RESUMO

The CERN-MEDICIS (MEDical Isotopes Collected from ISolde) facility has delivered its first radioactive ion beam at CERN (Switzerland) in December 2017 to support the research and development in nuclear medicine using non-conventional radionuclides. Since then, fourteen institutes, including CERN, have joined the collaboration to drive the scientific program of this unique installation and evaluate the needs of the community to improve the research in imaging, diagnostics, radiation therapy and personalized medicine. The facility has been built as an extension of the ISOLDE (Isotope Separator On Line DEvice) facility at CERN. Handling of open radioisotope sources is made possible thanks to its Radiological Controlled Area and laboratory. Targets are being irradiated by the 1.4 GeV proton beam delivered by the CERN Proton Synchrotron Booster (PSB) on a station placed between the High Resolution Separator (HRS) ISOLDE target station and its beam dump. Irradiated target materials are also received from external institutes to undergo mass separation at CERN-MEDICIS. All targets are handled via a remote handling system and exploited on a dedicated isotope separator beamline. To allow for the release and collection of a specific radionuclide of medical interest, each target is heated to temperatures of up to 2,300°C. The created ions are extracted and accelerated to an energy up to 60 kV, and the beam steered through an off-line sector field magnet mass separator. This is followed by the extraction of the radionuclide of interest through mass separation and its subsequent implantation into a collection foil. In addition, the MELISSA (MEDICIS Laser Ion Source Setup At CERN) laser laboratory, in service since April 2019, helps to increase the separation efficiency and the selectivity. After collection, the implanted radionuclides are dispatched to the biomedical research centers, participating in the CERN-MEDICIS collaboration, for Research & Development in imaging or treatment. Since its commissioning, the CERN-MEDICIS facility has provided its partner institutes with non-conventional medical radionuclides such as Tb-149, Tb-152, Tb-155, Sm-153, Tm-165, Tm-167, Er-169, Yb-175, and Ac-225 with a high specific activity. This article provides a review of the achievements and milestones of CERN-MEDICIS since it has produced its first radioactive isotope in December 2017, with a special focus on its most recent operation in 2020.

Many regulations prohibit using veterinary drugs in feedingstuffs to protect consumers and animals alike. Within this investigation we developed a simple, cost-efficient primary screening method for detecting antibiotics and coccidiostats in animal feeds. Thirty-two veterinary drugs were originally considered. Following matrix-free testing to optimise detection, an assay based on matrix extraction with methanol/acetonitrile/phosphate buffer followed by inoculation and diffusion in agar plates was developed. Final validation was performed with 14 representative drugs (one per drug class) and four bacteria (Escherichia coli ATCC11303 and ATCC27166, Staphylococcus aureus ATCC6538P, Micrococcus luteus ATCC9341) in bovine, lamb and swine fodder, measuring growth inhibition zones. Of the original drugs tested, 27 remained detectable in feed matrices at or below 20 mg kg(-1). Of the 14 validated representatives, two had estimated minimum detectable concentrations of 10-11 mg kg(-1), others of 5 mg kg(-1) or lower, an earlier minimum European Union inclusion rate for many veterinary drugs. No significant matrix effect on inhibition zones was detected. Per cent wrong negative deviations ranged from 0% (nine of 14 compounds) to 20-27% (two of 14), while inter-day precision based on inhibition zones had relative standard deviations (RSDs) of 6-109% (mean of 40%). When setting a 1 mm inhibition zone, the maximum observed for negative controls, as a cut-off level, no false-positives were found. While not all targeted antibiotics were detectable in complex matrices, the majority of veterinary drugs were detected with reasonable sensitivity, indicating that this method could be suitable for screening feedingstuffs prior to further confirmatory investigation of positive findings such as by LC-MS/MS.

A simple multi-residue method was developed for detecting and quantifying 33 analytes from 13 classes of antibiotics (tetracyclines (3), quinolones (7), penicillins (3), ionophore coccidiostats (7), macrolides (3), sulfonamides (1), quinoxalines (2), phenicols (2), lincosamides (1), diaminopyrimidines (1), polypeptides (1), streptogramins (1) and pleuromutilins (1)) in animal feeds. Extraction and clean-up procedures were optimized with spiked piglet feed. Samples were extracted by ultrasonic-assisted extraction with a mixture of methanol/acetonitrile/McIlvaine buffer at pH 4.6 (37.5/37.5/25, v/v/v) containing 0.3% of EDTA-Na(2), followed by a clean up using a dispersive solid-phase extraction (d-SPE) with PSA (primary secondary amine). Detection of antibiotics was achieved by liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS) within 28 min using both positive and negative ESI mode. Average recoveries ranged from 51% (oxytetracycline) to 116% (tilmicosin) with associated relative standard deviations of 7.3% and 9.0% and an overall mean of 87%. Limits of quantification ranged from 3.8 ngg(-1) (lincomycin) to 65.0 ngg(-1) (bacitracin). Following optimization, the method was further verified for bovine and lamb feeding stuffs; negative matrix effects were evaluated and overcome by a standard addition method.

Contamination levels of PCBs, and of the heavy metals cadmium (Cd), lead (Pb) and mercury (Hg) were analyzed in four fish species from seven rivers in the North of Luxembourg. During August and September 2007, 85 samples of fish were collected belonging to four species: the stone loach (Barbatula barbatula, n=12 pools), the chub (Squalius cephalus, n=36), the barbel (Barbus barbus, n=23) and eel (Anguilla anguilla, n=14). The concentration of seven indicator PCBs ( summation operator(7)PCBs) reached a mean of 39ngg(-1) and varied between 4.0 and 346.2ngg(-1) (wet wt) depending on the site and species. Fish collected at Wallendorf on the Our River and sites on the Wiltz and the Clerve rivers showed the highest concentrations for PCBs. In comparison with 1994, PCB levels in fish decreased strongly during the last decade in these rivers. Lead was detected at low levels (0-181.4ngg(-1) wet wt). Mercury concentrations ranged between 10.3 and 534.5ngg(-1) (wet wt) exceeding maximum tolerable levels for human consumption of 500ngg(-1) in two fish out of 85. Chubs and eels from the Sûre River were the most contaminated by mercury. Cadmium levels varied between 4.0 and 103.9ngg(-1) (wet wt). In addition to mercury in fish, cadmium was the most problematic pollutant on the Our, the Wiltz, the Clerve and the Troine Rivers, because values found in 20% of fish exceeded the threshold of about 10-50ngg(-1) (wet wt) recommended for human health. The total PCB level predicted to accumulate in livers from otter potentially feeding on these fish based on a previously published mathematical model is 37.7microgg(-1) (lipid wt), which is between a proposed "safe level" and a "critical level" for otters. Rivers in the North of Luxembourg are thus to some extent polluted, and the establishment of otter populations could be affected by current levels of contamination.

Populations of widely distributed species can be subjected to unequal selection pressures, producing differences in rates of local adaptation. We report a laboratory experiment testing tolerance variation to UV-B and polycyclic aromatic hydrocarbons (PAHs) among common frog (Rana temporaria) populations according to their natural exposure level in the field. Studied populations were naturally distributed along two gradients, i.e. UV-B radiation with altitude and level of contamination by PAHs with the distance to emitting sources (road traffic). Tadpoles from eight populations were subjected to (1) no or high level of artificial UV-B; (2) four concentrations of benzo[a]pyrene (BaP) (0, 50, 250, 500microgL(-1)); (3) simultaneously to UV-B and BaP. Since both stressors are genotoxic, the number of micronucleated erythrocytes (MNE) in circulating red blood cells was used as a bioindicator of tadpole sensitivity. High-altitude populations appear to be locally adapted to better resist UV-B genotoxicity, as they showed the lowest MNE numbers. Conversely, no correlation was observed between levels of PAH contamination in the field and tadpole tolerance to BaP in the laboratory, indicating the absence of local adaptation for BaP tolerance in these populations. Nevertheless, the decrease of MNE formation due to BaP exposure with altitude suggests that high-altitude populations were intrinsically more resistant to BaP genotoxicity. We propose the hypothesis of a co-tolerance between UV-B and BaP in high-altitude common frog populations: local adaptation to prevent and/or repair DNA damage induced by UV-B could also protect these highland populations against DNA damage induced by BaP. The results of this study highlight the role of local adaptation along pollutant gradients leading to tolerance variation, which implies that is it necessary to take into account the history of exposure of each population and the existence of co-tolerance that can hide toxic effects of a new pollutant.
