RESUMEN

The pro-health action of germinated lentils could be useful to be added with wheat flour in the production of box bread. In this work, we spectroscopically evaluate the germinated and non-germinated lentils, and use them at the concentrations of 5 and 10% for the production of box bread. The chemical and physical tests of the bread and its determination of phenolic acids and flavonoids (by HPLC) were also performed. As well as the evaluation of the quality of flour and dough used to produce the bread and the acceptance of the germinated lentil bread with a population of 20 people with diabetes or with diabetic relatives It is shown that: (1) The amplitude of photoacoustic signal obtained by photoacoustic spectroscopy is modified as a function of the percentage of germinated lentil (GL) flour (0, 5 or 10%) add to the bread; being higher the photoacoustic amplitude to higher concentration of GL in the absorption band of 300-425 nm, which is related to higher content of phenols and flavonoids. (2) The contents of phenolic acids (Sinapinic, ß- resorcylic, Chlorogenic and Ferulic) and flavonoids (Quercetin and Isorhamnetin) tended to increase in the germinated lentil bread with 10% concentration of germinated lentil flour with respect to the control bread (0% GL). (3) The addition of germinated lentils flour to 5 and 10% into wheat flour to produce bread with higher hardness and less cohesiveness than bread based on wheat flour only. The Falling number indicate that there is no significant difference between the control sample and the 5% GL flour, while in the 10% GL flour there was a reduction of 21 s, with respect to the control. The effect of the germinated lentil flour percentage on the pasting properties of the flours was significant between the control and 10% GL flour. In general, the quality of the dough and flour are modified due to the addition of germinates lentils, and this affectation increases with the increase in the concentration of GL. (4) The bread added with germinated lentil has sensory acceptance with a group of people with diabetes and/or diabetic relatives in their attributes in general. The obtained results thus support the production of wheat bread with mixed germinated lentils flour, as a nutraceutical option for human consumption.

RESUMEN

Maya civilization developed in Mesoamerica and encompassed the Yucatan Peninsula, Guatemala, Belize, part of the Mexican states of Tabasco and Chiapas, and the western parts of Honduras and El Salvador. This civilization persisted approximately 3,000 years and was one of the most advanced of its time, possessing the only known full writing system at the time, as well as art, sophisticated architecture, and mathematical and astronomical systems. This civilization reached the apex of its power and influence during the Preclassic period, from 2000 BCE to 250 CE. Genetic variation in the pre-Hispanic Mayas from archaeological sites in the Mexican states of Yucatan, Chiapas, Quintana Roo, and Tabasco and their relationship with the contemporary communities in these regions have not been previously studied. Consequently, the principal aim of this study was to determine mitochondrial DNA (mtDNA) variation in the pre-Hispanic Maya population and to assess the relationship of these individuals with contemporary Mesoamerican Maya and populations from Asia, Beringia, and North, Central, and South America. Our results revealed interactions and gene flow between populations in the different archaeological sites assessed in this study. The mtDNA haplogroup frequency in the pre-Hispanic Maya population (60.53%, 34.21%, and 5.26% for haplogroups A, C, and D, respectively) was similar to that of most Mexican and Guatemalan Maya populations, with haplogroup A exhibiting the highest frequency. Haplogroup B most likely arrived independently and mixed with populations carrying haplogroups A and C based on its absence in the pre-Hispanic Mexican Maya populations and low frequencies in most Mexican and Guatemalan Maya populations, although this also may be due to drift. Maya and Ciboneys sharing haplotype H10 belonged to haplogroup C1 and haplotype H4 of haplogroup D, suggesting shared regional haplotypes. This may indicate a shared genetic ancestry, suggesting more regional interaction between populations in the circum-Caribbean region than previously demonstrated. Haplotype sharing between the pre-Hispanic Maya and the indigenous populations from Asia, the Aleutian Islands, and North, Central, and South America provides evidence for gene flow from the ancestral Amerindian population of the pre-Hispanic Maya to Central and South America.

RESUMEN

Slowing down or stopping the natural process of cataractogenesis is certainly a challenge for those who today propose an option other than surgery. Addressing the same problem in different ways constitutes a new approach to solving what is today the number one cause of reversible blindness worldwide. The technological revolution, as well as the advances in the biological sciences, allows us to conceive mechanisms never thought of before to stop the process that, as a common pathway, constitutes opacification of the crystalline lens. A new dawn for cataracts is coming through molecular, newly-discovered mechanisms. Cataractogenesis and molecular pathways have reactive free oxygen species as a common pathway. Surgical removal is today's gold standard, but perhaps not for much longer.

RESUMEN

BACKGROUND: Thiopurine S-Methyltransferase (TPMT) catalyses the S-methylation of thiopurine drugs, such as 6-mercaptopurine, 6-thioguanine and azathioprine, leading to their inactivation. Individuals who carry TPMT allele variants are more likely to experience life-threatening toxicity when these drugs are given at a standard dose. Wildtype phenotype TPMT*1 exhibits high level of catalytic activity, while all variants manifest with a decreased enzymatic activity. Ethnic-related differences in the distribution of TPMT variant alleles have been found. In Mexico, limited information is available; so far only two studies have been published and clear differences exist between them. MATERIAL AND METHODS: Allelic variants and genotypes of the TPMT gene were determined in 240 Mexican children with leukemia and solid tumors using DNA extracted from peripheral blood. Polymorphisms G460A and A719G were identified by PCR-RFLP and G238C by the specific-allele PCR assay. The enzyme variants were detected by allelic discrimination. RESULTS: Homozygous wild-type genotype TPMT*1/TPMT*1 was found in 173 patients (72.1%); 67 cases (27.9%) were heterozygous: 18 with genotype TPMT*1/TPMT*3B (7.5%), 17 TPMT*1/TPMT*3C (7.1%), 16 TPMT*1/TMPT*2 (6.7%), 14 TPMT*1/TPMT*3A (5.8%), and 2 (0.8%) were homozygous for two variants: TPMT*2/ TPMT*3B in both. The allele frequencies were TPMT*1 in 411 (85.62%), TPMT*3B in 20 (4.1%), TPMT*2 in 18 (3.75%), TPMT*3C in 17 (3.55%) and TPMT*3A in 14 (2.9%). CONCLUSIONS: A high frequency and diversity of variant TPMT genotypes was found in this series with predominance of the TPMT*3B allele.

RESUMEN

Enteroaggregative Escherichia coli (EAEC) and enterohemorrhagic E. coli (EHEC) are E. coli pathotypes associated with unmanageable diarrhea in children and adults. An alternative to the treatment of infections caused by these microorganisms is the use of the bacteria of the Lactobacillus genus; however, the beneficial effects on the intestinal mucosa are specific to the strain and species. The interest of this study consisted of analyzing the coaggregation properties of Lactobacillus casei IMAU60214, as well as the effect of cell-free supernatant (CSF) on growth and anti-cytotoxic activity in a cell model of the human intestinal epithelium for an agar diffusion assay (HT-29) and the inhibition of biofilm formation on plates of DEC strains of the EAEC and EHEC pathotypes. The results showed that L. casei IMAU60214 exhibits time-dependent coaggregation (35-40%) against EAEC and EHEC that is similar to the control E. coli ATCC 25922. The CSF showed antimicrobial activity (20-80%) against EAEC and EHEC depending on the concentration. In addition, the formation and dispersion of biofilms of the same strains decrease, and the proteolytic pre-treatment with catalase and/or proteinase K (1 mg/mL) of CSF reduces the antimicrobial effect. When evaluating the effect in HT-29 cells pre-treated with CFS on the toxic activity induced by the EAEC and EHEC strains, a reduction of between 30 and 40% was observed. The results show that L. casei IMAU60214 and its CSF have properties that interfere with some properties associated with the virulence of the EAEC and EHEC strains that cause intestinal infection, which supports their use for the control and prevention of infections caused by these bacteria.

RESUMEN

Lactobacilli species are an effective biotherapeutic alternative against bacterial infections and intestinal inflammatory disorders. However, it is important to evaluate their beneficial properties, before considering them as probiotics for medical use. In this study we evaluated some probiotic properties of Lactobacillus rhamnosus GG, Lactobacillus rhamnosus KLSD, Lactobacillus helveticus IMAU70129, and Lactobacillus casei IMAU60214 previously isolated from dairy products and as control Lactobacillus casei Shirota. Experimental evaluations revealed that all strains expressed hydrophobicity (25-40%), auto-aggregation (55-60%), NaCl tolerance (1-4%), adhesion to Caco-2 cells (25-33%), partial inhibition on adherence of Escherichia coli ATCC 35218, Salmonella Typhimurium ATCC 14028, and Staphylococcus aureus ATCC 23219. Cell-free supernatants (CFS) of Lactobacilli also inhibit growth of these pathogens. In immunomodulatory properties a reduction of interleukin-8 (IL-8) and nitric oxide (NO) release was observed in assays with Caco-2 cells stimulated with interleukin-1ß (1 ng/mL), or lipopolysaccharide (0.1 µg/mL). On the other hand, the damage induced to Caco-2 cells with sodium dodecyl sulfate (SDS) was attenuated when the cultured cells were pretreated with L. rhamnosus KLDS, L. helveticus IMAU70129 and L. casei IMAU60214. These Lactobacilli possess probiotic properties determined by both an antagonistic activity on pathogenic bacteria and reduction in the inflammatory response of cells treated with SDS, a pro-inflammatory stimulant.

RESUMEN

There is evidence that high circulating levels of IL-6 and IL-8 are markers of a poor prognosis in various types of cancer, including NB. The participation of these cytokines in the tumor microenvironment has been described to promote progression and metastasis. Our objective was to evaluate the prognostic role of genetic polymorphisms and serum levels of IL-6 and IL-8 in a cohort of Mexican pediatric patients with NB. The detection of the SNPs rs1800795 IL-6 and rs4073 and rs2227306 IL-8 was carried out by PCR-RFLP and the levels of cytokines were determined by the ELISA method. We found elevated circulating levels of IL-8 and IL-6 in NB patients compared to the control group. The genotype frequencies of the rs1800795 IL-6 and rs4073 IL-8 variants were different between the patients with NB and the control group. Likewise, the survival analysis showed that the GG genotypes of rs1800795 IL-6 (p = 0.014) and AA genotypes of rs4073 IL-8 (p = 0.002), as well as high levels of IL-6 (p = 0.009) and IL-8 (p = 0.046), were associated with lower overall survival. We confirmed the impact on an adverse prognosis in a multivariate model. This study suggests that the SNPs rs1800795 IL-6 and rs4073 IL-8 and their serum levels could be promising biomarkers of a poor prognosis, associated with overall survival, metastasis, and a high risk in Mexican children with NB.

RESUMEN

An improved understanding of how tree species will respond to warmer conditions and longer droughts requires comparing their responses across different environmental settings and considering a multi-proxy approach. We used several traits (tree-ring width, formation of intra-annual density fluctuations - IADFs, wood anatomy, Δ13C and δ18O records) to retrospectively quantify these responses in three conifers inhabiting drought-prone areas in northwestern Mexico. A fir species (Abies durangensis) was studied in a higher altitude and slightly rainier site and two pine species were sampled in a nearby, lower drier site (Pinus engelmannii, Pinus cembroides). Tree-ring-width indices (TRWi) of the studied species showed a very similar year-to-year variability likely indicating a common climatic signal. Wood anatomy analyses done over 3.5 million measured cells, showed that P. cembroides lumen area was much smaller than in the other two species and it remained constant along all the studied period (over 64 years). Instead, cell wall thickness was widest in P. engelmannii and this species presented the highest amount of intra-annual density fluctuations. Climate and wood anatomy correlations pointed out that lumen area was positively affected by winter precipitation for all studied species, while cell-wall thickness was negatively affected by this season's precipitation in all species but P. cembroides. Stable isotope analysis showed significantly lower values of Δ13C for P. cembroides and no significant δ18O differences between the three species, although they shared a common decreasing trend. With very distinct wood anatomical traits (smaller cells, compact morphology), P. cembroides stood out as the better adapted species in its current environment and could be less affected by future drier climate. P. engelmannii and A. durangensis showed high plasticity at wood anatomical level, allowing them to promptly respond to seasonal water availability but likely gives few advantages on future climate scenarios with longer and frequent drought spells.

RESUMEN

Mediterranean climate promotes two distinct growth peaks separated by summer quiescence in trees. This bimodal pattern has been associated to favourable growing conditions during spring and autumn when mild temperatures and soil-water availability enhance cambial activity. Climatic models predict progressive warming and drying for the Mediterranean Basin, which could shorten or shift the spring and autumn growing seasons. We explored this idea by comparing two sites with different Mediterranean climate types (continental/dry and coastal/wet) and studied how climate drives the bimodal growth pattern in Aleppo pine (Pinus halepensis). Specifically we investigated the intra-annual changes in wood anatomy and the corresponding formation of density fluctuations (IADF). Trees on both sites were analyzed by dendrometer monitoring and by developing chronologies of wood anatomical traits. Radial-increment dynamics followed a similar bimodal pattern in both sites but coastal trees showed higher increments during the spring and autumn growth peaks, especially in autumn. The summer rest of cambium activity occurs almost one month earlier in the coastal than in the inland site. Lumen area and cell-wall thickness were significantly smaller in the continental site, while the increment rate of cell-wall thickness during an IADF event was much higher in the coastal pines. The accumulated soil moisture deficit was the main climatic constraint of tracheid enlargement in continental pines. Intra-annual density fluctuations were more frequent in the coastal trees where wood anatomy features recover to average values after such events, meanwhile inland trees presented a much lower recovery rate. Growth bimodality and the formation of density fluctuations were linked, but mild climate of the coastal site allows a longer growing season, which explains why trees in this area showed higher and more variable growth rates.

RESUMEN

Understanding how climate affects xylem formation is critical for predicting the impact of future conditions on tree growth and functioning in the Mediterranean region, which is expected to face warmer and drier conditions. However, mechanisms of growth response to climate at different temporal scales are still largely unknown, being complicated by separation between spring and autumn xylogenesis (bimodal temporal pattern) in most species such as Mediterranean pines. We investigated wood anatomical characteristics and carbon stable isotope composition in Mediterranean Pinus pinea L. along tree-ring series at intra-ring resolution to assess xylem formation processes and responses to intra-annual climate variability. Xylem anatomy was strongly related to environmental conditions occurring a few months before and during the growing season, but was not affected by summer drought. In particular, the lumen diameter of the first earlywood tracheids was related to winter precipitation, whereas the size of tracheids produced later was influenced by mid-spring precipitation. Diameter of latewood tracheids was associated with precipitation in mid-autumn. In contrast, tree-ring carbon isotope composition was mostly related to climate of the previous seasons. Earlywood was likely formed using both recently and formerly assimilated carbon, while latewood relied mostly on carbon accumulated many months prior to its formation. Our integrated approach provided new evidence on the short-term and carry-over effects of climate on the bimodal temporal xylem formation in P. pinea. Investigations on different variables and time scales are necessary to disentangle the complex climate influence on tree growth processes under Mediterranean conditions.

The human eye is highly exposed to light of all types of electromagnetic waves, the metabolic stress in the elimination of cellular damage, as well as its accumulation, constitute the major oxidative stress due to ultraviolet radiation. The objective was to document the new scientific evidence in epigenetics regarding ultraviolet radiation and oxidative stress in cataract formation. A review of the literature from May 1, 2021 to May 1, 2022 was performed with meta-search engines in English and Spanish. Cumulative biochemical damage at the histone level is considered the primary environmental insult in cataract formation. The immunomodulatory potential of human lens epithelium cells is a promising therapeutic target, due to being the main cell line affected in ultraviolet radiation. The technological, biochemical and physiological advance will allow promoting a different solution, by a different concept of surgery, for the cure of the most prevalent condition in the world for reversible blindness: cataract(AU)

We present the case of a 28-year-old male patient, heterosexual, with negative HIV who developed an intense skin rash characterized by the presence of confluent vesicles, pustules, and umbilicated papules on the penis and pubic region with little spread to the rest of the body, without lymphadenopathy or systemic manifestations, four days after having sexual relations with an apparently healthy prostitute. The swabbing of the dermal lesions revealed the presence of MPOX virus DNA by real-time PCR. The immunological tests were the following: negative fourth-generation ELISA and Western Blot for HIV, IgM antibodies to herpes simplex1=1.2 U/ ml, IgM antibodies to herpes simplex2=1.9 U/ml, IgM antibodies to varicella zoster=0.5 S/CO and negative FTA-ABS (IgM) for syphilis. The intensity of skin lesions on the penis and the need to perform tests to rule out MPOX in prostitutes are highlighted.

RESUMEN

Using the conventional Haberkorn approach, it is evaluated the recombination of the radical pair (RP) singlet spin state to study theoretically the cytoprotective effect of an extremely-low-frequency electromagnetic field (ELF-EMF) on early stages of hepatic cancer chemically induced in rats. The proposal is that ELF-EMF modulates the interconversion rate of singlet and triplet spin states of the RP populations modifying the products from the metabolization of carcinogens. Previously, we found that the daily treatment with ELF-EMF 120 Hz inhibited the number and area of preneoplastic lesions in chemical carcinogenesis. The singlet spin population is evaluated diagonalizing the spin density matrix through the Lanczos method in a radical pair mechanism (RPM). Using four values of the interchange energy, we have studied the variations over the singlet population. The low magnetic field effect as a test of the influence over the enzymatic chemical reaction is evaluated calculating the quantum yield. Through a bootstrap technique the range is found for the singlet decay rate for the process. Applying the quantum measurements concept, we addressed the impact toward hepatic cells. The result contributes to improving our understanding of the chemical carcinogenesis process affected by charged particles that damage the DNA.

ABSTRACT Diabetic nephropathy (DN) is a public health problem whose prevalence is constantly increasing, it represents a significant economic, social and health burden, its diagnosis is usually late and not routine. Early and timely diagnosis slows disease progression, improves quality of life, increases life expectancy, and reduces high costs of care. It is characterized by elevated urinary albumin excretion (≥30 mg / g urinary creatinine) and / or estimated glomerular filtration rate <60 ml / min / 1.73 m2. The pathophysiology of DN is complex, determined by persistent hyperglycemia, and includes renal hemodynamic changes, spinal ischemia, tissue inflammation, and activation of the renin-angiotensin-aldosterone system. Among the non-modifiable risk factors associated with DN are gender, chronological age, age at diagnosis of DM2, duration of DM2, family history of DN, ethnicity and genetic factors. Among the modifiable poor prognostic factors for DN, the modifiable risk factors for DN are diabetes education, smoking, high glycosylated hemoglobin levels, arterial hypertension, dyslipidemia, abdominal obesity, and bad prognostic factors would be anemia. retinopathy, neuropathy, and elevated uric acid levels. A review of the literature of the factors associated with DN is carried out and the importance of its early and timely diagnosis is emphasized.

RESUMEN

Forecasted warmer and drier conditions will probably lead to reduced growth rates and decreased carbon fixation in long-term woody pools in drought-prone areas. We therefore need a better understanding of how climate stressors such as drought constrain wood formation and drive changes in wood anatomy. Drying trends could lead to reduced growth if they are more intense in spring, when radial growth rates of conifers in continental Mediterranean climates peak. Since tree species from the aforementioned areas have to endure dry summers and also cold winters, we chose two coexisting species: Aleppo pine (Pinus halepensisMill., Pinaceae) and Spanish juniper (Juniperus thuriferaL., Cupressaceae) (10 randomly selected trees per species), to analyze how growth (tree-ring width) and wood-anatomical traits (lumen transversal area, cell-wall thickness, presence of intra-annual density fluctuations-IADFs-in the latewood) responded to climatic variables (minimum and maximum temperatures, precipitation, soil moisture deficit) calculated for different time intervals. Tree-ring width and mean lumen area showed similar year-to-year variability, which indicates that they encoded similar climatic signals. Wet and cool late-winter to early-spring conditions increased lumen area expansion, particularly in pine. In juniper, cell-wall thickness increased when early summer conditions became drier and the frequency of latewood IADFs increased in parallel with late-summer to early-autumn wet conditions. Thus, latewood IADFs of the juniper capture increased water availability during the late growing season, which is reflected in larger tracheid lumens. Soil water availability was one of the main drivers of wood formation and radial growth for the two species. These analyses allow long-term (several decades) growth and wood-anatomical responses to climate to be inferred at intra-annual scales, which agree with the growing patterns already described by xylogenesis approaches for the same species. A plastic bimodal growth behavior, driven by dry summer conditions, is coherent with the presented wood-anatomical data. The different wood-anatomical responses to drought stress are observed as IADFs with contrasting characteristics and responses to climate. These different responses suggest distinct capacities to access soil water between the two conifer species.

Cataract comprises opacification of the crystalline lens, which may progressively affect the cortex and the anterior subcapsular nucleus, secondary to accumulation of damaged proteins on this level, with loss of balance between production and elimination of free reactive oxygen species. The importance of delaying or identifying specific markers, as well as promoting a new therapeutic target, is the object of study and analysis of a variety of research lines. A review was conducted of the literature published from 1 January to 20 July 2020. Use was made of PubMed, Infomed, Clinical Key, Lilacs, EBSCO, SciELO, Prisma and UpToDate metasearch engines in English and Spanish to identify new scientific evidence about oxidative stress and its involvement in cataract formation. The crystalline lens barrier serves as a medium for exchange between various molecules, preventing entrance of antioxidants into the nucleus, which results in opacification. Mitochondria on the crystalline lens cortex allow oxygen removal. Oxidative phosphorylation then forms free superoxide radicals which naturally accumulate on this level with the passing of time. With aging, adaptive homeostasis loses its ability to respond to oxidative stress changes, but the prophylactic, targeted use of antioxidants may change the ultimate fate of this condition. Lack of balance in oxidation-reduction processes is the cause of cataract formation(AU)

RESUMEN

BACKGROUND: Genetic polymorphisms in patients with acute lymphoblastic leukemia (ALL) may influence the toxicity of chemotherapeutic agents. Due to the importance of the transport P-glycoprotein and methylenetetrahydrofolate reductase in the metabolism of chemotherapeutic agents, we analyzed the MDR1 rs1045642 and MTHFR rs1801133 polymorphisms and their associations with clinical outcomes in Mexican childhood ALL patients. METHODS: A total of 109 patients participated in this study. The clinical evaluation consisted of a physical examination and a laboratory test. Genotyping of MDR1 rs1045642 (3435 C>T) and MTHFR rs1801133 (677 C>T) was performed by polymerase chain reaction/restriction fragment length polymorphism. Statistical analyses were performed using SPSS 14.0. The odds ratios and 95% confidence intervals (CI) were estimated by logistic regression. RESULTS: Individuals who were CC homozygotes at MDR1 rs1045642 had lower risk of having methotrexate plasma concentrations >1 µM and leukopenia grade I (odds ratio [OR] = 0.30; 95% CI = 0.13-0.72 and OR = 0.32; 95% CI = 0.14-0.72, respectively). Patients who were CC homozygotes at MTHFR rs1801133 had a higher risk of developing mucositis (OR = 3.61; 95% CI = 1.42-9.14). CONCLUSION: MDR1 rs1045642 and MTHFR rs1801133 should be considered as diagnostic candidates for the identification of pediatric patients with a high risk of suffering adverse events during ALL treatment.

ABSTRACT The history of pandemics and modern man have marked a before and after in current medicine, as was the case with the Spanish flu in 1918. The last pandemic recorded worldwide is coronavirus, identified as COVID-19, caused by the viral agent SARS-CoV-2. From its onset on 30 December 2019 this disease has had a close connection to ophthalmology. The purpose of the study was to document the new scientific evidence about this condition and its ocular manifestation through a literature review covering the period extending from 1 January to 25 April 2020, using search terms in English and Spanish. Respiratory symptoms are among the transmission routes most commonly dealt with. However, some ocular manifestations are characterized by the presence of the SARS-CoV-2 virus in tears and in the conjunctiva of patients infected with COVID-19. The most typical manifestation is nonspecific viral conjunctivitis. Mortality rate is a topic of special relevance. The statistics accumulated suggest greater fatality and thus the need to implement social measures and actions aimed at population control. The current fatality rate is 7.2 percent worldwide, 5.8 percent in Latin America and 4.2 percent in Cuba. The present paper provides a comprehensive analysis of COVID-19 based on international recommendations for ophthalmology personnel and facilities, as well as the risk factors, clinical status, new treatments and reports about ongoing clinical studies(AU)
