OBJECTIVE: To develop a clinically applicable technique for recording cord dorsum potentials (CDPs) following stimulation of the radial and ulnar nerves and establish reference values for radial and ulnar sensory nerve conduction velocities (SNCVs) in the wings of ducks. ANIMALS: 8 clinically normal adult female mallard ducks (Anas platyrhynchos). PROCEDURES: Radial and ulnar compound nerve action potentials (CNAPs) and CDPs were recorded following distal sensory nerve stimulation. The CDPs were recorded from the interarcuate space between the last cervical vertebra and the first thoracic vertebra. Surgical dissection and transection of the brachial plexus in 1 anesthetized duck were performed to identify nerve root location and confirm functional loss of nerve conduction assessed by loss of the CDP. RESULTS: Radial and ulnar CNAPs and CDPs were consistently recorded in all birds. Median radial SNCV was 38.3 m/s (range, 36.0 to 49.0 m/s), and ulnar SNCV was 35.3 m/s (range, 28.0 to 40.0 m/s). Surgical transection of the brachial plexus resulted in complete loss of the CDP. CONCLUSIONS AND CLINICAL RELEVANCE: Measurement of radial and ulnar SNCV or CDP is feasible in isoflurane-anesthetized mallard ducks. The CDP accurately reflects sensory nerve conduction through the brachial plexus. Assessment of brachial plexus function in mallard ducks via evaluations of SNCVs and CDPs may have application for diagnosis of traumatic injuries to the brachial plexus, evaluation of neuropathies associated with exposure to toxic chemicals, and assessment of the efficacy of interventions such as brachial plexus nerve blockade.

An adult male black-necked swan (Cygnus melanocoryphus) was evaluated for a routine physical examination and West Nile virus vaccination. Nine days later, the swan was hospitalized for weakness, dehydration, tremors, and leukocytosis. Gradual clinical improvement was seen over a 2-wk course of supportive care. However, the animal became acutely weak and was found dead. Histopathology revealed severe granulomatous pneumonia and air sacculitis with dissemination to the liver, spleen, intestine, and coelom. Mycobacterial infection was confirmed by identification of acid-fast bacilli within tissue sections and by polymerase chain reaction. Concurrent West Nile virus infection was identified in sections of brain by using immunohistochemistry.

Physical examination of an asymptomatic 20-yr-old intact female spotted hyena (Crocuta crocuta) revealed a midabdominal mass. A complete blood count (CBC) revealed peripheral lymphocytosis. Abdominal ultrasonography and laparoscopy confirmed severe splenomegaly. Cytologic examination of a bone-marrow core and histologic examination of spleen and liver biopsy samples revealed neoplastic small lymphocytes. Immunohistochemical staining of liver and spleen samples with the use of leukocyte-specific monoclonal antibodies showed that the neoplastic lymphocytes were immunoreactive to T-lymphocyte CD3 receptor and immunonegative to B-lymphocyte CD79a receptor. The morphology and distribution of neoplastic T-lymphocytes within the spleen, liver, peripheral blood, and bone marrow was most consistent with chronic T-lymphocytic leukemia. Treatment with chlorambucil and prednisone effectively decreased the lymphocyte count, but was associated with thrombocytopenia, which resolved after chlorambucil treatment was temporarily discontinued. Chemotherapy was resumed with a single dose of L-asparaginase, followed by a lower dosage of chlorambucil and continued prednisone. Two years after initial diagnosis, the hyena developed a hemoabdomen and was euthanized. Neoplastic T-lymphocytes were present in spleen, liver, visceral and peripheral lymph nodes, lungs, heart, kidney, adrenal glands, mesentery, intestines, pancreas, brain, and bone marrow.

Diagnosis of avian mycobacteriosis, caused by Mycobacterium genavense or species belonging to the Mycobacterium avium complex (MAC), is problematic. Polymerase chain reaction (PCR) offers rapid and sensitive detection of minute quantities of DNA, and conventional protocols have been used for evaluating avian specimens. The recent development of real-time PCR offers several advantages over conventional PCR. In attempts to improve diagnosing avian mycobacteriosis, a real-time TaqMan PCR assay was developed targeting the 65-kD heat shock protein gene of M. genavense and MAC spp. Nineteen reference isolates, 16 clinical isolates, and 32 avian tissue samples were used to evaluate the assay. When sufficient amplicons were produced, the species of mycobacteria was determined by standard sequencing of TaqMan PCR products and compared with results from commercial mycobacteriology laboratories and/or standard sequencing of conventional PCR products. The TaqMan PCR detected DNA from reference isolates of M. genavense, MAC spp., and Mycobacterium tuberculosis complex spp. Of the clinical isolates, the TaqMan PCR detected DNA from 10 of 12 Mycobacterium avium avium isolates and two of three Mycobacterium avium intracellulare isolates. For the tissue samples, the TaqMan PCR amplified DNA in six of nine samples that were identified by sequencing of conventional PCR products and/or by commercial mycobacteriology laboratories as being MAC spp. positive and three of four samples that were positive for M. genavense. There was some disagreement between speciation results from the TaqMan PCR and those from commercial mycobacteriology laboratories or conventional PCR or both. This disagreement was suspected to be because of relatively small numbers of base pairs in the TaqMan PCR products. The TaqMan PCR may provide a useful tool for evaluating clinical samples for DNA from mycobacteria species that most commonly infect birds; however, further refinement is needed in order to improve sensitivity and provide more accurate speciation.

In 1996, lymphosarcoma was observed in a captive adult female white-tailed deer (Odocoileus virginianus) from northeastern Kansas (USA). A subcutaneous mass on the deer's left cheek was surgically removed and lymphosarcoma was diagnosed. The mass recurred within 3 wk. A second surgical removal was attempted but the tumor had grown much larger, had become intimately involved with the buccal mucosa, and was beginning to interfere with mastication. For these reasons, the deer was euthanized. At postmortem examination the only abnormal findings were the primary tumor and enlarged ipsilateral parotid and mandibular lymph nodes. Histologically these tissues demonstrated changes characteristic of lymphosarcoma but no other organs had evidence of neoplastic disease. A diagnosis of focal lymphosarcoma with local metastasis was made. The organ distribution of lymphosarcoma in this deer differs from previously described cases of lymphosarcoma in cervids.

Avian vacuolar myelinopathy (AVM) is a neurologic disease that has been diagnosed in free-ranging birds in the southeastern United States. Bald eagles (Haliaeetus leuocephalus), American coots (Fulica americana), and mallards (Anas platyrhynchos) have been affected. Previous investigations have not determined the etiology of this disease. In November and December 2002, we attempted to induce AVM in game-farmed mallards through four, 7-day exposure trials. Mallards were housed in six groups of eight, with two of these groups serving as controls. One group was housed with AVM-affected coots; one group was tube fed daily with water from the lake where affected coots were captured; one group was tube fed daily with aquatic vegetation (Hydrilla verticillata) from the same lake; and another group was tube fed daily with sediment from the lake. No ducks exhibited clinical neurologic abnormalities consistent with AVM and no evidence of AVM was present at histopathologic examination of brain tissue. Although limitations in sample size, quantity of individual doses, frequency of dose administration, duration of exposure, and timing of these trials restrict the interpretation of the findings, AVM was not readily transmitted by direct contact, water, hydrilla, or sediment in this investigation.
