RESUMEN

OBJECTIVES: Shellfish waste is a primary source for making N-acetyl-D-glucosamine. Thus, establishing a high-efficiency and low-cost bioconversion method to produce N-acetyl-D-glucosamine directly from shellfish waste was promising. RESULTS: A mutant C81 was obtained from Chitinolyticbacter meiyuanensis SYBC-H1 via 60Co-γ irradiation. This mutant C81 showed the highest chitinase activity of 9.8 U/mL that was 85% higher than the parent strain. The mutant C81 exhibted improved antioxidant activities, including total antioxidant capacity, superoxide radical ability, and hydroxyl radical scavenging ability, compared to that of the parent strain. Four out of nine organic solvents increased the chitinase activity by 1.9%, 6.8%, 11.7%, and 15.8%, corresponding to methylbenzene, n-heptane, petroleum ether, and n-hexane, respectively. The biphase system composed of aqueous and hexane presented a five-fold reduction of cell viability compared to the control. Using a continuous fermentation bioconversion process, 4.2 g/L GlcNAc was produced from crayfish shell powder with a yield of 80% of the chitin content. CONCLUSIONS: This study demonstrated that the mutant C81 is suitable for converting crayfish shell powder into GlcNAc in an aqueous-organic system.

RESUMEN

Two new milbemycin metabolites, 13α-hydroxymilbemycin ß13 (1) and 26-methyl-13α-hydroxymilbemycin ß13 (2), were isolated from the fermentation broth of a genetically engineered strain Streptomyces avermitilis AVE-H39. Their structures were determined by the comprehensive spectroscopic data, including 1 D, 2 D NMR, MS spectral analysis and the comparison with data from the literature. Compounds 1 and 2 not only exhibited potent acaricidal activities against Tetranychus cinnabarinus, but also had nematocidal activity against Bursaphelenchus xylophilus.

RESUMEN

Two new milbemycin derivatives, milbemycin M (1) and milbemycin N (2), were isolated from the culture of a genetically engineered strain Streptomyces bingchenggensis BCJ60. Their structures were elucidated through the interpretation of NMR and HR-ESI-MS spectroscopic data, as well as comparison with previous reports. The acaricidal and nematicidal activities of them against Tetranychus cinnabarinus and Bursaphelenchus xylophilus were tested. The results showed that compounds 1-2 possessed potent acaricidal and nematocidal activities.

Two new naphthalenone derivatives, 5-hydroxy-4-oxo-2-(2-oxopropyl)-1,2,3,4-tetrahydronaphthalen-1-yl acetate (1) and 5-hydroxy-2-(2-hydroxypropyl)naphthalene-1,4-dione (2), together with two new anthrone derivatives, (S)-2,5-dihydroxy-2-methyl-1,2,3,4-tetrahydroanthracene-9,10-dione (3) and 4,5-dihydroxy-2-methyl-9H-xanthen-9-one (4), were isolated from the fermentation broth of endophytic Micromonospora sp. NEAU-gq13. Their structures were determined by 1D-NMR, 2D-NMR, and HR-ESI-MS analysis. Compounds 2 and 3 exhibited strong cytotoxic activity against human central nervous system cancer (SF-268) with the IC50 values of 3.04 and 5.66 µg/ml, respectively. Moreover, compound 2 also displayed potent activity against human liver cancer (HepG2) with an IC50 value of 1.01 µg/ml.

Most of the currently mature methods that are used globally for population spatialization are researched on a single level, and are dependent on the spatial relationship between population and land covers (city, road, water area, etc.), resulting in difficulties in data acquisition and an inability to identify precise features on the different levels. This paper proposes a multi-level population spatialization method on the different administrative levels with the support of China's first national geoinformation survey, and then considers several approaches to verify the results of the multi-level method. This paper aims to establish a multi-level population spatialization method that is suitable for the administrative division of districts and streets. It is assumed that the same residential house has the same population density on the district level. Based on this assumption, the least squares regression model is used to obtain the optimized prediction model and accurate population space prediction results by dynamically segmenting and aggregating house categories.In addition, it is assumed that the distribution of the population is relatively regular in communities that are spatially close to each other, and that the population densities on the street level are similar, so the average population density is assessed by optimizing the community and surrounding residential houses on the street level. Finally, the scientificalness and rationality of the proposed method is proved by spatial autocorrelation analysis, overlay analysis, cross-validation analysis and accuracy assessment methods.

A novel Gram-stain-positive, motile, endospore-forming, rod-shaped bacterium, designated strain NEAU-3TGS17T, was isolated from the head of an ant (Lasius fuliginosus). The isolate grew at 0-35 °C (optimum 28-30 °C), at pH 6.0-11.0 (optimum pH 7.0-8.0) and with 0-6 % (w/v) NaCl (optimum 0 %). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain NEAU-3TGS17T belonged to the genus Psychrobacillus. Sequence similarities between strain NEAU-3TGS17T and members of the genus Psychrobacillus with validly published names (Psychrobacillus psychrotolerans DSM 11706T, Psychrobacillus insolitus DSM 5T, Psychrobacillus psychrodurans DSM 11713T and Psychrobacillus soli NBRC 110600T) were 98.4-99.1 %. DNA-DNA relatedness values between strain NEAU-3TGS17T and its closest relatives were below 70 %. The major cellular fatty acids (>5 %) were iso-C14 : 0, anteiso-C15 : 0, C16 : 0 and iso-C15 : 0. The major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The cell-wall peptidoglycan type was A4ß with ornithine as the diamino acid and the predominant menaquinones were MK-8 and some MK-7. The DNA G+C content was 35.8 mol%. On the basis of phenotypic data and phylogenetic inference, strain NEAU-3TGS17T was classified as representing a novel species in the genus Psychrobacillus, for which the name Psychrobacillus lasiicapitis sp. nov. is proposed. The type strain is NEAU-3TGS17T (=DSM 100484T=CGMCC 1.15308T).

RESUMEN

During an investigation exploring potential sources of novel species and natural products, a novel actinomycete with antifungal activity, designated strain NEAU-Gz11T, was isolated from a soil sample, which was collected from Gama, Chad. The isolate was found to have morphological and chemotaxonomic characteristics typical of members of the genus Streptomyces. 16S rRNA gene sequence similarity studies showed that strain NEAU-Gz11T belongs to the genus Streptomyces with high sequence similarity to Streptomyces hiroshimensis JCM 4098T (98.0 %). Similarities to other type strains of the genus Streptomyces were lower than 98.0 %. However, the physiological and biochemical characteristics and low levels of DNA-DNA relatedness could differentiate the isolate genotypically and phenotypically from S. hiroshimensis JCM 4098T. Therefore, the strain is concluded to represent a novel species of the genus Streptomyces, for which the name Streptomyces gamaensis sp. nov. is proposed. The type strain is NEAU-Gz11T (=CGMCC 4.7304T=DSM 101531T).

The Chinese Gaofen-3 (GF-3) mission was launched in August 2016, equipped with a full polarimetric synthetic aperture radar (SAR) sensor in the C-band, with a resolution of up to 1 m. The absolute positioning accuracy of GF-3 is of great importance, and in-orbit geometric calibration is a key technology for improving absolute positioning accuracy. Conventional geometric calibration is used to accurately calibrate the geometric calibration parameters of the image (internal delay and azimuth shifts) using high-precision ground control data, which are highly dependent on the control data of the calibration field, but it remains costly and labor-intensive to monitor changes in GF-3's geometric calibration parameters. Based on the positioning consistency constraint of the conjugate points, this study presents a geometric cross-calibration method for the rapid and accurate calibration of GF-3. The proposed method can accurately calibrate geometric calibration parameters without using corner reflectors and high-precision digital elevation models, thus improving absolute positioning accuracy of the GF-3 image. GF-3 images from multiple regions were collected to verify the absolute positioning accuracy after cross-calibration. The results show that this method can achieve a calibration accuracy as high as that achieved by the conventional field calibration method.

A new naphthalenepropanoic acid analog (1) was isolated from the broth of the actinomycetes Micromonospora sp. HS-HM-036. The structure of compound 1 was determined based on MS and extensive NMR analysis. A preliminary investigation of the biological activity of compound 1 was also described.

Two new macrocyclic lactones, 4,25-diethyl-4,25-demethyl-milbemycin ß3 (1) and 27-formaldehyde-milbemycin ß14 (2), were isolated from a genetically engineered strain Streptomyces bingchenggensis BCJ60. Their structures were determined on the basis of spectroscopic analysis, including 1D and 2D NMR techniques as well as ESI-MS and comparison with data from the literature. The acaricidal and nematocidal capacities of compounds 1 and 2 were evaluated against Tetranychus cinnabarinus and Bursaphelenchus xylophilus, respectively. The results showed that the two new macrocyclic lactones 1 and 2 possessed potent acaricidal and nematocidal activities.

RESUMEN

A new spectinabilin derivative (1) was isolated from the fermentation broth of the ant-derived Streptomyces sp. 1H-GS5, and the structure was elucidated by extensive spectroscopic analysis. Compound 1 showed cytotoxicity against human tumor cell lines A549, HCT-116, and HepG2 with IC50 values of 9.7, 12.8, and 9.1 µg/ml, respectively, which was relative higher than that of spectinabilin.

A novel actinomycete, designated strain 1C-HV12T, was isolated from the cuticle of Camponotus japonicas Mayr and characterized using a polyphasic approach. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that the organism should be assigned to the genus Promicromonospora and that it forms a monophyletic clade with the closest relatives Promicromonospora umidemergens JCM 17975T(99.17 % 16S rRNA gene sequence similarity), Promicromonospora vindobonensis V45T (98.88 %) and Promicromonospora iranensis HM 792T (98.85 %). Moreover, morphological and chemotaxonomic properties of strain 1C-HV12T also confirmed the affiliation of the isolate to the genus Promicromonospora. The peptidoglycan hydrolysates contained alanine, glutamic acid and lysine, and whole-cell sugars were galactose, glucose, rhamnose and ribose. The polar lipid profile consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside, an unidentified phosphoglycolipid, an unidentified phospholipid and an unidentified glycolipid. The major menaquinones were MK-9(H4) and MK-9(H2). The predominant cellular fatty acids were anteiso-C15 : 0, iso-C15 : 0 and anteiso-C17 : 0. The DNA G+C content was 71.6 mol%. However, the low level of DNA-DNA relatedness and some phenotypic characteristics allowed the isolate to be differentiated from closely related species. Therefore, it is concluded that strain 1C-HV12T represents a novel species of the genus Promicromonospora, for which the name Promicromonospora alba sp. nov. is proposed. The type strain is 1C-HV12T ( = CGMCC 4.7283T = DSM 100490T).

A novel actinobacterium, designated strain NEAU-ND5T, was isolated from the rhizosphere of corn (Zea mays L.) collected in Heilongjiang Province, north-east China, and characterized using a polyphasic approach. 16S rRNA gene sequence analysis showed that strain NEAU-ND5T was a member of the genus Nonomuraea, with highest sequence similarities to Nonomuraea jabiensis A4036T (98.29 %), Nonomuraea rosea GW12687T (98.25 %), Nonomuraea candida HMC10T (98.22 %), Nonomuraea rhizophila YIM 67092T (98.04 %) and Nonomuraea kuesteri NRRL B-24325T (98.04 %). Similarities to other type strains of the genus Nonomuraea were lower than 98 %. Morphological and chemotaxonomic properties of strain NEAU-ND5T were also consistent with the description of the genus Nonomuraea. The cell wall contained meso-diaminopimelic acid and the whole-cell sugars were glucose, ribose and madurose. The phospholipid profile consisted of diphosphatidylglycerol, phosphatidylmonomethylethanolamine, phosphatidylethanolamine, hydroxy-phosphatidylmonomethylethanolamine, hydroxy-phosphatidylethanolamine, phosphatidylinositol and phosphatidylinositol mannoside. The major menaquinones were MK-9(H4), MK-9(H2) and MK-9(H0). The predominant cellular fatty acids were iso-C16:0 and 10-methyl C17:0. A combination of DNA-DNA hybridization results and some phenotypic characteristics demonstrated that strain NEAU-ND5T was clearly distinguished from its closely related Nonomuraea species. Consequently, it is concluded that strain NEAU-ND5T represents a novel species of the genus Nonomuraea, for which the name Nonomuraea zeae sp. nov. is proposed. The type strain is NEAU-ND5T (=CGMCC 4.7280T=DSM 100528T).

Two novel actinobacteria, designated strains NEAU-Jh1-4T and NEAU-Wp2-0T, were isolated from muddy soil collected from a riverbank in Jiaohe and a dandelion root collected from Harbin, respectively. A polyphasic study was carried out to establish the taxonomic positions of these two strains. The phylogenetic analysis based on the 16S rRNA gene sequences of strains NEAU-Jh1-4T and NEAU-Wp2-0T indicated that strain NEAU-Jh1-4T clustered with Streptosporangium nanhuense NEAU-NH11T (99.32 % 16S rRNA gene sequence similarity), Streptosporangium purpuratum CY-15110T (98.30 %) and Streptosporangium yunnanense CY-11007T (97.95 %) and strain NEAU-Wp2-0T clustered with 'Streptosporangium sonchi ' NEAU-QS7 (99.39 %), 'Streptosporangium kronopolitis' NEAU-ML10 (99.26 %), 'Streptosporangium shengliense' NEAU-GH7 (98.85 %) and Streptosporangium longisporum DSM 43180T (98.69 %). Moreover, morphological and chemotaxonomic properties of the two isolates also confirmed their affiliation to the genus Streptosporangium. However, the low level of DNA-DNA hybridization and some phenotypic characteristics allowed the isolates to be differentiated from the most closely related species. Therefore, it is proposed that strains NEAU-Jh1-4T and NEAU-Wp2-0T represent two novel species of the genus Streptosporangium, for which the name Streptosporangium jiaoheense sp. nov. and Streptosporangium taraxaci sp. nov. are proposed. The type strains are NEAU-Jh1-4T (=CGMCC 4.7213T=JCM 30348T) and NEAU-Wp2-0T (=CGMCC 4.7217T=JCM 30349T), respectively.

A novel actinomycete, designated strain 3H-GS17T, was isolated from the head of an ant (Lasius fuliginosus L.) and characterized using a polyphasic approach. 16S rRNA gene sequence similarity studies showed that strain 3H-GS17T belongs to the genus Actinocorallia with high sequence similarity to Actinocorallia glomerata JCM 9376T (98.13 %) and Actinocorallia longicatena JCM 9377T (97.64 %). The chemotaxonomic properties of strain 3H-GS17T were also consistent with those of members of the genus Actinocorallia. The cell wall contained meso-diaminopimelic acid and whole-cell sugars were ribose, mannose, glucose, galactose and madurose. The predominant menaquinones were MK-9(H4), MK-9(H6) and MK-9(H8). The phospholipid profile consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol and phosphatidylinositol mannoside. The major fatty acids were C16 : 0 and C18 : 1ω7с. A combination of DNA-DNA hybridization experiments and phenotypic tests were carried out between strain 3H-GS17T and its closely related strains, which further clarified their relatedness and demonstrated that 3H-GS17T could be distinguished from these strains. Therefore, strain 3H-GS17T is concluded to represent a novel species of the genus Actinocorallia, for which the name Actinocorallia lasiicapitis sp. nov. is proposed. The type strain is 3H-GS17T (=DSM 100595T=CGMCC 4.7282T).

A novel actinomycete, designated strain NEAU-gxj3T, was isolated from soybean root [Glycine max (L.) Merr.] collected from Harbin, Heilongjiang Province, China, and characterized using a polyphasic approach. The 16S rRNA gene sequence of strain NEAU-gxj3T showed highest similarity to those of Micromonospora equina Y22T (98.2 %) and Plantactinospora endophytica YIM 68255T (98.0 %). Phylogenetic analysis based on the 16S rRNA gene and gyrB gene demonstrated that the isolate clustered with the members of the genus Plantactinospora. The chemotaxonomic properties of strain NEAU-gxj3Twere also consistent with those of members of the genus Plantactinospora. The cell wall contained meso-diaminopimelic acid and whole-cell sugars were xylose, glucose and galactose. The predominant menaquinones were MK-10(H6), MK-9(H8), MK-10(H2) and MK-10(H4). The polar lipid profile consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol and phosphatidylinositol mannoside. The major fatty acids were identified as anteiso-C17 : 0, iso-C16 : 0, iso-C15 : 0 and C15 : 0. A combination of DNA-DNA hybridization result and some phenotypic characteristics indicated that strain NEAU-gxj3Tcould be differentiated clearly from its closest phylogenetic relatives. Therefore, the strain is concluded to represent a novel species of the genus Plantactinospora, for which the name Plantactinospora soyae sp. nov. is proposed. The type strain is NEAU-gxj3T (=CGMCC 4.7221T=DSM 46832T).

RESUMEN

A novel strain of actinobacteria, designated strain NEAU-ZJC8T, was isolated from a saline-alkaline soil collected from Heilongjiang Province, north-east China and characterized using a polyphasic approach. Strain NEAU-ZJC8T exhibited morphological, cultural and chemotaxonomic features consistent with its classification as representing a member of the genus Streptomyces. Growth occurred at 16-35 °C, pH 5.0-11.0 and in the presence of 0-11 % (w/v) NaCl. The cell-wall peptidoglycan contained ll-diaminopimelic acid and glycine. Whole-cell hydrolysates mainly contained glucose, galactose and ribose. Predominant menaquinones were MK-9(H6), MK-9(H4) and MK-9(H8). The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositol mannoside and two unidentified phospholipids. The major cellular fatty acids (>10 %) were iso-C16 : 0, iso-C15 : 0, anteiso-C17 : 0 and C16 : 0. The G+C content of the DNA was 72.7 mol%. A phylogenetic tree based on 16S rRNA sequences showed that strain NEAU-ZJC8T formed a distinct clade within the genus Streptomyces and was closely related to Streptomyces haliclonae DSM 41970T (98.56 % similarity) and Streptomyces marinus NBRC 105047T (97.96 % similarity). A combination of DNA-DNA hybridization results and some phenotypic characteristics demonstrated that strain NEAU-ZJC8T could be distinguished from its closest phylogenetic relatives. Therefore, strain NEAU-ZJC8T represents a novel species of the genus Streptomyces, for which the name Streptomyces daqingensis sp. nov. is proposed. The type strain is NEAU-ZJC8T ( = CGMCC 4.7178T = JCM 30057T).

A novel actinobacterium, designated strain NEAU-FJL2(T), was isolated from a tomato root (Solanum lycopersicum L.) and characterised using a polyphasic approach. Morphological and chemotaxonomic properties of strain NEAU-FJL2(T) are consistent with the description of the genus Dactylosporangium. Finger-shaped sporangia were observed to form on short sporangiophores branching from the substrate hyphae. The cell wall peptidoglycan was found to contain meso- and 3-hydroxy-diaminopimelic acids; arabinose, mannose, rhamnose and xylose were found as whole-cell sugars. The phospholipid profile was found to consist of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol and phosphatidylinositol. The predominant menaquinones were identified as MK-9(H8) and MK-9(H6). The major fatty acids were identified as iso-C16:0, C16:1 ω7c and iso-C15:0. Phylogenetic analysis based on 16S rRNA gene sequences demonstrated that strain NEAU-FJL2(T) belongs to the genus Dactylosporangium, with Dactylosporangium sucinum JCM 19831(T) (99.3 % 16S rRNA gene sequence similarity), Dactylosporangium darangshiense JCM 17441(T) (99.2 %), Dactylosporangium fulvum JCM 5631(T) (98.9 %) and Dactylosporangium roseum JCM 3364(T) (98.8 %) as the nearest phylogenetic relatives. However, a combination of DNA-DNA hybridization results and some phenotypic characteristics demonstrated that strain NEAU-FJL2(T) can be distinguished from them. Consequently, it is proposed that strain NEAU-FJL2(T) represents a novel species of the genus Dactylosporangium, for which the name Dactylosporangium solaniradicis sp. nov. is proposed. The type strain is NEAU-FJL2(T) (=CGMCC 4.7302(T) = DSM 100814(T)).

A novel actinomycete, designated strain 2C-HV3(T), was isolated from the cuticle of Camponotus japonicus Mayr collected from Harbin, Heilongjiang province, north China and characterised using a polyphasic approach. The 16S rRNA gene sequence of strain 2C-HV3(T) showed that it has high sequence similarities with Microbispora bryophytorum NEAU-TX2-2(T) (99.9 %), Microbispora amethystogenes JCM 3021(T) (98.9 %) and Microbispora rosea subsp. rosea JCM 3006(T) (98.6 %). Phylogenetic analysis based on 16S rRNA and gyrB gene sequences demonstrated that strain 2C-HV3(T) clusters with M. bryophytorum NEAU-TX2-2(T) using two tree-making algorithms. Moreover, key morphological and chemotaxonomic properties also confirmed the affiliation of strain 2C-HV3(T) to the genus Microbispora. Longitudinal paired spores were observed to be born on short sporophores branching from the aerial hyphae. The cell wall was found to contain meso-diaminopimelic acid as the diagnostic diamino acid; madurose was found in the whole cell hydrolysate. The polar lipid profile was found to consist of diphosphatidylglycerol, phosphatidylmonomethylethanolamine, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositolmannoside, ninhydrin-positive glycophospholipids, an unidentified phospholipid and an unidentified glycolipid. The predominant menaquinones were identified as MK-9(H2) and MK-9(H4). The major fatty acids were identified as 10-methyl C17:0 and iso-C16:0. However, a combination of DNA-DNA hybridization results and some phenotypic characteristics demonstrated that strain 2C-HV3(T) can be distinguished from its closely related relatives. Consequently, it is proposed that strain 2C-HV3(T) represents a new species of the genus Microbispora, for which the name Microbispora camponoti sp. nov. is proposed. The type strain is 2C-HV3(T) (=CGMCC 4.7281(T) = DSM 100527(T)).

A novel endophytic actinomycete, designated strain NEAU-HZ10(T) was isolated from moss and characterised using a polyphasic approach. The strain was found to have morphological and chemotaxonomic characteristics typical of the genus Streptomyces. Strain NEAU-HZ10(T) formed grayish aerial mycelia, which differentiated into straight to flexuous chains of cylindrical spores. The cell wall peptidoglycan was found to contain LL-diaminopimelic acid. Predominant menaquinones were identified as MK-9(H6) and MK-9(H8). The polar lipid profile was found to consist of phosphatidylethanolamine, phosphatidylinositol and two unidentified phospholipids. The major fatty acids were identified as iso-C16:0, anteiso-C15:0 and C16:0. 16S rRNA gene sequence similarity studies showed that strain NEAU-HZ10(T) belongs to the genus Streptomyces and exhibits high sequence similarity to Streptomyces cocklensis DSM 42063(T) (98.9 %). Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain NEAU-HZ10(T) clustered with S. cocklensis DSM 42063(T), Streptomyces yeochonensis CGMCC 4.1882(T) (98.7 %), Streptomyces paucisporeus CGMCC 4.2025(T) (98.4 %) and Streptomyces yanglinensis CGMCC 4.2023(T) (98.1 %). However, a combination of DNA-DNA hybridisation results and some phenotypic characteristics indicated that strain NEAU-HZ10(T) can be distinguished from its phylogenetically closely related strains. Therefore, it is proposed that strain NEAU-HZ10(T) represents a novel species of the genus Streptomyces for which the name Streptomyces bryophytorum sp. nov. is proposed. The type strain is NEAU-HZ10(T) (= CGMCC 4.7151(T) = DSM 42138(T)).
