RESUMEN

The formation of multi-pistil flowers reduces the yield and quality in Japanese apricot (Prunus mume). However, the molecular mechanism underlying the formation of multi-pistil flowers remains unknown. In the current study, overexpression of PmKNAT2/6-a, a class I KNOTTED1-like homeobox (KNOX) member, in Arabidopsis (Arabidopsis thaliana) resulted in a multi-pistil phenotype. Analysis of the upstream regulators of PmKNAT2/6-a showed that AGAMOUS-like 24 (PmAGL24) could directly bind to the PmKNAT2/6-a promoter and regulate its expression. PmAGL24 also interacted with Like Heterochromatin Protein 1 (PmLHP1) to recruit lysine trimethylation at position 27 on histone H3 (H3K27me3) to regulate PmKNAT2/6-a expression, which is indirectly involved in multiple pistils formation in Japanese apricot flowers. Our study reveals that the PmAGL24 transcription factor, an upstream regulator of PmKNAT2/6-a, regulates PmKNAT2/6-a expression via direct and indirect pathways and is involved in the formation of multiple pistils in Japanese apricot.

Japanese apricot is an important subtropical deciduous fruit tree in China, widely distributed in different altitude areas. How does it adapt to the different temperature environments in these areas? In this study, we identified a low-temperature transcription factor PmCBF03 on chromosome 7 through adaptive analysis of populations at different altitudes, which has an early termination single nucleotide polymorphism mutation. There were two different types of variation, PmCBF03A type in high-altitude areas and PmCBF03T type in low-altitude areas. PmCBF03A gene increased the survival rate, Fv/Fm values, antioxidant enzyme activity, and expression levels of antioxidant enzyme genes, and reducing electrolyte leakage and accumulation of reactive oxygen species in transgenic Arabidopsis under low temperature and freezing stress. Simultaneously, PmCBF03A gene promoted the dormancy of transgenic Arabidopsis seeds than wild-type. Biochemical analysis demonstrated that PmCBF03A directly bound to the DRE/CRT element in the promoters of the PmCOR413, PmDAM6 and PmABI5 genes, promoting their transcription and enhanced the cold resistance and dormancy of the overexpressing PmCBF03A lines. While PmCBF03T gene is unable to bind to the promoters of PmDAM6 and PmABI5 genes, leading to early release of dormancy to adapt to the problem of insufficient chilling requirement in low-altitude areas.

OBJECTIVES: By retrospective study of the epidemiological characteristics of sports-related sudden death (SrSD), the risk factors associated with SrSD were analyzed and explored to provide a scientific basis for comprehensive prevention and treatment of SrSD. METHODS: The personal information (sex, age, occupation, etc.), case information (time, place, type of sports, relative time between SrSD occurrence and exercise, etc.), death related information (sign or prodrome, medical history and surgical history, etc.), rescue situation (witnesses, on-site assistance, the availability of paramedics, etc.) of 374 SrSD cases in Guangdong Province from 2017 to 2021 were collected. Statistical analysis was conducted aiming at the key factors. RESULTS: In the 374 cases, there were significantly more males than females (19.78:1); the number of people aged between >39 and 59 was the largest (151, 40.37%); non-manual workers (68.98%) were more than manual workers; the top three sports with the highest number cases were basketball (34.49%), running (19.52%) and badminton (12.03%); from 3 pm to 9 pm (63.10%) was the time period with the highest incidence of events; sudden death mainly occurred during exercise (75.27%) and within 1 h after exercise (20.05%); the on-site rescue rate was very low (6.15%); the rate of autopsies was extremely low (1.07%); sudden cardiac death was the most common cause (67.11%). CONCLUSIONS: SrSD is most common in males aged >39 to 59 years old, mostly in non-manual workers, and usually occurs in basketball and running. Sudden death is more likely to occur during exercise and within 1 h after exercise. Therefore, the above potential risk factors should be focused on and studied in daily comprehensive prevention and treatment to provide scientific basis for accurate prevention and first aid of such sudden death.

RESUMEN

Physical exercise can reduce the overall risk of cardiovascular disease, prolong lifespan and improve the quality of life, but some studies have shown that there is a certain correlation between vigorous physical exercise and sudden cardiac death. A number of retrospective or prospective studies on sports-related sudden cardiac death (SrSCD) have been conducted at home and abroad. This article reviews the related studies on the definition, epidemiological characteristics, common causes of SrSCD and effects of excercise on cardiovascular function, pre-exercise screening and evaluation of SrSCD, in order to understand the latest research progress on SrSCD and provide clues and references for SrSCD research.

The objectives of this study were to clarify the distribution as well as the removal mechanism of antibiotic resistance genes (ARGs) within three sludge treatment wetlands (STWs) during a loading period of two years. Three STW units were constructed and run during the loading period: Unit 1 (U1) built with aeration tubes, Unit 2 (U2) built with aeration tubes and reeds, and Unit 3 (U3) built with reeds only. All targeted ARGs, intI1, and 16S rRNA were detected in residual sludge in the order of magnitude: 16S rRNA>sul1>intI1>sul2>tetC>tetA>ermB. The abundance of the five targeted ARGs, intI1, and 16S rRNA increased in residual sludge, during the loading period, which may be due to the increase in bacteria caused by the continuous import of exogenous nutrients. However, STWs can also remove ARGs from sewage during the loading period and the mean removal efficiency of five resistance genes was 73.0%. The removal rates of intI1 and 16S rRNA were 73.5% and 78.6%, respectively. Positive correlations were detected in abundance of most ARGs and intI1, as well as 16S rRNA (P < 0.05), indicating intI1 plays a vital part in the propagation of ARGs. The removal of bacteria harboring these genes also occurs in the STW units.

Prunus zhengheensis, an extremely rare population of apricots, originated in warm South-East China and is an excellent material for genetic breeding. However, most apricots and two related species (P. sibirica, P. mandshurica) are found in the cold northern regions in China and the mechanism of their distribution is still unclear. In addition, the classification status of P. zhengheensis is controversial. Thus, we generated a high-quality haplotype-resolved genome for P. zhengheensis, exploring key genetic variations in its adaptation and the causes of phylogenetic incongruence. We found extensive phylogenetic discordances between the nuclear and organelle phylogenies of P. zhengheensis, which could be explained by incomplete lineage sorting. A 242.22-Mb pan-genome of the Armeniaca section was developed with 13 chromosomal genomes. Importantly, we identified a 566-bp insertion in the promoter of the HSFA1d gene in apricot and showed that the activity of the HSFA1d promoter increased under low temperatures. In addition, HSFA1d overexpression in Arabidopsis thaliana indicated that HSFA1d positively regulated plant growth under chilling. Therefore, we hypothesized that the insertion in the promoter of HSFA1d in apricot improved its low-temperature adaptation, allowing it to thrive in relatively cold locations. The findings help explain the weather adaptability of Armeniaca plants.

Photocurable 3D printing technology has outperformed extrusion-based 3D printing technology in material adaptability, resolution, and printing rate, yet is still limited by the insecure preparation and selection of photoinitiators and thus less reported. In this work, we developed a printable hydrogel that can effectively facilitate various solid or hollow structures and even lattice structures. The chemical and physical dual-crosslinking strategy combined with cellulose nanofibers (CNF) significantly improved the strength and toughness of photocurable 3D printed hydrogels. In this study, the tensile breaking strength, Young's modulus, and toughness of poly(acrylamide-co-acrylic acid)D/cellulose nanofiber (PAM-co-PAA)D/CNF hydrogels were 375 %, 203 % and 544 % higher than those of the traditional single chemical crosslinked (PAM-co-PAA)S hydrogels, respectively. Notably, its outstanding compressive elasticity enabled it to recover under 90 % strain compression (about 4.12 MPa). Resultantly, the proposed hydrogel can be utilized as a flexible strain sensor to monitor the motions of human movements, such as the bending of fingers, wrists, and arms, and even the vibration of a speaking throat. The output of electrical signals can still be collected through strain even under the condition of energy shortage. In addition, photocurable 3D printing technology can provide customized services for hydrogel-based e-skin, such as hydrogel-based bracelets, fingerstall, and finger joint sleeves.

ABSTRACTThe distribution and removal pathways of heavy metals within different sludge treatment wetlands (STWs) during different running periods in Northeast China have not been well studied. In this study, we examined three STWs, i.e. an STW with aeration tubes only (unit 1; U1), an STW with reeds and aeration tubes (unit 2; U2), and an STW with reeds only (unit 3; U3). The results showed that the levels of Cu as well as Zn accumulated faster within STW residual sludge, whereas the levels of Cd, Cr, Ni, and Pb accumulated more slowly and decreased slightly over time. The removal rates of heavy metals from the influent sludge by STWs ranged from 64.5% (Cr) to 92.2% (Zn). Reeds removed heavy metals from the STWs by direct absorption, and Zn was highly enriched in the reeds. The presence of reeds also promoted the spreading of heavy metals to the substrate layer and improved the removal of heavy metals in STWs. The mass of each heavy metal accumulated within the residual sludge of U2 and U3 was lower than that of U1, indicating that reeds could facilitate the removal of heavy metals. The STWs removed heavy metal mainly by substrate adsorption, and the mass percentage of heavy metals accumulated in the substrate ranged from 35.8 to 63.6%.

The Knotted1-like Homeobox gene is crucial for plant morphological development and growth. Physicochemical characteristics, phylogenetic relationships, chromosomal localization, cis-acting elements, and tissue-specific expression patterns of the 11 PmKNOX genes found in the Japanese apricot genome in this study were examined. Proteins of 11 PmKNOX were soluble proteins with isoelectric points between 4.29 and 6.53, molecular masses between 15.732 and 44.011 kDa, and amino acid counts between 140 and 430. The identified PmKNOX gene family was split into three subfamilies by jointly constructing the phylogenetic tree of KNOX proteins in Japanese apricot and Arabidopsis thaliana. Combined outcomes of the analyzed conserved motifs and gene structures of the 11 PmKNOX genes from the same subfamily displayed comparable gene structure and motif patterns. The 11 PmKNOX members were distributed across six chromosomes, while two sets of PmKNOX genes were found to be collinear. Analysis of the 2000 bp promoter upstream of the coding region of the PmKNOX gene revealed that most PmKNOX genes might be involved in the physiological metabolism, growth and development processes of plants. The PmKNOX gene expression profile revealed that these genes were expressed at varying levels in different tissues, and most of them were linked to the meristems of leaf and flower buds, suggesting that PmKNOX may be involved in plants' apical meristems. In Arabidopsis thaliana, functional validation of PmKNAT2a and PmKNAT2b revealed that these two genes might be involved in regulating leaf and stem development. In addition to laying the groundwork for future research on the function of these genes, understanding the evolutionary relationships between members of the PmKNOX gene family provides opportunities for future breeding in Japanese apricots.
