Anthraquinone is a redox mediator that can effectively catalyze the degradation of azo dyes by promoting the electron transfer. In this study, a mediator membrane with poly (vinylidene fluoride) (PVDF) as the membrane support and 1,8-dichloroanthraquinone (1,8-AQ) and graphene oxide (GO) as the additives was prepared and characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), mercury intrusion porosimetry (MIP), atomic force microscopy (AFM) and water contact angle. The introduction of GO increases the pure water flux of the membrane to 258.56±12.93 L/(m2·h). Its catalytic performances for the biodegradation of azo dyes were evaluated. Under the optimized conditions, the 1,8-AQ/GO/PVDF composite membrane is able to improve the dye degradation efficiency 2.2 times for reactive red X-3B and 1.1 times for acid red B, as compared with PVDF membrane. In addition, the mediator membrane maintains stable and high catalytic efficiency in the cyclic test and over 90 % dye degradation efficiency is still obtained after 5 cycles of decolorization. These results suggest the great application potentials of the 1,8-AQ/GO/PVDF membrane in the dye wastewater treatment.

Stress granules (SGs) are dynamic, membrane-less organelles. With their formation and disassembly processes characterized, it remains elusive how compositional transitions are coordinated during prolonged stress to meet changing functional needs. Here, using time-resolved proteomic profiling of the acute to prolonged heat-shock SG life cycle, we identify dynamic SG proteins, further segregated into early and late proteins. Comparison of different groups of SG proteins suggests that their biochemical properties help coordinate SG compositional and functional transitions. In particular, early proteins, with high phase-separation-propensity, drive the rapid formation of the initial SG platform, while late proteins are subsequently recruited as discrete modules to further functionalize SGs. This model, supported by immunoblotting and immunofluorescence imaging, provides a conceptual framework for the compositional transitions throughout the acute to prolonged SG life cycle. Additionally, an early SG constituent, non-muscle myosin II, is shown to promote SG formation by increasing SG fusion, underscoring the strength of this dataset in revealing the complexity of SG regulation.

Eukaryotic stress granules (SGs) are highly dynamic assemblies of untranslated mRNAs and proteins that form through liquid-liquid phase separation (LLPS) under cellular stress. SG formation and elimination process is a conserved cellular strategy to promote cell survival, although the precise regulation of this process is poorly understood. Here, we screened six E3 ubiquitin ligases present in SGs and identified TRIM21 (tripartite motif containing 21) as a central regulator of SG homeostasis that is highly enriched in SGs of cells under arsenite-induced oxidative stress. Knockdown of TRIM21 promotes SG formation whereas overexpression of TRIM21 inhibits the formation of physiological and pathological SGs associated with neurodegenerative diseases. TRIM21 catalyzes K63-linked ubiquitination of the SG core protein, G3BP1 (G3BP stress granule assembly factor 1), and G3BP1 ubiquitination can effectively inhibit LLPS, in vitro. Recent reports suggested the involvement of macroautophagy/autophagy, as a stress response pathway, in the regulation of SG homeostasis. We systematically investigated well-defined autophagy receptors and identified SQSTM1/p62 (sequestosome 1) and CALCOCO2/NDP52 (calcium binding and coiled-coil domain 2) as the primary receptors that directly interact with G3BP1 during arsenite-induced stress. Endogenous SQSTM1 and CALCOCO2 localize to the periphery of SGs under oxidative stress and mediate SG elimination, as single knockout of each receptor causes accumulation of physiological and pathological SGs. Solid-state nuclear magnetic resonance (SSNMR), a technique capable of studying solid or semisolid biological samples, was first applied to study the cell differentiation and mineralization using the whole-cell sample. Mesenchymal stromal cells (MSCs) with multipotent differentiation capacity were induced to differentiate into osteoblasts. The whole differentiation process, osteoblast mineralization and the mineral maturation, was investigated using SSNMR, providing intact, atomic level information on the cellular mineral structural transformation. Our research indicated the extent of osteoblast mineralization could vary significantly for different cell populations whereas the difference was not easily shown by other means of characterization. The SSNMR spectra revealed hydroxylapatite (or hydroxyapatite [HAP]) formation around 2 to 4 weeks after osteogenic induction for MSCs with a high differentiation potency. The early mineral phase deposit before HAP formation contained a high amount of HPO4 2-. The structures of minerals in the extracellular matrix (ECM) of osteoblasts could evolve for a period of time, even after the incubation of cells has been stopped. This observation was only possible by studying the sample in an intact state, where ECM was not disturbed. These findings improved our understanding of MSCs, which had wide applications in bone regeneration and tissue engineering. Meanwhile, this work demonstrated the advantage of studying these cellular systems as a whole without any mineral extraction, which had been largely overlooked. © 2022 The Authors. In mid-May 2008 a serious green tide caused mainly by floating Ulva prolifera (Müller) J. Agardh (Chlorophyta, Ulvales) thalli struck the coastal area of Qingdao, China. To understand the present physiological conditions of the floating alga, in this work both laboratory and field investigations were conducted on the floating U. prolifera thalli in comparison with the attached U. prolifera thalli collected from the area. The floating thalli of three distinctively different colors and attached thalli at three different stages of sporangium formation process were characterized under a microscope, while their photosynthetic parameters were determined with chlorophyll fluorescence technology. On the other hand, the sporangium formation status of the floating U. prolifera thalli was surveyed both in the laboratory and in the field. Comparisons showed that both of the paired morphological characteristics and the paired physiological parameters of the floating and attached U. prolifera thalli were consistent. Furthermore, some spores were confirmed in the field and some motile particles were found within the floating thalli. These results suggest that the floating U. prolifera thalli with different colors could be at different stages of sporangium formation. However, our results also showed that the floating alga thalli have only a limited reproductive potential. This might limit the duration and the further geographic expansion of the green algal bloom.

To develop an economic and sustainable biochar application strategy for mitigating methane emission from paddy fields, a four-year field experiment was conducted to compare two biochar amendment methods. The annual low (AL) rate pyrolyzed biochar returning method used the same amount of biochar as was harvested from rice straw in the field, 2.8 t ha-1 yr-1. The high single (HS) biochar returning method consisted of a single application of 22.5 t ha-1 biochar only in the first year, 2015. Our results showed that the AL biochar returning strategy prevailed over the HS strategy in mitigating methane emission from paddy fields. On average, AL and HS could reduce methane emissions by 41% and 38.25% in four years, respectively. Methane accumulation per unit rice production was 45.8% and 43.1% in AL and HS, respectively. AL showed a stable effect on mitigating methane emission over four successive years, which resulted from the continuously increasing methanotrophs due to annual fresh biochar application. Aged biochar weakened the promotion of methanotrophs, leading to lower methane reduction rates in HS than in AL in the 4 years. Our results indicate that AL is a highly sustainable strategy for methane mitigation in paddy fields due to its high efficiency, practical operation, and economical acceptance.

The effects of an on-site biomass (rice straw) equivalent biochar-returning strategy (RSC) on rice yield, soil nutrients and bacterial community composition were examined in a four-year field trial (2013-2016) conducted in a paddy field in south China. Three treatments were set up including annual on-site biomass return (RS, rice straw at 8 t ha-1 yr-1), annual on-site biomass equivalent biochar-return (RSC, rice straw biochar at 2.8 t ha-1 yr-1 with a 35 % carbonization rate) and control (CK, no rice straw or biochar added). Results showed that a low rate of biochar application (RSC) could significantly increase rice yield in four successive years. The increase in rice yield was mainly attributed to the increase in soil potassium and magnesium contents resulting from the presence of the unique surface functional groups of biochar. As a result of biochar amendment, soil bacterial cooperative relationships were improved in the RSC, compared to those in the RS treatment. Our study indicated that RSC might be promoted as a promising strategy to enhance rice productivity and soil fertility in a sustainable way.

Biochar application to soil has attracted extensive attention worldwide due to its carbon (C) sequestration and fertility-enhancing properties. However, the lack of biochar accumulation in highly disturbed agroecosystems challenges the perceived long-term stability of biochars in soil. This 9-year field experiment was conducted in rice paddy fields to understand the temporal degradation of biochars produced from two contrasting feedstocks (rice straw vs. bamboo) at a high temperature (600 °C). Obvious physical alterations, surface oxidation, and transformation of condensed aromatic C occurred in biochars in the disturbed paddy field with frequent redox cycles. Increase in O/C atomic ratio, levels of high-temperature-sensitive degradable components, H/C ratio, and linear alkyl-C content were observed, which were indicative of time-dependent molecular changes and degradative transformation of biochars. Biochar degradation was characterized by the loss of labile C at an early stage and the degradation of aromatic C at a later stage. Based on the massive loss of C content in biochars (10.3-11.8%) and considerable degradation of aromatic C (5.0-8.7%) in 9 years, we argue that current biphasic C dynamic models probably overestimate the stability of biochars in agroecosystems such as rice paddy fields. Long-term field experiments (>5 years) are required to assess biochar's potential for C sequestration. This study provides long-term field data regarding the temporal changes in biochar physicochemical properties, which may facilitate the development of a robust assessment scheme on the long-term persistence of biochars in agroecosystems.

A sustainable biochar strategies on increasing crop yield and mitigating CH4 emissions over successive years is unknown. Thus, on-site equivalent rice straw biochar-returning (ERSC, biochar at 2.8 t ha-1 annual) were compared with on-site equivalent rice straw- returning (RS, rice straw at 8 t ha-1 annual) and high application rate biochar-returning (RSCH, biochar at 22.5 t ha-1 only in the first year). The RS and RSCH treatments increased rice production by 10.1% and 11.8% on average, respectively. The ERSC treatment continually increased rice production by 8.0%, 1.6% and 7.3% in three successive years. The ERSC treatment had a cumulative effect on the soil nutrients phosphorus (P), potassium (K), and magnesium (Mg), as well as increasing total carbon (TC) and total nitrogen (TN) and continuously reducing the effect of soil available aluminum (Al). The RS treatment significantly promoted CH4 emissions while the ERSC treatment reduced methane emissions by 43%, 31% and 30% and the RSCH treatment reduced methane emissions by 52%, 22% and14% in three successive years. Compared with RSCH, ERSC showed the best long-term stable effect on methane emission mitigation in three successive years. This might result from the fact that fresh biochar promoted anaerobic oxidation of methane. This research gives us scientific evidence that an on-site equivalent rice straw biochar-returning strategy may be a promising method for sustaining rice production and mitigating methane emissions.

Ulva prolifera, a common green seaweed, is one of the causative species of green tides that occurred frequently along the shores of Qingdao in 2008 and had detrimental effects on the preparations for the 2008 Beijing Olympic Games sailing competition, since more than 30 percent of the area of the games was invaded. In view of the rapid accumulation of the vast biomass of floating U. prolifera in green tides, we investigated the formation of sporangia in disks of different diameters excised from U. prolifera, changes of the photosynthetic properties of cells during sporangia formation, and development of spores. The results suggested that disks less than 1.00 mm in diameter were optimal for the formation of sporangia, but there was a small amount of spore release in these. The highest percentage of area of spore release occurred in disks that were 2.50 mm in diameter. In contrast, sporangia were formed only at the cut edges of larger disks (3.00 mm, 3.50 mm, and 4.00 mm in diameter). Additionally, the majority of spores liberated from the disks appeared vigorous and developed successfully into new individuals. These results implied that fragments of the appropriate size from the U. prolifera thalli broken by a variety of factors via producing spores gave rise to the rapid proliferation of the seaweed under field conditions, which may be one of the most important factors to the rapid accumulation of the vast biomass of U. prolifera in the green tide that occurred in Qingdao, 2008.
