Play behavior is a promising welfare indicator in dairy calves because it decreases in negative situations such as pain or hunger and increases in positive contexts such as in appropriate social environments. Directly measuring play is time consuming because it is performed in irregular bouts and can be inconsistent over days. To facilitate automatic recording of play, previous studies fitted triaxial accelerometers to the hind legs of calves and measured the velocity of movements in large arenas; high correlations were reported between vertical axis peak duration and the duration of locomotor play. The current study aimed to validate accelerometers for recording spontaneous locomotor play in calves' home pens over longer periods. Data were collected from 48 Holstein Friesian calves, housed in groups of 3 in pens of 10 m2, at either 4 or 8 wk of age. Acceleration at the vertical axis of the hind leg was recorded at a rate of 1 Hz. One active time period for each calf was randomly selected (mean duration ± standard deviation = 34 ± 9 min). From video of the corresponding time period, the frequency of locomotor play events, consisting of run, turn, and buck/buck-kick, was recorded using behavior sampling. Combined counts of play events were highly correlated (Pearson r = 0.91) with counts of acceleration peaks. However, for calves with higher levels of locomotor play, this method underestimated the extent of play. Alternatively, run, turn, and buck events obtained from video were transformed by creating intervals of 10 s and then classifying each 10-s interval as comprising events of play ("play") or not comprising events of play ("no play"). The corresponding accelerometer data for all 10-s periods, equaling 10 consecutive readings each, were classified into play or no play by using quadratic discriminant analysis; 79% of periods with locomotor play were correctly classified. Counts of observed play intervals correlated with the counts of play periods from accelerometers (r = 0.87), but the discriminant analysis consistently overestimated play. In conclusion, accelerometer measurements at 1 Hz (in 1-s intervals) and at the vertical axis cannot be used alone to exactly quantify absolute levels of locomotor play in the home pen. However, counts of peak accelerations can provide a rough estimate of inter-individual differences in play events, and discriminant analysis can be used as a proxy for one-zero sampling of inter-individual differences in locomotor play.

Play behaviour in pre-weaned piglets has previously been shown to vary consistently between litters. This study aimed to determine if these pre-weaning litter differences in play behaviour were also consistent in the post-weaning period. Seven litters of commercially bred piglets were raised in a free farrowing system (PigSAFE) and weaned at 28 days post-farrowing (+/-2 days). Post-weaning piglets were maintained in litter groups in the PigSAFE pen. Analyses have been adjusted for sex both within and between litter as the only statistically significant covariate to play behaviour. Litter differences were observed in locomotor play in both the pre- and post-weaning stage (Pre: F(6,76) = 5.51 P < 0.001; Post: F(6,69) = 4.71, P < 0.001) and run (Pre: F(6,76) = 4.96, P < 0.001; Post: F(6,69) = 4.58, P < 0.001; the major element of locomotor play). Twenty eight% of the variance for a single observed animal in pre-weaning locomotor play and 26% of variance post-weaning could be attributed to the litter. There was no statistical evidence of differences in social play between litters at either stage with only 8% of pre-weaning variance, and 1% of post-weaning variance being attributable to the litter level. However non-harmful fighting (the major element of social play), showed strong evidence of litter differences in both periods (Pre: F(6,76) = 2.38, P = 0.037; Post: F(6,69) = 2.60, P = 0.025), and was the only aspect of the play behaviour to correlate between the pre- and post-weaning periods (r = 0.765, df = 5, P = 0.045). On average play increased post-weaning. Litters showed a 'litter weaning effect' by differing in their locomotor play behavioural response to weaning, measured as the change in locomotor play behaviour from pre- to post-weaning (F(6,70) = 5.95, P < 0.001). These results generally confirm previous work showing litter differences in aspects of play behaviour in both the pre and post-weaning period. However, there was no consistency in litter differences between pre- and post-weaning periods in the categories of play behaviour with the exception of non-harmful fighting. We demonstrated a 'litter weaning effect' where litters respond as a 'unit' to weaning in terms of their locomotory play behaviour. In general these results add further support to the use of play as a sensitive welfare indicator in neonatal pigs.

The selection of farm animals through breeding for human benefit has a very long history. In more recent times the practice of animal breeding has become highly sophisticated and the speed of change in 'production traits' such as rate of growth and milk yield has correspondingly increased dramatically. This narrow focus on production traits led to a number of well-documented examples of 'unfavourable' correlated responses such as negative fertility and health issues in high-yielding dairy cattle, with concerns that animal breeding is inherently antagonistic to animal welfare. In this paper the authors explore some of the questions surrounding breeding and welfare and, specifically, how to conceptualise and improve the 'fit' between the selected animal and the environment, or system, in which the animal is reared and managed. The authors conclude that there is a need for a better understanding of genotype x environment effects on health and welfare traits in order to inform the development of breeding programmes that lead to improved environmental fit in animals. They also see the need for the development of valid traits for assessing health and welfare, greater consideration of early life effects that can also potentially affect environmental fit and a need to consider the impacts of climate change on breeding programmes.

The effect of proximity to a dominant cow on a low-ranking cow's willingness to feed was assessed using choice tests. The main aim of the experiment was to determine the feeding space allowance at which the majority of subordinate cows would choose to feed on high-palatability food (HPF) next to a dominant cow rather than feeding alone on low-palatability food (LPF). Thirty Holstein-Friesian cows were used in the study. Half of the cows were trained to make an association between a black bin and HPF and a white bin and LPF, and the other half were trained with the opposite combination. Observations of pair-wise aggressive interactions were observed during feeding to determine the relative social status of each cow. From this, dominant and subordinate cows were allocated to experimental pairs. When cows had achieved an HPF preference with an 80% success rate in training, they were presented with choices using a Y-maze test apparatus, in which cows were offered choices between feeding on HPF with a dominant cow and feeding on LPF alone. Four different space allowances were tested at the HPF feeder: 0.3, 0.45, 0.6, and 0.75 m. At the 2 smaller space allowances, cows preferred to feed alone (choices between feeding alone or not for 0.3- and 0.45-m tests were significantly different). For the 2 larger space allowances, cows had no significant preferences (number of choices for feeding alone or with a dominant). Given that low-status cows are willing to sacrifice food quality to avoid close contact with a dominant animal, we suggest that the feeding space allowance should be at least 0.6m per cow whenever possible. However, even when space allowances are large, it is clear that some subordinate cows will still prefer to avoid proximity to dominant individuals.

The behavior of groups of housed, lactating dairy cattle was observed over 2 winter housing periods on 20 organic farms and 20 conventional farms in the United Kingdom. Three methods were used: (1) 6 video-clips of 10 min duration were captured of cows feeding at sections of the feed-bunk face during the peak feeding period (0 to 90 min) and continuously observed for aggressive interactions among cows; (2) the proportion of cows at the feed-bunk face was scanned every 15 min for 4.5h to include the peak feeding period (0 to 255 min); and (3) all nonfeeding behaviors were scanned every 15 min for 2.5h after the peak feeding period (120 to 255 min). The latter scans were analyzed post hoc for measures of cow comfort (freestall farms only). Management and health data were collected on each farm. On farms with open-fronted feed-bunk faces, a greater number of aggressive interactions occurred at the feed-bunk face at peak feeding time on organic farms than on conventional farms (organic = 36.3 ± 4.4; conventional = 29.1 ± 3.0). Higher proportions of cows were at the feed-bunk face at peak feeding on organic farms than on conventional farms (organic=0.58 ± 0.04; conventional=0.48 ± 0.03). Housing type (freestall versus straw pen) explained most differences in postfeeding behavior (proportion of ruminating cow in alleyways: freestalls=0.16±0.06 vs. straw-pen=0.08 ± 0.03), with few differences between organic and conventional herds. On freestall farms, the proportions of cows on organic farms lying down postfeeding was smaller than in conventional herds (organic=0.38 ± 0.09 vs. conventional=0.43 ± 0.07). Differences in behavior around peak feeding time could be associated with the reduction in food "quality" on organic farms compared with the energy requirement of the cows, with cows on organic farms being highly motivated to feed. A correlation was observed between farms that had high amounts of lying and farms that had high lameness prevalence (R(2)=55.3), suggesting a complex relationship between comfort and pain. Overall, the behavior of dairy cows on organic farms was not different from that of conventionally reared cows, and the results suggest that most behavioral welfare problems relating to housing could be alleviated by management practices.

Adolescents' interactions with animals are of increasing interest and their beneficial developmental outcomes are well known. However, negative interactions such as perpetrating cruelty toward animals during childhood and adolescence have been related with child abuse, domestic violence, and later interpersonal violence. Cruelty toward animals by adolescents has been reported predominately in criminal and clinical samples, and links have been made between animal cruelty and interpersonal violence. However, studies often lack a clear definition of animal cruelty and the animal involved. The present study addresses methodological shortcomings by providing a clear definition of the cruelty acts and the animals involved and the time frame within which cruelty acts have been taken place. Therefore, we investigated the prevalence of animal cruelty of 979 British adolescents (419 male, 497 female, Mage = 15.1 ± 1.57 years) by means of a survey questionnaire administered in school. Animal cruelty was investigated encompassing deliberate and nondeliberate cruelty acts, a clear definition of the target animals was included and a time frame was provided. Furthermore, acceptability of animal cruelty, engaging in antisocial behavior, and family affluence were investigated. Results show high reliabilities for the measures applied. Exploratory factor analysis reveals different types of animal cruelty. Gender differences were observed for deliberate and accidental cruelty acts, with boys reporting higher levels than girls. Younger adolescents reported higher accidental cruelty acts than older ones. Acceptance of animal cruelty played a significant role in predicting animal cruelty, together with antisocial behaviors and place of living. The present study shows for the first time the importance of distinguishing between different types of animal cruelty and defining the animals involved.

In this study, we aimed to validate existing plasma assays to measure biomarkers for maternal signalling in milk and saliva of lactating sows. These biological samples are minimally invasive to the animal and could give a physiological profile of maternal qualities available to their piglets. Sows were farrowed in a zero-confinement system, and their colostrum and milk samples were manually collected during naturally occurring let-downs (i.e. not induced) over the lactation period. Saliva sampling involved sows voluntarily accepting cotton buds to chew without restraint. Commercial kits designed for blood plasma were tested, and any modifications and results are given. We successfully measured total protein, cortisol, tumour necrosis factor-α (TNF-α) and oxytocin in pig milk and saliva and immunoglobulin G (IgG) in pig milk samples. We were unsuccessful at measuring relaxin and serotonin in these biological samples. We observed higher levels of biomarkers in milk than in saliva. The measurement of TNF-α in pig milk for the first time revealed increased levels with larger litters. This development will allow more detailed understanding of biomarkers in milk. There was also evidence that the minimally invasive technique of using saliva sampling did not interrupt natural oxytocin production around parturition.

The numbers of organic dairy farms are increasing in the United Kingdom and in other parts of the world. On organic farms, the use of veterinary medicines is restricted. Because of this, there is concern that cow health is poorer on these farms. As udder health is primarily maintained by the use of antimicrobials, the effect of organic status on mastitis and somatic cell counts (SCC) is important to investigate. The aim of this study was therefore to determine whether the organic status and other management factors affect SCC. A group of 80 dairy farms was used in the study: 40 organic farms and 40 nonorganic farms. The farms were recruited in pairs, and each organic:nonorganic pair was matched for herd size, housing type, genetic merit for milk production and geographical location. Somatic cell count data were extracted from national databases for a standard year (2004), and analyzed using stepwise logistic regression models. The organic status of the farm did not appear in the final model, indicating no major influence of organic status on SCC. There were, however, several effects of management on SCC. Somatic cell counts were lower on farms where the udders were not cleaned or cleaned only when dirty. Somatic cell counts were also lower on farms that kept cows in larger management groups and where the majority, but not all cases of mastitis are treated with antimicrobials. It can be concluded that the control measures used on the organic farms in this study are at least as effective as those used on nonorganic farms in controlling SCC. Other management factors are influential and attention to these factors will allow farmers to reduce SCC.

AIM: Background: High blood pressure is an independent risk factor for cerebrovascular, renal and cardiovascular disease. According to World Health Organization treatment to target Blood Pressure (BP) of <140/90 mmHg has been associated with decrease in morbidity and mortality. Despite this BP control has been very poor even in developed economies. OBJECTIVE: We set to assess level of BP control among hypertensive patients on treatment in Dutse, Jigawa state, and to identify treatable causes of failure to achieve target for better management. METHODS: It is a cross sectional study of all hypertensives for more than one year attending medical out patients clinic who have consented. An interviewer administered questionnaire was used to obtain information from the patients. RESULTS: A total of 123 patients of which 45% were females with mean age, duration of hypertension of 51.9 and 5.9 years respectively and BMI of 40.9Kg/ m2. Eighty-three per cent, 91% and 94% were aware of salt restriction, cessation of smoking and alcohol moderation as lifestyle modifications respectively. The mean Systolic and Diastolic BP were 142mmHg and 86mmHg respectively. Fifty-two per cent were on two drugs combination including a diuretic while 4.87% were on three drugs or more. Less than a third (27.6%) had their BP controlled at <140/90mmHg. There was no significant difference in the demographic and clinical data between patients with controlled and uncontrolled BP. CONCLUSION: This study found that control of BP is still poor in our setting. This could be due to physician inertia in the treatment, use of inappropriate combination of anti hypertensives or failure to reinforce lifestyle modifications.

The presence of hock injury was assessed in the milking herds of 80 dairy farms (40 organic, 40 nonorganic) across the United Kingdom. A wide range of information on farm management and husbandry was gathered via interview to assess the factors contributing to hock damage for all 80 farms, and a comprehensive building appraisal was conducted for 40 farms visited during the winter housing period. The prevalence of hock lesions was lower on organic compared with nonorganic farms (37.2 vs. 49.1%). Prevalence of hock damage was greater in the spring than fall (59.9 vs. 21.6%) and cows housed in free-stalls had a greater prevalence of hock lesions than those housed on straw (46.0 vs. 25.0%). Prevalence of hock damage increased with lactation number. In the analysis of fall/spring data, the age first mated, herd biosecurity, duration of summer grazing, and cow milk yield were significant factors relating to herd hock damage. Larger herds had a greater proportion of cows with hock swellings. Farms with a shorter calving interval had more cows with hock swellings. Factors relating to housing conditions that were positively associated with the prevalence of hock damage were low feed face space per cow, inferior passageway cleanliness, low total standing area per cow, and the type of bedding added to the free-stall. To assess whether free-stall versus straw-pen or organic versus nonorganic farms had different sets of risk factors, the data were reanalyzed for these types separately. For the straw-pen farms a high culling rate was associated with more hock damage. Within nonorganic farms, the length of summer grazing was significant, with longer periods meaning less hock injury. The prevalence of hock injuries on many UK farms, both organic and non-organic, exceeded levels that are deemed acceptable for cow comfort. Efforts are needed to improve housing standards to reduce the prevalence of hock injury and consequently improve cow welfare.

Survival is reduced in low birth weight piglets, which display poor thermoregulatory abilities and are slow to acquire colostrum. Our aim was to identify additional behavioural and physiological indicators of piglet survival incorporating traits reflective of both the intrauterine and extrauterine environment. Data were collected from 135 piglets from 10 Large White x Landrace sows to investigate which physiological measurements (e.g. individual placental traits), and which behavioural measurements (e.g. the quantification of piglet vigour), were the best indicators of piglet survival. Generalised linear models confirmed piglet birth weight as a critical survival factor. However, with respect to stillborn mortality, piglet shape and size, as measured by ponderal index (birth weight/(crown-rump length)(3)), body mass index (birth weight/(crown-rump length)(2)), respectively, and farrowing birth order were better indicators. With respect to live-born mortality, postnatal survival factors identified as crucial were birth weight, vigour independent of birth weight, and the latency to first suckle. These results highlight the importance of the intrauterine environment for postnatal physiological and behavioural adaptation and identify additional factors influencing piglet neonatal survival.

Environmental enrichment (EE) is widely used to study the effects of external factors on brain development, function and health in rodent models, but very little is known of the effects of EE on the brain in a large animal model such as the pig. Twenty-four young pigs (aged 5 weeks at start of study, 1:1 male: female ratio) were housed in environmentally enriched (EE) pens and provided with additional enrichment stimulation (a bag filled with straw) once daily. Litter, weight and sex matched controls n= (24) were housed in barren (B) conditions. Behaviour was recorded on alternate days from study day 10. After 21 days, RNA-sequencing of the frontal cortex of male piglets culled one hour after the enrichment stimulation, but not those at 4 h after stimulation, showed upregulation of genes involved in neuronal activity and synaptic plasticity in the EE compared to the B condition. This result is mirrored in the behavioural response to the stimulation which showed a peak in activity around the 1 h time-point. By contrast, EE piglets displayed a signature consistent with a relative decrease in microglial activity compared to those in the B condition. These results confirm those from rodents, suggesting that EE may also confer neuronal health benefits in large mammal models, through a potential relative reduction in neuroinflammatory process and increase in neuroprotection driven by an enrichment-induced increase in behavioural activity.

Dairy farms vary a great deal in the feeding and management systems that are used. These differences affect the performance of the cows, and some genotypes may be affected more than others. If effects of such genotype-by-environment interactions (GxE) are large, then farmers must be made aware of them to make informed breeding decisions. To investigate GxE, a classification system for farm environments was developed based on national- and fine-level data from dairy herds across the United Kingdom. The national data included herd and yield characteristics and local weather information. The fine-level data included information on feeding and management systems on farms, and was obtained from survey results from 778 farms. A principal components analysis of the surveys identified 2 major dimensions characterizing the data. The first dimension explained 14.6% of the variation and was related to the level of production intensity. The second dimension explained 11.5% of the variation and was related to climate. Information on milk yield, herd characteristics, and climate was then extracted from national databases for the survey farms. A canonical correlation analysis was used to relate the survey data to the variables extracted from the national data set to determine the most relevant variables. The canonical correlation between the chosen sets of national data and survey variables was 0.62. This environmental classification was then used to determine how the farm environment affects the life span of dairy cows. The life span of the daughters of 1,000 sires was related to the type of farm environment. The daughters of a majority of sires showed a "plastic" response, with increased life span in less intensive farms. The daughters of a smaller number of sires showed a more generalized response, with life span being less affected by the environment. This GxE suggested that sires vary in the sensitivity of their daughters to different farm environments. This variation in response could allow breeding companies and farmers to match sires to particular farm environments.

The aim of this study was to assess the effect of grazing (G) vs. zero-grazing (ZG), level of milk production, and quality and type of housing system [free stalls (FS) and straw yards (SY)] on the prevalence of lameness and leg injuries in dairy cows. Observations were made on 37 commercial dairy farms across Great Britain. A single visit of 5 d duration was made to each farm. During this visit, lameness scores and the incidence of swellings, rubs, and injuries to hocks and knees were recorded on all the peak- or mid-lactation cows. Aspects of the quality of housing and management that were likely to affect foot and leg health were recorded. There were more lame cows on ZG farms (39 +/- 0.02%) than on grazing (G) farms (15 +/- 0.01%), and lameness scores were higher on FS farms compared with SY farms (0.25 +/- 0.01 vs. 0.05 +/- 0.01). Cows on SY farms had fewer hock and knee injuries compared with FS farms. The frequency of knee swellings was higher on ZG farms (0.31 +/- 0.02) than on G farms (0.15 +/- 0.01). Aspects of the free-stall design affected foot and leg health. The number of hock swellings increased with increasing stall gradient (0.16 +/- 0.01 with no slope vs. 0.39 +/- 0.02 at a 0 to 1.5% slope). There was an interaction between the length of the free-stall lunging space and the hip width of the cow, indicating that the incidence of lameness is generally highest on farms with small free stalls and heavy cows. High levels of milk production did not affect lameness or leg injury. The results indicate that housing cows throughout the year potentially has a detrimental effect on foot and leg health. However, good free-stall design may reduce lameness and leg lesions.

To limit tail biting incidence, most pig producers in Europe tail dock their piglets. This is despite EU Council Directive 2008/120/EC banning routine tail docking and allowing it only as a last resort. The paper aims to understand what it takes to fulfil the intentions of the Directive by examining economic results of four management and housing scenarios, and by discussing their consequences for animal welfare in the light of legal and ethical considerations. The four scenarios compared are: 'Standard Docked', a conventional housing scenario with tail docking meeting the recommendations for Danish production (0.7 m2/pig); 'Standard Undocked', which is the same as 'Standard Docked' but with no tail docking, 'Efficient Undocked' and 'Enhanced Undocked', which have increased solid floor area (0.9 and 1.0 m2/pig, respectively) provision of loose manipulable materials (100 and 200 g/straw per pig per day) and no tail docking. A decision tree model based on data from Danish and Finnish pig production suggests that Standard Docked provides the highest economic gross margin with the least tail biting. Given our assumptions, Enhanced Undocked is the least economic, although Efficient Undocked is better economically and both result in a lower incidence of tail biting than Standard Undocked but higher than Standard Docked. For a pig, being bitten is worse for welfare (repeated pain, risk of infections) than being docked, but to compare welfare consequences at a farm level means considering the number of affected pigs. Because of the high levels of biting in Standard Undocked, it has on average inferior welfare to Standard Docked, whereas the comparison of Standard Docked and Enhanced (or Efficient) Undocked is more difficult. In Enhanced (or Efficient) Undocked, more pigs than in Standard Docked suffer from being tail bitten, whereas all the pigs avoid the acute pain of docking endured by the pigs in Standard Docked. We illustrate and discuss this ethical balance using numbers derived from the above-mentioned data. We discuss our results in the light of the EU Directive and its adoption and enforcement by Member States. Widespread use of tail docking seems to be accepted, mainly because the alternative steps that producers are required to take before resorting to it are not specified in detail. By tail docking, producers are acting in their own best interests. We suggest that for the practice of tail docking to be terminated in a way that benefits animal welfare, changes in the way pigs are housed and managed may first be required.

Aggressive behaviour exhibited by domestic pigs following encounters with unfamiliar individuals is a serious welfare and economical problem. Aggression resulting in skin lesions is similarly prevalent in prepubertal pigs of either sex. Little is known about the neural circuits and neuropeptides that control aggression in the pig. Because there is evidence for the involvement of the vasopressin and serotonergic systems in the regulation of aggressive behaviour in male mammals, we sought differences using quantitative in situ hybridisation of vasopressin and serotonin 1A receptor (5-HT1A) mRNA expression within specific brain regions of aggressive and nonaggressive prepubertal female pigs. The number of cells expressing vasopressin mRNA was significantly higher in aggressive pigs in the medial amygdala, lateral septum (LS) and showed a similar trend in the bed nucleus of the stria terminalis (BnST) but not the paraventricular nucleus (PVN) or supraoptic nucleus. The 5-HT1A receptor was widely expressed through the porcine brain and a significantly lower intensity (silver grain density) of 5-HT1A mRNA expression was observed in the BnST. In the medial amygdala and LS fewer cells expressed 5-HT1A mRNA in aggressive pigs but no differences were found in the PVN. In the absence of inbred strains or selection lines, these findings have shown that prior identification of phenotypic behavioural extremes in a population in advance of neural studies is a useful technique. Moreover, these findings support a central role for vasopressin and serotonin in the mediation of high trait aggression in prepubertal female pigs.

The frequency of human intervention during lambing was assessed for 708 lambs and 162 ewes in up to four deliveries. In total, 21.8 per cent of the lambs born alive required some assistance at birth and 9.4 per cent were delivered manually. Assistance was required less frequently by twin lambs and ewe lambs, and Scottish blackface lambs required assistance less frequently than Suffolk lambs. Approximately one-third of the ewes were assisted in at least one delivery and 10 per cent were assisted in all their deliveries; however, less than 20 per cent of ewes were assisted more than once. A quarter of the lambs needed some human intervention to suck successfully, and Suffolk lambs needed help more frequently. The lambs born to almost half the first-parity ewes needed help to suck, but at later deliveries the lambs born to less than a third of the ewes needed assistance. In blackface flocks the survival of 100 lambs required 4.6 hours of human intervention whereas in Suffolk flocks 28 hours were required.

This study aimed to investigate if pregnancy-induced hypoalgesia occurs in the sow, and to examine the role of endogenous opioids which are known to be released in response to nociception. Sixteen Large White x Landrace multiparous sows were tested in straw bedded pens (2.5 x 2.5 m) during weeks 4, 8 and 12 of pregnancy and over the farrowing period. Testing involved thermal stimulation of eight areas on the rear-quarters of the sows with a CO2 infra-red laser until a physical response was seen (tail flick, leg move or muscle twitch) or for a maximum of 16 s. Over the farrowing period testing was more frequent, and at 3.75 h after the birth of the first piglet, half the sows received an injection (i.m.) of an opioid antagonist naloxone (N) (1 mg kg(-1) body weight) with the remainder receiving a control dose of saline (S). Responses were recorded 15 and 30 min post-injection. There was no significant difference between response times over weeks 4, 8 and 12 of pregnancy (P = 0.152), however a significant rise was seen from week 12 to 5 days before parturition (P = 0.002). Response times continued to rise until the birth of the first piglet by which time the majority of sows had stopped responding within 16 s (P < 0.001). Response times fell over days 1, 2 and 7 post-partum. After administration of naloxone response times fell compared to control animals at 15 min (P < 0.001) and 30 min (P < 0.01) post-injection. These results suggest that nociceptive threshold increases during late pregnancy in the sow, perhaps as an endogenous defence against labour pain, and that during parturition this change in nociceptive threshold is, at least in part, opioid-mediated. Oxytocin is known to be inhibited by endogenous opioids at parturition, thus future research should consider the potential role of increased nociception at birth as a negative feedback to oxytocin release.

Oxytocin plays an important role at parturition due to its involvement in uterine contractions, foetal expulsion and the onset of maternal behaviour. The role of the related neurohypophysial hormone, vasopressin, is less clear; however, there is some evidence that it is also involved in maternal behaviour and its role in osmotic regulation is well established. The aim of this study was to investigate the inhibitory effects of endogenous opioids on these hormones during the expulsive phase of parturition in the pig, and to examine how opioid restraint interacts with environmental restriction. The subjects of this study were 31 Large Whitex Landrace primiparous sows (gilts). An indwelling jugular catheter was implanted under general anaesthesia at 12 days before the expected parturition day (EPD). From 5 days before the EPD 15 of the gilts were individually housed in a restrictive parturition crate without straw and 16 were individually housed in a straw-bedded pen. Blood samples were taken with increasing frequency towards and during parturition through a catheter extension to reduce disturbance. At 7.5 min after the birth of the first piglet half of the gilts in each environment received a dose of the opioid receptor antagonist naloxone (1 mg/kg, i.v.) with the remaining gilts receiving saline as a control. Overall, there was no effect of environment on either circulating oxytocin or vasopressin. However, both oxytocin and vasopressin were inhibited by endogenous opioids during the expulsive phase. The inhibitory effects of opioids on these hormones did not appear to have any adverse effects on the progress of parturition as judged by cumulative piglet birth intervals. The regulation of the opioid inhibition of oxytocin and vasopressin during parturition is discussed in relation to other neurotransmitters and whether opioid inhibition of these neurohypophysial hormones is part of the 'normal' physiological response to parturition or whether it is stress-induced.

Nest-building behaviour occurs 6-24 h before parturition in pigs (gestation=116 days). Pseudopregnancy in pigs (induced with oestradiol valerate injections) lasts 50-80 days. We have shown that prostaglandin F2alpha (PG) administration on day 47 of pseudopregnancy induces nest-building and changes to plasma prolactin, oxytocin, cortisol and progesterone similar to those seen before normal parturition. Peripheral prolactin has been proposed as a modulator of nest-building. This study assessed nest-building behaviour in prolactin-deprived gilts. Jugular vein catheters were inserted on day 39 of pseudopregnancy and blood samples collected daily from days 40-48. Animals were injected im with either 40 mg bromocriptine in 2 ml 70% ethanol (n=8) or vehicle (n=7) at 17.00 h on day 46 and 09.00 h on day 47 of pseudopregnancy. PG (15 mg Lutalyse: Upjohn) was injected im at 11.00 h on day 47. Blood and behavioural samples were taken from 90 min before PG to 6 h post-PG. Plasma prolactin increased in control but not bromocriptine treated animals following PG (P<0.05). Elevations in oxytocin, cortisol and progesterone (P<0.05) above pre-PG concentrations were also seen, but of these only progesterone showed between group differences [greater (P<0.05) in control gilts on both days 47 and 48]. PG significantly (P<0.05) increased both the rate and proportion of total time spent performing straw/floor-directed behaviours not including foraging (an index of nesting behaviour) in both treatment groups with no significant differences between groups. There were also no significant differences between groups in time spent performing pen fixture directed activities before or after PG. Bromocriptine suppressed the rise in prolactin concentrations after PG without suppressing nest-building behaviour. We conclude that peripheral prolactin is not an essential component of the nest-building complex in pigs.
