RESUMEN

The Lebranche mullet Mugil liza is a marine fish of great importance for artisanal and industrial fishing, as well as aquaculture. The use of live feeds during the larviculture phase of marine fish is a significant component of production costs. The present study evaluated the effects of the feeding transition on different larval stages of M. liza, from the combined supply of live feed (Brachionus rotundiformis + Artemia franciscana) and live + inert feed (Artemia franciscana + inert diet) until the weaning phase to only the inert diet. A total of 3240 M. liza larvae (weight 0.0133 ± 0.0062 g and length 0.793 ± 0.160 cm) were distributed among the 12 experimental units (n = 270), resulting in four groups with three replicates each. Treatment groups consisted of feed transition with A. franciscana (enriched metanauplii) to commercial inert feed starting weaning at four different larval ages: 28, 31, 34, and 37 days post hatching. Zootechnical performance indexes and intestinal histomorphometry were evaluated. Mortality, condition factor, and length variation coefficient did not show significant differences between treatments. Final weight, final length, weight gain, and length gain were significantly greater in larvae that started weaning at 31, 34, and 37 days post hatching. Weight coefficient of variation was significantly higher in larvae that started weaning at 28 days (67.51 ± 11.70) compared to 37 days (34.40 ± 7.30). In intestinal histology, villi height (180.3 ± 4.4) was significantly higher in larvae that started weaning at 37 days post hatching. Considering the evidence found in the present study, it is recommended to start weaning M. liza on the 31st day post-hatching, using a 2-day co-feeding protocol (31st and 32nd days). From the 33rd day after hatching, M. liza larvae can receive only commercial feed.

The aim of the present study was to evaluate the effects of taurine on endoplasmic reticulum stress, inflammatory cytokine expression and mitochondrial oxidative stress induced by high glucose in primary cultured muscle cells of olive flounder (Paralichthys olivaceus). Three experimental groups were designed as follows: muscle cells of olive flounder incubated with three kinds of medium containing 5 mM glucose (control), 33 mM glucose (HG) or 33 mM glucose + 10 mM taurine (HG + T), respectively. Results showed that taurine addition significantly alleviated the decreased activity of superoxide dismutase (SOD) and the ratio of reduced to oxidized glutathione (GSH/GSSG) induced by high glucose. The increase of cellular reactive oxygen species (ROS), malondialdehyde content and cell apoptosis induced by high glucose were alleviated by taurine. Besides, gene expression of glucose-regulated protein 78, PKR-like ER kinase, tumor necrosis factor-α, interleukin-6, interleukin-1ß, interleukin-8, muscle atrophy F-box protein and muscle RING-finger protein 1 were significantly up-regulated in the HG group, and taurine addition decreased the expression of these genes. High glucose led to the swelling of the endoplasmic reticulum (ER). Meanwhile, the nuclear translocation of nuclear factor κB (NF-κB) and the release of cytochrome C from mitochondria induced by high glucose were suppressed by taurine addition. These results demonstrated that taurine alleviated ERS, inflammation and mitochondrial oxidative stress induced by high glucose in olive flounder muscle cells. The ROS production, NF-κB signaling pathway and mitochondria function were the main targets of the biological effects of taurine under high glucose condition.

Artificial breeding of small yellow croaker (Larimichthys polyactis) was recently achieved, providing a bright future for its commercial farming. In May 2019, a disease outbreak occurred among small yellow croakers in an aquaculture farm near Xiangshan Bay, charactering by white spots spotted on the surface of fish skin, gills and fins. The parasite was preliminarily identified as Cryptocaryon irritans based on morphological feature of the parasite and the symptoms on fish. However, the previously published specific primer pairs failed to confirm the existence of C. iriitans. Six nucleotides mismatches were discovered after mapping specific forward primer back to targeted gene. Therefore, an updated PCR specific primer was developed within the 9th highly variable region of 18S rRNA gene and conserved in all C. irritans sequences available in GenBank database. The specificity was verified in silico by Primer-BLAST against GenBank nucleotide. Laboratory cultured ciliates (Mesanophrys, Pseudokeronopsis and Uronema) as well as natural microbial community samples collected from sea water and river water was used as negative control to verify the specificity of the primer in situ. Besides, tank transfer method was used to evaluate the treatment of the parasite infection. By tank transfer method, 2.00 ± 0.61 out of 10 fish that already sever infected were successfully survived after 8 days treatment, meanwhile the control group died out at d 6. More loss to the treatment group during first five days was observed and may attribute to the combined effect from infection and stress the recent domesticated fish suffered during rotation. Therefore, tank transfer method was also effective to prevent small yellow croaker from further infection, however the loss of the small yellow croaker suffered from stress during rotation also needs to be carefully concerned. In conclusion, this study reported the first diagnose of C. irritans infection on small yellow croaker, provided updated specific primer to detect C. irritans infection on fish body and reported the effect of tank transfer on small yellow croaker treatment.

Antibiotic resistance represents a global health crisis for humans, animals, and for the environment. Transmission of antibiotic resistance through environmental pathways is a cause of concern. In this study, quantitative PCR and culture-dependent bacteriological methods were used to detect the abundance of antibiotic resistance genes (ARGs) and the quantity of culturable heterotrophic antibiotic-resistant bacteria (ARB) in marine fish farming areas. The results indicated that sul and tet family genes were widely distributed in marine fish farming areas of Hainan during both rearing and harvesting periods. Specifically, sul1 and tetB were the most dominant ARGs. The total abundance of ARGs increased significantly from the rearing to the harvesting period. A total of 715 ARB strains were classified into 24 genera, within these genera Vibrio, Acinetobacter, Pseudoalteromonas, and Alteromonas are opportunistic pathogens. High bacterial resistance rate to oxytetracycline (OT) was observed. The numbers of OT- and enrofloxacin-resistant bacteria dropped significantly from rearing the period to the harvesting. The co-occurrence pattern showed that Ruegeria and tetB could be indicators of ARB and ARGs, respectively, which were found in the same module. Redundancy analysis indicated that salinity was positively correlated with the most dominant ARB, and was negatively correlated with the most dominant ARGs. These findings demonstrated the prevalence and persistence of ARGs and ARB in marine fish farming areas in China.

ABSTRACT: Essential oils of plants whose main compound is citral showed beneficial effects when added to fish feed. The objective of the present study was to evaluate the dietary effect of the addition of citral on zootechnical parameters and digestive enzyme activities of Centropomus undecimalis. Juveniles were fed for 45 days with diets containing different amounts of citral (0.0 - control, 0.5, 1.0, and 2.0 mL per kg of diet). The water quality parameters were kept stable during the experiment and no mortality was observed. At the end of the experimental period, the treatment 0.5 mL citral per kg of diet had the lowest weight gain and specific growth rate, and the highest feed conversion, while the same parameters did not differ between the other treatments. Pepsin activity was higher in the stomach of fish fed with 0.5 mL citral per kg of diet and amylase activity was higher in the intestine of fish fed with 0.5 and 2.0 mL citral per kg of diet compared to the control group. Intestinal lipase activity was higher in all groups that were fed with citral compared to the control group. Chymotrypsin and trypsin activities showed no difference between groups. Consequently, dietary addition of citral at any of the levels tested is not recommended for common snook.


RESUMO: Óleos essenciais de plantas, cujo composto majoritário é o citral, mostraram efeitos benéficos quando adicionados à ração para peixes. O objetivo do presente estudo foi avaliar o efeito da adição de citral sobre os parâmetros zootécnicos e atividades das enzimas digestivas de Centropomus undecimalis. Os juvenis foram alimentados por 45 dias com dietas contendo diferentes quantidades de citral (0,0 - controle, 0,5, 1,0 e 2,0 mL por kg de ração). Os parâmetros de qualidade da água foram mantidos estáveis ​​durante o experimento e nenhuma mortalidade foi observada. Ao final do período experimental, o tratamento 0,5 mL citral por kg de ração teve o menor ganho de peso e taxa de crescimento específico e a maior conversão alimentar, enquanto os mesmos parâmetros não diferiram entre os demais tratamentos. A atividade da pepsina foi maior no estômago de peixes alimentados com 0,5 mL de citral por kg de ração e a atividade de amilase foi maior no intestino de peixes alimentados com 0,5 e 2,0 mL citral por kg de ração comparado ao grupo controle. A atividade da lipase intestinal foi maior em todos os grupos que foram alimentados com citral, em comparação ao grupo controle. As atividades de quimotripsina e tripsina não mostraram diferença entre os grupos. Consequentemente, a adição de citral na ração em qualquer um dos níveis testados não é recomendada para robalo flecha.

O desempenho reprodutivo de exemplares selvagens de beijupirá capturados no litoral de Pernambuco e aclimatados ao cativeiro foi avaliado durante 10 meses. Um plantel composto por duas fêmeas (peso de 14 a 16kg) e quatro machos (cerca de 12kg) mantidos em um único tanque de 70t produziu 48,7 milhões de ovos no período de outubro a junho. As fêmeas foram capazes de desovar 21 vezes, com uma frequência média (±DP) de uma desova a cada 11,9 dias (±2,9). Em média, cada desova produziu 2,4 milhões de ovos, com fertilização de 52,4 por cento, o que resultou em aproximadamente 1,0 milhão de larvas por desova. Os resultados demonstram a viabilidade da metodologia empregada na formação do plantel e no manejo de reprodutores e confirmam a relativa facilidade de obtenção de desovas espontâneas do beijupirá em cativeiro. Também foi confirmado que o período reprodutivo dessa espécie em cativeiro se estende além do observado na natureza. Este é o primeiro relato sobre o desempenho reprodutivo do beijupirá em cativeiro no Brasil...


The reproductive performance of wild cobia caught off the coast of Pernambuco, northeastern Brazil, was assessed. Six breeders (two females with 14-16kg and four males with approximately 12kg each) were maintained in a 70 ton tank from October to June and produced 48.7 million eggs. Females were able to spawn every 11.9 days (±2.9), which resulted in 21 spawns. On average each spawn produced 2.4 million eggs with a fertilization of 52.4 percent, which resulted in approximately 1.0 million larvae per spawn. The present results demonstrate the feasibility of the methodology employed here in the formation and management of a cobia breeding stock and confirm the ease of spawning cobia in captivity. It was also confirmed that the spawning season may be extended under captivity when compared to the wild stock. This is the first report on the reproductive performance of cobia in captivity in Brazil...

A substituição do óleo de peixe em dietas para beijupirá (Rachycentron canadum) foi avaliada usando-se cinco proporções de óleo de peixe e soja. Duzentos juvenis (±12g) foram distribuídos aleatoriamente em 20 tanques e alimentados até a saciedade aparente durante 42 dias. À medida que se aumentou o teor de óleo de peixe nas dietas, houve aumento proporcional no desempenho. Os juvenis alimentados com dietas que continham 75 e 100 por cento de óleo de peixe apresentaram ganhos de peso mais altos que os das dietas com 0 e 25 por cento de óleo de peixe, enquanto os alimentados com a dieta com 50 por cento de óleo de peixe apresentaram crescimento intermediário. Não foram observadas diferenças nas concentrações de proteína e lipídio na carcaça dos peixes, mas os teores de umidade e cinzas diferiram entre os tratamentos. A inclusão de teores relativamente altos de ingredientes de origem vegetal diminuiu o consumo alimentar, o que afetou negativamente o crescimento. Os resultados indicam a possibilidade de substituição de até 50 por cento do óleo de peixe por óleo de soja em dietas para juvenis do beijupirá.


The replacement of fish oil for cobia (Rachycentron canadum) in diets was evaluated using five proportions of fish oil:soybean oil. Two hundred juveniles (±12g) were randomly distributed in twenty 480L tanks and fed to apparent satiety twice daily for 42 days. Diets contained 12 percent lipids with different proportions of fish to soybean oils and were therefore named OP0, OP25, OP50, OP75 and OP100 according to the inclusion content of fish oil (0, 25, 50, 75 and 100 percent, respectively). Increasing dietary fish oil levels resulted in higher survival, growth and feed intake. Fish fed diets OP75 and OP100 presented higher weight gain than those fed diets OP0 and OP25, whereas fish fed diet OP50 had an intermediate weight gain. Overall, performance was enhanced at higher contents of dietary fish oil. No significant differences on protein and lipid content of fish carcass were observed, but moisture and ash content differed significantly between treatments. The inclusion of relatively higher contents of plant ingredients affected feed intake, which resulted in lower growth rates. The present results suggest the possibility of replacing up to 50 percent of fish oil with soybean oil in diets for cobia juveniles.
