Results of biocide efficacy testing on laboratory-generated microbial biofilms containing Legionella bozemanii is presented. These show that chlorine is effective at recommended concentrations in cooling towers; at least 48 h contact between free chlorine and the biofilm is necessary. Of five commercial biocides tested, those containing isothiazolinones and dibromonitrilo-proprionamide were most effective against sessile L. bozemanii.

The construction and operation of a laboratory biofilm generator designed to grow sessile populations of legionellas for biocide efficacy testing is described. Some examples of static biocide testing on both sessile and planktonic populations of Legionella bozemanii are discussed along with other potential applications of the apparatus.

The respiratory quinone composition of 18 strains of obligate methane-utilizing bacteria was examined. All of the strains contained lipoquinones which on examination by tlc co-chromatographed with coenzyme Q. On the basis of chromatographic and physicochemical analyses the lipoquinones produced by 10 of the strains corresponded to Q-8. Reverse-phase partition and argentation hplc demonstrated the quinone produced by the remaining 8 strains did not correspond to any known coenzyme Q prenologue. On the basis of mass spectrometry, 1H and 13C nuclear magnetic resonance spectrometry the novel quinone was shown to correspond to 2,3-dimethoxy-5-methyl-6-(18-methylene-3,7, 11,15,19,23,27,31-octamethyldotriacontahepta-2,6,10,14,22,26,30 enyl-)-1, 4-benzoquinone.

The genomic relatedness among 36 strains of pink-pigmented facultatively methylotrophic bacteria (PPFMs) was estimated by determination of DNA base composition and by DNA:DNA hybridization studies. A reproducible hybridization system was developed for the rapid analysis of multiple DNA samples. Results indicated that the PPFMs comprise four major and several minor homology groups, and that they should remain grouped in a single genus, Methylobacterium.
