RESUMO

Inhibition of overexpressed enzymes is among the most promising approaches for targeted cancer treatment. However, many cancer-expressed enzymes are "nonlethal," in that the inhibition of the enzymes' activity is insufficient to kill cancer cells. Conventional antibody-based therapeutics can mediate efficient treatment by targeting extracellular nonlethal targets but can hardly target intracellular enzymes. Herein, we report a cancer targeting and treatment strategy to utilize intracellular nonlethal enzymes through a combination of selective cancer stem-like cell (CSC) labeling and Click chemistry-mediated drug delivery. A de novo designed compound, AAMCHO [N-(3,4,6-triacetyl- N-azidoacetylmannosamine)-cis-2-ethyl-3-formylacrylamideglycoside], selectively labeled cancer CSCs in vitro and in vivo through enzymatic oxidation by intracellular aldehyde dehydrogenase 1A1. Notably, azide labeling is more efficient in identifying tumorigenic cell populations than endogenous markers such as CD44. A dibenzocyclooctyne (DBCO)-toxin conjugate, DBCO-MMAE (Monomethylauristatin E), could next target the labeled CSCs in vivo via bioorthogonal Click reaction to achieve excellent anticancer efficacy against a series of tumor models, including orthotopic xenograft, drug-resistant tumor, and lung metastasis with low toxicity. A 5/7 complete remission was observed after single-cycle treatment of an advanced triple-negative breast cancer xenograft (~500 mm3).

A high-power all-fiber radially polarized laser system is demonstrated, in which an integrated nanograting mode convertor (S-wave plate) is used for the generation of radially polarized beam. Experimentally, a 1-W radially polarized beam was used as the seed laser, whose mode purity and mode extinction ratio (MER) were 96.5% and 98.3%, respectively. A single-stage few-mode fiber amplifier was employed to boost the 1-W seed laser to an average power of 113.2 W, when the pump power was 160 W. The corresponding slope efficiency and beam quality factor (M2) were approximately 72% and 2.3%, respectively. Moreover, the mode purity and MER of the amplified radially polarized laser were measured to be 95.7% and 97%, respectively. To the best of our knowledge, this is the highest output power from an all-fiber radially polarized laser system without obvious degradations of the mode purity and MER.

BACKGROUND Menopausal hormone therapy (MHT) has been receiving increasing attention in developed countries. The purpose of this study was to investigate understanding of menopause and acceptance of MHT in Qinhuangdao, China. MATERIAL AND METHODS We analyzed data from 186 perimenopausal patients on topics including menopausal symptoms and acceptance of and adherence to MHT treatment. We also surveyed 100 medical staff on menopausal-related knowledge. RESULTS Group A consisted of 41 patients treated with MHT for more than 1 cycle, group B consisted of 49 patients who had received MHT but had stopped it for more than 3 months, and group C consisted of 96 patients who never received MHT. There was a significant difference among them in modified Kupermann scores before treatment (P<0.05), but the difference disappeared after MHT (P>0.05). In group C, 32 patients (33%) were unaware of MHT, 60 (62.5%) were worried about the risk of breast/endometrial cancer, 24 (25%) were worried about high costs, and 67 (70%) had no obvious symptoms and did not want MHT. Similarly, in group B, most people stopped MHT for fear of breast or endometrial cancer. A survey targeting 100 medical staff in our hospital found 14 people (14%) knew about and were willing to accept MHT, 44 people (44%) knew about MHT but were afraid to use it, and 42 people (42%) did not know about MHT at all. CONCLUSIONS MHT has not yet been accepted by the majority of people, even medical staff, in Qinhuangdao, China, and much further progress is needed.

In this paper, we investigate a 1018 nm gain-switched ytterbium-doped fiber oscillator at a low repetition rate in terms of theory and experiment. Theoretically, a numerical model applicable to a 1018 nm gain-switched ytterbium-doped fiber laser was established. The influence of the pump peak power and active fiber lengths on the 1018 nm gain-switched ytterbium-doped fiber laser was numerically simulated. Experimentally, a compact 1018 nm all-fiber-structured pulsed laser oscillator is constructed, in which a pulse width of 110 ns and a single-pulse energy of 0.1 mJ were obtained. Moreover, the experimental results are in agreement with the numerical simulation ones. To the best of our knowledge, this is the first time that gain-switching technology has been applied to 1018 nm fiber lasers to generate nanosecond pulsed lasers. The model and experimental results can provide a reference for the engineering design of the same type of low repetition rate fiber lasers below the kilohertz level.

To investigate the effects of exogenous steroid hormones on growth, body color, and gonadal development in the Opsariichthys bidens (O. bidens), synthetic methyltestosterone (MT) and 17ß-estradiol (E2) were used for 28 days' treatment of 4-month-old O. bidens before the breeding season. Our results suggested that MT had a significant growth-promoting effect (P < 0.05), whereas E2 played an inhibitory role. On the body surface, the females in the MT group showed gray stripes, and the fish in other groups showed no obvious stripes. The males with MT treatment displayed brighter blue-green stripes compared to the CK and E2 groups. The histological analysis showed that the MT significantly promoted testes development in males, blocked oocyte development, and caused massive apoptosis in females, whereas the E2 group promoted ovarian development and inhibited testes development. Based on qRT-PCR analysis, in females, the expression of igf-1, dmrt1, and cyp19a1a genes revealed that E2 treatment resulted in down-regulation of igf-1 expression and up-regulation of cyp19a1a expression. In males, igf-1 and dmrt1 were significantly up-regulated after MT treatment, and E2 treatment led to down-regulation of igf-1. Therefore, this study demonstrates that MT and E2 play an important role in reversing the morphological sex characteristics of females and males.

In this paper, we propose a method for narrowing the spectrum in high-power narrow-linewidth polarization-maintaining (PM) fiber amplifiers and investigate its potential for suppressing the stimulated Brillouin scattering (SBS). In this method, in addition to common phase modulation to suppress SBS, precisely designed amplitude modulation is induced to generate self-phase modulation in a high-power PM fiber amplifier. In this co-modulation way, the spectrum can be gradually compressed along the fiber. Compared to phase modulation alone or fiber-Bragg-gratings (FBGs) based narrow-linewidth fiber oscillator schemes, in which the spectrum remains the same or broadens, this scheme can achieve a higher SBS threshold for the same output spectral linewidth. Experiments on a ∼ 3 kW peak power quasi-continuous wave (QCW) fiber amplifier show that the co-modulation scheme can compress the spectrum from 0.25 nm to 0.084 nm as output peak power increases from 13 W to 3.2 kW and enhances the SBS threshold by ∼1.7 times compared to traditional FBGs-based fiber oscillator schemes, and by ∼1.4 times compared to common phase modulation schemes. This co-modulation scheme has the potential for mitigating SBS in high-power fiber amplifiers.

In this work, a narrow-linewidth polarization-maintaining (PM) all-fiber amplifier with near-diffraction-limited beam quality and record output power is presented. First, a 4.45-kW PM fiber amplifier with a 3-dB linewidth of 0.08 nm and root mean square (rms) linewidth of 0.22 nm is achieved based on optimized phase modulation. However, the sideband of the spectrum broadens significantly during the amplification process, which is mainly caused by the additional intensity variation of the injected signal. Meanwhile, an up to 5.04-kW linearly polarized fiber laser with a relatively stable spectral bandwidth is achieved by effectively suppressing spectral broadening. At the maximum output power, the rms linewidth is 0.2 nm, the beam quality factor M2 is less than 1.3, the polarization extinction ratio (PER) is 16.5 dB, and the signal-to-noise ratio (SNR) is approximately 53 dB. The further power scaling of the amplifier is mainly limited by the pump power. To the best of our knowledge, this is the maximum output power of a narrow linewidth linearly polarized fiber amplifier to date.

Coherent beam combining (CBC) of cylindrical vector beams (CVBs) based on an active phase controller is proposed and demonstrated. Experimentally, two pieces of spatially variant wave plates (S-wave plate) were employed as vector mode convertors for two individual 1064-nm fiber amplifier channels. When the system was in a closed loop, a combined output power of 10 W and a CBC efficiency of higher than 94% were achieved, for both TM01 (radially polarized) mode and TE01 (azimuthally polarized) mode cases. Moreover, the laser system showed a high vector mode purity, which was measured to be ∼97.4% (TM01) and ∼97.3% (TE01). To the best of knowledge, this could possibly be the first demonstration of CBC of CVBs, which paves the way for power scaling of CVBs.

The CO2 aqueous foams stabilized by bioresource-derived ultra-long chain surfactants have demonstrated considerable promising application potential owing to their remarkable longevity. Nevertheless, existing research is still inadequate to establish the relationships among surfactant architecture, environmental factors, and foam properties. Herein, two cases of ultra-long chain tertiary amines with different tail lengths, N-erucamidopropyl-N,N-dimethylamine (UC22AMPM) and N-oleicamidopropyl-N,N-dimethylamine (UC18AMPM), were employed to fabricate CO2 foams. The effect of temperature, pressure and salinity on the properties of two foam systems (i.e., foamability and foam stability) was compared using a high-temperature, high-pressure visualization foam meter. The continuous phase viscosity and liquid content for both samples were characterized using rheometry and FoamScan. The results showed that the increased concentrations or pressure enhanced the properties of both foam samples, but the increased scope for UC22AMPM was more pronounced. By contrast, the foam stability for both cases was impaired with increasing salinity or temperature, but the UC18AMPM sample is more sensitive to temperature and salinity, indicating the salt and temperature resistance of UC18AMPM-CO2 foams is weaker than those of the UC22AMPM counterpart. These differences are associated with the longer hydrophobic chain of UC22AMPM, which imparts a higher viscosity and lower surface tension to foams, resisting the adverse effects of temperature and salinity.

Rapid RAFT polymerization can significantly improve production efficiency of PAM with designed molecular structure. This study shows that ideal Reversible Addition-Fragmentation Chain Transfer (RAFT) polymerization of acrylamide is achieved in dimethyl sulfoxide (DMSO) solution at 70 °C. The key to success is the appropriate choice of both a suitable RAFT chain transfer agent (CTA) and initiating species. It is illustrated that dodecyl trithiodimethyl propionic acid (DMPA) is a suitable trithiocarbonate RAFT CTA and is synthesized more easily than other CTAs. Compared to other RAFT processes of polymers, the reaction system shortens reaction time, enhances conversion, and bears all the characteristics of a controlled radical polymerization. The calculation result shows that high concentrations can reduce high conversions, accelerate the reaction rate, and widen molecular weight distributions slightly. This work proposes an excellent approach for rapid synthesis of PAMs with a restricted molecular weight distribution.

The hydrophobically associating polyacrylamide (HAPAM) is an important kind of water-soluble polymer, which is widely used as a rheology modifier in many fields. However, HAPAM products prepared in a traditional method show disadvantages including poor water solubility and the need for hydrocarbon solvents and appropriate surfactants, which lead to environmental pollution and increased costs. To solve these problems, we reported a novel kind of HAPAM "water-in-water" (w/w) emulsion and its solution properties. In this work, a series of cationic hydrophobic monomers with different alkyl chain lengths were synthesized and characterized. Then, HAPAM w/w emulsions were prepared by the aqueous dispersion polymerization of acrylamide, 2-methylacryloylxyethyl trimethyl ammonium chloride and a hydrophobic monomer. All these emulsions can be stored more than 6 months, showing excellent stability. An optical microscopy observation showed that the particle morphology and the particle size of the HAPAM emulsion were more regular and bigger than the emulsion without the hydrophobic monomer. The solubility tests showed that such HAPAM w/w emulsions have excellent solubility, which took no more than 180 s to dilute and achieve a homogeneous and clear solution. The rheology measurements showed that the HAPAM association increases with a hydrophobe concentration or the length of hydrophobic alkyl chains, resulting in better shear and temperature resistances. The total reduced viscosity was 124.42 mPa·s for cw101, 69.81 mPa·s for cw6-1, 55.38 mPa·s for cw8-0.25, 48.95 mPa·s for cw12-0.25 and 28 mPa·s for cw16-0.25 when the temperature increased from 30 °C to 90 °C. The cw8-2.0 that contains a 2 mol% hydrophobe monomer has the lowest value at 19.12 mPa·s due to the best association. Based on the excellent stability, solubility and rheological properties, we believe that these HAPAM w/w emulsions could find widespread applications.

pH-responsive viscoelastic fluids are often achieved by adding hydrotropes into surfactant solutions. However, the use of metal salts to prepare pH-responsive viscoelastic fluids has been less documented. Herein, a pH-responsive viscoelastic fluid was developed by blending an ultra-long-chain tertiary amine, N-erucamidopropyl-N, N-dimethylamine (UC22AMPM), with metal salts (i.e., AlCl3, CrCl3, and FeCl3). The effects of the surfactant/metal salt mixing ratio and the type of metal ions on the viscoelasticity and phase behavior of fluids were systematically examined by appearance observation and rheometry. To elucidate the role of metal ions, the rheological properties between AlCl3- and HCl-UC22AMPM systems were compared. Results showed the above metal salt evoked the low-viscosity UC22AMPM dispersions to form viscoelastic solutions. Similar to HCl, AlCl3 could also protonate the UC22AMPM into a cationic surfactant, forming wormlike micelles (WLMs). Notably, much stronger viscoelastic behavior was evidenced in the UC22AMPM-AlCl3 systems because the Al3+ as metal chelators coordinated with WLMs, promoting the increment of viscosity. By tuning the pH, the macroscopic appearance of the UC22AMPM-AlCl3 system switched between transparent solutions and milky dispersion, concomitant with a viscosity variation of one order of magnitude. Importantly, the UC22AMPM-AlCl3 systems showed a constant viscosity of 40 mPa·s at 80 °C and 170 s-1 for 120 min, indicative of good heat and shear resistances. The metal-containing viscoelastic fluids are expected to be good candidates for high-temperature reservoir hydraulic fracturing.

Hydrophobically associating polymers have found widespread applications in many domains due to their unique rheological behavior, which is primarily dictated by the hydrophobe content. However, the low fraction of hydrophobic monomers in polymers makes this parameter's precise and straightforward measurement difficult. Herein, a variety of hydrophobically associating polyacrylamides (HAPAM) with different alkyl chain lengths (L) and hydrophobic contents ([H]) were prepared by post-modification and accurately characterized by 1H NMR spectroscopy. The maximal fluorescence emission intensity (I) of 8-anilino-1-naphthalenesulfonic acid, which is sensitive to hydrophobic environments, was then detected in those polymer solutions and shown as a ratio to that in the polymer-free solution (I0). It was found that I/I0 for 0.5 wt% HAPAM can be scaled versus CH, which is a variate related to both L and [H], as I/I0 = 1.15 + 1.09 × 108CH3.42, which was also verified to be applicable for hydrophobic associating hydrolyzed polyacrylamide (HHAPAM). This relationship provides a handy method for determining the hydrophobic content of hydrophobically associating polymers, particularly for field applications.

Generally, the solution viscosity of wormlike micelles (WLMs) assembled from common surfactants decreases upon an increase in the temperature, following the Arrhenius law. However, abnormal thermo-thickening behavior has been repeatedly observed for WLMs formed by ultra-long-chain (≥C18) surfactants. It would be useful to unravel the mechanism behind this phenomenon. Here, three C22-tailed surfactants with an erucyl tail, two of them containing carboxylate or sulfonate head groups (UC22DAB and UC22DAS) and a cationic one with an iodide counterion (UC22DAI), and two C18-tailed betaines with olemido- and stearamidopropyl hydrophobic chains (UC18DAS and C18DAS) were characterised in terms of their viscosity and viscoelastic behavior with increasing temperature to examine the roles of head groups, tail length and tail nature. Their micellar structures were elucidated at various temperatures using small angle neutron scattering (SANS), small angle neutron X-ray scattering (SAXS) and molecular dynamics simulation. It is found that the thermo-thickening behavior of ultra-long-chain surfactants is ascribed to the prolonged persistence length and increased entanglement points. When the temperature is increased, an increase in viscosity is always accompanied by a longer persistent length, thus a larger hydrodynamic volume. The iceberg structure around the hydrophobic tail of surfactants can be destroyed at high temperatures leading to the self-assembly of these surfactants. In this self-assembly process, compared to cationic surfactants, zwitterionic surfactants can form WLMs more readily due to weak electrostatic repulsions between their head groups; the longer tails give the surfactants enhanced hydrophobicity to form WLMs with a long breaking time; the cis-unsaturation and the resulting kink in the hydrophobic tails give the surfactants good solubility, which is not conducive to the formation of micelles. In brief, the zwitterionic, longer tail and saturated tail surfactants can form WLMs with a prolonged persistence length at elevated temperatures.

For the alignment problem of strapdown inertial navigation system (SINS) under the complex environment of unknown latitude, angular oscillation interference, and line interference, the ant colony simulated annealing algorithm of gravity vector optimization is proposed to obtain the gravity apparent motion vector optimization equation, and the polynomial fitting method is proposed to simultaneously perform latitude estimation and self-alignment in combination with the alignment principle of SINS. Simulations and experiments show that the proposed method has more robust anti-interference capability than the traditional interference-based alignment method, the latitude estimation accuracy is improved by six times, the self-alignment yaw angle error RMSE value after obtaining the latitude is within 0.7°, and the roll angle and pitch angle error values are within 0.1°.

Water-soluble polymers as drag reducers have been widely utilized in slick-water for fracturing shale oil and gas reservoirs. However, the low viscosity characteristics, high operating costs, and freshwater consumption of conventional friction reducers limit their practical use in deeper oil and gas reservoirs. Therefore, a high viscosity water-soluble friction reducer (HVFR), poly-(acrylamide-co-acrylic acid-co-2-acrylamido-2-methylpropanesulphonic acid), was synthesized via free radical polymerization in aqueous solution. The molecular weight, solubility, rheological behavior, and drag reduction performance of HVFR were thoroughly investigated. The results showed that the viscosity-average molecular weight of HVFR is 23.2 × 106 g⋅mol-1. The HVFR powder could be quickly dissolved in water within 240 s under 700 rpm. The storage modulus (G') and loss modulus (G″) as well as viscosity of the solutions increased with an increase in polymer concentration. At a concentration of 1700 mg⋅L-1, HVFR solution shows 67% viscosity retention rate after heating from 30 to 90 °C, and the viscosity retention rate of HVFR solution when increasing CNaCl to 21,000 mg⋅L-1 is 66%. HVFR exhibits significant drag reduction performance for both low viscosity and high viscosity. A maximum drag reduction of 80.2% is attained from HVFR at 400 mg⋅L-1 with 5.0 mPa⋅s, and drag reduction of HVFR is 75.1% at 1700 mg⋅L-1 with 30.2 mPa⋅s. These findings not only indicate the prospective use of HVFR in slick-water hydrofracking, but also shed light on the design of novel friction reducers utilized in the oil and gas industry.

As compared to China's overall oil reserves, the reserve share of offshore oilfields is rather significant. However, offshore oilfield circumstances for enhanced oil recovery (EOR) include not just severe temperatures and salinity, but also restricted space on offshore platforms. This harsh oil production environment requires polymers with relatively strong salt resistance, solubility, thickening ability, rapid, superior injection capabilities, and anti-shearing ability. As a result, research into polymers with high viscosity and quick solubility is recognized as critical to meeting the criteria of polymer flooding in offshore oil reservoirs. For the above purposes, a novel hydrophobically associating polymer (HAP) was prepared to be used for polymer flooding of Bohai offshore oilfields. The synthetic procedure was free radical polymerization in aqueous solutions starting at 0 °C, using acrylamide (AM), acrylic acid (AA), 2-acrylamido-2-methylpropane sulfonic acid (AMPS), and poly(ethylene glycol) octadecyl methacrylate (POM) as comonomers. It was discovered that under ideal conditions, the molecular weight of HAP exceeds 2.1 × 107 g⋅mol-1. In a simulated reservoir environment, HAP has substantially greater solubility, thickening property, and salt resistance than conventional polyacrylamide (HPAM), with equivalent molecular weight. Finally, the injectivity and propagation of the two polymers in porous media were investigated. Compared with HPAM, which has a similar molecular weight, HAP solution with the concentration of 0.175% had a much better oil displacement effect in the porous medium, which can enhance oil recovery by 8.8%. These discoveries have the potential to pave the way for chemical EOR in offshore oilfields.

We demonstrate the mitigation of stimulated Brillouin scattering (SBS) in a double-clad single mode Yb-doped optical fiber amplifier through external phase modulation of narrow linewidth laser radiation using optimized periodic waveforms from an arbitrary waveform generator. Such optimized phase modulation waveforms are obtained through a multi-objective Pareto optimization based on a comprehensive model for SBS in high power narrow linewidth fiber amplifiers using Brillouin parameters determined from controlled measurements. The ability of our approach to mitigate SBS is tested experimentally as a function of RMS linewidth of the modulated optical radiation, and we measure an enhancement in SBS threshold with respect to optical linewidth of ∼ 10 GHz-1. Furthermore, we discuss the dependence of the SBS threshold enhancement on key parameters such as the amplifier length and the period of the optimized waveforms. Through simulations we find that waveforms of sufficiently long periods and optimized for a relatively long fiber (10 m) are effective for SBS suppression for shorter fibers as well. We also investigate the effect of increase in the bandwidth and amplitude of the modulation waveform on the SBS threshold enhancement observed at higher optical linewidth.

Bioinspired honeycomb-like porous films with switchable properties have drawn much attention recently owing to their potential application in scenarios in which the conversion between two opposite properties is required. Herein, the CO2-gas-triggered ON/OFF switching wettability of biocompatible polylactic acid (PLA) honeycomb porous films is fabricated. Highly ordered porous films with diameters between 2.0 and 2.8 µm are separately prepared from complexes of nonresponsive PLA and a CO2-sensitive melamine derivative [N2,N4,N6-tris(3-(dimethylamino)propyl)-1,3,5-triazine-2,4,6-triamine, MET] via the breath figure method. The hydrophilic CO2-sensitive groups can be precisely arranged in the pore's inner surface and/or top surface of the films by simply changing the PLA/MET ratio. The sensitive groups in the pore's inner surface act as a switch triggered by CO2 gas controlling water to enter the pores or not, thus resulting in ON/OFF switching wettability. The largest response of the water contact angle of honeycomb films reaches 35°, from 100 to 65°, leading to an obvious hydrophobic-hydrophilic conversion. The improved surface wettability enhances the interaction between the cell and honeycomb film surface, thus resulting in a better cell attachment. Such smart properties accompanying the biocompatible polymer and biological gas trigger facilitate possible biomedical and bioengineering applications in the future for these films.

In this paper, we demonstrate a high-power, narrow-linewidth, polarization-maintaining fiber amplifier with near-diffraction-limited beam quality. By optimizing the phase modulation signal, a nearly top-hat-shaped spectrum was generated for self-pulsing suppressing. That results in doubling the self-pulsing threshold we got from conventional white noise signal phase modulation with the same optical linewidth. Based on an optimized signal and a high power, polarization-maintaining, counter-pumped fiber amplifier, we obtain a 3.25 kW narrow-linewidth linearly polarized laser output with a linewidth of ∼20GHz, the polarization extinction ratio is about 15 dB, and the M2 is less than 1.22 at the maximum output power. To the best of our knowledge, this is the first demonstration of a narrow-linewidth, linear polarization, all-fiber amplifier with 3.25 kW laser output.
