This study was conducted to evaluate the effect of ß-glucan on feed intake, digestibility of nutrients and ruminal fermentation in Thai native beef cattle that were fed low-quality roughage. Four, Thai native beef cattle with initial body weight (BW) of 100 ± 20.0 kg were randomly assigned according to a 4 × 4 Latin square design to receive four treatment by ß-glucan at 0, 1.6, 3.1 and 4.7 g/day respectively. Supplementation of ß-glucan at 0-4.7 g in cattle did not affect the intake of DM, OM, CP, NDF or ADF. However, supplementation with ß-glucan significantly improved rice straw intake (g/kg BW0.75 ), compared to the nonsupplemented group (p < 0.05), so rice straw intake was higher than in the group without ß-glucan supplementation. In addition, total feed intake increased with the higher level of ß-glucan supplementation. The estimated energy intake was not altered by ß-glucan supplementation (p > 0.05). The experimental diet had no effect (p > 0.05) on the apparent digestibility of DM, OM, NDF or ADF. However, the digestibility of CP was significantly different among treatment and supplementation at 4.7 g/day ß-glucan which was the highest level (63.2%) among all diets. ß-glucan supplementation did not affect rumen pH, temperature or ruminal NH3 -N concentration. In addition, blood urea-N levels were similar among experimental groups. The population of protozoa at 4 hr post-feeding increased along with ß-glucan supplementation and was highest at 4.7 g of ß-glucan. Based on this experiment, it could be concluded that supplementation of ß-glucan at 4.7 g improved rice straw intake, as well as total intake and digestibility of CP in Thai native beef cattle.

The objective of this research was to evaluate the effect of feeding fresh cassava root (CR) along with a feed block containing high was to sulfur (FBS) on feed intake, digestibility, rumen fermentation, and blood thiocyanate concentration in Thai native beef cattle. Four Thai male native beef cattle, initial body weight (BW) of 130 + 20.0 kg, were used in this study. The experiments were randomly assigned according to a 2 × 2 factorial arrangement in a 4 × 4 Latin square design. The main factors were supplemented fresh CR levels (1.0 and 1.5% BW) and across to a feed block supplemented with sulfur added 2% (FBS-2) and 4% (FBS-4). Intakes of rice straw, concentrate diets, and FBS were not affected by treatments. Intakes of CR, sulfur, and total intake were significantly altered by the FBS treatment. The apparent dry matter and organic matter digestibility coefficient were significantly higher in animals fed FBS-4 than in those fed FBS-2. The ruminal ammonia nitrogen concentration was not affected by treatment and ranged from 15.6 to 17.6 mg/dl. Populations of protozoa and fungal zoospores were similar across treatments, whereas the bacterial population was significantly different between sulfur levels in the feed block. Feeding CR with FBS did not change total volatile fatty acid (VFA) concentrations and VFA profiles except for the propionic acid concentration, which was higher in the group with CR supplementation at 1.5% BW. Cattle fed CR with FBS showed similar blood urea nitrogen concentration at various feeding times and overall. In contrast, CR supplementation at 1.5% BW with FBS-2 increased blood thiocyanate concentrations. Therefore, supplementation of FBS-2 was beneficial to Thai native beef cattle fed with 1.5% BW fresh CR as it improved digestibility and rumen fermentation presumed, because HCN from fresh cassava root was converted into thiocyanate, which is nontoxic to farm animals.

OBJECTIVE: This experiment was conducted to investigate the effects of crude glycerin from waste vegetable oil (CGWVO) on performance, carcass traits, meat quality, and muscle chemical composition. METHODS: Twenty-four crossbred (Thai Native×Anglo Nubian) uncastrated male goats (16.8± 0.46 kg body weight [BW]) were assigned to a completely randomized design and subjected to four experimental diets containing 0%, 2%, 4%, and 6% of CGWVO (63.42% of glycerol and 47.78% of crude fat) on a dry matter (DM) basis. The diets were offered ad libitum as total mixed rations twice daily. The feed intake, feeding behavior, growth performance, carcass and meat traits, and muscle chemical composition were evaluated. RESULTS: Based on this experiment, there were significant differences (p>0.05) among groups regarding DM intake, growth performance, and carcass traits where goats receiving 6% of CGWVO had lower daily DM intake, growth performance, and carcass traits than those fed on 0%, 2%, and 4% of CGWVO. There were no effects of CGWVO on carcass length, carcass width, Longissimus muscle (LM) area, Warner-Bratzler shear force, pH and color of LM at 45 min after slaughter, as well as on other carcass cut and muscle chemical composition. CONCLUSION: In conclusion, the addition of up to 4% of DM in the diets for crossbred finishing goats seems to be the most interesting strategy, since it promotes greatest animal performance. Moreover, this study was a suitable approach to exploit the use of biodiesel production from waste vegetable oil for goat production.

OBJECTIVE: This study was an attempt to investigate the effect of levels of fungal (Lentinus sajor-caju) treated oil palm fronds (FTOPF) on digestibility, rumen fermentation, and nitrogen balance in goats. METHODS: Four 16 month old male crossbred (Thai Native×Anglo Nubian) goats with initial body weights of 33.5±1.7 kg were randomly assigned according to a 4×4 Latin square design. Four levels of FTOPF were assigned for feed intake. The experimental treatments consisted of 0%, 33%, 67%, and 100% of oil palm fronds (OPF) being replaced by FTOPF. RESULTS: The results revealed that total dry matter intake and nutrient intake were not influenced (p>0.05) by the inclusion of FTOPF. However, the efficiency values of the digestibility of the dry matter, organic matter, crude protein, neutral detergent fiber, acid detergent fiber, and acid detergent lignin on FTOPF were higher (p<0.05) in treatments with 33%, 67%, and 100% of FTOPF compared with 0% of FTOPF. FTOPF feeding did not change the rumen pH, temperature, and NH3-N. However, the FTOPF levels did affect the total volatile fatty acid (VFA), molar proportion of acetate, propionate, butyrate, ratio of acetic (propionic acid and acetic) plus butyric (propionic acid), and production of CH4. The totals of VFA and propionate was lower in goat fed with 0% of FTOPF than in those of the other groups (p<0.05). The amount of nitrogen retention based on g/d/animal or the percentage of nitrogen retained was the lowest the goat fed with 0% of FTOPF (p<0.05), whereas nitrogen intake, excretion, and absorption were not changed among treatments. CONCLUSION: Based on this study, FTOPF could be effectively used as an alternative roughage source in total mixed ration diets, constituting at least up to 100% of OPF.

The aim of this experiment was to determine the effect of dried rumen digesta pellet levels on feed intake, digestibility, rumen ecology, and blood metabolites in swamp buffalo. Four 2-year-old male swamp buffalo with an initial body weight (BW) of 150 ± 10.0 kg were randomly assigned according to a 4 × 4 Latin square design to receive four levels of dried rumen digesta pellets (DRDPs). The dietary treatments were supplementation of DRDP at 0, 50, 100, and 150 g dry matter/day, respectively. Total feed intake was significantly different among treatments (p < 0.05) and was highest in the 150 g/day DRDP supplement (2.68 kg/day). Intakes of neutral detergent fiber (NDF) and acid detergent fiber did not affect DRDP levels, while intakes of organic matter and crude protein (CP) were altered significantly when 150 g of DRDP was used (p < 0.05). Buffalo fed with DRDP at 150 g/day had the highest CP and NDF digestibility (p < 0.05). DRDP supplementation did not affect rumen pH, and temperature and the concentration of ruminal ammonia-nitrogen and blood urea nitrogen were not altered among the treatments. The mean value of fungal zoospores in the buffalo was significantly different among treatments and was highest in supplementation with DRDP at 150 g. The mean value of propionic acid was significantly different at various levels of DRDP; it was highest in the group fed with 150 g DRDP (p < 0.05). Thus, supplementation of DRDP at 150 g improved feed use and increased fungal zoospore population. In addition, DRDP feeding is recommended, since it has positive economic impacts and helps control environmental pollution.

OBJECTIVE: This experiment was studied the effects of various levels of crude glycerin (CG) in dairy goat diet on daily intake, milk yield, milk composition, some physical properties and some quality changes of goat milk after sterilization. METHODS: Twelve 75% Saanen dairy goats (body weight = 49±3 kg; days in milk = 60±12 d) were randomly assigned in a completely randomized design to evaluate the effects of three experimental diets consisting of 0%, 5%, and 10% CG (dry matter basis) which were formulated to meet or exceed the nutrient requirements of goats. Experimental dairy goats were evaluated for feed and milk yield. Milk samples were analyzed for their composition, including fatty acids, casein profile, fat globule size, and color, and were sterilized to evaluate milk heat stability. RESULTS: There were no significant differences between 0% and 5% CG treatments infeed. Increasing CG supplementation from 0% to 5% increased milk yield from 2.38±0.12 to 2.64±0.23 kg/goat/d. In addition, milk samples from 5% CG treatment had the highest total solids, fat content and lactose content, and largest fat globule size. Increasing CG to 10% resulted in a decrease in milk fat. After sterilizing at 116°C, F 0 = 3 min, goat milk samples from 5% CG treatment had slightly higher sediment content and comparatively higher degree of browning. CONCLUSION: Considering milk yield, milk fat content and quality of sterilized milk, 5% CG supplementation in a total mixed ration has a potential for implementation in dairy goats.

OBJECTIVE: This experiment was conducted to investigate the effects of fungal treated oil palm fronds (FTOPF) on performance, carcass traits, meat quality, and muscle chemical composition. METHODS: Eighteen growing crossbred male goats (Thai Native×Anglo Nubian) with 18.7±2.0 kg of initial body weight (BW) were stratified and blocked by BW in a randomized complete block design. Three diets containing 30% of oil palm fronds (OPF) either untreated (UOPF) or treated with Lentinussajor-caju (FTOPF) with or without urea (FTOPFU) were used as roughage sources in total mixed rations (TMRs). The diets were offered ad libitum and weight gain was determined. At the end of the experimental period, the harvest data and carcass characteristics of the goats were recorded, and muscular longissimus dorsi composition was determined. RESULTS: No significant effect of fungal treated (FT) inclusion was observed in any of the feed intake, growth performance, and carcass characteristics. Likewise, no apparent effects on carcass composition and muscle chemical composition were detected in this study, except for hind leg and chump were affected (p<0.05) by FT inclusion. CONCLUSION: In conclusion, feeding of fungal (Lentinussajor-caju) treated oil palm frond in TMR diet did not affect performance and carcass characteristics in finishing goats.

This experiment was evaluation of the effects of increasing concentrations of crude glycerin from waste vegetable oil (CGWVO) in diets on feed intake, digestibility, ruminal fermentation characteristics, and nitrogen balance of goats. Four crossbred male (Thai Native × Anglo Nubian) goats, with an average initial body weight (BW) of 31.5 ± 1.90 kg, were randomly assigned according to a 4 × 4 Latin square design. The dietary treatments contained 0, 2, 4, and 6 % of dietary dry matter (DM) of CGWVO. Based on this experiment, there were significantly different (P > 0.05) among treatment groups regarding DM intake and digestion coefficients of nutrients (DM, OM, CP, EE, NDF, and ADF), which goats receiving 6 % of CGWVO had lower daily DMI and nutrient intake than those fed on 0, 2, and 4 % of CGWVO. Ruminal pH, NH3-N, and blood urea nitrogen (BUN) concentration were unchanged by dietary treatments, except that for 6 % of CGWVO supplementation, NH3-N, and BUN were lower (P < 0.05) than for the diets 0 % of CGWVO, while the differences between the diets 0, 2, and 4 % of CGWVO were not significant. The amounts of N absorption and retention were similar among treatments, except that for 6 % of CGWVO which N absorption was lower (P < 0.05) than among treatments while the difference between the diets 0, 2, and 4 % of CGWVO were not significant. Based on this study, CGWVO levels up to 4 % in total mixed ration could be efficiently utilized for goats. This study was a good approach in exploiting the use of biodiesel production from waste vegetable oil for goat production.

OBJECTIVE: The objective of the experiment was to study yeast supplementation (yeast, Y) and dried kratom leaves (DKTL) on the digestibility, ruminal fermentation, blood metabolites and nitrogen balance in goats. METHODS: Four of 7 to 8 months old male crossbred (50% Thai Native-Anglo Nubian) goats with average liveweight 20±0.13 kg were randomly assigned according to a 2×2 factorial arrangement in a 4×4 Latin square design to receive four diets ad libitum basis. The study investigated the effects of two levels of yeast (Y) supplementation (Y, 0 and 0.5g/kg dry matter [DM]) along with two levels of DKTL supplementation (DKTL, 0 and 4.44g/kg DM). The experimental groups were as follows: T1 = control group with 0Y+0DKTL, T2 = 0Y+4.44 DKTL, T3 = 0.5Y+0DKTL, and T4 = 0.5Y+4.44 DKTL. RESULTS: The results showed that there were no interactions between Y levels and DKTL levels with respect to total DM intake, but there were significant effects (p<0.05) by levels of Y; goats receiving 0.05 g/kg DM Y had higher than goats fed 0.0 g/kg DM on average (kg/d). A percentage of body weight (% BW) and grams per kilogram of metallic weight (g/kg w0.75) had no influence on yeast levels and DKTL, but there was a difference (p<0.05) by yeast level Y at 0.5 g/kg DM, being higher compared to the non-supplemented group. Apparent digestibility coefficient of nutrition in the form of (DM, organic matter, crude protein, neutral detergent fiber, and acid detergent fiber) was an increased trend in the Y-level complementary group at 0.5 g/kg DM and DKTL at 4.44 g/kg DM, respectively. Protozoa populations decreased in the group receiving Y levels at 0.5 g/kg DM and DKTL levels at 4.44 g/kg DM when compared to group T1. The acetic acid concentration and methane gas generation decreased (p<0.05) in the group receiving Y levels of 0.5 g/kg DM and DKTL levels of 4.44 g/kg DM, while the amount of propionic acid increased (p<0.05). CONCLUSION: Effects of feeding combinations of Y and DKTL supplementation on feed showed no interaction effect (Y×DKTL) on feed intake, rumen fermentation, bacterial and fungi population. The effect on protozoal populations was lower in the group that was supplemented with DKTL at 4.44 g/kg DM related to synthetic CH4 was reduced.

Feed is the most expensive component in goat production. Hence, lowering it is crucial to increasing producer profitability. The microbial community in rumen is vital for nutritional digestion and absorption in ruminants. Live yeast and yeast-based products generated from the strain Saccharomyces cerevisiae (commercial strain) are actively being used and investigated. The purpose of this study was to investigate the effects of substituting soybean meal (SBM) in concentrate diets with yeast-fermented palm kernel cake protein (YFPKCP) on dry matter intake, digestibility, blood markers, and nitrogen balance. Five crossbred Thai Native-Anglo-Nubian goats (50% Thai Native goats with 50% Anglo-Nubian goats) weighing an average of 27 ± 2 kg were randomly allocated to one of five diets using a 5 × 5 Latin square design: 0, 25, 50, 75, and 100% YFPKCP replacement for SBM. Plicatulum hay (Paspalum plicatulum Michx.) was provided ad libitum. There were no significant differences in dry matter (DM) intake among treatments, but the apparent digestibility of DM, crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) were affected (p < 0.05) by including YFPKCP in diets. They also tended to be slightly lower for goats fed the diet containing 100% YFPKCP replacement for SBM compared to other treatments. Ruminal pH, ammonia-nitrogen (NH3-N), blood glucose, and packed cell volume were equivalent among treatments. On the other hand, replacement YFPKCP reduced digestibility and N absorption by up to 75% (p < 0.05). Furthermore, there was no difference in total volatile fatty-acid concentration among goats fed YFPKCP as a substitute for SBM. According to the results of this study, the level of YFPKCP in the concentrate replacement of SBM for goats fed plicatulum hay should be 75%.

This study aimed to evaluate the inclusion levels of extracted oil palm meal (EOPM) from biodiesel byproducts in the total mixed ration (TMR) for goats on feed intake, nutrient digestibility, nitrogen balance, blood parameters, and rumen fermentation characteristics. The EOPM was included at 0%, 10%, 20%, and 30% dry matter (DM) to formulate into four dietary treatments and assigned to goats with an average age of 12 months and an initial body weight of 23 ± 0.5 kg in a 4 × 4 Latin square design. The PROC MIXED procedure was used to analyze all data, and the orthogonal polynomial was tested for EOPM levels using the SAS program. The DM intake, nitrogen, and metabolizable energy linearly (p < 0.05) increased when there was increased EOPM inclusion. Increasing EOPM inclusions resulted in (p < 0.05) a linear increase in fiber digestibility but did not affect DM, crude protein, or ether extract. Total volatile fatty acids increased (p < 0.05) linearly with increasing EOPM inclusions. The EOPM inclusions did not alter the bacteria, fungi, and protozoal populations. Blood parameters were not affected by the inclusions of EOPM except globulin, triglyceride, and MCV concentration. This study revealed that EOPM could be included in up to 20% DM in TMR for goats with no negative impact.

Once improperly managed, the citric acid production industry generates waste, which contributes to pollution and other environmental issues. We proposed that, with sufficient quality improvement, citric acid by-product (CAP) might be used for animal feed, thereby reducing the environmental impact. The aim of the present study was to ferment citric acid by-product (CAP) by inoculation with lactic acid bacteria (LAB) and a fibrolytic enzyme mixture for quality improvement and crude fiber reduction in the waste products. LAB inoculants were L. casei TH14, and the additive enzyme used was a fibrolytic enzyme mixture (glucanase, pectinase, and carboxymethylcellulase) of a small-scale fermentation method. The seven treatments employed in this study were as follows: (1) control (untreated), (2) CAP-inoculated L. casei TH14 at 0.01% DM, (3) CAP-inoculated L. casei TH14 at 0.05% DM, (4) CAP-inoculated enzymes at 0.01% DM, (5) CAP-inoculated enzymes at 0.05% DM, (6) CAP-inoculated L. casei TH14 at 0.01% DM with enzymes at 0.01% DM, and (7) CAP-inoculated L. casei TH14 at 0.05% DM with enzymes at 0.05% DM. The samples were taken on days 1, 7, 14, 21, and 28 of ensiling, both before and after. Four replications were used. The results of the chemical composition of the CAP before and after ensilage inoculated with L. casei TH14 did not show any differences in crude protein, ether extract, ash, or gross energy, but the enzymes significantly (p < 0.05) decreased crude fiber and increased nitrogen-free extract. The combination was especially effective at improving the characteristics of CAP, with a reduction in crude fiber from 21.98% to 22.69%, of neutral detergent fiber (NDF) from 16.01% to 17.54%, and of acid detergent fiber (ADF) from 13.75% to 16.19%. Furthermore, the combination of L. casei TH14 and the enzyme increased crude protein from 1.75% to 2.24% at 28 days of ensiling. Therefore, CAP-inoculated L. casei TH14 did not change in chemical composition, while crude fiber, NDF, and ADF decreased when CAP was inoculated with enzyme. The combination of L. casei TH14 and the enzyme is more effective at improving chemical composition and reducing crude fiber and enhancing carbohydrate breakdown in the CAP. Finally, by enhancing the CAP's quality, it may be possible to use it in animal feed and minimize its impact on the environment.

This research examines the impact of adding dried coffee cherry pulp (CoCP) to goat feed on the digestibility of the feed, rumen fermentation, hematological, and nitrogen balance. A goat feeding experiment employed four male crossbreds (Thai Native × Anglo Nubian) aged 12 months and weighing 21.0 ± 0.2 kg each. The treatment was conceived as a 4 × 4 Latin square with four specific CoCP levels at 0, 100, 200, and 300 g/day. Dry matter intake (DMI), organic matter intake (OMI), and crude protein intake (CPI) were unaffected by the addition of CoCP. However, across treatment groups, there was a linear increase in ether extract intake (EEI) (p < 0.01), neutral detergent fiber intake (NDFI) (p = 0.06), and acid detergent fiber intake (ADFI) (p = 0.04), as well as a quadratic effect on DMI% BW (p = 0.04). The findings showed that rumen temperature, pH, ammonia-nitrogen, or pack cell volume did not change with CoCP supplementation. Total volatile fatty acid showed linear effects on acetate (p = 0.03) and was quadratically affected by propionate concentration (p = 0.02), acetate to propionate ratio (p = 0.01), acetic plus butyric to propionic acid ratio (p = 0.01), and methane estimation (p = 0.01). With increased CoCP supplementation, there was a linear decrease in protozoa count by about 20.2% as the amount of CoCP supplemented increased (p = 0.06). CoCP supplementation in animal feed resulted in a linear decrease in urinary nitrogen (p = 0.02) and a quadratic effect on absorbed nitrogen (p = 0.08) among treatment groups, with greater N utilization values found in goats fed 200 g/d CoCP. In light of this, supplementing CoCP into animal feed may improve animal digestion and rumen fermentation effectiveness while having no effect on feed intake, rumen microbes, or blood metabolites.

The objective of this study was to see how dried Mitragyna speciosa Korth leaves (DKTL) affected growth, hematological parameters, carcass characteristics, muscle chemical composition, and fatty acid profile in finishing goats. In a randomized complete block design, twenty crossbred males (Thai Native x Boer) weaned goats (17.70 ± 2.50 kg of initial body weight (BW)) were provided to the experimental animals (5 goats per treatment) for 90 days. Individual dietary treatments of 0, 2.22, 4.44, and 6.66 g/d of DKTL on a dry matter basis were given to the goats. The diets were provided twice daily as total mixed rations ad libitum. In comparison to the control diet, DKTL supplementation had no effect on BW, average daily gain (ADG), feed conversion ratio (FCR), carcass composition, meat pH, or meat color (p > 0.05). After DKTL treatment, the hot carcass weight, longissimus muscle area, oleic acid (C18:1n9), monounsaturated fatty acid (MUFA), and protein content increased, but saturated fatty acids (SFA) and ether extract decreased (p < 0.05). To summarize, DKTL supplementation can improve goat meat quality.

Upgrading the nutritive value of rice straw (RS) is necessary to increase its contribution to enhancing meat and milk production. Present work verified whether novel Crabtree negative yeast inoculant could promote RS utilization, rumen fermentation, and milk quality in tropical crossbred lactating Holstein cows. The new stain of Crabtree negative yeasts (Pichia kudriavzevii KKU20 and Candida tropicalis KKU20) was isolated from the rumen of dairy cattle. This study used 6 multiparous crossbreds between Holstein Frisian × Zebu dairy cows in their mid-lactation period. Dairy cows were randomly allocated to three ensiled RS with various yeast stains including Saccharomyces cerevisiae, P. kudriavzevii KKU20, and C. tropicalis KKU20 according to a 3 × 3 replicated Latin square design. Crabtree-negative yeast (P. kudriavzevii and C. tropicalis) increased the apparent digestibility of dry matter by about 6.9% when compared with Crabtree-positive yeast (S. cerevisiae). Bacterial populations were highest with ensiled RS by C. tropicalis KKU20. Ensiled RS with Crabtree-negative yeasts were significantly increased with total volatile fatty acids, but they did not affect volatile fatty acid profiles. Milk protein precentage was highest at 35.6 g/kg when C. tropicalis was fed, and lowest when applied with S. cerevisiae and P. kudriavzevii KKU20 in ensiled RS at 34.5 and 34.1 g/kg, respectively. Thus, feeding ensiled RS with novel Crabtree negative yeast could improve RS digestion, rumen fermentation, and milk protein content in dairy cows.

Saccharomyces cerevisiae is a yeast strain often used to improve the feed quality of ruminants. However, S. cerevisiae has limited capacity to provide biomass when inoculated with carbon sources and a low ability to produce cellulase enzymes. Here, we hypothesized that yeast in the rumen produces a large amount of biomass and could release cellulase enzymes to break down fiber content. Therefore, the aim of this study was to screen, isolate and identify yeast from the rumen fluids of Holstein Friesian steers and measure the efficiency of biomass production and cellulase activity. A fermentation medium containing sugarcane molasses as a carbon source and urea as a nitrogen source was optimized. Two fistulated-crossbred Holstein Friesian steers averaging 350 ± 20 kg body weight were used to screen and isolate the ruminal yeast. Two experiments were designed: First, a 12 × 3 × 3 factorial was used in a completely randomized design to determine biomass and carboxymethyl cellulase activity. Factor A was the isolated yeast and S. cerevisiae. Factor B was sugarcane molasses (M) concentration. Factor C was urea (U) concentration. In the second experiment, potential yeasts were selected, identified, and analyzed for 7 × 4 factorial use in a completely randomized design. Factor A was the incubation times. Factor B was the isolated yeast strains, including codes H-Khon Kaen University (KKU) 20 (as P. kudriavzevii-KKU20), I-KKU20 (C. tropicalis-KKU20), and C-KKU20 (as Galactomyces sp.-KKU20). Isolation was imposed under aerobic conditions, resulting in a total of 11 different colonies. Two appearances of colonies including asymmetric colonies of isolated yeast (indicated as A, B, C, E, and J) and ovoid colonies (coded as D, F, G, H, I, and K) were noted. Isolated yeast from the rumen capable of providing a high amount of biomass when inoculant consisted of the molasses 15% + urea 3% (M15 + U3), molasses 25% + urea 1% (M25 + U1), molasses 25% + urea 3% (M25 + U3), and molasses 25% + urea 5% (M25 + U5) when compared to the other media solution (p < 0.01). In addition, 11 isolated biomass-producing yeasts were found in the media solution of M25 + U1. There were 4 isolates cellulase producing yeasts discovered in the media solution of M25 + U1 and M25 + U5 whereas molasses 5% + urea 1% (M5 + U1), molasses 5% + urea 3% (M5 + U3), molasses 5% + urea 5% (M5 + U5), molasses 15% + urea 1% (M15 + U1), molasses 15% + urea 3% (M5 + U3), and M25 + U3 were found with 2, 3, 1, 2, 1, and 2 isolates, respectively. Ruminal yeast strains H-KKU20, I-KKU20, and C-KKU20 were selected for their ability to produce biomass. Identification of isolates H-KKU20 and I-KKU20 revealed that those isolates belonged to Pichia kudriavzevii-KKU20 and Candida tropicalis-KKU20 while C-KKU20 was identified as Galactomyces sp.-KKU20. Two strains provided maximum cell growth: P. kudriavzevii-KKU20 (9.78 and 10.02 Log cell/mL) and C. tropicalis-KKU20 (9.53 and 9.6 Log cells/mL) at 60 and 72 h of incubation time, respectively. The highest ethanol production was observed in S. cerevisiae at 76.4, 77.8, 78.5, and 78.6 g/L at 36, 48, 60, and 72 h of incubation time, respectively (p < 0.01). The P. kudriavzevii-KKU20 yielded the least reducing sugar at about 30.6 and 29.8 g/L at 60 and 72 h of incubation time, respectively. The screening and isolation of yeasts from rumen fluids resulted in 11 different yeasts being obtained. The potential yeasts discovered in the rumen fluid of cattle were Pichia kudriavzevii-KKU20, Candida tropicalis-KKU20, and Galactomyces sp.-KKU20. P. kudriavzevii-KKU20 had higher results than the other yeasts in terms of biomass production, cellulase enzyme activity, and cell number.

The objective of this study was to compare the effects of Crabtree-negative ruminal yeast and Crabtree-positive yeast in ensiled rice straw (RS) on the ensilage quality, nutritive value, and microorganism composition, including the evaluation of the ensiled RS using the in vitro gas production technique. The experiment was conducted in a 4 × 3 factorial arrangement in a randomized complete design. Factor A was yeast species with no inoculant, Crabtree-negative yeasts (Pichia kudriavzevii KKU20 and Candida tropicalis KKU20), and Crabtree-positive yeast (Saccharomyces cerevisae), whereas factor B was ensilage times (7, 14, and 21 days). The rate of growth was revealed to be lower in Crabtree-positive yeasts than the other Crabtree-negative yeast strains (p < 0.01). RS ensiled with S. cerevisiae showed decreased dry matter (DM) content by 9.0% when compared to the sample without a yeast inoculant. In addition, organic matter (OM) content was greater (p < 0.01) for P. kudriavzevii KKU20 than C. tropicalis KKU20 and without an inoculant. Neutral detergent fiber (NDF) content was significantly decreased (p < 0.01) by yeast inoculants by about 2.75% when compared to the control group. Lactic acid bacteria (LAB) and aerobic bacteria were low (p < 0.05) when yeasts were added. However, no interaction was found between yeast and ensilage times on the quality of ensiled RS (p > 0.05). The P. kudriavzevii KKU20 addition was associated with the highest value (p < 0.01) of gas produced-an insoluble fraction (b), potential extent of gas production (a + b), and cumulative gas production at 96 h-when compared with S. cerevisiae or the control group. The highest in vitro dry matter digestibility and in vitro neutral detergent fiber digestibility was observed in RS ensiled with P. kudriavzevii KKU20 for 14 days (p < 0.01, p < 0.05). The maximum total volatile fatty acids (VFAs) at 4 and 8 h of incubation and the mean value were observed in RS ensiled with P. kudriavzevii KKU20 (p < 0.01). RS ensiled with all yeast strains showed an increased propionate concentration at 8 h (p < 0.01). In conclusion, ensiling RS with isolated Crabtree-negative ruminal yeasts could benefit feed digestion and in vitro gas production more than Crabtree-positive yeast does. P. kudriavzevii KKU20, an isolated Crabtree-negative ruminal yeast used to treat RS, had the highest potential for increasing cumulative gas production and enhancing in vitro digestibility.

The influence of sulfur included in fermented total mixed ration (FTMR) containing fresh cassava root on rumen characteristics, microbial protein synthesis, and blood metabolites in cattle was evaluated. Four Thai native beef cattle were randomly assigned according to a 2 × 2 factorial in a 4 × 4 Latin square design, and dietary treatments were as follows: factor A included a level of sulfur at 1% and 2% in total mixed ration, and factor B featured ensiling times at zero and 7 days. Digestibility of dry matter was increased when FTMR was supplemented with 2% sulfur. Blood thiocyanate increased by 69.5% when ensiling time was 7 days compared to no ensiling (p < 0.01). Bacterial populations were significantly different in the FTMR containing sulfur at 2% and 7 days of ensiling. Furthermore, microbial crude protein and efficiency of microbial protein synthesis were higher in the FTMR containing 2% sulfur and 7 days of ensiling (p < 0.01). Thus, high levels of hydrocyanic acid from fresh cassava root could be detoxified by a sulfur addition with an ensiling process to become nontoxic to cattle.

The object of this present work was to determine the effects of supplementation with pellets containing royal poinciana seed meal (PEREM) on feed use, ruminal fermentation efficiency, microbial protein synthesis, blood metabolites and mitigation of methane (CH4) emissions in cattle. The animals used in this experiment were four male Thai native beef cattle (Bos indicus) with initial body weights (BWs) of 125 ± 5.0 kg. Each of the animals were randomly assigned to receive PEREM doses at 0, 50, 100 and 150 g/d, respectively, according to a 4 × 4 Latin square design. Concentrates were fed at 0.5% BW daily, and rice straw was fed ad libitum. There were no significant differences (p > 0.05) on intakes of rice straw, concentrate and total diet. The intake of nutrients did not change among the levels of PEREM supplementation (p > 0.05), except for an intake of crude protein, which was linearly enhanced when increasing the dose of PEREM (p < 0.05). The inclusion of different doses of PEREM did not adversely affect the digestibility of dry matter, organic matter, crude protein, neutral detergent fiber and acid detergent fiber (p > 0.05). Adding various doses of PEREM did not alter ruminal pH and ruminal temperature, while concentrations of ammonia-nitrogen were significantly increased with an increased dose of PEREM supplementation (p < 0.01). The increasing doses of PEREM linearly reduced protozoal numbers (p < 0.01), with the lowest concentration when PEREM was added at 150 g. PEREM supplementation did not change (p > 0.05) the concentration of acetic acid or butyric acid or the ratio of acetic acid to propionic acid. Nevertheless, the total volatile fatty acid and propionic acid content were changed among PEREM levels (p < 0.05), which were linearly increased with an increasing dose of PEREM. At 4 h post feeding, the CH4 concentrations in the rumen of the animal were linearly reduced when the dose of pellets was increased (p < 0.01). In addition, the inclusion of PEREM did not adversely affect other blood metabolites, namely total protein, creatinine and albumin (p > 0.05). Furthermore, microbial crude protein and efficiency of microbial N synthesis were linearly enhanced when increasing levels of PEREM were added. The feeding of PEREM at 150 g/d might be an alternative with the potential to improve rumen fermentation efficiency and reduce the environmental effects produced by ruminants.
