RESUMO

Herein, smart coatings based on photo-responsive polymer nanocapsules (NC) and deposited by laser evaporation are presented. These systems combine remotely controllable release and high encapsulation efficiency of nanoparticles with the easy handling and safety of macroscopic substrates. In particular, azobenzene-based NC loaded with active molecules (thyme oil and coumarin 6) were deposited through Matrix-Assisted Pulsed Laser Evaporation (MAPLE) on flat inorganic (KBr) and organic (polyethylene, PE) and 3D (acrylate-based micro-needle array) substrates. SEM analyses highlighted the versatility and performance of MAPLE in the fabrication of the designed smart coatings. DLS analyses, performed on both MAPLE- and drop casting-deposited NC, demonstrated the remarkable adhesion achieved with MAPLE. Finally, thyme oil and coumarin 6 release experiments further demonstrated that MAPLE is a promising technique for the realization of photo-responsive coatings on various substrates.

Doping of biocompatible nanomaterials with magnetic phases is currently one of the most promising strategies for the development of advanced magnetic biomaterials. However, especially in the case of iron-doped magnetic hydroxyapatites, it is not clear if the magnetic features come merely from the magnetic phases/ions used as dopants or from complex mechanisms involving interactions at the nanoscale. Here, we report an extensive chemical-physical and magnetic investigation of three hydroxyapatite nanocrystals doped with different iron species and containing small or no amounts of maghemite as a secondary phase. The association of several investigation techniques such as X-ray absorption spectroscopy, Mössbauer, magnetometry, and TEM allowed us to determine that the unusual magnetic properties of Fe2+/3+-doped hydroxyapatites (FeHA) occur by a synergy of two different phenomena: i.e., (i) interacting superparamagnetism due to the interplay between iron-doped apatite and iron oxide nanoparticles as well as to the occurrence of dipolar interactions and (ii) interacting paramagnetism due to Fe3+ ions present in the superficial hydrated layer of the apatite nanophase and, to a lesser extent, paramagnetism due to isolated Fe3+ ions in the apatite lattice. We also show that a major player in the activation of the above phenomena is the oxidation of Fe2+ into Fe3+, as induced by the synthesis process, and their consequent specific positioning in the FeHA structure.

Candida rugosa lipase (CRL) was deposited by matrix assisted pulsed laser evaporation (MAPLE) in order to immobilize the enzyme with a preserved native conformation, which ensures its catalytic functionality. For this purpose, the composition of the MAPLE target was optimized by adding the oil phase pentane to a water solution of the amino acid 3-(3,4-dihydroxyphenyl)-2-methyl-l-alanine (m-DOPA), giving a target formed by a frozen water-lipase-pentane microemulsion. Fourier transform infrared (FTIR) spectroscopy and atomic force microscopy (AFM) were used to investigate the structure of MAPLE deposited lipase films. FTIR deconvolution of amide I band indicated a reduction of unfolding and aggregation, i.e., a better preserved lipase secondary structure in the sample deposited from the frozen microemulsion target. AFM images highlighted the absence of big aggregates on the surface of the sample. The functionality of the immobilized enzyme to promote transesterification was determined by thin layer chromatography, resulting in a modified specificity.

This paper presents the results of several experimental methods (FT-IR spectroscopy, UV-vis spectroscopy, fluorescence microscopy (FM), Atomic Force Microscopy (AFM)) evidencing structural changes induced in extremely diluted solutions (EDS), which are prepared by an iterated process of centesimal (1:100) dilution and succussion (shaking). The iteration is repeated until an extremely high dilution is reached, so that the composition of the solution becomes identical to that of the solvent--in this case water--used to prepare it. The experimental observations reveal the presence of supramolecular aggregates hundreds of nanometres in size in EDS at ambient pressure and temperature, and in the solid state. These findings confirm the hypothesis--developed thanks to previous physico-chemical investigations--that formation of water aggregates occurs in EDS. The experimental data can be analyzed and interpreted with reference to the thermodynamics of far-from-equilibrium systems and irreversible processes.

Bioinspired strategies for scaffold design and optimization were improved by the introduction of Additive Manufacturing (AM), thus allowing for replicating and reproducing complex shapes and structures in a reliable manner, adopting different kinds of polymeric and nanocomposite materials properly combined according to the features of the natural host tissues. Benefiting from recent findings in AM, a Matrix-Assisted Pulsed Laser Evaporation (MAPLE) technique was employed for obtaining graphene-like material (GL) uniform coatings on 3D scaffolds for tissue repair strategies, towards the development of a new concept 3D scaffold with controlled morphological/architectural and surface features and mechanical and biological properties. The effect of the material-design combination through an integrated technological approach (i.e., MAPLE deposition of GL on 3D AM PCL scaffolds) was assessed through scanning electron microscopy, atomic force microscopy, contact angle measurements, mechanical measurements and biological analyses (cell viability assay and alkaline phosphatase activity) in conjunction with confocal laser scanning microscopy. The differentiation of hMSCs towards the osteoblast phenotype was also investigated analysing the gene expression profile. The obtained findings provided a further insight into the development of improved strategies for the functionalization or combination of GL with other materials and 3D structures in a hybrid fashion for ensuring a tighter adhesion onto the substrates, improving cell fate over time, without negatively altering the mechanical properties and behaviour of the neat constructs. In particular, the results provided interesting information, making 3D AM GL-coated scaffolds potential candidates for bone tissue engineering.

This work reports the production of biocompatible thin layers for biomedical applications based on a graphene-like material (GL), a graphene-related material (GRM) obtained from carbon black. GL was combined in a hybrid fashion with polydopamine (pDA), a mussel-inspired water-resistant wet adhesive bonding obtained by the oxidative polymerization of dopamine (DA), and polyvinyl pyrrolidinone (PVP), a nontoxic synthetic polymer with intrinsic adhesion properties, to obtain a tighter adhesion of the thin layer to the substrate (silicone slices). Matrix-assisted pulsed laser evaporation (MAPLE) was used to coat PDMS slices with thin films of GL-pDA and GL-PVP directly from their frozen suspensions in water. The results indicate that the relevant chemical-physical characteristics of both thin films (evidenced by FTIR and AFM) were maintained after MAPLE deposition and that the films exhibit uniformity also at the nanometric level. After deposition, the GL-pDA and GL-PVP films underwent a biological survey toward murine fibroblasts (NIH3T3), human keratinocytes (HaCAT), and human cervical adenocarcinoma epithelial-like (HeLa) cells to assess the feasibility of this approach. Results indicate that both the GL-pDA and GL-PVP films did not perturb the biological parameters evaluated, including cytoskeleton alterations. Both hybrid films enhanced the effects of GL on cellular vitality across all cell lines. Specifically, the GL-pDA film exhibited a more stable effect over time (up to 72 h), whereas the GL-PVP film behaved similarly to the GL film in NIH3T3 and HeLa cell lines after long-term exposure. These promising results make the GL-pDA and GL-PVP films potential candidates for the manufacture of coated flexible devices for biomedical applications.

Combining magnetic nanoparticles (MNPs) with high-voltage processes to produce ultra-thin magnetic nanofibers (MNFs) fosters the development of next-generation technologies. In this study, polycarbonate urethane nanofibers incorporating magnetic particles were produced via the electrospinning technique. Two distinct types of magnetic payload were used: (a) iron oxide nanoparticles (IONPs) with an average size and polydispersity index of 7.2 nm and 3.3%, respectively; (b) nickel particles (NiPs) exhibiting a bimodal size distribution with average sizes of 129 nanometers and 600 nanometers, respectively, and corresponding polydispersity indexes of 27.8% and 3.9%. Due to varying particle sizes, significant differences were observed in their aggregation and distribution within the nanofibers. Further, the magnetic response of the IONP and/or NiP-loaded fiber mats was consistent with their morphology and polydispersity index. In the case of IONPs, the remanence ratio (Mr/Ms) and the coercive field (Hc) were found to be zero, which agrees with their superparamagnetic behavior when the average size is smaller than 20-30 nm. However, the NiPs show Mr/Ms = 22% with a coercive field of 0.2kOe as expected for particles in a single or pseudo-single domain state interacting with each other via dipolar interaction. We conclude that magnetic properties can be modulated by controlling the average size and polydispersity index of the magnetic particles embedded in fiber mats to design magneto-active systems suitable for different applications (i.e., wound healing and drug delivery).

In this study, matrix-assisted pulsed laser evaporation (MAPLE) was used to deposit graphene-like materials (GL), a new class of biocompatible graphene-related materials (GRMs) obtained from a controlled top-down demolition of a carbon black, on silicone slices to test their potential use as functional coating on invasive medical devices as indwelling urinary catheters. Results indicate that the relevant chemical-physical features of the deposit (controlled by FTIR and AFM) were maintained after MAPLE deposition. After deposition, GL films underwent a biological survey toward target cellular lines (murine fibroblast NIH3T3, human keratinocytes HaCAT and the human cervical adenocarcinoma epithelial-like HeLa). Results indicate that the GL films did not lead to any perturbations in the different biological parameters evaluated. The presented results and the possibility to further functionalize the GL or combine them with other functional materials in a hybrid fashion to assure a tighter adhesion onto the substrate for use in harsh conditions open the door to practical applications of these new-concept medical devices (drug delivery, next generation flexible devices, multifunctional coatings) paving the way to the prevention of nosocomial infections driven by catheterization through antibiotics-free approaches.

The effects of cobalt incorporation in spherical heterostructured iron oxide nanocrystals (NCs) of sub-critical size have been explored by colloidal chemistry methods. Synchrotron X-ray total scattering methods suggest that cobalt (Co) substitution in rock salt iron oxide NCs tends to remedy their vacant iron sites, offering a higher degree of resistance to oxidative conversion. Self-passivation still creates a spinel-like shell, but with a higher volume fraction of the rock salt Co-containing phase in the core. The higher divalent metal stoichiometry in the rock salt phase, with increasing Co content, results in a population of unoccupied tetrahedral metal sites in the spinel part, likely through oxidative shell creation, involving an ordered defect-clustering mechanism, directly correlated to core stabilization. To shed light on the effects of Co-substitution and atomic-scale defects (vacant sites), Monte Carlo simulations suggest that the designed NCs, with desirable, enhanced magnetic properties (cf. exchange bias and coercivity), are developed with magnetocrystalline anisotropy which increases due to a relatively low content of Co ions in the lattice. The growth of optimally performing candidates combines also a strongly exchange-coupled system, secured through a high volumetric ratio rock salt phase, interfaced by a not so defective spinel shell. In view of these requirements, specific absorption rate (SAR) calculations demonstrate that the rock salt core sufficiently protected from oxidation and the heterostructure preserved over time, play a key role in magnetically mediated heating efficacies, for potential use of such NCs in magnetic hyperthermia applications.

Aluminum Josephson junctions are the building blocks for the realization of superconducting quantum bits. Attention has been also paid to hybrid ferromagnetic Josephson junctions, which allow switching between different magnetic states, making them interesting for applications such as cryogenic memories, single-photon detectors, and spintronics. In this paper, we report on the fabrication and characterization of high-quality ferromagnetic Josephson junctions based on aluminum technology. We employed an innovative fabrication process inspired by niobium-based technology, allowing us to obtain very high-quality hybrid aluminum Josephson junctions; thus, supporting the use of ferromagnetic Josephson junctions in advanced quantum circuits. The fabrication process is described in detail and the main DC transport properties at low temperatures (current-voltage characteristic, critical current as a function of the temperature, and the external magnetic field) are reported. Here, we illustrate in detail the fabrication process, as well as the main DC transport properties at low temperatures (current-voltage characteristic, critical current as a function of the temperature, and the external magnetic field).

Skin aging is a very well-known process setting a gradual worsening of skin mechanical features due to a decline in the production of the extra-cellular matrix machinery and to a concurrent change in the contraction process. To slow this progression, it is crucial to induce the expression of several proteins able to promote elastic fibers formation and tissue repair. Here, the Oenothera biennis cell culture aqueous extract has been investigated from a chemical point of view and then it was tested in vitro, in cell, and in ex vivo experiments as adjuvant in counteracting skin aging. Accordingly, it has been shown that the Oenothera biennis extract was able, by increasing MYLK gene expression, to promote matrix collagen contraction, actin polymerization, and the production of essential ECM proteins.

PURPOSE: The importance of polymer-based composite materials to make multifunctional substrates for tissue engineering and the strategies to improve their performances have been stressed in the literature. Bioactive features of sol-gel synthesized poly(ε-caprolactone)/TiO2 or poly(ε-caprolactone)/ZrO2 organic-inorganic hybrid materials are widely documented. Accordingly, the aim of this preliminary research was to develop advanced composite substrates consisting of a poly(ε-caprolactone) matrix reinforced with sol-gel synthesized PCL/TiO2 or PCL/ZrO2 hybrid fillers. METHODS: Micro-computed tomography and atomic force microscopy analyses allowed to study surface topography and roughness. On the other hand, mechanical and biological performances were evaluated by small punch tests and Alamar Blue™ assay, respectively. RESULTS: Micro-computed tomography and atomic force microscopy analyses highlighted the effect of the preparation technique. Results from small punch tests and Alamar Blue™ assay evidenced that PCL reinforced with Ti2 (PCL=12, TiO2=88 wt%) and Zr2 (PCL=12, ZrO2=88 wt%) hybrid fillers provided better mechanical and biological performances. CONCLUSIONS: PCL reinforced with Ti2 (PCL=12, TiO2=88 wt%) and Zr2 (PCL=12, ZrO2=88 wt%) hybrid fillers could be considered as advanced composite substrates for hard tissue engineering.

ß-glucosidase (BG) plays a key role in determining the efficiency of the enzymatic complex cellulase for the degradation of cellulose into sugars. It hydrolyses the cellobiose, an inhibitor of the enzymatic complex. Therefore, the immobilization of BG is a great challenge for the industrial application of cellulases. Cellulases usually contains a BG amount insufficient to avoid inhibition by cellobiose. Here the BG was immobilized by matrix assisted pulsed laser evaporation (MAPLE) technique. The frozen matrix was composed of water, water/m-DOPA and water/m-DOPA/quinone. The effect of the excipients on the final conformation of the enzyme after the MAPLE processing was determined. The enzyme secondary structure was studied by FTIR analysis. The catalytic performances of the deposited films were tested in the cellobiose hydrolysis reaction. The results demonstrate that the presence of the oxidized form of m-DOPA, the O-quinone form, can protect the protein native structure, with the laser inducing little or no damage. In fact, only the samples deposited from this target preserved the secondary structure of the polypeptide chain and allowed a complete hydrolysis of cellobiose for four consecutive runs, showing a high operational stability of the biocatalyst.

The identification of alternative biocompatible magnetic NPs for advanced clinical application is becoming an important need due to raising concerns about iron accumulation in soft issues associated to the administration of superparamagnetic iron oxide nanoparticles (NPs). Here, we report on the performance of previously synthetized iron-doped hydroxyapatite (FeHA) NPs as contrast agent for magnetic resonance imaging (MRI). The MRI contrast abilities of FeHA and Endorem® (dextran coated iron oxide NPs) were assessed by 1H nuclear magnetic resonance relaxometry and their performance in healthy mice was monitored by a 7 Tesla scanner. FeHA applied a higher contrast enhancement, and had a longer endurance in the liver with respect to Endorem® at iron equality. Additionally, a proof of concept of FeHA use as scintigraphy imaging agent for positron emission tomography (PET) and single photon emission computed tomography (SPECT) was given labeling FeHA with 99mTc-MDP by a straightforward surface functionalization process. Scintigraphy/x-ray fused imaging and ex vivo studies confirmed its dominant accumulation in the liver, and secondarily in other organs of the mononuclear phagocyte system. FeHA efficiency as MRI-T2 and PET-SPECT imaging agent combined to its already reported intrinsic biocompatibility qualifies it as a promising material for innovative nanomedical applications. STATEMENT OF SIGNIFICANCE: The ability of iron-doped hydroxyapatite nanoaprticles (FeHA) to work in vivo as imaging agents for magnetic resonance (MR) and nuclear imaging is demonstrated. FeHA applied an higher MR contrast in the liver, spleen and kidneys of mice with respect to Endorem®. The successful radiolabeling of FeHA allowed for scintigraphy/X-ray and ex vivo biodistribution studies, confirming MR results and envisioning FeHA application for dual-imaging.

Graphene is an attractive component for high-performance stimuli-responsive or 'smart' materials, shape memory materials, photomechanical actuators, piezoelectric materials and flexible strain sensors. Nanocomposite fibres were produced by electrospinning high molecular weight Polyvinylpyrrolidone (PVP-1300 kDa) in the presence of noncovalently functionalised graphene obtained through tip sonication of graphite alcoholic suspensions in the presence of PVP (10 kDa). Bending instability of electrospun jet appears to progressively increase at low graphene concentrations with the result of greater fibre stretching that leads to lower fibre diameter and possibly conformational changes of PVP. Further increase of graphene content seams having the opposite effect leading to greater fibre diameter and Raman spectra similar to the pure PVP electrospun mats. All this has been interpreted on the basis of currently accepted model for bending instability of electrospun jets. The graphene addition does not lower the very high sound absorption coefficient, α, close to unity, of the electrospun PVP mats in the frequency range 200⁻800 Hz. The graphene addition affects, in a non-monotonous manner, the bell shaped curves of α versus frequency curves becoming sharper and moving to higher frequency at the lower graphene addition. The opposite is observed when the graphene content is further increased.

We have explored the synthesis of iron oxide particles, tubes, and fibrils within the pores of nanoporous polycarbonate and alumina membranes. The membranes contain uniformly distributed cylindrical pores with monodispersed diameters (varying between 20 and 200 nm) and thicknesses of 6 and 60 microm, respectively. By hydrolysis and polymerization of iron salts, particles of different sizes and phases were formed in the pores, building iron oxide particle nanowires. Alternatively, by the sol-gel technique, using as reagents metalloorganic compounds, fibrils and tubes of different iron oxide phases were prepared. Structural and morphological investigations performed using scanning electron microscopy and transmission electron microscopy revealed ordered iron oxide particle wires, tubes, and fibrils formed inside the membrane nanopores. Magnetic characterization was accomplished with a vibrating sample magnetometer. Below the blocking temperature (T(B)), the magnetic behavior of the nanowires was governed by dipolar interaction between nearest-neighbor nanoparticles inside the pore, whereas the energy barrier, and therefore the T(B) value, was mainly governed by dipolar interaction between magnetic moments over larger (interpore) distances. As expected, crystalline iron oxide nanotubes exhibited magnetic perpendicular anisotropy due to their magnetocrystalline and shape anisotropy.

Disordered hyperuniform (DH) media have been recognized as a new state of disordered matter that broadens our vision of material engineering. Here, long-range correlated disordered two-dimensional patterns are fabricated by self-assembling of spherical diblock-copolymer (BCP) micelles. Control of the self-assembling parameters leads to the formation of DH patterns of micelles that can host nanoscale material inclusions, therefore providing an effective strategy for fabricating multimaterial DH structures at molecular scale. Centroidal patterns are accurately determined by virtue of BCP micelles loaded with metal nanoparticles. Our analysis reveals the signature of nearly ideal DH BCP assemblies in the local density fluctuation and a dominant linear scaling in the local number fluctuation.

We describe a simple mechanical approach for low-density polyethylene film coating by multilayer graphene. The technique is based on the exfoliation of nanocrystalline graphite (few-layer graphene) by application of shear stress and allows to obtain thin graphene layers on the plastic substrate. We report on the temperature dependence of electrical resistance behaviors in films of different thickness. The experimental results suggest that the semiconducting behavior observed at low temperature can be described in the framework of the Efros-Shklovskii variable-range-hopping model. The obtained films exhibit good electrical conductivity and transparency in the visible spectral region. PACS: 72.80.Vp; 78.67.Wj; 78.66.Qn; 85.40.Hp.

Poly(epsilon-caprolactone) (PCL 6, 12 and 24 wt%) and zirconium (ZrO(2)) organic-inorganic hybrid materials have been synthesized by the sol-gel method from a multicomponent solution containing zirconium propoxide [Zr(OC(2)H(7))(4)], poly(epsilon-caprolactone) (PCL), water, chloroform (CHCl(3)). Sodium ampicillin was incorporate in the hybrid materials to verify the effect as local controlled drug delivery system. The structure of interpenetrating network is realized by hydrogen bonds between Zr-OH group (H donator) in the sol-gel intermediate species and carboxylic group (H-acceptor) in the repeating units of the polymer. The presence of hydrogen bonds between organic/inorganic components of the hybrid material was proved by FTIR analysis. The morphology of the hybrid material was studied by scanning electron microscope (SEM). The structure of a molecular level dispersion has been disclosed by atomic force microscope (AFM), pore size distribution and surface measurements. The bioactivity of the synthesized hybrid material has been showed by the formation of a layer of hydroxyapatite on the surface of PCL/ZrO(2) samples soaked in a fluid simulating the composition of the human blood plasma. Release kinetics in a simulate body fluid (SBF) have been subsequently investigated. The amount of sodium ampicillin released has been detected by UV-VIS spectroscopy and SEM. The release kinetics seems to occur in more than one stage. HPLC analysis has also been taken to ensure the integrity of ampicillin after the synthetic treatment.
