RESUMEN

At the seedling and adult plant phases, pot experiments were carried out to enhance the physiological-biochemical characteristics of cucumber, guarantee its high yield, and ensure its cultivation of quality. Trichoderma harzianum conidia agents at 104, 105, 106, and 107 cfu g-1 were applied in accordance with the application of Fusarium oxysporum powder at concentrations of 104 cfu/g on the protective enzyme activity, physiological and biochemical indices, seedling quality, resilience to Fusarium wilt, quality, and yield traits. Fusarium oxysporum powder at 104 cfu g-1 was used to treat CK1, while Fusarium oxysporum powder and T. harzianum conidia agents were not used to treat CK2. The results show that different T. harzianum agents improved the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT), and peroxidase (POD) in cucumber seedlings, improved chlorophyll content, root activity, root-shoot ratio, and seedling strength index, and decreased malondialdehyde (MAD) content (P < 0.05). T3, a combination of 104 cfu g-1 Fusarium oxysporum powder and 106 cfu g-1 T. harzianum conidia agents, had the greatest promoting effect. The effects of different T. harzianum conidia agents and their application amounts on the control of cucumber Fusarium wilt were explored. T3 had the best promotion impact, and the control effect of cucumber Fusarium wilt at seedling stage and adult stage reached 83.98% and 70.08%, respectively. The quality index and yield formation of cucumber were also increased by several T. harzianum agents, with T3 having the strongest promotion effects. In comparison to CK1, the soluble sugar, Vc, soluble protein, and soluble solid contents of T3 cucumber fruit were 120.75%, 39.14%, 42.26%, and 11.64% higher (P < 0.05), respectively. In comparison to CK2, the soluble sugar, Vc, soluble protein, and soluble solid contents of T3 cucumber fruit were 66.06%, 24.28%, 36.15%, and 7.95% higher (P < 0.05), respectively. In comparison to CK1 and CK2, the yields of T3 cucumber were 50.19% and 35.86% higher, respectively. As a result, T. harzianum agents can enhance the physiological and biochemical traits of cucumber seedlings, raise the quality of cucumber seedlings, have a controlling impact on Fusarium wilt, and increase the yield and quality of cucumber fruit. The greatest effectiveness of T3 comes from its use. In this study, Trichoderma harzianum conidia agents demonstrated good impacts on cucumber yield formation and plant disease prevention, demonstrating their high potential as biocontrol agents.

The diversity of genetic programs and cellular plasticity of glioma-associated myeloid cells, and thus their contribution to tumor growth and immune evasion, is poorly understood. We performed single cell RNA-sequencing of immune and tumor cells from 33 glioma patients of varying tumor grades. We identified two populations characteristic of myeloid derived suppressor cells (MDSC), unique to glioblastoma (GBM) and absent in grades II and III tumors: i) an early progenitor population (E-MDSC) characterized by strong upregulation of multiple catabolic, anabolic, oxidative stress, and hypoxia pathways typically observed within tumor cells themselves, and ii) a monocytic MDSC (M-MDSC) population. The E-MDSCs geographically co-localize with a subset of highly metabolic glioma stem-like tumor cells with a mesenchymal program in the pseudopalisading region, a pathognomonic feature of GBMs associated with poor prognosis. Ligand-receptor interaction analysis revealed symbiotic cross-talk between the stemlike tumor cells and E-MDSCs in GBM, whereby glioma stem cells produce chemokines attracting E-MDSCs, which in turn produce growth and survival factors for the tumor cells. Our large-scale single-cell analysis elucidated unique MDSC populations as key facilitators of GBM progression and mediators of tumor immunosuppression, suggesting that targeting these specific myeloid compartments, including their metabolic programs, may be a promising therapeutic intervention in this deadly cancer. One-Sentence Summary: Aggressive glioblastoma harbors two unique myeloid populations capable of promoting stem-like properties of tumor cells and suppressing T cell function in the tumor microenvironment.

Water resource is in high demand within the Yangtze River Delta, given its developed economy. Long-term exploitation of this resource has posed risks of artificial pollution and seawater intrusion to the shallow groundwater. This study aims to reveal the hydrochemical characteristics and health risks of shallow groundwater in the coastal plain of the Yangtze River Delta, as well as to discuss the possible factors affecting groundwater quality. Standard methods for hydrochemical parameter measurements, water quality assessment, and health risk models were applied to fulfill the objectives of the study. The results showed that the shallow groundwater was slightly alkaline, and the average values of total dissolved solids (TDS) and total hardness (TH) were 930.74 mg/L and 436.20 mg/L, respectively. The main hydrochemical types of groundwater were HCO3-Ca·Mg and HCO3-Ca·Na, accounting for 44.3% and 47.5%, respectively. In addition, As concentration was generally high, with a mean value of 0.0115 mg/L. The principal factors affecting the groundwater components include water-rock interactions (especially silicate), cation exchange, seawater intrusion, and human activities. The data also showed that As is strongly influenced by the redox of Fe, Mn, and NO3-. The results of the groundwater quality evaluation indicated that the shallow groundwater in some regions was unsuitable for drinking and agricultural irrigation. Health risk assessment showed that 44.3% of the water samples had significant health risks, which was attributed to the high As concentration. Therefore, it is urgent to establish long-term As monitoring to maintain sustainable groundwater management and drinking water safety. The results of this study provide essential data for water resource management and human health security in the Yangtze River Delta.

Machine learning (ML) and Natural Language Processing (NLP) have achieved remarkable success in many fields and have brought new opportunities and high expectation in the analyses of medical data, of which the most common type is the massive free-text electronic medical records (EMR). However, the free EMR texts are lacking consistent standards, rich of private information, and limited in availability. Also, it is often hard to have a balanced number of samples for the types of diseases under study. These problems hinder the development of ML and NLP methods for EMR data analysis. To tackle these problems, we developed a model called Medical Text Generative Adversarial Network or mtGAN, to generate synthetic EMR text. It is based on the GAN framework and is trained by the REINFORCE algorithm. It takes disease tags as inputs and generates synthetic texts as EMRs for the corresponding diseases. We evaluate the model from micro-level, macro-level and application-level on a Chinese EMR text dataset. The results show that the method has a good capacity to fit real data and can generate realistic and diverse EMR samples. This provides a novel way to avoid potential leakage of patient privacy while still supply sufficient well-controlled cohort data for developing downstream ML and NLP methods.
