BACKGROUND & OBJECTIVES: Sikkim is a part of Eastern Himalayan biodiversity hotspot of India rich in bamboo flora harbouring over 30 different bamboo species. The present study was aimed to investigate the larval mosquito diversity in the bamboo stumps of Gangtok, Sikkim. Besides, efforts were also made to evaluate the propensity of particular species of mosquito towards specific bamboo species (if any). METHODS: A total of 75 bamboo stumps of four genera were surveyed and screened at five different sampling sites of Gangtok from July to October 2017. Mosquito species similarity between the five sampling sites and the four varieties of bamboo species was calculated using the Bray-Curtis similarity index. RESULTS: A total of 216 larvae were collected from 25 different bamboo stumps studied. The species identified were Aedes albopictus, Ae. atlanticus, Ae. aegypti, Orthopodomyia signifera, Oclerotatus japonicus, Oc. taeniorhynchus, Armigeres subalbatus, and Toxorhynchites splendens. The Oc. japonicus (34.5%) was found to be the most abundant species having distribution in Phyllostachys assamica, Dendrocalamus hamiltonii and Bambusa nutans. On the other hand, genus Armigeres subalbatus and Tx. splendens were found to breed only in the stumps of P. assamica. Based on Bray-Curtis similarity index highest species similarity was recorded between D. hamiltonii and P. assamica bamboo species. INTERPRETATION & CONCLUSION: The study may help to understand the bioecology of the mosquito larvae which may help to devise suitable mosquito control programmes. Future studies including the survey of large number of bamboo stumps both in urban and rural areas of Sikkim may provide better insight into the mosquito diversity in the bamboo stumps of Sikkim.

Rheumatoid arthritis (RA) is a complex, multifactorial, systemic autoimmune disease. Reports are suggestive of the role of HLA especially HLA-DRB1 alterations in RA pathogenesis. Existing data involving different geographical populations on the role of alterations in specific locus of HLA-DRB1 in RA susceptibility and severity are equivocal, with no data available from ethnically distinct North-east Indian population, where RA cases are alarmingly increasing. This study aimed to evaluate the association of HLA-DRB1 gene SNPs (rs13192471, rs660895 and rs6457617) with susceptibility and severity of RA in an ethnically distinct North-east Indian population. Whole blood was collected from clinically characterized RA cases (satisfying the American College of Rheumatology 1987 criteria) (n = 123) and community-based age and sex-matched healthy controls (n = 156) with informed consent. The HLA-DRB1 SNP analysis was performed for all the RA and control cases using ARMS-PCR using case and control genomic DNA as template. Statistical analysis was performed by SPSSv13.0 software. The HLA-DRB1 rs660895 showed both wild (AA) and heterozygote (AG) genotype but the heterozygote allele was found to be associated with reduced risk of RA compared to controls [OR = 0.531, p = .024]. The difference in distribution of rs6457617 polymorphism between RA and control cases was comparable [OR = 0.525, p = .079]. Significantly higher distribution of variant rs13192471 genotype was observed in RA cases (69.92%) compared to controls (46.75%) (p < .001) and was associated with increased risk of susceptibility to RA [OR = 2.576, p < .001] compared to controls, as well as progression to severity in RA cases [OR = 2.404, p = .048]. Combinatorially also, the presence of rs13192471 variant genotype was associated with increased risk of RA susceptibility [OR = 8.267, p = .026] and RA severity [OR = 3.647, p = .280]. Alterations in HLA-DRB1 are associated with RA susceptibility. HLA-DRB1 rs13192471 SNP plays a critical role in RA susceptibility and severity in North-east Indian cases and has prognostic significance in RA.

Laboratory investigation on vermicomposting efficacy of Perionyx excavatus for recycling of distillation waste biomass of java citronella (Cymbopogon winterianus Jowitt) was carried out in two seasonal trials i.e. summer and winter periods. The experiment was conducted in earthen pots using a mixture of citronella waste material and cowdung in the proportion of 5:1. A control treatment without earthworms was setup for comparison of the results. The vermicompost had shown 5.8 folds reduction in C/N ratio and 5.6 folds enhancement in ash content. The nutrient contents (N, P, K, Ca and Mg) in the vermicompost had increase in the range of 1.2 - 4.1 fold than the initial level. The FT-IR spectra of the vermicompost confirmed increase in nitrogen rich compounds and decrease in aliphatic/aromatic compounds as compared to the initial level of the biowaste materials. The vermicomposting process is influenced by seasonal variation and summer was more productive than winter.

Laboratory experiment on vermicomposting of distillation waste of java citronella (Cymbopogon winterianus Jowitt.) was carried out employing Eudrilus eugeniae, in two seasonal trials, covering summer and winter periods. Two vermicomposting treatments were conducted in earthen pots, one with citronella plant waste only (CW) and the other, a mixture of citronella waste and cowdung in the proportion 5:1 (CW+CD). Vermicomposting of citronella waste resulted reduction in C/N ratio (83.5-87.7%), enhancement of ash content and a number of macro and micronutrients. The FT-IR spectroscopy of the vermicompost revealed the reduction in aliphatic and aromatic compound as well as increase in amide group after the 105 days stabilization process. The vermicompost output was significantly enhanced in CW+CD treatment than CW treatment. Even, nutrient content of the vermicompost was also higher in CW+CD treatment than CW alone indicating the positive role of cowdung in improvement of quantity and quality.

RESUMO

Picroliv, a standardised iridoid glycoside fraction from the root and rhizome of Picrorhiza kurroa at a dose of 25 mg kg-1 p.o. inhibited passive cutaneous anaphylaxis in mice (82%) and rats (50-85%) and protected mast cells from degranulation (60-80%) in a concentration-dependent manner. Its effect was also studied in sensitised guinea pig ileum preparation in vitro (Schultz-Dale study) and in normal guinea pigs in vivo (Konzett-Rossler, in preparation). There was inhibition of the Schultz-Dale response in sensitised guinea pig ileum, but the bronchospasm induced by histamine could not be antagonised or prevented by Picroliv, indicating the absence of a direct post-synaptic histamine receptor blocking activity.
