RESUMO

Mitochondria form a complex, interconnected reticulum that is maintained through coordination among biogenesis, dynamic fission, and fusion and mitophagy, which are initiated in response to various cues to maintain energetic homeostasis. These cellular events, which make up mitochondrial quality control, act with remarkable spatial precision, but what governs such spatial specificity is poorly understood. Herein, we demonstrate that specific isoforms of the cellular bioenergetic sensor, 5' AMP-activated protein kinase (AMPKα1/α2/ß2/γ1), are localized on the outer mitochondrial membrane, referred to as mitoAMPK, in various tissues in mice and humans. Activation of mitoAMPK varies across the reticulum in response to energetic stress, and inhibition of mitoAMPK activity attenuates exercise-induced mitophagy in skeletal muscle in vivo. Discovery of a mitochondrial pool of AMPK and its local importance for mitochondrial quality control underscores the complexity of sensing cellular energetics in vivo that has implications for targeting mitochondrial energetics for disease treatment.

Work may influence the home domain and subsequently impact employee sleep. Past work found that negative spillover mediated the relationship between perceived unfairness about work and insomnia symptoms across 20 years. As an extension of past work, this study investigated whether negative spillover and positive spillover mediate the relationship between job demands (perceived unfairness, job discrimination) and job resources (coworker and supervisor support) on multidimensional sleep health. Two waves of survey data from a subset of full-time workers were obtained from the Midlife in the United States Study approximately 10 years apart. A sleep health composite captured irregularity, dissatisfaction, nap frequency, inefficiency, and suboptimal sleep duration (higher=more sleep health problems). PROCESS Macro evaluated cross-sectional (T1) and sequential (T1 exposureàT1 mediatoràT2 outcome) mediation pathways, adjusting for sociodemographic characteristics, physical health, neuroticism, and work hours. Both cross-sectionally and prospectively, higher negative spillover mediated the association of higher unfairness with more sleep health problems, and the association between higher discrimination and more sleep health problems. There was no support for positive spillover as a mediator between job resources and sleep health cross-sectionally or prospectively. Findings suggest that organizations should reduce the amount of negative spillover by limiting instances of unfairness and discrimination at work to promote specific aspects of employee sleep health such as sleep irregularity, dissatisfaction, efficiency, and nap frequency.

This paper examines the decision-making processes of physicians and intelligent agents within the healthcare sector, particularly focusing on their characteristics, architectures, and approaches. We provide a theoretical insight into the evolving role of artificial intelligence (AI) in healthcare, emphasizing its potential to address various healthcare challenges. Defining features of intelligent agents are explored, including their perceptual abilities and behavioral properties, alongside their architectural frameworks, ranging from reflex-based to general learning agents, and contrasted with the rational decision-making structure employed by physicians. Through data collection, hypothesis generation, testing, and reflection, physicians exhibit a nuanced approach informed by adaptability and contextual understanding. A comparative analysis between intelligent agents and physicians reveals both similarities and disparities, particularly in adaptability and contextual comprehension. While intelligent agents offer promise in enhancing clinical decisions, challenges with types of dataset biases pose significant hurdles. Informing and educating physicians about AI concepts can build trust and transparency in intelligent programs. Such efforts aim to leverage the strengths of both human and AI toward improving healthcare delivery and outcomes.

INTRODUCTION: Metabolic conditions such as diabetes, and dyslipidemia are prevalent in the United States (US), serving as potential risk factors for hepatocellular carcinoma (HCC). This study aimed to examine the association between various metabolic markers and Hounsfield Units (HU) from L1 vertebral CT scans as indicators of bone mineral density (BMD) in HCC patients under age 65. METHODS: A cross-sectional analysis was conducted on HCC patients under 65. Correlational and regression analyses were used to assess the association of metabolic markers and other health variables with HU scores. RESULTS: Race and age were significantly associated with HU scores in multivariate analyses, indicating these factors play a crucial role in bone health among HCC patients. Race showed a positive association, and age showed a negative association with HU scores. Fasting blood glucose had a significant negative correlation with BMD, but this relationship was not significant in univariate regression analysis. No significant correlations were found between HU scores and triglycerides, cholesterol, low-density lipoprotein (LDL), high-density lipoprotein (HDL), cholesterol/HDL ratio, LDL/HDL ratio, and hemoglobin A1C (HbA1c) levels. CONCLUSION: Traditional metabolic markers may not be strong predictors of osteoporosis in this specific population. Further research with larger, more diverse populations and longitudinal data is necessary to understand better the factors contributing to BMD variations in HCC patients.

Heavy rainfall is one of the major aspects of tropical cyclones (TC) and can cause substantial damages. Here, we show, based on satellite observational rainfall data and numerical model results, that between 1999 and 2018, the rain rate in the outer region of TCs has been increasing, but it has decreased significantly in the inner-core. Globally, the TC rain rate has increased by 8 ± 4% during this period, which is mainly contributed by an increase in rain rate in the TC outer region due to increasing water vapor availability in the atmosphere with rising surface temperature. On the other hand, the rain rate in the inner-core of TCs has decreased by 24 ± 3% during the same period. The decreasing trend in the inner-core rain rate likely results mainly from an increase in atmospheric stability.
