RESUMO

In mice, small oocytes (primordial oocytes) are enclosed within flattened granulosa cells to form primordial follicles around birth. A small number of primordial oocytes enter the growth phase, whereas others are quiescent. The mechanism regulating this selection of primordial oocytes is not well understood. The objective of the present study was to understand the role of p27(Kip1), which regulates cell cycle progression in somatic cells, in the growth initiation of primordial oocytes in neonatal mice. We studied the localization of p27(Kip1) in 0-, 3-, 5-, 7- and 21-day-old mouse ovaries by immunohistochemistry. Ovaries from 3-day-old mice were treated with p27(Kip1) siRNAs (small interfering RNAs), and knockdown of p27(Kip1) was determined by immunohistochemistry and Western blotting. Ovaries treated with siRNAs were organ-cultured for 6 days, and oocyte growth was estimated histologically. Expression of p27(Kip1) was undetectable in the primordial oocytes of newborn mice. In the 3-day-old ovaries (n=3), p27(Kip1) was demonstrated in the nucleus of 36 ± 6% primordial oocytes. The percentage of p27(Kip1)-positive primordial oocytes increased to 72 ± 8 (n=3), 85 ± 7 (n=3) and 93 ± 5 (n=3) in the 5-, 7- and 21-day-old mouse ovaries, respectively. After knockdown of the p27(Kip1) protein by siRNAs, a higher proportion of oocytes entered the growth phase in cultured ovaries than those in the control. These results suggest that p27(Kip1) negatively regulates primordial oocyte growth and that knockdown of p27(Kip1) leads primordial oocytes to enter the growth phase in vitro.

We report the swelling properties of chemically crosslinked poly(vinyl alcohol) (PVA) gels with high degrees of polymerization and hydrolysis. Physical crosslinking by microcrystallites was introduced in this chemical PVA gel by a simple dehydration process. The equilibrium swelling ratio was measured in several mixed solvents, which comprised two-components: a good solvent (water or dimethyl sulfoxide (DMSO)), and a poor organic solvent for PVA. In the case of aqueous/organic solvent mixtures subjected to a multiple-sample test, the swelling ratio decreased continuously when the concentration of the organic solvent increased, reaching a collapsed state in the respective pure organic solvents. In the case of DMSO, starting from a swollen state, the swelling ratio rapidly decreased by between 15 and 50 mol % when the concentration of the organic compound increased in a single-sample test. To understand the hysteresis phenomenon, the swelling ratio was measured in a DMSO/acetone mixed solvent, starting from a collapsed state in acetone. The reversibility of swelling in response to successive concentration cycles between DMSO and acetone was examined. As a result, an irreversible swelling behavior was observed in the first cycle, and the swelling ratio in acetone after the first cycle became larger than the initial ratio. Subsequently, the swelling ratio changed reversibly, with a large hysteresis near a specific molar ratio of DMSO/acetone of 60/40. The microstructures were confirmed by Fourier transform infrared spectroscopy during the cycles. The irreversible swelling behavior and hysteresis are discussed in terms of the destruction and re-formation of additional physical crosslinking in the chemical PVA gels.

In this paper, the formation and destruction of hydrogen bonds in gels and in aqueous solutions of N-isopropylacrylamide (NIPA) and sodium acrylate (SA) were studied using Fourier transform infrared (FTIR) spectroscopy with attenuated total reflection (ATR). Aqueous solutions of NIPA and SA monomers with different pHs were prepared, and the ATR-FTIR spectra were obtained immediately after preparing the solution and after having been stored at room temperature for 6 months. It was found that the IR spectra evidently changed after 6 months due to polymerization, and the viscosity of a solution in the lowest pH system increased with time and finally the solution became a gel. The detailed analysis of the IR spectra indicated that the network of the gel was formed by the formation of hydrogen bonds (without crosslinker). Moreover, this physical gel exhibited the re-swelling transition by increasing the pH of solvent. The transition was caused by the destruction of hydrogen bonds due to the dissociation of carboxyl groups, which was also confirmed by the IR spectroscopy.

Calnexin (CANX) and calreticulin (CALR) chaperones mediate nascent glycoprotein folding in the endoplasmic reticulum. Here we report that these chaperones have distinct roles in male and female fertility. Canx null mice are growth retarded but fertile. Calr null mice die during embryonic development, rendering indeterminate any effect on reproduction. Therefore, we conditionally ablated Calr in male and female germ cells using Stra8 (mcKO) and Zp3 (fcKO) promoter-driven Cre recombinase, respectively. Calr mcKO male mice were fertile, but fcKO female mice were sterile despite normal mating behavior. Strikingly, we found that Calr fcKO female mice had impaired folliculogenesis and decreased ovulatory rates due to defective proliferation of cuboidal granulosa cells. Oocyte-derived, TGF-beta family proteins play a major role in follicular development and molecular analysis revealed that the normal processing of GDF9 and BMP15 was defective in Calr fcKO oocytes. These findings highlight the importance of CALR in female reproduction and demonstrate that compromised CALR function leads to ovarian insufficiency and female infertility.

The cylindrical poly(sodium acrylate) gel (SA gel) was synthesized in the glass capillary using aluminum ions as the crosslinker. The swelling ratio of the gel was measured after the repeated exchange of solvent (distilled deionized water, about pH 5.8). The gel exhibited two relaxation processes; at first the gel swells rapidly as exchange of water (the swelling process), then shrinks very slowly (the shrinking process). In order to reveal the microscopic structural change (especially, the formation of hydrogen bonding) by water exchange, attenuated total refraction (ATR) Fourier transform infrared (FT-IR) spectroscopy was applied to the gels with different swelling ratio. The IR absorption peaks of the gel were assigned based on those of poly(sodium acrylate) aqueous solutions at different pH. On the swelling process, the carboxyl groups were gradually protonated, and the intermolecular hydrogen bonding started to form in the gel with maximum swelling ratio. On the shrinking process, the formation of hydrogen bonding gradually increased with long-time repeated water exchange which resulted in the shrinkage of the gel. Effects of the repeated water exchange on the swelling behavior were discussed in terms of the exchange of counter ions and the formation of hydrogen bonding.
