RESUMEN

Obesity has been reported to promote disordered folliculogenesis, but the exact molecular mechanisms are still not fully understood. In this study, we find that miR-133a is involved in obesity-induced follicular development disorder. After feeding with a high-fat diet (HFD) and fructose water for nine weeks, the mouse body weight is significantly increased, accompanied by an inflammatory state and increased expression of miR-133a in the adipose tissues and ovaries as well as accelerated follicle depletion. Although miR-133a is increased in the fat and ovaries of HFD mice, the increased miR-133a in the HFD ovaries is not derived from exosome transferred from obese adipose tissues but is synthesized by ovarian follicular cells in response to HFD-induced inflammation. In vivo experiments show that intrabursal injection of miR-133a agomir induces a decrease in primordial follicles and an increase in antral follicles and atretic follicles, which is similar to HFD-induced abnormal folliculogenesis. Overexpression of miR-133a modestly promotes granulosa cell apoptosis by balancing the expression of anti-apoptotic proteins such as C1QL1 and XIAP and pro-apoptotic proteins such as PTEN. Overall, this study reveals the function of miR-133a in obesity-induced ovarian folliculogenesis dysfunction and sheds light on the etiology of female reproductive disorders.

MAIN CONCLUSION: Identification of NnCER2 and NnCER2-LIKE from Nelumbo nucifera, which are required for the very-long-chain fatty acid elongation, provides new evidence that CER2 proteins are evolutionarily conserved across the eudicots. CER2-LIKE family proteins have been described as core components of the fatty acid elongase complex in Arabidopsis, maize, and rice, having specific function in synthesis of the C30 to C34 fatty acyl-CoA precursors of cuticular waxes. Little is known about the functional conservation in this gene family across species. In this study, two CER2-LIKE family proteins, NnCER2 and NnCER2-LIKE, were characterized from sacred lotus (Nelumbo nucifera), which is an ancient basal eudicot. The transcriptional expression of NnCER2 and NnCER2-LIKE was found in floating leaf blades, emergent petioles and vertical leaves, petals, and anthers. The NnCER2 and NnCER2-LIKE proteins were localized to the endoplasmic reticulum and nucleus. Overexpressing NnCER2 and NnCER2-LIKE in Arabidopsis led to alteration of cuticle wax structure in inflorescence stems, and this was associated with elevated 30, 32, and 34 carbon length wax compounds, and their derivatives. The different substrate specificities of NnCER2 and NnCER2-LIKE were explored using co-expression with AtCER6 in yeast cells. These findings provide clear evidence that the function of CER2 family proteins in producing VLCFAs is highly conserved across the eudicots.

Plants employ metal tolerance proteins (MTPs) to confer tolerance by sequestering excess ions into vacuoles. MTPs belong to the cation diffusion facilitator (CDF) family, which facilitates the transport of divalent transition metal cations. In this study, we conducted a comprehensive analysis of the MTP gene families across 21 plant species, including maize (Zea mays). A total of 247 MTP genes were identified within these plant genomes and categorized into distinct subgroups, namely Zn-CDF, Mn-CDF, and Fe/Zn-CDF, based on phylogenetic analyses. This investigation encompassed the characterization of genomic distribution, gene structures, cis-regulatory elements, collinearity relationships, and gene ontology functions associated with MTPs. Transcriptomic analyses unveiled stress-specific expression patterns of MTP genes under various abiotic stresses. Moreover, quantitative RT-PCR assays were employed to assess maize MTP gene responses to diverse heavy metal stress conditions. Functional validation of metal tolerance roles was achieved through heterologous expression in yeast. This integrated evolutionary scrutiny of MTP families in cereals furnishes a valuable framework for the elucidation of MTP functions in subsequent studies. Notably, the prioritized MTP gene ZmMTP6 emerged as a positive regulator of plant Cd tolerance, thereby offering a pivotal genetic asset for the development of Cd-tolerant crops, particularly maize cultivars.

Microsatellites (simple sequence repeats, SSRs) are co-dominant nuclear markers that are widely used in population genetic studies. Population genetic parameters from different studies might be significantly influenced by differences in marker number. In our study, 265 sequences with polymorphic microsatellites were obtained from SLAF-seq data. Then, subpopulations containing different numbers (5, 6, 7,…, 15, 20, 25, 30, 35, 40) of markers were genotyped 10 times to investigate the impact of marker numbers on population genetic diversity results. Our results show that genotyping with less than 11 or 12 microsatellite markers lead to significant deviations in the population genetic diversity or genetic structure results. In order to provide markers for population genetic and conservation studies for Rhododendron, 26 SSR primers were designed and validated in three species.

The substitution of chemical pesticides by biopesticides is crucial to ensure the quality of agricultural products and to foster environmental sustainability. This study takes the willingness and the behaviors of rice farmers on the application of biopesticides as the research object. The survey questionnaire was designed based on the theory of rational small-scale farmers from three aspects: "individual and family characteristics of farmers", "cognition of farmers" and "external factors". The survey was then conducted on 163 rice farmers in seven prefecture-level cities in Jilin Province of China. The logistic model was used to analyze the influencing factors resulting in the deviation of the behaviors of the rice farmers from their initial willingness on the application of biopesticides. The explanatory structure model (ISM) was used to analyze the logical hierarchical relationship among various influencing factors. The results show that: (1) For 45% of the farmers surveyed, there's a deviation between their willingness and behaviors regarding the application of biopesticides; (2) Among the significant factors leading to the deviation between farmers' willingness and behaviors concerning the application of biopesticides, the surface-level direct factor is biopesticide awareness. The mid-level indirect factors are agricultural product quality and safety awareness and the deep-level root cause is farmers' education level. (3) The primary reason for the deviation of the farmers' behaviors from their willingness is their lack of knowledge about biopesticides and the biopesticides' incomplete market structure. Based on the comprehensive analysis, it is recommended to improve the professionalization of the farmers, to strengthen the publicity of green production and to accelerate the formulation of the biopesticides market to further promote the usage of biopesticides.

In the past 30 years, China's industrialization level has developed rapidly, and agricultural green development (AGD) is facing severe challenges. The research on the temporal and spatial features of the coupling and coordination of industrialization and agricultural green development is a key issue to promote the sustainable development of agriculture. This paper takes China's industrialization and AGD level as the research object, and uses panel data from 31 provinces in China from 1990 to 2019 to construct an evaluation index system for industrialization and AGD. This paper uses the coupling coordination degree model and spatial autocorrelation analysis method to calculate, test and analyze the temporal and spatial features of the coupling coordination level of industrialization and AGD. The results show that: this paper compares the industrialization and AGD levels during the study period and finds that China's industrialization development level is on the rise as a whole, and the AGD level shows a downward trend first and then rises later. Through the horizontal comparison of different regions, this paper finds that there is a large regional imbalance in the level of industrialization and AGD. The coupling and coordination level of industrialization and AGD has always been primary. From the time point of view, coupling coordination degree shows a trend of first decline and then rise. From a spatial point of view, coupling coordination degree varies greatly among provinces, with the eastern, central and western regions decreasing successively. The level of coupling coordination has obvious positive autocorrelation in spatial distribution, and presents significant spatial agglomeration characteristics in space. The research results can provide a theoretical basis for regionally differentiated governance of the coordinated development of industrialization and AGD, and promote coordinated development.

Understanding the processes of divergence and speciation is an important task for evolutionary research, and climate oscillations play a pivotal role. We estimated the genetic structure and demographic history of two closely related species of Rhododendron, R. dauricum, and R. mucronulatum, distributed in northeastern China using 664,406 single nucleotide polymorphic loci of specific-locus amplified fragment sequencing (SLAF-seq) and 4 chloroplast DNA (cpDNA) fragments, sampling 376 individuals from 39 populations of these two species across their geographic distributions. The geographical distribution of cpDNA haplotypes revealed that R. dauricum and R. mucronulatum have different spatial genetic structures and haplotype diversity. Analysis of molecular variance (AMOVA) results showed that these two species have significant genetic differentiation and that the phylogeny demonstrates that these two species clustered a monophyletic group based on SLAF data, respectively, but not in cpDNA data. The evidence of significant gene flow was also detected from R. mucronulatum to R. dauricum. A deep divergence between the two species was observed and occurred during the early Oligocene. The niche models showed that the two species have different demographic histories. Thus, our results imply that geography and climate changes played important roles in the evolutionary process of R. dauricum and R. mucronulatum, and although there was an interspecific gene flow, the divergence was maintained by natural selection.
