RESUMO
We synthesised a novel membrane-insertable amphiphilic DNA. The amphiphilic DNA had a nine-nucleotide hydrophobic region at one end consisting of octyl phosphotriester linkages. The amphiphilic DNA bound to the lipid membrane by inserting the hydrophobic region; this process was facilitated by the presence of the complementary DNA strand.

The progress of a polymerase chain reaction (PCR) was sensitively monitored based on the increase in fluorescence of N,N'-bis(3-aminopropyl)-2,7-diamino-1,8-naphthyridine, which was covalently anchored on the cytosine bulge directly neighbouring the 5'-T_G-3'/5'-CCA-3' sequence in the hairpin tag at the 5' end of the PCR primer.

To investigate how protein interactions to DNA affects DNA-mediated hole transport, hole transport through the DNA bound to restriction endonuclease BamHI was examined. The DNA bound to BamHI retains a regular B-DNA type of conformation, but have a number of hydrogen bonding interaction. These hydrogen bondings in BamHI-DNA complex suppressed the guanine oxidation and the hole transport in sequence selective manner.

We examined the photoinduced DNA cleavage reaction of 32P-end labeled oligodeoxyribonucleotides which contain 5'-TGGGC-3', 5'-CGGGC-3', and 5'-TGGGT-3'. As for 5'-TGGGC-3' and 5'-TGGGT-3', the cleavage occurred most strongly at the middle of 5'-GGG-3' step, whereas the strongest cleavage occurred at the 5' side of 5'-GGG-3' in 5'-CGGGC-3' step. These results showed the 5' side base of 5'-GGG-3' step was very important for the sequence selectivity of 5'-GGG-3' cleavage.

The one-electron oxidation of PNA-DNA hybrid containing G-triplet sequence was examined. In DNA duplex G-triplet was selectively cleaved by oxidation, whereas in PNA-DNA hybrid cleavage efficiency was extremely lowered. These result suggested that cleavage efficiency of PNA-DNA hybrid was different from that of B-form DNA duplex.
