RESUMEN

Pneumonia in cattle is one of the causes of morbidity rates and economic loss. The host response to lung infections caused by Ureaplasma diversum in bovines is virtually unknown. Here in the immune response was evaluated in a murine model for an experimental pulmonary infection by U. diversum. Therefore, AJ, BALB/C and C57BL/6 mice received intratracheal inoculation of U. diversum and were evaluated after 1, 2, 3, 7 and 14 days and the clinical specimens were collected. In bronchoalveolar lavages (BAL) an increase of inflammatory cells was observed. Neutrophils were the main cells recruited to the site of infection and the infiltration was coincided with the production of pro-inflammatory cytokines. We found a large amount of neutrophil in this initial period, followed by a decrease 7 and 14 days post infection, accompanied by bacterial clearance. Our results evidenced the presence of U. diversum within the neutrophil that suggests a phagocytic role of this cell in the elimination of the infection. The immune response features reported here are the initial evidence that healthy immune systems may control these microorganisms. This may be the first step to design new strategies immune based to control the infections in naturally infected hosts.

The aim of this study was to identify force-velocity and power-velocity curves in climbing activity protocols, used as dynamic resistance exercise in rats. Eighteen 45-day-old male Wistar rats (weight = 211.9 ± 5.2 g) were evaluated. After familiarization to the climbing procedure, the animals performed an incremental climbing test (load relative to 75% of the body mass at first stage, followed by 30 g increments with and 120 s recovery between climbs) to determine the maximum carrying capacity (MCC). After this, the animals climbed with different loads (without load, 10%, 20%, 30%, 40%, 50%, 75%, 90%, and 100% of MCC) with 120 s recovery between climbs. Time for each climb was recorded to calculate the mechanical power. The peak power was reached at 30% of MCC. For the force-velocity curve, an inversely proportional relation was observed between force and velocity, as expected, greater forces were expressed in lower velocities. Therefore, our results suggest that training at 30% of MCC should be encouraged aiming the target for greater power output and 90%-100% of MCC should be the load aiming for strength training in climbing activities for rats.

J Strength Cond Res 32(1): 48-56, 2018-Various body positions have been used in the scientific literature when performing the isometric midthigh pull resulting in divergent results. We evaluated force production in the isometric midthigh pull in bent (125° knee and 125° hip angles) and upright (125° knee, 145° hip angle) positions in subjects with (>6 months) and without (<6 months) substantial experience using weightlifting derivatives. A mixed-design ANOVA was used to evaluate the effect of pull position and weightlifting experience on peak force, force at 50, 90, 200, and 250 ms. There were statistically significant main effects for weightlifting experience and pull position for all variables tested, and statistically significant interaction effects for peak force, allometrically scaled peak force, force at 200 ms, and force at 250 ms. Calculated effect sizes were small to large for all variables in subjects with weightlifting experience, and were small to moderate between positions for all variables in subjects without weightlifting experience. A central finding of the study is that the upright body position (125° knee and 145° hip) should be used given that forces generated are highest in that position. Actual joint angles during maximum effort pulling should be measured to ensure body position is close to the position intended.

BACKGROUND AND AIMS: Whether intensity or other characteristics of physical activity can better promote the release of nitric oxide (NO) and reduction of blood pressure in hypertensive older-adults is still unknown. In this study, the post-exercise blood pressure (BP) response and NO release after different intensities of aerobic exercise in elderly women were analyzed. METHODS: Blood pressure response and NO were analyzed in 23 elderly mildly hypertensive women. Participants underwent (1) high-intensity incremental exercise (IT); (2) moderate-intensity 20 min exercise at 90% of the anaerobic threshold (AT), and (3) control (CONT) session. BP was measured before and after interventions; volunteers remained seated for 1 h. NO estimates were made through NO2- analyses. RESULTS: After CONT session, both diastolic BP and mean arterial pressure (MAP) were significantly higher than during pre-exercise resting. Post-exercise hypotension (PEH) was observed after exercise at IT and 90% of AT. Although exercise in both sessions lowered SBP and MAP compared with CONT, exercise at the highest intensity (IT) was more effective on lowering systolic BP after exercise. In comparison with pre-exercise resting, NO2- increased significantly only after IT, but both exercise sessions caused NO2- to increase compared with CONT. CONCLUSION: Exercise intensity and NO release may exert a role in eliciting PEH in mildly hypertensive elderly women.

Cognitive decline usually begins after individuals reach maturity, which is more evident in late adulthood. Rapid and constant cognitive screenings allow early detection of cognitive decline and motivate individuals to participate in prevention interventions. Due to accelerated technological advances, cognitive screening and training are now available to the layperson using electronic devices connected to the internet. Large datasets generated by these platforms provide a unique opportunity to explore cognitive development throughout life and across multiple naturalistic environments. However, such data collection mechanisms must be validated. This study aimed to determine whether the data gathered by commercial visuospatial and phonological working memory tests (CogniFit Inc., San Francisco, USA) confirm the well-established argument that age predicts cognitive decline. Data from 3,212 participants (2,238 females) who were 45 years old or older were analyzed. A linear regression analysis explored the relationship between age and working memory while controlling for gender, sleep quality, and physical activity (variables that are known to affect working memory). We found that age negatively predicts working memory. Furthermore, there was an interaction between age and gender for visuospatial working memory, indicating that although male participants significantly outperformed females, the relationship between age and working memory differs for females and males. Our results suggest that the computerized assessment of visuospatial and phonological working memory is sensible enough to predict cognitive functions in aging. Suggestions for improving the sensitivity of self-reports are discussed. Further studies must explore the nature of gender effects on cognitive aging.

The consumption of high-protein diets (HPD) is associated with resistance training (RT) due to effects on metabolism. However, little is known about these effects on cardiac tissue. This study aimed to investigate effects of HPD and RT on cardiac biomarkers. 18 rats were divided into normo-protein (NPD), and HPD groups: NPD-Control, NPD-RT, HPD-Control, and HPD-RT. Interleukin-6 (IL-6), tumour necrosis factor alpha (TNF-a), nitric oxide (NO), activity of metalloproteinase-2 (MMP-2), and vascular factor (VEGF) were analysed. RT was effective in regulating body weight, increasing strength, and reducing food consumption (p < .05). HPD induces higher levels of interleukin 6 (p = .0169), and lowers NO (p < .0001). When associated with RT, the HPD decreases levels of tumour necrosis factor alpha, while enhances NO, and MMP activity (p < .05). The association of RT with HDP decreases inflammatory parameters and indicates an enhancement in the molecular parameters of cardiac tissue.

BACKGROUND: The absence of the I allele of the angiotensin converting enzyme (ACE) gene has been associated with higher levels of circulating ACE, lower nitric oxide (NO) release and hypertension. The purposes of this study were to analyze the post-exercise salivary nitrite (NO2-) and blood pressure (BP) responses to different exercise intensities in elderly women divided according to their ACE genotype. METHODS: Participants (n = 30; II/ID = 20 and DD = 10) underwent three experimental sessions: incremental test - IT (15 watts workload increase/3 min) until exhaustion; 20 min exercise 90% anaerobic threshold (90% AT); and 20 min control session without exercise. Volunteers had their BP and NO2- measured before and after experimental sessions. RESULTS: Despite both intensities showed protective effect on preventing the increase of BP during post-exercise recovery compared to control, post-exercise hypotension and increased NO2- release was observed only for carriers of the I allele (p < 0.05). CONCLUSION: Genotypes of the ACE gene may exert a role in post-exercise NO release and BP response.

Methods: Variables from 305 matches from 2019/2020 Bundesliga season were analyzed. Normality, mean comparisons, analysis of variance and effect sizes were performed. Results: After SD, lower values were observed for goals attempt and distance covered, while higher values were found for passes accuracy and fouls committed (p < 0.05). Home teams before SD presented higher number of goals scored at 2nd half and less fouls committed than visiting opponents, after SD away teams had higher number of goals scored at the same half (p = 0.043) and a similar number of fouls (p = 0.035). Home teams performed more sprints than visiting opponents before SD, but after SD no difference was observed between home and away teams (p = 0.042). Total distance covered for both home and away teams were lower after SD (p = 0.041). Home teams presented higher values of goal attempts to the target, but changing from small to trivial effects sizes (ES) after SD. However, home advantage was 50.32 ± 19.06% before SD, after SD comeback the home advantage was 40.37 ± 26.08% (ES = 0.45). Conclusions: The return to football pitch without stadium supporters, affected some game and physical variables, as well as home advantage.

The increasing demand for water, food and energy poses challenges for the world´s sustainability. Tropical palm oil is currently the major source of vegetable oil worldwide with a production that exceeds 55 million tons per year, while generating over 200 million tons of palm oil mill effluent (POME). It could potentially be used as a substrate for production of microalgal biomass though. In this study, the microalgal strain Chlamydomonas biconvexa Embrapa|LBA40, originally isolated from a sugarcane vinasse stabilization pond, was selected among 17 strains tested for growth in POME retrieved from anaerobic ponds of a palm oil industrial plant located within the Amazon rainforest region. During cultivation in POME, C. biconvexa Embrapa|LBA40 biomass productivity reached 190.60 mgDW • L-1 • d-1 using 15L airlift flat plate photobioreactors. Carbohydrates comprised the major fraction of algal biomass (31.96%), while the lipidic fraction reached up to 11.3% of dry mass. Reductions of 99% in ammonium and nitrite, as well as 98% reduction in phosphate present in POME were detected after 5 days of algal cultivation. This suggests that the aerobic pond stage, usually used in palm oil industrial plants to reduce POME inorganic load, could be substituted by high rate photobioreactors, significantly reducing the time and area requirements for wastewater treatment. In addition, the complete mitochondrial genome of C. biconvexa Embrapa|LBA40 strain was sequenced, revealing a compact mitogenome, with 15.98 kb in size, a total of 14 genes, of which 9 are protein coding genes. Phylogenetic analysis confirmed the strain taxonomic status within the Chlamydomonas genus, opening up opportunities for future genetic modification and molecular breeding programs in these species.

High-protein diets (HPDs) are widely used for health and performance. However, the combination of whey protein and natural foods (i.e., fruits) is still unclear. Thus, we evaluated the role of supplemental HPD with Bocaiuva (Acrocomia sp.) in metabolic and body composition parameters of rats submitted to resistance training (RT). Wistar rats (203.3 ± 30 g) were randomly allocated to five groups: normoproteic control (CON, n = 5), sedentary high-protein (SH, n = 5), RT + H (trained high-protein [TH], n = 5), sedentary+Bocaiuva (SH+B, n = 4), and RT+Bocaiuva (TH+B, n = 4) diet groups. After 12 weeks of RT, the maximal strength increased in both trained groups (P < .05). The TH + B group had lower values of adiposity index (AI) (3.8 ± 0.7% vs. 6.8 ± 1.3%) and visceral fat (0.038 ± 0.004 g/g vs. 0.067 ± 0.012 g/g) compared with the SH group, respectively (P < .05). The other groups did not show differences in values of AI (CON, 5.4 ± 1.6%, TH, 5.4 ± 1.3%, and SH+B, 5.5 ± 1.2%). In addition, the fasting glucose of trained groups (TH, 106.0 ± 4.5, and TH+B, 100.4 ± 13.5 dL/mg) was significantly lower when compared with controls (SH, 120.0 ± 14.4, and SH+B, 119 ± 6.4 dL/mg) (P < .05). Bocaiuva combined with an HPD reduced visceral fat and AI in addition to improving glucose tolerance of rats submitted to RT.

RESUMEN

In this paper we show that the absorption spectrum of the microalgae Nannochloropsis oceanica exhibits changes in response to the modulation of incident light. A model was used to analyze the contribution of different active pigments to the total absorption in the photosynthetically active radiation region and suggested consistent diel oscillations in the optical activity of carotenoids.

Sugarcane ethanol is produced at large scale generating wastes that could be used for microalgae biomass production in a biorefinery strategy. In this study, forty microalgae strains were screened for growth in sugarcane vinasse at different concentrations. Two microalgae strains, Micractinium sp. Embrapa|LBA32 and C. biconvexa Embrapa|LBA40, presented vigorous growth in a light-dependent manner even in undiluted vinasse under non-axenic conditions. Microalgae strains presented higher biomass productivity in vinasse-based media compared to standard Bold's Basal Medium in cultures performed using 15L airlift flat plate photobioreactors. Chemical composition analyses showed that proteins and carbohydrates comprise the major fractions of algal biomass. Glucose was the main monosaccharide detected, ranging from 46% to 76% of the total carbohydrates content according to the strain and culture media used. This research highlights the potential of using residues derived from ethanol plants to cultivate microalgae for the production of energy and bioproducts.

This study evaluated the feasibility of using the Ribulose Bisphosphate Carboxylase Large subunit gene (rbcL) and the Internal Transcribed Spacers 1 and 2 of the nuclear rDNA (nuITS1 and nuITS2) markers for identifying a very diverse, albeit poorly known group, of green microalgae from neotropical inland waters. Fifty-one freshwater green microalgae strains isolated from Brazil, the largest biodiversity reservoir in the neotropics, were submitted to DNA barcoding. Currently available universal primers for ITS1-5.8S-ITS2 region amplification were sufficient to successfully amplify and sequence 47 (92%) of the samples. On the other hand, new sets of primers had to be designed for rbcL, which allowed 96% of the samples to be sequenced. Thirty-five percent of the strains could be unambiguously identified to the species level based either on nuITS1 or nuITS2 sequences' using barcode gap calculations. nuITS2 Compensatory Base Change (CBC) and ITS1-5.8S-ITS2 region phylogenetic analysis, together with morphological inspection, confirmed the identification accuracy. In contrast, only 6% of the strains could be assigned to the correct species based solely on rbcL sequences. In conclusion, the data presented here indicates that either nuITS1 or nuITS2 are useful markers for DNA barcoding of freshwater green microalgae, with advantage for nuITS2 due to the larger availability of analytical tools and reference barcodes deposited at databases for this marker.

Staphylococcus aureus infection may lead to the development of soft tissue damage. It has been evaluated in other researches using different animal models. In addition, the inflammatory response developed by the host organism facing an infection by this pathogen has been analyzed and neutrophils have been linked to the immune response developed. In this study, we aimed to compare the inflammatory response developed by the host induced by an intradermal infection with a methicillin-resistant strain of Staphylococcus aureus (MRSA) or a methicillin-susceptible strain of Staphylococcus aureus (MSSA). Mice euthanasia occurred in the following times: 6, 24, 48 and 96 hours of infection; the cell number and the cytokine release were evaluated. Our results showed that infections by different strains of Staphylococcus aureus lead to different immune response degrees. Although MRSA infection induces higher neutrophil recruitment to the infection site and higher inflammatory response in the draining lymph node, the increased production of TNF-α, IFN-γ, IL-6 and IL-1ß in the lymph node 6 hours after the infection was observed only in MSSA infected animals. Considering the data, MSSA may have mechanisms to prevent neutrophil recruitment to the infection site.


Infecções causadas por Staphylococcus aureus podem causar o desenvolvimento de lesões no tecido frouxo. Isto tem sido avaliado em estudos que avaliam a resposta imune em diferentes modelos animais. Além disso, a resposta inflamatória desenvolvida pelo hospedeiro frente à infecção por este patógeno tem sido analisada e neutrófilos têm sido associados com a resposta imune desenvolvida. Neste estudo, nosso objetivo foi comparar a resposta inflamatória desenvolvida pelo organismo hospedeiro induzida por uma infecção intradérmica com uma cepa de Staphylococcus aureus resistente à meticilina (MRSA) ou uma cepa Staphylococcus aureus susceptível à meticilina (MSSA). A eutanásia dos camundongos ocorreu nos seguintes tempos: 6, 24, 48 e 96 horas de infecção; o número de células e a quantidade de citocinas foram avaliados. Nossos resultados mostraram que infecções por diferentes cepas de Staphylococcus aureus causam resposta imunológica com diferentes intensidades. Enquanto infecções por MRSA induzem maior recrutamento de neutrófilos para o sítio de infecção e maior resposta inflamatória no linfonodo, o aumento na produção de TNF-α, IFN-γ, IL-6 e IL-1ß no linfonodo 6 horas após a infecção foi observado somente nos animais infectados com MSSA. Considerando as análises, MSSA pode possuir mecanismos para prevenir o recrutamento de neutrófilos para o sítio de infecção.
