RESUMEN

INTRODUCTION: The risk for lung cancer is incremented in high degree dysplasia (HGD) and in subjects with hypermethylation of multiple genes. We sought to establish the association between them, as well as to analyze the DNA aberrant methylation in sputum and in bronchial washings (BW). METHODS: Cross sectional study of high risk patients for lung cancer in whom induced sputum and autofluorescence bronchoscopy were performed. The molecular analysis was determined on DAPK1, RASSF1A and p16 genes using Methylation-specific PCR. RESULTS: A total of 128 patients were enrolled in the study. Dysplasia lesions were found in 79 patients (61.7%) and high grade dysplasia in 20 (15.6%). Ninety eight patients out of 128 underwent molecular analysis. Methylation was observed in bronchial secretions (sputum or BW) in 60 patients (61.2%), 51 of them (52%) for DAPK1, in 20 (20.4%) for p16 and in three (3.1%) for RASSF1A. Methylated genes only found in sputum accounted for 38.3% and only in BW in 41.7%, and in both 20.0%. In the 11.2% of the patients studied, HGD and a hypermethylated gene were present, while for the 55.1% of the sample only one of both was detected and for the rest of the subjects (33.6%), none of the risk factors were observed. CONCLUSIONS: Our data determines DNA aberrant methylation panel in bronchial secretions is present in a 61.2% and HGD is found in 15.6%. Although both parameters have previously been identified as risk factors for lung cancer, the current study does not find a significative association between them. The study also highlights the importance of BW as a complementary sample to induced sputum when analyzing gene aberrant methylation.

The present study was carried out to evaluate the phytoplankton community and water quality in four parallel fishponds (FP1 to FP4) during one year. Some abiotic parameters of water column and phytoplankton was sampled in four sites, one in each fishpond. Similarity analysis showed that FP2 fishpond was markedly different for the others ponds with regard to phytoplankton community. Conductivity, orthophosphate, temperature, TSS and hardness parameters were significantly different (p<0.05) between fishponds and all limnological parameters were significantly different (p<0.01) among period (months). Organic matter and phosphorus of the sediment were significantly different (p<0.01) only among fishponds. In current study Chlorophyceae and Bacillariophyceae occurred in most months. Cyanobacteria density in April and May was higher in all fishponds, at FP3 represented 71.4% in April and FP1 represented 78.7% in May of total reported, with chlorophyll-a content lower than 41 mg.L-1. The physical and chemical parameters of fishponds water constituted relations between the parameters and algae occurrence. The present study however, indicated that Cyanobacteria was more frequent in the fishponds after the fish grow-out period (April), and the water quality should be monitored more frequently in this period, due to the high total phosphorus, orthophosphate and chlorophyll-a contents.

AIMS: To investigate the volatile fractions of 16 essential oils for activity against the more common fungi causing spoilage of bakery products, Eurotium amstelodami, E. herbariorum, E. repens, E. rubrum, Aspergillus flavus, A. niger and Penicillium corylophilum. METHODS AND RESULTS: The study applied 50 microl of pure essential oils in a sterilized filter paper, were carried out at pH 6 and at different water activity levels (0.80-0.90). First, a wheat flour based agar medium was used, where cinnamon leaf, clove, bay, lemongrass and thyme essential oils where found to totally inhibit all microorganisms tested. These five essential oils were then tested in sponge cake analogues, but the antifungal activity detected was much more limited. CONCLUSION: Five essential oils showed potential antifungal capacity against all species tested, over a wide range of water availability. Their activity, however, seems to be substrate-dependent. More research is needed to make them work in real bakery products, as in the preliminary study limited effectiveness was found. SIGNIFICANCE AND IMPACT OF THE STUDY: The potential of the cinnamon leaf, clove, bay, lemongrass and thyme essential oils against species belonging to Eurotium, Aspergillus and Penicillium genus has been demonstrated.
