[Pharmacokinetic and clinical research on a new aminoglycoside antibiotic: sisomicin]. / Ricerca farmacocinetica e clinica su un nuovo antibiotico aminoglicosidico: sisomicina.

Results obtained with sisomycin in 10 cases of purulent meningitis and 4 of bronchopulmonitis showed good tissue diffusion, together with CSF concentrations sufficient to inhibit the main aetiological agents in these forms: MIC 0.02 mg/ml for both D. pneumoniae (I-ATCC 6301) and N. meningitidis (C-ATCC 13101), i.e. liquor transfer of the antibiotic more than sufficient to handle these agents.

[Pharmacokinetics of a new aminoglycoside: kanendomycin]. / Farmacocinetica di un nuovo aminoglicoside: kanendomicina.

The pharmacokinetics of kanendomycin, a new aminoglycoside derived from kanamycin, has been assessed in 15 volunteers after i.m. administration of 100 and 300 mg and i.v. administration of 100 mg. Analysis of the tricompartmental model adopted for the pharmacokinetic study showed that kanendomycin half-life is similar and perhaps superior to that of gentamycin. The antibiotic's bioavailability is of the order of 70-80% of the dose used.

[Pharmacokinetic and clinical research of a new phenyl ester of carbecinillin: carfecillin]. / Ricerca farmacocinetica e clinica su un nuovo estere fenilico di carbenicillina: carfecillina.

Following a review of the latest methods of treating infections of the urinary ways, the pharmacokinetic and clinical results of a new antibiotic, Carfecillin, are reported. The examination was carried out on a total of 10 volunteers. The substance showed good serum levels and fair renal excretion with both the 250 mg and the 500 mg p.o. doses. The clinical study was carried out on a double blind basis versus a standard reference (indanyl ester of carbenicillin) in 26 patients and showed the electivity of the antibiotic in the oral treatment of urinary infections under well tolerated conditions.

[Pharmacokinetic and clinical research on a new antibiotic combination (amoxicillin and flucloxacillin in equivalent-weight dose)]. / Ricerca farmacocinetica e clinica su una nuova associazione antibiotica (amoxicillina e flucloxacillina in dosi equiponderali)

A controlled double-blind biometric and an open clinical trial were conducted to determine the therapeutic effectiveness of a new equal-dose w/w association of amoxicillin and flucloxacillin. The following conclusions were drawn. Both antibiotics were present in high serum levels; those of flucloxacillin were higher and more persistent. Analysis of variance on 89 patients pointed to the superiority of the association by comparison with amoxicillin alone. The clinical study made it clear that the broad and complementary spectrum of the association, its synergy, absence of toxicity and good gastric tolerance make it a valuable and effective therapeutic aid, also in presence of germs that produce beta-lactase.

[Pharmacokinetics and clinical aspects of a new ampicillin]. / Farmacocinetica e clinica di una nuova ampicillina

The chemical, physical, kinetic features, spectrum and clinical employment of amoxicillin (alpha-amino-p-hydroxbenzylpenicillin) are described. A kinetic study on 48 volunteers indicated its rapid gastroenteric absorption. In addition, the new drug quickly reached serum levels that were able double those obtained with corresponding ampicillin doses of equal weight. A very satisfactory therapeutic response and no more than trivial side-effects were noted in a clinical trial on 106 patients, most of whom presented bacterial infection of the upper and lower airways.
