Shiga toxin-producing and Enteropathogenic Escherichia coli are foodborne pathogens commonly associated with diarrheal disease in humans. This study investigated the presence of STEC and EPEC in 771 dairy cattle fecal samples which were collected from 5 abattoirs and 9 dairy farms in South Africa. STEC and EPEC were detected, isolated and identified using culture and PCR. Furthermore, 339 STEC and 136 EPEC isolates were characterized by serotype and major virulence genes including stx1, stx2, eaeA and hlyA and the presence of eaeA and bfpA in EPEC. PCR screening of bacterial sweeps which were grown from fecal samples revealed that 42.2% and 23.3% were STEC and EPEC positive, respectively. PCR serotyping of 339 STEC and 136 EPEC isolates revealed 53 different STEC and 19 EPEC serotypes, respectively. The three most frequent STEC serotypes were O82:H8, OgX18:H2, and O157:H7. Only 10% of the isolates were classified as "Top 7" STEC serotypes: O26:H2, 0.3%; O26:H11, 3.2%; O103:H8, 0.6%; and O157:H7, 5.9%. The three most frequent EPEC serotypes were O10:H2, OgN9:H28, and O26:H11. The distribution of major virulence genes among the 339 STEC isolates was as follows: stx1, 72.9%; stx2, 85.7%; eaeA, 13.6% and hlyA, 69.9%. All the 136 EPEC isolates were eaeA-positive but bfpA-negative, while 46.5% carried hlyA. This study revealed that dairy cattle are a major reservoir of STEC and EPEC in South Africa. A coprological survey was conducted to determine the types, prevalence, and intensity of infection of internal parasites in a population of captive African lions (Panthera leo) at a recreational game park in Zimbabwe. Individual fecal samples were collected on three occasions over a 4-month period from each of 30 lions (55%) out of 55 animals held. The samples were examined using flotation and sedimentation techniques to assess the presence and count of parasite eggs, oocysts, and cysts per gram of feces as well as larvae identification. The overall prevalence of helminth infections was 100% (30/30), and 80% (24/30) of fecal samples also were positive for protozoan parasite forms. Eggs of Ancylostoma spp. were found in the feces of 23 (76.7%) lions, Physaloptera sp. in 14 (46.7%) lions, Toxascaris leonina in 13 (43.3%) lions, Toxocara cati in 12 (40%) lions, and Gnathostoma spinigerum and Toxocara canis in 2 (6.7%) lions. Furthermore, eggs of Cylicospirura subequalis, Gnathostoma spp., Lagochilascaris major, Acanthocephalan and Linguatula spp. as well as larvae of Aelurostrongylus sp. were identified in the feces of one lion. Oocysts of five apicomplexan parasites and cysts of one mastigophoran protozoan parasite were recorded, namely, Cystoisospora leonina in 11 (36.7%) lions' feces, Cystoisospora spp. in 9 (30.0%) lions, Cystoisospora felis in 5 (16.7%) lions; Toxoplasma-like spp. in 5 (16.7 %) lions, and Giardia spp. in 8 (26.7%) lions. The majority of lions (28/30) showed mixed infections with different internal parasites, whereas only two animals had single parasite infections. The intensity of infection was relatively low. Some parasite forms observed and identified, such as Eimeria spp. oocysts, were spurious and probably originated from the prey species for the lions. Among the parasites identified were some of zoonotic importance that have health implications for at-risk personnel and visitors who get into contact with the animals.

Toxoplasma gondii is a major neglected parasitic infection occurring in settings of extreme poverty in Africa. Apart from causing reproductive failure in animals it is also a significant zoonotic concern. The objective of this study was to determine the seroprevalence and associated risk factors of T. gondii infection in cats, chickens, goats, sheep and pigs in the southeast of South Africa, of which little is known. Sera was obtained from 601 domestic animals including 109 cats, 137 chickens, 128 goats, 121 sheep and 106 pigs managed under different production systems in different agro-ecological regions and evaluated by the Toxoreagent, a latex agglutination test for T. gondii antibody detection. Household-level and animal-level data were collected by interviewing animal owners and/or herders using a closed-ended questionnaire. The study revealed an overall farm seroprevalence of 83.33% (125/150 farms) with the highest rate of infection for the parasite found in sheep with 64.46% (78/121), followed by goats with 53.91% (69/128), pigs with 33.96% (36/106), cats with 32.11% (35/109 cats) and chickens with 33.58% (46/137). The risk factors that were found to be statistically significant (p 0.05) to different species of seropositivites were age, location, climate, animal production system, rodent control, seropositive cat, cat-feed access and cat faecal disposal. The relatively high seroprevalence of T. gondii detected in this region suggests that domestic animals may pose a substantial public health risk through the consumption of T. gondii-infected raw meat as well as via contact with cat faeces.
